#1 - Immunomodulatory effects of dietary β-glucan in silver catfish (Rhamdia quelen)

Abstract in English:

The immunomodulatory effects of dietary β-glucan were evaluated in silver catfish. β-glucan was added to the diet (0.01%, and 0.1%) and fed to the fish for 21 days, to evaluate effects on blood and some innate immune parameter, or fed for 42 days, to evaluate growth rate and resistance to challenge with pathogenic Aeromonas hydrophila. We found that adding β-glucan to the diet had no effect on fish growth and no effect on blood cells, or serum bacterial agglutination and serum myeloperoxidase activity. However, fish that received β-glucan in the diet had the natural hemolytic activity of complement significantly higher compared to control fish. Furthermore, fish fed with β-glucan and challenged with A. hydrophila had fewer bacteria in blood and presented a significantly higher survival rate compared to control fish. Thus, we concluded that β-glucan might be explored as feed additive aiming to improve silver catfish innate immunity and resistance to specific pathogen.

O uso da β-glucana como suplemento alimentar foi avaliado em jundiás. A β-glucana foi adicionada à ração na proporção de 0.01%, e 0.1% e fornecida aos peixes por 21, para avaliar dados hematológicos e parâmetros do sistema imune natural, ou 42 dias, para avaliar ganho de peso e resistência ao desafio com Aeromonas hydrophila. A adição da β-glucana na dieta não afetou o ganho de peso e não induziu alterações hematológicas nem alterações nos níveis de aglutininas e mieloperoxidase sanguínea. No entanto, a atividade hemolítica natural do sistema do complemento foi significativamente maior nos peixes alimentados com β-glucana. Além disso, nos peixes alimentados com β-glucana e desafiados com A. hydrophila, o número de bactérias isoladas do sangue foi significativamente menor, e a sobrevivência ao desafio foi significativamente maior do que nos peixes que não receberam β-glucana. Consequentemente, concluímos que a β-glucana tem potencial imunomodulador quando adicionada à dieta, nas condições experimentais aqui indicadas, e contribui para aumentar imunidade natural e a resistência dos jundiás ao desafio com patógenos específicos.

E-mail: lckreutz@upf.br Knowledge on fish immunoglobulin (Ig) characteristics and the availability of monoclonal or polyclonal antibodies to fish Igs are essential to evaluate the humoral immune response and the Ig distribution on leukocyte cells. We demonstrated that silver catfish serum Ig is composed of one immunodominant H chain with approximately 75k Da and one L chain with approximately 28 kDa, similar to human IgM. Rabbit polyclonal antibodies to the catfish IgM-like Ig recognized both the H and L chain and were useful in developing an indirect ELISA to measure the production of antibodies in fish immunized with bovine serum albumin. Dot blot and western blot cross-reactivity studies indicated a wide degree of epitope sharing amongst Ig from several Siluriformes and Characiformes fish indigenous to Brazilian rivers. In these fish species, polyclonal antibodies reacted mostly with the H chain. The results presented here are central to the development of tools and strategies to investigate the antibody production to inoculated antigens and tissue distribution of Ig molecules in native fish species. Furthermore, because of the wide range of cross-reactivity, polyclonal antibodies to silver catfish IgM-like Ig might be used to develop immunoassays to measure the humoral immune response in other fish species.

Nesse trabalho nós demonstramos que a Ig do soro de jundiás é composta por uma cadeia pesada (H) imunodominante, de aproximadamente 75kDA e de uma cadeia leve (L) de aproximadamente 28 kDa, similar à IgM humana. Anticorpos policlonais produzidos contra a Ig do jundiá reconheceram a cadeia H e L e permitiram o desenvolvimento de um ELISA indireto para mensurar a produção de anticorpos em peixes imunizados com albumina sérica bovina. Estudos de reatividade cruzada, por meio de Dot blot e western blot, indicaram um alto grau de compartilhamento de epitopos entre as Igs de diversos peixes Siluriformes e Caraciformes nativos do Brazil. Nestas espécies de peixes, os anticorpos policlonais reconheceram principalmente a cadeia H. Os resultados deste estudo são fundamentais para o desenvolvimento de ferramentas e estratégias para investigar a produção de anticorpos subsequente à imunização e a distribuição tecidual de Igs em peixes nativos. Além disso, devido ao compartilhamento de epitopos entre as espécies de peixes avaliadas, os anticorpos policlonais anti Ig do jundiá poderão ser usados para desenvolver ensaios imunoenzimáticos para avaliar a resposta imune humoral nestas espécies.


