#1 - Evaluation of tear production in juvenile opossum using three different methods

Abstract in English:

The establishment of parameters for tear production in different species is important for better understanding eye´s health and is one of the components of the ophthalmic semiological technique. Particularities derived from the anatomophysiology of non-domestic species induce the search for more reliable methodologies. The aim was to evaluate and compare tear production of white-eared opossum (Didelphis albiventris) and Brazilian common opossum (Didelphis aurita) by three different methods. Fifteen individuals of each species, juveniles, healthy, of both sexes, with 60 to 90 days of life, were physically restrained. Phenol red thread test (PRTT), endodontic absorbent paper point tear test (EAPPTT) and modified -Schirmer tear test (mSTT) were performed. PRTT was the most difficult to perform because of the wire malleability, while EAPPTT was more feasible for both species. The median ± semi-quartile range for PRTT were 19.79±2.61mm/15 “and 5.22±2.92mm/15”, for EAPPTT were 16.25±1.82mm/min and 10.9±3.04mm/min, and for STTm were 0±1.63mm/min and 0±1.63mm/min for white-eared opossum and Brazilian common opossum respectively. There was no difference between the right and left eye neither sex. A significant difference was obtained for the same test to different species. No significant correlation was found between the tests for both species. The description of tear production parameters for juvenile white-eared opossum and Brazilian common opossum may be used as a tool, which will allow the early diagnosis of ocular diseases.

O estabelecimento do parâmetro de produção lacrimal nas diferentes espécies é importante para o entendimento da saúde do olho e é um dos componentes da semiotécnica oftálmica. Particularidades derivadas da anatomofisiologia das espécies não domésticas induzem a busca de metodologias que sejam mais fidedignas aos parâmetros. Objetivou-se com este estudo avaliar e comparar a produção lacrimal de gambás-de-orelha-branca (Didelphis albiventris) e gambás-de-orelha-preta (Didelphis aurita) por três diferentes métodos. Quinze indivíduos de cada espécie, juvenis, hígidos, de ambos os sexos, com 60 a 90 dias de vida, foram contidos fisicamente para realização do teste lacrimal do vermelho de fenol (TLVF), da ponta de papel absorvente estéril e do teste lacrimal de Schirmer modificado (TLSm). O TLVF foi o mais difícil de ser executado devido à maleabilidade do fio, enquanto a TEPA se mostrou mais exequível para ambas as espécies. A mediana ± intervalo semi-interquartil para o TLVF foi de 19,79±2,61mm/15” e 5,22±2,92mm/15”, para a TEPA foram de 16,25±1,82mm/min e 10,93±3,04mm/min, e para o TLSm foram de 0±1,63mm/min e 0±1,63mm/min, para gambás-de-orelha-branca e gambás-de-orelha-preta, respectivamente. Não houve diferença entre o olho direito e esquerdo e nem quanto ao sexo. Obteve-se diferença significativa para um mesmo teste entre as espécies. Não foi encontrada correlação significativa entre os testes para ambas as espécies. A quantificação da porção aquosa da lágrima poderá auxiliar no diagnóstico precoce de doenças oculares nas espécies estudadas.

Thirty-five healthy adult animals from a conservation breeding center in Brazil were used in this study. Modified Schirmer tear test, rebound tonometry and PFL measurements were performed in both eyes, with birds under physical restraint. Mean mSTT was 6.2±0.1mm/min and mean IOP was 6.4±0.1mmHg, while PFL was 10.1±0.1mm. A moderate correlation was seen between mSTT and PFL for OD (ρ=0.14) and OS (ρ=0.20). The results provide ophthalmic tests reference values for A. aestiva.

Foram avaliados 35 papagaios adultos e saudáveis, provenientes de um Criadouro Conservacionista do Brasil. Após avaliação clínica e laboratorial, as aves foram fisicamente contidas para aferição, em ambos os olhos, do TLSm, da PIO pela tonometria de rebote e do comprimento da FP utilizando-se um paquímetro digital. Valor médio do TLSm foi 6.2±0.1mm/min e da PIO foi 6.4±0.1 mmHg, enquanto a aferição da FP foi 10.1±0.1mm. Uma correlação moderada foi observada entre TLSm e a FP para olho direito (OD) (ρ=0.14) e olho esquerdo (OE) (ρ=0.20). Os resultados podem servir como valores de referência para testes oftálmicos para A. aestiva.

