#1 - An outbreak of fatal Pullorum disease (Salmonella Pullorum) in Guinea fowl keets (Numida meleagris)

Abstract in English:

Pullorum disease is described worldwide and is caused by Salmonella enterica subspecies enterica serovar Gallinarum biovar Pullorum (S. Pullorum). S. Pullorum infection is important in commercial poultry, provoking a systemic disease with high mortality rates. Its occurrence requires notification, and when it is diagnosed in commercial breeding flocks, its eradication is demanded. The aim of this study was to report a severe outbreak of Pullorum disease in young Guinea fowl (Numida meleagris), resulting in 100% mortality of keets (n=290) within the first two weeks of age. All examined keets had enlarged liver, kidneys and spleen (5/5), and the affected tissues were submitted to histological and bacteriological examination. On histopathology, random paratyphoid nodules characterized by areas of necrosis with fibrin and a moderate infiltrate of macrophages and heterophils were observed in the liver. In kidneys, discrete areas of necrosis associated with moderate multifocal infiltrates of lymphocytes, and plasma cells were observed. In the spleen, a moderate infiltrate of macrophages was noticed. Isolation of colonies suggestive of S. Pullorum from liver and spleen was performed in selective agars and, after biochemical tests, confirmed by specific duplex-PCR. The antimicrobial susceptibility test of the isolated strain revealed resistance to only sulfamethoxazole + trimethoprim among the tested antimicrobials. The S. Pullorum isolate recovered in the present study was highly pathogenic to N. meleagris and may represent a risk to other avian species, including industrial poultry.

#2 - Cutaneous fungal infections secondary to avian pox in Northeast Brazil

Abstract in English:

This study describes the epidemiological and clinical-pathological aspects of outbreaks of avian pox diagnosed in poultry associated with fungal co-infections. A retrospective study was carried out, and cases of avian pox with suspected associated fungal infection were selected. The slides were subjected to routine and special histochemical staining in cases of suspected fungal infection, in addition to performing the immunohistochemical technique. Macroscopically, there were nodular lesions with crust formation, and histologically, the lesions were characterized by hyperplasia of the spinous layer associated with multiple eosinophilic intracytoplasmic inclusions. In seven cases, morphologically compatible structures with fungi were verified through black impregnation with GMS and strongly stained in pink with PAS. The morphotintorial aspects were suggestive of fungi belonging to the genus Aspergillus sp. and Candida sp. The diagnosis of fungal co-infections was confirmed through immunohistochemistry, with positive immunostaining for fungi of the genus Aspergillus sp. (five cases) and Candida sp. (two cases). It is concluded that the occurrence of fungal co-infections secondary to the cutaneous lesions of avian pox represents a complicating factor of the disease, favoring the weakness of these animals and death. Therefore, the investigation of associated secondary agents is necessary.

#3 - Type C botulism outbreak in free-ranging waterfowl in Goiás

Abstract in English:

Botulism is generally a fatal disease caused by ingestion of neurotoxins produced by Clostridium botulinum. The present study describes the epidemiological, clinical, and laboratory aspects of a type C botulism outbreak in free-living aquatic birds residing in an urban park in Quirinópolis, Goiás, Brazil. Among a population of approximately 80 waterfowl, a total of 30 birds, including ducks (Cairina moschata), teals (Anas platyrhynchos), and geese (Anser cygnoides), died within 10 days. Of these, six birds showed signs of flaccid paralysis of the pelvic limbs, eyelids, neck, and wings. To confirm the suspicion of botulism, four lake water samples, two samples of the feed consumed by the birds, and samples of serum, intestinal content, stomach content, and liver tissue from two teals that died after presenting clinical signs were analyzed. Using bioassay and neutralization with homologous antitoxin in mice, it was possible to detect the presence of botulinum toxin type C in a water sample and in the intestinal content of one of the necropsied teals. Additionally, the presence of C. botulinum type C was identified in the lake water using polymerase chain reaction. Based on the clinical signs and laboratory results, a diagnosis of botulism caused by botulinum toxin type C was confirmed with probable transmission by lake water.

#4 - Molecular detection of Apicomplexa protozoa in tissues from Alouatta guariba clamitans

Abstract in English:

The brown howler monkey (Alouatta guariba clamitans) is a primate species widely distributed in South America. Infections by protozoa are common in primates. However, studies on protozoa in primates in Brazil are scarce, so the goal of this study was to investigate DNA from the apicomplexan protozoa Neospora caninum, Sarcocystis spp. and Toxoplasma gondii in tissues of A. guariba clamitans. DNA extraction was performed on tissue samples from the heart, brain, liver, spleen, lung and intestine of six A. guariba clamitans from Santa Maria, Central Region of Rio Grande do Sul, Brazil. Conventional PCR was performed using 18S rRNA gene general primers for Apicomplexa and also specific primers to amplify Neospora spp. and Toxoplasma gondii DNA. All animals were positive in the 18S PCR and the genetic sequencing confirmed the presence of Sarcocystis spp. DNA in the tissues of four animals belonging to at least two species (S. neurona and S. gigantea) and T. gondii DNA in the other two animals. One positive sample for T. gondii was genotypically characterized as atypical by the restriction fragment length polymorphism technique. N. caninum DNA was not detected in the tested samples. The presence of Apicomplexa protozoan DNA in the tissues of the six animals tested in this study highlights the importance of howler monkeys as maintainers of these pathogens in nature.