#3 - Celioscopic liver biopsy in silver catfish (Rhamdia quelen), 35(1):19-22

E-mail: mauriciovelosobrun@hotmail.com Endosurgery has been used for assessment of fish celomatic cavity, as well as for obtaining biopsies for organic analysis. Such minimally invasive access may also be used for the analysis of environmental impact on biomarkers of pollution. In Brazil, studies and literature regarding the use of celioscopy in fish are sparse. The purpose of the current study was to develop a two-port celioscopy technique to obtain liver biopsy in silver catfish (Rhamdia quelen). Six adult female silver catfish were used. The animals were anesthetized and the inspection of the celomatic cavity were performed using a telescope and celioscopic-guided liver biopsy were taken using laparoscopic Kelly forceps. On the early postoperative period, the animals were released in a confined water reservoir where mortality could be checked. The liver samples were sent for histological assessment. There were no complications during surgery on early postoperative period. It was possible to visualize meticulously several organs (liver, spleen, stomach, pancreas, swim bladder, ovaries, bowel and transverse septum). In conclusion, the surgical technique and the anesthetic protocol proposed were suitable to perform liver biopsies in silver catfish and provided low morbidity.

Esse acesso, minimamente invasivo, pode ainda ser empregado no estudo de impactos ambientais, a partir do uso desses animais como bioindicadores de poluentes. No Brasil, poucos são os estudos envolvendo a realização de celioscopia de peixes. Este trabalho teve como objetivo, desenvolver uma técnica de videocelioscopia, a partir da utilização de dois portais, na obtenção de biopsia hepática em jundiás (Rhamdia quelen). Para o estudo, foram utilizadas seis fêmeas adultas. Após os animais serem anestesiados, realizou-se a inspeção da cavidade celomática com endoscópio de 10mm e 00 para na sequência, realizar a coleta de tecido hepático com pinça de Kelly. Após a recuperação anestésica/cirúrgica, os animais foram liberados em açude que permitia o controle de possíveis óbitos. Todas as amostras de tecido hepático foram encaminhadas a exame histopatológico. Os procedimentos foram realizados sem a ocorrência de complicações trans ou pós-operatórias. Visualizou-se detalhadamente diferentes órgãos e estruturas intracavitárias (fígado, baço, estômago, pâncreas, bexiga natatória, ovários, intestino grosso e septo transverso. Conclui-se que a técnica cirúrgica e o protocolo anestésico proposto são adequadas para a realização de biópsias hepáticas em jundiás e estão associados a diminuta morbidade.


#4 - Pyrethroid toxicity in silver catfish, Rhamdia quelen, 32(12):1297-1303

E-mail: chicopm28@yahoo.com.br This study aimed to determine both the lethal and sublethal concentrations of Cypermethrin in young Silver Catfish (Brazilian “Jundiá”, Rhamdia quelen) on aquatic environment during 96 hours, as well as to determine the Cypermethrin and Deltamethrin sublethal concentrations during the initial embryonic development period of Rhamdia quelen, and to verify their respective rates of fertilization, hatching and survival. Pyrethroid nowadays is a widely used insecticide, which presents a high toxicity to fish. In order to determine lethal and sublethal concentrations, 120 silver catfish were used; each one had an average weight of 59.58±4.50g and an average size of 20.33±2.34cm. Concentrations used were 0, 1.0, 1.5, 2.0, 2.5, 3.0, 5.0, 10.0, 15.0 and 20.0mg of Cypermethrin per liter of water (mg/L). Fish were exposed to the product in 30-liter fish tanks. In each fish tank there were four fishes and the product was applied three times, i.e., a total of twelve fish were exposed to the product at each application, and a total of 120 fish during the entire experiment (n=120). In order to determine the Cypermethrin and Deltamethrin sublethal concentrations during the initial embryonic development, ovulation induction was performed on female fishes using hormones, and then and egg collection was performed. The eggs were then hydrated and fertilized in Cypermethrin and Deltamethrin in different concentrations: 0.001, 0.01, 0.1, 1.0 and 10.0mg/L of Cypermethrin and 0.001, 0.01, 0.1, 0.5 and 1.0mg/L of Deltamethrin, in addition to the control group (0mg/L). After fertilization, the eggs were kept in containers with the respective pesticides of Cypermethrin and Deltamethrin until hatching, when hatching rate was verified. Then the alevins, from the hatching, were kept on their respective concentrations of Cypermethrin and Deltamethrin so that the survival rate could be analyzed regarding the tested insecticides, during both 12-hour and 24-hour periods. Concerning the sublethal and lethal concentrations, Silver Catfish was sensitive to the tested concentrations of Cypermethrin, showing symptoms of poisoning, such as loss of balance, swimming alteration, dyspnea (they kept their mouths and opercula open), upright swimming and sudden spiral swimming movements. The intensity of such symptoms varied in proportion to the concentration used. The concentrations above 3.0mg/L were considered lethal to the species, since every animal exposed to concentrations between 3.0 and 20.0mg/L had died, while concentrations between 1.0 and 2.5mg/L were considered sublethal. Lethal concentration of Cypermethrin to Silver catfish, in 96 hours, was 1.71 milligram per liter of water. Concerning the sublethal concentration of Cypermethrin and Deltamethrin during the initial embryonic development, the results show that both pyrethroids had significantly decreased the analyzed parameters when comparing them with the control group. It was concluded that, even with the fish being more resistant to pyrethroids in comparison with other species, both the young animals and the ones in stage of embryonic development were susceptible to the effects of these pesticides.

