#11 - Serological investigation of antibodies against respiratory viruses in calves from Brazilian family farming and their relation to clinical signs of bovine respiratory disease

Abstract in English:

Bovine respiratory disease (BRD) is responsible for economic losses in cattle production. Viruses are categorized as primary etiological agents. The aims of this study were to evaluate the presence of antibodies against bovine viral diarrhea virus (BVDV), bovine herpes virus type 1 (BoHV-1), and bovine respiratory syncytial virus (BRSV) in healthy and BRD calves from family farming in relation to clinical signs of BRD. Hundred and forty-five calves were randomly selected and physical examination was performed. Only 123 animals were classified as healthy and BRD calves. Antibodies were evaluated by virus neutralization test. Person’s Chi-square test and Fisher’s exact test were performed as univariate analysis. Binary Logistic Regression was applied as multivariate analysis. Variables with P<0.10 were considered statistically significant. Variables with 0.15<P<0.10 were considered as statistical tendencies. Antibodies against BoHV-1, BVDV, and BRSV were detected in 32%, 23% and 37% animals. Antibodies against both three viruses were detected in 26.8% of calves. The presence of antibodies against BRSV were associated to normal heart rates (P=0.018) and unilateral airflow (P=0.035). Tendency was observed to unilateral airflow (P=0.06) Statistical tendencies were observed to Ab-BoHV-1 and body temperature (P=0.119) and breathing pattern (P=0.123). The profile of antibodies against respiratory viruses in calves from Brazilian family farming was firstly described. The results confirmed the importance of some clinical signs to viral infection.

A doença respiratória dos bovinos (DRB) é responsável por importantes perdas econômicas para a produção bovina, com maior impacto na agricultura familiar. Os vírus são comumente caracterizados como agentes etiológicos primários devido a mudanças na mucosa respiratória, na produção de citocinas e no funcionamento do sistema imune. Os objetivos deste estudo transversal foram avaliar a presença de anticorpos contra o vírus da diarreia viral bovina (VDVB), herpes vírus bovino tipo 1 (HVbo-1) e vírus respiratório sincicial bovino (VRSB) em bezerros sadios e com DRB de assentamentos em associação com a presença sinais clínicos de DRB. Cento e quarenta e cinco animais foram randomicamente selecionados e exame físico foi realizado. Somente 123 animais foram classificados em sadios e com DRB. Anticorpos foram identificados pelo teste de virusneutralização. Teste de qui-quadrado e teste exato de Fisher foram utilizados como análise univariada. A análise multivariada foi realizada pela regressão binária logística com o método Backward conditional. Variáveis com P<0,10 foram considerados significantes. Variáveis com 0,15<P<0,10 foram consideradas tendências. Anticorpos contra HVbo-1, VDVB e VRSB foram detectados em 32%, 23% e 37% dos animais. A presença concomitante de anticorpos contra os três vírus foi detectada em 26,8% dos bezerros. A presença de anticorpos contra VRSB foi associada à frequência cardíaca normal (P=0,018) e fluxo de ar nasal unilateral (P=0,035). Tendência estatística observada para anticorpos contra VDVB temperatura corporal (P=0,119) e padrão respiratório (P=0,123). Ao nosso conhecimento, este é o primeiro estudo que objetivou a avaliação da presença de anticorpos contra VDVB, VRSB e HVbo-1 em bezerros de assentamentos do estado de São Paulo, Brasil e sua relação com as manifestações clínicas da DRB.