E-mail: dra.paulagalera@gmail.com The aim of this study was to establish reference values for selected ophthalmic diagnostic tests in healthy blue-and-yellow macaws. We investigated a total of 35 adult macaws (70 eyes) of undetermined sex and with an average weight of 1 kg, who were living in captivity in the Federal District, Brazil. Tear production using the Schirmer tear test (STT), normal conjunctival flora, intraocular pressure (IOP) using a rebound tonometer and horizontal palpebral fissure length (HPFL) were evaluated. In this study, 84.1% of samples were positive for microbial growth. Bacteria, fungi and yeasts were isolated, and Staphylococcus spp. (21.9%) and Bacillus spp. (26.8%) were the most frequently isolated microorganisms. The mean value for STT was 7.6±4.6mm/min in the right eye (OD) and 6.6±4.4mm/min in the left eye (OS) (median = 7,11±0,76mm/min). Mean IOP was 11.4±2.5mm Hg OD and 11.6±1.8mm Hg OS (median = 11.49±0.22mm Hg), prior to anesthesia, and 7.6±2.4mm Hg OD and 7.8±1.8mm Hg OS (median 7.71±0.08mmHg) after anesthesia. The IOP was significantly lower when the animals were under anesthesia as compared to when they were conscious (p≤0.05). Horizontal palpebral fissure length was 11.7±0.1mm OD and 11.8±0.1mm OS (median = 11.72±0.07mm). The STT showed a positive correlation with palpebral fissure measurement for this species. These selected ophthalmic reference values will be particularly useful in diagnosing pathological changes in the eyes of blue-and-yellow macaws.

Aferiram-se a produção lacrimal pelo Teste lacrimal de Schirmer (TLS), a avaliação microbiológica da conjuntiva ocular, a pressão intra-ocular (PIO) utilizando a tonometria de rebote e o comprimento horizontal da rima palpebral Neste estudo 84.1% das amostras analisadas foram positivas para crescimento microbiológico. Bactérias, fungos e hifas foram isolados e Staphylococcus spp. (21.9%) e Bacillus spp. (26.8%) foram isolados mais frequentemente. Os valores médios do teste de Lacrimal de Schirmer (TLS) foram de 7.6±4.6 e 6.6±4.4mm/min para olhos direito (OD) e esquerdo (OE), respectivamente (média = 7,11±0,76mm/min). A pressão intraocular média foi de 11.4±2.5 (OD) e 11.6±1.8mmHg (OE) anteriormente à anestesia (média 11,49±0,22 mmHg) e 7.6 ± 2.4 mmHg (OD) e 7.8 ± 1.8 mm Hg (OE) (média 7,71±0,08mm Hg) após a anestesia, verificando-se que a PIO foi significativamente menor quando os animais se encontravam sob anestesia comparativamente ao momento em que não estavam anestesiados. O comprimento horizontal da rima horizontal palpebral foi de 11.7±0.1mm OD e de 11.8±0.1mm OE (média 11,72±0,07mm). Verificou-se correlação positiva do TLS com o comprimento da fissura palpebral para a espécie estudada. Estes valores de referencia serão úteis no diagnóstico de alterações oculares da Arara Canindé.

E-mail: cristiane.honsho@unifran.edu.br The purpose of this study was to establish baseline data on lacrimal quantity (STT-1) and intraocular pressure (IOP) in captive maned wolves. Ten healthy adult maned wolves were contained with a snare pole and muzzle and kept in decubitus of the left side. STT-1 measurement was performed on the lateral third of the lower conjunctival sac for one minute. The cornea was desensitized and intraocular pressure was measured with an tonopen. Average STT-1 in both eyes was 11±5mm.min-1, with no statistical difference between the left and right eye (p=0.960). Average IOP in both eyes was 20±6mmHg, with no statistical difference between the left and right eye (p=0.836). Average STT-1 was lower than, and IOP was the same as normal levels found in dogs. There was no statistical difference in the age of the animals, and STT-1 and IOP values. In the present paper, average maned wolf STT-1 levels were lower compared with those found in dogs, while the IOP was the same in maned wolves as in dogs. Due to the increased incidence in providing emergency care for maned wolf victims of road kill and fires, determination reference values of ocular parameters may improve the correct diagnosis and treatment of the disease.

