#1 - Evaluation of phosphorus levels in bones and serum of buffaloes (Bubalus bubalis) before and after supplementation with a selective mineral mixture

Abstract in English:

This study aimed to evaluate phosphorus (P) concentration in serum and bone, the percentage of ash and the specific bone density of buffaloes on Ilha de Marajó before and after mineral supplementation. For this study, 14 crossbred buffaloes of Murrah and Mediterranean descent aged between 18 and 36 months were used. The average values of P before supplementation in serum and bone, the percentage of bone ash and the specific bone density were 5.68mg/dL±1.18, 16.53%±0.53, 59.95%±1.96 and 1.52g/cm3±0.32, respectively, which demonstrated P deficiency in animals raised on Ilha de Marajó. After supplementation with P for a period of seven months, the values were 6.61mg/dL±0.87, 16.90%±0.56 and 60.30%±0.95 and 1.71g/cm3±0.21, respectively. These results showed a significant increase in P concentration in blood serum, specific bone density and percentage of P in ash (P<0.05), but there was no significant increase in the percentage of ash. The average increase in P in the serum and ash did not reach normal levels in all animals; however, 28.6% of the animals had normal values of P in serum and 50% in the ash, and 64.3% had normal specific bone density values. The nonre-establishment, in some of the animals, of the variables of P serum and bone after supplementation for seven months may have occurred as a result of the low intake of the mineral mixture and by the low concentration of P in the Brachiaria brizantha cv. Marandu used for feeding animals during the experiment.

Objetivou-se avaliar as concentrações de fósforo (P) no soro e no osso, o percentual de cinzas e a densidade óssea específica em búfalas da Ilha de Marajó antes e após suplementação mineral seletiva. Foram utilizadas 14 búfalas mestiças de Murrah com Mediterrânea, com idades entre 18 e 36 meses. Os valores médios de P, antes da suplementação, no soro, no osso, o percentual de cinzas e a densidade óssea específica foram de 5,68mg/dL±1,18, 16,53%±0,53, 59,95%±1,96 e 1,52g/cm3±0,32, respectivamente, o que demonstra deficiência de P nos animais criados na Ilha de Marajó. Após a suplementação com P por um período de sete meses os valores foram 6,61mg/dl±0,87, 16,90%±0,56 e 60,30%±0,95 e 1,71g/cm3±0,21 respectivamente. Esses resultados caracterizam um aumento significativo nas concentrações de P no soro sanguíneo, na densidade óssea específica e no percentual de P nas cinzas (P<0,05), porém não houve um aumento significativo no percentual de cinzas. O aumento médio nos valores de P no soro e nas cinzas não alcançou patamares de normalidade em todas as búfalas, entretanto 28,6% delas tinham valores normais de P no soro e 50% nas cinzas, 64,3% tinham valores normais da densidade óssea específica. O não restabelecimento, em parte dos animais, das variáveis ósseas e sanguíneas após suplementação com P durante sete meses pode ter ocorrido em virtude da baixa ingestão da mistura mineral e da baixa concentração de P em Brachiaria brizantha cv. Marandu utilizada para alimentação dos animais durante o experimento.


#2 - Enterotoxin-encoding genes in Staphylococcus aureus from buffalo milk

Abstract in English:

This paper investigated the occurrence of Staphylococcus aureus and the detection of enterotoxin-encoding genes of these strains in milk collected from 30 Murrah buffaloes used to produce dairy products in Brazil. A total of 68 strains of Staphylococcus aureus were found as identified by conventional laboratory tests, and thus screened for sea, seb, sec, sed, see, seg, seh and sei enterotoxin-encoding genes by polymerase chain reaction (PCR). Twelve strains containing enterotoxin-amplified genes were found, with higher expression for the sei and seh genes. These results can be attributed to animal health and inadequate cleaning of the equipment, indicating the need for better quality control in animal production and health lines. The results of this study with the presence of pathogens and their enterotoxigenic potential indicate a source of food poisoning, as well as being a pioneering study in the detection of new enterotoxins for buffalo milk.

