E-mail: davetpat@ufrgs.br Bovine viral diarrhea virus (BVDV) is responsible for different syndromes that affect cattle worldwide causing important economic losses. This study analyzed the clinical, pathological, immunohistochemical and viral aspects of persistent infection by BVDV in five animals of a farm located in the county of Viamão, Rio Grande do Sul, southern Brazil. The clinical signs included growth impairment, nasal and ocular discharge and, in two animals, congenital cataract. The main gross lesions observed at the necropsy were enlargement of mesenteric lymph nodes and Peyer’s patches, and in one case, pododermatitis and crusted lesions on nasal planum and periocular region. Microscopic findings were characterized mostly by mononuclear infiltrate in the lamina propria, primarily in the small intestine and lymphoid depletion with histiocytic infiltrate in follicular centers of lymph nodes and Peyer’s patches. Viral antigens were more frequently demonstrated in epidermal keratinocytes, epithelium of hair follicles and dendritic cells of the dermis of the ears and skin, histiocytes and lymphocytes in lymph nodes, thyroid follicular cells, in the cytoplasm of neurons and to a lesser extent, in glial cells in the cerebral cortex and hippocampus. Viral isolation from blood samples and organs confirmed the presence of non-cytopathic BVDV. Moreover, viral RNA was detected by RT-PCR in serum samples. Phylogenetic analysis of a partial fragment of the5’ non-translated region of the viral genome allowed the classification of the sample as BVDV type 2b. The present study strengthens the need to investigate and to characterize BVD outbreaks and to describe its different clinic-pathological presentations.

O presente trabalho analisou as características clínicas, patológicas e imuno-histoquímicas e virais de cinco bovinos persistentemente infectados pelo BVDV de uma mesma propriedade, localizada no Município de Viamão, Rio Grande do Sul. Dentre os sinais clínicos verificados destacaram-se subdesenvolvimento, secreções nasais e oculares, além de catarata congênita unilateral em dois bovinos. As principais lesões observadas durante a necropsia consistiram de aumento dos linfonodos mesentéricos, evidenciação das placas de Peyer e pododermatite e lesões crostosas no plano nasal e na região periocular em um animal. Os achados microscópicos caracterizavam-se, principalmente, por infiltrado mononuclear na lâmina do intestino delgado e rarefação linfoide com infiltrado histiocitário nos centrofoliculares de linfonodos e nas placas de Peyer. Antígenos virais foram detectados por imuno-histoquímica principalmente em queratinócitos da epiderme, no epitélio de folículos pilosos e células mononucleares da derme de orelhas e pele; histiócitos e em linfócitos dos linfonodos; células foliculares da tireoide; no citoplasma de neurônios e, em menor escala, em células da micróglia no córtex cerebral e no hipocampo. O isolamento viral de amostras de sangue e órgãos dos animais confirmou a presença de BVDV não citopático. Também foi possível detectar a presença do genoma viral por RT-PCR no soro dos animais. A análise filogenética do fragmento parcial da região 5’ não traduzida do genoma viral permitiu a classificação da amostra viral como BVDV tipo 2b. O presente estudo reforça a necessidade de investigar e caracterizar surtos de BVD e descrever suas diferentes formas de apresentação.

