#1 - Experimental poisoning by Crotalaria lanceolata and Crotalaria pallida seeds in broilers

Abstract in English:

Crotalaria lanceolata E. Mey. and Crotalaria pallida Aiton. are leguminous plants of family Fabaceae found in most of the Brazilian territory. They were initially used as green manure and due their easy spread they are currently considered weeds in crops. Soybean and corn contamination can occur through the mechanical harvesting of these grains along with seeds of the Crotalaria species, which end up in the formulation of feed for production animals. Crotalaria spp. genus has toxic pyrrolizidine alkaloids (PA). Most plant species belonging to this genus can cause acute or chronic liver injury. In a first stage, one-day old broilers were divided into three groups: Group A (C. pallida seeds), Group B (C. lanceolata seeds), and Group C (Control). Groups A and B were divided into five subgroups, each with eight broilers, which received the following doses of the respective seeds in feed as of the 7th day of age: daily doses of 0.4%, 0.8% and 2.5%, and single doses of 15% and 25%. Four broilers in each study group were euthanized at 28 days of age - completing 21 days of seed consumption, and the four remaining broilers were euthanized at 42 days of age - completing 35 days of seed consumption. In a second stage, experiments were conducted using seeds of both the aforementioned plants with 28-day old broilers. These were divided into three groups of four animals each: Group D (C. pallida seeds) and Group E (C. lanceolata seeds), which received the respective seeds at daily doses of 1% and 2% in feed for 20 days, and Group F (Control). These broilers were euthanized when they were 80 days old. C. lanceolata seeds showed higher toxicity to broilers than C. pallida seeds, both supplied as of the 7th day of life. Clinical signs included inappetence, ruffled feathers, and brown diarrhea. The following gross lesions were observed: subcutaneous edema, ascites, hydropericardium, yellowish liver with hypertrophy or atrophy and enhanced lobular pattern, and distended gallbladder. Histologic lesions present in all birds in varying degrees were characterized by tumefaction and vacuolar degeneration of hepatocytes. The following clinical conditions and gross lesions were observed in the broilers: hepatocyte megalocytosis and karyomegaly, slight biliary epithelial hyperplasia, eosinophilic spheroids, and nuclear invagination with loss of hepatocyte cord architecture.

Inicialmente foram utilizadas como adubação verde e devido sua fácil disseminação são consideradas invasoras de culturas. Através da colheita mecanizada da soja e milho pode ocorrer a contaminação destes grãos com sementes dessas espécies e entrar na formulação de ração para animais de produção. O gênero Crotalaria spp. possui alcaloides pirrolizidínicos de ação tóxica. A maioria das espécies desse gênero causam lesões hepáticas, com evolução aguda, ou, crônica. Frangos de corte de um dia de vida foram divididos em três grupos: Grupo A (sementes de C. pallida - doses diárias de 0,4%, 0,8%, 2,5% e doses únicas de 15% e 25%), Grupo B (sementes de C. lanceolata - doses diárias de 0,4%, 0,8%, 2,5% e doses únicas de 15% e 25%) e Grupo C (Controle). Os Grupos A e B foram divididos em cinco subgrupos, com oito frangos cada, que a partir do sétimo dia de vida, receberam as doses estabelecidas. Quatro frangos de cada grupo foram sacrificados aos 28 dias, e os quatro restantes aos 42 dias de vida. Também foram conduzidos experimentos com as sementes dessas duas plantas com aves de 28 dias, as quais foram divididas em três grupos: Grupo D (C. pallida - doses de 1% e 2% diariamente, durante vinte dias), Grupo E (C. lanceolata - doses de 1% e 2% diariamente, durante vinte dias) e Grupo F (Controle). Cada grupo composto por quatro aves. Estas aves foram sacrificadas ao completarem 80 dias de vida. Sementes de C. lanceolata demonstraram maior toxicidade para frangos de corte do que sementes de C. pallida. Os sinais clínicos foram inapetência, penas arrepiadas e diarreia acastanhada. Na macroscopia observou-se edema subcutâneo, ascite, hidropericárdio, fígado de coloração amarelada com hipertrofia e, ou, atrofia, evidenciação do padrão lobular e vesícula biliar distendida. As lesões histológicas presentes em todas as aves, em diferentes graus caracterizaram-se por tumefação e degeneração vacuolar de hepatócitos. Nas aves que manifestaram alterações clínicas e lesões macroscópicas, havia megalocitose, cariomegalia, hiperplasia do epitélio biliar, leve, esferoides eosinofílicos e invaginação nuclear com perda da arquitetura dos cordões de hepatócitos.

