Abstract in English:

Rodrigues A., Fighera R.A., Souza T.M., Schild A.L., Soares M.P., Milano J. & Barros C.S.L. 2005. [Outbreaks of trypanosomiasis in horses by Trypanosoma evansi in the state of Rio Grande do Sul, Brazil: epidemiological, clinical, hematological, and pathological aspects.] E-mail: claudioslbarros@uol.com.br Cases of trypanosomiasis by Trypanosoma evansi were diagnosed in horses in the state of Rio Grande do Sul, Brazil, between 2003 and 2004. In one stud farm (Farm A) with 125 horses, 52 died. Additionally, around 80 mares were sent to Farm A to be bred. Of those, 66 became ill and 56 died after being returned to their farms of origin. Twenty one horses clinically affected by the disease were observed. Clinical signs included loss of weight (despite voracious appetite), lethargy, incoordination and instability of hindlimbs, atrophy of the large muscles of the hindlimbs, muscle weakness and paleness of mucosae. Specimens of T. evansi were detected in the blood drawn from four affected horses. Normocytic normochromic anemia with PCVs ranging from 15 to 31%, leucocytosis due to lymphocytosis associated to large atypical lymphocytes was observed in several affected horses. High levels of antibodies against T. evansi were detected in the serum of six horses from Farm A. Eight horses presented encephalic neurological signs such as circling, ataxia, blindness, excitation, falls, listlessness, proprioception deficits and head tilt. One horse assumed a “dog-seating position”. Necropsy findings included muscle atrophy, enlargement and lymphoid hyperplasia of the spleen and lymphnodes, edema and softening of the white and grey matter of the brain. Histologically, an overwhelming necrotizing panencephalitis was observed in the seven horses with encephalic signs. This panencephalitis was characterized by marked edema, demyelination and necrosis and perivascular infiltrates of 6-10 layers of lymphocytes and plasm cells affecting both the white and gray matter. Several plasm cells in the inflammatory infiltrate contained numerous eosinophilic globules in their cytoplasm (Mott cells). Similar histological lesions were observed in the spinal cord of the horse with the “dog-seating position”. The brains of five horses with the encephalic signs were submitted to immunohistochemistry stain by the streptavidin-biotin technique. In all of those five brains moderate to abundant specimens of T. evansi in the perivascular spaces and neuropile were marked by the specific antibody. Epidemiological, clinical, hematological, and pathological aspects of equine trypanosomiasis caused by T. evansi are discussed.

#113 - Antimicrobial resistance and R-plasmid in Salmonella spp from swine and abattoir environments

Abstract in English:

Lázaro N.S., Tibana A., Rodrigues D.P., Reis E.M.F., Quintaes B.R. & Hofer E. 2004. Antimicrobial resistance and R-plasmid in Salmonella spp from swine and abattoir environments. Pesquisa Veterinária Brasileira 24(2):57-60. Depto Epidemiologia e Saúde Pública, Instituto de Veterinária, UFRRJ, Seropédica, RJ 23890-000, Brazil. E-mail: nslazaro@aol.com Salmonella serovars isolated from swine are of particular interest not only because of the pathogenic potential for this animal species, but also due to its relevance with regard to public health. On basis of the profile of resistance to antimicrobials, 13 Salmonella strains were selected which belonged to the serovars Muenster (7), Derby (4), Typhimurium (1), and Braenderup (1). They were isolated from healthy swine as well as from the abattoir environment in the state of Rio de Janeiro. All strains of Salmonella were subjected to bacterial conjugation, and the E. coli K12 Nalr Lac+ F standard strain was used as receptor, with the purpose to verify the ability to transfer the resistance marks. Gene transfer phenomenon was detected in seven strains, and except Salmonella Typhimurium which transconjugated to Sm, Tc and Su, the remaining ones were characterized by transferring mark Su only. By plasmidial analysis of strains used and their respective transconjugants, 63 Kb plasmid was found, which was probably related to S. Typhimurium resistance.