E-mail: eduardofurtadoflores@gmail.com Field isolates of bovine viral diarrhea virus (BVDV) display a high genetic and antigenic diversity that may difficult diagnosis and vaccine formulation. The present study presents a genetic and antigenic characterization of 20 BVDV isolates from Rio Grande do Sul, Brazil (2000-2010). The isolates were associated with a variety of clinical conditions that included respiratory or gastroenteric disease, skin lesions, abortions, animals with retarded growth, and persistently infected (PI) animals. Most isolates (19 or 95%) belong to the non-cytopathic biotype (NCP), one isolate (5%) had a mixture of viruses NCP and cytopathic (CP). Nucleotide sequencing of a region of 270 nucleotides of the 5’ untranslated region of the viral genome followed by phylogenetic analysis identified nine isolates of BVDV-2 (45%), eight BVDV-1 (40%). Three isolates were not classified in any genotype and were classified as atypical pestiviruses. It was not possible to associate genotypes or subgenotypes with clinical conditions: both BVDV-1 and BVDV-2 were involved in different clinico - pathological syndromes. Analysis of reactivity with a panel of 19 monoclonal antibodies (mAbs) revealed a marked variability in the major envelope glycoprotein (E2/gp53) among viruses of the same genotype, but especially among viruses from different genotypes. Virus neutralization assays (VN) with antisera of BVDV-1 and BVDV-2 reference strains against the isolates revealed variable levels of cross-reactivity between viruses of the same genotype, and lack or very low reactivity between viruses of different genotypes. These results indicate a similar frequency of BVDV-1 and BVDV-2 in the cattle population of Rio Grande do Sul, confirm the remarkable antigenic diversity of BVDV and reinforce the need to include viruses of genotypes BVDV-1 and BVDV-2 in vaccines. In addition, these results indicate the circulation of atypical pestiviruses in the cattle population of RS.

As amostras foram oriundas de uma variedade de condições clínicas, que incluíam doença respiratória ou gastroentérica aguda ou crônica, lesões cutâneas, abortos, animais com crescimento retardado, além de animais persistentemente infectados (PI). A maioria das amostras (19 ou 95%) pertence ao biótipo não-citopático (NCP); enquanto um isolado apresentou uma mistura de vírus NCP e citopático (CP). O sequenciamento e análise filogenética de uma região de 270 nucleotídeos da região 5’ não-traduzida do genoma viral permitiu identificar 9 isolados de BVDV-2 (45%) e 8 isolados de BVDV-2 (40%). Três amostras não agruparam filogeneticamente com nenhum dos genótipos, sendo classificados como pestivírus atípicos. Não foi possível associar os genótipos ou subgenótipos com as condições clínicas e, tanto os BVDV-1 quanto os BVDV-2 estavam envolvidos em diferentes síndromes clínico-patológicas. Análise de reatividade com um painel de 19 anticorpos monoclonais (AcMs) revelou uma variabilidade marcante na glicoproteína principal do envelope (E2) entre vírus do mesmo genótipo, e sobretudo, entre vírus de genótipos diferentes. Testes de neutralização viral (SN) com anti-soro de cepas de referência de BVDV-1 e BVDV-2 frente às amostras isoladas revelaram níveis variáveis de reatividade cruzada entre vírus do mesmo genótipo, e reatividade muito baixa ou ausente entre vírus de genótipos diferentes. Esses resultados indicam uma frequência semelhante de BVDV-1 e BVDV-2 na população estudada, confirmam a marcante variabilidade antigênica e reforçam a necessidade de se incluir vírus dos dois genótipos nas vacinas. Finalmente, indicam a presença de pestivírus atípicos circulantes na população bovina do RS.

E-mail: giovanacamillo@yahoo.com.br Neospora caninum is a major causative agent of reproductive losses in cattle and the diagnosis of neosporosis is essential for control and eradication programs. Therefore, this study aimed to: (1) standardize an indirect immunofluorescence assay (IFAT) for detection of antibodies against N. caninum in bovine milk, starting from a standard IFAT used for blood serum samples; (2) check the agreement between antibodies detection by IFAT in blood serum and in milk of cows; (3) evaluate the suitability of IFAT for detection of anti-N. caninum antibodies in bulk milk samples. We tested samples of blood serum and milk collected from 112 lactating cows for detection of antibodies against N. caninum, furthermore, six bulk milk samples, each one corresponding to each dairy farm studied. Agreement between the detection of antibodies in blood serum (with antibody titer ³50) and in milk, with 90% of sensitivity and 100% of specificity for the IFAT in milk samples were found in 78% of the animals. However, for cows with antibody titers ³100 in blood serum, the agreement, the sensitivity and the specificity of the IFAT in milk were of 100%. It is shown that, considering the dairy farm conditions. The most appropriate diagnostic approach to be adopted regarding the collection of blood serum or milk can elect for search anti-N. caninum antibodies by IFAT. Moreover, detection of antibodies in bulk milk samples can serve for diagnosis and screening of herds with infected animals.

