#1 - Serological survey of bluetongue virus in sheep from Minas Gerais

Abstract in English:

Bluetongue is an infectious, non-contagious disease that affects domestic and wild ruminants, caused by a virus from the Orbivirus genus, Reoviridae family, transmitted by arthropod vectors of the Culicoides genus. This paper aims to be the first serological survey of bluetongue in sheep from the Meso-regions of Campo das Vertentes and South and Southeast of Minas Gerais. Samples were collected from sheep from different properties. The serum samples were submitted to Agar Gel Immunodiffusion (AGID) and competitive Enzyme-Linked Immunosorbent Assay (cELISA). 303 serum samples were submitted to AGID and cELISA. In these samples, 164 (54.13%) were positive in the AGID technique, and 171 (56.44%) positive in the cELISA technique, with an almost perfect agreement between the techniques (kappa index = 0.887). In all visited properties, positive animals have been found in the herd. Animals acquired from properties of the studied mesoregions were more likely to be positive in IDGA and cELISA tests than animals acquired from properties in other regions of Brazil (p<0.001). These results suggest that bluetongue virus (BTV) is widespread in the mesoregions of Campo das Vertentes and South and Southeast of Minas Gerais.

A língua azul (LA) é uma doença infecciosa, não contagiosa, que acomete ruminantes domésticos e silvestres, causada por um vírus do gênero Orbivirus da família Reoviridae, transmitida por vetores artrópodes do gênero Culicoides. O presente estudo representa o primeiro trabalho a realizar um inquérito sorológico da língua azul em rebanhos ovinos nas Mesorregiões de Campo das Vertentes e Sul e Sudoeste de Minas Gerais. Foram coletadas amostras de soro de ovinos de diferentes propriedades. As amostras de soro foram submetidas aos testes de imunodifusão em gel de ágar (IDGA) e ensaio de imunoadsorção enzimática por competição (cELISA). Ao todo 303 amostras de soro foram submetidas ao IDGA e cELISA. Dessas amostras, 164 (54,13%) foram positivas na técnica de IDGA e 171 (56,44%) positivas na técnica de cELISA, havendo concordância quase perfeita entre as técnicas (índice kappa = 0,887). Em todas as propriedades visitadas, foram encontrados animais positivos no rebanho. Animais adquiridos de propriedades das Mesorregiões estudadas, tiveram mais chances de serem positivos nos testes de IDGA e cELISA do que animais adquiridos de propriedades de outras Regiões do Brasil (p<0,001). Esses resultados sugerem que o vírus da língua azul encontra-se disseminado em ovinos nas Mesorregiões de Campo das Vertentes e Sul e Sudoeste de Minas Gerais.


#2 - Morphometry of the mandibular foramen applied to local anesthesia in hoary fox (Lycalopex vetulus)

Abstract in English:

Great part of the veterinary care in wild carnivores is intended to treat the dental disorders, and oral cavity disorders may generally affect the animal health as a whole. It is factual that knowing the location of the mandibular foramen is vital for local anesthetic block of the inferior alveolar nerve, however, there is still no data on the morphometry of the hoary fox mandibular foramen. The aim was describing morphometry of the mandibular foramen of this species and associating its position with anatomic reference points in the mandible, thus providing support for more effective local anesthetic block of the inferior alveolar nerve in such species. Four adult jaws of Lycalopex vetulus were used. Radiography and biometrics of the hemimandibulas were performed. The rostral third of the jaw body in a lateral view presented three mental foramens, being a rostral, a medium and a caudal. Each hemimandibula presented I3/C1/PM4/M3. The angle of the mandible was marked by the masseteric fossa, the angular incisure, the angular process and the mandibular foramen, and this last one located perpendicularly to the dorsal edge of the angular incisures in medial view. In this view, the crevice to the milohyoid nerve, projected in the caudodorso rostral direction, was also evidenced. The ramus of the mandible was characterized by the presence of the condylar and coronoid processes, and by the dorsal and ventral mandibular incisions. Statistical analyzes did not present significant differences between the antimeres of the studied animals, and the penetration of the needle perpendicularly to the dorsal end of the angular incision on average 8.79mm, overlapped to the medial face of the angle of the mandible could be indicated. Alternatively, the access may also be achieved by inserting the needle in an average of 17.69mm perpendicular to the dorsal end of the angular process, in contact with the medial aspect of the angle of the mandible, and in caudo-rostral projection, also allowing a better anesthetic blockade of the inferior alveolar nerve in L. vetulus. It can also be concluded that the masseteric fossa, the dorsal and ventral mandibular angles, the crevice to the milohyoid nerve, the ramus of the mandible and the mandibular foramen presented differences in their topographic descriptions when compared to the other canids.