Este estudo investigou a ocorrência de isolados de Staphylococcus aureus e a detecção de genes que codificam a enterotoxigenicidade dessas cepas em leite de búfala utilizado na produção de laticínios no Brasil. As amostras foram coletados em 30 búfalos da raça Murrah, identificado por testes laboratoriais convencionais, foram identificados um total de 68 cepas de S. aureus e rastreados para os genes que codificam a enterotoxina sea, seb, sec, sed, see, seg, seh and sei por reação em cadeia da polimerase (PCR). Doze cepas contendo genes da enterotoxina foram amplificadas, com maior expressão para os genes sei e seh. Esses resultados podem ser atribuídos à saúde animal e à higiene inadequada do equipamento, indicando a necessidade de melhor controle de qualidade nas linhas de produção e saúde animal. Os resultados desta pesquisa, com a presença de patógenos e seu potencial enterotoxigênico, indicam uma fonte de intoxicação alimentar, além de ser uma pesquisa pioneira na detecção de novas enterotoxinas para o leite de búfala.

Nineteen buffalo cows in various stages of pregnancy, serologically positive for brucellosis and subjected to culling were used. Fetal age was determined by ultrasound examination and the size of fetuses was measured at necropsy. Fragments of lymph nodes were collected for histopathology and qPCR. The detection of B. abortus DNA in the lymph nodes was checked from the fourth month of pregnancy in seven buffaloes and in a post-calving buffalo. Acute to chronic lymphadenitis was histologically diagnosed. B. abortus DNA was detected in all evaluated groups of lymph nodes; the mammary lymph nodes were the most affected.

Foram utilizadas 19 búfalas em diversos estágios de gestação, sorologicamente positivas para brucelose, submetidas ao abate sanitário, das quais se coletou fragmentos de diversos linfonodos. A idade fetal foi determinada através de exames ultrassonográficos associados à mensuração dos fetos durante a necropsia. Amostras foram coletadas e submetidas à qPCR e histopatologia. A detecção de DNA de B. abortus nos linfonodos das búfalas avaliadas foi verificada a partir do quarto mês de gestação em sete búfalas e em uma búfala pós-parição. Os achados histológicos foram linfadenite aguda a crônica. A presença de DNA de B. abortus foi detectada em todos os grupos de linfonodos avaliados, sendo que os linfonodos mais acometidos foram os mamários.

E-mail: lucaspizauro@yahoo.com.br To evaluate the biochemical profile and protein concentration of whey from milk samples of healthy Murrah primiparous and pluriparous buffaloes, 30 female buffaloes were analyzed during a complete lactation. The animals were divided into three groups: G1 = 10 primiparous buffaloes, G2 = 10 pluriparous buffaloes with 2-3 lactations and G3 = 10 pluriparous buffaloes with >3 lactations. The lactation period was divided into: early stage (I: 1-3 months of lactation), intermediate stage (T: 4-6 months of lactation) and final stage (F: 7-9 months of lactation). Before milk sampling, physical examination of the mammary gland, strip cup test and California Mastitis Test (CMT) were performed. After mammary quarters asepsis, 20mL of milk were collected monthly from each mammary quarter, during a complete lactation, in sterilized plastic bottles without preservative, in order to perform microbiological isolation, biochemical profile and protein electrophoresis in sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), and 30mL of milk from each mammary quarter were collect, in sterilized plastic bottles containing preservative bronopol to perform the somatic cell count (SCC). A total of 1,042 milk samples were collected from the experimental groups during lactation, of which 923 samples showed negative reaction to CMT and negative microbiological isolation and were selected to biochemical profile analysis and protein electrophoresis in SDS-PAGE. There were influence of parity order and stage of lactation in biochemical profile and protein concentration of healthy Murrah buffaloes’ whey. Primiparous buffaloes (G1) showed higher gamma-glutamyltransferase (GGT: 2,346 U/L), alkaline phosphatase (ALP: 181 U/L), phosphorus (P; 56.6mg/dL), potassium (K; 32.0mg/dL) and &#945;-lactalbumin (458mg/dL). Buffaloes with 2-3 lactations (G2) showed higher SCC (70,700 cells/mL) and higher concentrations of total protein (1.55g/dL), albumin (100mg/dL), magnesium (Mg; 8.80mg/dL), chlorides (Cl; 176mg/dL), iron (Fe; 10.7&#956;g/dL), sodium (Na; 178mMol/L) and lactoferrin (59.5mg/dL). Bufalloes with >3 lactations (G3) showed higher concentrations of total calcium (Ca; 41.8mg/dL), ionized calcium (iCa; 2.92mMol/L), immunoglobulin A (IgA; 1.32mg/dL), serum albumin (99.1mg/dL), immunoglobulin G (IgG; 49.7mg/dL) and &#946;-lactoglobulin (1,068mg/dL). During lactation it was observed increase in SCC, GGT, ALP, total protein, albumin, P, Mg, Cl, Na, lactoferrin, serum albumin, IgG and &#945;-lactalbumin, as well as decrease in concentrations of Ca, Fe, iCa, K, IgA and &#946;-lactoglobulin in buffaloes’ whey. The results may be used as reference for buffaloes and to support diagnosis and prognosis of diseases common to lactation periods.