#5 - Detection of avian metapneumovirus subtype A from wild birds in the State of São Paulo, Brazil

Abstract in English:

The present study investigated the circulation of avian metapneumovirus (aMPV) in wild birds in Brazil. To do so, 131 samples from 366 oropharyngeal or cloacal swabs collected from 18 species of birds were tested individually or in pools by RT-PCR. Samples detected by RT-PCR were selected for DNA sequencing. Thirteen (9.9%) samples were detected by the RT-PCR targeting the N gene and four out of 13 samples were sequenced. Sequencing results showed a high identity with the aMPV subtype A. Our results confirm the circulation of the aMPV subtype A in wild birds in Brazil even five years after its last detection.

#6 - Ultrasonography aspects, biometry and Doppler of howler monkey (Alouatta fusca) ocular bulb

Abstract in English:

This study aimed to describe the sonographic features of normal ocular structures, the ocular biometry and Doppler parameters of the internal ophthalmic artery and central retinal artery of the Alouatta fusca. Twenty ocular ultrasonographic examinations were perform in ten primate species of the Alouatta fusca. Proceeded to the sonographic description of the ocular structures and later biometrics was obtained in four distances: (D1) corresponding to the anterior chamber (D2) lens thickness (D3) vitreous chamber and (D4) axial length of the eyeball. Doppler ultrasound evaluated internal ophthalmic artery and central retinal artery as for the resistivity Index (RI), pulsatility index (PI), peak systolic velocity (PSV) and end diastolic velocity (EDV). The values of biometrics and flowmetry underwent comparison test between genders and sonographic views, by Student t test. The anatomical sonographic description of the ocular structures and vasculature of the red howler were similar to species such as dog, cat and man. The biometric average values found were 2.1±0.38mm for D1, 3.7±0.30mm for D2, 10.4±0.78mm for D3 and 19.3±1.64mm for D4. The Doppler values of the ophthalmic artery and central retinal artery found were: PSV of 25.6cm/s and 14.6cm/s; VDF 15.8cm/s and 10.7cm/s; IR 0.7 and 0.5; IP 1.4 and 0.8.

#7 - Outbreak of cutaneous form of avian poxvirus disease in previously pox-vaccinated commercial turkeys

Abstract in English:

This study describes an outbreak of avian poxvirus disease in previously pox-vaccinated turkeys in Brazil. The turkeys had suggestive gross lesions of cutaneous avian poxvirus in the skin of the head and cervical area without changes in the flock mortality rates. In the slaughterhouse, 30 carcasses were removed from the slaughter line to collect tissue from cutaneous lesions for histological analyses and characterization of the virus. The virus was identified by conventional polymerase chain reaction (PCR) and subsequent gene sequencing. Acanthosis, hyperkeratosis, and hydropic degeneration were seen on skin histopathology. Eosinophilic intracytoplasmic inclusion bodies (Bollinger) on keratinocytes were observed in 46.6% of the samples. Avian poxvirus DNA was detected on PCR in 83.3% of the total samples. PCR associated with histopathology had 93.3% of positivity for avian poxvirus. In the phylogenetic study, samples show 100% matching suggesting that the outbreak occurred by a single viral strain and was different from those strains affecting other wild birds such as canaries and sparrows. A single mutation (Adenine for Guanine) was detected in our study’s strain and in the strains of turkey, chickens, and vaccine strains published in GenBank. Also, when the sequence strain of the present study and sequences from GenBank of canarypox and sparrowpox strains were aligned, a Thymine was found replacing the Adenine or Guanine. The in ovo vaccination method as single-use in turkeys of this study apparently did not provide adequate protection against avianpox disease, but additional vaccination administered by wing-web when turkeys were 45-60 days old in the new flocks controlled the disease. In the subsequent year, new cases of this disease were not found. It was not possible to confirm the source of the virus strain, but infection with a field strain derived from chickens is one possibility, considering the poultry farm population in the area and biosecurity aspects. For wide characterization of avipoxvirus and differentiation among strains, the complete sequence of the viral genome is required.

