E-mail: luciana.sonne@ufrgs.br A retrospective study has been conducted with the objective of determining the types of bovine neoplasms and their frequency. Necropsy and biopsy cases with diagnosis of neoplasm in cattle submitted to the Setor de Patologia Veterinária of Universidade Federal do Rio Grande do Sul (SPV-UFRGS) from January 2005 to December 2014 were analyzed. A total of 4,188 exams were performed in cattle, and 154 were diagnosed as neoplasm. From these 154 occurrences, 100 cases of neoplasm were the cause of death, 41 were collected from cattle in slaughter house and 13 were biopsies sent by field veterinarians. Neoplasms occurred more frequently in adult cattle (96/154), females (125/154) and the most affected breed was Holstein-Friesian (79/154). The most common neoplasms were lymphoma (73/154), squamous cell carcinoma (28/154), papilloma (9/154), melanoma (6/154) and pheochromocytoma (6/154). The most affected systems were hemopoietic (75/154) and integumentary (25/154). We concluded that lymphoma and squamous cell carcinoma are the most common neoplasms in cattle diagnosed in SPV-UFRGS, Brazil.

Foram analisados os casos de necropsias e biopsias de bovinos com diagnóstico de neoplasia, submetidos ao Setor de Patologia Veterinária da Universidade Federal do Rio Grande do Sul (SPV-UFRGS), entre janeiro de 2005 e dezembro de 2014. De um total de 4.188 exames realizados em bovinos, 154 foram diagnosticados como neoplasmas. Destes, 100 casos foram a causa da morte, 41 foram achados de frigorífico e 13 casos eram biopsias de cirurgias realizadas por veterinários de campo. Os neoplasmas ocorreram com maior frequência em bovinos adultos (96/154), fêmeas (125/154) e a raça mais acometida foi a Holandesa (79/154). Os neoplasmas mais frequentes foram linfoma (73/154), carcinoma de células escamosas (28/154), papiloma (9/154), melanoma (6/154) e feocromocitoma (6/154). Os sistemas mais acometidos foram o hemopoiético (75/154) e tegumentar (25/154). Conclui-se que o linfoma e o carcinoma de células escamosas são os neoplasmas mais frequentes em bovinos diagnosticados no SPV-UFRGS, Brasil.

E-mail: alfieri@uel.br Canine oral papillomavirus (COPV), also known as Canine Papillomavirus type 1 (CPV1), induces papillomas at the mucous membranes of the oral cavity and at the haired skin of dogs. The classification of Papillomavirus (PV) types is based on the L1 capsid protein and nucleotide sequence; so far, 14 CPV types have been described in several countries, but the molecular characterization of CPV in Brazil is lacking. This study investigated the presence of the PV in seven papillomas from four mixed breed dogs from Londrina/PR, Southern Brazil, by partial sequencing of the L1 gene. Seven exophytic cutaneous lesions were surgically removed and processed for histopathological and molecular characterization. Histopathology confirmed the lesions as viral papillomas due to typical histological features. Polymerase Chain Reaction (PCR) assay using the FAP59 and FAP64 primers targeted the L1 gene followed by sequence analysis of the amplicons identified CPV1 in all evaluated papilloma samples. This study represents the first description of CPV1 DNA associated with canine papillomatosis in Brazil.

Atualmente são descritos 14 tipos de CPV, no entanto, ainda faltam estudos moleculares relacionados à identificação dos tipos de CPV no Brasil. O objetivo deste estudo foi investigar a presença de PV em fragmentos de papilomas obtidos de quatro cães sem raça definida, provenientes de Londrina/PR, região sul do Brasil, e definir o tipo viral por meio da análise da sequência parcial de nucleotídeos do gene L1. Sete lesões cutâneas foram cirurgicamente removidas e processadas ​​para a caracterização histopatológica e molecular. O exame histopatológico confirmou as lesões como papilomas. Foi realizada reação em cadeia de polimerase (PCR), utilizando os primers FAP59 FAP64 para a amplificação parcial do gene L1, seguida por análise das sequências dos produtos amplificados, que confirmou a presença do CPV1 em todas as amostras avaliadas. Este estudo representa a primeira identificação do DNA de CPV1 associado com papilomatose canina no Brasil.