Os animais foram distribuídos em três grupos: G1 - 10 búfalas primíparas, G2 - 10 búfalas pluríparas com duas a três lactações e G3 - 10 búfalas pluríparas com mais de três lactações. O período de lactação foi dividido em: fase inicial (I: primeiro ao terceiro mês de lactação), fase intermediária (T: quarto ao sexto mês de lactação) e fase final (F: sétimo ao nono mês de lactação). Antes da colheita das amostras de leite foram realizados o exame físico da glândula mamária, o teste da caneca de fundo escuro e o California Mastitis Test (CMT). Após a assepsia dos quartos mamários, foram colhidas mensalmente, durante uma lactação completa, amostras de 20mL de leite de cada quarto mamário, em frascos plásticos esterilizados e sem conservante, para a realização do isolamento microbiológico, determinação do perfil bioquímico e fracionamento proteico por meio de eletroforese em gel de poliacrilamida contendo dodecil sulfato de sódio (SDS-PAGE), e amostras de 30mL de leite de cada quarto mamário, em frascos plásticos esterilizados contendo conservante bronopol, para contagem de células somáticas (CCS). Das 1.042 amostras de leite colhidas dos três grupos experimentais durante a lactação, 923 amostras de leite apresentaram reação negativa ao CMT e isolamento microbiológico negativo e foram selecionadas para as análises do perfil bioquímico e fracionamento proteico em SDS-PAGE. Notou-se influência da ordem de parto e da fase da lactação no perfil bioquímico e no proteinograma do soro lácteo de búfalas da raça Murrah sadias. As búfalas primíparas (G1) apresentaram maior atividade das enzimas gamaglutamiltransferase (GGT: 2.346U/L) e fosfatase alcalina (ALP: 181U/L) e maiores concentrações de fósforo (P: 56,6mg/dL), potássio (K: 32,0mg/dL) e &#945;-lactoalbumina (458mg/dL). As fêmeas com duas a três lactações (G2) apresentaram maior CCS (70.700 células/mL) e maiores concentrações de proteína total (1,55g/dL), albumina (100mg/dL), magnésio (Mg: 8,80mg/dL), cloretos (Cl: 176mg/dL), ferro (Fe: 10,7µg/dL), sódio (Na: 178mMol/L) e lactoferrina (59,5mg/dL). As fêmeas com mais de três lactações (G3) apresentaram maiores concentrações de cálcio total (Ca: 41,8mg/dL), cálcio ionizado (Cai: 2,92mMol/L), imunoglobulina A (IgA: 1,32mg/dL), albumina sérica (99,1mg/dL), imunoglobulina G (IgG: 49,7mg/dL) e b-lactoglobulina (1.068mg/dL). Durante a lactação foi observado aumento da CCS, aumento das atividades das enzimas GGT e ALP, aumento das concentrações de proteína total, albumina, P, Mg, Cl, Na, lactoferrina, albumina sérica, IgG, &#945;-lactoalbumina e redução das concentrações de Ca, Fe, Cai, K, IgA e b-lactoglobulina no soro lácteo das búfalas. Os resultados obtidos podem ser utilizados como referências para a espécie bubalina e auxiliar no diagnóstico e no prognóstico de doenças de ocorrência comum na fase de lactação.


#5 - Comparison of indirect ELISA and indirect immunofluorescence in detection of antibodies to Neospora caninum in buffaloes (Bubalus bubalis), 33(4):431-434

Pesquisa Veterinária Brasileira 33(4):431-434. Laboratório de Investigação e Diagnóstico de Enfermidades Animais, Universidade Federal do Pará, Belém, PA, Brazil. E-mail: spatroca@yahoo.com.br To compare two serologic tests for detection of antibodies against Neospora caninum in sera from buffaloes, samples were collected from 288 buffaloes of 2 to 10 years of age. To identify the presence of IgG, anti-N. caninum was used for the indirect immunofluorescence assay (IFAT), with title 200 as the cutoff, and for the immunoenzymatic test (indirect ELISA), considering as positive samples with ratio S/P &#8805;0.5. There were 153 (53.12%) animals positive for N. caninum by IFAT, whilst 50 (17.36%) animals were reactive in ELISA. The presence of antibodies anti-N. caninum demonstrates that the parasite is circulating among buffaloes raised in Pará State, Brazil. ELISA and IFAT tests could be used to diagnose immunoglobulins against this agent. However there was a weak correlation (Kappa = 0.36) between both tests, considering the IFAT as the gold standard.