O TLS foi realizado no terço médio da pálpebra inferior, durante um minuto. A córnea foi dessensibilizada e a PIO mensurada com tonopen. A média do TLS-1 dos olhos direitos e esquerdo foi 11±5mm/min, não havendo diferença significativa entre os olhos (p=0,960). A média da PIO dos olhos direitos e esquerdo foi 20±6mmHg, não observando diferença entre os olhos direitos e esquerdos (p=0,836). Média STT-1 foi menor do que, e PIO foi o mesmo que os níveis normais em cães. Não houve diferença estatística na idade dos animais e valores STT-1 e da PIO. No presente trabalho, os níveis médios de guará STT-1 foram mais baixos em comparação com as observadas nos cães, enquanto que a pressão intraocular foi a mesma nos lobos guará como em cães. Devido ao aumento da incidência na prestação de cuidados de emergência para vítimas de lobos-guará atropelamentos e incêndios, determinar os valores de referência dos parâmetros oculares podem melhorar o diagnóstico correto e tratamento de doenças.

A total of 22 adult male rabbits were used. The ophthalmic tests included evaluation of tear production with Schirmer tear test 1(STT1) and Endodontic absorbent paper point tear test (EAPPTT) using two different commercial brand materials. Applanation tonometry, Culture of the conjunctival bacterial flora, , conjunctival cytology and conjunctival histology were also performed. Mean (±SD) for STT1, EAPPTTa, EAPPTTb and IOP was 7.27±2.51mm/min, 12.43±1.69mm/min, 15.24±2.07mm/min, 12.89±2.80mm Hg, respectively. Staphylococcus epidermidis, Staphylococcus sp. and Bacillus sp. were predominant. The cytological evaluation revealed the presence columnar epithelial cells, superficial squamous keratinized cells, lymphocytes, heterophils, red blood cells, mucus and bacteria. The histological analysis revealed a stratified epithelium, characterized by the presence of columnar epithelial cells with a large number of goblet cells. The reported data can be used for therapeutic or experimental purposes.

A média e desvio padrão do TLS1, EAPPTT1, EAPPTT2 e pressão intraocular foi 7,27±2,51 mm/min, 12,43±1,69 mm/min, 15,24±2,07 mm/min e 12,89±2,80 mmHg, respectivamente. Staphylococcus epidermidis, Staphylococcus sp. e Bacillus sp. mostraram-se predominantes. A citologia conjuntival evidenciou a presença de células epiteliais colunares, células escamosas superficiais queratinizadas, linfócitos, heterofilos, células sanguíneas, muco e bactérias. A histologia revelou epitélio estratificado caracterizado pela presença de células epiteliais colunares com grande número de células caliciformes. Os achados deste estudo poderão ser utilizados com fins terapêuticos ou experimentais.

E-mail: acrodarte@gmail.com The investigation aimed to describe the most relevant morphological characteristics of the eyes of the striped owl (Asio clamator) and determine normal values for selected ophthalmic diagnostic tests. A total of 32 eyes of 16 young and adult free-ranging healthy striped owls of both sexes were investigated. Data of different morphological observations of the skull, eye globe and adnexa, clinical tests and parameters were collected, including Schirmer Tear Test (STT), normal conjunctival bacterial microbiota, esthesiometry, intraocular pressure (IOP), central corneal thickness (CCT), horizontal palpebral fissure length, horizontal, corneal diameter and fundoscopy. Twenty two types of bacteria were identified in twelve owls, the most frequent bacteriae were the Gram-positive. The mean value found for the STT was 5.03±3.28mm/min; horizontal palpebral fissure lengths was 21.24±1.17mm, and 15.7±2.74mm for horizontal corneal diameter. The mean value for the esthesiometry was 0.80±0.59cm, IOP was 13.81±5.62mmHg and CCT was 0.28±0.03cm. This study contributes to establish reference values for selected ophthalmic diagnostic tests in the striped owl, as well as for morphological characterization of the eyeball of Asio clamator; but it is still necessary to characterize furthermore its histologic features.

