#11 - Transmission of porcine circovirus 2 (PCV2) by semen and viral distribution in different piglet tissues, p.70-76

Among six different clinical manifestations involving respiratory, enteric, nervous and reproductive signs, the postweaning multisystemic wasting syndrome (PMWS) is the most important and studied disease. However, reproductive failures associated with PCV2 have been increasingly reported. Some studies have shown the possible contamination of sows by semen of PCV2 positive boars. In order to investigate the transmission of PCV2 by contaminated semen and its ability to infect the sow and piglets, 20 PCV2 negative sows were inseminated, 10 with negative boar semen and 10 with previously nested-PCR tested positive boar semen. The sows were weekly monitored and blood samples were collected. Based on the results, 4 out 20 sows were selected (1 sow was PCR negative and inseminated with a negative semen, 2 sows were PCR negative and inseminated with a positive semen and 1 sow was PCR negative and inseminated with a positive semen, but became PCR positive around the 30 days of pregnancy). After weaning, 12 male piglets, 3 of each sow, were selected and maintained under isolation. In order to investigate which organs harbored the virus, the young pigs were necropsied around 9 months of age. Samples of serum collected monthly were tested by immunocitochemistry (ICC), and all 12 pigs serum converted. Samples of lymphoid, systemic and reproductive organs were analyzed by nested-PCR and immunohistochemistry (IHC). Evaluation of the samples by nested-PCR, revealed that several tissues were positive in 10 of 12 pigs, mainly the lymph nodes, bone marrow and spleen. Various samples were positive by IHC in 8 of 12 piglets, being the lymph nodes, tonsils and bulbourethral glands the most frequently positive. Thus, the results of testing different samples, in the 3 tests (ICC, nested-PCR and IHC) were complementary. These results show that PCV2 transmission through semen to the sows and piglets may occur and may also represent a potential risk for the herd.

E-mail: mores@cnpsa.embrapa.br A study to evaluate the distribution of granulomatous lesions in the lymph nodes of swine carcasses was accomplished. The main objective was to compare the methods of isolation of mycobacteria of the Mycobacterium avium Complex (MAC), Ziehl-Neelsen (ZN) staining, histopathological examination and immunohistochemistry (IHC) for the diagnosis of granulomatous lymphadenitis caused by MAC. A total of 431 samples of lymph nodes from 394 swine carcasses kept in 12 cold storage rooms of southern Brazil, which had the Service of Federal Inspection (SFI), were analyzed. Lymph nodes with granulomatous lesions were submitted to histological examination, ZN staining and IHC with monoclonal antibody produced with cellular extract of M. avium. The agreement between the exams was measured by the Kappa test at a 95% confidence level. The macrocospic examination done by SFI correctly identified 90.3% of granulomatous lesions, when compared with histological examination, and the majority of carcasses (92.5%) showed the lesions only in lymph nodes of the alimentary chain. Histological examination confirmed the presence of granulomatous lesions in 90.3% of the lymph nodes. The agreements between histological examination, ZN staining (Kappa: 0.342), IHC and the isolation of MAC (Kappa: 0.102) were low, however high between IHC and histology with typical presence of granulomas in the lymph nodes (Kappa: 0.973). IHC together with histological examination revealed to be efficient for identification of the granulomatous lymphadenitis caused by MAC.

Histological lesions consisted mainly of lymphohistiocytic infiltration with multinucleate giant cells in lymph nodes, spleen, Peyer’s patches, kidney, lung and liver, depletion or lymphoid hyperplasia, as well as lymphohistiocytic interstitial pneumonia and areas of secondary bronchopneumonia. The diagnosis was established through observations of the symptoms and typical lesions, and was confirmed by immunohistochemical examination and PCR. The objective of this study was to characterize the epidemiological, clinical and pathological aspects of the outbreak of PMWS, because of the severe direct or indirect economical losses caused by the disease to the world pig industry.

#14 - Fatores de risco associados à ocorrência de linfadenite em suínos na fase de crescimento-terminação

E-mail: armando@cnpsa.embrapa.br An epidemiological study in 60 units of swine termination (UT), pertaining to producers integrated with the main Agribusiness of the States of Santa Catarina and Rio Grande do Sul. These units had been selected due to the occurrence of lymphadenitis in slaughtered pigs. The batch size studied in each UT varied from 22 to 960 pigs. The animals were considered positive or negative for lymphadenitis, following the evaluation routine of the Service of Federal Inspection (SIF) at slaughter. For data collection at the UTs, a questionnaire with 156 variables was applied at a single visit to the farms. In the 60 UT studied, the percentage of positive animals for lymphadenitis was 10.37%. Among the variables analyzed, nine risk factors were identified (explicative variables) and associated with the occurrence of lymphadenitis (objective variables). The explicative variables were: bad hygiene of the feeders and drinking fountains, untreated drinking water, bad conservation of the facilities, use of the same truck for ration and animal transportation, production of ration inside the farm, free access of other animals to the ingredients of ration, storage of ready usable ration in boxes or bags and handling of the production of animals in an continuous system.

