E-mail: aca2006@ica.ufmg.br This study evaluated the antimicrobial activity and acute or chronic toxicity of the extract of Stryphnodendron adstringens. The stem bark dry extract was obtained by static maceration with ethanol. Quantification of tannins was performed by the Folin-Denis method, which indicated a total tannin content of 32.7%. The antimicrobial activity of the dry extract of S. adstringens was evaluated by agar-based disk diffusion assay with Escherichia coli (ATCC 25922) and Staphylococcus aureus (ATCC 25923) in the concentration of 200, 400 and 600μL/mL. The results indicated that 600μL/mL inhibited microbial growth, i.e. had antimicrobial activity against these species. Acute and chronic toxic effects of S. adstringens was evaluated in Wistar rats treated with 200, 400, 600 and 800mg/kg of extract, administrated by gavage. Liver degeneration was observed in the group of rats receiving 800mg/kg in chronic exposure, what may indicate some degree of toxicity at this concentration. However, no systemic toxicity was observed at lower doses. Considering the broad use of S. adstringens as a phytotherapeutic agent for various human and animal diseases and the livertoxicity observed at high concentrations, attention should be paid to the possible adverse effect of using the extract from this plant at high concentration.

[Toxicidade aguda e crônica e atividade antimicrobiana do extrato de Stryphnodendron adstringens (Mart.) Coville.] Pesquisa Veterinária Brasileira 37(8):840-846. E-mail: aca2006@ica.ufmg.br Avaliou-se neste estudo a atividade antimicrobiana e a toxicidades aguda e crônica do extrato da entrecasca de Stryphnodendron adstringens. A partir do extrato seco, obtido através da maceração estática da casca do caule em etanol de cereais, foi realizada a quantificação de taninos totais (32,7%) pelo método de Folin-Denis. A atividade antimicrobiana do extrato seco extraído de cascas do caule de S. adstringens foi avaliada pela técnica de disco-difusão para os micro-organismos Escherichia coli (ATCC 25922) e Staphylococcus aureus (ATCC 25923) nas concentrações de 200, 400 e 600μL/mL. Os testes de concentração inibitória mínima revelaram que 600μL/mL inibiu o crescimento dos dois micro-organismos testados; o mesmo resultado foi observado para atividade bactericida na concentração de 600μL/mL sobre essas espécies. Efeitos tóxicos sistêmicos agudos e crônicos do extrato seco de S. adstringens administrados por gavagem foram estudados em ratos Wistar, utilizando as doses de 400, 600 e 800mg/kg. Foi observada degeneração hepática no grupo de animais que receberam 800mg/kg tanto no estudo da toxicidade aguda quanto crônica, que pode indicar algum grau de toxicidade de S. adstringens nessa concentração. Considerando o amplo uso de S. adstringens como fitoterápico para humanos e animais, atenção deve ser dispensada para ingestão em altas doses mediante os efeitos tóxicos observados neste estudo.


#2 - Experimental poisoning of rabbits by the leaves and extracts of Thiloa glaucocarpa (Combretaceae)

Abstract in English:

The dried leaves and extracts (tannins) of Thiloa glaucocarpa were given by stomach tube to 28 and 12 rabbits, respectively. In these experiments it was determined that the rabbit is susceptible to poisoning by the leaves of T. glaucocarpa. With a single administration 9 of 22 rabbits died; one rabbit was sacrificed at the end of the experiment. In experiments with repeated administrations, four of six rabbits died, two others being sacrified at the end of the experiment. In all experiments in which the rabbits died, the course of the poisoning was acute (maximum 33 hrs). The clinical-pathological picture of experiments with single or repeated administrations, showed changes predominantly in the gastrointestinal tract and the liver. Mild lesions were also seen in the kidney. It waalso verified that the rabbit is susceptible to poisoning by the extracts (tannins) obtained from the leaves of T. glaucocarpa. With a single administration three of five rabbits died, and with repeated administrations, four of seven rabbits died; three rabbits were sacrificed when the experiment was terminated. In all experiments with a single administration, ending with the death of the animals, the course of the poisoning was acute (maximum 24 hrs). In experiments with repeated administrations, ending with the death of the animals, the course was acute (12h) in one experiment, and longer (3 and a half to 6 days) in three others. In these experiments with single or repeated administrations, liver lesions predominated, with very slight renal lesions: repeated administrations also produced lesions of the digestive tract. It is concluded from these experiments, that tannins have a noxious action similar to that of the leaves when given to rabbits, and that tannins constitute the toxic principie of T. glaucocarpa, in regard to the rabbit. It is also concluded, that it can not be confirmed that these tannins are responsible for the clinical-pathological picture of poisoning by T. glaucocarpa in bovines, where lesions of the kidneys and the digestive tract predominate, with some hepatic lesions also being found. Experiments with other plants suggest that this hypothesis is correct, for instance, in the poisoning by Vernonia nwllissima, depeding on the animal species, the course of the poisoning and the dose given influence whether tiver or renal lesions predominate.

As folhas dessecadas e extratos (taninos) de Thiloa glaucocarpa Eichl. foram administrados por sonda gástrica a 28 e 12 coelhos, respectivamente. Nesses experin1entos foi verificado que o coelho é sensível à intoxicação pelas folhas de T. glaucocarpa. Com administrações únicas morreram 9 dos 22 coelhos; adicionalmente, um coelho foi sacrificado após o experimento; com administrações repetidas morreram 4 dos 6 coelhos; 2 outros foram sacrificados. Em todos os experimentos que terminaram com êxito letal, a evolução da intoxicação foi aguda (no máximo 33 horas). Tanto com as administrações únicas como as repetidas, predominavam, no quadro clínico-patológico, alterações relativas ao tubo digestivo e ao fígado e havia, em grau leve, lesões renais. Foi verificado que o coelho é também sensível à intoxicação pelos extratos (taninos) obtidos das folhas de T. glaucocarpa. Com administrações únicas morreram 3 dos 5, e com administrações repetidas, 4 dos 7 coelhos; 3 outros foram sacrificados após o experimento. Em todos os experimentos com administrações únicas e êxito letal, a evolução da intoxicação foi aguda (no máximo 24 h); nos de administrações repetidas com êxito letal, em um caso foi aguda (de 12 h) e nos outros 3 casos variou de 3 e meio a 6 dias. Tanto nas administrações únicas como nas repetidas, predominavam alterações hepáticas e havia, em grau muito leve, lesões renais; com administrações repetidas havia, ainda, alterações do tubo digestivo. Concluiu-se, por esses experimentos, que os taninos possuem ação nociva semelhante à das folhas quando administrados a coelhos, e que, portanto, taninos constituem o princípio tóxico de T. glaucocarpa em relação ao coelho. Concluiu-se, também, que não se pode afirmar que esses taninos sejam responsáveis pelo quadro clínico-patológico da intoxicação por Thiloa glaucocarpa nos bovinos, em que predominam alterações renais e do tubo digestivo, mas em que há também lesões hepáticas. No entanto, experimentos com outras plantas sugerem que essa hipótese seja verdadeira, visto que, na intoxicação por Vernonia molíssima Don, por exemplo dependendo da espécie animal, da evolução da intoxicação e da dose empregada, podem-se obter quadros predominantemente hepatotóxicos ou predominante nefrotóxicos.
