#1 - Determination of the reference interval of the C-reactive protein/albumin ratio and its efficiency, CRP and albumin as prognostic markers in dogs

Abstract in English:

The objective of this research was to creates a reference interval for C-reactive protein (CRP)/albumin ratio (CAR) in the canine species and to analyze the potential of CRP, albumin and the relationship between both, to serve as indicators of disease severity, length of hospital stay (LoS) and mortality in this species. For this, an outcome study was conducted in a Veterinary Teaching Hospital in southern Brazil. One hundred ninety dogs were included randomly, without distinction of gender, age, or breed, from June 2013 to November 2016. Plasma was collected from them and analyzed for assessment of CRP and albumin. The reference range stipulated for CAR in dogs was 0.36-0.60, as determined by the confidence interval of mean resamplings (in percentiles). The frequencies mean, and standard deviations of the variables, correlation analysis, and comparative analysis (Kruskal-Wallis in α = 5%) were calculated. Elevation (above reference) of CAR was determined to be proportional to the severity of the underlying disease, and CRP means were reasonable. Besides, hypoalbuminemia was indicative of systemic disease, but not of severity. Thus, CAR was a better marker of disease severity than were CRP and albumin, analyzed separately. Concerning LoS, there was a positive correlation with CAR (p<0.01) in patients, and the same was not observed with CRP and albumin. Concerning mortality, hypoalbuminemia was the only marker valid in animals with a critical illness (p=0.04). In conclusion, CAR is a better marker of disease severity and LoS in dogs than are CRP and albumin analyzed separately.

O objetivo desta pesquisa foi determinar um intervalo de referência para a relação proteína C reativa (PCR)/albumina (R:PCR/ALB) na espécie canina e analisar o potencial de PCR, albumina e a relação entre ambas como indicadores de gravidade de doença, tempo de internação (TI) e mortalidade nesta espécie. Para isso, um estudo foi realizado em um Hospital Veterinário Escola no sul do Brasil. Cento e noventa cães foram incluídos aleatoriamente, sem distinção de sexo, idade ou raça, de junho de 2013 a novembro de 2016. O plasma foi coletado e analisado para avaliação da PCR e albumina. O intervalo de referência estipulado para o R:PCR/ALB em cães foi de 0,36-0,60, conforme determinado pelo intervalo de confiança da média das reamostragens (em percentis). Foram calculadas as frequências, médias e desvios-padrões das variáveis, análises de correlação e análises comparativas (Kruskal-Wallis em α = 5%). Notou-se elevação (acima da referência) da R:PCR/ALB proporcional à gravidade da doença de base, sendo normais as médias da PCR. Adicionalmente, a hipoalbuminemia foi indicadora de doença sistêmica, mas, não de gravidade. Dessa forma, a R:PCR/ALB foi melhor indicadora de gravidade de doença do que a PCR e albumina, analisadas separadamente. Em relação ao TI, houve correlação positiva com a R:PCR/ALB (p<0,01) em doentes, não sendo observado o mesmo com a PCR e albumina. Em relação à mortalidade, a hipoalbuminemia foi a única marcadora válida em animais com doenças críticas (p=0,04). Conclui-se, portanto, que a R:PCR/ALB é melhor marcadora de gravidade de doença e TI em cães do que a PCR e albumina analisadas separadamente.

E-mail: alexamazza@yahoo.com.br A retrospective study including the analysis of the cerebrospinal fluid (CSF) of dogs neurologically affected was conducted by the Neurology Service of the Veterinary Hospital at the Institution, between 2004 and 2015. The aim of this study was to analyze the results of the CSF of dogs with neurological signs, and compare the changes in the CSF in two sampling sites in the same patient and see if this test helped the clinician to strengthen clinical suspicion of the major diseases of the central nervous system. Lymphocytic pleocytosis was present in 78.3% (29/37) of samples from dogs with distemper and in 23.2% (10/43) of samples from dogs with IVDD. The albumin cytologic dissociation (ACD) was found in 73% (19/26) of samples from dogs with IC tumors and in 64.3% (9/14) from dogs with tumors involving the SC. For dogs with IVDD, there was statistical significance (p<0.05) between the degree of neurological dysfunction and the total nucleated cells (TNC) and total protein (TP). In 29 dogs, CSF was collected from the cistern magna and the lumbar and in 12 (41.4%) the results were different between the samples of the same dog, where two cases (6,9%) showed alterations in the sample collected cranial to the injury. It can be concluded that the lymphocytic pleocytosis was the main alteration found in the CSF of dogs with distemper and IVDD and ACD in tumors. Dogs affected by IVDD had more severe neurological signs as TNC and TP increased and the CSF was altered even collected cranial to the lesion site and helped the clinician to strengthen the clinical suspicion, but not confirm, the major neurological diseases in dogs.

