#132 - Detection and dynamics of porcine circovirus type 2 in semen using conventional and real time PCR, 30(11):918-920

Abstract in English:

ABSTRACT.- Gerber P.F., Pinto F.F., Heinemann M.B. & Lobato Z.I.P. 2010. Detection and dynamics of porcine circovirus type 2 in semen using conventional and real time PCR. Pesquisa Veterinária Brasileira 30(11):918-920. Laboratório de Pesquisa em Virologia Animal, Departamento de Medicina Veterinária Preventiva, Universidade Federal de Minas Gerais, Avenida. Antônio Carlos 6627, Belo Horizonte, MG 30123-970, Brazil. E-mail: ziplobato@vet.ufmg.br The dynamics of porcine circovirus type 2 (PCV2) shedding in semen of naturally infected boars was studied. Semen was collected serially each 15 or 20 days during 62 days from 5 boars from a herd and from 11 boars from an artificial insemination center. All boars were positive for PCV2 DNA by nested polymerase chain reaction of raw semen in at least two sampling dates, and most of them had detectable shedding in all sampling dates. Real-time quantitative PCR was performed in 23 samples. All samples showed low amounts of PCV2 DNA, ranging from 98 to 652 PCV2 copies/mL. No differences between the frequencies of PCV2 DNA shed in semen were found considering herds and age of boars. PCV2 shedding in the semen can occur continuously or intermittently up to 60 days in naturally infected boars at 12 to 42 months old in absence of PCV2 clinical signs. These results demonstrate sporadic and long-term shedding patterns of low amounts of PCV2 DNA in semen from naturally infected boars.

#134 - Diversity of porcine rotavirus genotypes in São Paulo State, Brazil, 29(9):707-712

Abstract in English:

ABSTRACT.- Gregori F., Rosales C.A.R., Brandão P.E., Soares R.M. & Jerez J.A. 2009. [Diversity of porcine rotavirus genotypes in São Paulo State, Brazil.] Diversidade genotípica de rotavírus suínos no Estado de São Paulo. Pesquisa Veterinária Brasileira 29(9):707-712. Centro de Pesquisa e Desenvolvimento em Sanidade Animal, Instituto Biológico, Av. Cons. Rodrigues Alves 1252, São Paulo, SP 04014-002, Brazil. E-mail: fabiogregori@biologico.sp.gov.br Rotavirus is one the most common causes of diarrhea both in humans and different animal species. It was carried out a transversal study with 144 diarrheic fecal samples of piglets, from 16 commercial swine-producing units distributed among 10 municipalities of São Paulo State, Brazil, aiming at the detection of rotavirus occurrence and its molecular characterization according to G and P genotypes. A total of 43 samples (29.86%) were positive for rotavirus by Polyacrylamide Gel Electrophoresis (PAGE) and ELISA, in a parallel screening scheme. The nested-multiplex RT-PCR characterization revealed that, separately, the P[6] genotype was the most frequent, detected in 25.58% of the samples, followed by P[1] (11.63%) and P[7] (9.3%). Concomitant infection of the genotypes P[6]+P[7] (9.3%), P[1]+P[6] (4.65%), P[1]+P[6]+P[7] (2.33%) were also found. Similarly, the G[5] genotype was detected on 30.23% of the samples, followed by G[10] (20.93%), G[6] (4.65%) and G[5]+G[10] (18.6%). The genotype G[5]P[6] was the most frequent (11.63%), but other combinations and untypeable samples were also observed. Considering the diversity porcine rotavirus found in the surveyed population, specific prophylactic measures should take in charge, for its effectiveness, the cross-protection degree between the genotypes present on vaccine formulations and those that really circulates on a region.

