#1 - Occurrence of Salmonella spp. in fecal samples from foals with and without diarrhea in the state of São Paulo: microbiological diagnosis, antimicrobial susceptibility profile, and molecular detection

Abstract in English:

The present study investigated Salmonella spp. in the feces of 200 foals up to one year of age (100 with clinical signs of diarrhea and 100 without clinical signs of diarrhea). Bacteriological culture, serotyping, antimicrobial susceptibility, and real-time PCR (qPCR SYBR® Green or a TaqMan®) for detecting the invA gene (with and without a selective pre-enrichment step in tetrathionate broth) were performed. Bacterial culture revealed 15% (n=30) of positive animals (21 animals with diarrhea and nine without diarrhea). Among the 30 isolates, 13 different serovars were identified: S. Infantis, S. Minnesota, S. I.4,5,12:i:-; S. Anatum, S. Cerro, S. Oranienburg, S. Braenderup, S. Give, S. Newport, S. IIIb 61:c:z35, S. 109:-:1.5, S. I.4.12:d:-, S. I.6.8:-:-. Multidrug resistance was found in 43.33% (n=13) of the isolates, with one isolate obtained from animals without diarrhea and 12 isolates from animals with diarrhea. All qPCR techniques used in the study classified more samples as positive for Salmonella spp. than the bacterial culture of feces. In addition, all qPCR techniques detected more positive animals in the diarrhea group than in the diarrhea-free group. The results confirm the utility of the qPCR method without the pre-enrichment step in tetrathionate as a rapid test for Salmonella spp. in carrier animals. In animals with clinical signs of diarrhea, it can be combined with bacterial culture (antimicrobial susceptibility testing and serotyping). The isolation of Salmonella spp. in nine animals without diarrhea confirms the importance of asymptomatic carrier animals in the epidemiology of the disease. The multidrug resistance observed highlights the importance of rational antimicrobial use in horses and adopting biosecurity protocols that are efficacious in controlling the spread of infections between animals and zoonotic transmission in farms.

O presente estudo investigou a ocorrência de Salmonella spp. em fezes de 200 potros com até um ano de idade (100 com sinais clínicos de diarreia e 100 sem sinais clínicos de diarreia), utilizando as técnicas de cultivo bacteriológico e PCR em tempo real (qPCR) pelos métodos de corante fluorescente (SYBR® Green) e sonda específica (Taqman®) para a detecção do gene invA com e sem etapa de pré-enriquecimento seletivo em caldo de tetrationato. O cultivo bacteriológico revelou 15% (n=30) de animais positivos (21 animais com diarreia e nove animais sem diarreia). Dentre esses 30 isolados, 13 sorovares diferentes foram identificados: S. Infantis, S. Minnesota, S. I.4,5,12:i:-; S. Anatum, S. Cerro, S. Oranienburg, S. Braenderup, S. Give, S. Newport, S. IIIb 61:c:z35, S. 109:-:1.5, S. I.4.12:d:-, S. I.6.8:-:-. Multirresistência foi constatada em 43,33% (n=13) dos isolados, sendo um isolado obtido de animal sem diarreia e 12 isolados de animais com diarreia. Todas as técnicas de qPCR empregadas no estudo apresentaram maior número de amostras classificadas como positivas para Salmonella spp. comparadas ao cultivo bacteriológico de fezes. Adicionalmente, em todas as técnicas de qPCR houve maior número de animais detectados como positivos no grupo de animais com diarreia em relação aos animais sem diarreia. Os resultados confirmaram a utilidade do método qPCR sem a etapa de pré-enriquecimento em tetrationato, como um teste rápido para detecção de Salmonella spp. em animais portadores ou em animais com sinais clínicos de diarreia. O cultivo bacteriológico deve ser associado para a realização do teste de sensibilidade aos antimicrobianos e sorotipificação. O isolamento de Salmonella spp. em nove animais sem diarreia, confirma a importância dos animais portadores assintomáticos na epidemiologia da doença. A multirresistência observada evidencia a importância do uso racional de antimicrobianos em equinos e a importância da adoção de protocolos de biossegurança que sejam eficazes para controlar a disseminação de infecções entre animais e a transmissão zoonótica nas fazendas.


