#1 - In-vitro production and pre-validation of lyophilized canine platelet-rich plasma for therapeutic use

Abstract in English:

Platelet-rich plasma (PRP) has been considered a promising therapeutic alternative, since platelets are rich in growth factors that are used in the Regenerative Medicine field. However, fresh PRP cannot be stored for long periods. This study aimed to develop a protocol for obtaining lyophilized canine PRP capable of maintaining viability after its reconstitution. For that purpose, canine PRP extraction and lyophilization protocols were initially tested. Subsequently, assays were carried out to quantify the growth factors VEGF and TGF-β, before and after the lyophilization process, gelation test and the three-dimensional gel structure analysis of the reconstituted lyophilized PRP by electron microscopy, as well as in vitro cell proliferation test in lyophilized PRP gel. Additionally, the immunogenicity test was performed, using allogeneic samples of lyophilized PRP. The results showed that the lyophilized PRP had adequate therapeutic concentrations of growth factors VEGF and TGF-β (9.1pg/mL and 6161.6pg/mL, respectively). The reconstituted PRP gel after lyophilization showed an in vitro durability of 10 days. Its electron microscopy structure was similar to that of fresh PRP. In the cell proliferation test, an intense division process was verified in mesenchymal stem cells (MSCs) through the three-dimensional mesh structure of the lyophilized PRP gel. The immunogenicity test showed no evidence of an immune reaction. The findings were promising, suggesting the possibility of having a lyophilized canine PRP that can be marketed. New in vivo and in vitro studies must be carried out for therapeutic confirmation.

O plasma rico em plaquetas (PRP) é uma alternativa terapêutica promissora, pois as plaquetas são ricas em fatores de crescimento com ação na regeneração de tecidos. No entanto, o PRP fresco não pode ser armazenado por longos períodos. Esse trabalho teve como objetivo desenvolver um protocolo de obtenção de PRP liofilizado canino capaz de manter a viabilidade pós reconstituição. Portanto, foram testados diversos protocolos de extração e liofilização. Para validação do PRP canino liofilizado foi analisada a concentração dos fatores de crescimento VEGF e TGF-β antes e após o processo de liofilização, a estrutura tridimensional do PRP liofilizado reconstituído em forma de gel por microscopia eletrônica e seu efeito in vitro na proliferação de células-tronco mesenquimais. Os resultados demonstraram que o PRP liofilizado apresentou concentrações terapêuticas adequadas dos fatores de crescimento VEGF e TGF- β (9,1pg/ml e 6161,6pg/ml, respectivamente). O gel de PRP reconstituído após liofilização apresentou uma durabilidade in vitro de 10 dias, sua estrutura tridimensional mostrou-se semelhante ao PRP fresco e proporcionou intensa proliferação de células-tronco mesenquimais durante o cultivo. O teste de imunogenicidade não demonstrou evidências de reação imune. Os achados foram promissores, sugerindo a possibilidade de uso de PRP canino liofilizado para o mercado. Novos estudos in vivo e in vitro deverão ser conduzidos para comprovação terapêutica.


#2 - Effects of canine adipose-derived mesenchymal stem cells on the epithelialization of rabbits’ skin autograft (Oryctolagus cuniculus)

Abstract in English:

The present study aimed to evaluate the effects of mesenchymal stem cells derived from canine adipose tissue in the healing process of full-thickness mesh skin grafts in rabbits. The stem cells were collected from young dogs; and, after characterization, remained in cryopreservation, in independent doses containing 2 x 106 cells. The mesh distal limb graft technique was performed in 60 rabbits, divided into three groups, CG (Control Group), GT1 (Intralesional Stem Cell Treated Group), and GT2 (Intravenous Stem Cell Treated Group), containing 20 animals each. After grafting, each group was randomly divided into four subgroups according to euthanasia time 3, 7, 14, and 30 days, containing five animals in each group. Animals of GT1_14, GT1_30, and GT2_14, GT2_30 subgroups received a second dose of xenogeneic cells on the seventh day. Meanwhile, animals from GT1_30 and GT2_30 received the third dose of xenogeneic cells on day 14. The groups treated with xenogeneic stem cells positively affected type III collagen re-epithelialization and deposition, and possibly GT1 had a controlled inflammatory response. However, no effect on angiogenesis. Thus, it was possible to demonstrate tolerance and therapeutic action of mesenchymal stem cells from canine adipose tissue in skin grafts in rabbits.

