#1 - Morphology and biometry of the reproductive organs of adult males of Trachemys scripta elegans reared in São Paulo state, Brazil

Abstract in English:

Trachemys scripta elegans is an American underwater chelonian illegally marketed in Brazilian pet shops. When abandoned in nature, it compromises native species, threatening local biodiversity. However, little is known about the body development and structure of its reproductive tract. The objective of the present study was to investigate the morphology and biometry of testis, epididymis and penis, as well as the biometry of the body and secondary sexual characters in this species. Twenty-seven adult males were used aiming to contribute to preservation actions in captivity, population control, and scientific research, as well as to interspecific comparisons. Sex identification by the third claw length was effective, and the specimens presented harmonious and positive body development between mass, carapace, plastron, and height, with unimodal tendency and higher frequency of maximum carapace length at 15cm. The testes and epididymides presented biometric similarity between the antimeres and anatomical and histological structure similar to that of other species of chelonians and mammals, except for the type of epithelium. The findings suggest that there is conserved morphology between slider turtles and homology in relation to mammals. Histological similarity to the reproductive organs of other amniotes, including humans, may give rise to scientific and comparative studies, essential for the establishment of conservation strategies in reptiles.

Trachemys scripta elegans é um quelônio subaquático americano ilegalmente comercializado em pet shops brasileiros. Ao ser abandonado na natureza, compromete as espécies nativas, ameaçando à biodiversidade local. No entanto, pouco se conhece sobre o desenvolvimento corporal e a estrutura do seu aparelho reprodutor. O objetivo do presente trabalho foi investigar a morfologia e a biometria dos testículos, epidídimos e pênis, a biometria corporal e dos caracteres sexuais secundários. Foram utilizados 27 machos adultos desta espécie, visando contribuir com ações de preservação em cativeiro, controle populacional e pesquisas científicas, além de comparações interespecíficas. A identificação sexual pelo comprimento da terceira garra foi efetiva e os espécimes apresentaram desenvolvimento corporal harmônico e positivo entre massa, carapaça, plastrão e altura, com tendência unimodal e maior frequência de comprimento máximo de carapaça em 15,0cm. Testículos e epidídimos apresentaram semelhança biométrica entre os antímeros e estrutura anatômica e histológica semelhantes à de outras espécies de quelônios e mamíferos, excetuando-se pelo tipo de epitélio. Os achados sugerem haver morfologia conservada entre os cágados e homologia em relação aos mamíferos. A semelhança histológica com os órgãos reprodutivos de outros amniotas, incluindo os humanos, pode dar ensejo a estudos científicos e comparativos, essenciais para estabelecimento de estratégias de conservação em répteis.


#2 - Effects of vincristine sulphate on albino rat testis treated in the prepuberal phase

Abstract in English:

This study evaluated the vincristine sulfate effect on rat testes treated in pre pubertal stage, especially regarding the changes of Sertoli cells and germ cells. Thirty control rats and 30 rats treated with vincristine sulfate were used. The drug application occurred at 15 days of life, and euthanasia at 40, 64 and 127 days of life to enable evaluation at different stages of reproductive development. Biometric measurements were performed (body and testicular weights), testicular morphometric measures (major and minor testicular axis and of seminiferous tubule and seminiferous lumen) and stereological (testicular volumes and volume densities of the tubular and testicular interstitial tissue). The biometric measurements were made on all rats in the experiment, and morphometric and stereological analysis was carried out in 200 seminiferous tubules. The results demonstrate that vincristine sulfate reduces biometric parameters such as body weight, testicular weight and the total testicular volume. Morphometric and stereological variables as diameter of the seminiferous tubules, height of the seminiferous epithelium and volume of the seminiferous tubules were also reduced. The most affected cell types were spermatogonia, late spermatids and Sertoli cells.

Este trabalho visou avaliar os efeitos de sulfato de vincristine sobre os testículos de ratos tratados na fase pré púbere, sobretudo quanto às alterações das células de Sertoli e das células germinativas. Foram utilizados 30 animais controles e 30 tratados com sulfato de vincristine. As aplicações da droga ocorreram aos 15 dias de vida, e a eutanásia aos 40, 64 e 127 dias de vida para possibilitar a avaliação em diferentes estágios de desenvolvimento reprodutivo. Foram realizadas medidas biométricas (pesos corpóreos e testiculares), medidas morfométricas testiculares, (eixos testiculares maiores e menores, diâmetros testiculares de túbulo e lúmen seminíferos, e altura do epitélio seminífero) e estereológicas (volumes testiculares e as densidades de volume do tecido tubular e do tecido intersticial testicular). As medidas biométricas foram feitas em todos os animais do experimento, e as avaliações morfométricas e estereológicas foram realizadas em 200 túbulos seminíferos. Os resultados demonstraram que sulfato de vincristine reduz parâmetros biométricos como peso corpóreo, peso testicular e volume testicular total. Variáveis morfométricas e estereológicas como diâmetro dos túbulos seminíferos, altura do epitélio seminífero e volume dos túbulos seminíferos também foram reduzidos. Os tipos celulares mais atingidos foram as espermatogônias, espermátides tardias e células de Sertoli.