#2 - Evaluation of skin prick test, exclusion diet and dietary challenge in the diagnosis of food allergy in dogs with chronic pruritus

Abstract in English:

Allergic dogs are commonly sensitized to food allergens. We evaluated the use of the prick test to diagnose food allergies in dogs with pruritus and the efficacy of an exclusion diet based on the test. The prick test was performed in 10 healthy dogs and 34 dogs with pruritus, of which 25 received an exclusion diet for 60 days and was challenged with positive food on the test. pVAS and CADESI-4 were assessed on days 0, 30, and 60 after re-exposure. As a result, two control group dogs reacted to a single food allergen, milk and wheat. Of the 25 dogs with pruritus that reacted to food allergens, 24 (96%) reacted to more than one food allergen, and only one (4%) reacted to a single food protein: pork. In the test group (n=25), there was a significant improvement of pVAS and CADESI-4 after 30 and 60 days of dietary exclusion, with significant worsening of the scores with food allergen challenge. In conclusion, the prick test can be used for screening food allergens to make an exclusion diet.

Cães alérgicos são comumente sensibilizados a alérgenos alimentares. Nós avaliamos o uso do teste de puntura para o diagnóstico de alergias alimentares em cães com prurido e a eficácia de uma dieta de exclusão baseada no teste. O teste de puntura foi realizado em 10 cães saudáveis e em 34 cães com prurido, dos quais 25 receberam uma dieta de exclusão por 60 dias e depois desafiados com o alimento que foi positivo no teste. pVAS e CADESI-4 foram realizados nos dias 0, 30, 60 e depois da reexposição. Como resultados, dois cães do grupo controle reagiram a um único alérgeno alimentar, leite e trigo. Dos 25 cães com prurido que reagiram aos alérgenos alimentares, 24 (96%) reagiram a mais de um alérgeno alimentar e apenas um (4%) reagiu a uma única proteína, que foi porco. No grupo teste (n=25), houve uma significativa melhora do pVAS e CADESI-4 depois de 30 e 60 dias de dieta de exclusão, com significante piora dos scores com o desafio alimentar. Como conclusão, o teste de puntura pode ser usado para selecionar alérgenos alimentares para fazer uma dieta de exclusão.


#3 - Bacterial identification, somatic cell count, antimicrobial profile and toxigenic Staphylococcus strains search from mastitic cow milk samples on small farms properties

Abstract in English:

Bovine mastitis has a negative impact on milk production and can pose risks to public health. The present study aimed to evaluate the quality of bovine milk from small farms in the Botucatu/SP region. Somatic cell counts (SCC), identification of pathogens involved in mastitis, and sensitivity antimicrobial profile of staphylococci isolated were performed. The presence of enterotoxin encoding genes in isolates of staphylococci obtained from milk was investigated. Milk samples from individual mammary quarters of cows were submitted to the California mastitis test (CMT) and SCC. Of the 239 dairy cows from 21 dairy herds evaluated (mean = 11.4 animals/property), two cows (0.8%) presented clinical mastitis and 86 (35.9%) subclinical mastitis. Bacterial culture was performed in 177 quarter milk samples. Staphylococci were identified in 55 (31.1%), corynebacteria in 45 (25.4%), streptococci in 25 (14.1%) and coliforms in four (2.3%) milk samples. Average SCC from culture-positive samples was 1598x103 cells/mL, in case of staphylococci was 1362x103 cells/ml, streptococci was 2857x103 cells/mL, corynebacteria was 976x103 cells/mL and in the cases of coliforms 1161x103 cells/mL were obtained. Staphylococci showed a high sensitivity (>95%) to cephalothin, cotrimoxazole, enrofloxacin, and gentamicin, with a 41.2% resistance to penicillin and 11.8% to oxacillin. Both coagulase positive (CPS) and negative staphylococci (CNS) carried genes encoding enterotoxins in 21.6% of the first group and 41.9% in the second. The sea gene was the most detected 45.8% (n=24) between them, followed by seb with 29.2% and sec with 25.0%. The sed gene was not identified. We highlight the potential risk to public health in the possibility of strains of Staphylococcus spp. enterotoxin-producing genes that can cause staphylococcal food poisoning.

