E-mail: natalia.silva@ufra.edu.br The objective of this study was to describe the epidemiological, clinical and pathological aspects of an outbreak of periodontitis in sheep that occurred on a rural property in Benevides, in the state of Pará, Brazil. The first clinical signs of this disease in the herd – visible nodular swollen mandible - were observed about one month after the sheep began grazing on a recently renovated Panicum maximum cv. Massai pasture; the animals were also supplemented with elephant grass (Pennisetum purpureum). From the 545 adult Santa Ines, Dorper, Texel and crossbred sheep, 20 (3.7%) showed facial bulging, particularly of the mandible. These 20 sheep, which were generally over 36 months of age, had low body condition scores as well as dull and ruffled coats; many of them had loose or missing pre, upper or lower molars, showed the formation of abscesses and fistulas, exhibited pain upon palpation and had difficulty chewing. In the same herd, the prevalence of periodontal lesions at the premolars and molar teeth was evaluated via post-mortem examination of 39 young and 17 adult sheep. In 51.3% (20/39) of adolescents and in 100% of adults, periodontal lesions were found at least at one tooth of the dental arch, with unilateral or bilateral periodontal lesions at the maxillary and mandibular teeth. Histopathological analysis of 13 sheep revealed pyogranulomatous inflammation. The second and third maxillary premolars were the teeth that were most affected in young animals, whereas the third pre-molar and the molars, of the maxilla and mandible, were most often affected in adults. These lesions led to severe bone destruction, alterations in the dental arch, occlusion, tooth loss and periodontal abscesses. The epidemiological and clinic-pathological characteristics of this periodontitis outbreak, described here for the first time in sheep in Brazil, were similar to those for bovine periodontitis (“cara inchada”).

Massai, que havia sido reformada recentemente, e suplementados com capim Elefante (Pennisetum purpureum). De 545 ovinos adultos, das raças Santa Inês, Dorper, Texel e seus mestiços, 20 (3,7%) apresentaram abaulamento facial, predominantemente mandibular. Os 20 animais, na sua maioria com idades acima dos 36 meses, apresentavam baixo escore corporal, pelos arrepiados e sem brilho, alguns com afrouxamento e perda dos dentes pré-molares e molares inferiores e superiores, formação de abscesso e fístula, demonstrando dor à palpação e dificuldade na mastigação. No mesmo rebanho, a prevalência de lesões periodontais nos dentes pré-molares e molares foi avaliada por meio do exame post-mortem em 39 ovinos jovens e 17 adultos. Assim, 51,3% (20/39) dos jovens e todos os adultos apresentavam lesões periodontais em pelo menos um dos dentes da arcada, com ocorrência uni ou bilateral nos maxilares e mandibulares. A análise histopatológica de 13 animais revelou processo inflamatório piogranulomatoso. Os dentes mais afetados nas arcadas dos animais jovens foram os segundo e terceiro pré-molares maxilares. Já nos adultos foram o terceiro pré-molar e os molares, tanto maxilares quanto mandibulares. As lesões causaram severa destruição óssea, alteração na arcada dentária, na oclusão, perda dental e ocorrência de abscessos periodontais. As características epidemiológicas e clínico-patológicas do surto de periodontite em ovinos, aqui descritos originalmente nesta espécie animal no País, assemelham-se às da periodontite bovina (“cara inchada”).