E-mail: nslazaro@aol.com Thirty-eight strains of Salmonella Muenster, isolated from swine and the abattoir environment, in the State of Rio de Janeiro, Brazil, from March 1991 to February 1992, were analyzed for the presence of plasmids. The strains were selected according to their profile regarding the antimicrobials: streptomycin, tetracycline, sulphonamide and sulfametoxazole-trimethoprim. Thirteen strains were resistant to one or several antimicrobials, 18 with intermediate degree and seven were sensitive. Plasmids varying in size from 1.2 Kb to 42 Kb were detected in 37 (97.36%) of the 38 samples, corresponding to 11 different profiles (P1- P11), varying from 1 to 6 plasmids per model. The number and plasmids diversity was greater than the resistance marks for strains. The plasmid of 2.85 Kb was the most frequent, being present in 83.78% of the 37 strains; only the one of 7.5 Kb was detected at the two slaughterhouses. There was no parallelism between resistance pattern and plasmidial profile, and a same antibiotype was found in several plasmidial profiles. The results of the present investigation, allowed us to conclude that the plasmid characterization is an useful and simple tool for the epidemiological typing of this sorovar.

#115 - LD50 in fish and rat produced by powdered roots of Derris spp and ecotoxicological implications, 18(2):53-56

Abstract in English:

ABSTRACT.- Mascaro U.C.P., Rodrigues L.A., Bastos J.K., Santos E. & Costa J.P.C. 1998. [LD50 in fish and rat produced by powdered roots of Derris spp and ecotoxicological implications.] True "timbós" from the Brazilian Amazon region, plants belonging to the genus Derris of the Leguminosae family, have there considerable importance, because they contain rotenoids which are toxic to fish and mammals. The aim of the study was to establish the lethal doses (LD50) for three phylogenetical different species of fish and also for the rat. The lethal dose for Collosoma macropomum (tambaqui) was 2.6 mg/ml, for Oreochromis niloticus (tilápia) 4.8 mg/ml, for Plecostomus sp (cascudo) 14.2 mg/ml and for Rattus norvegicus 100 mg/kg. The large differences between the lethal doses for the fish and the one for the rat are possibly due to different barrier tissues against the action of rotenoids in the mammal when given orally.

RESUMO.- Mascaro U.C.P., Rodrigues L.A., Bastos J.K., Santos E. & Costa J.P.C. 1998. [LD50 in fish and rat produced by powdered roots of Derris spp and ecotoxicological implications.]

#116 - Effect of a management system on grazing behaviour, ponderal growth and parasitie infestation of Suffolk ewes

Abstract in English:

Grazing behaviour, ponderal growth and level of parasitic infestation were studied in Suffolk breed sheep, from 1994 to 1995, in Nova Odessa, São Paulo. Two management systems were compared: restricted grazing, where the animals were released to the pastures at 9:50am and confined at 5:30pm, and 24 hour grazing, where the animals were maintained all the time in the paddocks, but with access to shelters. Thirty-four adult ewes were used in the summer period (17 under restricted grazing and 17 full-time grazing), and 42 ewes in the winter period (21 with restricted grazing and 21 full-time grazing). Also, for both seasons, 12 traceranimals, six in each grazing system, were used to obtain worm counts from their digestive tracts. For three consecutive days, grazing behaviour, that is, whether grazing or not, was observed in January/February (summer) and July/August (winter) at 30 minute intervals from 7:00am to 5:30pm, also whether the animals stayed in the sun or shade, irrespective of whether they were grazing or not. The level of parasitic infestation was evaluated under each system by eggs per gram countings (EPG) of the herd and the traceranimals, as well as by larvae count. It was concluded that restriction of grazing time by itself does not provide any effective control of parasitic infestation in sheep, however a better control was obtained in the summer period. Restricted grazing time was compensated by the greater activity of the animals during the hotter hours of the day, however, this behaviour affected the animal performance, resulting in lower weight gains. Greater forage availability in relation to estimated consumption may explain the similarity between the grazing times observed in both management systems, either in the summer or in the winter.