A maior parte dos atendimentos veterinários em carnívoros silvestres destina-se ao tratamento das afecções dentárias, sendo que doenças de cavidade oral podem afetar a saúde do animal como um todo. É factível que o conhecimento da localização do forame mandibular é essencial para o bloqueio anestésico local do nervo alveolar inferior, entretanto, ainda são inexistentes dados sobre a morfometria do forame mandibular de raposa-do-campo. Objetivou-se descrever a morfometria do forame mandibular desta espécie, e correlacionar sua posição com pontos de referência anatômica na mandíbula, oferecendo subsídio para um bloqueio anestésico local mais efetivo do nervo alveolar inferior nesta espécie. Foram utilizadas quatro mandíbulas de cadáveres adultos de Lycalopex vetulus. Realizou-se a radiografia e as biometrias das hemimandíbulas. O terço rostral do corpo da mandíbula em uma vista lateral apresentou três forames mentuais, sendo um rostral, um médio e um caudal. Cada hemimandíbula apresentou I3/C1/PM4/M3. O ângulo da mandíbula foi marcado pela fossa massetérica, pela incisura angular, pelo processo angular e pelo forame mandibular, e este último localizado perpendicularmente ao extremo dorsal da incisura angular em vista medial. Nesta vista, também se evidenciou o sulco para o nervo milohióideo, projetado em sentido caudodorsorostral. O ramo da mandíbula foi caracterizado pela presença dos processos condilar e coronóide, e pelas incisuras mandibulares dorsal e ventral. As análises estatísticas não apresentaram diferenças significantes entre os antímeros dos animais estudados, podendo-se indicar a penetração da agulha perpendicularmente ao extremo dorsal da incisura angular em média 8,79mm, justaposto à face medial do ângulo da mandíbula. Alternativamente, o acesso também poderá ser realizado introduzindo a agulha em média 17,69mm de forma perpendicular ao extremo dorsal do processo angular, em contato com a face medial do ângulo da mandíbula, e em projeção caudorostral, permitindo também um melhor bloqueio anestésico do nervo alveolar inferior na L. vetulus. Também se pode concluir que a fossa massetérica, as incisuras angular, mandibulares dorsal e ventral, o sulco para o nervo milohióideo, o ramo da mandíbula e o forame mandibular apresentaram diferenças em suas descrições topográficas quando comparados aos demais canídeos.


#3 - Evaluation of tear production in juvenile opossum using three different methods

Abstract in English:

The establishment of parameters for tear production in different species is important for better understanding eye´s health and is one of the components of the ophthalmic semiological technique. Particularities derived from the anatomophysiology of non-domestic species induce the search for more reliable methodologies. The aim was to evaluate and compare tear production of white-eared opossum (Didelphis albiventris) and Brazilian common opossum (Didelphis aurita) by three different methods. Fifteen individuals of each species, juveniles, healthy, of both sexes, with 60 to 90 days of life, were physically restrained. Phenol red thread test (PRTT), endodontic absorbent paper point tear test (EAPPTT) and modified -Schirmer tear test (mSTT) were performed. PRTT was the most difficult to perform because of the wire malleability, while EAPPTT was more feasible for both species. The median ± semi-quartile range for PRTT were 19.79±2.61mm/15 “and 5.22±2.92mm/15”, for EAPPTT were 16.25±1.82mm/min and 10.9±3.04mm/min, and for STTm were 0±1.63mm/min and 0±1.63mm/min for white-eared opossum and Brazilian common opossum respectively. There was no difference between the right and left eye neither sex. A significant difference was obtained for the same test to different species. No significant correlation was found between the tests for both species. The description of tear production parameters for juvenile white-eared opossum and Brazilian common opossum may be used as a tool, which will allow the early diagnosis of ocular diseases.

