#1 - Effects of a single trace minerals injection on antioxidant and immune response, resistance to endoparasites, health and growth of newborn Boer kids

Abstract in English:

This study evaluated the effects of injectable trace minerals (ITM) on antioxidant and immune response, resistance to endoparasites, health and growth of newborn Boer kids. Forty-six Boer kids [24 males and 22 females; 3.94±1.03kg of body weight (BW); 6.2±2.4 d of age] were enrolled in the study. Kids were stratified by type of birth (twins or singlet), sex, and BW and assigned to 1 of 2 treatments: one subcutaneous injection (0.1mL/4.5kg of BW) of (1) saline solution or (2) ITM (60, 10, 5, and 15mg/mL of Zn, Mn, Se and Cu, respectively). Blood samples were collected on d 0, 7, 14, 28 and 56. Feces samples were collected on d 56 and BW on d 0, 28 and 56. Kids were checked daily for signs of diarrhea. ITM kids had greater (P<0.01) plasma concentration of superoxide dismutase and tended (P=0.06) to have greater plasma concentration of glutathione peroxidase. ITM kids had greater (P=0.05) concentration of eosinophils, but no differences (P≥0.11) were observed for other hemogram variables. The ITM application did not affect (P≥0.11) the EPG count. However, ITM kids had less (P=0.02) cumulative incidence of diarhea until d 42 (3.85 vs. 25.93±6.8% for ITM vs. Saline kids, respectively) but no differences (P>0.10) were observed after d 42. The ITM application did not affect (P≥0.40) the growth of kids (0.071 vs. 0.065±0.005kg/day for ITM vs. Saline kids, respectively). Thus, the ITM application, increased the plasma concentration of antioxidant enzymes and eosinophils, decreased the incidence of diarrhea only in the middle of the experiment, but did not affected the EPG count and growth of Boer kids.

Este estudo avaliou os efeitos de microminerais injetáveis (ITM) na resposta antioxidante e imune, resistência a endoparasitas, saúde e crescimento de cabritos Boer recém-nascidos. Quarenta e seis cabritos [24 fêmeas e 22 machos; 3,94±1,03kg de peso corporal (PC); 6,2±2,4 dias de idade] foram incluídos no estudo. Os animais foram estratificados por tipo de nascimento (gêmeos ou singular), sexo e peso ao nascimento (PN) e atribuídas a 1 de 2 tratamentos. Uma injeção subcutânea (0,1ml/4,5 de PC de (1) Solução salina ou (2) ITM (60,10,5 e 15mg/ml de Zn, Mn, Se e Cu, respectivamente). As amostras de sangue foram coletadas nos dias 0, 7, 14, 28 e 56. As amostras de fezes foram coletadas no dia 56 e PC nos dias 0, 28 e 56. Os recém-nascidos foram verificados diariamente quanto a sinais de diarreia. Os cabritos ITM apresentaram maior (P<0.01) concentração de superóxido desmutase no plasma e tenderam (P=0,06) a ter maior concentração de glutationa peroxidase no plasma. Os animais ITM apresentaram maior (P=0,05) concentração de eosinófilos, mas não foram observadas diferenças (P≥0.11) para outras variáveis do hemograma. A aplicação de ITM não afetou (P≥0.11) a contagem de EPG. No entanto, os cabritos ITM apresentaram menor incidência cumulativa de diarreia (P=0,02) ate d 42 (3,85 vs. 25,93±6,8% para animais ITM vs. animais salina, respectivamente), mas nenhuma diferença (P>0.10) foi observada após d 42. A aplicação do ITM não afetou (P≥0.40) o crescimento dos animais (0.071 vs. 0.065±0.005kg/dia para ITM vs. Salina, respectivamente). Assim, a aplicação do ITM aumentou a concentração plasmática de enzimas antioxidantes e eosinófilos, diminuiu a incidência de diarreia somente na metade do experimento, mas não afetou a contagem de OPG e crescimento de cabritos Boer recém-nascidos.

E-mail: teresa@colpos.mx This study was conducted to assess the changes in some indicators of oxidative status during pregnancy and lactation in sheep of different parity. Dorset x Suffolk ewes were classified by number of parity: 1, 2 and &#8805;4. They were sampled before pregnancy and on the first, second, third, and fourth months, then on day 143 of pregnancy, as well as on day 5 after birth and after one month of lactation. Antioxidant capacity was found to have two reductions, the first during the second month of pregnancy and the second on day 5 of lactation. Susceptibility to lipid oxidation decreased with an increased number of parturitions. A reduction in lipid oxidation was observed on day 143 of gestation relative to the other samplings during gestation and lactation. Total glutathione peroxidase activity increased when the two reductions in antioxidant capacity took place. Ascorbic acid decreased during lactation and gestation; the lowest values were recorded in the third month of gestation. It is concluded that susceptibility to lipid oxidation decreases with the number of parturitions and that in ewes, during gestation and lactation, there is a mechanism that prevents lipid oxidation involving changes in antioxidant capacity, glutathione peroxidase and ascorbic acid.

