#1 - Body condition, external morphology, parasitology, and histological and biometrical study of the gastrointestinal tract of Sporophila nigricollis and Sporophila caerulescens seized from trafficking in Northeastern Brazil

Abstract in English:

The objective was to analyze Sporophila nigricollis and Sporophila caerulescens in terms of body conditions, parasitological, bacteriological, external biometric measurements, and histological analysis of the gastrointestinal tract. We used 115 individuals apprehended from 2020 to 2021 and sent to the Wild Animal Triage Center, Paraíba, Brazil. Concerning weight, S. nigricollis, females and males presented 9.36±1.36g and 9.70±1.39g, and S. caerulescens with 10.5±0.70g and 11.75±1.28g, being the second heaviest. The body condition in 69.62% of S. nigricollis and 62.50% of S. caerulescens was good. In the fecal microbiological examination, 40.25% of S. nigricollis and 40% of S. caerulescens were positive for Isospora sp. Dispharynx sp. was reported for the first time in S. nigricollis and S. caerulescens. In fecal bacteriology, 95.65% of S. nigricollis were identified with Gram-positive cocci and 73.91% with Gram-positive bacilli. Forty-one animals died and were placed under refrigeration for 48 hours; of these, 31.16% of S. nigricollis and 37.50% of S. caerulescens presented inadequate body conditions. 19.51% had gastrointestinal tract engorgement with hemorrhagic points, and 14.63% had hepatic alteration due to yellowish coloration. The study contributes with subsidies for the taxonomic elucidation of the genus Sporophila, in addition to the knowledge of the conditions in which birds are found in sorting centers, once the destination of most of the seized animals is the release, thus, a potential source of pathogens to the natural environment.

O objetivo foi analisar Sporophila nigricollis e Sporophila caerulescens quanto às condições corporais, parasitológicas, bacteriológicas, medidas biométricas externas e análise histológica do trato gastrointestinal. Foram utilizados 115 indivíduos apreendidos entre 2020 e 2021 e encaminhados ao Centro de Triagem de Animais Silvestres, Paraíba, Brasil. Quanto ao peso, S. nigricollis, fêmeas e machos apresentaram, 9,36±1,36g e 9,70±1,39g, e S. caerulescens com 10,5±0,70g e 11,75±1,28g, sendo o segundo mais pesado. A condição corporal em 69,62% de S. nigricollis e 62,50% de S. caerulescens foi boa. No exame microbiológico fecal, 40,25% de S. nigricollis e 40% de S. caerulescens foram positivos para Isospora sp. Dispharynx sp. foi relatada pela primeira vez em S. nigricollis e S. caerulescens. Na bacteriologia fecal, 95,65% dos S. nigricollis foram identificados com cocos Gram-positivos e 73,91% com bacilos Gram-positivos. Quarenta e um animais morreram e foram colocados sob refrigeração por 48 horas; destes, 31,16% de S. nigricollis e 37,50% de S. caerulescens apresentaram condições corporais inadequadas. 19,51% apresentaram ingurgitamento do trato gastrointestinal com pontos hemorrágicos e 14,63% alteração hepática devido à coloração amarelada. O estudo contribui com subsídios para a elucidação taxonômica do gênero Sporophila, além do conhecimento das condições em que as aves se encontram nos centros de triagem, uma vez que a maior parte dos animais apreendidos tem como destino a soltura, sendo assim, uma potencial fonte de patógenos para o ambiente natural.


#2 - Occurrence of clinical laminitis after adaptation to confinement: effects on morphology, density, and mineral composition of the hoof of Nellore cattle after finishing

Abstract in English:

Laminitis is a disease that affects the dermis and epidermis of the bovine hoof, generating changes in the hoof capsule. This study evaluated the effects of clinical laminitis diagnosed after the adaptation phase to confinement on the morphology, density, and mineral composition of the hoof of Nellore cattle after finishing. The animals were separated in the first weeks of confinement into a sick group (SG), with clinical laminitis, and a healthy group (HG). SG animals had higher heel length, dorsal wall length, toe height, and diagonal hoof length (p<0.05) than healthy animals. The dermal laminae had similar measurements for thickness, length, and spacing between them between SG and HG. Animals with laminitis showed congestion, hemorrhage, and basement membrane irregularities on histology. Computed microtomography (µCT) revealed that the hoof density of sick animals is lower than healthy ones. According to the mineral composition by energy-dispersive X-ray fluorescence (ED-XRF) spectrometry, the hooves of animals with laminitis (SG) and healthy ones (HG) were not biochemically different. Therefore, the occurrence of clinical laminitis in Nellore cattle in the first weeks of confinement causes an increase in the morphometric parameters of the hoof capsule and a reduction in the density of the abaxial hoof wall evaluated after the finishing period. This disease does not promote changes in the histomorphometric parameters of the dermal laminae and the percentage of minerals in the abaxial hoof wall.

