E-mail: silva_rcd@yahoo.com.br Toxoplasmosis is caused by the obligate intracellular protozoan parasite Toxoplasma gondii and affects warm-blooded vertebrates, including pets and man. Dogs are epidemio-logically important since they act as sentinels for the infection in humans. The present study aimed to determine the presence of antibodies to T. gondii in 205 serum samples from dogs in Ubatuba, Sao Paulo state, Brazil, through indirect immunofluorescence reaction (IFAT), as well as the risk factors related to toxoplasmosis in the animals such as breed, age, sex, access to outdoors, homemade food ingestion, access to untreated water, and contact with rodents. Toxoplasmosis-positive samples accounted for 52/205 (25.4%), with titers ranging from 16 to 256. The serological results presented significant association (P<0.05) with homemade food ingestion (45/118; 38.1%; CI95% 29.9%-47.2%) (OR=7.0; CI95% 3.0-16.6), and with access to outdoors where those that do not have access to the street were prevalent (37/121; 30.6%; CI95% 23.1%-39.3%) (OR=0.5; CI95% 0.2-1.0). These results show that toxoplasmosis in this region is related to problems of sanitary education, mainly concerning the appropriate cooking of foods, since most positive animals did not show significant association with the presence of rodents or untreated water consumption but showed, instead association with ingestion of homemade food. Thus, toxoplasmosis is a public health problem in the studied region, and sanitary measures are needed to control the infection due to the strict relationship between man and dog and the presented risk factors.

O cão apresenta importância epidemiológica por atuar como sentinela da infecção para o homem. O presente estudo objetivou determinar a ocorrência de anticorpos para T. gondii em 205 amostras de soro de cães do município de Ubatuba, SP, Brasil, pela reação de imunofluorescência indireta (RIFI), assim como os fatores de risco relacionados à infecção nos animais, como raça, idade, sexo, acesso a rua, ingestão de comida caseira, acesso a água não tratada e presença de roedores. 52/205 (25,4%) amostras foram positivas para toxoplasmose, com títulos variando de 16 a 256. Os resultados sorológicos apresentaram associação significativa (P<0,05) com consumo de comida caseira (45/118; 38,1%; IC95% 29,9%-47,2%) (OR=7,0; CI95% 3,0-16,6), e acesso a rua, em que aqueles que não tinham acesso a rua foram prevalentes (37/121; 30,6%; IC95% 23,1%-39,3%) (OR=0,5; CI95% 0,2-1,0). Estes resultados demonstram que a toxoplasmose na região está relacionada com problema de educação sanitária, principalmente quanto ao adequado cozimento dos alimentos, visto que a maioria dos animais positivos não apresentou associação significativa com presença de roedores ou consumo de água não tratada, porém os mesmos permaneciam em casa aos quais fora oferecida comida caseira. Portanto, a toxoplasmose consiste em um problema de saúde pública na região estudada, sendo necessárias medidas sanitárias para o controle da infecção, visto a estreita relação homem-cão e os fatores de risco presentes.

E-mail: fonteque@cav.udesc.br The objective was to evaluate the electrophoretic profile of proteins and serum concentration of immunoglobulin G (IgG) in Saanen goats with mastitis experimentally induced by Staphylococcus aureus (dl-a-tocopherol acetated). 14 adult goats, (supplemented with vitamin E DL-a-tocopherol) primiparous pregnant, seronegative for caprine arthritis encephalitis (CAEV), clinically healthy, were divided into two groups of seven animals: Not supplemented group (G1) and group supplemented with 2.000 UI of DL-a-tocopherol (G2 Vit E), by intramuscular injection on the day of the parturition and seven days later. At the 9th day after delivery 300 UFCs of the S. aureus ATCC 225923 strain were inoculated into the left half of the mammary gland of each animal. The mastitis was determined through collection of milk samples for evidence of infection by means of bacteriological examination, somatic cell count (SCC) and California Mastitis Test (CMT). Then samples were collected after 12, 24, 48 and 72 hours, antimicrobial intra-mammary gland treatment was initiated, with new evaluation 48 hours after treatment. The electrophoretic profile of serum protein of the goats, showed five fractions, as follows: albumin and globulin (a, b1, b2 e g-globulin). There was an increase in the production of g-globulin and lower production of b2-globulin fraction 12 hours after infection, and faster decrease in the supplemented group, showing the influence of vitamin E in the production of acute phase proteins. There was no influence of vitamin E in the serum concentration of immunoglobulin G (IgG) in supplemented animals. The supplementation with vitamin E increased the concentration of immunoglobulin and decreased the production of acute phase proteins, probably the antioxidant effect minimizing the tissue injury during the inflammatory process in the mammary gland.