E-mail: reginacrpaz@gmail.com The pampas cat Leopardus colocolo (Molina, 1782) is a species of the Felidae family, widely distributed in South America, included on CITES Appendix II and classified as Near Threatened on the IUCN Red List, with population trend decreasing. Based on this information, the objective of this study is to describe morphologically the testes and epididymal ducts of pampas cat. The animal, coming from the Federal University of Mato Grosso Zoo, Brazil, had died after anesthesia procedure and the male reproductive system was dissected to collect the testicles. The samples taken were fragmented and histologically examined. From the microscopic analysis of the testes were identified: vaginal and tunica albuginea, formed by dense connective tissue modeled with large amount of collagen fibers. The tunica albuginea fibrous septa emits into the body. The seminiferous tubules are coiled and coated internally by spermatogenic epithelium consisting of Sertoli cells, surrounded by a basement membrane in the presence of myoid cells. The interstitial tissue between the seminiferous tubules, is composed of loose connective tissue, blood and lymph vessels, and Leydig cells in polyhedral shape. The epididymal ducts showed pseudostratified columnar epithelium with secretory cells of which stereocilia design, situated on a basement membrane filled by myoid cells. This epithelium has principal and basal cells, the main cell design stereocilia toward the lumen of the epididymal duct.

O animal, oriundo do Zoológico da Universidade Federal de Mato Grosso, Brasil, veio a óbito após procedimento anestésico e o sistema reprodutor masculino foi dissecado para coleta dos testículos. As amostras retiradas foram fragmentadas e histologicamente examinadas. A partir das análises microscópicas dos testículos foram identificados: a túnica vaginal e albugínea, formada por tecido conjuntivo denso modelado, com grande quantidade de fibras de colágeno. A túnica albugínea emite septos fibrosos para o interior do órgão. Os túbulos seminíferos são enovelados e revestidos internamente por epitélio estratificado constituído por células espermatogênicas e células somáticas de Sertoli, envolvidos por uma membrana basal com presença de células mioides. O tecido intersticial, entre os túbulos seminíferos, é constituído de tecido conjuntivo do tipo frouxo com vasos sanguíneos e linfáticos, e células de Leydig em formato poliédrico. Os ductos epididimários apresentaram epitélio cilíndrico pseudoestratificado com células secretoras dos quais projetam estereocílios, situados sobre uma membrana basal repleta por células mióides. Este epitélio apresenta células principais e basais, cujas células principais projetam estereocílios em direção ao lúmen do ducto epididimário.

E-mail: mirley.souza@gmail.com This study aimed to assess whether there are differences in Doppler velocimetry parameters between different sizes. Twenty dogs were equally divided into small and large groups used in this study. The dogs were evaluated using Triplex ultrasound. Testicular artery was located by Colour Doppler in the spermatic cord, marginal to the testes and intratesticular segments and then, spectral Doppler were used to calculate: peak systolic velocity (PSV), end diastolic velocity (EDV), resistance index (RI) and pulsatility index (PI). The mean testicular volume in the left side was significantly higher than the right side, in both groups. Doppler examination showed higher velocities (EDV) at spermatic cord in large dogs; marginal to the testes was observed higher velocities in small dogs; intratesticular region no differences were observed (P < 0.05) and within the groups differences between segments of the artery were also observed for each parameter. The results showed that there are differences in Doppler velocimetry parameters between different sizes.

E-mail: mirley.souza@gmail.com Este trabalho teve como objetivo verificar se existem diferenças nos parâmetros dopplervelocimétricos entre cães de diferentes portes. Para tanto, foram utilizados 20 cães, sendo 10 cães de pequeno porte e 10 cães de grande porte. Foram avaliados pelo Triplex Doppler para localização da artéria testicular nos segmentos de cordão espermático, marginal ao testículo e intratesticular. Após isso, o Doppler espectral foi acionado para cálculo dos parâmetros de velocidade de pico sistólico (VPS), velocidade diastólica final (VDF) e índices de resistência (IR) e pulsatilidade (IP). O volume testicular médio do testículo esquerdo foi significativamente maior que o direito em ambos os grupos. Ao Doppler, foram observadas diferenças dos valores entre os portes, sendo as velocidades no cordão espermático superiores nos animais de grande porte (P < 0,05) e dentro dos grupos também foram observadas diferenças entre as regiões da artéria testicular. Diante do exposto, conclui-se que existem diferenças nos parâmetros dopplervelocimétricos quando comparados animais de diferentes portes, além disso, os parâmetros dopplervelocimétricos são diferentes dependendo da região em que são mensurados.