A pleocitose linfocítica esteve presente em 78,3% (29/37) das amostras de cães com cinomose e em 23,2% (10/43) de cães com DDIV. Houve dissociação albuminocitológica (DAC) em 73% (19/26) das amostras de cães com tumores IC e em 64,3% (9/14) de cães com tumores envolvendo a ME. Em cães com DDIV, houve significância estatística (p<0,05) entre o grau de disfunção neurológica e o total de células nucleadas (TCN) e total de proteínas (TP). Em 29 cães, houve a colheita do LCE da cisterna magna e da cisterna lombar e em 12 (41,4%) os resultados foram diferentes entre as duas amostras colhidas do mesmo cão, onde dois (6,9%) apresentaram alteração na amostra colhida cranial à lesão. Pode-se concluir que a pleocitose linfocítica foi a principal alteração encontrada no LCE de cães com cinomose e DDIV e DAC nas neoplasias, IC e ME, cães acometidos pela DDIV apresentaram sinais neurológicos mais severos conforme o TCN e o TP aumentaram e o LCE sofreu alteração, mesmo colhido cranial ao local da lesão e auxiliou o clínico em reforçar a suspeita clínica, mas não confirmou, as principais doenças neurológicas em cães.

E-mail: cacavann@usp.br The study of the organic interaction and physiological adaptations during pregnancy is of utmost importance for clinical evaluation and diagnosis of pathological conditions of pregnant bitches. The aim of the present study was to compare the serum protein profile (total protein, albumin, a1, a2, b and g globulin) of pregnant and diestrous bitches. For this purpose, 40 healthy 2 to7-year-old Great Dane bitches were used. The bitches were allocated in two experimental groups: Non-pregnant group (NP; n=20) and pregnant group (P; n=20). From each female, blood was drawn weekly during 9 weeks, from the diestrous onset until the beginning of anestrus or parturition, respectively from NP and P groups. The concentration of total protein, albumin and a1, a2, b and g globulins were determined from serum samples. No statistical difference was found for total protein between 1st and 6th weeks of pregnancy or diestrus. In both groups, there was a progressive decline in total protein concentration. At the end of pregnancy (7th to 9th week), a significant increase in total protein was verified, suggesting an anabolic process. Albumin concentration decreased between the 1st and 9th week in both groups, however, more markedly in the P group (with significant difference between groups at 7th, 8th and 9th week). The levels of &#945;1 globulin in pregnant bitches increased significantly from the 2nd week on. Statistical difference was observed between groups only at the 8th and 9th week, during which a preparatory phase for parturition occurs. A significant rise in &#945;2 globulin was shown between the 2nd and 3rd week of gestation, however without difference from the NP group. There was significant difference for &#945;2 globulin between groups at the 4th, 5th, 6th, 8th and 9th week. Serum concentration of b globulin in diestrous bitches was superior than pregnant bitches at the 2nd, 4th and 7th week. During early gestation (1st to 3rd week) there was a decrease in g globulin, simultaneously to the preimplantation period and formation of implantation sites. An increase in g globulin was shown between the 8th and 9th week in pregnant bitches, possibly due to the increase in immunoglobulin synthesis targeting the mammary gland to form the colostrum. In conclusion, markedly changes in protein profile occur during gestation. It was possible to state different biological function of blood proteins in pregnant and diestrous bitches. The determined proteins are enrolled in the inflammatory stimulus during gestation, as well as in the hormonal regulatory mechanisms and maternal preparatory process to lactation.