E-mail: isdutra@fmva.unesp.br Periodontitis in cattle is an infectious purulent progressive disease associated with strict anaerobic subgingival biofilm and is epidemiologically related to soil management at several locations of Brazil. This study aimed to detect Treponema species in periodontal pockets of cattle with lesions deeper than 5mm in the gingival sulcus of 6 to 24-month-old animals considered periodontally healthy. We used paper cones to collect the materials, after removal of supragingival plaques, and kept frozen (at -80°C) up to DNA extraction and polymerase chain reaction (PCR) using T. amylovorum, T. denticola, T. maltophilum, T. medium and T. vincentii primers. In periodontal pocket, it was possible to identify by PCR directly, the presence of Treponema amylovorum in 73% of animals (19/26), T. denticola in 42.3% (11/26) and T. maltophilum in 54% (14/26). Among the 25 healthy sites, it was possible to identify T. amylovorum in 18 (72%), T. denticola in two (8%) and T. maltophilum in eight (32%). Treponema medium and T. vincentii were not detected over all 51 evaluated samples. The presence of Treponema amylovorum, T. maltophilum and, in particular, the widely recognized T. denticola in subgingival microflora brings an original and potencially important contribution in studies of the bovine periodontitis.

O trabalho teve por objetivo detectar espécies de Treponema presentes na bolsa periodontal de bovinos com lesões de profundidade maior que 5mm e do sulco gengival de animais com idade de 6 a 24 meses e considerados periodontalmente sadios. Os materiais foram colhidos por meio de cones de papel, após a remoção do biofilme supragengival, e mantidos sob congelamento (-80°C) até a extração do DNA e realização da reação em cadeia da polimerase (PCR) com o emprego de iniciadores de T. amylovorum, T. denticola, T. maltophilum, T. medium e T. vincentii. Na bolsa periodontal de 73% (19/26) dos animais foi possível detectar diretamente, pela PCR, a presença de Treponema amylovorum, de 42,3% (11/26) T. denticola e de 54% (14/26) T. maltophilum. Dos 25 sítios sadios, em 18 (72%) foi possível identificar T. amylovorum, em dois (8%) T. denticola e em oito (32%) T. maltophilum. Treponema medium e T. vincentii não foram detectados nas 51 amostras avaliadas. A presença de Treponema amylovorum, T. maltophilum na microbiota subgengival, e em especial do amplamente reconhecido periodontopatógeno T. denticola, traz uma contribuição original de importância potencial nos estudos da periodontite bovina.

E-mail: andrea@ufv.br There are several methods for inducing periodontal disease in animal models, being the bone defect one of the most reported. This study aimed to evaluate this model, through clinical, radiographic, tomographic and histological analyzes, thus providing standardized data for future regenerative works. Twelve dogs were subjected to the induction protocol. In a first surgical procedure, a mucoperiosteal flap was made on the buccal aspect of the right third and fourth premolars and a defect was produced exposing the furcation and mesial and distal roots, with dimensions: 5mm coronoapical, 5mm mesiodistal, and 3mm buccolingual. Periodontal ligament and cementum were curetted and the defect was filled with molding polyester, which was removed after 21 days on new surgical procedure. Clinical and radiographic examinations were performed after the two surgeries and before the collection of parts for dental tomography and histological analysis. All animals showed grade II furcation exposure in both teeth. Clinical attachment level increased after induction. Defect size did not change for coronoapical and buccolingual measurements, while mesiodistal size was significantly higher than at the time of defect production. Radiographic analysis showed decreased radiopacity and discontinuity of lamina dura in every tooth in the furcation area. The horizontal progression of the disease was evident in micro-computed tomography and defect content in the histological analysis. Therefore, it is concluded that this method promotes the induction of periodontal disease in dogs in a standardized way, thus being a good model for future work.