E-mail: davetpat@ufrgs.br A retrospective study of 24 cases of papillomas in dogs was performed from January 2001 to March 2011. Additionally, immunohistochemistry (IHC) was used to characterize and evaluate the samples. We found that disease was observed more in mixed breed dogs, ages ranging from 6 months to 10 years (mean 3.1 years), and there was no gender predilection. The main lesion sites were the skin (75%), lips (16.7%), and eyelids (8.3%). Upon histological evaluation, we observed papillary exophytic proliferation of squamous epithelium and papillary endophytic proliferation (inverted) in 87.5% and 12.5% of cases, respectively. The tumors were characterized by spinous layer hyperplasia (87.5%) with koilocytes (70.8%) and intranuclear pale basophilic inclusions bodies (8.3%), prominent granular layer with large amounts of keratohyalin granules (95.8%), and hyperkeratosis in the stratum corneum (100%). Positive immunostaining for Papillomavirus was found in 83.3% of cases, which were distributed between the granular layer and the stratum corneum. These findings indicate the following: that papillomas in dogs are caused by Papillomavirus, the viral cytopathic effect induces epithelial lesions, viral particles are found inside the cell nuclei, and inclusions bodies are rare.

Cães sem raça definida foram os mais afetados, a idade média foi de 3,1 anos, com variação de 6 meses a 10 anos e não houve predileção sexual. Quanto à localização das lesões, 75,0% estavam na pele, 16,7% no lábio e 8,3% em pálpebra. Na avaliação histológica havia proliferação papilar exofítica do epitélio escamoso em 87,5% e papilar endofítica (invertido) em 12,5%. O tumor era caracterizado por hiperplasia do estrato espinhoso (87,5%) com coilócitos (70,8%) e inclusões intranucleares basofílicas pálidas (8,3%); o estrato granular estava proeminente com grande quantidade de grânulos de querato-hialina (95,8%); e havia hiperqueratose do estrato córneo (100%). Na avaliação IHQ para Papillomavirus houve marcação nos estratos granuloso e córneo em 83,3%. Estes achados indicam que os papilomas em cães são causados por Papillomavirus, as lesões epiteliais são decorrentes do efeito citopático viral, as partículas virais estão no núcleo das células e corpúsculos de inclusão são raros.

E-mail:glaukommers@yahoo.com Bovine papillomavirus type-4 (BPV-4) is responsible for the development of papillomas in the upper digestive tract (UDT) of cattle. The necessary steps for the development, growth and regression of papillomas are intimately related with the immunological status of the animal. The consumption of bracken fern (Pteridium aquilinum) has been pointed out as the major factor involved in the persistence of BPV-4 infection of the UDT. The theory involving the relationship between alimentary papillomas and the formation of squamous cell carcinomas (SCCs) suggests the persistence of alimentary papillomatosis through a chronic immunosupressive lymphopenic state induced by bracken fern. The papillomas would serve then as a development site of SCCs through the interaction of BPV-4 oncoproteins and bracken carcinogens. The objective of this study was to evaluate the relationship between lymphopenia and alimentary papillomatosis in spontaneous cases of chronic bracken fern poisoning of cattle. The age, intensity of papillomatosis and the leukon were evaluated of 40 cattle with SCCs of the UDT. Three animals had leukopenia and one had neutrophilia. The mean lymphocyte count (and standart deviation) of mild, moderate and severe papillomatosis was 5,395(±1,696)/mm3, 4,560(±1,561)/mm3 and 5,007(±1,786)/mm3, respectively. There were no statistically significant differences between the intensity of alimentary papillomasis, age and the lymphocyte count. Immunosupression by lymphopenia was a sporadic finding (three cases) in the present study. The results demonstrate that the persistence of alimentary papilomatosis in spontaneous cases of bracken fern-related SCCs of the UDT is not related to lymphopenia and it may be related to other immunological factors.

