ABSTRACT.- Galindo C.M., Hemckmeier D., Biondo N., Parizotto L.H., Ogliari D. & Gava A. 2017. [Spontaneous and experimental poisoning by tifton 68 (Cynodon nlemfuensis Vanderyst) in cattle.] E-mail: clau.diem@uol.com.br Tifton 68 (Cynodon nlemfuensis Vanderyst) is a grass cultivated in southern Brazil which causes peracute clinical manifestations including dyspnea, swallowing difficulties, muscular tremors, tympanism and recumbency in cattle. Animal’s death occurs rapidly after the onset of clinical signs and neither macroscopic nor microscopic lesions have been found. This paper describes the epidemiology, clinical findings and lesions of spontaneous poisoning by tifton 68 in cattle in the municipalities of Rio do Sul, Pouso Redondo, Taió and Rio do Campo in the state of Santa Catarina, Brazil, in 1996, 1997, 1998 and 2010 respectively. The presence of hydrocyanic acid was experimentally estimated in green and dried leaves through the picric acid paper test and poisoning treatment using a specific antidote solution of sodium thiosulfate and sodium nitrite. The poisoning was experimentally reproduced by the administration of tifton 68 green leaves to two cattle at a starting dose of 10.3 g/kg. The cyanide poisoning was confirmed by the immediate response to treatment with intravenous antidote. The samples of tifton 68 green leaves gave positive response in the picric acid paper test. The hay of tifton 68 showed no toxicity even at high doses (18 and 27 g/kg) and it is safe for cattle consumption.

E-mail: garciajuanagustin@hotmail.com A retrospective analysis (2000 to 2013) of cattle poisoning caused by toxic plants and other compounds was carried out in the Pampas region of Argentina by the Animal Health Group of INTA-EEA, Balcarce. During this period, 1263 reports of diseases of different etiologies (infectious, parasitic, toxic, metabolic and miscellaneous) were recorded in cattle, by collecting anamnestic, clinical and pathological information. A toxic etiology was diagnosed in 21.1% of these reports. Iatrogenic poisoning caused by ionophores was the most frequently recorded etiology. Consumption of toxic plants (Wedelia glauca, Solanum glaucophyllum, among others), mycotoxins (Claviceps purpurea, Claviceps paspali, Epichloë coenophiala, among others), and plants producing cyanide and nitrates/nitrites were also commonly diagnosed. The high frequency of toxic episodes and the difficulties in their diagnosis by practitioners in our livestock production systems emphasizes the importance of this report.

#64 - Genotoxicity assessment of fipronil (frontline plus®) in Canis familiaris, 37(3):257-260

Abstract in English:

ABSTRACT.- Ziliotto L., Luna S.P.L., Filho D.A.A., Resende L.O., Aun A.G. & Braz M.G. 2017. Genotoxicity assessment of fipronil (frontline plus®) in Canis familiaris. [Avaliação de genotoxicidade do fipronil (frontline plus®) in cães.] Pesquisa Veterinária Brasileira 37(3):257-260. Faculdade de Medicina de Botucatu, Universidade Estadual Paulista, Avenida. Prof. Mário Rubens Guimarães Montenegro s/n, Botucatu, SP 18618-687, Brazil. E-mail: mgbraz@hotmail.com Fipronil is a pesticide widely used for controlling fleas and ticks in domestic animals, and its short-term exposure can lead to serious effects on animals. However, the possible genotoxic effect of this compound has not been investigated in target animals. Based on the hypothesis that fipronil can induce genotoxicity, this study evaluated the effect of fipronil on DNA damage in peripheral blood cells. For that purpose, ten dogs of both sexes were used in the study. The product (6.7mg/kg) was applied on the dorsal neck region of each animal. Peripheral blood samples were collected immediately prior to application of the product, and at 3, 8 and 24 hours after the application. Samples were processed for comet assay. No statistically significant differences were found among the four time points. The current study suggests for the first time that a single exposure to this pesticide does not induce systemic genotoxic effect in dogs.