E-mail: aclauren@bio.feis.unesp.br Two experiments were conducted to evaluate the use of dried seeds of Piper cubeba in the diets of 1 to 21-day-old broilers and its effect on biochemical blood profile and biometry of the organs. In each experiment, 240 one-day-old male broiler chicks Cobb were distributed in a completely randomized design with five treatments and four replicates of 12 birds per experimental plot. The first experiment evaluated the use of Piper cubeba in considered highly digestible diets based on corn and soybean meal, and the second evaluated the use of pepper in low digestibility diets, which were obtained with the inclusion of meat and bone meal. Regarding the blood chemistry profile data of the experiment I, with the exception of gamma glutamyl transferase, all other serum levels were within recommended limits for poultry, and only triglyceride levels differed between treatments (P <0.05). In the second experiment there was significant difference in albumin levels, cholesterol and uric acid, but this was not the result of metabolic disorders, because except for the gama glutamyl transferase levels, all variables were within recommended levels. In both experiments, there were no differences (P> 0.05) for the biometry of organs. The conditions under which the experiments were performed at inclusion of Piper cubeba seeds in feed for broilers has not provided any toxicity to poultry.

No primeiro experimento foi avaliado o uso da Piper cubeba em dietas consideradas de alta digestibilidade, a base de milho e farelo de soja, e no segundo avaliou-se o uso da pimenta em dietas de baixa digestibilidade, as quais foram obtidas com a inclusão de farinha de carne e ossos. Com relação aos dados de perfil bioquímico sanguíneo do experimento I, com exceção da gama glutamil transferase, todos os demais níveis séricos se apresentaram dentro dos limites recomendados para aves, e apenas os níveis de triglicerídeos diferiram entre os tratamentos (P<0,05). No experimento II houve diferença significativa para os níveis de albumina, colesterol e ácido úrico, porém isso não foi resultado de desordens metabólicas, visto que com exceção da gama glutamil transferase, todos os níveis encontraram-se dentro dos limites recomendados. Em ambos os experimentos não foram observadas diferenças (P>0,05) para a biometria dos órgãos. Nas condições em que os experimentos foram realizados a inclusão das sementes de Piper cubeba na ração para frangos de corte não proporcionou alterações bioquímicas e biométricas que possam limitar seu uso como material vegetal para os estudos fitogênicos.


#3 - Alternative control of helminthes, parasites of Astyanax cf. zonatus, using phytotherapy with seeds of Cucurbita maxima and Carica papaya, 32(1):5-10

E-mail: fabriciomramos@gmail.com The objective of this study was to evaluate the effectiveness of an alternative method with pumpkin and papaya seeds, dried and ground, for control of helminth parasites of Astyanax cf. zonatus. Sixty fish were distributed into twelve containers, with one fish/liter. The experiment consisted of four treatments and three replications: TJ = fish in fasting; TRC = fish fed with commercial diet; TSA = fish fed ad libitum with pumpkin seed, and TSM = fish fed ad libitum with papaya seeds. After seven days of feeding, all fish were weighed and blood was taken for blood smears. Efficacy was determined by checking the presence of parasites in the gills, stomach and intestine, in 40% of fishes per treatment (n=6). TJ and TSM showed 39% and 25% of weight loss respectively. The TSA showed better efficacy against the nematode in intestine and stomach (95.26% and 92.48%). The TSM promoted 72% of efficiency on monogeneans control. Hematology revealed an increase of monocyte values to treatment TSM. A decrease of eosinophil values was observed in treatments TSA, TSM and TRC. Thus, feeding fishes with pumpkin is a effective alternative method of low cost for control of nematodes in Astianax cf. zonatus.

