#1 - Dry-eye syndrome in Shih Tzu dog: Ocular surface clinical and morphological aspects in the different stages of keratoconjunctivitis sicca

Abstract in English:

This study analyzed clinical and cyto-histomorphological parameters of the ocular surface of Shih Tzu dogs, non-carriers and carriers of quantitative keratoconjunctivitis sicca (KCS) at different stages. Thirty-five eyes from 23 male and female Shih Tzu dogs between two and eight years were evaluated in four groups: control group (CG – without KCS), mild KCS group (KCS1), moderate KCS group (KCS2), and severe KCS group (KCS3). Most clinical variables among KCS carrier groups worsened at the more advanced stages of the disease, with a negative correlation between the Schirmer tear test (STT-1) and tear film break-up time (TBUT). Squamous metaplasia, lymphoplasmacytic inflammatory infiltrate, and decrease in conjunctival goblet cells on histopathological examination comprised disease severity parameters. Quantitative KCS non-carried Shih Tzu dogs have qualitative and tear film distribution changes. The cytomorphological exam is limited to evaluating the inflammatory infiltrate and quantifying conjunctival goblet cells. However, intermediate epithelial cells were higher in healthy eyes compared to eyes with KCS in Shih Tzu dogs. Also, moderate and severe KCS carrier Shih Tzu dogs have qualitative dysfunction of the tear film. KCS in Shih Tzu dogs is common and chronic and may be responsible for the loss of vision of these animals. Early identification of the disease and routine evaluation can improve these dogs’ quality of life and ocular health.

#2 - Cytometric blood phenotyping in free-living white-eyed parakeet (Psittacara leucophthalmus)

Abstract in English:

This study aimed at performing cytometric phenotyping of the blood samples from free-living, young white-eyed parakeets (Psittacara leucophthalmus), stained with 3,3-dihexyloxacarbocyanine [DiOC6(3)]. DiOC6(3)-stained whole blood samples from 19 free-living, young white-eyed parakeets were analyzed by flow cytometry and cell types were distinguished by their typical fluorescence in blue laser channel (FL-1) and SSC (side scatter). It was possible to differentiate erythrocytes (58.3±13.6) from leukocytes (32.4±13.1) and some of the leucocyte subpopulations: lymphocytes/thrombocytes (29.7±7.7), monocytes (30.6±8.5), and granulocytes (5.9-26). However, lymphocytes and thrombocytes could not be sorted in the plots. Our study determined that the predominant population in white-eyed parakeet (P. leucophthalmus) was lymphocytes, thrombocytes, and monocytes in the leucocytes gates in comparison to the granulocyte population. The cytometry method and use of DiOC6(3) stain was available for parakeets blood samples and can be studied and applied to other species of parrots.

#3 - Macroscopic morphological aspects, ecometry and rebound tonometry of the eye bulb in sloth (Bradypus variegatus)

Abstract in English:

The field related to the visual system of wild animals is deeply scarce. Settling anatomical and physiological parameters for these animals is still a descriptive vision for Bradypus variegatus (Schinz, 1825). Thus, our research aimed to determine patterns of normal eye for this species. For this purpose, eight eye bulbs were dissected from the carcasses obtained by natural death, and then performed an overview of ocular anatomical. Rebound tonometry (RBT) and ocular B-mode ultrasonography were also applied for eight eyes in four animals from “Parque Estadual Dois Irmãos”, situated in the city of Recife, state of Pernambuco (PE), to estimate the intraocular pressure and ocular ecobiometry. The ocular morphology of sloth is similar as described for other species, however, with some peculiarities. They present a third eyelid emerging in the nasal region of the inferior conjunctival sac and retina and also contain little differentiated blood vessels. Medium the intraocular pressure (IOP) was 4.25mmHg with no difference for both eyes. Ultrasonography of ocular anatomy is also similar regarding other species. Ecobiometric patterns were evaluated to determine the anterior chamber depth, lens width, vitreous chamber depth, and axial length (AL) of ocular globe and the averaged as shown 0.63±1.11mm, 3.73±0.24mm, 6.15±0.41mm, 3.70±0.27mm, and 8.48±0.22mm, respectively. There was no difference between the right and left eyes. The RBT and ocular B-mode ultrasonography are fast exams and easy for animal testing. This study contributed to the characterization of ocular anatomy as well as settling medium values of IOP and intraocular measures; however, further research on physiology and histology is necessary to better understand the visual function of the species.