E-mail: fabio.mendonca@pq.cnpq.br The aim of this paper is to describe the first report of spontaneous poisoning by Prosopis juliflora in sheep. From flock of 500 sheep at risk, four adult male sheep were affected. One died spontaneously and three other were examined, euthanized and necropsied. Neurologic examination focused particularly on motor and sensory-cranial nerve function, complete blood counts, serum biochemistry and urinalysis were done. The evolution of the disease was chronic and to present signs of poisoning, sheep had to ingest a diet containing at least 80% of P. juliflora pods during 21 months. The biochemistry revealed a substantial increase in creatine phosphokinase levels. Clinical signs included drooling of saliva, dropped jaw, tongue protrusion and loss of food from the mouth. Gross and histological lesions were similar to those previously reported in cattle and goats. Sheep are more resistant to poisoning by P. juliflora considering that it took 21 months of pod consumption to show clinical signs. There is no specific treatment for P. juliflora poisoning in ruminants.

#66 - Senecio brasiliensis poisoning in calves: pattern and evolution of hepatic lesions

Abstract in English:

Poisoning by Senecio spp. plants are an important cause of death in cattle in the State of Rio Grande do Sul, Brazil. It is estimated that over 50% of deaths caused by toxic plants are due to the ingestion of Senecio spp. The epidemiological, clinical and anatomopathological aspects of a spontaneous outbreak of Senecio brasiliensis in calves are described here. Characterization and evolution of liver lesions during the outbreak are also described. Two hundred and six 4-month-old female calves were weaned and placed in a 25-hectare pasture heavily infested by S. brasiliensis at of varying growth stages. The calves remained in this pasture for three months and were later transferred to another area where six calves aged 7-8- month-old developed clinical signs including unthriftiness, diarrhea, and subcutaneous dependent edema in the submandibular region. All these six affected calves died within 15 days from the onset of clinical signs and one of those was necropsied. Grossly the first of three patterns of hepatic lesions described in this report was observed and it was characterized by an enlarged liver, with round borders and an orange hue to the capsular and cut surfaces. Histologically there was marked diffuse vacuolar hepatocellular degeneration, bilestasis, hepatocellular megalocytosis, and mild bile duct proliferation and periportal fibrosis. Two months after the onset of the outbreak, in another visit to the farm, it was observed that an additional 28 calves got sick and 14 died. Affected calves observed in this second on-site visit were 9-10 month-old and their main clinical sign was photosensitization characterized by varying degrees of photophobia, tearing from the eyes, marked drooling, mucopurulent ocular discharge, increased nasal secretion, and ventral diphtheric glossitis. Two of the fourteen calves that died were necropsied and displayed the second pattern of hepatic lesions observed in the outbreak. Liver had smooth and whitish capsular surfaces and a moderate decrease in size. There was moderate irregularity to the cut surface of hepatic parenchyma and pale areas intercalated with red ones. Histologically there was marked periportal fibrosis, marked bile duct proliferation, hepatocellular megalocytosis, focal areas of hepatocellular necrosis, and hemorrhage and veno-occlusive changes. In this second on-site visit, GGT serum activity was determined in 15 affected calves and 13 of them had increased values (26-175 U/L). Seven months after the onset of the outbreak, a third visit was made to the farm when was learned that and additional 27 calves have been affected, 23 of which died within a period of 40 days. At the necropsy of three of these calves the third pattern of hepatic lesions were observed. The liver was decreased in size and markedly firm. The capsular surface was smooth and whitish and the capsular surface was diffusely tan. Histological lesions similar to those observed in the second pattern were seen in in the liver of these three calves. A fourth on-site visit to the farm was made and a liver biopsy was performed in each of the 163 calves remaining in the herd and hepatic lesions characteristic of Senecio poisoning were observed in the liver biopsy of 103 of them. Affected calves were classified in groups according to the severity and morphologic aspects of the observed hepatic lesions. The diagnosis of Senecio poisoning in calves of this study was based on epidemiology, clinical and anatomopathological findings associated with the history of consumption of the plant, and on the chronological development of the lesions.