Para identificar a presença de imunoglobulina G anti-N. caninum utilizou-se à reação de imunofluorescência indireta (RIFI), tendo o título 200 como ponto de corte, e o Ensaio Imunoenzimático indireto (ELISA-indireto), considerando-se positiva as amostras que obtiveram razão S/P&#8805;0,5. Observaram-se 153 (53,12%) animais soropositivos para N. caninum, através da RIFI, enquanto que 50 (17,36%) animais foram reagentes no ELISA. A ocorrência de anticorpos anti-N. caninum demonstram que o parasito esta circulando entre búfalas criadas no estado do Pará, sendo que ambos os teste de RIFI e ELISA podem ser utilizados para diagnosticar imunoglobulinas contra este agente. No entanto observou-se uma fraca correlação (Kappa=0,36) entre ambos os testes, considerando a RIFI como padrão ouro.

E-mail: rinaldo.mota@hotmail.com The aim of this study was to evaluate the somatic cell profile in the subclinical mastitis in dairy buffaloes in the Brazilian Northeastern. One thousand eight hundred ninety six milk samples from 474 buffaloes proceeding from four dairy farms located in the states of Pernambuco, Alagoas, Bahia and Ceará were analyzed. The lactic secretion was submitted to the California Mastitis Test (CMT) and the positive samples, from two crosses on were collected for the somatic cell count (SCC) and microbiologic exam. It was observed that the positive samples at the microbiologic exam presented SCC between 280,000 and 401,000 cells/mL with median of 328,000 cells/mL. It was concluded that SCC values above 280,000 cells/mL is an indication of mammary gland infection, however, the microbiologic exam of the milk is the best method for the diagnostic of subclinical mastitis in buffaloes. The observation of Staphylococcus coagulase-negative exerting influence in the SCC elevation must be subject of further studies in order to elucidate the pathogenicity of this group of microorganisms in mastitis of buffaloes.

RESUMO.- Objetivou-se com esse estudo avaliar o perfil de células somáticas na mastite subclínica em búfalas leiteiras no Nordeste do Brasil. Foram analisadas 1896 amostras de leite de 474 búfalas procedentes de quatro propriedades de exploração leiteira nos Estados de Pernambuco, Alagoas, Bahia e Ceará. A secreção láctea de cada teto foi submetida ao Califórnia Mastitis Test (CMT) e as amostras positivas, a partir de duas cruzes foram coletadas para realização da contagem de células somáticas (CCS) e exame microbiológico. Observou-se que as amostras positivas no exame microbiológico apresentaram CCS entre 280.000 a 401.000 cel/mL com mediana de 328.000 cel/mL. Conclui-se que valores de CCS acima de 280.000 cel/mL é um indicativo de infecção da glândula mamária, contudo o exame microbiológico do leite é o melhor método para diagnóstico da mastite subclínica em búfalas. A observação dos Staphylococcus coagulase negativa exercendo influência na elevação da CCS deve ser objeto de outros estudos para elucidar a patogenicidade desse grupo de microrganismos na mastite de búfalas.

E-mail: spatroca@yahoo.com.br To investigate the occurrence of antibodies IgG anti-Neospora caninum and Toxoplasma gondii, blood samples were collected from 374 adult female water buffaloes originated from 14 production units located in 13 counties of the state of Pará. The sera were subjected to indirect fluorescent antibody (IFA), using titers of 200 and 64 as IFA cut off points for N. caninum and T. gondii, respectively. Hundred and fifty three (40.9%) buffaloes were sero-positive for N. caninum, with 100% of the properties being sources of infection. Four buffaloes (1.1%) in four farms were seropositive for T. gondii, and one buffalo cow (0.27%) was seropositive for the two parasites. The presence of antibodies indicates that these protozoan parasites are circulating among buffaloes from the properties studied, representing a source of infection to other animals, as well as a possible cause of reproductive disorders in this species.