E-mail: cacavann@usp.br O estudo das interações orgânicas da gestação e as mudanças fisiológicas que estão ocorrendo nesta fase são de extrema importância para a avaliação clínica da fêmea gestante ou para estabelecer o diagnóstico de processos patológicos em andamento. O objetivo do presente estudo foi comparar o perfil das diversas proteínas sanguíneas (frações protéicas do soro - albumina, a1, a2, b, g globulinas e proteína total) durante o período gestacional e no diestro em cadelas. Foram utilizadas 40 fêmeas caninas da raça Dogue Alemão, em idade variando entre 2 a 7 anos, clinicamente saudáveis. Os animais foram separados em dois grupos, denominados Grupo não gestante (NG) e Grupo gestante (G), constituídos por 20 fêmeas em diestro e 20 fêmeas gestantes, respectivamente. Preconizou-se colheita de sangue semanalmente de cada animal durante 9 semanas. Nas fêmeas do grupo NG, as amostras foram colhidas a partir do início do diestro, até a detecção do início do anestro; no grupo G, as amostras foram colhidas do início do diestro até o momento da parição. A partir do soro sanguíneo, foram determinadas as concentrações de proteína total, albumina, a1, a2, b e g globulinas. Não houve diferença significativa quanto aos resultados de proteína sérica total entre a 1ª e 6ª semanas de gestação e diestro, havendo decréscimo gradual em ambos grupos. Já ao final da gestação (entre a 7ª e 9ª semanas), houve acréscimo significativo dos valores de proteína total, sugerindo ação anabólica. Os níveis de albumina sofreram queda da 1ª a 9ª semana, tanto no grupo gestante, como não-gestante, com diferença estatística entre os dois grupos na 7ª, 8ª e 9ª semanas. As concentrações de &#945;1 globulina nas fêmeas gestantes sofreram acréscimo significativo a partir da 2ª semana, contudo, diferença estatística entre os grupos NG e G ocorreu somente na 8ª e 9ª semanas, coincidindo com a fase de preparação à parição. Houve aumento no perfil de &#945;2 globulina entre a 2ª e 3ª semanas de gestação, porém tais valores não diferiram das fêmeas em diestro. Diferença significativa de &#945;2 globulina foi observada apenas durante a 4ª, 5ª, 6ª, 8ª e 9ª semanas entre os dois grupos avaliados. As concentrações séricas de &#946; globulina nas cadelas não-gestantes foi superior às das fêmeas gestantes durante a 2ª, 4ª e 7ª semanas. Durante o primeiro terço da gestação (1ª a 3ª semana), foi observado queda dos valores de &#947; globulina, coincidente ao período pré implantacional e durante a formação dos sítios de implantação. Entre a 8ª e 9ª semanas de gestação, houve acréscimo significativo de &#947; globulina, possivelmente conseqüente ao aumento da produção de imunoglobulinas direcionadas à glândula mamária, como constituinte do colostro. Em conclusão, as proteínas alteram-se de forma evidente durante o período de gestação. Foi possível inferir diferenças nas funções biológicas das proteínas sanguíneas em cadelas gestantes e não gestantes. As proteínas determinadas estão envolvidas com o estímulo inflamatório durante a gestação, além dos mecanismos de regulação hormonal e preparação do organismo materno à lactação.

There are many studies to find better results for the organic sources of minerals, and one of the reasons for that is to avoid the negative interactions. The metabolic profile helps in assessment of production indices, therefore analyzes blood biochemical components. The objective of this study was to investigate the effects of organic and inorganic copper and sulphur supplementation on blood biochemical parameters in sheep. The experiment was conducted at the FZEA/USP. Total 40 weaned sheep were divided in a completely randomized design with 10 treatments: 1) basal diet; 2) basal diet plus Mo; 3) basal diet + inorganic Cu + inorganic S; 4) basal diet + inorganic Cu + organic S; 5) basal diet + organic Cu + inorganic S; 6) basal diet + organic Cu + organic S; 7) diet plus Mo + inorganic Cu + inorganic S; 8) diet plus Mo + inorganic Cu + organic S; 9) diet plus Mo + organic Cu + inorganic S; 10) diet plus organic Cu + organic S. According each treatment was added 10mg kg-1 MS inorganic or organic Cu or 10mg kg-1 MS Mo or 0.2% S inorganic or organic. The animals were fed twice a day, a total of 3% of body weight. The experiment lasted 84 days, blood was sampled every 28 days to study glucose, urea, albumin, cholesterol and triglycerides. The parameters were analyzed with factorial structure 2 x 2 x 2 (with and without Mo, organic and inorganic Cu and organic and inorganic S) and a basal diet and a basal diet plus Mo, with 5% significance. Serum levels of glucose, urea, albumin and cholesterol showed no significant difference between treatment and time (p> 0.05). It observed means of 64.8mg dL- 1; 30.0mg dl -1; 2.78mg L -1; 72.2mg dL- 1 respectively for glucose, urea, albumin and cholesterol. Serum levels of triglycerides in the treatments (28.8; 34.8; 30.8; 36.9; 34.3; 27.0; 31.6; 32.1; 34.6; 31.1mg dL- 1) were influenced by interaction Cu x S, triglycerides decreased with Cu and S organic.