O estabelecimento do parâmetro de produção lacrimal nas diferentes espécies é importante para o entendimento da saúde do olho e é um dos componentes da semiotécnica oftálmica. Particularidades derivadas da anatomofisiologia das espécies não domésticas induzem a busca de metodologias que sejam mais fidedignas aos parâmetros. Objetivou-se com este estudo avaliar e comparar a produção lacrimal de gambás-de-orelha-branca (Didelphis albiventris) e gambás-de-orelha-preta (Didelphis aurita) por três diferentes métodos. Quinze indivíduos de cada espécie, juvenis, hígidos, de ambos os sexos, com 60 a 90 dias de vida, foram contidos fisicamente para realização do teste lacrimal do vermelho de fenol (TLVF), da ponta de papel absorvente estéril e do teste lacrimal de Schirmer modificado (TLSm). O TLVF foi o mais difícil de ser executado devido à maleabilidade do fio, enquanto a TEPA se mostrou mais exequível para ambas as espécies. A mediana ± intervalo semi-interquartil para o TLVF foi de 19,79±2,61mm/15” e 5,22±2,92mm/15”, para a TEPA foram de 16,25±1,82mm/min e 10,93±3,04mm/min, e para o TLSm foram de 0±1,63mm/min e 0±1,63mm/min, para gambás-de-orelha-branca e gambás-de-orelha-preta, respectivamente. Não houve diferença entre o olho direito e esquerdo e nem quanto ao sexo. Obteve-se diferença significativa para um mesmo teste entre as espécies. Não foi encontrada correlação significativa entre os testes para ambas as espécies. A quantificação da porção aquosa da lágrima poderá auxiliar no diagnóstico precoce de doenças oculares nas espécies estudadas.


#4 - Closed suction drain in total unilateral mastectomy in bitches

Abstract in English:

The aim of this study was to evaluate the use of closed suction drain in surgical wounds of total unilateral mastectomy in dogs. Eighteen dogs were used with an average weight of 14.87±9.29kg, average age 9.34±2.68 years, from the hospital routine, which were selected through clinical and oncological evaluation. The animals were divided into two groups: Conventional (CG, n=9) which underwent total unilateral mastectomy surgery without drain and Drain group (DG, n=9) which underwent complete unilateral mastectomy with placement closed suction drain (Biovácuo). The diameter of the drain was determined according to the animal weight. Five evaluations were performed during the postoperative period: clinical evaluation of the animals daily through the heart rate (HR), respiratory rate (RR), rectal temperature (RT), color of mucous membranes, capillary refill time (CRT), degree of dehydration, blood count, chemistry profile (urea, creatinine, cholesterol, triglycerides and glucose); qualitative assessment of the drain; qualitative assessment of the surgical wound, is also being held on 14 and 30 days for the presence or absence of exudate, edema, skin integrity, dehiscence, necrosis and scarring; quantitative evaluation of the drained fluid; qualitative assessment of fluid drained, as the cellularity type and cell morphology. Surgical time was recorded in both groups. There were an increase in average values of density, neutrophils, lymphocytes, and increased fibrinogen median value of drained fluid in 72 hours compared to 24 hours in DG group. There was an increase of exudate within 24, 48 and 72 hours in DG when compared to GC. There was an increase in the volume of drained fluid in 60 and 72 hours compared to 12 hours in DG. There was a reduction in the average value of red blood cells within 72 hours from baseline in DG group. There was an increase in average hemoglobin and hematocrit in 24 hours in the control group compared to the DG. There was an increase in the average value of neutrophils targeted at baseline in DG when compared to CG. It follows that, the closed suction drain placement technique presents more exudate. It is recommended that the drain vacuum is redone 12 in 12 hours, initially being performed 6 hours after surgery. The closed suction drain does not generate additional discomfort to the patient; it is recommended that it be removed 72 hours after the surgery, when the fluid production decreases, but even so some individuals may present seroma formation in the late postoperative.