O diagnóstico dessa infecção é de grande importância, principalmente para programas de erradicação e controle. Sendo assim, os objetivos deste estudo foram: (1) adaptar uma reação de imunofluorescência indireta (RIFI) para detecção de anticorpos anti-N. caninum no leite, a partir de uma RIFI padronizada para a detecção desses anticorpos no soro sanguíneo, (2) analisar a concordância entre a detecção desses anticorpos pela RIFI no soro sanguíneo e no leite de fêmeas bovinas, (3) avaliar a viabilidade da RIFI para a detecção de anticorpos anti-N. caninum em amostras coletivas de leite. Foram testadas amostras de soro sanguíneo e de leite, coletadas de 112 vacas em lactação, e seis amostras coletivas de leite, correspondentes a cada uma das propriedades avaliadas. Encontrou-se 78% de concordância entre a detecção de anticorpos no soro sanguíneo (com título de anticorpos ³50) e no leite, com sensibilidade de 90% e especificidade de 100% para a RIFI nas amostras de leite. Entretanto, para as vacas com títulos de anticorpos ³100 no soro sanguíneo, tanto a concordância como os valores de sensibilidade e especificidade da RIFI no leite foram de 100%. Todas as amostras coletivas de leite foram positivas na RIFI. Isso demonstra que, conforme a propriedade pode-se eleger com segurança qual a melhor abordagem diagnóstica a ser adotada em relação à coleta de soro sanguíneo ou de leite para a pesquisa de N. caninum pela RIFI. Além disso, a determinação da presença de anticorpos em amostras coletivas de leite pode servir para diagnóstico e triagem de rebanhos com animais infectados.


#24 - A thymidine kinase-negative bovine herpesvirus 5 is highly attenuated for rabbits, but is neuroinvasive and establishes latent infection, 31(5):389-397

A thymidine kinase-negative bovine herpesvirus 5 is highly attenuated for rabbits, but is neuroinvasive and establishes latent infection. Pesquisa Veterinária Brasileira 31(5):389-397. E-mail: eduardofurtadoflores@gmail.com Mutant viral strains deleted in non-essential genes represent useful tools to study the function of specific gene products in the biology of the virus. We herein describe an investigation on the phenotype of a bovine herpesvirus 5 (BoHV-5) recombinant deleted in the gene encoding the enzyme thymidine kinase (TK) in rabbits, with special emphasis to neuroinvasiveness and the ability to establish and reactivate latent infection. Rabbits inoculated with the parental virus (SV-507/99) (n=18) at a low titer (105.5TCID50) shed virus in nasal secretions in titers up to 104.5TCID50 for up to 12 days (average: 9.8 days [5-12]) and 5/16 developed neurological disease and were euthanized in extremis. Rabbits inoculated with the recombinant BoHV-5TKD at a high dose (107.1TCID50) also shed virus in nasal secretions, yet to lower titers (maximum: 102.3TCID50) and for a shorter period (average: 6.6 days [2-11]) and remained healthy. PCR examination of brain sections of inoculated rabbits at day 6 post-infection (pi) revealed a widespread distribution of the parental virus, whereas DNA of the recombinant BoHV-5TKD-was detected only in the trigeminal ganglia [TG] and olfactory bulbs [OB]. Nevertheless, during latent infection (52pi), DNA of the recombinant virus was detected in the TGs, OBs and also in other areas of the brain, demonstrating the ability of the virus to invade the brain. Dexamethasone (Dx) administration at day 65 pi was followed by virus reactivation and shedding by 5/8 rabbits inoculated with the parental strain (mean duration of 4.2 days [1 – 9]) and by none of seven rabbits inoculated with the recombinant virus. Again, PCR examination at day 30 post-Dx treatment revealed the presence of latent DNA in the TGs, OBs and in other areas of the brain of both groups. Taken together, these results confirm that the recombinant BoHV-5TKD is highly attenuated for rabbits. It shows a reduced ability to replicate in the nose but retains the ability to invade the brain and to establish latent infection. Additional studies are underway to determine the biological and molecular mechanisms underlying the inability of BoHV-5TKD to reactivate from latency.