A laminite é uma doença que afeta a derme e epiderme do casco de bovinos gerando alterações no estojo córneo. O estudo avaliou os efeitos da laminite clínica diagnosticada após a fase de adaptação ao confinamento na morfologia, densidade e composição mineral do casco de bovinos da raça Nelore após terminação. Nas primeiras semanas de confinamento, os animais foram separados em um grupo doente (GD) com laminite clínica e em um grupo saudável (GS). Os animais do GD apresentaram maior comprimento de talão, comprimento da parede dorsal, altura da pinça e comprimento diagonal do casco (p<0,05) do que os saudáveis. As lâminas dérmicas tiveram medidas semelhantes para espessura, comprimento e espaçamento entre elas entre GD e GS. Animais doentes apresentaram congestão, hemorragia e irregularidades da membrana basal na histologia. A microtomografia computadorizada (µCT) revelou que a densidade do casco de animais doentes é menor do que o saudável. Para a composição mineral por meio da espectrometria de fluorescência de raio-X por dispersão de energia (ED-XRF), o casco dos animais doentes (GD) e dos saudáveis (GS), não se mostraram diferentes bioquimicamente. Conclui-se que a ocorrência de laminite clínica em bovinos da raça Nelore nas primeiras semanas de confinamento ocasiona aumento de parâmetros morfométricos do estojo córneo e redução da densidade da parede abaxial do casco, avaliados após o período de terminação. Essa enfermidade não promove modificações nos parâmetros histomorfométricos das lâminas dérmicas e na porcentagem de minerais da parede abaxial do casco.


#3 - Anatomopathological aspects and the use of immunohistochemistry in slaughter pigs with cutaneous lesions of erysipelas

Abstract in English:

Swine erysipelas is a disease of worldwide distribution, responsible for causing economic losses in swine and considered an occupational zoonotic disease. It is estimated that 30% to 50% of pigs are carriers and stress can predispose the appearance of clinical disease. The diagnosis of erysipelas in slaughter pigs becomes a challenge for pathologists, since scalding and dehairing, routine procedures in slaughterhouses, generate histological artifacts that often make the final diagnosis impossible. This study describes the anatomopathological aspects and evaluate the use of immunohistochemistry as a diagnostic tool in these cases. Forty-three cases of erysipelas in slaughter pigs were analyzed. Grossly, the cutaneous lesions were characteristic pink, red, or purple raised rhomboid, rectangular or square lesions (“diamond skin”). Histologically, in the dermis and subcutaneous tissue, there were suppurative vasculitis, hidradenitis and folliculitis, as well as degeneration and necrosis of the vessel wall, thrombosis and multifocal areas of necrosis. Suppurative vasculitis and damage to the blood vessel wall were observed in all cases, with varying degrees of severity. The immunohistochemical technique proved to be an effective complementary method of diagnosis, with positive immunostaining in 93%. In most cases, we observed mild immunostaining (57.5%), moderate in 22.5% and marked in 20%.

A erisipela suína é uma doença de distribuição mundial, responsável por causar prejuízos econômicos na suinocultura, além de ser uma doença zoonótica com caráter ocupacional. Estima-se que 30%-50% dos suínos sejam portadores e fatores estressantes podem predispor o aparecimento da doença clínica. O diagnóstico de erisipela em suínos de abate torna-se um desafio aos patologistas, uma vez que os processos de escaldagem e depila, rotineiros em abatedouros frigoríficos, geram artefatos histológicos que muitas vezes impossibilitam o diagnóstico final. Este trabalho descreve os aspectos anatomopatológicos e avalia o uso da imuno-histoquímica como uma ferramenta diagnóstica nestes casos. Foram analisados fragmentos de pele de 43 casos de erisipela em suínos de abate. Macroscopicamente, eram múltiplas lesões cutâneas romboides, retangulares ou quadradas rosa, vermelho ou roxo característicos (“pele de diamante”). Histologicamente, na derme e subcutâneo, havia vasculite, hidradenite e foliculite supurativas, bem como degeneração e necrose da parede de vasos, trombose e áreas de necrose multifocais. A vasculite supurativa e a lesão na parede de vasos sanguíneos foram observadas em todos os casos, em diferentes graus de severidade. A técnica imuno-histoquímica se mostrou um eficaz método complementar de diagnóstico, com imunomarcação positiva em 93%. Na maior parte dos casos observamos marcação discreta (57,5%), moderada em 22,5% e acentuada em 20%.