A ingestão de samambaia (Pteridium aquilinum) tem sido relacionada como o principal fator envolvido na persistência da infecção pelo BPV-4 no TAS. A teoria que estabelece a relação entre papilomatose alimentar e a formação de CCEs sugere a produção de um estado imunossupressivo crônico pela planta, permitindo a persistência dos papilomas no TAS. Os papilomas serviriam então como sítios de desenvolvimento dos CCEs através da interação entre as proteínas do BPV-4 e os carcinógenos da samambaia. O objetivo deste estudo foi avaliar a relação entre a quantidade de linfócitos circulantes e a papilomatose alimentar em casos de intoxicação espontânea crônica por P. aquilinum em bovinos com CCE no TAS. Quarenta bovinos com CCEs no TAS foram avaliados quanto à idade, à intensidade da papilomatose alimentar no TAS e ao leucograma. Três bovinos tinham leucopenia e um apresentava neutrofilia. A média de linfócitos foi de 5.395 (±1.696) na papilomatose leve, 4.560 (±1.561) na moderada e 5.007 (±1.786) na acentuada. Não houve diferença estatisticamente significativa entre o grau de papilomatose, a idade e a quantidade de linfócitos circulantes. Imunossupressão por linfopenia foi um achado esporádico (três casos) neste estudo. Os resultados indicam que a persistência da papilomatose alimentar em casos espontâneos de intoxicação crônica por samambaia em bovinos não tem relação com a quantidade de linfócitos circulantes e que talvez esteja relacionada a outros fatores imunológicos.


#5 - Digital dermatitis of the accessory digits of dairy cows, 30(3):246-248

E-mail: celrodri@fmva.unesp.br Esse estudo caracteriza lesões de dermatite digital (DD) nos dígitos acessórios de vacas leiteiras, além de apresentar a terapia aplicada. Foram utilizados 15 bovinos leiteiros da raça Holandês com DD nos dígitos acessórios dos membros pélvicos. Os animais eram provenientes de quatro fazendas leiteiras com histórico prévio de dermatite digital (DD). Todos os quinze animais foram tratados da mesma forma: após excisão das lesões e sutura das feridas cutâneas, aplicou-se oxitetraciclina pó topicamente sob bandagem e oxitetraciclina (20mg/kg) de longa ação, via intramuscular. Obtiveram-se amostras de tecidos para histopatologia, inclusive por microscopia eletrônica de transmissão (MET). Observou-se cicatrização em todos os animais após 15 dias do procedimento cirúrgico. A maioria das lesões macroscópicas foram projeções papilomatosas ou em forma de verrugas. Os achados histopatológicos de todas as amostras revelaram hiperplasia da epiderme com hiperceratose, inúmeras mitoses no estrato basal, com invasões alongadas em forma de rede na derme superficial e intermediária. A MET evidenciou organismos com formas longas, afiladas e espiraladas, presumivelmente espiroquetas. Tanto as características morfológicas, quanto a resposta à terapia das lesões foram comparáveis às descritas para DD.

E-mail: celrodri@fmva.unesp.br This report characterizes the digital dermatitis (DD) lesions in the accessory digits of dairy cows and presents data on the applied therapy. Fifteen Holstein cattle with DD affecting the accessory digits of the hindlimbs from four dairy farms with previous history of DD were evaluated. Lesions were excised, the wounds were sutured, and a topical application of oxytetracycline powder covered by bandaging was associated with a single parenteral administration of long acting oxytetracycline IM (20mg/kg). Tissue samples were obtained for histopathology and transmission electronic microscopy (TEM). Lesions from all the animals were recuperated 15 days after surgical procedure. Overal, most DD lesions were papillomatous epidermal projections or wartlike verrucous lesions. Histopathologically, samples revealed hyperplasia of epidermis with hyperkeratosis, several mitoses in the stratum basale and elongated rete ridges in the superficial and middle dermis. TEM revealed long, thin spirochete-like bacteria. Morphologic features of lesions and its response to therapy were comparable to those described for DD.