Sendo compilados dados referentes a observações morfológicas do crânio, bulbo ocular e anexos, além de mensuração de testes oftálmicos, incluindo, Teste Lacrimal de Schirmer (TLS), cultura da microbiota normal da conjuntiva, estesiometria, pressão intraocular (PIO), espessura de córnea central (ECC), diâmetro horizontal da rima palpebral, diâmetro horizontal da córnea e oftalmoscopia indireta. Vinte e dois tipos de bactérias foram identificados em 12 corujas havendo predominância de microrganismos Gram-positivos. A média encontrada para o TLS foi de 5,03±3,28mm/min, para o diâmetro horizontal da rima palpebral em 16 corujas foi 21,24±1,17mm, e 15,7±2,74mm para o diâmetro horizontal da córnea. O valor médio para o teste de estesiometria foi de 0,80±0,59cm, a PIO média de 13,81±5,62mmHg e ECC média de 0,28±0,03cm. O estudo contribuiu para a caracterização da morfologia ocular e para o estabelecimento de valores de referências de testes diagnósticos oftálmicos em corujas-orelhudas, sendo necessário ainda o desenvolvimento de estudos complementares sobre histologia ocular desta espécie.


#7 - Comparison of two methods of tear sampling for protein quantification by Bradford method, 33(2):261-264

E-mail: paulaeye@unb.br The aim of this study was to compare two methods of tear sampling for protein quantification. Tear samples were collected from 29 healthy dogs (58 eyes) using Schirmer tear test (STT) strip and microcapillary tubes. The samples were frozen at -80oC and analyzed by the Bradford method. Results were analyzed by Student’s t test. The average protein concentration and standard deviation from tears collected with microcapillary tube were 4.45mg/mL ±0.35 and 4,52mg/mL ±0.29 for right and left eyes respectively. The average protein concentration and standard deviation from tears collected with Schirmer Tear Test (STT) strip were and 54.5mg/mL ±0.63 and 54.15mg/mL ±0.65 to right and left eyes respectively. Statistically significant differences (p<0.001) were found between the methods. In the conditions in which this study was conducted, the average protein concentration obtained with the Bradford test from tear samples obtained by Schirmer Tear Test (STT) strip showed values higher than those obtained with microcapillary tube. It is important that concentration of tear protein pattern values should be analyzed according the method used to collect tear samples.

E-mail: paulaeye@unb.br Compararam-se dois métodos de coleta de lágrima para quantificação proteica, utilizando-se 58 olhos de 29 cães hígidos. As amostras foram coletadas utilizando-se fitas de teste da lágrima de Schirmer (Schirmer tear test - STT) e tubos microcapilares. Após obtidas, as amostras foram congeladas a -80o C e posteriormente analisadas pelo método de Bradford. Os resultados foram analisados pelo teste T de Student. A média da concentração proteica e desvio padrão das amostras obtidas com microcapilar foi de 4,45mg/mL ±0,35 e 4,52mg/mL ±0,29 para olhos direito e esquerdo, respectivamente. Para as amostras obtidas com STT os resultados foram 4,45mg/mL ±0,35 and 4,52mg/mL ±0,29 para olhos direito e esquerdo, respectivamente. Diferenças estatisticamente significativas (p<0,001) foram encontradas entre os dois métodos. Nas condições em que o trabalho foi conduzido, a média da concentração proteica pelo método de Bradford das amostras obtidas através das tiras do STT foi superior à obtida com o tubo microcapilar. Os valores padrão da concentração protéica da lágrima obtida pelo método de Bradford devem ser analisados considerando-se o método de coleta da lágrima, uma vez que este interfere significativamente nos resultados obtidos.