O presente estudo teve como principal objetivo avaliar os efeitos das células-tronco mesenquimais derivadas do tecido adiposo de cães no processo de cicatrização de autoenxertos de pele de espessura total em malha em coelhos. As células-tronco foram coletadas de cães jovens, após a caracterização estas permaneceram em criopreservação, em doses individuais contendo 2 x 106 células. A técnica de enxerto em malha na região distal do membro foi realizada em 60 coelhos, divididos em três grupos, GC (Grupo Controle), GT1 (Grupo tratado com células-tronco intralesional) e GT2 (Grupo tratado com células-tronco via endovenosa), contendo 20 animais cada. Imediatamente após a enxertia, cada grupo foi dividido aleatoriamente em quatro subgrupos, de acordo com o tempo de eutanásia 3, 7, 14 e 30 dias contendo cinco animais cada. Animais dos subgrupos GT1_14, GT1_30 e GT2_14, GT2_30 receberam uma segunda dose de células xenógenas no sétimo dia. Ademais, animais do GT1_30 e do GT2_30 receberam a terceira dose de células xenógenas no dia 14. Os grupos tratados com células-tronco xenógenas tiveram um efeito positivo na reepitelização e deposição de colágeno tipo III, e possivelmente, o GT1 teve uma resposta inflamatória controlada, entretanto o efeito na angiogênese não foi observado. Dessa forma, foi possível demonstrar que houve tolerância e ação terapêutica das células-tronco mesenquimais derivadas do tecido adiposo de cães em enxertos de pele em coelhos.


#3 - Ozonized solutions favor the repair of experimentally induced skin wounds in rats

Abstract in English:

This study aimed to evaluate and compare the effects of ozonized solutions on tissue wound repair in rats. Treatments consisted of ozonized water (GA), 0.9% sodium chloride (GCL), ozonized oil (GO), and 0.2% allantoin cream (GAL). The morphometric evaluation showed that wounds of the GA group presented a higher degree of retraction (p<0.05) at three and eight days of treatment (37.96 and 84.81%, respectively). Picrosirius red staining showed that groups GA and GO presented higher deposition (p<0.05) of type I collagen at 15 and 22 days of treatment, respectively. The neovascularization was higher in wounds of group GO on days 3, 8, and 15 (p<0.05), with higher VEGF immunostaining. (p<0.05). Thus, ozonized water enhances wound retraction and assists in the maturation and remodeling phase, while ozonized oil promotes higher neovascularization during tissue repair and higher deposition of type I collagen from the third week of treatment.

O objetivo deste estudo foi avaliar e comparar os efeitos de soluções ozonizadas sobre o reparo tecidual de feridas em ratos. Foram realizados os tratamentos: água ozonizada (GA), cloreto de sódio 0,9% (GCL), óleo ozonizado (GO) e creme de alantoína 0,2% (GAL). À avaliação morfométrica, as feridas do grupo GA apresentaram maior grau de retração (p<0,05) aos três e oito dias de tratamento (37,96% e 84,81%, respectivamente). A coloração de picrosirius red mostrou que os grupos GA e GO apresentaram maior deposição (p<0,05) de colágeno do tipo I aos 15 e aos 22 dias de tratamento, respectivamente. Já a variável neovascularização foi maior (p<0,05) nas feridas do grupo GO nos dias três, oito e 15, o que fora ratificado à imunoistoquímica, com maior imunomarcação de VEGF nas feridas do grupo GO (p<0,05). Conclui-se que a água ozonizada potencializa a retração da ferida e auxilia na fase de maturação e remodelamento, enquanto o óleo ozonizado promove maior neovascularização durante o reparo tecidual e maior deposição de colágeno do tipo I a partir da terceira semana de tratamento.