#4 - Concentrations of matrix metalloproteinase-2 and interleukin-1 β in the aqueous humor of dogs with normal and cataractous eyes

Abstract in English:

We aimed to determine the concentration of MMP-2 and IL-1β in the aqueous humor of dogs with healthy eyes (n=8) and in those with mature (n=8) and hyper mature cataracts (n=8). Correlations between cytokines, cytokines, and intraocular pressure (IOP), as well as cytokines with ages of patients of each group, were also assessed. In patients with cataract, aqueous humor was collected at the end of the electroretinographic procedure. In healthy dogs, aqueous humor was collected before elective surgeries. Cytokine levels were determined using ELISA. IOP was assessed by applanation tonometry. IOP of patients with mature and hyper mature cataracts were lower than the ones measured in normal eyes (P=0.158). MMP-2 aqueous humor concentration was higher in patients with hyper mature cataracts, in comparisons with healthy patients (P=0.04). Average IL-1 β aqueous concentration was higher in patients with cataracts (P<0.0001). Significant higher values of IL-1 β were observed in patients with hyper mature, than in the ones with mature cataracts (P=0.0085). Correlations between MMP-2 and IL-1 β (r=-0.38, P=0.06), MMP-2 and IOP (r=-0.149, P=0.484), and IL-1 β and the ages of patients were not observed (P>0.05). IL-1 β and IOP correlated negatively (r=-0.42, P=0.04). MMP-2 and the ages of patients correlated only in dogs with mature cataracts (r=0.772, P=0.02). It can be concluded that the increment in the aqueous humor concentration of IL-1 β found in dogs with mature and hyper mature cataracts, in addition to the negative relationship of this cytokine with IOP, suggests that IL-1 β is involved in the pathogenesis of LIU. Despite dogs with hypermature cataracts presented significant higher concentrations of MMP-2 in the aqueous humor, such cytokine did not correlate with IL-1 β and IOP. In our study, a relationship between aqueous humor cytokines and the ages of patients was only confirmed between MMP-2 and the ages of dogs with mature cataracts.

#5 - Antimicrobial susceptibility and minimal inhibitory concentration of bacteria isolated from the eyes of dogs with keratoconjunctivitis sicca

Abstract in English:

The objective of this study was to evaluate the antimicrobial susceptibility profile of bacteria isolated from the eyes of dogs with keratoconjunctivitis sicca (KCS). We evaluated 65 dogs diagnosed with KCS and 30 healthy dogs (Control Group). Conjunctival swab samples were collected after KCS was diagnosed. Microbiological examinations were performed, including aerobic culture, antimicrobial susceptibility testing and minimum inhibitory concentration (MIC) determination for chloramphenicol, tobramycin, ofloxacin and moxifloxacin. MICs of the fifteen most resistant strains of Staphylococcus pseudintermedius (Staphylococcus intermedius Group, SIG) and the fifteen most resistant strains of gram-negative bacteria were determined. By percentage, the microorganisms exhibited the highest susceptibility to polymyxin B, tobramycin and chloramphenicol and the lowest to tetracycline. Three multi drug-resistant strains of SIG were detected: one displayed isolated susceptibility to cefazolin, another to vancomycin, and another to polymyxin B and amikacin. The species of bacteria isolated from the eyes of dogs with KCS presented variable susceptibility to the antibiotics tested. We found evidence of the emergence of quinolone-resistant strains of SIG and further evidence of increased ocular prevalence. These findings reinforce the need to identify the bacteria involved and their antimicrobial susceptibility profile, as secondary infections can serve as exacerbating and perpetuating factors in KCS.