O perfil metabólico auxilia na avaliação de índices produtivos, para tanto, se faz análises de componentes bioquímicos do sangue. O objetivo foi estudar os efeitos que fontes orgânicas e inorgânicas de cobre e enxofre possuem nos parâmetros bioquímicos. O experimento foi realizado na FZEA/USP, para tanto 40 ovinos foram distribuídos em 10tratamentos: 1) dieta basal; 2) dieta contendo Mo; 3) dieta basal+ Cu inorg + S inorg; 4) dieta basal + Cu inorg + S org; 5) dieta basal + Cu org + S inorg; 6) dieta basal + Cu org + S org; 7) dieta com Mo + Cu inorg + S inorg; 8) dieta com Mo + Cu inorg + S org; 9) dieta com Mo + Cu org + S inorg; 10) dieta com Mo + Cu org + S org. De acordo com cada tratamento houve a inclusão de 10mgkg-1 de MS de Cu inorgânico ou orgânico ou 10 mg kg-1 de MS de Mo ou 0,2% de S inorgânico ou orgânico. Os animais receberam dieta única duas vezes ao dia com inclusão de volumoso num total de 3% do peso vivo. O experimento teve duração de 84 dias, com coletas a cada 28 dias para estudo de glicose, ureia, albumina, colesterol, triglicerídeos. Os parâmetros foram analisados com estrutura fatorial 2 x 2 x 2 (com e sem Mo, Cu orgânico e inorgânico e S orgânico e inorgânico) e uma dieta basal e uma basal mais molibdênio, em delineamento inteiramente casualizado, com um nível de significância de 5%. Os teores séricos de glicose, ureia, albumina e colesterol não apresentaram diferença significativa entre tratamentos e tempo, tão pouco foram influenciados (p>0.05) pelos tratamentos, sendo as médias 64,8mg dL-1; 30,0mg dL-1; 2,78mg L-1; 72,2mg dL-1respectivamente, apresentando-se dentro da normalidade. Os teores de triglicerídeos nos tratamentos (28.8; 34.8; 30.8; 36.9; 34.3; 27.0; 31.6; 32.1; 34.6; 31.1mg dL-1) foram influenciados pela interação Cu x S.

E-mail: carolina.cavalcante@pucpr.br Hyperadrenocorticism is one of the commonest endocrinopathies in dogs, and it is characterized by the excessive exposure of glucocorticoids excreted by adrenals. Chronic hypercortisolemia may promote several complications, including systemic hypertension and glomerulonephritis. Glomerulonephritis may initiate several variable degrees of proteinuria and leading to the development of chronic kidney disease. The loss of proteins through urine, mainly predominant albumin, is a characteristic of glomerular diseases and the determination of laboratorial variables, such as the urinary protein-to- creatinine ratio (UPC), urinary albumin-to-creatinine ratio (UAC; ELISA test) and electrophoresis of urinary proteins are recommended to elucidate the diagnosis. Therefore, the goal of this study is to evaluate the relationship between proteinuria and systemic arterial hypertension in dogs with hyperadrenocorticism and to determine through evaluation of albuminuria and molecular weight of urinary proteins, the segment of the nephron that could be damaged. Thirty dogs with hyperadrenocorticism were evaluated and subdivided into groups; 13 dogs with systemic arterial hypertension (group I) and 17 normotensive (group II). The UPC was determined, as well as UAC and the urine protein electrophoresis by polyacrylamide gel technique, containing dodecyl sodium sulphate (SDS-PAGE). The results were compared with data obtained from 30 clinically healthy dogs. No association between systemic arterial hypertension and albuminuria was detected in dogs with hyperadrenocorticism as well as no alterations of proteins patterns or molecular weights bands of low (<60 kDa) or high molecular weight (> 60 kDa) was found. However, dogs with hyperadrenocorticism may develop glomerular and tubular injuries that were characterized by the presence of albuminuria and proteins of low and high molecular weights, independently of systemic arterial hypertension. In conclusion, the quantitative (UPC and UAC) and qualitative (SDS-PAGE) evaluation of urinary proteins could add information to indicate the possible segments of the nephrons that caused the loss of those proteins.