#117 - Factors which influence the toxicity of Baccharis coridifolia (Compositae): an experimental study in rabbits

Abstract in English:

Experimental studies were performed on Baccharis coridifolia D.C. (Compositae), a small shrub with the popular name "mio-mio", one of the more important poisonous plants of southern Brazil, Uruguay, Argentina and Paraguay, in arder to find out possible variations in toxicity due to its origin, sex, plant parts and annual variability. Doses which varied from 0.085 to 5.44 g/kg of the dried powdered aerial parts of B. coridifolia were administered in aqueous suspension by stomach tube to 49 adult rabbits. The plant material was collected in five different localities, from four sites in Brazil and one in Uruguay, in the flowering/seeding stage and separated by sex; in some regions collections were made in two successive years. From three of the sites additionally only the leaves of female plants were used for the experiments. Sixteen animals died and one that showed severe symptoms was euthanized together with the contral animal. Nineteen animals that showed slight to severe symptoms, recovered. First symptoms after the administration of the plant were seen, in the fatal cases from 6h32min to 1 day 6h15min, and in the non-fatal cases from 4h03min to 24h00min, after ingestion. The clinical course, in the fatal cases, varied from approximately 6h00min to 4 days 20h23min and, in the non-fatal cases, from 9h25min to 11 days 3h45min. The length of time from first administration of the plant till death varied from 14h20min to 5 clays 2h55min. The lethal dose varied from 0.17 to 5.44 g/kg. The main symptoms were anorexia, modifications in the form, size and consistency of the faeces and hair loss. The main post-mortem alterations were found in the stomach, caecum and liver. There was red colouration of the stomach mucosa. ln the caecum the surface of the mucosa was rough and red and the wall was oedematous. The liver was clearer than normal with perceptible lobulation. Histopathologically these lesions correspond to 1) coagulative necrosis associated with haemorrhages and oedema of the stomach mucosa, 2) haemorrhagic necrosis of the mucosa and oedema and/or haemorrhages of the submucosa of the caecum and 3) cloudy swelling and/or vacuolization of the liver. Greater toxicity of B. coridifolia was associated with more southerly sites of collection in Brazil. The female plant was up to 32 times more poisonous than the male plant; the whole aerial pa1ts of the plant were up to 16 times more poisonous when compared with only the leaves. There was up to a fourfold variation in toxicity from year to year. These differences can be explained by variations in the type and the amount of macrocyclic trichothecenes accumulated in the different parts of the plant, which are believed to be absorbed from the soil fungus Myrothecium verrucaria which produces them. B. coridifolia was compared with B. megapotamica var. weirii, which does not show these variations.

#118 - Chronic copper poisoning in sheep grazed in an apple orchard

Abstract in English:

Cases of chronic copper poisoning (CCP) were observed in a flock of 120 Ile de France ewes grazed on white clover ( Trifolium repens) pasture in an apple orchard. The orchard was sprayed with copper sulphate 4 weeks after the ewes were introduced. A month later some ewes showed depression, jaundice and hemoglobinuria. Twenty one ewes died representing 17.5 % of the flock. The clinical signs, necropsy and microscopic findings were typical of CCP. The sernm levels of aspartate aminotransferase (AST) in 11 ewes varied from 159 to 510 iu/1, anda sample from the pasture revealed 60 mg/kg of copper. High copper levels of 1313 and 300 mg/kg, respectively, were found in the liver and kidney collected from one of the two sheep on which post-mortem examination was performed. It is suggested that the poisoning was caused by the high level of copper in the pasture, together with the higher susceptibility of mutton sheep to copper poisoning.