O experimento constituiu de quatro tratamentos e três repetições: TJ = peixes deixados em jejum; TRC = peixes alimentados com ração comercial; TSA = peixes alimentados ad libitum com abóbora, e TSM = peixes alimentados ad libitum com mamão. Após sete dias de alimentação, todos os peixes foram pesados, e o sangue foi retirado para extensão sanguínea. A eficácia foi determinada, verificando a presença de parasitos nas brânquias, no estômago e no intestino. Os peixes do TJ e TSM apresentaram perda de peso (39% e 25%, respectivamente). O TSA apresentou melhor eficácia no controle de nematóides do intestino e do estômago (95,26% e 92,48%). No controle de monogenéticos TSM promoveu 72% de eficácia. Na hematologia observou-se aumento de monócitos nos peixes do TSM e os valores de eosinofilos diminuíram nos tratamentos TSA, TSM e TRC. Assim pode-se concluir que a alimentação com abóbora pode ser utilizado como um controle alternativo eficaz de nematóides intestinais do lambari.

E-mail: moleta@ufmt.br Hepatocellular necrosis are reported in swine after consumption of diets containing grains of “sorgo-granífero” (Sorghum bicolor) accidentally contaminated with Crotalaria spectabilis seeds in the municipality of Juscimeira, MT. Clinical signs began 24-48 hours after consumption of contaminated ration and were characterized by depression, lethargy, apathy, loss of appetite, vomiting, pale or jaundiced mucous membranes, ascites, lateral recumbency, the lateral position with paddling and convulsions, clinical outcome was 48-60 hours followed by death. 76 pigs died in four properties. The main gross lesions were liver increased in size and lobular illustration with red-brown central areas interspersed with yellowish areas, ascites and hydrothorax with reddish-yellow liquid containing filaments with aspect of fibrin, enlarged lymph nodes and interlobular pulmonary edema. The disease was experimentally reproduced with 16 pigs divided into six groups that received seeds of C. spectabilis in different doses. Hepatocellular necrosis occurred in seven pigs, two of which received daily doses of 2.5g/kg and five who received single doses of 5 and 9.5g/kg. Ten daily doses of 0.5 and 1.25g/kg caused liver fibrosis.

As Principais alterações macroscópicas foram fígado aumentado de tamanho com evidenciação do padrão lobular, ascite e hidrotórax com líquido de coloração amarelo avermelhado contendo filamentos com aspecto de fibrina, linfonodos aumentados e edema pulmonar interlobular. A doença foi reproduzida utilizando-se 16 suínos divididos em seis grupos que receberam sementes de C. spectabilis em diferentes doses. Necrose hepatocelular ocorreu em sete suínos, sendo dois que receberam doses diárias 2,5g/kg e cinco que receberam doses únicas de 5,0 e 9,5g/kg. Dez doses diárias de 0,5 e 1,25g/kg causaram fibrose hepática.

#6 - Aspectos clínico-patológicos da intoxicação experimental pelas sementes de Crotalaria mucronata (Fabaceae) em bovinos, p.149-156

[Experimental poisoning by Crotalaria mucronata (Fabaceae) seeds in cattle.] Aspectos clínico-patológicos da intoxicação experimental pelas sementes de Crotalaria mucronata (Fabaceae) em bovinos. Pesquisa Veterinária Brasileira 27(4):149-156. Projeto Sanidade Animal Embrapa/UFRRJ, Seropédica, RJ 23890-000, Brazil. E-mail: mailto:mubogho@hotmail.com Experiments were performed to define the clinical and pathological picture of prolonged administration of the seeds of Crotalaria mucronata Desv. to cattle, in order to obtain additional information about this toxicosis. The ground seeds were administered orally to 9 bovines. Doses of 1g/kg/day, 2g/kg/day, 3g/kg/day each in one bovine, and 5g/kg/day in two of three bovines, given for 61- 63 days, did not cause poisoning. Doses of 5g/kg, in one bovine, 7.5g/kg in two bovines and 10g/kg in one bovine, given for 47-61 days, caused symptoms between 47 and 80 days after the first administration and caused death between 3 hours and 5 days after the onset of symptoms. The main clinical signs were positive venous pulse of the jugular vein, abdominal breath, tachycardia, loss of appetite, dry feces, sub-mandibular edema and weakness. Bovines that did not die, were slaughtered 8 or 9 months after first administration. At necropsy pulmonary paleness, hydropericardium, hydrothorax, hydroperitoneum, mesenteric edema, augmented hepatic consistency, discoloration of the liver, right cardiac ventricle dilatation and ruminal wall edema were seen. The main histological lesions were thickening of the alveolar walls and of the arterioles with narrowing of their lumen, and periarteriolar fibrosis, besides hepatic and cardiac lesions of minor importance. It is concluded, that the lesions caused by ingestion of the seeds of C. mucronata over a long period are caused by the difficulties of blood passage through the pulmonar vessels due to fibrosis and thickening of the arteriolar walls through the pneumotoxic action of the plant.