Este estudo objetiva avaliar esse modelo em cães, por análises clínica, radiográfica, tomográfica e histológica, fornecendo assim dados padronizados para trabalhos futuros. Doze cães foram submetidos ao protocolo de indução. Em um primeiro procedimento cirúrgico, um retalho mucoperiosteal foi produzido na face vestibular do terceiro e do quarto dentes pré-molares direitos e foi criado um defeito, expondo a furca e parte das raízes mesial e distal, com as dimensões: 5 mm corono-apical, 5 mm mesio-distal e 3 mm vestíbulo-lingual. O ligamento periodontal e o cemento foram curetados e o defeito foi então preenchido com poliéster de moldagem, que foi removido após 21 dias em um novo procedimento cirúrgico. Exames clínicos e radiográficos foram realizados após as duas cirurgias e antes da coleta dos dentes e tecidos associados para análise tomográfica e histológica. Todos os animais apresentaram exposição de furca grau II em ambos os dentes. O nível clínico de inserção aumentou após a indução. O tamanho do defeito não apresentou alteração nas medidas corono-apical e vestíbulo-lingual, enquanto o tamanho mesio-distal foi significativamente maior que o produzido. Análise radiográfica mostrou diminuição da radiopacidade e descontinuidade da lamina dura na região da furca dos dois dentes. A progressão horizontal da doença foi evidente na microtomografia e pelo conteúdo do defeito nas análises histológicas. Assim, conclui-se que este modelo promove a indução de doença periodontal em cães de forma padronizada, sendo um bom modelo para trabalhos futuros.

These have been provided by a synthetic cell biding peptide (P-15), associated to a anorganic bone matrix (ABM) to form ABM/P-15. The aim of this study was to evaluate the attachment loss and periodontal pocket response in teeth after 3 and 6 months with ABM/P-15 graft application. Twenty one dogs from the Veterinary Hospital, University of São Paulo, were anesthetized in order to accomplish periodontal treatment and 132 teeth faces with attachment loss were treated. From these, 36.4% (48 faces) received cell binding peptide and 63.6% (84 faces) compounded the control group that received conventional treatment (muco-gingival flap and root planning). The procedure was documented by intra-oral radiography and all periodontal probings were photographed. After 3 and 6 months, the animals were re-anesthetized in order to accomplish new photography, radiography and periodontal probing exams. The 48 attachment loss faces that received graft material exhibited 40% of regeneration rate after 6 months. The control faces did not change their attachment level. The palatal face presented the better regeneration rates (40%) and the canines and molars teeth showed the better responses (57.14% and 65%, respectively). There was no post-surgical infection related to absence of oral home care. It can be concluded that ABM/P-15 helps a more rapidly periodontal structure re-attachment and regeneration, including alveolar bone. Its application was easy and practical, and the post-surgical complications incidence was low. Nevertheless, more work is necessary to evaluate the amount and the quality of formed bone and periodontal ligament.

Additionally, 4 jaguars (P. onca) proceeding from wild life were captured for the study on Farm Sete, municipality of Miranda, Mato Grosso do Sul, in the southern Pantanal. All animals surveyed in captivity showed various degrees of oral lesions related to periodontal disease. The animals living in the wild did not present any clinical signs of oral lesions.

#16 - "Cara inchada" and cellular immunity in cattle

Abstract in English:

Attempts were made to study possible effects of the periodontal disease "cara inchada" (CI) on the cellular immunity of cattle, using adherence-, chemotaxis- and phagocytosis-determinations. Adherence of Bacteroides melaninogenicus to bovine granulocytes was significantly decreased in animals with CI. Phagacytosis of B. melaninogenicus by polymorphonuclear granulocytes (PMN) was also decreased in CI-diseased animals. Chemotaxis of the granulocytes appeared to be slightly increased in animals with CI and animals without CI coming from CI-affected herds.

Foram realizados estudos para verificar os possíveis efeitos da periodontite dos bezerros ("cara· inchada") sobre a imunidade celular, utilizando-se a determinação da aderência, fagocitose e quimiotaxia de Bacteroides melaninogenicus frente a granulócitos de animais com e sem lesões da enfermidade. A aderência de B. melaninoganicus foi significativamente inferior quando testada frente a granulócitos isolados de bezerros afetados pela "cara inchada" (CI). Da mesma forma, a fagocitose de B. melaninogenicuspor polirnorfonucleares (PMN) apresentou-se diminuida em animais com CI. Por outro lado, frente a B. melaninogenicus, o índice leucotático de PMN de bezerros com CI e de animais sadios do mesmo rebanho, pareceu levemente superior quando comparado com o de animais de área indene.