#67 - Experimental poisoning by Enterolobium contortisiliquum in sheep

Abstract in English:

Ingestion of Enterolobium contortisiliquum pods causes digestive disturbances, secondary hepatogenous photosensitization and abortions in ruminants. Pods were administered to sheep via a ruminal cannula to characterize acute poisoning. In Experiment 1, a single dose of 12g/kg of body weight (BW) was administered to three sheep in one experiment. One sheep died, and the other two recovered after presenting clinical signs. In Experiment 2, 10g/kg BW were administered daily to 15 sheep until the onset of clinical signs or for three consecutive days. Fourteen sheep showed mild to severe signs after the ingestion of 1-3 doses. Two sheep died, and the others recovered. Clinical signs in both experiments were diarrhea, anorexia, rumen atony, apathy, dehydration and tachypnea. The main macroscopic findings were an orange, frothy ruminal content witch contained pods fragments. The intestinal content was liquid. Detachment of the mucosa from the submucosa and ballooning degeneration of mucosal cells were observed histologically in the forestomachs. Evaluation of ruminal contents revealed acute lactic ruminal acidosis (ALRA). Bromatological analysis of E. contortisiliquum pods revealed 537.8g/kg DM (dry matter) of non-fibrous carbohydrates, which is sufficient to cause ALRA. Only one sheep in Experiment 2 had liver failure, characterized by jaundice, elevated serum activity of liver enzymes and histological lesions in liver biopsies. It is concluded that the administration of E. contortisiliquum pods in forage-fed sheep at doses of 10g/kg BW or higher may cause ALRA. The induction of liver failure in one sheep suggests that liver damage may occur in those sheep that do not develop acidosis.

Abstract in Portuguese:

A avaliação do conteúdo ruminal revelou acidose ruminal láctica aguda (ARLA).

E-mail: a2ag@cav.udesc.br This paper describes the toxic potential of the green leaves of Crotalaria pallida and the clinical signs in experimentally poisoned sheep. Green leaves of C. pallida were collected in São João do Sul, Santa Catarina, and administered orally to six adult sheep, as single dose of 40, 20, 10, 5, 2.5 and 2.5g/kg, and for two sheep at doses of 0.63 and 1.25g/kg during 30 days. The poisoning was reproduced at lethal doses of 2.5g/kg. One of the sheep that received a single dose of 2.5g/kg and another that received daily doses did become ill. All sheep wich developed clinical signs died between 13 and 22 hours after administration of the plant. Slight signs started in about 11 hours after ingestion of the plant, and clinical signs included loose stool, restlessness and increased heart and respiratory rates. A few minutes preceding death, pronounced abdominal breathing and severe dyspnea was observed. The main gross lesions were restricted to the thoracic cavity, as severe pulmonary edema and hydrothorax. Lung microscopy showed marked edema in the diffuse pleural surface, the interlobular spaces and around the peribronchial arteries. Multifocal edema was observed within alveoli and around the bronchi and bronchioles, and mild multifocal infiltration of neutrophils. The clinical and pathological alerations of the experimental poisoning was similar to that observed in the spontaneous poisoning, characterized by marked pulmonary edema and death shortly after the onset of clinical signs.