#4 - Behaviour of diferent types of chitosan membranes implanted in horses

Abstract in English:

Chitosan has been successfully used as a biomaterial with several purposes in many species. In this study, chitosan membranes were produced with six different types of materials, and their behavior were evaluated upon implantation in the subcutaneous tissue of the flank of twelve healthy horses. We assessed chitosan membranes obtained from commercial chitosan, impregnated or not with silver nanoparticles, sterilized with ethylene oxide (CCEO, n=3; CCSNEO, n=3) or by ultraviolet radiation (CCUR, n=3; CCSNUR, n=3), and chitosan membranes obtained from squid gladius, sterilized with ethylene oxide (SCEO, n=6) or by ultraviolet radiation (SCUR, n=6). The same animals were randomly used in two experimental groups, with a minimum interval of 60 days between procedures, respecting the fact of only one flank side, left or right, be under evaluation by experimental period. After preparation of the membranes and implantation in the flank subcutaneous tissue of the horses, macroscopic and ultrasonographic evaluations of the implant regions were performed, as well as physical examination, blood count and fibrinogen measurement. No clinical or laboratory abnormalities were observed. All animals that received commercial chitosan membranes, regardless of the preparation technique, showed rejection to the biomaterials, considering that 100% of the surgical wounds presented dehiscence of suture and expulsion of the implants. The animals that received squid gladius chitosan membranes showed success in the treatment, with healing by primary intention of the surgical wound. We conclude that squid gladius chitosan membranes are biocompatible and biodegradable when implanted in the subcutaneous tissue of the flank of healthy horses.

A quitosana tem sido utilizada, com sucesso, como biomaterial para diversas espécies e finalidades. Neste estudo foi avaliada a confecção de membranas de quitosana, produzidas a partir de seis tipos de materiais diferentes e foi estudado seu comportamento quando implantadas no tecido subcutâneo do flanco de doze equinos sadios. Foram avaliadas membranas de quitosana obtidas de quitosana comercial, impregnadas ou não com nanopartículas de prata, esterilizadas com óxido de etileno (QCOE, n=3; QCNPOE, n=3) ou por radiação ultravioleta (QCRU, n=3; QCNPRU, n=3) e membranas de quitosana obtidas do gládio de lula, esterilizadas com óxido de etileno (GLOE, n=6) ou por radiação ultravioleta (GLRU, n=6). Os mesmos animais foram utilizados em dois grupos experimentais, de forma aleatória, com um intervalo mínimo de sessenta dias entre os procedimentos, respeitando-se o fato de apenas um lado do flanco, esquerdo ou direito, estar em avaliação por período experimental. Após preparo das membranas e implantação no tecido subcutâneo do flanco dos equinos, foram realizadas avaliações macroscópicas e ultrassonográficas das regiões de implante, além de exames físicos, hemogramas e fibrinogênio. Não foram observadas alterações clínicas e laboratoriais. Todos os animais que receberam membranas de quitosana comercial, independente da técnica de preparo, demonstraram rejeição dos biomateriais, uma vez que 100% das feridas cirúrgicas apresentaram deiscência da sutura e expulsão dos implantes. Os animais que receberam as membranas de quitosana de gladio de lula demonstraram sucesso no tratamento, com cicatrização das feridas cirúrgicas por primeira intenção. Conclui-se que membranas de quitosana de gládio de lula são biocompatíveis e biodegradáveis, quando implantadas no tecido subcutâneo do flanco de equinos sadios.