E-mail: claudioslbarros@uol.com.br A spontaneous outbreak of a neurological disease in swine caused by the ingestion of Aeschynomene indica seeds and the reproduction of the disease in the same animal species are reported. The natural outbreak occurred in a pig-raising facility in the central region of the state of Rio Grande do Sul, Brazil. On the premises there were 100 pigs (20 breeding sows and 80 young weaned pigs from several categories) that were fed a ration made by mixing 50% of corn bran, 25% of soybean bran, 5% of a commercial mix of vitamins and minerals, and 20% of broken rice contaminated with 40% of A. indica seeds. Although all pigs apparently ingested the same ration, only 45-day-old pigs were affected; morbidity, mortality and lethality rates were respectively 25%-40%, 8.5%-20%; and 25%-66%. Clinical signs appeared 24 hours after the beginning of feeding of A. indica seeds contaminated ration and included variable degrees of incoordinated gait, falls, sternal recumbency with the hind limbs in a wide base stance, lateral recumbency and death. It was not possible to ascertain how many pigs recovered nor the time frame of recovery. One pig was euthanatized and necropsied in the premises. The poisoning was reproduced in 5 young pigs (A-E) which were fed a ration containing 10% (Pig A), 15% (Pig B) and 20% (Pigs C-E) of A. indica seeds, and in one older pig (Pig F) which was fed a ration with 16.5% of A. indica seeds. Pigs A, B and F were euthanatized and Pigs C-E died of an acute disease respectively 16, 21 and 24 hours after the beginning of the experiment. Clinical signs were similar to those observed in pigs of the spontaneous outbreak. Necropsy findings included marked hyperemia of the encephalic leptomeninges of all pigs; there were large amounts of A. indica seeds in the stomach and reddening of the intestinal wall and bloody intestinal content in Pigs C-E. A hematoma was observed in the lungs of Pig C. The histopathological findings in the brain of pigs fed rations with larger concentrations (20%) of A. indica seeds (C-E) included congestion, edema and hemorrhage and swollen vascular endothelia with focal symmetrical distribution in several brain nuclei and in the telencephalic cortex. In Pigs A and B, and in Pig F, the case which received the lower dosage of the seeds of A. indica, and in the pig from the spontaneous outbreak, histopathological changes in the brain consisted of discrete focal symmetrical areas of malacia in which closely packed Gitter cells and astrocytosis, and capillaries with swollen endothelium obliterated the normal neuropil. The symmetrical malacic foci caused by the ingestion of A. indica seeds in swine affected cerebellar and vestibular nuclei, putamen, and the mesencephalic oculomotor and red nuclei. This indicates that the A. indica seeds ingestion was responsible for the neurological condition, that it may be fatal and seems to affect equally young and older swine. The clinical outcome and pathological changes were dose-dependent, and the brain lesions progressed from damaged blood vessels to vasogenic edema, hemorrhage and malacia.

#8 - Experimental poisoning by the ground seeds of Ricinus communis (Euphorbiaceae) in the rabbit, 17(1):1-7