The main limitation in the use of rock phosphate in swine diets is the fluorine contente that may be toxic when used for long periods. The objective of this study was to evaluate the effects of monocalcium phosphate (MP) and Tapira phosphate (TP) to sows and their offspring when fed during four reproductive cycles, as compared to the traditional use of dicalcium phosphate (DP). The study involved 13, 16 and 13 sows for DP, MP and TP phosphates, respectively. The litters were also fed with the sarne respective phosphate treatments until they reached 95 kg of body weight. The variables analyzed were: macroscopic tooth lesions score in sows, histological evaluation of bone tissue, the thyroid and parathyroid glands, and the evaluation of bone mineral contents and bone morphometrical characteristics. The results showed that TP is toxic for sows when used continuously during 4 reproductive cycles and causes lesions of chronic fluorosis in the teeth and bone tissue. TP also affected the litters with some minor lesions of fluorosis in bone tissue at the 4th reproductive cycle. MP induced some minor inespecific lesions only in bone tissue of the sows. No deleterious effects were observed in sows and litters fed DP.

Pesquisa Veterinária Brasileira 19(1):1-6. Embrapa - Centro Nacional de Pesquisa de Suínos e Aves, Cx. Postal 21, Concórdia, SC 89700-000, Brazil. A limitação principal do uso de fosfatos naturais na alimentação de suínos é a presença de flúor que pode ser tóxico quando ingerido por longos períodos. O objetivo deste trabalho foi avaliar os eventuais efeitos deletérios dos fosfatos Monocálcico (MP) e Tapira (TP), sobre porcas e suas proles alimentadas com esses fosfatos durante quatro ciclos reprodutivos consecutivos, comparativamente com o tradicional fosfato bicálcico (DP). Foram usadas 13 porcas alimentadas com FB (testemunha) 16 com MP e 13 com TP. As leitegadas oriundas de cada ciclo também foram alimentados com os respectivos fosfatos até a idade de abate. As seguintes variáveis foram avaliadas: lesões macroscópicas nos dentes das porcas, alterações histológicas no tecido ósseo e nas glândulas tireóide e paratireóide, conteúdo de minerais no tecido ósseo e características morfométricas do osso. Os resultados obtidos mostraram que o uso de TP por 4 ciclos reprodutivos, na alimentação de suínos, provocou lesões de fluorose crônica nos dentes e tecido ósseo das porcas. As leitegadas alimentadas com este fosfato também apresentaram discretas lesões de fluorose, mas somente evidentes no 4° ciclo reprodutivo. Somente algumas alterações inespecíficas ocorreram nos ossos das porcas que receberam o MP.O DP não provocou alterações patológicas no tecido ósseo, tanto nas porcas como nas suas leitegadas.


#16 - Experimental intoxication in pigs fed with low concentrations of Senna occidentalis (Leg. Caes.) in the ration

Abstract in English:

Thirty pigs with mean weight of 27.5 kg were divided at random in tive groups. Group 1 (control) was fed with a growing ration from day O to day 42, and with a finishing ration from day 43 to day 90. Groups 2, 3, 4 and 5 received the sarne ration as the control group plus 0.25%, 0.50%, 0.75% and 1.00% respectively of ground Senna occidentJJljs (L.) Link. seeds. The regression analysis showed a negative linear regression of gowth rates Ŷ = 0.75 - 0.36x, r2 =0,92, (P<0,01)) and food consumption (Ŷ = 1,98 - 0.82x, r2 = 0,93, (P<0.07)) of growing and finishing pigs in relation with the concentration of S. occidentalis in the food. Four pigs of group 5 and one of group 3 had clinical sigos at 54, 59, 78 (two pigs) and 81 days after the beginning of the experiment. These pigs were killed in extremis until 72 hours after the onset of clinical sigos. Three pigs from group 5 and three from group 4 were killed 56 and 57 days after the beginning of the experiment. The other pigs, except three from the control group, were killed in extremis at the end of the experiment. Post mortem findings were enlargment of the liver, which had pale areas of discoloration and accentuation of its lobular pattern, in 9 pigs from groups 3, 4 and 5, one of which had a nutmeg appearance on the cut surface. No gross lesions were observed in skeletal muscles or myocardium in any of the pigs. Histological lesions of the liver were characterized by hepatocellular fat metamorphosis and discrete bile duct proliferation. One pig had centrilobular necrosis probably resultant from cardiac failure, Degeneration and segmentai necrosis of fibers were observed in skeletal muscles. The degree of these lesions was correlated directly with the quantity of S. occidentalis seeds in the ration. Mild degenerative lesions were observed in the myocardium of some pigs from groups 3, 4 and 5.