Este estudo relata uma investigação do fenótipo, em coelhos, de uma cepa recombinante do herpesvírus bovino tipo 5 (BoHV-5) defectiva na enzima timidina quinase (TK), com ênfase para a neuroinvasividade e capacidade de estabelecer e reativar a infecção latente. Coelhos inoculados com o vírus parental (SV-507/99, n=18) em baixo título (105,5TCID50) excretaram o vírus nas secreções nasais em títulos de até 104,5TCID50/mL por até 12 dias (média: 9,8 dias [5-12]) e 5/16 desenvolveram doença neurológica e morreram ou foram eutanasiados in extremis. Em contraste, coelhos inoculados com o recombinante BoHV-5TKD em alto título (107,1TCID50) excretaram o vírus em títulos inferiores (máximo 102,3TCID50/mL), por um período menor (média: 6,6 days [2-11]) e permaneceram saudáveis. A realização de PCR em seções do encéfalo no dia 6 pós-infecção (pi) revelou uma ampla distribuição do DNA do vírus parental, enquanto o DNA do vírus recombinante foi detectado apenas nos gânglios trigêmeos [TGs] e nos bulbos olfatórios [OBs]. Não obstante, durante a infecção latente (52pi), o DNA do vírus recombinante foi detectado nos TGs, OBs e em outros locais do encéfalo, demonstrando que o vírus recombinante mantém a neuroinvasividade. Tratamento com dexamethasona (Dx) no dia 65 pi resultou em reativação e excreção viral por 5/8 dos coelhos inoculados com o vírus parental (duração média de 4,2 dias [1-9]) e por nenhum dos sete coelhos inoculados com o vírus recombinante. No entanto, PCR realizado no dia 30 pós-Dx revelou a presença de DNA latente do BoHV-5TKD nos TGs, OBs e em outras áreas do encéfalo. Esses resultados confirmam que o recombinante BoHV-5TKD é altamente atenuado para coelhos. A sua capacidade de replicação na mucosa nasal é reduzida, mas mantém a capacidade de invadir o encéfalo e estabelecer infecção latente. Estudos adicionais estão em andamento para elucidar os mecanismos responsáveis pela incapacidade do recombinante de reativar a infecção latente.


#25 - Experimental infection of calves with recombinants of bovine herpesvirus 5 defective in glycoprotein E (gE), thymidine kinase (TK) and both, gE/TK, 31(4):319-325

E-mail: eduardofurtadoflores@gmail.com This article describes an investigation on the virulence/attenuation of bovine herpesvirus type 5 (BoHV-5) recombinants deleted in the genes encoding glycoprotein E (BoHV-5gED), thymidine kinase (BoHV-5TKD), and both gE and TK (BoHV-5gEDTKD). Seronegative calves (80 to 90 days-old) inoculated with the parental strain (SV-507/99, n=5) shed virus in nasal secretions for up to 15 days (average 10.8 days). Duration of virus shedding was 11 days for BoHV-5gD, 9.6 days for BoHV-5TKD and 6.2 days for BoHV-5gEDTKD groups. The highest titers were observed between days 1 and 6 post-infection (pi) for SV-507/99 (106.8TCID50/mL), 105.1TCID50/mL (BoHV-5gED), 105.9TCID50/mL (BoHV-5TKD) and 104.7TCID50/mL (BoHV-5gEDTKD). Calves inoculated with the parental virus presented anorexia, profound apathy and loss of body condition. Two calves were euthanized in extremis on days 10 and 11 pi; infectious virus was recovered from several areas of the brain. In contrast, calves inoculated with the recombinants remained healthy and a few presented a mild and transient nasal secretion. Dexamethasone (Dx) administration at day 42 pi resulted in virus shedding by all controls calves (mean duration 3.7 days), by 2/5 of BoHV-5TKD calves (two days) and 2/5 of BoHV-5gED (one day). No virus shedding was detected in BoHV-5gEDTKD calves upon Dx treatment. PCR examination of brain sections of calves euthanized at day 30 post Dx treatment revealed the presence of latent viral DNA widely distributed in the brain of SV-507/99 calves. Latent viral DNA was detected in a few sections (3/30) of the brains of BoHV-5gED calves and was not detected in the brains of calves inoculated with BoHV-5TKD and BoHV-5gEDTKD. These results show that the single BoHV-5 mutants (gE and tk-deleted) are attenuated for calves and establish and/or reactivate latent infection inefficiently. The double mutant BoHV-5gEDTKD is fully attenuated and appears not to establish or not reactivate efficiently from latent infection. Thus, these recombinants, especially the double mutant BoHV-5gEDTKD, display an adequate phenotype for use in modified-live vaccine formulations.