O objetivo deste estudo foi avaliar a utilização de dreno de sucção fechado em feridas cirúrgicas de mastectomias unilaterais totais em cadelas. Foram utilizadas 18 cadelas com peso médio de 14,87±9,29kg, idade média 9,34±2,68 anos, provenientes da rotina hospitalar, as quais foram selecionadas por meio de avaliação clínica e oncológica. Os animais foram alocados em dois grupos: Convencional (GC, n=9) que foram submetidas à cirurgia de mastectomia unilateral total, sem colocação de dreno e grupo Dreno (GD, n=9) que foram submetidas à mastectomia unilateral total com colocação do dreno de sucção fechado (Biovácuo), sendo o diâmetro do dreno determinado conforme peso do animal. Foram realizadas cinco avaliações durante o período pós-operatório: avaliação clínica diária dos animais através da frequência cardíaca (FC), frequência respiratória (f), temperatura retal (TR), coloração de mucosas, tempo de preenchimento capilar (TPC), grau de desidratação, hemograma, perfil bioquímico (ureia, creatinina, colesterol, triglicérides e glicose); avaliação qualitativa do dreno; avaliação qualitativa da ferida cirúrgica, sendo esta também realizada em 14 e 30 dias quanto à presença ou não de exsudato, edema, integridade da pele, deiscência, necrose e fibrose cicatricial; avaliação quantitativa do fluido drenado e avaliação qualitativa do fluido drenado, quanto à celularidade, tipo e morfologia celular. Houve aumento dos valores médios de densidade, neutrófilos, linfócitos, e aumento do valor de mediana de fibrinogênio do fluido drenado em 72 horas em relação às 24 horas no grupo GD. Ocorreu aumento do exsudato em 24, 48 e 72 horas no GD quando comparado ao GC. Houve aumento do volume de fluido drenado em 60 e 72 horas em relação às 12 horas no grupo GD. Houve redução do valor médio de eritrócitos em 72 horas em relação ao basal no grupo GD. Observou-se aumento nos valores médios de hemoglobina e hematócrito em 24 horas no grupo GC quando comparado ao GD. Houve aumento no valor médio de neutrófilos segmentados no basal no GD quando comparado ao GC. Conclui-se que a técnica de colocação de dreno de sucção fechado apresenta maior teor de exsudato. Recomenda-se que o vácuo do dreno seja refeito de 12 em 12 horas, sendo inicialmentrealizado 6 horas após o término da cirurgia. O dreno de sucção fechado não gera desconforto adicional ao paciente; recomenda-se que o mesmo seja removido 72 horas após o término da cirurgia, quando a produção de fluido diminui, porém mesmo assim alguns indivíduos podem apresentar formação de seroma no pós-operatório tardio.


#5 - Improved method for diagnosis of Nerium oleander poisoning in necropsy tissues

Abstract in English:

Nerium oleander is an ornamental cardiotoxic plant found in tropical and subtropical areas of the World. Its toxicity is related to the content of cardioactive glycosides, mainly oleandrin, found throughout the plant. The present study aimed to describe a new and improved method for oleandrin detection in tissue samples. The determination of oleandrin was made after extraction with a modified QuEChERS technique and measurement by UFLC-MS/MS. A total of 36 guinea pigs (Cavia porcellus) were distributed into 3 groups (n=12): control group that received only water orally (CON), and two treated groups that received hydroalcoholic oleander extract at doses of 150mg.kg-1 (OLE 150) and 300mg.kg-1 (OLE 300) in single oral dose. After three hours, fragments of heart, kidneys, liver and brain were collected for determination of oleandrin levels. The extraction and chromatographic procedures were effective for oleandrin detection and quantification in tissues, with retention time of 1.2 min and detection limit of 0.001µg g-1. The chromatographic analysis of treated guinea pigs indicated that oleandrin is distributed equally among the analyzed tissues. The developed methodology is a reliable, effective and rapid form of diagnosis of N. oleander poisoning based on necropsy tissue samples.