#5 - Body and testicular development in males of meat- and egg-type quails of 25 to 360 days

Abstract in English:

Meat- and egg-type quails were analyzed to determine reproductive development, puberty and the onset of senility. For this purpose, 288 quails (144 meat- and 144 egg-type quails) were followed from hatch to 360 days old. The birds were distributed by age in 18 groups, 8 quails/group/type of quail. After 35 days quails were maintained under long daylength conditions (17L: 7D). The live weight and the macro and microscopical testicular characteristics were determined in each period. The data were analyzed to determine the growth curve and the behavior of parameters. The model that best fits the data was the non-linear Gompertz model (Y = A exp [-B and (-kt)]). Live weight and testicular morphometric parameters had an allometric behavior among themselves, and at approximately 60 days the males had sexually development, and body weight stabilized at about 100 days. The right testicle was more cranial than the left, differing in the length and width, but no difference in testicular weight (P>0.05) was observed. The meat quails had a larger body weight and testicular weight than the egg quails, but the egg-quail had a higher percentage of testes in relation to body weight. During the whole adult period the males could be considered sexually fit. Breeders had active sex characteristics until 360 days of age, represented by testicular size and by cellular activity in the seminiferous tubules.

Codornas do tipo carne e tipo ovos foram analisadas para determinar o desenvolvimento reprodutivo, a puberdade e o início da senilidade. Para tal, 288 codornas (144 codornas de corte e 144 de postura) foram acompanhadas desde a eclosão até os 360 dias de idade. As aves foram distribuídas por idade em 18 grupos, sendo 8 codornas/grupo/tipo de codorna. Após 35 dias as codornas foram mantidas em condições de fotoperíodo de dias longos (17luz: 7escuro). O peso vivo e os valores morfométricos e histológicos testiculares foram determinados em cada período. Os dados obtidos foram analisados para determinar a curva de crescimento e o comportamento dos parâmetros analisados. O modelo que mais se adequou aos dados foi o modelo não linear de Gompertz (Y=A exp [-B e (-kt)]). O peso vivo e as características testiculares macro e microscópicas apresentaram comportamento alométrico entre si, sendo que, aproximadamente aos 60 dias os machos apresentaram-se sexualmente desenvolvidos, e estabilizaram o peso corporal por volta dos 100 dias. O testículo direito é mais cranial que o esquerdo e diferem em relação a comprimento e largura, porém não foi observada diferenças (P>0,05) para peso testicular. As codornas de corte apresentaram peso corporal e peso testicular maiores que as codornas de postura, porém as codornas de postura apresentaram peso relativo testicular maior. Durante todo o período analisado os machos puderam ser considerados sexualmente aptos. Os reprodutores apresentaram características sexuais ativas até os 360 dias de idade, representadas pelo tamanho testicular e pela atividade celular nos túbulos seminíferos.


#6 - Cortisol influence on testicular testosterone secretion in domestic cat: An in vitro study, 30(10):887-890

E-mail: ggenaro@ffclrp.usp.br The aim of the present experiment was to investigate the effect of corticosteroids (exogen) on in vitro testosterone secretion after stress by transportation (40 minutes). Feline testes (Felis silvestris catus) were incubated in the following media: TCM 199; TCM 199 + hCG 10–7M; TCM 199 + hydrocortisone 10–7M, or TCM 199 + hCG + hydrocortisone. The animals (n=21) were allocated into three groups: (S) that arrived at 3 h prior to surgery, (A) that remained in the laboratory for 36 h before being submitted to surgical procedure, and (C) that were also allowed to remain for 36 hours in the laboratory before the surgical procedure, but whose testes had been incubated with hydrocortisone prior to incubation in the referred media. The results showed that group S secreted higher levels of testosterone, regardless of the culture media. It is noteworthy that the suppressing action of hydrocortisone sodium succinate led to a reduction in the testosterone concentration, despite the presence of hCG.