Bezerros soronegativos (80-90 dias de idade) inoculados com o vírus parental SV-507/99 (n=5) excretaram o vírus em secreções nasais por até 15 dias (média de 10,8 dias). Nos animais inoculados com os recombinantes, a duração da excreção viral foi de 11 dias (BoHV-5gED), 9,6 dias (BoHV-5TKD) e 6,2 dias (BoHV-5gEDTKD). Os maiores títulos foram observados entre os dias 1 e 6 pós-inoculação (pi), sendo de 106,8TCID50/mL para o SV-507/99, 105,1TCID50/mL (BoHV-5gED), 105,9TCID50/mL (BoHV-5TKD) e 104,7TCID50/mL (BoHV-5gEDTKD). Os bezerros inoculados com o vírus parental apresentaram anorexia e apatia; três deles mostraram apatia profunda e perda da condição corporal. Dois bezerros foram eutanasiados in extremis nos dias 10 e 11 pi, respectivamente e o vírus foi isolado de várias regiões do encéfalo. Já os bezerros inoculados com os recombinantes permaneceram saudáveis; alguns apresentaram uma secreção nasal serosa transitória. Administração de dexametasona (Dx) no dia 42 pi resultou em excreção viral por todos os bezerros inoculados com o vírus parental (duração média de 3,7 dias), por 2 de 5 bezerros dos grupos BoHV-5TKD (dois dias) e BoHV-5gED (um dia). Os bezerros inoculados com o duplo mutante BoHV-5gEDTKD não excretaram o vírus após o tratamento com Dx. Pesquisa de DNA viral por PCR no dia 30 pós-Dx revelou uma ampla distribuição do DNA do vírus parental no encéfalo; poucas seções (3/30) foram positivas no encéfalo dos animais do grupo BoHV-5gED, e não detectou-se DNA latente no encéfalo dos animais dos grupos BoHV-5TKD e BoHV-5gEDTKD. Esses resultados demonstram que os mutantes simples (gE and tk-deletados) são atenuados para bezerros e estabelecem e/ou reativam infecção latente ineficientemente. Já o duplo mutante BoHV-5gEDTKD é atenuado e parece não estabelecer e/ou não reativar eficientemente a infecção latente. Portanto, os vírus recombinantes, e em especial o duplo mutante BoHV-5gEDTKD apresentam um fenótipo compatível com a sua inclusão em vacinas vivas modificadas.