Nerium oleander é uma planta cardiotóxica ornamental encontrada em áreas tropicais e subtropicais do mundo. Sua toxicidade é relacionada á presença de glicosídeos cardioativos, principalmente a oleandrina, encontrada em toda a planta. O presente estudo objetiva descrever um novo e aprimorado método para detecção da oleandrina em amostras de tecido. A determinação da oleandrina foi feita após extração utilizando técnica modificada de QuEChERS e mensuração por UFLC-MS/MS. Um total de 36 cobaios (Cavia porcellus) foi distribuído em três grupos (n=12): grupo controle que recebeu apenas água por via oral (CON), e dois grupos tratados que receberam extrato hidroalcóolico de oleander nas doses de 150mg.kg-1 (OLE 150) e 300mg.kg-1 (OLE 300) em uma única dose oral. Após três horas, fragmentos do coração, rins, fígado e cérebro foram coletados para determinação dos níveis de oleandrina. A extração e procedimentos cromatográficos foram eficientes na detecção e quantificação da oleandrina nos tecidos, com tempo de retenção de 1,2min e limite de detecção de 0,001µg g-1. A análise cromatográfica dos animais tratados indicou que a oleandrina é distribuída de forma equalizada pelos tecidos analisados. A metodologia desenvolvida representa uma forma de diagnóstica segura, efetiva e rápida da intoxicação por N. oleander a partir de amostras de tecidos de necropsia.


#6 - Value of transfaunation for the treatment of induced ruminal lactic acidosis in goats and sheep

Abstract in English:

The aim of this study was to evaluate the value of transfaunation in the treatment and recovery of goats and sheep with acute rumen lactic acidosis (ARLA) experimentally induced. Six goats (41.2±5.6kg) and six sheep (46.8±4.57kg), non-pregnant and non-lactating, received 15g of sucrose per kilogram of body weight for the disease induction. The ARLA was induced in each animal twice with an interval of 30 days after full recovery of the first induction. Therapeutic procedures consisted in removal of the rumen fluid content by washing and siphoning with an esophageal tube and correction of metabolic acidosis with intravenous electrolyte solutions containing sodium lactate or sodium bicarbonate. The transfaunation was part of only one of each animal treatment and consisted in administration of two liters of rumen fluid colleted from a healthy cow. Complete recovery was assessed by physical examinations and rumen fluid examinations up to four days after therapeutic procedures. The efficacy of treatment protocols, with or without transfaunation was compared. The protocol used was effective in induce the disease and goats and sheep showed clinical signs of maximum intensity (apathy, rumen stasis, abdominal distension, diarrhea and moderate dehydration) 16 hours after the intraruminal administration of sucrose. At this moment, the characteristics of ruminal fluid were similar to the typical ARLA. The frequency of ruminal movements became normal on the third day after therapeutic procedures, with no difference between species, and regardless transfaunation. The ruminal fluid transfer did not accelerate the recovery of appetite, considered normal only on the fourth day after treatment in both species. When transfauntation was received, goats and sheep showed recovery of characteristics of color, odor and consistency faster than when they did not receive. The fermentative activity of the rumen microbiota was not impacted by transfaunation and normalized two and three days after therapeutic procedures in sheep and goats, respectively. The transfaunation promoted faster return of the protozoa population, which was observed 24 hours after therapeutic procedures in both species. When the animals did not receive transfaunation, the protozoa returns occurred only on the second day after therapeutic procedures. Full recovery of the animals occurred within four days, regardless of transfaunation. In conclusion, the ruminal fluid transfer can not be considered crucial for the treatment and convalescence of goats and sheep affected by ARLA.