#8 - Evaluation of the biofilm formation capacity of Pasteurella multocida strains isolated from cases of fowl cholera and swine lungs and its relationship with pathogenicity, 37(10):1041-1048

Abstract in English:

ABSTRACT.- Emery B.D., Furian T.Q., Pilatti R.M., Chitolina G.Z., Borges K.A., Salle C.T.P. & Moraes H.L.S. 2017. Evaluation of the biofilm formation capacity of Pasteurella multocida strains isolated from cases of fowl cholera and swine lungs and its relationship with pathogenicity. Pesquisa Veterinária Brasileira 37(10):1041-1048. Centro de Diagnóstico e Pesquisa em Patologia Aviária, Faculdade de Medicina Veterinária, Universidade Federal do Rio Grande do Sul, Av. Bento Gonçalves 8824, Porto Alegre, RS 91540-000, Brazil. E-mail: brunnadeemery@hotmail.com Pasteurella multocida is a Gram-negative bacillus that causes economic losses due to the development of respiratory diseases in several animal species. Among the mechanisms of virulence, the formation of biofilms is an important factor for bacterial survival in hostile environments. Studies of biofilm formation by P. multocida are needed because P. multocida is an important pathogen involved in respiratory infections. However, in contrast to other microorganisms, few studies of biofilm formation have examined P. multocida. Studies comparing the pathogenicity of microbial strains as a function of their biofilm production capacity are also rare. Consequently, the aim of this study was to evaluate the biofilm formation capacity of 94 P. multocida strains isolated from cases of fowl cholera and from swine lungs on polystyrene plates. The associations of the biofilm formation capacity with the pathogenicity index (PI) in vivo and with the presence of four genes (screened by PCR) of the tad locus (tadB, tadD, tadE and tadG), described as adhesion markers, were also determined. Strains from both animal origins were able to form biofilms. However, most of the specimens (52.13%) were classified as weak producers, and more than 40% of the strains of P. multocida (40.42%) did not produce biofilms. There was no significant difference (p>0.05) in the degree of biofilm production between the two sources of isolation. Of the analyzed strains, 56.52% contained all four genes (tadB, tadD, tadE and tadG). The PI arithmetic mean of the strains classified as non-biofilm producers was significantly different (p<0.05) from the PI of moderate-producer strains. The PI of specimens classified as weak biofilm producers also differed significantly (p<0.05) from that of the moderate-producer strains. The results indicate that even though the P. multocida strains isolated from cases of fowl cholera and swine lungs formed biofilms on polystyrene surfaces, adhesion was usually weak. The genes tadB, tadD, tadE and tadG were not significantly associated (p>0.05) with the production of biofilms and with the origin of a given strain. Finally, low virulence strains may suggest a higher biofilm formation capacity on polystyrene plates.

Evaluation of the biofilm formation capacity of Pasteurella multocida strains isolated from cases of fowl cholera and swine lungs and its relationship with pathogenicity.

#10 - Avipoxvirus infecction in Bubo virginianus (great horned owl), 36(7):630-633

Abstract in English:

ABSTRACT.- Echenique J.V.Z., Bandarra P.M., Brauner R.K., Soares M.P., Coimbra M.A.A. & Schild A.L. 2016. [Avipoxvirus infecction in Bubo virginianus (great horned owl).] E-mail: gmpsoares@gmail.com This paper describes a case of mixed infection by pox virus and Aspergillus fumigatus in Bubo virginianus (Owl Jacurutu). An adult male Bubo virginianus was referred to the Núcleo de Reabilitação da Fauna Silvestre, Instituto de Biologia, Universidade Federal de Pelotas (NURFS/CETAS/UFPEL). The owl was active and had a good body condition but with flight disability. After three days of their admission at NURFS the owl developed crusty and verrucous lesions at the dorsal surface of their feet. Also it had nodes on the left eyelid and cera with the same aspect. The owl died 15 days after its arrival. Necropsy and histopatological examination were carried out. The warty lesions had hyperplasia of the epithelium and intracytoplasmic Bollinger-like inclusion bodies in the basal, spinal, granulosa layer and cornea. Viral particles characteristic of pox viruses were shown by electron microscopy. This case includes Bubo virginianus as a host of the avipoxvirus. There were also a mononuclear inflammatory cell infiltrate and bacterial colonies in the dermis. In the lugs, there was congestion and presence of granulomas with intralesional fungal hyphae. With the Grocott stain those structures showed dichotomous branching which was later identified in mycological culture as characteristic for A. fumigates. The diagnosis of avipoxvirus infection can contribute to studies related to the occurrence of this disease in free-living populations and as auxiliary information for the management and conservation of this raptor species. It is also suggested to include the use of X-rays in rehabilitation center protocols as screening test to diagnose aspergillosis in birds of prey with good body condition but inability to fly.

RESUMO.- Echenique J.V.Z., Bandarra P.M., Brauner R.K., Soares M.P., Coimbra M.A.A. & Schild A.L. 2016. [Avipoxvirus infecction in Bubo virginianus (great horned owl).]