In addition to afford protection, herpesvirus vaccines should allow serological differentiation of vaccinated from naturally, latently infected animals. We previously reported the construction and characterization in vitro of a double mutant BoHV-5 (BoHV-5gE/TK”D) lacking the genes encoding thymidine kinase (tk) for attenuation, and glycoprotein E (gE) as the antigenic marker, as a vaccine candidate strain (Brum et al. 2010a). The present article reports an investigation on the attenuation and immunogenicity of this recombinant in calves. In a first experiment, 80 to 90-day-old seronegative calves (n=6) inoculated intranasally with the recombinant (titer of 107.5TCID50) shed virus in low to moderate titers in nasal secretions for up to 6 days, yet did not develop any respiratory, systemic or neurological signs of infection. At day 30 post-infection (pi) all calves had BoHV-5 specific neutralizing (VN) antibodies in titers of 4 to 8 and were negative for anti-gE antibodies in a commercial ELISA test. Administration of dexamethasone (0.1mg/kg/day during 5 days) to four of these calves at day 42 pi did not result in virus shedding or increase in VN titers, indicating lack of viral reactivation. Secondly, a group of 8-month-old calves (n=9) vaccinated intramuscularly (IM) with the recombinant virus (107.5TCID50/animal) did not shed virus in nasal secretions, remained healthy and developed VN titers from 2 to 8 at day 42 post-vaccination (pv), remaining negative for gE antibodies. Lastly, 21 calves (around 10 months old) maintained under field conditions were vaccinated IM with the recombinant virus (titer of 107.3TCID50). All vaccinated animals developed VN titers from 2 to 16 at day 30 pv. A boost vaccination performed at day 240 pv resulted in a rapid and strong anamnestic antibody response, with VN titers reaching from 16 to 256 at day 14 post-booster. Again, serum samples remained negative for gE antibodies. Selected serum samples from vaccinated animals showed a broad VN activity against nine BoHV-5 and eight BoHV-1 field isolates. These results show that the recombinant virus is attenuated, immunogenic for calves and induces an antibody response differentiable from that induced by natural infection. Thus, the recombinant BoHV-5gE/TKD is an adequate candidate strain for a modified live vaccine.

No dia 30 pós-infecção (pi), todos os animais possuíam anticorpos neutralizantes contra o BoHV-5, em títulos entre 4 e 8, mas permaneceram negativos para anticorpos contra a gE. Administração de dexametasona (Dx) a quatro desses bezerros no dia 42 pi (0.1mg/kg/dia durante 5 dias) não resultou em excreção viral ou em aumento dos títulos de anticorpos, indicando ausência de reativação viral. Em um segundo experimento, vacinação intramuscular (IM) de bezerros com 8 meses de idade (n=9) com o recombinante (107,5TCID50/animal) não resultou em excreção viral ou em manifestações clínicas. Os animais vacinados desenvolveram anticorpos neutralizantes em títulos de 2 a 8 no dia 42 pós-vacinação (PV) e permaneceram negativos para anticorpos anti-gE. Finalmente, 21 bezerros (aproximadamente 10 meses de idade) mantidos a campo foram vacinados com o recombinante (107,3TCID50) pela via IM. Todos os animais vacinados desenvolveram anticorpos neutralizantes em títulos de 2 a 16 no dia 30pv. Revacinação desses animais no dia 240 pv provocou uma resposta anamnéstica rápida e intensa, resultando em títulos neutralizantes entre 16 e 256 no dia 14 pós-revacinação. Serological studies have demonstrated the widespread presence of the virus in Brazil, however, clinical information of BT in South America are limited. This article describes clinical and pathological changes observed in sheep naturally infected by BTV in southern Brazil. In two outbreaks on different farms, 15 sheep showed clinical signs such as severe hyperthermia, apathy, swelling of the face and submandibular area, difficulty in swallowing with regurgitation, greenish mucopurulent nasal secretion, severe weight loss, and erosions in the oral mucosa. Necropsy findings in six sheep included subcutaneous edema of the face and ventral region of the chest, greenish nasal discharge, and dilated esophagus filled with abundant food contents, collapsed lungs with areas of anteroventral consolidation, and trachea and bronchi filled by foamy material mixed with food. In the heart and base of the pulmonary artery there were foci of hemorrhage. Histologically, the main changes were in cardiac and skeletal muscles and consisted of biphasic lesions characterized by hyaline and floccular degeneration/necrosis of myofibers associated with micro-mineralization and mononuclear cell infiltration. Pneumonia associated with the presence of organic matter and bacteria in the lumen of the bronchi was also observed. The diagnosis of BT was confirmed by detection of the viral genome by duplex RT-PCR in blood of affected animals, followed by the identification of BTV, serotype 12 by nucleotide sequencing.