O objetivo deste trabalho foi avaliar a importância da transfaunação no tratamento e recuperação de cabras e ovelhas com acidose lática ruminal aguda (ALRA) induzida experimentalmente. Seis cabras (41,2±5,6kg) e seis ovelhas (46,8±4,57kg), não gestantes e não lactantes, receberam 15g de sacarose por quilo de peso corporal para indução da enfermidade. A ALRA foi induzida duas vezes em cada animal, com intervalo de 30 dias após recuperação total da primeira indução. Os procedimentos terapêuticos consistiram na remoção do conteúdo ruminal líquido por lavagem e sifonamento com auxílio de sondagem esofágica, e na correção da acidose metabólica com soluções eletrolíticas, contendo lactato ou bicarbonato de sódio, infundidas por via intravenosa. A transfaunação fez parte de apenas um dos tratamentos de cada animal e consistiu na administração por sondagem esofágica de 2L de suco ruminal de um bovino sadio. A recuperação completa foi avaliada por exames físicos e exames do suco ruminal realizados até quatro dias após os procedimentos terapêuticos. A eficácia dos protocolos de tratamento, com ou sem transfaunação, foi comparada. O protocolo de indução foi efetivo em induzir a enfermidade e as cabras e ovelhas apresentaram sinais clínicos de intensidade máxima (apatia, atonia ruminal, distensão abdominal, diarreia de consistência pastosa a líquida e desidratação moderada) 16 horas após a administração intrarruminal de sacarose, sem distinção entre as espécies. Neste momento, as características do suco ruminal mostraram-se semelhantes aos quadros típicos de ALRA. A frequência de movimentos ruminais se normalizou no terceiro dia após os procedimentos terapêuticos, sem diferença entre as espécies, e independente de terem recebido a transfaunação ou não. A transferência de suco ruminal também não acelerou a recuperação do apetite, que foi considerado normal somente no quarto dia após os procedimentos terapêuticos, em ambas as espécies. Em relação ao líquido ruminal, quando receberam a transfaunação, os caprinos e ovinos apresentaram recuperação das características de cor, odor e consistência mais rapidamente do que quando não receberam. A atividade fermentativa da microbiota ruminal não sofreu influência da transfaunação e se normalizou dois e três dias após os procedimentos terapêuticos nas ovelhas e cabras, respectivamente. A transfaunação promoveu o retorno mais rápido da população de protozoários ruminais, que já foram observados 24 horas após os procedimentos terapêuticos, em ambas as espécies. Quando os animais não receberam a transfaunação, o retorno dos protozoários só ocorreu no segundo dia após os procedimentos terapêuticos. A recuperação completa dos animais estudados ocorreu em até quatro dias, independente da realização da transfaunação ou não. Conclui-se que a transferência de suco ruminal não pode ser considerada medida crucial para o tratamento e convalescença de caprinos e ovinos acometidos por de ALRA.


#7 - Diagnostic utility of an immunochromatography test for the detection of Leptospira IgM antibodies in domestic dogs, 37(7):708-712

E-mail: luciaazocaraedo@gmail.com A cross-sectional study using 99 serum samples of dogs from southern Chile was conducted to determine the diagnostic utility of a rapid immunochromatography assay for the detection of Leptospira specific IgM antibodies as screening test and as a potential aid in the diagnosis of leptospirosis in animals with and without clinical suspicion of the disease. The Microscopic Agglutination Test (MAT) was used as reference assay. Anti-Leptospira antibodies were detected in 37.3% of the dogs with MAT. Using the immunochromatography test, specific IgM antibodies were found in 13.1% of sampled dogs. The sensitivity of the rapid test as screening assay was 29.7% (95% Confidence Interval=16.4-47.2) and the specificity was 96.7% (95% Confidence Interval=87.8-99.4). 40.0% of the canines with clinical suspicion of leptospirosis and 37.1% of dogs without clinical signs were serological reactors to MAT, but none of MAT reactive dogs with clinical suspicion tested positive in the rapid test. Rapid and user-friendly diagnostic procedures for canine leptospirosis such as this immunochromatography assay could be important tools to use in clinical practice, however, further studies are needed to obtain more information about their utility, considering that diagnostic tests could not have similar performances in different geographic locations, clinical and epidemiological contexts.