#17 - Microelements and the periodontal disease "cara inchada" of cattle

Abstract in English:

"Cara inchada" (swollen face), a periodontal disease of cattle (CI), is considered to be of dietetic origin. Several nutritional factors have been investigated as possible causes, including pasture micronutrient imbalances. The present study includes the assessment of microelement levels in liver and pasture samples from CI farms in central-westem and northem Brazil. Copper, molybdenum, cobalt, zinc, manganese and iron levels were determined in 83 liver samples, of which 61 were from CI animals, 17 from healthy and 4 from recovered ones. The 17 healthy bovines were also from farms positive for CI and the 4 recoverd bovines had been transferred to CI negative farms. Copper, molybdenum, sulphur, zinc, manganese and iron were analysed in 76 pasture samples, 48 from CI positive and 28 from CI negative farms. Results indicate low levels of copper in liver samples of both the diseased and healthy animals. High levels of zinc found in the livers of CI animals probably reflect their poor condition. Analysis of Cu, Mo and S in pasture samples indicate a possible influence of sulphur concentration on copper availability. It was concluded that a copper deficiency exists in the pastures where most samples were obtained, independent of the occurrence of CI in the animals. This copper deficiency, predominant in areas of fertile soils, is probably related to a Cu-Mo-S interaction and is not associated with the occurrence of CI.

A "cara inchada" dos bovinos (CI) é considerada como doença peridentária de origem alimentar. Várias hipóteses foram aventadas, levando em conta um possível desequilíbrio de microelementos na dieta animal. No presente estudo foram determinadas as concentrações de microelementos em amostras de fígado e pastagens de fazendas onde ocrre a CI, nas regiões Centro-Oeste e Norte do Brasil. Teores de cobre, molibdênio, cobalto, zinco, manganês e ferro foram dosados em 83 amostras de fígado bovino, sendo 61 amostras de animais afetados pela CI, 17 de animais sadios e 4 de animais curados. Os 17 animais sadios eram também de fazendas CI-positivas e os 4 animais curados tinham sido transferidos para fazendas indenes. Foram coletadas 48 amostras de pastos onde ocorreu a CI, e 28 de pastegens indenes. Os teores de cobre, molibdênio, enxofre, manganês e ferro foram analisados. Os resultados mostraram teores baixos de cobre tanto nos fígados de bovinos com CI, como nos bovinos sadios. Teores altos de zinco foram encontrados nos fígados dos bovinos com CI, provavelmente resultantes do mau estado nutricional dos animais. Os teores de cobre, molibdênio e enxofre encontrados nas amostras de pastagens mostraram a possível influência da concentração do enxofre sobre a disponibilidade do cobre. Concluiu-se que existe deficiência de cobre na região de onde a maioria das amostras foram coletadas, independente da ocorrência da CI. Esta deficiência, predominantemente nas regiões de terras férteis, está provavelmente relacionada à interação Cu-Mo-Se não é fator desencadeador da CI nos bovinos.


#18 - Outbreaks of periodontitis in calves ("Cara inchada") related to soil cultivation

Abstract in English:

"Cara inchada" of cattle (CI) is a periodontitis which occurs in young animals during tooth eruption and is related to tree clearance atid sowing pasture grasses in certain areas of Central Brazil once covered by forest or savanna vegetation. The findings of outbreaks of CI on two farms in the State of São Paulo are reported. In these outbreaks the calves showed severe emaciation, diarrhea, various periodontal lesions, halitosis and loss of premolar teeth of the upper jaw. On one farm the disease occurred after feeding sugar cane harvested from a newly cultivated area, in which the prevalence of periodontal lesions was 61.5% and mortality 22.1%. On the other farm, the outbreak occurred after cattle grazed on recently improved pastures. In the dry season the animals were also fed supplements of forage and silage grown on the sarne farm. The prevalence of CI in the latter was 51.9% and mortality 3.6%. It is concluded that there is a relationship between new outbreaks of periodontitis and soil and pasture improvement, i.e. ploughing, harrowing, application of lime and fertilizers.