#6 - The use of targeted selective treatments on controlling gastrointestinal nematodes in different sheep categories under grazing system

Abstract in English:

Infections with gastrointestinal nematodes can impair both the animal health and performance. Thereby, the aim of this study was to evaluate the use of targeted selective treatments as the FAMACHA© system and faecal egg count on controlling gastrointestinal nematodes in different sheep categories under grazing system. Thirty Santa Inês sheep, six in each category (six weaning, six weaned lambs, six non-pregnant ewes, six pregnant ewes and six lactating ewes were used. Sheep were weighed, and blood and faecal samples were collected on day 0, 15, 30, 45, 60 and 75 for determination of faecal egg count (FEC), packed cell volume (PCV), total plasma protein (PPT) and the mucosa colour by FAMACHA© system in the morning (1) and afternoon (2). The faecal egg count was higher (p=0.0039) in weaning and weaned lambs compared to the non-pregnant, pregnant and lactating ewes. Also, FEC differed between ewes with higher counts in lactating ewes. The observation of eye mucosae showed a frequency of 100% of FAMACHA© scores ranged of 2.45 to 2.91 in the morning, differing (p=0.0494) among the categories, where weaned lambs showed better score. The animal categories and period interaction had a significant (p=0.0148) effect on FAMACHA© score 1 with better score in weaned lambs compared to the pregnant ewes at day 0, 15 and 75. In addition, weaned lambs were presented better FAMACHA© score compared with weaning lambs and lactating ewes at day 45 than weaning lambs at day 60 and 75. Packed cell volume in the morning and in the afternoon was influenced by days of collection with lower percentage at 30 days of experiment. The FAMACHA© system seems to be a suitable method to detect parasite infections. However, use of all available methods increases the accuracy of the diagnosis and promotes better control of gastrointestinal nematodes in grazing sheep.

#7 - Biometric, B-mode and color Doppler ultrasound assessment of eyes in healthy dogs

Abstract in English:

B-scan ultrasonography is an important diagnostic tool that allows characterization of internal organ anatomy and, when complemented by Doppler ultrasound, allows vascular hemodynamic assessment, increasing the diagnostic accuracy. Thus, the aim of the present study was the B-scan ultrasound characterization and measurement of the eyeball segments and assessment of the external ophthalmic artery by color and pulsed Doppler. Sixty eyeballs were assessed from 30 dogs of different breeds using an 8.5MHz microconvex transductor. First, biometry was performed by B-scan of the following segments: axial length (M1), anterior chamber depth (M2), lens thickness (M3), lens length (M4), vitreous chamber depth (M5), optical disc length (M6) and optic nerve diameter (M7). Colored Doppler identified the external ophthalmic article and pulsed Doppler assessed its flow, and the following were measured: systolic peak velocity (VPS), final diastolic velocity (VDF), resistivity index (IR) and pulse index (IP). No statistical difference was observed for the biometric values of the eye segments between the right and left eyes (p>0.05). The vitreous chamber depth (M5) was shown to be the biometric variable with greatest bilateral symmetry, varying from 0.79 to 0.87cm and 0.78 to 0.86cm for the right and left eye, respectively. The ophthalmic artery was visualized over the optic nerve towards the eyeball, with flow stained red. There was no significant statistical difference between the Doppler velocimetric values for the ophthalmic artery between the right and left eye of the animals assessed (p>0.05). The mean resistivity index (RI) showed average values equal to 0.63±0.03, bilaterally. The mean base velocity was 17.50cm/s and 18.18cm/s at the systolic peak and 6.21cm/s and 6.68cm/s at the end of the diastole, for the right and left eyes respectively. The anatomic, biometric and hemodynamic characterization using the ultrasound B-scan and the Doppler modalities permitted the ultrasonographic and Doppler velocimetric assessment of the eyeball components in dogs of different breeds, and it can be used in ophthalmic clinical routine to identify eye pathologies.

#8 - Sarcocystis spp. in red deer (Cervus elaphus), fallow deer (Dama dama), and pudu (Pudu pudu) in southern Chile, 37(8):874-876

Abstract in English:

ABSTRACT.- Reyes E., Paredes E. & Navarrete-Talloni M.J. 2017. Sarcocystis spp. in red deer (Cervus elaphus), fallow deer (Dama dama), and pudu (Pudu pudu) in southern Chile. Pesquisa Veterinária Brasileira 37(8):874-876. Unidad de Anatomía Patológica, Instituto de Patología Animal, Facultad de Ciencias Veterinarias, Universidad Austral de Chile, Campus Isla Teja, Valdivia, Chile. E-mail: majose.navarrete@uach.cl Worldwinde, cervids are considered an important source of infection and dissemination of a wide variety of pathogens, both for farm animals and humans. Among this diseases is sarcosporidiosis, which is a parasitic disease caused by Sarcocystis spp. (Protozoa: Apicomplexa). Most frequent clinical signs are hemolytic anemia, weakness, weight loss and decrease of growth and some species of Sarcocystis might cause abortions. The clinical disease in ruminants is fairly rare but the infection is very frequent. Infections are accumulative and the parasite does not generate immunity in any of the hosts. Ovine sarcosporidiosis is a serious issue in the some regions of Chile due to the macrocysts located in the muscle which means condemnation of the whole carcass. Sarcocystis spp. has been widely reported in red deer and other cervid species but in Chile the situation remains unknown. Nowadays there is little to no evidence of Sarcocystis in foreign deer in Chile and there is only one report of the parasite on pudu. The main goal of this study is to demonstrate the presence of Sarcocystis spp. in myocardium of red deer and fallow deer in Chile, and confirm the presence of Sarcocystis spp. in pudu. All cervid cases from 1994 to 2013 of the Institute of Animal Pathology of the Universidad Austral de Chile were reviewed. The animals selected were those in which a myocardium sample was taken. From the histopathological samples observed, it was found that 5 of the 9 red deer, 1 of the 4 fallow deer and in 11 of the 23 pudu there were Sarcocystis cysts in the myocardium. This study represents the first record for Chile of Sarcocystis spp. in myocardium of red deer and fallow deer. Stablishing the red deer, fallow deer and pudu as hosts of Sarcocystis aids to have a better understanding of the parasite epidemiology in Chile and the role of wild and captive cervids in the maintenance and spread of these parasites.

RESUMO.- Reyes E., Paredes E. & Navarrete-Talloni M.J. 2017. Sarcocystis spp. in red deer (Cervus elaphus), fallow deer (Dama dama), and pudu (Pudu pudu) in southern Chile.

#9 - Paratuberculosis: new histopathological findings in red deer (Cervus elaphus) and fallow deer (Dama dama) in Chile, 37(7):749-753

Abstract in English:

ABSTRACT.- Reyes E., Paredes E. & Navarrete-Talloni M.J. 2017. Paratuberculosis: new histopathological findings in red deer (Cervus elaphus) and fallow deer (Dama dama) in Chile. Pesquisa Veterinária Brasileira 37(7):749-753. Unidad de Anatomía Patológica, Instituto de Patología Animal, Facultad de Ciencias Veterinarias, Universidad Austral de Chile, Isla Teja, Valdivia, Chile. E-mail: majose.navarrete@uach.cl Paratuberculosis is a disease caused by Mycobacterium avium subsp. paratuberculosis (MAP) that affects domestic and wild ruminants. The most common gross lesions are emaciation and corrugation and thickening of the mucosa of the small intestine. Mesenteric lymph nodes might be enlarged. For the present study, 14 red deer and 9 fallow deer from game reserves or venison farms were analyzed. The lesions found correspond to those found by other authors in other geographic locations, except for some differences in histopathological examinations. Among these differences, stands out that intestinal lesions were concentrated mostly in the ileum and granulomas were shown to be more frequent in this section of the intestine than in the corresponding lymph node. Furthermore, in multibacillary lesions the inflammatory infiltrate in the lymph nodes was mainly composed of macrophages. These differences may be due to individual variations of the animals, the stage of disease or a different strain of the pathogen. This study allowed to obtain basic information about the disease and to describe patterns of lesions found in red deer and fallow deer with prediagnosis of clinical paratuberculosis which were not described in the literature before.

E-mail: arianneoria@ufba.br A Yellow-headed Caracara (Milvago chimachima) was submitted to the Ophthalmology Service of the Federal University of Bahia with a corneal abnormality. During ophthalmic evaluation the right cornea was stained positively with fluorescein; a blurred bullous lesion, with irregular surface, compatible with the diagnosis of bullous keratopathy was found. This is a rare condition in a bird which was treated successfully with a modified third eyelid flap associated with antibiotic and hyaluronic acid eye drops. The adopted therapeutic proved to be simple to implement and viable for repair of the bullous keratopathy in the Yellow-headed caracara.