#4 - Macroscopic and microscopic morphology of the trachea and lungs of giant anteater (Myrmecophaga tridactyla)

Abstract in English:

Giant anteater (Myrmecophaga tridactyla) is a wild mammal distributed in Central and South America; nowadays, it is classified as an endangered species. Research about the macroscopic and histomorphological aspects of its respiratory tract is scarce, and, sometimes, it limits the treatment provided to sick animals and impairs species preservation. Thus, the present study aims to describe the macroscopic and microscopic morphology of its lower respiratory tract, including trachea and lungs. To do so, 12 adult giant anteaters from “Centro de Triagem de Animais Silvestres de Goiânia” (CETAS-GO), Goiás State, Brazil, were used in the research after natural death or euthanasia. Three of these animals were used for macroscopic assessments; they were fixed in 10% buffered formalin and dissected. Trachea and lung tissue samples were collected from nine animals right after death and fixed in 10% buffered formalin for histomorphological analysis; they were processed, embedded in paraffin, and inked with hematoxylin-eosin (HE), periodic acid-Schiff (PAS), and Masson’s trichrome. The macroscopic analysis showed that the trachea in this species is proportionally short and presents from 19 to 27 tracheal cartilages. The right lung presents four lobes and the left one, two. The microscopic analysis evidenced respiratory epithelium of the ciliated cylindrical pseudostratified type, without evident goblet cells in the mucosa layer of the trachea and bronchi. The pulmonary visceral pleura is thick, similar to other large domestic mammals - complete septa extend from the pulmonary visceral pleura. In conclusion, the macroscopy and histomorphology of giant anteater’s lower respiratory tract, represented by trachea and lungs, are similar to that of other domestic and wild mammals. Pulmonary histomorphology is mainly similar to that of pigs and ruminants: it has thick visceral pleura that emits complete septa of conjunctive tissue, which enable lobular parenchymal architecture.

O tamanduá-bandeira (Myrmecophaga tridactyla) é um mamífero silvestre com distribuição na América Central e do Sul e, atualmente, encontra-se classificado como ameaçado de extinção. Pesquisas acerca dos aspectos macroscópicos e histomorfológicos do seu sistema respiratório são escassas, o que, por vezes, limita o tratamento e o manejo de eventuais animais doentes, bem como a preservação da espécie. Desse modo, o objetivo deste estudo foi descrever a morfologia macroscópica e microscópica do aparelho respiratório inferior do tamanduá-bandeira, incluindo traqueia e pulmões. Para tanto, foram utilizados 12 tamanduás-bandeiras adultos, provenientes do Centro de Triagem de Animais Silvestres de Goiânia (CETAS-GO), Goiás, Brasil, após morte natural ou eutanásia. Destes, três foram utilizados para o estudo macroscópico, sendo fixados em formalina tamponada a 10% e dissecados. Para a análise histomorfológica, amostras teciduais da traqueia e do pulmão foram colhidas de nove animais logo após o óbito, fixadas em formalina tamponada a 10%, processadas, incluídas em parafina e coradas com hematoxilina e eosina (HE), ácido periódico-Schiff (PAS) e tricrômico de Masson. À análise macroscópica notou-se que a traqueia é proporcionalmente curta, apresentando 19 a 27 cartilagens traqueais. O pulmão direito apresenta quatro lobos e o esquerdo dois. À análise microscópica foi constatado epitélio respiratório do tipo pseudoestratificado cilíndrico ciliado, sem células caliciformes evidentes na camada mucosa da traqueia e dos brônquios. A pleura visceral pulmonar é espessa, assim como nos grandes mamíferos domésticos, e, a partir desta, estendem-se septos completos. Conclui-se que a macroscopia e a histomorfologia do sistema respiratório inferior do tamanduá-bandeira, representado pela traqueia e pulmões, são semelhantes àquelas de outros mamíferos domésticos e silvestres. A histomorfologia pulmonar é especialmente semelhante à de suínos e ruminantes, com a pleura visceral espessa e emitindo septos completos de tecido conjuntivo, que conferem arquitetura parenquimal lobular.


#5 - Testicular tumors in 190 dogs: clinical, macroscopic and histopathological aspects

Abstract in English:

This study aimed to characterize the prevalence and clinical, macroscopic and histopathological aspects of dogs affected by testicular tumors based on biopsy specimens from the Laboratório de Patologia Veterinária of the Universidade Federal de Santa Maria (LPV-UFSM) over 19 years. Parameters regarding the age, size, and breed of the affected dogs were also established. Of all dogs with some type of neoplasm submitted to histopathological analysis at the LPV over these 19 years (n=1,900), 213 (11.2%) had at least one testicular neoplasm. The tissues of 190 dogs (with 220 neoplasms) were available for histological reassessment. The dogs in this study had different types of testicular tumors with relatively similar frequencies. In descending order, the most frequent testicular neoplasms were seminomas (88/220), Leydig (interstitial) cell tumor (LCT; 64/220), Sertoli cell tumor (SCT; 61/220), and mixed germ cell-sex cord stromal tumor (MGSCT) (07/220). Among the dogs of defined breed (119 cases), large breeds had the largest number of cases (50/119), followed by small (47/119) and medium-sized (22/119) breeds. The ages of dogs affected by testicular tumors ranged from 10 months to 18 years. Increased testicular volume was the most common clinical manifestation. Eleven dogs presented information about clinical signs suggestive of testicular feminization syndrome. In seminomas, the diffuse pattern predominated over the intratubular pattern. Two sites (luminal and basal compartments) suggestive of the onset of neoplastic transformations in germ cells were observed in intratubular seminomas. They corroborate the hypothesis that canine seminomas possibly have pathogenesis similar to that observed in human spermatocytic seminomas. The SCTs and LCTs presented high cell morphology variation. SCTs had neoplastic cells organized in five different histological arrangements. As for LCT, solid-diffuse and cystic-vascular histological patterns were the most commonly observed. Through this study, it was possible to establish some of the leading clinical, macroscopic, and histopathological aspects of testicular neoplasms diagnosed over 19 years in the area covered by the LPV-UFSM.

Este estudo teve por objetivo caracterizar a prevalência, aspectos clínicos, macroscópicos e histopatológicos dos cães acometidos por neoplasmas testiculares, a partir dos espécimes de biópsias do Laboratório de Patologia Veterinária da Universidade Federal de Santa Maria (LPV-UFSM) em 19 anos. Parâmetros quanto à idade, porte, raça dos cães acometidos também foram estabelecidos. De todos os cães com algum tipo de neoplasma submetido à análise histopatológica no LPV nesses 19 anos (n=1.900), 213 (11,2%) tinham ao menos um neoplasma testicular. Os tecidos de 190 cães (com 220 neoplasmas) estavam disponíveis para reavaliação histológica. Os cães deste estudo apresentaram diferentes tipos de neoplasmas testiculares com frequências relativamente semelhantes. Em ordem decrescente, os neoplasmas testiculares mais frequentes foram: seminomas (88/220), leydigomas (64/220), sertoliomas (61/220) e o tumor misto de células germinativas e do estroma do cordão sexual (MGSCT; 07/220). Dentre os cães com raça definida (119 casos), as raças de grande porte tiveram o maior número de casos (50/119), seguido das raças de pequeno (47/119) e médio porte (22/119). As idades dos cães acometidos por neoplasmas testiculares variaram de 10 meses a 18 anos. Aumento de volume testicular foi a manifestação clínica mais comum. Onze cães tinham informações sobre sinais clínicos sugestivos da síndrome da feminilização. Nos seminomas, houve o predomínio do padrão difuso sobre o intratubular. Dois locais (compartimentos luminal e basal) sugestivos de início das transformações neoplásicas nas células germinativas foram observados nos seminomas intratubulares, corroborando com a hipótese de que os seminomas caninos possivelmente tem patogênese semelhante à observada nos seminomas espermatocíticos humanos. Sertoliomas e leydigomas foram neoplasmas com alta variação na morfologia celular. Os sertoliomas tinham células neoplásicas dispostas em cinco arranjos histológicos distintos. Quanto aos leydigomas, os padrões histológicos sólido-difuso e cístico-vascular foram os mais comumente observados. Através deste estudo foi possível estabelecer alguns dos principais aspectos clínicos, macroscópicos e histopatológicos dos neoplasmas testiculares diagnosticados em 19 anos na área de abrangência do LPV-UFSM.


#6 - Cattle rabies: the effect of clinical evolution, viral genetic lineage, and viral load on the severity of histological lesions

Abstract in English:

Our objective was the characterization and staging of histological lesions in different anatomical sites of the central nervous system (CNS) of rabid cattle. The severity of the lesions was compared with the clinical stages of the disease, the variants of viral isolates, and with the load of virus. Thirty-one spontaneously affected rabid cattle the state of Santa Catarina underwent clinical follow-up and were eventually necropsied. CNS tissues were sampled and submitted to direct fluorescent antibody technique (DFAT), immunohistochemistry (IHC), routine histopathology with hematoxylin and eosin stain (HE), reverse transcriptase polymerase chain reaction (RT-PCR), and polymerase chain reaction in quantitative reverse transcriptase in real time (qRT-PCR). Affected cattle were allotted in four groups according to their clinical stage when euthanized: G1, euthanized while standing; G2, euthanized when in sternal recumbence; G3, euthanized when in lateral recumbence; and G4, affected cattle with natural death. In order to evaluate the degree of severity of the lesions and the presence of Negri bodies (NBs), the brain was sectioned at 9 sites. Additionally, spinal cord and trigeminal ganglion sections were examined. The intensity of the lesions was graded as either absent, mild, moderate, or marked, and the presence or absence of the NBs was noted. Histological lesions were characterized by lymphocytic and monocytic meningoencephalitis with NBs in 28 cases. In all analyzed groups, intensities of histological lesions ranging from mild to severe were observed. Brain regions with the highest inflammatory lesion intensity were the medulla at the level of obex, followed by the colliculus and thalamus. NBs were observed in a higher percentage in the cerebellum, followed by medulla at the obex level, striatum complex, and frontal telencephalon. The duration of the clinical course of the disease did not influence the intensity of the inflammatory lesion, but it did influence the presence of NBs, with a higher percentage of these inclusions in cattle that died naturally than in euthanized cattle. All isolated rhabdovirus included in this study were genetically compatible with samples from hematophagous bats Desmodus rotundus. The evaluation by qRT-PCR did not demonstrate a correlation between lesion intensity and the amount of virus.