E-mail: teresa@colpos.mx Este estudo foi conduzido para avaliar as mudanças em alguns indicadores do estado oxidativo durante a gestação e lactação em ovelhas com diferentes números de partos. Ovelhas Dorset x Sufolk foram classificadas pelo número de partos: 1, 2 e &#8805;4. Amostras foram coletadas antes da prenhez e no primeiro, segundo, terceiro e quarto mês e no dia 143 de gestação, assim como no dia 5 após o parto e com um mês de lactação. Encontrou-se que a capacidade antioxidativa teve duas reduções, a primeira durante o segundo mês de gestação e a segunda no dia 5 de lactação. A atividade total da glutationa peroxidase aumentou quando se deram as reduções de capacidade antioxidante. O ácido ascórbico diminuiu durante a gestação e lactação, com o valor mais baixo foi no terceiro mês de gestação. Conclui-se que a suscetibilidade a oxidação diminui com o número de partos, e que nas ovelhas durante a gestação e lactação há um mecanismo que previne a oxidação lipídica ocasionando mudanças na capacidade antioxidante, e das atividades glutationa peroxidase e ácido ascórbico.


#3 - Expression of P-glycoprotein, multidrug resistance-associated protein, glutathione-S-transferase pi and p53 in canine transmissible venereal tumor, 34(1):71-78

E-mail: daniel.gerardi@ufrgs.br The overexpression of proteins P-glycoprotein (P-gp), multidrug resistance-associated protein (MRP1), mutant p53, and the enzyme glutathione-S-transferase (GSTpi) are related to resistance to chemotherapy in neoplasms. This study evaluated the expression of these markers by immunohistochemistry in two groups of canine TVT, without history of prior chemotherapy (TVT1, n=9) and in TVTs presented unsatisfactory clinical response to vincristine sulfate (TVT2, n=5). The percentage of specimens positively stained for P-gp, MRP1, GSTpi and p53 were, respectively 88.8%, 0%, 44.5% and 22.2% in TVT1 and 80%, 0%, 80% and 0% in TVT2. In TVT1, one specimen presented positive expression for three markers and four specimens for two markers. In TVT2, three specimens expressed P-gp and GSTpi. In conclusion, the canine TVTs studied expressed the four markers evaluated, but just P-gp and GSTpi were significantly expressed, mainly at cytoplasm and cytoplasm and nuclei, respectively, either before chemotherapy as after vincristine sulfate exposure. Future studies are needed to demonstrate the function of these two markers in conferring multidrug resistance (MDR) or predict the response to chemotherapy in canine TVT.

Este estudo avaliou a expressão, por meio da imuno-histoquímica desses marcadores em espécimes de TVT caninos sem histórico de quimioterapia prévia (TVT1, n=9) e em TVT caninos que apresentaram resposta clínica insatisfatória ao sulfato de vincristina (TVT2, n=5). A porcentagem de espécimes positivos para Gp-P, MRP1, GSTpi e p53 foram, respectivamente 88,8%, 0%, 44,5% e 22,2% no grupo TVT1 e 80%, 0%, 80% e 0% no grupo TVT2. No TVT1, um espécime apresentou expressão positiva para três marcadores e quatro para dois marcadores. No TVT2, três espécimes expressaram a Gp-P e GSTpi. Em conclusão, os TVTs caninos estudados expressaram os quatro marcadores avaliados, no entanto apenas a Gp-P e GSTpi foram significativamente expressas, principalmente no citoplasmas e no citoplasma e no núcleo, respectivamente, tanto antes da quimioterapia quanto após à exposição ao sulfato de vincristina. Estudos futuros são necessários para demonstrar a função desses dois marcadores em conferir resistência à multiplas drogas (RMD) ou predizer a resposta a quimioterapia no TVT canino.

E-mail: refarinelli@yahoo.com.br Oxidative stress by physical exercise induces a lipidic peroxidation reaction in cellular membranes and damages in proteins and nucleic acids. Malondialdehyde (MDA) is one of the final products of this reaction. Erythrocytic reduced glutathione (GSH), a multifunctional antioxidant, is present in the plasma and erythrocyts and it represents the total antioxidant suitability of the organism after an oxidative stress. The aim of this study was to evaluate the oxidative stress in different physical exercise conditions through serum MDA and erythrocytic GSH measurement in 45 American Trotter and crossbred horses distributed in three groups: G1 (no physical exercise), G2 (until 6 months of regular physical exercise) and G3 (more than 12 months of regular physical exercise). In the animals with no physical exercise MDA values were lower than the others. There was no difference in GSH values corrected by hemoglobin (Hb) or hematocrit (VG) values among the groups. However, there was a tendency of greater values in G2, where their antioxidant system is adapting to the regular physical exercises and their consequent damages. We conclude that physical exercises induce cellular damage by oxidative stress, but the antioxidant system keeps the homeostasy and it is capable to adapt itself to the free radicals harm.

A glutationa reduzida (GSH), considerada um antioxidante multifuncional, está presente no plasma e principalmente nas hemácias e tem importância pelo fato de ser um dos índices da capacidade total antioxidante do corpo após um estresse oxidativo. Com o objetivo de avaliar o estresse oxidativo em diferentes condições de treinamento físico, determinaram-se a concentração de MDA sérico e GSH eritrocitária em 45 cavalos da raça American Trotter e mestiços divididos em três grupos: G1 (sem treinamento), G2 (até 6 meses de treinamento) e G3 (treinamento há mais de 12 meses). Observou-se que o MDA teve um valor significativamente menor no grupo de animais sem treinamento físico. Não houve diferença estatística significante para GSH corrigida pela Hb e para GSH corrigida pelo VG entre os grupos analisados, mas houve uma aparente tendência a maiores valores no G2, no qual o sistema antioxidante está em fase de adaptação ao treinamento físico constante e suas consequentes injúrias. Conclui-se que a atividade física acarreta danos celulares frente ao estresse oxidativo, mas o sistema antioxidante tem papel fundamental nesta homeostasia observando uma adaptação às injúrias causadas pelos radicais livres.