#12 - Seroprevalence of Toxoplasma gondii in equidae from Northeast Brazil

Abstract in English:

This paper reports seroprevalence of toxoplasmosis in horses kept in different forms of breeding system in the state of Pernambuco. For that, 400 blood serum samples from clinically healthy horses were analyzed through the test of modified agglutination (MAT) considering cut-off of 1:25. Data related to the characteristics of the animals and herds, breeding system, presence of other animals, age, gender, breed, aptitude, and physical conditions were collected throughout investigative surveys. IgG anti-Toxoplasma gondii antibodies were detected in 12.5% (50/400) of the analyzed animals. In the 12 studied towns, there was a positivity in 91. 67% (11/12) with a variation between 4% and 33.3%. When the risk factors were evaluated, only the mesoregion factor (p=0.029) had an association with the infection, particularly the Zona da Mata region (OR=3), followed by the Recife Metropolitan Area (OR=2.2), Agreste region (OR=1.7) and Sertão region (OR=1). The results shows the presence of the parasites on the studied area, which may represent a link with the transmission chain of toxoplasmosis which has influence on the public health system, considering that Brazil is the eighth greatest exporter of equine meat in the world.

Este estudo teve como objetivo determinar a soroprevalência da toxoplasmose em equídeos mantidos em diferentes formas de manejo no estado de Pernambuco. Para tanto, um total de 400 amostras de soro sanguíneo de equídeos clinicamente saudáveis foram analisados através do teste de aglutinação modificado (MAT) considerando-se cut-off de 1:25. Dados referentes às características dos animais e dos rebanhos, sistema de criação, presença de outros animais, idade, sexo, raça, aptidão, condição física foram coletados por meio de questionários investigativos. Anticorpos IgG anti-Toxoplasma gondii foram detectados em 12,5% (50/400) dos animais analisados. Dos 12 municípios estudados, houve positividade em 91,67% (11/12) com variação entre 4,4% e 33,3%. Quando avaliados os fatores de risco, apenas o fator mesorregião (p=0,029) apresentou associação com a infecção, particularmente Zona da Mata (OR=3), seguida de Região Metropolitana do Recife (OR=2,2), Agreste (OR=1,7) e Sertão (OR=1). Os resultados revelam a presença do parasito na área estudada, o que pode representar um elo na cadeia de transmissão da toxoplasmose a qual tem repercussão em saúde pública tendo em vista que o Brasil é o oitavo maior exportador de carne equina do mundo.

E-mail: elmiro@vm.uff.br MG-F protects chickens from MG Mycoplasmosis and monitoring is done by serology (SAR and ELISA) and PCR. Histopathology is used to evaluate bird response to MG. This study evaluated MG-F profile vaccination in SPF chicken. This trial used 100 chickens, being 40 unvaccinated (G1), 40 eye-drop vaccinated at 8 weeks of age with MG-F ( Ceva Animal Health , São Paulo , SP , Brazil ) (G2) and 20 immunized by contact (G3) . Samples were obtained on the 8th, 12th, 15th, 18th, 20th and 24th week for SAR, ELISA and PCR. Fragments of trachea and air sac, for microscopy, were got after necropsies on the 15th and 24th week. Up to 12 weeks there was no significant difference among groups by SAR. SAR reactions appeared from the 15th week with these averages: G1 (1.7, 1.76 , 0.1, 0.15) , G2 (7.81, 7.65, 8.25, 6.29) and G3 (8.1, 8.5, 9.5, 6.16), while by ELISA it occurred after the 18th week with optical densities averages: G1 (0.19, 0.14, 0.16) , G2 (0.47, 0.45, 0.41) and G3 (0.55, 0.51, 0.51) . Positivity in G3 by PCR occurred seven weeks after exposure. At the 15th week the air sac score means were 0.20, 0.55, and 0.32 and 24th week were 0.15, 0.80 and 0.66 (p>0.05). For trachea, G2 (0.48) yielded higher score average than G1 (0.10) and G3 (0.00) on the 15th week. Changes in G3 were seen only at 24th week, being this average (1.00) significantly different (p<0,05) from G1 (0.08) and G2 (0.46). SAR and PCR detected MG-F in G3 earlier than ELISA. Higher tracheal changes for G2 and G3 as compared to G1 could be ascribed to MG-F vaccine infection.