E-mail: chicopm28@yahoo.com.br O presente estudo teve o objetivo de determinar a concentração letal e subletal da cipermetrina por via hídrica em 96 horas em jundiás (Rhamdia quelen) jovens, determinar a concentração subletal da cipermetrina e da deltametrina durante o período inicial do desenvolvimento embrionário do Rhamdia quelen e verificar suas respectivas taxas de fertilização, eclosão e sobrevivência. Os piretróides são inseticidas amplamente utilizados atualmente e apresentam alta toxicidade para peixes. Para determinação da concentração letal e subletal foram utilizados 120 jundiás com peso médio de 59,58±4,50g e comprimento médio de 20,33±2,34cm. As concentrações utilizadas foram 0; 1,0; 1,5; 2,0; 2,5; 3,0; 5,0; 10,0; 15,0 e 20,0mg de cipermetrina por litro de água (mg/L). Os peixes foram expostos ao produto dentro de aquários com capacidade para 30 litros. Em cada aquário foram colocados quatro peixes e cada tratamento teve três repetições, totalizando doze peixes por tratamento. Para determinar a concentração subletal da cipermetrina e deltametrina na fase de desenvolvimento embrionário foi realizada a indução da ovulação em fêmeas de jundiá através de hormônios, com posterior coleta dos ovos. Os ovos foram hidratados e fertilizados em soluções de cipermetrina e deltametrina em diferentes concentrações, sendo estas: 0,001; 0,01; 0,1; 1,0 e 10,0mg/L para cipermetrina e 0,001; 0,01; 0,1; 0,5 e 1,0mg/L para deltametrina, além do grupo controle (0mg/L). Após a fertilização, os ovos permaneceram em recipientes com os respectivos pesticidas até o momento da eclosão onde foi analisada a taxa de eclosão. Em seguida as larvas, a partir da eclosão dos ovos, foram mantidas nas respectivas concentrações de cipermetrina e deltametrina para analisar a taxa de sobrevivência frente aos inseticidas testados, durante 12 e 24 horas. Em relação as concentrações letais e subletais de jundiás jovens, estes apresentaram-se sensíveis à cipermetrina nas concentrações testadas, apresentando sinais de intoxicação como perda de equilíbrio, alteração na natação, dispnéia mantendo a boca e os opérculos abertos, nados verticais e movimentos súbitos de natação em forma de espiral. A intensidade destes sinais variou proporcionalmente a concentração utilizada. As concentrações acima de 3,0mg/L foram consideradas letais para a espécie em 96 horas, pois todos os animais expostos a concentrações entre 3,0 e 20,0mg/L vieram a óbito, enquanto que as concentrações entre 1,0 e 2,5mg/L são subletais. A CL50 96 horas da cipermetrina para o jundiá foi 1,71mg/L. Em relação aos efeitos da cipermetrina e deltametrina sobre a fase de desenvolvimento embrionário, os resultados mostraram que ambos os piretróides diminuíram significativamente os parâmetros analisados em comparação com o grupo controle. Conclui-se que mesmo o jundiá mostrando ser mais resistente aos piretróides em comparação com outras espécies, tanto animais jovens como animais em fase de desenvolvimento embrionário mostraram-se sensíveis aos efeitos destes pesticidas.

E-mail: mateus.costa@univasf.edu.br Aiming the evaluation of sensitivity profiles of pathogen bacteria responsible for fish diseases, 51 bacterial isolates from Jundiá (Rhamdia quelen) belonging to the genus Acinetobacter spp. (8), Aeromonas spp. (15), Edwardsiella spp. (2), Enterobacter spp. (2), Klebsiella spp. (1), Plesiomonas spp. (5), Pseudomonas spp. (1), Staphylococcus spp. (11), and Vibrio spp. (6), were tested against antimicrobial agents used for treatment of bacterial fish diseases. All samples were processed at the Laboratory of Bacteriology, Department of Preventive Veterinary Medicine, UFSM. From 51 bacteria isolated from jundiá fishes (Rhamdia quelen) 51 (100%) were sensitive to gentamycin, 49 (96,08%) to sulphazotrin, 47(92,16%) to chloramphenicol, 43 (84,31%) to tetracylin, 43 (84,31%) to naldixic acid, 31 (60,78%) to nitrofurantoin, 22 (43,14%) to erytromycin, 22 (43,14%) to ampicillin, 15 (29,41%) spiramycin, 13 (25,50%) to cholystin, and 5 (3%) to penicillin G. With exception of an isolate of Staphylococcus spp., the bacteria analyzed in the present study were resistant to one or more antimicrobial agents tested. Knowledge of the sensitivity profile of bacteria involved in infectious processes in fish will allow rational antimicrobial treatment that will contribute to the control of fish diseases in Rhamdia quelen without causing great risks to public health and the environment.