Trinta sumos com peso inicial médio de 27,15 kg foram distribuídos aleatoriamente em cinco grupos de tratamento, O grupo 1 (controle) recebeu ração de crescimento do dia O ao 42º dia de experimento e ração· de terminação do 43° ao 90º dia. Os grupos 2, 3, 4 e 5 receberam, durante o mesmo período, a mesma ração do grupo controle, porém contendo 0,25%, 0,50%, 0,75% e 1,00% de sementes de Senna occidentalis (L.) Link., respectivamente. A análise de regressão demonstrou uma relação linear negativa para o ganho de peso (Ŷ = 0,75 - 0,36x, r2 = 0,92, (P<0,01)) e o consumo alimentar (Ŷ = 1,98 - 0,82x, r2 = 0,93, (P<0,07)), em relação à concentração de S. occidentalis na ração, nas fases de crescimento e terminação. Cinco animais, quatro do grupo 5 e um do grupo 3, apresentaram sintomas de incapacitação muscular no 54°, no 59º, no 78º (dois animais) e 81º dias de experimento. Esses animais foram sacrificados in extremis até 72 horas após o início dos sintomas. Três suínos do grupo 4 e três do grupo 5 foram sacrificados no 56º e no 57° dias após o início do experimento (um desses com sintomas) e os restantes nos últimos 12 dias do experimento, com exceção de 3 animais controle que não foram sacrificados. Nas necropsias, o fígado de nove sumos dos grupos 3, 4 e 5 apresentou aumento de tamanho e áreas de coloração mais clara com acentuação do padrão lobular; em um sumo foi observado aspecto de noz-moscada na superfície de corte desse órgão. Nos másculos esqueléticos e cardíaco não foram observadas lesões macroscópicas. Histologicamente observou-se uma moderada degeneração gordurosa e uma leve proliferação de células epiteliais dos duetos biliares no fígado. Um animal apresentou acentuada necrose centrolobular, provável mente devido a insuficiência cardíaca. Os músculos esqueléticos apresentaram necrose segmentar e alterações regenerativas. O grau de lesão muscular esteve positivamente correlacionado com a concentração de S. occidentalis na ração. O músculo cardíaco apresentou processo degenerativo pouco acentuado em alguns animais dos grupos 3, 4 e 5.


#17 - Effect of pigs diet protein levei and/or acidification in the Escherichia coli post-weaning diarrhea

Abstract in English:

An experimental study was conducted to evaluate the possibility of preventing Escherichia coli post-weaning diarrhea reducing the protein level and/or acidifying the diet. Sixteen SPF (Specific Pathogen Free) Landrace piglets, 36 days old (weaning day) were used in four treatments (T) as follow: TA) control ration with 20% crude protein (CP); TB) ration similar to TA with 2% citric acid; TC) ration with 16% CP plus lisine up to 0.9%; and TD) ration similar to TC plus 2% citric acid. Rations were formulated with corn, soybean meal, minerals and vitamins. The pigs received the experimental diets from Do (weaning day) to D10 (slaughter day). All piglets were inoculated with 20 ml of E. coli culture 0141 K85 with 109 CFU/ml on the day D3 and D4. The parameters analysed were, occurrence of diarrhea, faeces elimination and intestinal colonization with the E. coli sample inoculated, gastrointestinal content pH and small intestine histology. The addition of citric acid (TB) as well as CP reduction (TC) in the diet caused a benefic effect on the E. coli diarrhea. This effect was more evident with the CP reduction and addition of the acid simultaneously (TD). The diarrhea was prevented, probably, by the reduction of stomach and jejunum pH and by avoiding an exaggerated E. coli multiplication in the intestinal content.