A mastite bovina impacta negativamente a produção leiteira e pode acarretar riscos à saúde pública. O presente estudo teve como objetivo a avaliação da qualidade do leite bovino proveniente de pequenas propriedades na região de Botucatu/SP. Foi realizada a contagem de células somáticas (CCS), identificação dos patógenos envolvidos nas mastites, e realizado o perfil de sensibilidade aos antimicrobianos dos estafilococos isolados. Pesquisou se a presença de genes codificadores de enterotoxinas em isolados de estafilococos obtidos a partir do leite mastítico. Amostras de leite de quartos mamários individuais de vacas foram submetidas ao “California mastitis test” (CMT) e à CCS. Das 239 vacas em lactação provenientes de 21 rebanhos leiteiros avaliados (média = 11,4 animais/propriedade), dois (0,8%) animais apresentaram mastite clínica e, 86 (35,9%) mastite subclínica. 177 amostras de leite foram cultivadas em ágar sangue bovino 5% e ágar MacConckey e obteve-se 55 (31,1%) Staphylococcus spp., 25 (14,1%) Streptococcus spp., 45 (25,4%) Corynebacterium spp. e quatro (2,3%) coliformes. A média da CCS das amostras procedentes de todos os quartos mamários infectados avaliados foi de 1598x103 células/mL, enquanto que nos casos que foram isolados Staphylococcus spp. foi de 1362x103 células/mL, Streptococcus spp. 2857x103 células/mL, Corynebacterium spp. de 976x103 células/mL e nos casos de coliformes 1161x103 células/mL. Os estafilococos revelaram grande sensibilidade (>95%) à cefalotina, cotrimoxazol, enrofloxacina e gentamicina, com resistência de 41,2% à penicilina e 11,8% à oxacilina. Tanto estafilococos coagulase positivos (ECP) como negativos (ECN) revelaram genes codificadores de enterotoxinas em 21,6% do primeiro grupo e 41,9% no segundo. O gene sea foi o mais detectado 45,8% (n=24), seguido pelo seb com 29,2% e sec com 25,0%. O gene codificador da sed não foi identificado. Frente aos resultados, destaca-se o risco potencial à saúde pública pela possibilidade de veiculação de linhagens de Staphylococcus spp. carreadores de genes produtores de enterotoxinas, podendo ocasionar toxi-infecções alimentares.


#4 - Insulin favors acute inflammatory reaction in alloxan‑diabetic tilapia during infectious aerocystitis

Abstract in English:

In vertebrates, the inflammatory reaction is responsible for modulating the initial nonspecific defense until specific immunity is acquired. In this context, numerous studies in mammals have demonstrated the participation of insulin in the inflammatory response, favoring cell proliferation and the migratory capacity of endothelial cells, vascular smooth muscle cells and monocytes, as well as mediating the expression of pro-thrombotic and pro-fibrotic factors. However, little is known about the effect of this peptidic hormone on the inflammatory reaction in teleostean fish. In order to evaluate the participation of insulin in the acute inflammatory response of Nile tilapia, Oreochromis niloticus, during aerocystitis induced by Aeromonas hydrophila, and 48 aloxane-diabetic tilapia were used, constituting two groups: diabetics treated with insulin and diabetics without treatment. After six, 24, and 48 hours of inflammatory stimulation, tilapia were submitted to deep anesthesia for euthanasia and necropsy, and thus, obtaining exudate and harvesting of the swim bladder for analysis of the inflammatory reaction. Based on this premise, the present study demonstrated the participation of insulin in the acute inflammatory reaction of alloxan-diabetic tilapia by favors the cellular accumulation in the exudate, the proliferative effect of fibrous tissue and neovascularization in the inflamed site. Such findings reinforce the old hypothesis that insulin plays an important role in the innate immune response during acute inflammatory reaction, being an important pro-inflammatory hormone. However, Nile tilapia proved to be a promising experimental model for studies and advances in research involving diabetes mellitus.