[Lectin histochemistry evaluation of liver and mesenteric lymph node of buffaloes kept on Brachiaria spp. pastures.] E-mail: ileanamiranda@gmail.com Animals grazing Brachiaria spp. commonly present foamy macrophages isolated or grouped in the liver, and crystals within biliary ducts. The pathogenesis of formation and the nature of the material stored in these cells however are not completely known. Through lectin histochemistry evaluation, steroidal saponins (secondary glycosylated metabolites) have been identified in the crystals and within the cytoplasm of the foam cells, which are probably liable for damaging the liver, leading to accumulation of phylloerythrin. This study aims to standardize and characterize the use of lectin histochemistry to detect glycosylated metabolites in tissues of buffaloes kept on different Brachiaria spp. pastures in Brazil. Fragments of liver and mesenteric lymph node from 40 buffaloes were analyzed: 10 buffaloes that were kept in predominant pasture of B. decumbens for 12 months; 10 buffaloes that were kept in pasture with a predominance of B. brizantha for 18 months; 10 buffaloes that were kept on pasture of B. brizantha for about four years; and as a negative control, 10 buffaloes that were maintained on native pasture without Brachiaria spp. since birth. Fourteen lectins were tested (Con-A, SBA, WGA, DBA, UEA, RCA, PNA, GSL-I, PSA, LCA, PHA-E, PHA-L, SJA and SWGA), in a total of 1120 evaluated samples. Previous studies demonstrated that PNA showed great binding reactivity for foamy macrophages in cattle and sheep. In the present study, SWGA showed high specificity and marked binding reactivity for foamy macrophages; WGA, GSL, PHA-E and PHA-L showed moderate to marked reactivity, but low specificity for foamy macrophages. The other lectins had not relevant reactivity or specificity. Moreover there was no relevant reactivity difference between the collected samplesd from buffaloes that grazed B. decumbens for 12 months and Brachiaria brizantha for 18 months. However the decreased presence of foamy macrophages and its lectin histochemical binding in animals that fed on B. brizantha for a longer time, indicates that the buffaloes can pass through an adaptation process according to the plant intake time. Lectin histochemistry analysis can be used to characterize the material stored in foamy macrophages present in liver and mesenteric lymph node of buffaloes that graze on Brachiaria spp. pastures and helps to clarify the pathogenesis of these cells.

RESUMO.- Miranda I.C., Riet-Correa G., Oliveira Júnior C.A., Riet-Correa B., Peixoto P.V., Costa S.Z.R., d’Avila M.S., Cid G.C. & França T.N. 2016. [Lectin histochemistry evaluation of liver and mesenteric lymph node of buffaloes kept on Brachiaria spp. pastures.]

Abstract in English:

ABTSRACT.- Olurin K.B., Mbaka G.O. & Agbato O.A. 2016. Histopathological effect of sub-lethal concentration of aluminum phosphide (phostoxin) on Clarias gariepinus juveniles. Pesquisa Veterinária Brasileira 36(7):574-580. Department of Anatomy, Lagos State University College of Medicine, Ikeja, Lagos, Nigeria. E-mail: mbaaka2gm@gmail.com The study evaluated the effect of sub-lethal concentration of phostoxin on Clarias gariepinus juveniles. C. gariepinus juveniles belonging to the same cohort (40.1±1.2g; 18.1±1.1cm) from a commercial fish farm were randomly placed ten in each of 15 plastic tanks containing 15 liters of water. They were exposed for 96 hrs to three sub-lethal concentrations (treatments) of phostoxin (0.125, 0.250, 0.5mg L-1) and a phostoxin free control. At the end of 96 hrs exposure, they were dissected and the tissues need for histopathology removed and fixed in Bouin’s fluid. The gill filament exhibited fusion at the secondary lamella that was progressive with concentration. At the highest concentration of exposure, the secondary lamellae showed marked pyknotic and necrotic changes characterized by epithelia detachment. The hepatic tissue showed mild inflammatory changes at lower concentrations while at the highest concentration of exposure there was marked inflammation with observed hydropic degeneration. In the kidney, an inflammatory change was only observed in the interstices at the highest dose of exposure with the convoluted tubules showing partial shrinkage. Phostoxin showed to have significantly caused alterations in cyto-architecture of the gills and to a considerable extent liver and kidney of C. gariepinus.