Nosso objetivo foi a caracterização e estadiamento de lesões histológicas em diferentes locais anatômicos do sistema nervoso central (SNC) de bovinos raivosos. A gravidade das lesões foi comparada com os estágios clínicos da doença, as variantes dos isolados virais e com a quantidade de vírus. Trinta e um bovinos do estado de Santa Catarina, afetados naturalmente por raiva, foram acompanhados clinicalmente e, ao final, necropsiados. Os tecidos do SNC foram amostrados e submetidos a imunofluorescência direta, imunohistoquímica, histopatologia de rotina, reação em cadeia da polimerase via transcriptase reversa (RT-PCR) e reação em cadeia da polimerase em transcriptase reversa quantitativa em tempo real (qRT-PCR). Os bovinos afetados foram distribuídos em quatro grupos, de acordo com sua fase clínica: G1, eutanasiados quando ainda se mantinham em pé; G2, eutanasiados quando em decúbito esternal; G3, eutanasiados quando em decúbito lateral; e G4, bovinos afetados com morte natural. Para avaliar o grau de gravidade das lesões e a presença de corpúsculos de Negri (CNs), o cérebro foi seccionado em 9 locais. Além disso, seções da medula espinhal e do gânglio trigêmeo foram examinadas. A intensidade das lesões foi graduada como ausente, leve, moderada ou acentuada, e a presença ou ausência dos CNs foi anotada. Lesões histológicas foram caracterizadas por meningoencefalite linfocítica e monocítica com CNs em 28 casos. Em todos os grupos analisados foram observadas intensidades de lesões histológicas variando de leve a grave. As regiões cerebrais com maior intensidade de lesão inflamatória foram o bulbo no nível do obex, seguido do colículo e tálamo. CNs foram mais prevalentes no cerebelo, seguido pelo bulbo ao nível do óbex, corpo estriado e telencéfalo frontal. A duração do curso clínico da raiva não influenciou a intensidade da lesão inflamatória, mas influenciou a presença de CNs, com maior porcentagem dessas inclusões em bovinos que morreram naturalmente do que em bovinos sacrificados. Todos os isolados rabdovírus obtidos neste estudo eram geneticamente compatíveis com amostras provenientes de morcegos hematófagos Desmodus rotundus.


#7 - Uterine secretory activity and histology of prepubertal female dogs in an animal model of partial ablation of endometrial glands development

Abstract in English:

Inadequate exposure of the female reproductive system to steroids in uterine developmental periods can partially inhibit the development of endometrial glands in dogs. However, the effects of steroids on the formed glands functionality remain unknown, as well as the possible occurrence of endometrial fibrosis. This study aimed to evaluate the secretory activity of endometrial glands in prebubertal female dogs submitted to a protocol of partial ablation of the uterine adenogenesis. Sixteen females of non-specific breed were distributed into two groups; MPA (n=8), females that received applications of medroxyprogesterone acetate every 3 weeks; and C (n=8) untreated control females. Ovariohysterectomy was performed in all animals at the age of 6 months and evaluated the uterine horns by histological and histochemistry exams. The secretion intensity (degrees 1-4) was evaluated using periodic acid-Schiff (PAS) and alcian blue (AB) pH 2.5. Histological evaluation was performed using Masson’s trichrome and toluidine blue. Only degree 1 and 2 marks for PAS were observed in both groups, with no difference of uterine secretion intensity between the groups regarding the degrees found. However, the MPA group revealed higher intensity of uterine secretion compared to group C (p<0.05). Staining with AB pH 2.5 also revealed only degree 1 and 2 marks in both groups, with no statistically significance between them. Masson’s trichrome staining revealed no marks in the periglandular region in both groups. A higher among of mast cells was observed in the myometrial region of the uterus in both groups. Prepubertal female dogs with partial ablation of the uterine adenogenesis present minimal uterine secretory activity, absence of periglandular fibrosis and increased presence of mast cells in the myometrium compared to endometrium.