The Microscopic Agglutination Test (MAT) was used as reference assay. Anti-Leptospira antibodies were detected in 37.3% of the dogs with MAT. Using the immunochromatography test, specific IgM antibodies were found in 13.1% of sampled dogs. The sensitivity of the rapid test as screening assay was 29.7% (95% Confidence Interval=16.4-47.2) and the specificity was 96.7% (95% Confidence Interval=87.8-99.4). 40.0% of the canines with clinical suspicion of leptospirosis and 37.1% of dogs without clinical signs were serological reactors to MAT, but none of MAT reactive dogs with clinical suspicion tested positive in the rapid test. Rapid and user-friendly diagnostic procedures for canine leptospirosis such as this immunochromatography assay could be important tools to use in clinical practice, however, further studies are needed to obtain more information about their utility, considering that diagnostic tests could not have similar performances in different geographic locations, clinical and epidemiological contexts.

E-maol: ceambrosio@usp.br The increasing interest for the use of stem cells in regenerative medicine, is based on their self-renewal capacity, tissue repair and immunomodulation. The therapy with stem cells is a science in formation considered a promising therapeutic alternative for the treatment of various degenerative, hereditary and inflammatory disorders. The search for alternative sources of stem cells of easy obtention and isolation has led to the study of various tissues, includng embryonic attachments such as the amniotic membrane. However for effective therapeutic application are needed studies of the characteristics in vitro and preclinical and clinical trials. Thus, the aim of this study was a literature review regarding the characterization and clinical application of stem cells from the amniotic membrane in several domestic animal species.

A terapia utilizando células-tronco é uma ciência em formação considerada uma alternativa terapêutica promissora no tratamento de diversas patologias degenerativas, hereditárias e inflamatórias. A busca por fontes alternativas de CTs que possuam características como fácil obtenção e isolamento, tem levado ao estudo dos mais variados tecidos, onde se inclui os anexos embrionários como a membrana amniótica. Porém, para sua efetiva aplicação terapêutica são necessários estudos de suas características “in vitro”, além de testes pré-clínicos e clínicos. Com isso, o objetivo deste trabalho é realizar uma revisão bibliográfica sobre a caracterização e aplicação clínica das células-tronco da membrana amniótica em diversas espécies de animais domésticos.

E-mail: andrea@ufv.br Tissue engineering has been a fundamental technique in the regenerative medicine field, once it permits to build tri-dimensional tissue constructs associating undifferentiated mesenchymal cells (or mesenchymal stromal cells - MSCs) and scaffolds in vitro. Therefore, many studies have been carried out using these cells from different animal species, and rabbits are often used as animal model for in vivo tissue repair studies. However, most of the information available about MSCs harvesting and characterization is about human and murine cells, which brings some doubts to researchers who desire to work with a rabbit model in tissue repair studies based on MSCs. In this context, this study aimed to add and improve the information available in the scientific literature providing a complete technique for isolation, expansion and differentiation of MSCs from rabbits. Bone marrow mononuclear cells (BMMCs) from humerus and femur of rabbits were obtained and to evaluate their proliferation rate, three different culture media were tested, here referred as DMEM-P, DMEM´S and &#945;-MEM. The BMMCs were also cultured in osteogenic, chondrogenic and adipogenic induction media to prove their multipotentiality. It was concluded that the techniques suggested in this study can provide a guideline to harvest and isolate MSCs from bone marrow of rabbits in enough amount to allow their expansion and, based on the laboratory experience where the study was developed, it is also suggested a culture media formulation to provide a better cell proliferation rate with multipotentiality preservation.