E-mail: jllaus@fcav.unesp.br The study aimed to evaluate tear production by means of modified Schirmer tear tes-1 (mSTT-1) in neonate cats. Likewise, correlation between mSTT-1 and STT-1 was assessed in vitro. Standard SST strips were cut in half and after eye lid opening, tear production of neonates (n=15) was daily measured in both eyes (mSTT-1), until the 7th day, and at day 14, 21, and 28. Animals were daily weighted until 28 days of age. Results were compared statistically (P<0.05). During the first 7 days, the overall mSTT-1 mean was 0.76 wetting/minute. Significant differences between right and left eyes were not observed at any time point (P=1.00). Tear secretion increased significantly, from the 14th to 28th day, in comparison with 7 first days (P<0.05). Positive correlation between maturity parameters and tear secretion was observed (P<0.0001). Distance between slopes of each strip changed significantly (P<0.0001). It was concluded that tear secretion in the neonatal period of cats is very below the reference values described for young and adults of the same species. It is not possible to extrapolate results obtained with mSTT-1 to standard STT-1.

Tiras pa-dronizadas para realização do TLS foram partidas ao meio e após a abertura das pálpebras, a produção lacrimal dos neonatos (n=15) foi aferida em ambos os olhos (TLS-1m), diariamente, até o sétimo e aos 14, 21 e 28 dias. Os animais foram pesados diariamente até os 28 dias de idade. Os resultados foram comparados estatisticamente (p<0,05). Durante os primeiros sete dias de avaliação, a média geral obtida pelo TLS-1m foi de 0,76±0,08 mm/minuto. Não se constatou diferença significativa entre os olhos direito e esquerdo, em nenhum dos períodos avaliados (p=1,00). A produção lacrimal elevou-se significativamente, do 14º até o 28º dia, comparativamente aos primeiros sete dias (p<0,05). Observou-se correlação positiva entre parâmetros de maturidade e a produção lacrimal (p<0,0001). A distância entre as linhas de cada tira foi significativa (p<0,0001). Conclui-se que a produção lacrimal no período neonatal em gatos é bem inferior aos valores de referência descritos para adultos e jovens da mesma espécie. Não é possível extrapolar valores obtidos com o TLS-1 modificado para o TLS-1 padrão.

E-mail: jllaus@fcav.unesp.br This study aimed to compare the normal intraocular pressure (IOP) and tear secretion, by means of applanation tonometry and the Schirmer tear test-1 (STT-1), in goats of the Saanen breed with different ages, and at different time points. Thirty six goats, free of ocular abnormalities, were grouped into three different age categories (n=12), animals with 45, 180 and 549 days of age. STT-1 and IOP measurements were carried out always at 9:00am and 7:00pm, during three consecutive days. Results were evaluated statistically (P<0.05). Regarding the time of the day, overall IOP values were significantly lower at 7:00 pm (P<0.001) in individuals with 45 days of age; whereas STT-1 values were significantly higher at 7:00pm (P=0.004) in goats with 549 days of age. Considering the sum of three days, both parameters were significantly lower in individuals with 45 days of age (P<0.001). Intraocular pressure and tear secretion values increase until 180 days of age in the Saanen breed of goats.

E-mail: jllaus@fcav.unesp.br Objetivou-se comparar os valores normais de pressão intraocular (PIO) e de produção lacrimal, através de tonometria de aplanação e do teste da lágrima de Schirmer-1 (TLS-1) em cabras da raça Saanen, com diferentes idades e momentos. Trinta e seis cabras da raça Saanen, livres de afecções oculares, foram agrupadas em três categorias etárias (n=12), compreendendo animais com 45, 180 e 549 dias de idade. O TLS-1 e a PIO foram aferidos sempre às 9:00 e às 19:00, durante três dias consecutivos. Os resultados foram avaliados estatisticamente (p<0,05). Relativamente ao horário do dia, a PIO foi significativamente menor às 19:00 (p<0,001) nos animais com 45 dias de idade; os valores do TLS-1 foram significativamente maiores às 19:00 (p=0,004) nas cabras com 549 dias de idade. Considerando-se a somatória dos três dias, ambos os parâmetros foram significativamente menores nos indivíduos com 45 dias de idade (p<0,001). A pressão intraocular e a produção lacrimal em cabras da raça Saanen se elevam de forma significativa até os 180 dias de idade.