Grupo não suplementado (G1) e grupo suplementado com 2.000 U.I. de acetato de dl-a-tocoferol (G2 Vit E) via intramuscular no dia do parto e sete dias após o parto. Ao nono dia do pós-parto foram inoculados 300 UFCs da cepa de S. aureus ATCC 225923, na metade esquerda da glândula mamária de cada animal. A mastite foi determinada pela colheita das amostras de leite para a comprovação da infecção, por meio de exames bacteriológicos, contagem de células somáticas (CCS) e California Mastitis Test (CMT), a partir deste momento foram efetuadas colheitas às 12, 24, 48 e 72 horas, sendo posteriormente instituído o tratamento intramamário com antimicrobiano e nova avaliação 48 horas após o tratamento. O perfil eletroforético em gel de agarose das proteínas séricas das cabras, apresentaram cinco frações, sendo: albumina e globulinas (a, b1, b2 e g). Houve aumento na produção de g-globulina e menor produção da fração b2-globulina 12 horas após a infecção, com os valores reduzindo mais rapidamente no grupo suplementado, evidenciando a influência da vitamina E na diminuição da produção das proteínas de fase aguda. Não houve influência da vitamina E na concentração sérica de imunoglobulina G (IgG) nos animais suplementados. A suplementação com vitamina E aumentou a concentração de imunoglobulinas e diminuiu a produção de proteínas de fase aguda, provavelmente pelo efeito antioxidante minimizando a lesão tecidual durante o processo inflamatório localizado na glândula mamária.

E-mail: hlangoni@fmvz.unesp.br Nowadays, farmers have great interest in milk production within an organic system due to the consumers’ claim for natural products. The purpose of this paper was to study the cytological and microbiological quality of milk produced by four farms in the county of Botucatu, SP, which use organic system management. A total of 150 mammary gland milk samples were tested by CMT and Somatic Cell Count (SCC/mL milk), microbiological examination, and Colony Former Unit Count (CFU mesophile microrganisms/mL milk). From these individual milk samples, 66 (44.0%) were CMT positive. Corynebacterium bovis was isolated from 37.9% milk samples, Staphylococcus aureus from 18.2%, S. epidermidis from 15.2%, Streptococcus uberis from 3.0%, and S. dysgalactiae from 3.0%. Association of at least two different bacteria species was observed in 7.6% samples. In respect to the whole milk of each farm, the SCC/mL values were in normal limits on three of them (i.e. lower than 400x103), and considering the CFU/mL, on three farms high levels were observed (8,5x105; 1,5x106; 4,1x105). From the individual milk samples, contagious mastitis pathogens were isolated predominantly, but environmental microorganisms were detected from the whole milk, as Escherichia coli and Pseudomonas aeruginosa, probably due to contamination during and/or after milking. The results suggest that more than mastitis problems, the four farms studied have hygienic deficiencies, what strengthens the importance of sanitary education to achieve a better milk hygiene quality.

Estudaram-se os aspectos citológicos e microbiológicos do leite no sistema orgânico de produção em quatro propriedades no município de Botucatu, SP, utilizando métodos como CMT, exame microbiológico das amostras positivas, contagem de células somáticas (CCS/mL de leite) e Contagem de Unidades Formadoras de Colônias (UFC de microrganismos mesófilos/mL de leite) em amostras individuais de leite em animais com pelo menos um teto positivo ao CMT. Foi também realizado a CCS/mL de leite e UFC/mL de leite, e exame microbiológico de amostras de leite do conjunto (tanque) de cada propriedade. Das 150 glândulas mamárias examinadas, 66 (44,00%) amostras foram positivas ao CMT, com isolamento de Corynebacterium bovis em 37,90%, Staphylococcus aureus (18,20%), S. epidermidis (15,20%), Streptococcus uberis (3,00%) e S. dysgalactiae (3,00%), e isolamento de mais de um agente bacteriano em 7,60% das amostras. Os valores de CCS/mL das amostras do leite de conjunto estiveram dentro dos limites de normalidade em três das quatro propriedades (< 400x103), por outro lado considerando a UFC/mL em três das quatro propriedades observou-se altos índices (8,5x105; 1,5x106; 4,1x105). Obteve-se o isolamento de microrganismos ambientais, como Escherichia coli e Pseudomonas aeruginosa, sugerindo a contaminação do leite durante ou após a ordenha, o que reforça a importância de atividades de educação sanitária para obtenção higiênica do leite.