[Experimental poisoning by the ground seeds of Ricinus communis (Euphorbiaceae) in the rabbit.] Intoxicação experimental pelas sementes trituradas de Ricinus communis (Euphorbiaceae) em coelhos. Pesquisa Veterinária Brasileira 17(1):1-7. Projeto Saúde Animal Embrapa/UFRRJ, Km 47, Seropédica, Rio de Janeiro 23851- 970, Brazil. The ground seeds of Ricinus communis L., given by stomach tube in single doses to rabbits, caused severe symptoms of poisoning, lethal in three rabbits which received the dose of 2 g/ kg and in one of four rabbits which received 1 g/kg; the other three rabbits which received the lower dose, showed slight to moderate symptoms and recovered. Three other rabbits, which received 0.5 g/kg, showed only slight symptoms. The period between administration of the seeds and death or recovery, varied from 12h47min to 68h08min, and from 3 to 6 days, respectively. First clinical symptoms after the administration of the seeds were observed about 8 hours in the lethal cases and in those where the animals showed more than slight symptoms, and about 24 hours in the cases with only slight symptoms. The course of the poisoning varied from 4 to 56 hours in the lethal cases and froin 2 to 5 and half days in the cases with recovery. The clinical signs consisted mainly in digestive distress; the animals showed inappetence or anorexia. Faeces were generally scarce, with bolus altered in form and size, dark, sometimes soft, with mucus. There were manifestations of colic. The most evident post-mortem findings were in the small intestine and cecum, which had liquid contents; its wall was congested and edematous, and fibrine covered the mucosa as pseudomembranes or was found in the lumen as flakes and or filaments. The most important histological changes were seen also in the small intestine and cecum. In the former there was coagulative necrosis associated with congestion/ hemorrhages of the mucosa and submucosa which also showed edema. Similar lesions were seen in the cecum, but thése were less marked, with exception of the edema of the submucosa. In colon and rectum the changes were slight or absent. Necrosis with marked caryorhexis of the macrophages, which migrated to the upper mucosal layer, was seen in the lymphoid follicles of the appendix vermiformis and in one case also of the rudimentary cecum.

As sementes trituradas de Ricinus communis L., administradas em doses únicas por sonda intragástrica a coelhos, causaram graves sintomas de intoxicação com êxito letal nos três coelhos que receberam a dose de 2 g/kg e em um dos quatro que receberam 1 g/kg. Os outros três coelhos que receberam 1g/kg desenvolveram sintomas discretos a moderados e se recuperaram, e os três que receberam 0,5g/kg só mostraram sintomas discretos. Havia nítidas manifestações de cólica. Os achados de necropsia mais evidentes foram constatados no intestino delgado e ceco. O conteúdo destes segmentos era líquido. A parede apresentou congestão e edema e havia fibrina recobrindo a mucosa sob forma de pseudomembranas, ou na luz sob forma de flocos e/ou filamentos. As alterações histológicas mais importantes foram verificadas no intestino delgado e ceco. No intestino delgado observou-se necrose de coagulação associada a congestão/hemorragias na mucosa. Havia ainda congestão/hemorragias e edema na submucosa. Lesões semelhantes foram vistas no ceco onde porém, com exceção do edema da submucosa, eram menos acentuadas. No cólon e reto as lesões eram leves ou ausentes. #9 - Pathological aspects of poisoning by Crotalaria spectabilis (Fabaceae) seeds in swine, 17(1):12-18

[Pathological aspects of poisoning by Crotalaria spectabilis (Fabaceae) seeds in swine.] Aspectos patológicos da intoxicação de suínos por sementes de Crotalaria spectabilis (Fabaceae). Pesquisa Veterinária Brasileira 17(1):12-18. Depto Patologia Animal, Faculdade de Ciências Agrárias e Veterinárias, Unesp-Campus de Jaboticabal, Rodovia Carlos Tonanni Km 5, Jaboticabal, SP 14870-000, Brazil. A feeding experiment was undertaken using 24 weanling pigs, distributed in four treatments, with 6 animals in each group. The animals of group 1, 2, 3 and 4 were fed with rations containing 0.0, 0.2, 0.4 and 0.6% of Crotalaria spectabilis seeds, respectively, during a period of 90 days. Postmorten examination was performed immediately after death in the animals which died during the experiment. The pigs which survived were sacrificed on the last day of the experiment. During the autopsy fragments of liver, kidneys, lungs and stomach were collected for histological examination. The main clinical signs of the intoxication were subcutaneous oedema, affecting particulary the face, limbs and neck, pale ocular and oral mucous membranes, ruffled bristle, emaciation and apathy. Hydropericardium was the most common finding at necropsy, occurring in severa) intoxicated animals. The major microscopic changes were fibrosis, bile ductulus proliferation and megalocytosis in the liver, bronchopneumonia and chronic bronchitis, renal tubular cell megalocytosis, nephrosis and chronic nephritis, as well as gastritis and gastric ulcerations. The experimental findings showed that rations contaminated with C. spectabilis seeds, at the used rate, were toxic to swine.