Em vertebrados, a reação inflamatória é responsável por modular a defesa inicial não-específica, até que imunidade específica seja adquirida. Neste contexto, inúmeros estudos em mamíferos têm demonstrado a participação da insulina sobre a resposta inflamatória, favorecendo a proliferação celular e a capacidade migratória das células endoteliais, células do músculo liso vascular e dos monócitos, além de mediar a expressão de fatores pró-trombótico e pró-fibrótico. Porém, pouco se conhece o efeito deste hormônio peptídico sobre a reação inflamatória em peixes teleósteos. Para avaliar a participação da insulina sobre a resposta inflamatória aguda em tilápias do Nilo, Oreochromis niloticus, na aerocistite induzida por Aeromonas hydrophila, foram utilizadas 48 tilápias aloxano-diabéticas, constituindo dois grupos: dos diabéticos tratados com insulina e diabéticos sem tratamento. Após, seis, 24 e 48 horas do estimulo inflamatório, as tilápias foram submetidas à anestesia profunda para eutanásia e necropsia, e assim, obtenção de exsudato e colheita da bexiga natatória para analise da reação inflamatória. Partindo-se desta premissa, o presente estudo demonstrou a participação da insulina na reação inflamatória aguda infecciosa de tilápias do Nilo aloxano-diabéticas por favorecer o acúmulo positivo celular no exsudato, assim como o efeito proliferativo de tecido fibroso e a neovascularização no local inflamado. Tais achados reforçam a hipótese de que a insulina desempenha importante papel na resposta imune inata na reação inflamatória aguda, sendo um importante hormônio pró-inflamatório. Contudo, a tilápia do Nilo demonstrou ser um modelo experimental promissor para estudos e avanços em pesquisas envolvendo o diabetes mellitus.


#5 - Frequency, serotyping and antimicrobial resistance pattern of Salmonella from feces and lymph nodes of pigs, 36(12):1165-1170

E-mail: josepaes@fmvz.unesp.br Salmonellosis is a foodborne disease caused by bacteria of the genus Salmonella, being pigs and pork-products potentially important for its occurrence. In recent decades, some serovars of Salmonella have shown increase of resistance to conventional antimicrobials used in human and animal therapy, with serious risks for public health. The aim of this study was to evaluate feces (n=50), mediastinal (n=50), mesenteric (n=50) and mandibular (n=50) lymph nodes obtained from slaughter houses for Salmonella spp. Positive samples were serotyped and subjected to an in vitro antimicrobial susceptibility test, including the extended-spectrum beta-lactamase (ESBL) production. Salmonella species were identified in 10% (20/200) of total samples. From these, 20% (10/50) were identified in the submandibular lymph nodes, 18% (9/50) in the mesenteric lymph nodes, 2% (1/50) in feces and 0% (0/50) in the mediastinal lymph nodes. The serotypes found were Salonella Typhimurium (55%), S. enterica subsp. enterica 4,5,12: i: - (35%), S. Brandenburg and S. Derby with 5% (5% each). All strains showed resistance to at least one antimicrobial; 90% were resistant to four or more antimicrobials, and 15% were multidrug-resistant. Resistance to ciprofloxacin, tetracycline and nalidixic acid was particularly prevalent amongst the tested serovars. Here, we highlighted the impact of pigs in the epidemiological chain of salmonellosis in domestic animals and humans, as well as the high antimicrobial resistance rates of Salmonella strains, reinforcing the necessity for responsible use of antimicrobials for animals as an emergent One Health issue, and to keep these drugs for human therapy approaches.