Esse trabalho descreve alterações clínico-patológicas em ovinos acometidos pela LA no Sul do Brasil. Em dois surtos, em propriedades distintas, 15 ovinos apresentaram como principais sinais clínicos hipertermia, apatia, aumento de volume da face e região submandibular, dificuldade de deglutição com regurgitação, secreção nasal mucopurulenta esverdeada, alterações respiratórias, além de acentuada perda de peso e erosões na mucosa oral. Os achados de necropsia em seis ovinos afetados incluíram edema subcutâneo na face e região ventral do tórax, secreção nasal esverdeada, esôfago dilatado preenchido por grande quantidade de conteúdo alimentar, pulmões não colabados com áreas consolidadas anteroventrais, bem como luz da traquéia e brônquios preenchida por espuma misturada com conteúdo alimentar. No coração e base da artéria pulmonar, havia focos de hemorragia. Histologicamente, as principais alterações observadas ocorriam no tecido muscular cardíaco e esquelético, especialmente no esôfago e consistiam de lesões bifásicas caracterizadas por degeneração/necrose hialina e flocular de miofibras associadas com micro-calcificação e infiltrado inflamatório mononuclear. Pneumonia aspirativa associada à presença de material vegetal e bactérias na luz de brônquios também foi observada. O diagnóstico de LA foi confirmado pela detecção do genoma viral por duplex RT-PCR em amostras de sangue de animais afetados, seguido da identificação do VLA, sorotipo 12 por sequenciamento.


#28 - Evidence of mixed persistent infections in calves born to cows challenged with a pool of bovine viral diarrhea virus isolates, 30(12):1053-1057

E-mail: eduardofurtadoflores@gmail.com Pregnant cows infected with noncytopathic (NCP) isolates of bovine viral diarrhea virus (BVDV) between days 40 and 120 days of gestation frequently deliver immunotolerant, persistently infected (PI) calves. We herein report the characterization of PI calves produced experimentally through inoculation of pregnant cows with a pool of Brazilian BVDV-1 (n=2) and BVDV-2 isolates (n=2) between days 60 and 90 of gestation. Two calves were born virus positive, lacked BVDV antibodies, but died 7 and 15 days after birth, respectively. Six other calves were born healthy, seronegative to BVDV, harbored and shed virus in secretions for up to 210 days. Analysis of the antigenic profile of viruses infecting these calves at birth and 30 days later with a panel of monoclonal antibodies indicated two patterns of infection. Whereas three calves apparently harbored only one isolate (either a BVDV-1 or BVDV-2), co-infection by two antigenically distinct challenge viruses was demonstrated in three PI calves. Moreover, testing the viruses obtained from the blood of PI calves by an RT-PCR able to differentiate between BVDV-1 and BVDV-2 confirmed the presence/persistence of two co-infecting viruses of different genotypes (BVDV-1 and BVDV-2) in these animals. These findings indicate that persistent infection of fetuses/calves - a well characterized consequence of fetal infection by BVDV – may be established concomitantly by more than one isolate, upon experimental inoculation. In this sense, mixed persistent infections with antigenically distinct isolates may help in understanding the immunological and molecular basis of BVDV immunotolerance and persistence.

Análise antigênica dos vírus presentes nos bezerros PI com anticorpos monoclonais, ao nascimento e 30 dias após, revelou dois padrões de infecção. Três bezerros apresentavam apenas um dos vírus utilizados no desafio (BVDV-1 ou BVDV-2), enquanto outros três bezerros apresentavam co-infecção com mais de um vírus. Além disso, testando-se o sangue dos animais PI com um PCR capaz de diferenciar entre BVDV-1 e BVDV-2 observou-se a presença concomitante de vírus dos dois genótipos (BVDV-1 e BVDV-2) nestes três animais. Esses resultados indicam que infecção persistente - uma consequência bem conhecida da infecção fetal pelo BVDV - pode ser estabelecida concomitantemente por mais de um vírus, após infecção experimental. Nesse sentido, infecções persistentes mistas com isolados antigenicamente distintos podem ser úteis para o estudo das bases imunológicas e moleculares da imunotolerância e infecção persistente pelo BVDV.