E-mail: rossi.silmara@yahoo.com.br The green turtle Chelonia mydas feeds and nests in the Brazilian coastal area and is considered an endangered species by the World Conservation Union (IUCN 2009) and threatened by the Red List of Brazilian Fauna (Ministério do Meio Ambiente 2009). Fibropapillomatosis is a disease characterized by benign skin tumors (fibropapillomas), and it is one of the main threats to the survival of this species. Studies suggest the involvement of viruses as infectious agents associated with environmental and genetic factors. Blood samples were collected from 45 turtles captured in the coastal area of the state of Sao Paulo, Brazil. From these, 27 were affected by fibropapillomas and 18 were tumor free. Biometrical data on the turtles, size, location and quantity of tumors were recorded. The area occupied by fibropapillomas per animal was calculated and four groups were determined according to severity of the disease or its absence. The objective of the study was to compare hemogram results of the sea turtles classified in these four groups. The lowest hematocrit value was observed in severely affected animals. In the hemoglobin assay, the highest value was observed in the group of tumor free turtles and the lowest, in animals severely affected. Lymphocyte counts and curved carapace length were on the verge of statistical significance.

A fibropapilomatose, doença caracterizada por tumores cutâneos benignos (fibropapilomas), é uma das mais importantes ameaças à sobrevivência dessa espécie. Pesquisas sugerem o envolvimento de agentes infecciosos virais em associação com fatores ambientais e genéticos. Foram colhidas amostras sanguíneas de 45 tartarugas provenientes do litoral do estado de São Paulo, Brasil, sendo 18 sem fibropapilomas e 27 acometidas. Dados de biometria das tartarugas, quantidade, localização e tamanho dos tumores foram anotados. Foi realizado o cálculo da área de fibropapilomas por animal e foram estipulados 4 grupos de acordo com o grau de acometimento e sua ausência. O objetivo foi realizar uma comparação entre os hemogramas das tartarugas marinhas classificadas nos 4 grupos. Animais de grau grave apresentaram o menor valor para hematócrito. Para dosagem de hemoglobina, observou-se que o maior valor foi para o grupo de tartarugas sem fibropapilomas e o menor para o de grau grave. Os valores de linfócitos e comprimento curvilíneo da carapaça beiraram a significância estatística.

The entire viral genome was sequenced in 2007. Additionally, a variant of BPV-8, BPV-8-EB, was also identified from papillomatous lesions of a European bison in Slovakia. In Brazil, despite the relatively common occurrence of BPV infections, the identification and determination of viral types present in cattle is still sporadic. The aim of this study is to report the occurrence of the recently described BPV-8 in Brazil. The virus was identified in a skin warts obtained from a beef cattle herd located in Parana state, southern Brazil. The papilloma had a macular, non-verrucous gross aspect and was located on the dorsal thorax of a cow. Polymerase chain reaction (PCR) was performed using generic primers for partial amplification of L1 gene. The obtained amplicon (480bp) was cloned and two selected clones were sequenced. The nucleotide sequence was compared to existing papillomaviral genomic sequences, identifying the virus as BPV type 8. This study represents the first report of BPV-8 occurrence in Brazil, what suggests its presence among Brazilian cattle.

Aural plaques occurred in 57% of Mangalarga and in 35% of Quarter breeds. Clinically the lesions consisted of flat, desquamated and hypochromic plaques formed by coalescence of small papules. The main histopathological findings were epidermal hyperplasia and hypomelanosis with abrupt change between the normal and the affected epithelium.

In all papillomas examined, a product with the expected molecular size was amplified. Phylogenetic analysis of the PCR products identified BPV-2 in three samples, BPV-1 in one, and BPV-6 in five papillomas. BPV-6 was detected in cutaneous papillomas of the teat and in other body parts as well. In one animal, from which more than one sample was collected, a concomitant infection by BPV-1 and BPV-2 was identified. The five positive BPV-6 samples showed a nucleotide identity of 100% with the sequence of the reference strain available in GenBank. However, differences among BPV-2 and BPV-1 Brazilian samples and the respective reference sequences deposited in GenBank were observed. Molecular comparison of the two BPV-2 strains identified showed the involvement of two viral variants. This study revealed the diversity of BPV types circulating in the state of Paraná.