E-mail: valeriasb@fmvz.unesp.br The aim of the present study was to compare tissue repair of skin defects in rabbits submitted to autologous and homologous grafts treated or not with platelet-rich plasma (PRP). We selected nine rabbits and constituted four groups, designated as G1, in which were performed autologous graft treated with PRP; G2, autologous graft only; G3, homologous graft treated with PRP; and G4, homologous graft only. Macroscopic and histomorphometric evaluation was realized. The histomorphometric evaluation was performed by Hematoxylin/Eosin and Masson´s Trichrome staining with quantification of collagen fibers, macrophages, fibroblasts and vessels. The autologous graft treated with PRP showed positive influence on the early stage of the tissue repair process at the macroscopic evaluation, characterized by rosy color and cosmetic appearance. At the histomorphometric evaluation, there was no statistical difference in the number of macrophages and fibroblasts between the treated grafts or not with the PRP, as well as the quantification of vessels and collagen fibers. It can be concluded that PRP promotes a positive influence on the initial phase or “take” of the graft.

Foram realizadas avaliações macroscópica e histomorfométrica, por meio das colorações de Hematoxilina/Eosina e Tricômio de Masson, incluindo quantificação de fibras colágenas, contagem de macrófagos, fibroblastos e vasos. O uso do enxerto autólogo com PRP influenciou positivamente na fase inicial do processo de reparação tecidual à avaliação macroscópica, caracterizada por coloração rósea e de aspecto cosmético. À avaliação histomorfométrica, não houve diferença estatística quanto ao número de macrófagos e fibroblastos entre os enxertos tratados ou não com o PRP, bem como quanto às contagens de vasos e a quantificação das fibras colágenas. Conclui-se que o PRP sob a promoveu influência positiva na fase inicial ou de “pega” do enxerto autólogo.


#6 - Healing of brain lesions in sheep recovered from amprolium-induced polioencephalomalacia

Abstract in English:

To evaluate the outcome of acute lesions in the brains of sheep that completely clinically recover from acute polioencephalomalacia (PEM), ten sheep were used in this experiment. Eight of those sheep received varying doses of amprolium to induce PEM. Four sheep were treated intramuscularly with 40mg/kg/body weight with thiamine to allow recovery and four sheep were left untreated. Two control sheep did not receive either amprolium or thiamine and were kept along with the other eight sheep for the duration of the experiment. Except for the two drugs, the diet and water source were the same for the ten sheep. Two sheep receiving high daily doses of amprolium and one sheep receiving a lower dose had acute deaths and developed acute brain lesions consisting of neuronal laminar cortical necrosis (red neurons), edema, reactive astrocytes, swollen endothelial cells and gitter cells infiltration. Four sheep that recovered from lower doses of amprolium-induced PEM after being treated with thiamine and another one that recovered spontaneously were euthanatized six months after clinical recovery and had gross changes consisting of segmental absence of cortical tissue. Histologically these segmental cortex-deprived areas corresponded to quasi-empty spaces where only vessels and gitter cells existed. No changes were seen in the brains of the two control sheep.

Para avaliar a evolução das lesões agudas no cérebro de ovinos que se recuperam clinicamente de polioencefalomalacia aguda (PEM), dez ovinos foram usados neste experimento. Oito desses ovinos receberam doses variáveis de amprólio para induzir PEM. Quatro ovinos foram tratados intramuscularmente com 40mg/kg/peso corporal de tiamina para permitir a recuperação, e outros quatro ficaram sem tratamento. Dois ovinos controles não receberam amprólio nem tiamina e foram mantidos com os outros oito ovinos durante a duração do experimento. Exceto pelas duas drogas, a dieta e a fonte de água eram as mesmas para os dez ovinos. Dois ovinos que receberam doses diárias altas de amprólio, e um que recebeu doses menores, tiveram mortes agudas e desenvolveram lesões cerebrais constituídas por necrose neuronal laminar cortical (neurônios vermelhos), edema, tumefação de células endoteliais, astrócitos reativos, tumefação de células endoteliais e infiltração por células gitter. Quatro ovinos que se recuperam da PEM induzida por amprólio, após tratamento com tiamina, e outro que se recuperou espontaneamente, permaneceram clinicamente normais e foram submetidos a eutanásia seis meses após a recuperação clínica. Na necropsia, apresentavam alterações macroscópicas caracterizadas por ausência segmentar de tecido corticocerebral. Histologicamente, essas áreas privadas de tecido cortical consistiam de espaços praticamente vazios onde apenas vasos e células gitter eram vistos. Não foram encontradas alterações no encéfalo das duas ovelhas controle.