A exposição inadequada do sistema reprodutor feminino a esteróides em períodos do desenvolvimento uterino pode inibir parcialmente o desenvolvimento das glândulas endometriais em cães. Entretanto, não se conhece os efeitos dos esteróides sobre a funcionalidade das glândulas formadas, bem como a possível ocorrência de fibrose endometrial. Objetivou-se avaliar a atividade secretória das glândulas endometriais de cadelas pré-púberes submetidas a protocolo de ablação parcial da adenogênese uterina. Foram utilizadas 16 fêmeas, sem-raça-definida, distribuídas nos grupos MPA (n=8), fêmeas que receberam aplicações de acetato de medroxiprogesterona a cada 3 semanas, e C (n=8), fêmeas controle não tratadas. Aos seis meses de idade, foi realizada ovariohisterectomia em todos os animais, e avaliados os cornos uterinos pelo exame histológico e de histoquímica. Para avaliar a intensidade de secreção (graus 1-4), foram utilizadas periodic acid-Schiff e alcian blue (AB) pH 2,5. Para a avaliação histológica foram utilizados tricrômico de Masson e azul de toluidina. Apenas marcações graus 1 e 2 foram observadas para PAS em ambos os grupos, sem diferença na intensidade de secreção uterina entre grupos com relação aos graus encontrados. Entretanto, o grupo MPA apresentou maior intensidade de secreção uterina em relação ao grupo C (p<0,05). Com relação ao AB pH 2,5, em ambos os grupos também foram encontradas apenas marcações de graus 1 e 2, sem diferença estatística entre grupos. Não foram observadas marcações para a coloração de tricrômico de Masson na região periglandular, em ambos os grupos. Foi observada maior quantidade de mastócitos presentes no útero na região do miométrio, em ambos os grupos. Conclui-se que cadelas pré-púberes com ablação parcial da adenogênese uterina apresentam mínima atividade secretória uterina, ausência de fibrose periglandular e maior presença de mastócitos no miométrio em relação ao endométrio.


#8 - Morphological description of ovary and uterus of the nurse shark (Ginglymostoma cirratum) caught off at the Fortaleza coast, Northeast Brazil

Abstract in English:

The nurse shark, Ginglymostoma cirratum (Bonnaterre, 1778) is one of the most studied species of elasmobranchs. However, the knowledge of their reproductive biology is still relatively rare, particularly in the western South Atlantic. This study aimed to describe the morphology of the uterus and the ovary of G. cirratum, based on specimens caught off at the Fortaleza/CE coast, northeast Brazil. Samples were collected from September 2012 to June 2013, from regular landings of artisanal fishing, which commercialize this species freely. A total of ten females were collected. The methodologies followed for analyzing the ovaries and uterus of those females included both macroscopic and histological analysis. G. cirratum has internal type ovary morphology, with invaginations of connective tissue, which defines compartments and separate oocyte groups in ovigerous lots. The epithelium lining the ovary changes from simple columnar ciliated in the area without ovigerous lots, which turns into a simple cubic epithelium in the coating portion of the epigonal organ where ovarian tissue is absent. The uterine mucosa has secretory cells denoted by Alcian Blue staining, indicating the production of mucopolysaccharides, even in immature individuals. This lecithotrophic shark has a uterine vascularized mucosa that is one characteristic of viviparous elasmobranch species.

O tubarão-lixa, Ginglymostoma cirratum (Bonnaterre, 1778) é uma das espécies mais estudadas de elasmobrânquios. O conhecimento de sua biologia reprodutiva, no entanto, ainda é relativamente raro, particularmente no Oeste do Atlântico Sul. Este estudo objetivou a descrição morfológica do útero e ovário de G. cirratum, baseado em espécimes capturados na costa de Fortaleza/CE, no Nordeste do Brasil. Um total de dez fêmeas foi coletado de setembro de 2012 a junho de 2013, nos desembarques regulares da pesca artesanal, que comercializam essa espécie livremente. A metodologia foi de acordo com as analises macroscópica e histológica dos ovários e útero. G. cirratum tem o tipo de morfologia de ovário interno, com invaginações de tecido conjuntivo, que define compartimentos e separa grupos de oócitos em lotes ovígeros. O epitélio que reveste o ovário varia de epitélio simples colunar ciliado na área que não apresenta lotes ovígeros para epitélio simples cúbico na porção de revestimento do órgão epigonal, onde o tecido ovariano é ausente. A mucosa uterina possui células secretoras, evidenciadas pela coloração em Alcian Blue, indicando a produção de mucopolissacarídeos, mesmo em indivíduos imaturos. Este tubarão lecitotrófico apresenta a mucosa uterina vascularizada, sendo esta uma característica de espécies de elasmobrânquios vivíparos.