Testículos foram incubados nos seguintes meios de cultura: TCM 199; TCM 199 + hCG 10–7M; TCM 199 + hydrocortisona 10–7M e TCM 199 + hCG + hidrocortisona. Os animais (n=21) foram alocados em 3 sub-grupos: (S) animal admitido 3 horas antes da orquiectomia, (A) animais orquiectomizados após 36 horas de permanência no biotério e (C) animais que permaneceram por 36 horas no biotério antes da cirurgia e que tiveram seus testículos pré-incubados em hidrocortisona. Os resultados demonstraram que o grupo S secretou maiores valores de testosterona em todas as condições estudadas. É válido mencionar que a supressão promovida pela hidrocortisona também promoveu redução na concentração de testosterona no meio TCM 199, a despeito da presença de hCG.


#7 - Influence of the bipartite scrotum on the testicular and scrotal temperatures in goats, 29(10):797-802

E-mail: machadojunior2@yahoo.com.br O objetivo deste trabalho foi avaliar a influência do grau de bipartição escrotal e do período do ano sobre a termorregulação escroto-testicular em caprinos criados no Estado do Piauí. Foram utilizados 18 reprodutores caprinos machos, divididos em três grupos de seis animais: O Grupo I contendo caprinos com escroto simples, o Grupo II, caprinos com escroto bipartido até 50% do comprimento testicular e o Grupo III, caprinos com bipartição superior a 50% do comprimento testicular. Os parâmetros avaliados foram as temperaturas do escroto, testículo e funículo espermático, obtidas de forma invasiva, com um termômetro digital termoacoplável, e não invasiva, com um pirômetro, nos terços proximal, médio e distal. Os dados foram coletados nos períodos seco (outubro-novembro) e chuvoso (fevereiro-março) do ano, bem como, nos turnos da manhã (6h00 às 7h00) e tarde (14h00 às 15h00). Os resultados foram submetidos à análise de variância (ANOVA) seguida do teste SNK para comparação das médias (p<0,05). O período do ano interferiu na termorregulação escroto-testicular, pois no período seco as temperaturas do escroto, testículo e funículo espermático foram mais elevadas que as observadas no período chuvoso. O grau de bipartição do escroto foi outro fator que modificou a temperatura escroto-testicular, já que os caprinos que apresentaram escroto com maior grau de bipartição demonstraram as menores médias das temperaturas escroto-testiculares em ambos os períodos e turnos avaliados. Conclui-se, portanto, que tanto o período do ano quanto o grau de bipartição do escroto influenciaram o processo de termorregulação escroto-testicular em caprinos.

Eighteen male goats at mating age were accomplished in this study and arranged into three Groups (6 animals each) obeying the classification as goats presenting no scrotal bipartition (Group I), goats showing scrotal bipartition at 50% of the testicular length (Group II), and goats with more than 50% of scrotal bipartition (Group III). The scrotal, testicular and spermatic funiculi temperatures were evaluated invasively with the aid of a digital thermometer and non-invasive with a pyrometer in the proximal, medial and distal portion. The data were acquired during the dry (October-November) and rainy (February-March) period of the year, measured in two shifts: morning (6h00-7h00) and afternoon (14h00-15h00). The results were submitted to variance analysis (ANOVA) following the SNK test for average comparison (p<0.05). The year period interfered on the scrotal-testicular thermal regulation, due to increased temperatures of the scrotal, testicular and spermatic funiculi during the dry period in comparison with the rainy period. The bipartition level was also a factor which contributed to the influence of scrotal-testicular temperature regulation, due to lower average scrotal-testicular temperature rates observed during both periods in the goats with higher levels of scrotal bipartition (>50%). It is possible to conclude that with the experimental conditions applied on this study, the level of scrotal bipartition and the climatic conditions interfere with the scrotal-testicular thermal regulation in goats.

It was found that: (1) at 4 months of age the seminiferous tubules were poorly developed, appeared as seminiferous cords without lumen, lined by a low epithelium, and showed undifferentiated Sertoli cells and scarce interstitial tissue; (2) at 5 months the seminiferous tubules began to differentiate with increase in tubular diameter and lumen, the other tubular structures remaining similar to those previous referred; (3) at 6 and 7 months of age spermatocytogenesis began to appear, Leydig cells were large, polyhedral in shape, with vacuolated cytoplasm and clear nuclei, resting on a sparse interstitial tissue with few blood vessels; (4) 1-year-old cats showed testicular histological features of an adult animal, had seminiferous tubules of large diameter and high seminiferous epithelium with small lumen, and Leydig cells of different sizes, with polyhedral shape, vacuolated cytoplasm, clear nuclei and evident nucleoli resting in a sparse interstitial tissue with some blood vessels; (5) in Group 1 the average diameter of the seminiferous tubules was 160.58µm, and 185.94µm in Group 2; (6) the height of the seminiferous epithelium was 49.51µm for Group 1 and 63.29µm for Group 2; (7) the largest measures of the analyzed parameters were found in animals of Group 2, with functional reproductive organs.