#7 - Role of the autologous mesenchymal stem cells compared with platelet rich plasma on cicatrization of cutaneous wounds in diabetic mice, 36(7):617-624

E-mail: argolo_napoleao@ufpi.edu.br Chronic cutaneous lesions affect 15% of diabetic human patients and represent a risk 15 to 46 times larger of limb amputations compared to people with normal glycemia. It is assumed that half of these amputations could be prevented by early treatment of wounds, for example, with proper cell therapy. Objectives: In this study, the action of the autologous transplant of mesenchymal stem-cells (MSC) was evaluated compared to the treatment with autologous platelet rich plasma (PRP) in the cicatrization of cutaneous lesions induced in diabetic mice. These animals were previously treated with streptozootocin to induce diabetes mellitus and round wounds of 1.5cm in diameter were created in the posterior region. Diameters of the wounds and healing time were evaluated during 30 days and the results were submitted to variance analysis and Tukey’s test average. It was noticed that the animals treated with MSC presented a more accelerated cicatrization of the cutaneous lesion than the animals treated with PRP. However, the treatment with PRP presented better results than just the daily asepsis of the lesions with saline or covering them with semi-permeable bandage. Besides, the use of semi-permeable bandage kept the cutaneous lesions of diabetic mice did not interfere negatively with cicatrization, proved to be harmless to use, but kept the cutaneous lesions more hydrated than the ones exposed to the environment.

Objetivos: Neste estudo, a ação do transplante autólogo de células estaminais mesenquimais (MSC) foi avaliada em comparação com o tratamento com plasma rico em plaquetas autólogo (PRP) na cicatrização de lesões cutâneas induzidas em camundongos diabéticos. Estes animais foram previamente tratados com estreptozotocina para induzir diabetes mellitus e feridas redondas de 1,5 cm de diâmetro foram criadas na região posterior. Os diâmetros dos ferimentos e tempo de cicatrização foram avaliados durante 30 dias e os resultados foram submetidos à análise de variância e média pelo teste de Tukey. Verificou-se que os animais tratados com MSC apresentam uma cicatrização mais acelerada da lesão cutânea que do que os animais tratados com PRP. No entanto, o tratamento com PRP apresentou melhores resultados do que apenas a assepsia das lesões diariamente com solução salina ou cobrindo-os com atadura semi-permeável. Além disso, a utilização de atadura semi-permeável mantidas as lesões cutâneas de camundongos diabéticos não interfere negativamente com a cicatrização, provou ser inofensiva para usar, mas manteve as lesões cutâneas hidratadas mais do que os expostos ao meio ambiente.

E-mail: msouza@ufv.br The objective of the study was to evaluate the effect of platelet-rich plasma (PRP) in the treatment of induced tendinitis in the superficial digital flexor tendon (SDFT) of horses through ultrasonographic evaluation. Thus, six 8 to15-year-old healthy gelding horses (=12 years) were used. Tendinitis of the SDFT was provoked in both forelimbs via intratendineous administration of 2.5mg of collagenase (2.5mg.mL-1), what was considered as beginning of the experimental phase. Twelve days after induced tendinitis, the horses were submitted to two treatments: (1) in the lesion caused in the right SDFT (treated group, TG), 2.5mL of PRP activated with calcium chloride at 0.0125mol.L-1, at concentrations from 320,000 to 500,000 platelets.µL-1, were administered; (2) in the tendinitis of the left SDFT (control group, CG), 2.5mL of 0.9% saline solution was administered. After 5 days, the animals were submitted to controlled and progressive physical activity during 30 days. Ultrasonographic examinations were carried out before and after tendinitis induction (48 hours after and on experimental days 7, 12, 14, 21 28, 35 and 42), with tendon cross-sectional area (CSA), lesion cross-sectional area (L-CSA), L-CSA percentage, severity and echogenicity of the lesion, as well as collagen fiber alignment being evaluated. The result revealed reduction (P<0.05) in the L-CSA and in the degree of lesion echogenicity in function of time, but with difference (P<0.05) between groups only for L-CSA, with smaller values in TG. The treatment with PRP promotes greater reduction in the area of the lesion measured by ultrasound.

