#1 - Comparison of two methods of tear sampling for protein quantification by Bradford method, 33(2):261-264

E-mail: paulaeye@unb.br The aim of this study was to compare two methods of tear sampling for protein quantification. Tear samples were collected from 29 healthy dogs (58 eyes) using Schirmer tear test (STT) strip and microcapillary tubes. The samples were frozen at -80oC and analyzed by the Bradford method. Results were analyzed by Student’s t test. The average protein concentration and standard deviation from tears collected with microcapillary tube were 4.45mg/mL ±0.35 and 4,52mg/mL ±0.29 for right and left eyes respectively. The average protein concentration and standard deviation from tears collected with Schirmer Tear Test (STT) strip were and 54.5mg/mL ±0.63 and 54.15mg/mL ±0.65 to right and left eyes respectively. Statistically significant differences (p<0.001) were found between the methods. In the conditions in which this study was conducted, the average protein concentration obtained with the Bradford test from tear samples obtained by Schirmer Tear Test (STT) strip showed values higher than those obtained with microcapillary tube. It is important that concentration of tear protein pattern values should be analyzed according the method used to collect tear samples.

E-mail: paulaeye@unb.br Compararam-se dois métodos de coleta de lágrima para quantificação proteica, utilizando-se 58 olhos de 29 cães hígidos. As amostras foram coletadas utilizando-se fitas de teste da lágrima de Schirmer (Schirmer tear test - STT) e tubos microcapilares. Após obtidas, as amostras foram congeladas a -80o C e posteriormente analisadas pelo método de Bradford. Os resultados foram analisados pelo teste T de Student. A média da concentração proteica e desvio padrão das amostras obtidas com microcapilar foi de 4,45mg/mL ±0,35 e 4,52mg/mL ±0,29 para olhos direito e esquerdo, respectivamente. Para as amostras obtidas com STT os resultados foram 4,45mg/mL ±0,35 and 4,52mg/mL ±0,29 para olhos direito e esquerdo, respectivamente. Diferenças estatisticamente significativas (p<0,001) foram encontradas entre os dois métodos. Nas condições em que o trabalho foi conduzido, a média da concentração proteica pelo método de Bradford das amostras obtidas através das tiras do STT foi superior à obtida com o tubo microcapilar. Os valores padrão da concentração protéica da lágrima obtida pelo método de Bradford devem ser analisados considerando-se o método de coleta da lágrima, uma vez que este interfere significativamente nos resultados obtidos.


#2 - Evaluation of tear production with modified Schirmer tear test-1 during the neonatal period in cats, 31(4):350-354

E-mail: jllaus@fcav.unesp.br The study aimed to evaluate tear production by means of modified Schirmer tear tes-1 (mSTT-1) in neonate cats. Likewise, correlation between mSTT-1 and STT-1 was assessed in vitro. Standard SST strips were cut in half and after eye lid opening, tear production of neonates (n=15) was daily measured in both eyes (mSTT-1), until the 7th day, and at day 14, 21, and 28. Animals were daily weighted until 28 days of age. Results were compared statistically (P<0.05). During the first 7 days, the overall mSTT-1 mean was 0.76 wetting/minute. Significant differences between right and left eyes were not observed at any time point (P=1.00). Tear secretion increased significantly, from the 14th to 28th day, in comparison with 7 first days (P<0.05). Positive correlation between maturity parameters and tear secretion was observed (P<0.0001). Distance between slopes of each strip changed significantly (P<0.0001). It was concluded that tear secretion in the neonatal period of cats is very below the reference values described for young and adults of the same species. It is not possible to extrapolate results obtained with mSTT-1 to standard STT-1.

Outrossim, estabelecer in vitro a existência de correlação entre o TLS e o TLS-1m. Tiras pa-dronizadas para realização do TLS foram partidas ao meio e após a abertura das pálpebras, a produção lacrimal dos neonatos (n=15) foi aferida em ambos os olhos (TLS-1m), diariamente, até o sétimo e aos 14, 21 e 28 dias. Os animais foram pesados diariamente até os 28 dias de idade. Os resultados foram comparados estatisticamente (p<0,05). Durante os primeiros sete dias de avaliação, a média geral obtida pelo TLS-1m foi de 0,76±0,08 mm/minuto. Não se constatou diferença significativa entre os olhos direito e esquerdo, em nenhum dos períodos avaliados (p=1,00). A produção lacrimal elevou-se significativamente, do 14º até o 28º dia, comparativamente aos primeiros sete dias (p<0,05). Observou-se correlação positiva entre parâmetros de maturidade e a produção lacrimal (p<0,0001). A distância entre as linhas de cada tira foi significativa (p<0,0001). Conclui-se que a produção lacrimal no período neonatal em gatos é bem inferior aos valores de referência descritos para adultos e jovens da mesma espécie. Não é possível extrapolar valores obtidos com o TLS-1 modificado para o TLS-1 padrão.


#3 - Intraocular pressure and tear secretion in Saanen goats with different ages, 30(9):798-802

E-mail: jllaus@fcav.unesp.br This study aimed to compare the normal intraocular pressure (IOP) and tear secretion, by means of applanation tonometry and the Schirmer tear test-1 (STT-1), in goats of the Saanen breed with different ages, and at different time points. Thirty six goats, free of ocular abnormalities, were grouped into three different age categories (n=12), animals with 45, 180 and 549 days of age. STT-1 and IOP measurements were carried out always at 9:00am and 7:00pm, during three consecutive days. Results were evaluated statistically (P<0.05). Regarding the time of the day, overall IOP values were significantly lower at 7:00 pm (P<0.001) in individuals with 45 days of age; whereas STT-1 values were significantly higher at 7:00pm (P=0.004) in goats with 549 days of age. Considering the sum of three days, both parameters were significantly lower in individuals with 45 days of age (P<0.001). Intraocular pressure and tear secretion values increase until 180 days of age in the Saanen breed of goats.

[Pressão intraocular e produção lacrimal em cabras da raça Saanen com diferentes idades.] Pesquisa Veterinária Brasileira 30(9):798-802. Departamento de Clínica e Cirurgia Veterinária, Universidade Estadual Paulista, Via de acesso Professor Paulo Donato Castellane s/n, Jaboticabal, SP 14870-000, Brazil. E-mail: jllaus@fcav.unesp.br Objetivou-se comparar os valores normais de pressão intraocular (PIO) e de produção lacrimal, através de tonometria de aplanação e do teste da lágrima de Schirmer-1 (TLS-1) em cabras da raça Saanen, com diferentes idades e momentos. Trinta e seis cabras da raça Saanen, livres de afecções oculares, foram agrupadas em três categorias etárias (n=12), compreendendo animais com 45, 180 e 549 dias de idade. O TLS-1 e a PIO foram aferidos sempre às 9:00 e às 19:00, durante três dias consecutivos. Os resultados foram avaliados estatisticamente (p<0,05). Relativamente ao horário do dia, a PIO foi significativamente menor às 19:00 (p<0,001) nos animais com 45 dias de idade; os valores do TLS-1 foram significativamente maiores às 19:00 (p=0,004) nas cabras com 549 dias de idade. Considerando-se a somatória dos três dias, ambos os parâmetros foram significativamente menores nos indivíduos com 45 dias de idade (p<0,001). A pressão intraocular e a produção lacrimal em cabras da raça Saanen se elevam de forma significativa até os 180 dias de idade.