Staphylococcus aureus (25.7%), Streptococcus spp. (21.4%), Corynebacterium bovis (12.9%), Streptococcus agalactiae (4.3%) and Staphylococcus spp (4,3%) were the more-frequent microorganisms isolated from animals with mastitis. Aspergillus spp. was isolated from one animal. Ceftiofur (95.2%), oxacillin (84.2%), gentamicin (76.3%) and cefoperazone (70.3%) were the more effective drugs. High resistance of isolates were found to penicillin (53.5%), ampicillin (41.6%) and neomycin (38.6%). Multiple drug resistance to three or more drug was observed in 40 (39.6%) isolates. Media of somatic cell count encountered in animals with mastitis and controls were 175,742.67cs/mL and 58,227.6 cs/mL, respectively. Antimicrobials residues in milk were detected in four (2.7%) animals. The present findings showed the low somatic cell count of animals, indicative of good quality of milk. However, pointed the need of control measures for contagious pathogens of bovine mastitis and more attention for prohibition of antimicrobial use in organic dairy farms.

Samples were collected in 17 properties located in different municipalities. Neospora agglutination test (NATe”25) was used to determine the presence of Neospora caninum in all animals. Epidemiological and reproductive data were obtained through a questionnaire which was administered in all goat pens. Significance level was set at 5% for all statistical tests. N. caninum positivity rate was 19.77%. Only one of the properties had no seropositive animal revealing N. caninum diffusion in the state. Seropositivity frequency did not correlate with sex, age, or reproductive problems. However, the presence of dogs was associated with a higher frequency of N. caninum seropositivity. A choroplethic map with hatch patterns showing the geographical distribution of goats seropositive for Neospora caninum may considerably contribute to geographic epidemiological studies and the development of disease control strategies.

E-mail: jrmodolo@fmvz.unesp.br The study evaluated the anti-Toxplasma gondii antibodies occurrence in dairy goats in São Paulo State, and also verified likely associations with age, sex, presence of cats, occurrence of reproductive problems, and potential risks to public health. Serum was obtained from 923 animals of both sex, over three months of age from 17 properties in different cities. For the diagnosis, the indirect immunofluorescence reaction (RIFIe”16) and a questionnaire about health to collect epidemiological information and the reproductive cycle of every goat were used. Among all properties, 15 foci of T. gondii were diagnosed with seropositivity ranging from 2.70% to 81.25%. Neither association between the frequency of seropositivity and the sex of animals nor occurrence of reproduction failure of goats was identified. There was positive influence of anti-T. gondii antibody rate due to the older age of the animals and the presence of cats in pens. The risk of transmitting T. gondii to the public health must be considered, once in some properties where the positive serology was verified natural, products such as milk and/or meat are commercialized. Results were discussed at 5% level of significance.

E-mail: jrmodolo@fmvz.unesp.br The study evaluated the anti-Toxplasma gondii antibodies occurrence in dairy goats in São Paulo State, and also verified likely associations with age, sex, presence of cats, occurrence of reproductive problems, and potential risks to public health. Serum was obtained from 923 animals of both sex, over three months of age from 17 properties in different cities. For the diagnosis, the indirect immunofluorescence reaction (RIFIe”16) and a questionnaire about health to collect epidemiological information and the reproductive cycle of every goat were used. Among all properties, 15 foci of T. gondii were diagnosed with seropositivity ranging from 2.70% to 81.25%. Neither association between the frequency of seropositivity and the sex of animals nor occurrence of reproduction failure of goats was identified. There was positive influence of anti-T. gondii antibody rate due to the older age of the animals and the presence of cats in pens. The risk of transmitting T. gondii to the public health must be considered, once in some properties where the positive serology was verified natural, products such as milk and/or meat are commercialized. Results were discussed at 5% level of significance.