E-mail: msouza@ufv.br The objective of the study was to evaluate the effect of platelet-rich plasma (PRP) in the treatment of induced tendinitis in the superficial digital flexor tendon (SDFT) of horses through ultrasonographic evaluation. Thus, six 8 to15-year-old healthy gelding horses (=12 years) were used. Tendinitis of the SDFT was provoked in both forelimbs via intratendineous administration of 2.5mg of collagenase (2.5mg.mL-1), what was considered as beginning of the experimental phase. Twelve days after induced tendinitis, the horses were submitted to two treatments: (1) in the lesion caused in the right SDFT (treated group, TG), 2.5mL of PRP activated with calcium chloride at 0.0125mol.L-1, at concentrations from 320,000 to 500,000 platelets.µL-1, were administered; (2) in the tendinitis of the left SDFT (control group, CG), 2.5mL of 0.9% saline solution was administered. After 5 days, the animals were submitted to controlled and progressive physical activity during 30 days. Ultrasonographic examinations were carried out before and after tendinitis induction (48 hours after and on experimental days 7, 12, 14, 21 28, 35 and 42), with tendon cross-sectional area (CSA), lesion cross-sectional area (L-CSA), L-CSA percentage, severity and echogenicity of the lesion, as well as collagen fiber alignment being evaluated. The result revealed reduction (P<0.05) in the L-CSA and in the degree of lesion echogenicity in function of time, but with difference (P<0.05) between groups only for L-CSA, with smaller values in TG. The treatment with PRP promotes greater reduction in the area of the lesion measured by ultrasound.

This oil is used for the treatment of skin wound due to its recognized antiinflammatory and wound healing effects. Despite, its popular use, there are few published data about the therapeutic effect of this medicinal plant. The aim of the study was to evaluate the topic treatment effect of the Copaíba oil on the process of skin repair inflammation induced by a foreign body subcutanously implanted. Sixty BALB/c mice were submitted to a 1cm linear incision and a 12mm circle coverslip was subcutaneously implanted. Four treatments groups were established: control, sterile saline (C); vehicle control, sterile mineral oil, (VC); treatment 1 (T1), mineral oil plus copaiba oil (V/V), and treatment 2 (T2) copaiba oil. The evaluations were performed at pre-determined time points (1, 3, 5, 7 and 14 days). It was possible to find fibroblasts, epithelial cells proliferation, re-epithelization and newly formed blood vessels in all groups, however, all oil treated groups (T1 and T2) did not present re-epithelization at three days post surgical incision. On days 5 and 7, a higher intensity of edema and hyperemia on the groups T1 and T2 was observed, besides that, the T1 and T2 groups presented a serous cellular scab on the wounds that was absent on the C and VC groups. The inflammatory reactions among the groups C and VC showed more mononuclear cells than the T1 and T2 groups that presented a mixed cell patter composed from both mono and polymorphonuclear cells. Although the surgical wounds were re-epithelizaded, in the groups T1 and T2, they were covered by a serous cellular crust and the dermis tissue still presented an intense mononuclear cell inflammatory focus. Fourteen days after of the surgical incision, the gross aspects on groups C and VC were similar and on groups T1 and T2, despite wound to be completely closed and without crusts, the skin those animals was thickened. Furthermore, the dermis on group T2 presented moderate fibrosis, while the other groups presented slightly ones. The results demonstrated that topical treatment with C. langsdorffii oil debilitated the normal process of a wound repair in the presence of a foreign body.