Soroaglutinação Rápida (SAR), ELISA e PCR são testes no monitoramento da micoplasmose, enquanto a histopatologia, mesmo não sendo rotineira, é usada para avaliar a resposta das aves à infecção por MG. O objetivo deste estudo foi avaliar a transmissibilidade, soroconversão e alterações teciduais de MG-F em galinhas. Um total de 100 galinhas SPF foi utilizado, sendo 40 delas não vacinadas (G1), 40 vacinadas na 8ª semana de idade com MG-F (Ceva Saúde Animal, São Paulo/SP, Brasil) (G2) e 20 imunizadas por contato com aves do G2 (G3). Soros e suabes traqueais foram obtidos na 8ª, 12ª, 15ª, 18ª, 20ª, 24ª semana para monitoramento por SAR, ELISA e PCR. Fragmentos de traqueia e saco aéreo, para microscopia, foram feitas após necropsias na 15ª e 24ª semana. Até a 12ª semana não houve diferença significativa entre os grupos pela SAR. Houve reação a SAR a partir da 15ª semana com as seguintes médias: G1 (1,7; 1,76; 0,1; 0,15), G2 (7,81; 7,65; 8,25; 6,29) e G3 (8,1; 8,5; 9,5; 6,16), enquanto por ELISA a soroconversão ocorreu a partir da 18ª semana com médias de densidades óticas de G1 (0,19; 0,14; 0,16), G2 (0,47; 0,45; 0,41) e G3 (0,55; 0,51; 0,51). Todas as aves do G3 apresentaram positividade pela PCR sete semanas após exposição. Não houve diferença significativa entre as medias dos escores de saco aéreo entre os grupos, na 15ª semana (0,20; 0,55; 0,32) e 24ª semana (0,15; 0,80 e 0,66). Em relação à traqueia, G2 apresentou média maior na 15ª semana (0,48) que G3 (0,00) e G1 (0,10). Alterações em G3 foram observadas somente na 24ª semana onde as médias foram de 0,08(G1); 0,46 (G2) e 1,00 (G3), havendo significância (p<0,05) entre G1 e G3. SAR e PCR foram capazes de detectar a transmissão de MG-F de forma precoce em relação ao ELISA. Em relação ao G1 (controle negativo) as reações teciduais para os grupos vacinados foram mais intensas na 24ª semana, o que tudo indica sendo resposta à vacinação.

Like all reptiles, the black caiman has special features for energy use to control his body temperature according to climatic variations. These variations can reveal the current metabolic status of the animals, explaining the importance to study such parameters. We determined the biochemical profile of some blood compounds of 12 alligators, weighing 77.66±8.92 kg, as there are: urea (3.41±0.66), albumin (24.85±0.63), total protein (63.83±16.97), uric acid (0.25±0.09), glucose (6.99±0.77), total cholesterol (2.20±0.27), triglycerides (0.58±0.07), TGO (138.83±9.16), and ALT (12.47±2.82.

E-mail: fassisle@gmail.com The presence of anti leptospiral agglutinins (microscopic agglutination test - MAT) and DNA of leptospires was investigated in the kidney and urine (Polymerase Chain Reaction - PCR) in samples collected at the time of slaughter of cattle originating from the dairy basin of Parnaíba, Piauí, Brazil, as also the lesions in kidney, lung, liver, uterus, ovary and placenta (histopathology and immunohistochemistry). In the MAT, Hardjo was the predominant serovar with the highest number of reagent animals for the strain Hardjobovis/Sponselee. Anti-leptospiral antigens were scored in epithelial cells, interstitial vascular endothelium, endothelium of glomerular capillaries and Bowman’s capsule of 20 positive animals. Inflammatory cells were more common in the kidney. PCR was positive in urine and kidney tissue.

Antígenos anti leptospira foram marcados em 20 animais positivos nas células epiteliais e do endotélio vascular, endotélio dos capilares glomerulares e na cápsula de Bowman, somente nos animais infectados. O infiltrado inflamatório foi maior no rim do que nos demais órgãos. A PCR foi positiva em amostras de urina e tecido renal.