Deste modo, o objetivo do presente trabalho foi pesquisar a presença de Salmonella a partir de fezes (n=50), linfonodos mediastinos (n=50), mesentéricos (n=50) e submandibular (n=50) oriundos de um abatedouro suíno. As cepas isoladas foram sorotipadas e testadas quanto a resistência antimicrobiana. A presença de Salmonella isolada foram em 10% (20/200) do total de amostras, sendo 20% dos linfonodos submandibulares, 18% dos linfonodos mesentéricos e 2% das fezes. Os sorotipos encontrados foram S. Typhimurium (55%), S. enterica subsp. enterica 4,5,12: i: - (35%), S. Brandenburg (5%) e S. Derby (5%). Todas a cepas apresentaram resistência a pelo menos um antimicrobiano testado, sendo 90% resistente pelo menos quatro antimicrobianos. Destes, 15% foram classificadas como multidrogas resistentes. Os antimicrobianos mais resistentes entre os sorovares isolados foram a ciprofloxacina, tetraciclina e o ácido nalidixico. A presença de cepas de Salmonella resistente a antimicrobianos na espécie suína tem gerado um grande impacto epidemiológico entre homem e animal, reforçando cada vez mais a necessidade do uso adequado de drogas principalmente relacionado com o tema “One Health”.


#6 - Osteology of Melanosuchus niger (Crocodylia: Alligatoridae) and the evolutionary evidence, 36(10):1025-1044

Pesquisa Veterinária Brasileira 36(10):1025-1044. E-mail: luceliabio@yahoo.com.br The objective was an anatomical description of the skeleton of Melanosuchus niger, in order to contribute with evolutionary information about the species. Three adult specimens of M. niger with an average length of 2.40m were used, originating from the biologic collection of Lapas-UFU. In the forelimb, the scapula is bigger than the coracoid. Regarding the hindlimbs, the pubic does not participate in the formation of the acetabulum; the contact with the ilium is made by ligaments, and its articulation with the rump allows dorsal-ventral movements. Regarding the forelimbs, the humerus is a stylopodium element, and the ulna and radius a zeugopodium element. The carpus exibits the ulnar-radial+intermedium fusion, fusion of the distal carpals 3+4+5, and the pisiform. It has five metacarpals, numbered lateromedially as metacarpal 1, 2, 3, 4, and 5. The phalangeal formula is 2:3:4:3:2. Regarding the pelvic limbs, the stylopodium is formed by the femur, and the zeugopodium by tibia and fibula. In tarsus has four bones: fusion of the intermedium+centrale, fibulare, distal tarsal 3, and distal tarsal 4. It has four long metatarsals I, II, III and IV, with metatarsal II and III being relatively longer than the others. Metatarsal V is a very small bone; the feet have the phalangeal formula 2:3:4:4. At the skull, the nasal opening is only the palatine bones, vomer, pterygoid, premaxilla and maxilla forming the bone structure of the secondary palate; the parietal bone is the only element on the cranial roof. In the pos- axial skeleton ist distinct pairs of ribs which articulate with the cervical, dorsal, lumbar, sacral and caudal vertebrae. The gastralia consists of seven rows of fine bone located between the pubic bone and caudal part of the sternum.