E-mail: spatroca@yahoo.com.br Para determinar a ocorrência de anticorpos IgG anti-Neospora caninum e Toxoplasma gondii em 14 unidades produtivas de búfalos, situadas em 13 municípios no estado do Pará, foram coletadas amostras de soro sanguíneo de 374 fêmeas adultas. Os soros foram submetidos à reação de imunofluorescência indireta (RIFI), utilizando-se os títulos 200 e 64, respectivamente como ponto de corte para N. caninum e T. gondii. Obteve-se 153 (40,9%) de animais soropositivos para N. caninum com 100% das propriedades com focos da infecção, enquanto que quatro búfalas (1,1%) foram soropositivas em quatro fazendas para T. gondii e uma búfala (0,27%) foi soropositiva para ambos parasitos. A presença de anticorpos é um indicativo da circulação desses protozoários em búfalos das propriedades estudadas, representando uma fonte de infecção para outros animais, assim como o possível envolvimento em distúrbios reprodutivos nessa espécie.

The corion is a simple layer of circle cells, with spherical nucleus, named trofoblastics; there is another cellular type, named trophoblastic giant cells, with two or more nucleus. Both of them have many cytoplasmic and reticule endoplasmic vesicles on transmission electron microscopy. The allantoic has many vessels filled with eritrocytes, elongated cells which form a stratified simple epithelium. The amnion is transparent or sometimes whitish, consisting of a stratified simple epithelium. The main difference between the allantoic and amnion is that the latter is avascular. The yolk sac is an opaque membrane that disappears during gestation, and is the only membrane that is not in contact with the others; it presents three different types of cells which form three distinct layers (endoderm, mesothelium and mesenquimal).

The means were: right cranial papilla (RCP) 20.95g; right caudal papilla (RCaP) 21.49g; left cranial papilla (LCP) 23.52g; and left caudal papilla (LCaP) 21.35g. Papillary duct average length, numbers of folds and papillary cisterns length were, respectively: 0.67/5,5 e 2.17cm for RCP; 0.62/6.0 e 2.26cm for RCaP; and for the left antimere: RCP: 0.71/6.0 e 2.26cm and for RCaP: 0.74/5.7 e 2.57cm. Pearson’s analysis revealed negative correlation between body weight and mammary glandular weight. Sampling of proximal, middle and distal area was taken and fixed into a 10% formaldehyde solution, followed by mesoscopical investigation through 10, 20 and 40µm thickness cuts stained by Azan, F3BA Picrosirus and Weigert-Van Gieson techniques. Histological and morphometric analyses were applied on 5µm thick samples and showed that the muscular layer was disposed into two arrangements, internal circular and external longitudinal with a bi-stratified cuboidal epithelium. A well developed muscular layer were related to the middle portion revealing thickness of 22.90mm with a great amount of connective tissue (7.10mm).

E-mail: diomedes@ufpa.br Eight lactating buffaloes of the Murrah breed were used from 60 days before up to 60 days after calving, in order to evaluate alterations in the concentration of blood glucose, total plasmatic protein, haematocrit and the presence of ketonic bodies in the urine. The 5 to 10-year old buffaloes were clinically healthy and had had at least two lactations. During the experimental period the animais were maintained on Brachiaria brizantha pasture and · supplemented with a mineral mixture and water. The media concentrations of blood glucose and the media values of the haematocrit diminished significantly after calving (p<0,05). The concentrations of total plasmatic protein did not show significant variation during the pre and post-parturient period. The ketonic bodies in the pre-parturient period were only detected in the urine of one buffaloe, but after the 32nd day of lactation ketonic bodies were detected in ali animais. There was a direct relationship between the color of the urine positive by the Rothera test and blood glucose concentrations. lt can be concluded that at the beginning of lactation the buffaloes had an energetic deficit, characterized by a decline of blood glucose concentrations and the presence of ketonic bodies in the urine, and that lactation caused a progressive decline of the hematocrit, but that the concentration of total plasmatic protein did not vary during the pre and post-parturient period.

As concentrações médias de glicose sangüínea e o valor médio do hematócrito diminuíram significativamente no pós-parto (p<0,05). As concentrações de proteína plasmática total não sofreram variações significativas do pré para o pós-parto. No período préparto os corpos cetônicos só foram detectados na urina de uma búfala; entretanto, a partir do 32º dia de lactação foram detectados em todos os animais. Houve uma relação direta entre a coloração da urina positiva para o teste de Rothera e as concentrações de glicose sangüínea. Pode-se concluir que na fase inicial da lactação as búfalas utilizadas sofreram um déficit energético, caracterizado pela diminuição nas concentrações sangüíneas de glicose e presença de corpos cetônicos na urina, e que a lactação causou um declínio progressivo no hematócrito, enquanto que as concentrações da proteína plasmática total não sofreram variações do pré para o pós-parto.