#9 - Impact of high-energy diets on the rumen environment and digital cushion in confined cattle

Abstract in English:

This study aimed to identify gross and microscopic changes, caused by high-energy diets, in the rumen environment and hoof of confined beef cattle. The study sample comprised 40 confined heifers (Bos taurus) with no disease history divided into four experimental groups using different diets: Group 1 (D1, control), 48:52 forage:concentrate ratio; Group 2 (D2), 30:70 forage:concentrate ratio; Group 3 (D3), 30:70 forage:concentrate ratio + sucrose; Group 4 (D4), 100% concentrate. All animals underwent clinical examination, assessment of ruminal fluid pH and lameness, and sample collection after slaughter for histopathology of the hoof laminae and digital cushion and ruminal tissue. All dependent variables of the study were compared using the SPSS 20.0 statistical software. The variables that did not show normality (HR, RM, ST, and CRT) were compared with application of the Kruskal-Wallis test followed by the Dunnet’s multiple comparison test. All other variables were submitted to analysis of variance (ANOVA) followed by Tukey’s test. The different diets had an impact on the rumen environment (p<0.05) of the heifers assessed, with momentary general depression in the first 12 h after sucrose induction (D3), as well as mild clinical signs in D4. The animals in D3 and D4 presented lower motility (p<0.05) and ruminal pH (p<0.01) than those in D1. Of the 40 heifers, 27.5% (n=11) showed gross lesions in the epithelium of ruminal pillars, whereas 22.5% (n=9) of those in D3 and D4 presented these lesions. Sole corium thickness varied between heifers in D3 compared with those in D1 and D2 (p<0.05). Therefore, high-energy diets, as used in this study, alter some clinical parameters and the rumen environment, causing lesions in the rumen mucosa, and of lesser intensity, in the hoof corium and laminae, suggestive of laminitis.

O objetivo deste trabalho foi identificar alterações macroscópicas e microscópicas provocadas por dietas de alto valor energético no ambiente ruminal e casco de bovinos de corte (Bos taurus) em confinamento. Foram utilizadas 40 novilhas mantidas em confinamento, sendo divididas em 4 grupos experimentais: Grupo D1(controle) - 48% de volumoso e 52% de concentrado; Grupo D2 - 30% de volumoso e 70% de concentrado; Grupo D3 - 30% de volumoso e 70% de concentrado + sacarose; Grupo D4 - 100% de concentrado. Foi realizado exame clínico dos animais, avaliação do pH ruminal, da claudicação, além de coletas de amostras post-mortem para análises histopatológicas da região laminar do casco, coxim digital e do rúmen. Todas as variáveis dependentes do estudo foram comparadas por meio do software estatístico SPSS 20.0. As variáveis sem normalidade (FC, MR, Tc e TPC) foram comparadas pelo teste de Kruskal Wallis, seguido pelo teste de comparação múltipla de Dunnet. Todas demais foram submetidas a análise de variância (ANOVA), seguida pelo teste de Tukey. Foi observado impacto das distintas dietas ao ambiente ruminal (p<0,05). Os grupos D3 e D4 exibiram menor motilidade (p<0,05) e pH ruminal (p<0,01). 27,5% (n=11) dos animais tiveram registros macroscópicos de lesões no epitélio dos pilares ruminais enquanto que os submetidos às dietas 3 e 4 foi observada ocorrência de 22,5% (n=9) de lesões macroscópicas no rúmen. As espessuras do córium solear variaram entre os grupos que receberam a indução de sacarose em relação ao grupo controle e novilhas alimentadas com a dieta 2 (p<0,05). Sendo assim dietas com elevada densidade energética, na condição em que foi empregada, alteraram alguns parâmetros clínicos e o ambiente ruminal, provocando lesões na mucosa do rúmen e de pouca intensidade no córium e tecido laminar, sugestiva de laminite.


#10 - Ultrasonographic, macroscopic and histological characterization of the proximal insertion of the suspensory ligament in Crioulo horses

Abstract in English:

Although ultrasound (US) is a routine diagnostic modality, it still presents limitations for the diagnosis of lesions such as those in the proximal insertion of the suspensory ligament (PISL) because of its composition, which includes muscle fibers and adipose tissue interspersed with the ligament fibers. The objective of the present study was to describe the ultrasonographic, macroscopic and histological aspects of the PISL of thoracic limbs (TL) and pelvic limbs (PL) in Crioulo horses (CH). We selected 34 specimens of TL (right and left) and 10 specimens of PL of horses with a mean age of 5.7 years, from a private clinic or sent to the Department of Veterinary Pathology of UFSM, which died from different causes. The animals had no previous history of lameness in selected limbs associated with PISL injuries. The 34 specimens of PISL of TL were divided into CH (n=25) and Thoroughbred horses (TBH) (n=9), which composed the control group, and 11 specimens of PISL of PL were divided into CH (n=8) and TBH (n=3), which also served as control. The US examination was performed in the PISL using a Sonosite Edge device, 5-10 MHz linear transducer, with cross-sectional and longitudinal palmaromedial and palmarolateral images of the proximal surface of metacarpus III, II and IV (MCIII/MCII/MCIV). In PL, the evaluation was performed four centimeters below the chestnut in the plantaromedial aspect of metatarsus III and II (MTIII/MTII). PISL lobulated shape and size were compared with those of the contralateral limb, as well as the regularity of the palmar bone surface of MC III, II and IV. Subsequently, dissection of the PISL lobes was performed, as well as its macroscopic evaluation, which preceded the histological processing of the samples. In specimens of the CH breed, PISL showed echogenicity varying from peripheral dorsal hyperechogenic zones that merge into echogenic and hypoechogenic zones, where lobulation occurs. In the samples from the TBH group, PISL was also lobulated, but with differences in the echogenicity pattern such as diffuse hypoechogenicity and echogenicity. Macroscopically, CH samples presented a large amount of adipose tissue that corresponds to the dorsal peripheral zone of PISL, which ends in the connective tissue that delimits the ligamentous lobes. On a macroscopic cross-section of PISL, muscle fibers in red are mixed with white ligament fibers in the center of the ligament. This macroscopic finding was not observed in TBH samples, in which muscle fibers overlap ligament fibers throughout the ligament extension and a small amount of fat is present in the dorsal periphery of the ligament. PISL of PL had a triangular shape with echogenicity characteristics very similar to those observed in TL. In ultrasonographic, macroscopic and histological evaluation, PISL samples of TL and PL in CH showed a larger amount of peripheral dorsal adipose tissue, as well as a larger number of merged ligament and muscle fibers compared with those in TBH.

Embora uma modalidade diagnóstica rotineira, a ultrassonografia ainda possui algumas limitações para o diagnóstico de lesões como as que afetam a inserção proximal do ligamento suspensório (IPLS). Uma dessas limitações é relacionada à composição desse ligamento que inclui fibras musculares e tecido adiposo intercalados entre as fibras ligamentares. O objetivo do presente trabalho foi descrever os aspectos ultrassonográficos (US), macroscópicos e histológicos da IPLS de membros torácicos (MT) e membros pélvicos (MP) de equinos da raça crioulo (CC). Foram selecionados 34 espécimes de MT (direito e esquerdo) e 10 espécimes de MP de equinos com idade média de 5,7 anos, que vieram a óbito por diferentes causas, oriundos de uma clínica privada ou destinados ao Laboratório de Patologia Veterinária da UFSM. Não havia histórico prévio de claudicações nos membros selecionados que pudessem estar relacionadas a lesões da IPLS. Os 34 espécimes da IPLS MT foram divididos pela raça CC (n=25) e Puro Sangue Inglês (PSI) (n=9), o qual serviu como grupo controle e, 11 espécimes da IPLS do MP divididos em raça CC (n=8) e PSI (n=3) também como grupo controle. O exame US foi realizado na IPLS com um aparelho Sonosite Edge, transdutor linear de 5-10 MHz, com imagens transversais e longitudinais palmaromedial e palmarolateral da face proximal do metacarpiano (MC) III, II e IV. No MP a avaliação foi realizada quatro centímetros abaixo da castanha no aspecto plantaromedial do metatarsiano III e II (MTIII / MTII). Foram também observadas à forma lobulada da IPLS e o tamanho em comparação ao membro contralateral, bem como a regularidade da superfície óssea palmar do MC III, II, IV. Posteriormente foi realizada a dissecação dos lobos IPLS, bem como a avaliação macroscópica dos mesmos que antecedeu o processamento das amostras para histologia. Em espécimes CC, a IPLS possui uma ecogenicidade que varia de zonas periféricas dorsais hiperecogênicas que se mesclam a zonas ecogênicas e hipoecogênicas onde ocorre a sua lobulação. Nas amostras do grupo PSI, a IPLS também é lobulada, mas com diferenças no padrão de ecogenicidade como, hipoecogenicidade e ecogenicidade difusas. Macroscopicamente, as amostras CC apresentaram uma grande quantidade de tecido adiposo que corresponde à zona periférica dorsal da IPLS, a qual termina no tecido conjuntivo que delimita os lobos ligamentares. Em uma secção transversal macroscópica da IPLS as fibras musculares em vermelho se mesclam as fibras ligamentares brancas no centro do ligamento. Este achado macroscópico não foi observado na raça PSI, onde as fibras musculares intercalam as fibras ligamentares em toda a extensão do ligamento e pequenas quantidades de gordura estão presentes na periferia dorsal do mesmo. A IPLS no MP possui um formato triangular com características de ecogenicidade muito similares as citadas no MT. Na avaliação US, macroscópica e histológica as amostras da IPLS em MT e MP de equinos na raça CC demonstraram uma maior quantidade de tecido adiposo dorsal periférico bem como, uma maior quantidade de fibras musculares e ligamentares mescladas em comparação às amostras PSI.