The aim of the present work was to evaluate the association among the kinetics of humoral immune response, the recovery ofviable leptospiras and the intensity ofthe tissue lesions in mice selected for high (H) and low (L) production of antibodies (selection N-A) and in mice from the outbreed Balb/c Iine, inoculated with pathogenic Leptospira interrogans serovar icterohaemorrhagiae. Toe H and L lines showed modifications in some steps ofthe immune response, mainly in relation to the macrophagic activity, representing extreme phenotypes found in heterogeneous natural populations. Mice were sacrificed in eight moments after the infection. Analysis of the results revealed that from the 7th day after the infection, on Iine H mice presented antibodies titles significantly higher as compared to the L line mice, maintaining the multispecific effect Balb/c tine mice showed intermediàte results between the two Iines. Antibodies production worked as a timiting factor to the infection, becausewhen a greater leptospiras recovery was obtained, at the initial phase of the infection, the titles of antibodies were elevated. Inflammatory and degenerative process led to similar lesions in the organs ofinfected mice. Only a variation in the degree oftissue compromising was observed according to the tine. H tine mice showed more extensive lesions than L and Balb/c lines mice. For the Balb/c tine mice, the lesions were moderate. As a mie, H and Balb/c mice lines showed a Th2 profile of immune response, with higher indexes of antibody production and gravity ofthe lesions, while L line mice presented a Th 1 profile ofimmune response.

O presente trabalho teve por finalidade associar a cinética da resposta humoral à recuperação de leptospiras viáveis e à intensidade das lesões teciduais em camundongos geneticamente selecionados para bons (High) e maus (Low) produtores de anticorpos (seleção IV-A), além de camundongos outbreed, Balb/c, inoculados com amostra patogênica de Leptospira interrogans sorovar icterohaemorrhagiae. As linhagens High e Low (seleção IV-A) apresentam modificações em alguns compartimentos da resposta imune, principalmente em relação à atividade macrofágica, representando fenótipos extre mos encontrados em populações naturais heterogêneas. Os camundongos foram sacrificados em oito momentos após a infecção. A análise dos resultados revelou que a partir do 7º dia após a infecção, os camundongos da linhagem High apresentaram elevação nos títulos de anticorpos estatisticamente significantes quando comparados aos camundongos da linhagem Low, mantendo assim o efeito multiespecífico. Os camundongos Balb/c apresentaram resultados intermediários entre as duas linhagens. A produção de anticorpos colaborou como fator limitante à infecção, pois quando obtevese maior recuperação de leptospiras, na fase inicial da infecção, os títulos de anticorpos encontravam-se em elevação. As lesões observadas nos órgãos de camundongos infectados consistiram basicamente nos mesmos processos inflamatórios e degenerativos, que não se alteraram, variando apenas o grau de comprometimento tecidual, de acordo com a linhagem. A linhagem high apresentou lesões mais extensas que as apresentadas pelas linhagens low e Balb/c, sendo que nesta última as lesões foram moderadas. De forma geral a linhagem High e Balb/c apresentaram um perfil de resposta Th2, com o maior índice de produção de anticorpos e gravidade das lesões, enquanto a linhagem Low apresentou um perfil de resposta Th 1.


#28 - Etiology and treatment of bovine mastitis with the help of dimethyl sulfoxide

Abstract in English:

A study was performed in 2 dairy farms on the etiology of bovine mastitis, its control by sanitary measures as well as by treatment with drugs, alone or in combination with dimethyl sulfoxide (DMSO). Staphylococci, streptococci and coliform bacteria were responsible for 79,7% of the cases of mastitis; staphylococci alone caused 55,6%. The measures adopted reduced levels of mastitis from 38 and 21% respectively to 2,7 and 3,1% on the 2 farms. Statistical analysis showed that the combination of drugs with 20% DMSO significantly improved treatments with nitrofurantoin (P < 0.01), chloranphenicol (P < 0.05) and penicilin+streptomycin (P=0,1-0,2). recommending the use of DMSO as vehicle for drugs in the treatment of mastitis.

Em duas granjas leiteiras procedeu-se à Pesquisa sobre a etiologia das mastites bovinas, seu controle por medidas de manejo sanitário e terapêutico com drogas comuns e por sua associação ao dimetilsulfóxido (DMSO), durante 9 trimestres. Os principais agentes foram estafilococos, estreptococos e bactérias coliformes que foram responsáveis por 79 ,7% dos casos, devendo-se aos estafilococos 55 ,6% das mastites. As medidas adotadas reduziram as taxas de mastites de 38 e 21 % respectivamente, para 2,7 e 3,1% em ambas as granjas. A análise estatística mostrou que a associação das drogas ao DMSO a 20% melhorou o índice de curas, com significância (P < 0,01) para o furacin, (P < 0,05) para o cloranfenicol e (P = 0,1-0,2) para a penicilina +estreptomicina, recomendando seu uso como veículo para drogas antimastíticas.