Estudou-se experimentalmente a possibilidade de se prevenir a diarréia pós-desmama por Escherichia coli através da redução da proteína e/ou acidificação da dieta. Utilizaram-se 16 leitões Landrace SPF (Specific Pathogen Free) com 35 dias de idade (dia da desmama), distribuídos em quatro tratamentos (T): TA - ração controle com 20% de PB; T B - ração do TA com 2% de ácido cítrico; TC - ração com 16% PB mais lisina até nível de 0,9%; TD - ração do TC mais·2% do ácido cítrico. As rações foram formuladas com milho, farelo de soja, minerais e vitaminas. Os leitões receberam as dietas experimentais do D0 (dia da desmama) até D10 (dia do sacrifício). Todos os leitões foram inoculados com 20 ml de um cultivo de E. coli 0141 K85 contendo 109 UFC/ml, nos dias D3 e D4. Os parâmetros analisados foram: ocorrência de diarréia, eliminação nas fezes e colonização intestinal da amostra de E. coli inoculada, pH do conteúdo gastrointestinal e histologia do intestino delgado. Tanto a adição do ácido cítrico (TB) como a redução da PB (TC) na dieta tiveram efeito benéfico na ocorrência da diarréia por E. coli. Este efeito mostrou-se mais evidente com a redução da PB e adição do ácido simultaneamente (T D). A diarréia foi prevenida, possivelmente pela redução do pH do estômago e jejuno e por evitar a multiplicação exagerada de E. coli no conteúdo intestinal.


#18 - Role of the superior brachygnathia in the early diagnosis of atrophic rhinitis in pigs

Abstract in English:

The objective of this work was to determine the sensitivity (α) and the specificity (β) of the superior brachygnathia (SB) test in the early diagnosis of atrophic rhinitis (AR) in pigs. 287 crossbred pigs (LW x L), 70 days old, were examined for the presence or absence of SB and AR lesions. AR positive pigs were those which had turbinate atrophy observed in cross sections of the nose. The results obtained indicated low sensitivity (α = 0,27) and high specificity (β = 0,95) to SB test for the early diagnosis of AR. Utilizing these parameters, the estimated prevalence was ṗ = 0,2115 ± 0,0823, with an error of 38,91% Thus, SB can not be used as a test in the early diagnosis of AR because of its low sensitivity and its very high level of error.

O objetivo deste trabalho foi determinar a sensibilidade (α) e especificidade (β) do teste de braquignatia superior (BS), com o intuito de avaliar sua eficiência no diagnóstico precoce da rinite atrófica (RA). Para isto, utilizaram-se 287 leitões cruzados (Large White x Landrace), com 70 dias de idade, os quais foram examinados quanto à presença ou ausência de BS e de lesões de RA. Considerou-se como suínos com RA positiva todo aquele que apresentava qualquer grau de atrofia dos cometos observados em cortes transversais do nariz. Os resultados obtidos indicaram baixa sensibilidade (α= 0,27) e alta especificidade (β = 0,95) para o teste de BS no diagnóstico precoce de RA. Utilizando-se estes parâmetros, a prevalência estimada foi ṗ = 0,2115 ± 0,823, com um erro de 38,91%. Então, as BS não pode ser usada como um teste no diagnóstico precoce da RA, ,devido à sua baixa sensibilidade e ao seu alto nível de erro.


#19 - Atrophic rhinitis of swine: effect of vaccination against Bordetella bronchiseptica in piglets challenged at an early age

Abstract in English:

The effectiveness of a bacterin in the prevention of swine atrophic rhinitis (AR) was tested. The bacterin was prepared with Bordetella bronchiseptica and adsorved to aluminum hydroxide. Six sows were vaccinated at 60 and 100 days of gestation and their litters at seven and 28 days of age. Five sows and their litters were used as an invaccinated control group. All piglets were challenged with B. bronchiseptica at three, four and five days of age. The vaccination significantly reduced the clinical signs (P < 0.001), occurrence and severity (P < 0.01) of nasal turbinate atrophy, but it did not reduce the rate of infection at 70 days of age.

Testou-se a eficiência de uma bacterina preparada com B. bronchiseptica e adsorvida a hidróxido de alumínio, na prevenção da rinite atrófica dos suínos. Seis porcas foram vacinadas, aos 60 e 100 dias de gestação, e suas leitegadas, aos sete e 28 dias de idade. Cinco leitegadas não vacinadas, nascidas de porcas não vacinadas, serviram de controle. Todos os leitões foram inoculados com B. bronchiseptica aos três, quatro e cinco dias de idade. A vacinação contribuiu para reduzir significativamente os sintomas clínicos da doença (P < 0,001) e a ocorrência e gravidade das lesões dos cornetos nasais (P < 0,01), mas não eliminou a infecção aos setenta dias de idade.