"Cara inchada" dos bovinos (CI) é uma periodontite que os acomete na fase de dentição e está associada à formação de pastagens em determinadas áreas antes cobertas de mata ou com vegetação de cerrado no Brasil Central. Foram registrados dois surtos da doença em propriedades situadas no Estado de São Paulo, após a reforma de áreas anteriormente positivas para CI, e utilização das forrageiras cultivadas na alimentação animal. Nas duas propriedades surtos da doença haviam sido observados há mais de 10 anos atrás, após a formação inicial das pastagens em área de cerrado; no entanto, nos últimos anos não havia mais registros da ocorrência da periodontite. No ressurgimento da doença os bezerros apresentaram emagrecimento acentuado, diarréia, lesões peridentárias de diferentes intensidades, odor fétido bucal e perda dos dentes premolares decíduos maxilares. Numa das propriedades a doença ocorreu após a utilização de cana-de-açúcar, cultivada no ano anterior, na alimentação dos bezerros. A prevalência de animais com lesões peridentárias foi de 61,5%, com coeficiente de mortalidade de 22,1%. Na outra propriedade o surto ocorreu após a reforma dos pastos e das áreas destinadas à capineira e silagem de milho. A prevalência da doença foi de 51,9%, com mortalidade de 3,6%. Numa análise retrospectiva pode-se associar o ressurgimento da periodontite em bezerros nas duas propriedades com a reforma das áreas através de aragem, gradeação, calagem e adubação do solo.


#19 - Efficiency of Virginiamycin for the recovery of calves from the periodontal disease "Cara inchada"

Abstract in English:

"Cara inchada" of young bovines (CI) is an important economic and health problem for cattle raising in certain areas of newly cultivated pastures in Brazil. Affected calves develop a purulent periodontitis that leads to loss of premolar teeth, mainly of the upper jaw, to emaciation and frequently to death. To study the efficiency of Virginiamycin for the recovery of calves kept on a farm under conditions which led to an prevalence of 61.5% of CI, an experiment was performed. Seventy seven calves with progressive periodontal lesions received orally during 8 consecutive weeks 0.032g of Virginiamycin/animal 3 times a week. Two control groups of diseased non-treated calves were used: the first consisting of 10 calves maintained within the treated group of CI animals and a second group of 95 calves affected by the disease. After the 8 week period the treated calves showed a very good recovery, but there was aggravation of symptoms in the 2 groups of nontreated animals, such as bad odor from the buccal cavity, weight loss, diarrhea and shedding of premolar teeth. It was concluded that Virginiamycin was efficient for the treatment of calves with CI.

A "cara inchada" dos bovinos (CI) constitui-se num importante problema econômico-sanitário da pecuária em determinadas áreas de pastagens recém-formadas no Brasil. Bezerros acometidos desenvolvem uma periodontite purulenta progressiva, levando à perda de dentes pré-molares maxilares. Quando mantidos em propriedades com alta incidência da doença, um considerável número dos animais afetados vem a morrer por emaciação. O presente experimento teve por finalidade verificar a eficácia da Virginiamicina na recuperação de bezerros com CI mantidos sob as condições que levaram a 61,5% de incidência da doença. Assim, 77 bezerros com lesões peridentárias progressivas receberam durante 8 semanas consecutivas, por via oral, 0,032g de Virginiamicina por dose, 3 vezes por semana. Como controle foram utilizados dois grupos: um constituído por 10 bezerros que permaneceram no mesmo lote de tratamento, porém sem receber a Virginiamicina, e outro, por 95 bezerros, todos mantidos nas mesmas condições sob as quais ocorreu a doença na propriedade. Ao final do período de administração do antibiótico, o lote tratamento apresentou melhora acentuada, ao passo que os animais dos outros dois lotes tiveram agravamento do quadro, com perda de dentes, mau odor da cavidade bucal, diarréia e emagrecimento progressivo. Concluiu-se que a Virginiamicina foi eficiente na recuperação de bezerros acometidos pela CI, quando estes parâmetros foram considerados.