E-mail: eduardofurtadoflores@gmail.com The activity of three anti-herpetic drugs (Acyclovir [ACV], Gancyclovir [GCV] and Foscarnet [PFA]) was tested against bovine herpesvirus 1 (BoHV-1), 2 (BoHV-2) and 5 (BoHV-5) in vitro using the plaque reduction assay. Different drug concentrations were tested against one hundred 50% tissue culture infectious dose (TCID50) of the respective viruses. Drug concentrations lower than 200µg/mL resulted in viability rates of more than 80% for MDBK and Hep2 cells in the MTT test (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide). The selectivity index (IS) of the drugs was calculated dividing the concentration of the drug that is cytotoxic for 50% of the cells (CC50) by the concentration of the drug that was effective in reducing by 50% the number of viral plaques (EC50) for the three herpesviruses. Thus, ACV was shown to be moderately active against BoHV-1 (EC50: 112.9mg/mL; IS: 4.5), BoHV-2 (EC50: 114.2mg/mL; IS: 4.5) and BoHV-5 (EC50: 96.9mg/mL; IS: 5.3). GCV was effective against BoHV-2 (EC50: 33.5mg/mL; IS: 16.6), moderately effective against BoHV-5 (EC50: 123.2mg/mL; IS: 4.5) and poorly active against BoHV-1 (EC50: 335.8mg/mL; IS: 1.7). PFA exhibited the highest antiviral activity, being the only drug that, at concentration of 100mg/mL, completely inhibited plaque formation by all three viruses. PFA was the most effective in vitro against BoHV-1 (EC50: 29.5mg/mL; IS: 42.2), BoHV-2 (EC50: 45.2mg/mL; IS: 27.6) and BoHV-5 (EC50: 7.8mg/mL; IS: 160.6). Thus, the results indicate that PFA is a promising candidate for experimental therapeutic testing in vivo against bovine herpesviruses.

Para isso, utilizou-se o teste de redução de placas virais em cultivo celular, testando-se diferentes concentrações dos fármacos frente a 100 doses infectantes para 50% dos cultivos celulares (DICC50) dos respectivos vírus. Pelo teste de MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide), verificou-se que concentrações inferiores a 200µg/mL dos três antivirais resultaram em índices de viabilidade de células MDBK e Hep2 superiores a 80%. Com base na concentração citotóxica para 50% das células (CC50) e na concentração dos fármacos efetiva para inibir em 50% o número de placas virais (EC50), calculou-se o índice de seletividade (IS) dos antivirais para os três herpesvírus. Assim, o ACV demonstrou ser moderadamente ativo frente ao BoHV-1 (EC50: 112,9mg/mL e IS: 4,5), ao BoHV-2 (EC50: 114,2 mg/mL e IS: 4,5) e BoHV-5 (EC50: 96,9mg/mL e IS: 5,3). O GCV apresentou atividade moderada frente ao BoHV-2 (EC50: 33,5mg/mL e IS: 16,6) e, em menor grau, contra o BoHV-5 (EC50: 123,2mg/mL e IS: 4,5), sendo ineficaz frente ao BoHV-1 (EC50: 335,8mg/mL e IS: 1,7). Rabbits inoculated with BoHV-5 in the conjunctival sac (IC) and treated with PFA (100 mg/kg/day) from day 1 to 17 post-inoculation (pi) shed less virus between days 2 and 6 pi comparing to untreated controls; this difference was significant at day 3 pi [F(9,108) = 2,23; P<0.03]. The morbidity and mortality rates of rabbits inoculated with BoHV-5 IC or intranasally (IN) were also significantly reduced in PFA-treated rabbits (50%; 11/22) comparing to untreated controls (95.4%; 21/22) (P<0.0008). In rabbits inoculated IC with BoHV-1, a reduction in virus shedding was observed in PFA-treated animals between days 1 and 4 pi; 6 and 7 pi. In addition, PFA-treated rabbits presented a longer incubation period and a shorter clinical course comparing to untreated controls (P<0.005 and P<0.04, respectively). The frequency and severity of ocular signs were also reduced in the PFA-treated group. These results demonstrate that PFA is effective against BoHV-1 and BoHV-5 in vivo and open the way towards its use in experimental therapy of herpetic infections in domestic animals.