Serum samples of 3,102 cows (age ³ 24 months) from 373 herds were analyzed by complement fixation test. Totally, 44 (1.42%) animal were positive with titers ³ 32. The seroprevalence of Chlamydophila spp. in the herds was 8.82% (6.15%-12.17%). Four variables were associated with seroprevalence for Chlamydophila spp. in the final model of logistic regression: confined or semi-confined breeding (OR=3.339, P=0.004), farms with less than 35 cows (OR=3.339, P=0.017), abortion in the pasture (OR=2.372, P=0.037) and pasture rent (OR=3.398, P=0.006) were risk factors for Chlamydophila spp. This bacterium infected a small number of cattle from herds with abortion in Paraná state. Chlamydophila spp impact as abortion cause is reduced in this state.

E-mail: adivaldo@ufrrj.br Spirochaetes transmitted by ticks are microorganisms of worldwide distribution, which infect wild, domestic animals and human beings. A total of 300 equine sera from four municipalities: Ananideua (58), Belém (61), Castanhal (131), and Santa Izabel do Pará (50), were evaluated for Borrelia burgdorferi by an Elisa test. There were no significant differences (P<0.05) among municipalities, breed, sex or husbandry. A total of 80 (26.7%) horses were B. burgdorferi positive with titles of 1:800, 72 (90%) horses, 1:1.600, 6 (7.5%) horses, and 1:3.200, 2 (2.5%) horses. The results were similar to those in the USA, where related frequencies ranged from 7 to 75% in asymptomatic seropositive horses. The presence of anti-B.burgdorferi homologous antibodies in horses from four municipalities in the metropolitan mesorregion of Belém suggests the possibility of occurrence of human cases in the region.

A total of 300 equine sera from four municipalities: Ananideua (58), Belém (61), Castanhal (131), and Santa Izabel do Pará (50), were evaluated for Borrelia burgdorferi by an Elisa test. There were no significant differences (P<0.05) among municipalities, breed, sex or husbandry. A total of 80 (26.7%) horses were B. burgdorferi positive with titles of 1:800, 72 (90%) horses, 1:1.600, 6 (7.5%) horses, and 1:3.200, 2 (2.5%) horses. The results were similar to those in the USA, where related frequencies ranged from 7 to 75% in asymptomatic seropositive horses. The presence of anti-B.burgdorferi homologous antibodies in horses from four municipalities in the metropolitan mesorregion of Belém suggests the possibility of occurrence of human cases in the region.

The highest antibody titers predicted Australis as the infecting serogroup due to reactions against a reference strain of serovar Bratislava and a canine local isolate of serovar Australis, characterized as Leptospira noguchii. The data presented in this report highlight that a considerable fraction of capybaras in captivity may behave as reservoir for pathogenic leptospires emphasizing the occupational risk of those who deal with animal farming and slaughter.

There was an optimum agreement of results between IFAT and MAT evidenced by a Kappa test of 0.86. These results validate these tests for the detection of T. gondii infection in pigs. IFAT and MAT despite methodologies with different characteristics and readings have similar accuracy in pig serum samples.

Many species of domestic and wild animals are affected. The dog is an important reservoir in the domiciliary environment. The disease affects the joints and causes alterations in other organs. Blood serum samples from 143 dogs, living in the counties of Seropédica, Itaguaí and Rio de Janeiro, were analyzed with the indirect ELISA test, to search for antibodies against Borrelia burgdorferi latu sensu. Average positive results were 48.25%, 41.9% in Seropédica, 4.9% in Itaguaí and 1.4% in Rio de Janeiro. The titres varied from 1:400 to 1:6400. The prevalence was lower (p< 0.05) in dogs up to one year of age than in older dogs, with significant difference. Between males and females was no statistical difference with positives titres. The result of this survey should draw attention to the occurrence of Borrelia sp in the area studied, considering the importance of Lyme disease as an emerging zoonosis.