Assim, muitos estudos têm sido realizados utilizando estas células oriundas de diferentes espécies animais, e os coelhos são frequentemente utilizados como um modelo animal para estudos in vivo de reparação tecidual. No entanto, a maioria das informações disponíveis sobre a coleta e caracterização de CEMs referem-se às células humanas e murinas, o que traz algumas dúvidas para pesquisadores que desejam trabalhar com coelhos em estudos de reparação de tecidos baseados em CEMs. Neste contexto, o presente estudo objetivou contribuir e aprimorar as informações disponíveis na literatura científica fornecendo uma técnica completa para o isolamento, expansão e diferenciação das MSCs de coelhos. Células mononucleares da medula óssea (CMMOs) do úmero e fêmur de coelhos foram obtidas e, para avaliar sua taxa de proliferação, três meios de cultura diferentes foram testadas, aqui referidos como DMEM-P, DMEM’S e &#945;-MEM. As CMMOs também foram cultivadas em meios de indução osteogênico, condrogênico, e linhagens adipogênico para provar a sua multipotencialidade. Concluiu-se que as técnicas sugeridas neste estudo podem fornecer um guia para a coleta e isolamento de CEMs da medula óssea de coelhos em quantidade suficiente para permitir a sua expansão e, com base na experiência de laboratório onde o estudo foi desenvolvido, é também sugerida uma formulação de meio de cultivo para proporcionar uma melhor taxa de proliferação celular com preservação da multipotencialidade.

The culture of these cells allows studies about their behavior and metabolism. Thus, cell culture is the basis for cell therapy and tissue engineering researches. A major concern regarding the use of cultivated stem cell in human or veterinary clinical routine is the risk of carcinogenesis. Cellular activities require a balanced redox state. However, when there is an imbalance in this state, oxidative stress occurs. Oxidative stress contributes to cytotoxicity, which may result in cell death or genomic alterations, favoring the development of cancer cells. The aim of this study was to determine whether there are differences in the behavior of cultured mesenchymal stem cells from canine adipose tissue according to its site of collection (omentum and subcutaneous) evaluating the rate of proliferation, viability, level of oxidative stress and cytotoxicity over six passages. For this experiment, two samples of adipose tissue from subcutaneous and omentum where taken from a female dog corpse, 13 years old, Pitbull. The results showed greater levels of oxidative stress in the first and last passages of both groups, favoring cytotoxicity and cell death.

A possibilidade de cultivá-las permite o estudo de seu comportamento e metabolismo. Assim, o cultivo celular representa a base para pesquisas de terapia celular e engenharia de tecidos. Uma das principais preocupações relativa ao uso de células-tronco cultivas na rotina clínica humana ou veterinária é a reprogramação dessas células em tumores benignos ou malignos. As atividades celulares necessitam de um estado redox balanceado e quando há algum desequilíbrio nessas reações ocorre o estresse oxidativo. O quadro de estresse oxidativo contribui pra a citotoxicidade podendo resultar em morte celular e até mesmo em alterações genômicas e ocorrência de células cancerígenas. O objetivo deste trabalho foi verificar se há diferenças no comportamento de células-tronco mesenquimais estromais de tecido adiposo de cão de acordo com o seu tecido de coleta (omento e subcutâneo) avaliando o cultivo dessas células quanto a sua taxa de proliferação, viabilidade, estresse oxidativo e citotoxicidade ao longo de seis passagens. Para a execução deste experimento foram utilizadas duas amostras de tecido adiposo coletas do subcutâneo e omento do cadáver de um cão, fêmea, 13 anos de idade, da raça Pitbull. O cadáver era oriundo do Hospital Veterinário Universitário e sofreu eutanásia devido a complicações no seu quadro de cardiomiopatia. As duas amostras foram encaminhadas para o isolamento e cultura celular. Os resultados mostraram que a primeira e última passagem em ambos os grupos são as passagens mais submetidas ao estresse oxidativo ficando mais sujeitas à citotoxicidade.