#20 - Effect of spiramycin in the prophylaxis of "Cara inchada" of cattle

Abstract in English:

"Cara inchada" of cattle (CI), which means "swollen face", is a purulent inflarnrnatory process of the periodontium of calves, resulting frequently in chronic ossifying periostitis and uni -or bilateral apposition of maxillary bone. Pí:evious isolation trials have demonstrated the involvement of anaerobic bacteria, rnainly of the genus Bacteroides, in the pathogenicity of the disease. To determine the effects of spiramycin in the prophylaxis of CI, two experiments were conducted on two farms with a high prevalence of the disease in previous years. On the first farm, 130 Nellore cows with their newborn calves were divided in two treatrnent groups: 1) Mineral supplementation with the addition of spirarnycin at 50g of the antibiotic per 50kg of the mineral mix; 2) the sarne mineral mix without addition of spirarnycin. On the second farm 168 Nellore cows and their calves received a mineral mix with the addition of 25g of spirarnycin per 50kg of mix, while another 274 cows and their calves were used as controls, receiving only the mineral mix without the antibiotic. The evaluation of the efficiency of the prophylactic measures was by the periodic exarnination of the maxillary gingiva of the calves. In each treatment group. The last exarnination was performed at weaning, when calves were 7 to 8 months old. No periodontal lesions were found in the calves from the mineral mix plus antibiotic treatrnent groups, whereas in the control groups the prevalence of periodontal lesions was 10.8 and 5.1 per cent respectively. The results of those experiments indicated that the periodontal lesions and the proper CI can be avoided by prophylactic adrninistration of spirarnycin. This gives evidence of the role of the bacteriae in the development of the periodontitis in calves.

A "cara inchada" dos bovinos (CI) caracteriza-se por processo inflamatório purulento do periodôncio de bezerros, que freqüentemente resulta em periostite crônica ossificante e conseqüente abaulamento uni ou bilateral da face. Trabalhos de isolamento mostraram a participação de bactérias anaeróbias, sobretudo do gênero Bacteroides, na patogenia da doença. Com a finalidade de testar a eficiência de espiramicina na profilaxia da CI, foram conduzidos experimentos em duas fazendas com elevada prevalência da doença em períodos anteriores (54% e 30% respectivamente). Na primeira propriedade foram utilizadas 130 vacas Nelore, com bezerros recém-nascidos, que foram distribuídos em dois lotes, um recebendo mineral e o outro mistura mineral contendo espiramicina, na proporção de 50g do antibiótico por 50kg de suplemento mineral. Na segunda propriedade foram utilizadas 168 vacas Nelore com bezerros recém-nascidos, recebendo mistura mineral com 25g de espiramicina por 50kg de suplemento mineral; outros 274 vacas com seus bezerros serviram de controle, recebendo apenas mistura mineral. A avaliação da eficácia da medida na profilaxia da CI foi feita através de exames clínicos da gengiva maxilar dos bezerros dos diferentes lotes. O exame final foi feito ao desmame dos bezerros, quando eles tinham aproxirnadarnente 8 meses de idade. Não foram encontradas lesões peridentárias nos bezerros dos lotes que receberam espiramicina no suplemento mineral, ao passo que nos lotes de controle a prevalência dás lesões peridentárias foi de 10,8% e 5,1 %, respectivamente. Os resultados desta experimentação indicam que as lesões peridentárias e, com isso, a CI podem ser evitados pela administração profilática de espiramicina. Isso evidencia o papel das bactérias no desenvolvimento da periodontite e suas conqüências nos bezerros.