Utilizaram-se três espécimes adultos de M. niger, com comprimento médio de 2,40m, provenientes da coleção biológica do Lapas-UFU. Na cintura peitoral, a escápula é maior do que o coracóide. Já nos elementos da cintura pelvina, o púbis não participa da formação do acetábulo, o contato com o ilío, ocorre por ligamentos, e sua articulação com o ísquio, permite movimentos dorso-ventrais. Nos membros torácicos, o úmero figura como elemento do estilopódio, a ulna e rádio como elementos do zeugopódio. No carpo há o ulnar do carpo, fusão do radial+intermédio, fusão dos distais do carpo 3+4+5 e o pisiforme; possui cinco metacarpos, numerados lateromedialmente e a fórmula falângica 2:3:4:3:2. Nos membros pelvinos, o estilopódio é formado pelo fêmur e o zeugopódio pela tíbia e fíbula. No tarso há a fusão do intermédio+central, fibular do tarso, distal do tarso 3, distal do tarso 4; possui quatro metatarsos longos I, II, III e IV, sendo os metatarsos II e III maiores que os demais. O metatarso V é um osso bastante reduzido e o pé possui a fórmula falângica 2:3:4:4. No crânio, a abertura nasal é única, o palatino, vômer, pterigóide, pré-maxila e maxila formam a estrutura óssea do palato secundário; o osso parietal é o único elemento no teto craniano. No esqueleto pós- axial em pares de costelas distintas que se articulam com as vértebras cervicais, dorsais, lombares, sacrais e caudais. A gastrália é formada por sete fileiras de ossos finos localizados entre o púbis e a região caudal do esterno.

E-mail: fabianocl21@hotmail.com Skull represents the segment with conspicuous adaptations that, in lizards, may be conservative or promoted by selective pressures. The aim of assisting the morphological knowledge of reptiles, we provide a detailed description of the neurocranium of Iguana iguana iguana based on analysis of three dried adult skeletons. The skull of this species has basal characteristics in lizards without closure of cranial openings and general triangular shape. Bony structures that form the caudal base have many fusions, especially on the floor. In the caudal face the exoccipital and the opisthotic are fused and form the otooccipital, which contributes to the formation of the lateral part of the condyle. The central part is formed by the condyle supraocciopital. Fusions and skeletal structures in Iguana are similar to other lizards. There are no autopomorphies in the neurocranium for this species.

E-mail: fabianocl21@hotmail.com O crânio representa o segmento com conspícuas adaptações que, nos lagartos, podem ser conservativas ou impulsionadas por pressões seletivas. Objetivando subsidiar o conhecimento morfológico dos répteis, fornecemos uma descrição detalhada dos ossos que formam o neurocrânio de Iguana iguana iguana com base na análise de três esqueletos secos de espécimes adultos. O crânio da referia espécie possui características basais entre os lagartos sem o fechamento das aberturas cranianas e formato geral triangular. As estruturas ósseas que formam a base craniana apresentam muitas fusões, principalmente no assoalho. Na face caudal o exoccipital e o opistótico estão fundidos e formam o otoccipital, que contribui para a formação dos terços laterais do côndilo occipital. A parte central do côndilo é formada pelo supraoccipital. Fusões e estruturas esqueléticas presentes em Iguana são similares aos demais lagartos. Não foram descritas autapomorfias no neurocrânio para esta espécie.

E-mail: adriana.maciel@ufra.edu.br The goal of this study was to describe gross and microscopic lung lesions associated with the parasitism by Sebekia oxycephala between 100 specimens of black caiman (Melanosuchus niger) slaughtered in the Mamirauá Sustainable Development Reserve, situated in the Amazonas State, Brazil. During the macroscopic evaluation, specimens of parasites found in the lung tissue were collected individually in AFA (ethyl alcohol - formalin - glacial acetic acid) and 5% formalin for parasitological evaluation and taxonomic classification. Lung tissue samples of all the animals, regardless of the occurrence of gross lesions, were fixed in 10% formalin and embedded in paraffin. Histological sections obtained from the samples stained by Hematoxylin-Eosin were evaluated by light microscopy. Grossly, 4 of the 100 animals (4%) presented pentastomid specimens in the lung parenchyma. These parasites were taxonomically classified as Sebekia oxycephala. No macroscopic alterations were observed, however microscopically pulmonary lesions were found in 37% of the cases, and inflammatory lesions associated with the parasitism corresponded to 75.6% of them (28/37). In these cases, degenerate segments and eggs of S. oxycephala were surrounded by a fibrous capsule and inflammatory infiltrate composed predominantly by foreign body giant cells. Three specimens exhibited thickening of the alveolar septa and seven specimens presented multifocal inflammatory granulocytic infiltration in the lung parenchyma. Generally, the associated parasitic lesions had mild intensity and did not appear to represent an important cause of lung disease in the studied population. This is the first description of lung lesions in M. niger associated with parasitism by S. oxycephala in the Brazilian Amazon.