Foram utilizados 24 suínos desmamados, distribuídos em quatro tratamentos, com seis animais em cada. Os suínos dos tratamentos 1, 2, 3 e 4 receberam, durante 90 dias, ração com respectivamente: 0,0, 0,2, 0,4 e 0,6% de sementes de Crotalaria spectabilis. Os animais que morreram durante o período experimental foram necropsiados logo após a morte. Os sobreviventes foram sacrificados no último dia do experimento. Durante as necropsias, foram colhidos fragmentos do fígado, rim, pulmão e estômago, para realização de exame histopatológico. Os principais sintomas da intoxicação foram edemas subcutâneos, principalmente nos membros, na face e região do pescoço, mucosas oral e ocular pálidas, cerdas eriçadas, caquexia e apatia. O hidropericárdio foi a lesão macroscópica mais comum, acometendo vários animais que receberam a ração contaminada. As principais lesões microscópicas foram fibrose, proliferação de ductos biliares e megalocitose no fígado, broncopneumonia e bronquite crônicas, megalocitose de células epiteliais tubulares renais, nefrose e nefrite crônica, bem como gastrite e ulceração gástrica. Os resultados destes experimentos indicam que ração contaminada com sementes de C. spectabilis, nas proporções utilizadas, foi tóxica para suínos.


#10 - Cross-immunity by the seeds of Abrus precatorius and Ricinus communis in cattle, 17(1):25-35

Abstract in English:

ABSTRACT.- Tokarnia C.H. & Döbereiner J. 1997. [Cross-immunity by the seeds of Abrus precatorius and Ricinus communis in cattle.] Imunidade cruzada pelas sementes de Abrus precatorius e Ricinus communis em bovinos. Pesquisa Veterinária Brasileira 17(1):25-35. Projeto Saúde Animal Embrapa /UFRRJ, Km 47, Seropédica, RJ 23851-970, Brazil. Five bovines immunized against the toxicity of the seeds of Abrus precatorius L. (jequiriti bean) did not get poisoned or only slightly so when the seeds of Ricinus communis L. (castor bean) were given in amounts that in bovines, which never before ingested the seeds of A. precatorius or R. communis, caused moderate to severe poisoning or even death. A sixth bovine, which was not well immunized against the toxic action of the seeds of A. precatorius, was severely poisoned when given a high dose of the seeds of R. communis. On the other side, from five bovines immunized against the toxic action of the seeds of R. communis four were everely poisoned and the fifth one moderately when the seeds of A. prectitorius were given in doses that in bovines, which never before ingested seeds of R. communis or A. precatorius, caused slight to severe poisoning. It is concluded that bovines immunized against the toxic action of the seeds of A. precatorius are resistant to the toxic action of the seeds of R. com munis, but that the contrary is not the case; this is, bovines immunized against the toxic action of the seeds of R. communis were not protected against the poisoning by A. precatorius. Earlier studies by other authors had shown that the toxalbumins of the seeds of A. Precatorius and R. communis, respectively abrin and ricin, are different as to their antigenic properties. A possible explanation for the difference in results can be.that in the present study polygastric animals were used which received the seeds orally, but in the earlier studies monogastric animals received the seeds or toxins by parenteral route. The administration of the fresh leaves or the pericarp of the fruits R. communis to bovines, which had been immunized against the action of the seeds of the plant, had the sarne toxic effect as to not immunized animals, showing that the immunity due to ricin does not give protection against the action of ricinin, the toxic principie of the leaves and the pericarp.

Estudos anteriores por outros autores mostraram que as toxalbuminas de A. precatorius e R. communis, respectivamente abrina e ricina, são diferentes do ponto de vista antigênico. Uma explicação para a divergência desses resultados com os nossos poderia estar no fato de que no presente estudo foram usados poligástricos que receberam as sementes por via oral, enquanto que nos estudos anteriores foram usados monogástricos em que as sementes ou as toxinas foram aplicadas por via parenteral. A administração de folhas frescas ou do pericarpo do fruto de R. communis a bovinos imunizados contra a ação das sementes desta planta tiveram o mesmo efeito tóxico que em animais não imunizados, demonstrando que a imunidade conferida pela ricina não inibe a ação da ricinina, o princípio tóxico das folhas e do pericarpo.