#119 - Experimental intoxication in pigs fed with low concentrations of Senna occidentalis (Leg. Caes.) in the ration

Abstract in English:

Thirty pigs with mean weight of 27.5 kg were divided at random in tive groups. Group 1 (control) was fed with a growing ration from day O to day 42, and with a finishing ration from day 43 to day 90. Groups 2, 3, 4 and 5 received the sarne ration as the control group plus 0.25%, 0.50%, 0.75% and 1.00% respectively of ground Senna occidentJJljs (L.) Link. seeds. The regression analysis showed a negative linear regression of gowth rates Ŷ = 0.75 - 0.36x, r2 =0,92, (P<0,01)) and food consumption (Ŷ = 1,98 - 0.82x, r2 = 0,93, (P<0.07)) of growing and finishing pigs in relation with the concentration of S. occidentalis in the food. Four pigs of group 5 and one of group 3 had clinical sigos at 54, 59, 78 (two pigs) and 81 days after the beginning of the experiment. These pigs were killed in extremis until 72 hours after the onset of clinical sigos. Three pigs from group 5 and three from group 4 were killed 56 and 57 days after the beginning of the experiment. The other pigs, except three from the control group, were killed in extremis at the end of the experiment. Post mortem findings were enlargment of the liver, which had pale areas of discoloration and accentuation of its lobular pattern, in 9 pigs from groups 3, 4 and 5, one of which had a nutmeg appearance on the cut surface. No gross lesions were observed in skeletal muscles or myocardium in any of the pigs. Histological lesions of the liver were characterized by hepatocellular fat metamorphosis and discrete bile duct proliferation. One pig had centrilobular necrosis probably resultant from cardiac failure, Degeneration and segmentai necrosis of fibers were observed in skeletal muscles. The degree of these lesions was correlated directly with the quantity of S. occidentalis seeds in the ration. Mild degenerative lesions were observed in the myocardium of some pigs from groups 3, 4 and 5.

#120 - Occurrence of antibodies for avian viruses in broilers in a region of high density poultry production

Abstract in English:

Blood samples were obtained at slaughter from sixty 43 to 56 day old broilers, from each of 60 healthy flocks located in a region of high density poultry production. The flocks tested corresponded to approximately 10% of the total number existing in the region and were located within a radius of 50 km. All broilers had been vaccinated against Marek's disease atone day of age. Sera were tested in the immunodiffusion test for antibodies to avian reovirus (ARV), infectious bursal disease vírus (IBDV), fowl poxvirus (FPV), herpes virus of turkeys (HVT), avian adenovirus of groups 1 (AAV-1) and 2 (AAV-2). The sarne sera were tested in the micro serum neutralization test in plates for antibodies to infectious bronchitis virus (IBV) and infectious laryngotracheitis virus (ILTV), and for hemagglutination inhibition antibodies to Newcastle disease virus (NDV). Antibodies to ARV, detected in 2172 (63.6%) of 3418 sera, and to AAV-1, in 2701 (78.2%) of 3456 sera, were found in all 60 flocks, while antibodies to IBDV, in 3086 (89.4%) of 3452 sera, were found in only 57 of the sarne flocks. These results indicated that the three viruses are ubiquitous in broilers. Antibodies to FPV in 14 (0.5%) of 2935 sera, and to NDV in 6 (0.2%) of 3320 sera, were demonstrated in five of 59 and in one of 60 flocks, respectively, indicating that these two viruses occur only rarely. Antibodies to HVT, in 315 (9.0%) of 3384 sera, were detected in 39 of 60 flocks tested, suggesting poor serological response to vaccination and low field exposure to MDV. Antibodies to IBV, in 61 (2.1 %) of 2925 sera, were found in 12 of 57 flocks tested. However, in only one flock was the percentage of reactors significant (41 %); in the 11 flocks the percentage of broilers with antibody ranged from 1.7 to 16.1 %. Finally, no antibodies were found in 3491 sera from 60 flocks tested for AAV-2, nor in 3035 sera from 59 flocks when tested for ILTV, indicating that these two viruses do not exist in the geographical area tested. The significance of these findings is discussed.