Durante a avaliação macroscópica, exemplares dos parasitos encontrados no tecido pulmonar foram coletados individualmente em AFA (Álcool etílico - Formalina - Ácido acético glacial) e formol a 5% para avaliação parasitológica e classificação taxonômica. Amostras de tecido pulmonar de todos os animais, independentemente da ocorrência de lesões macroscópicas, foram fixadas em formol 10% e incluídas em parafina. Secções histológicas coradas por Hematoxilina-Eosina destas amostras foram avaliadas por meio de microscopia óptica. Macroscopicamente, 4 dos 100 animais (4%) apresentaram espécimes de pentastomídeos no parênquima pulmonar. Os parasitos foram classificados taxonomicamente como pertencentes à espécie Sebekia oxycephala. Nenhuma alteração macroscópicas foi observada, porém, microscopicamente, lesões pulmonares foram encontradas em 37% dos casos, sendo que, lesões inflamatórias associadas ao parasitismo corresponderam a 75,6% dos mesmos (28/37). Nestes, segmentos degenerados e ovos de S. oxycephala encontravam-se envolvidos por cápsula de tecido conjuntivo fibroso e infiltrado inflamatório predominantemente composto por células gigantes do tipo corpo estranho. Três espécimes apresentaram espessamento de septos alveolares e sete exemplares continham infiltrado inflamatório granulocítico multifocal no parênquima pulmonar. As lesões associadas ao parasitismo, de modo geral, apresentaram intensidade discreta e parecem não representar uma causa importante de doença pulmonar entre a população estudada. Esta é a primeira descrição de lesões pulmonares em M. niger associadas ao parasitismo por S. oxycephala na Amazônia brasileira.

E-mail: spatroca@yahoo.com.br To compare two serologic tests for detection of antibodies against Neospora caninum in sera from buffaloes, samples were collected from 288 buffaloes of 2 to 10 years of age. To identify the presence of IgG, anti-N. caninum was used for the indirect immunofluorescence assay (IFAT), with title 200 as the cutoff, and for the immunoenzymatic test (indirect ELISA), considering as positive samples with ratio S/P ≥0.5. There were 153 (53.12%) animals positive for N. caninum by IFAT, whilst 50 (17.36%) animals were reactive in ELISA. The presence of antibodies anti-N. caninum demonstrates that the parasite is circulating among buffaloes raised in Pará State, Brazil. ELISA and IFAT tests could be used to diagnose immunoglobulins against this agent. However there was a weak correlation (Kappa = 0.36) between both tests, considering the IFAT as the gold standard.

Para identificar a presença de imunoglobulina G anti-N. caninum utilizou-se à reação de imunofluorescência indireta (RIFI), tendo o título 200 como ponto de corte, e o Ensaio Imunoenzimático indireto (ELISA-indireto), considerando-se positiva as amostras que obtiveram razão S/P≥0,5. Observaram-se 153 (53,12%) animais soropositivos para N. caninum, através da RIFI, enquanto que 50 (17,36%) animais foram reagentes no ELISA. A ocorrência de anticorpos anti-N. caninum demonstram que o parasito esta circulando entre búfalas criadas no estado do Pará, sendo que ambos os teste de RIFI e ELISA podem ser utilizados para diagnosticar imunoglobulinas contra este agente. No entanto observou-se uma fraca correlação (Kappa=0,36) entre ambos os testes, considerando a RIFI como padrão ouro.