Assim, o objetivo do estudo é avaliar a relação entre proteinúria e hipertensão arterial sistêmica em cães com hiperadrenocorticismo e verificar, pela avaliação da albuminúria e do peso molecular das proteínas urinárias, o segmento do néfron que foi comprometido ou lesado. Foram avaliados 30 cães com diagnóstico de hiperadrenocorticismo, subdivididos em 13 cães com hipertensão arterial sistêmica (grupo I) e 17 cães normotensos (grupo II). Foram determinados a RPC; a albuminúria pela avaliação da albumina normalizada e razão albumina:creatinina urinária (RAC) e a eletroforese de proteínas pela técnica em gel de poliacrilamida, contendo dodecil sulfato de sódio (SDS-PAGE). Os resultados foram comparados com os dados obtidos de 30 cães clinicamente saudáveis. Foi constatado que não houve influência da hipertensão arterial sistêmica nos cães com hiperadrenocorticismo em relação à quantificação da albuminúria, determinada pelo método ELISA, e nem na qualidade e quantidade das bandas de proteínas de baixo (<60 kDa) e de alto peso molecular (>60 kDa). No entanto foi determinado que cães com hiperadrenocorticismo podem desenvolver lesões glomerulares e tubulares, caracterizadas pela presença de albuminúria e de proteínas de alto e de baixo pesos moleculares, independentemente da presença de hipertensão arterial sistêmica. Conclui-se que a avaliação quantitativa (RPC e RAC) e qualitativa (SDS-PAGE) das proteínas urinárias traz informações adicionais que indicam os possíveis segmentos comprometidos dos néfrons que causaram as perdas de proteínas na urina.

Cows were distributed into three groups as follows: early (30-120 days), middle (121-210 days) and end of lactation (more than 211 days). The proteinogram consisted of total proteins concentration determined by biuret method and polyacrymalide gel electrophoresis (SDS-PAGE). A gradual and significant decrease of some fractions of the whey was observed during the lactation cycle albumin, lactoferrin, immunoglobulins, &#946;-lactoglobulin and &#945; -lactoalbumin. The normal ranges obtained for the whey proteins of Jersey cows were: whey total proteins 569.0 to 713.0mg/dL, lactoferrin 36.0 to 49.0mg/dL, albumin 24.0 to 34.0mg/dL, immunoglobulin’s heavy chain 38.0 and 51.0mg/dL; immunoglobulin’s light chain 59.0 to 95.0mg/dL, &#946;-lactoglobulin 207.0 to 256.0mg/dL, &#945;-lactoalbumin 117.0 to 157.0mg/dL; protein with 226 KDa 5.80 to 12.0mg/dL, and protein with 118 KDa 2.30 to 6.80mg/dL.

Knowledge of the physiology of these animals and serum proteins concentration changes are important for early identification of major diseases which lead to states of hypoproteinemia and hyperproteinemia. The objective was to determine the concentration of total protein and serum protein electrophoresis profile of Crotalus durissus terrificus (rattlesnake) in captivity. Blood samples were taken from the ventral coccygeal vein of 21 adult and healthy snakes divided into groups: Group 1 with 12 males, weighing in average 588.89±193.55g, and Group 2 with nine females, weighing in average 708.33±194.04g. The total serum concentration of protein was determined by the method of refractometry and agarose gel electrophoresis. The total protein values in the serum for females was 4.82±0.72, for males 4.51±0.50 and males and females 4.64±0.61, identified by four fractions (g/dL): albumin, a, b and g-globulin. Additionally the albumin/globulin ratio was calculated. The female snakes showed higher values for the variables, albumin and the albumin/globulin (AG) differed significantly (P<0.05) from the group of male snakes, but there was no clinical significance.