Abstract in English:

ABSTRACT.- Asanome W., Koller F.L., Nottar E., Carregaro F.B., Corrêa A.M.R., Zlotowski P., Driemeier D. & Barcellos D.E.S.N. 2008. [Association between bacterial otitis and porcine circovirus 2 (PCV2) infection in pigs.] E-mail: davidbarcellos@terra.com.br The occurrence and bacteriology of purulent otitis in groups of pigs affected by three different clinical conditions are described: postweaning multisystemic wasting syndrome (PMWS), depressed growth (attrition) and normal growth (control). A total of 385 animals, 60-130 days of age, were examined. Diagnosis of PMWS was confirmed by the presence of clinical signs and pathological findings compatible with the disease, and through detection of viral antigens in tissues by immunohistochemistry. From 242 pigs with PMWS, 57 (23.5%) showed purulent lesions in the middle ear. Among 119 pigs with attrition, only 1 (0.7%) presented the lesion. In 24 control pigs, middle ear lesions were not detected. The most frequently isolated agents from the lesions were Arcanobacterium pyogenes (43%), á–hemolytic Streptococci (37.2%) and Pasteurella multocida (27.9 %) out of 86 materials. The frequent occurrence of purulent lesions in the middle ear of PMWS affected pigs suggests that PCV2 infection may increase the susceptibility to bacterial otitis. The low occurrence of this disease in piglets with attrition suggests that otitis does not represent a significant cause for depressed growth in pigs from growing and finishing age. The isolation of A. pyogenes, á-hemolytic Streptococci and P. multocida from most of the lesions emphasizes the importance of these organisms as causal agents of otitis media in swine.

E-mail: ticianafranca@terra.com.br Outbreaks of infection of swine with Lawsonia intracelullaris in the state of Rio de Janeiro are described. The symptomathology was characterized by bloody diarrhea with peracute course. Postmortem findings were pale organs and tissues, reticulate aspect of the ileum with its mucosa moderately thickened nad contained large amounts of partially coagulated blood. Histopathological examination revealed hyperplastic epithelial cell proliferation of the Lieberkühn crypts, sometimes associated with inflammatory and necrotic alterations. Immunohistochemistry demonstrated the presence of large amounts of bacteria in the cytoplasm of the cryptal epithelial cells. L. intracellularis was also seen by ultramicroscopic examination. These outbreaks occurred in 1987 and this is the first report of the disease in the state of Rio de Janeiro.

Laboratório de Anatomia da União Educacional do Planalto Central, Campus II, Área Especial nº 2, Setor Leste, Gama, DF 72460-000, Brazil. E-mail: crisruiz@scamilo.edu.br The experimental use of coronary arteries of swine to study drug action for clinical and surgical application is an important tool. For this anatomic study about coronary arteries, thirty hearts of Landrace pigs, of both sexes, 5 to 6 months old, weighing 80 to 110 kg, were fixed in 10% formalin. The coronary arteries and their branches were dissected until the visible ramifications. There was one left coronary artery, with length of 0.4-1.2cm, giving off 2 (80%) or 3 (20%) branches. The paraconal interventricular branch, with length of 10-16cm, emitted 16-25 branches, 52.3% for the right ventricle and 47.7% for the left ventricle. The circumflex branch, with length of 7-15cm, emitted 4-13 branches, 55.6% for the left ventricle and 44.4% for the left atrium. There was one right coronary artery, with length of 7.5-11.5cm, which emitted 12-21 branches, 57.4% for the right ventricle and 42.6% for the right atrium. The subsinuosus interventricular branch, length of 3-11cm, emitted 9-22 branches, 50.9% for the right ventricle and 49.1% for the left ventricle. The number of branches of the paraconal interventricular branch and the number of branches of the subsinuosus interventricular branch was similar for both ventricles. The results obtained in the present study compared with the literature indicate that the distribution of the coronary branches in swine and in humans is similar.

Departamento de Cirurgia, Setor de Anatomia dos Animais Domésticos e Silvestres, Faculdade de Medicina Veterinária e Zootecnia, Universidade de São Paulo, Av. Prof. Dr. Orlando Marques de Paiva 87, Cidade Universitária, São Paulo, SP 05508-000, Brazil. E-mail: avidotti@usp.br The sinoatrial node, for being topographically installed as the initial component of the conduction system, is responsible for the production of the nervous impulses, which determines the cardiac contraction. There have been made studies related to the node’s morphology in order to know the origin, distribution and quantification of the vases in this tissue, however, in spite of the results and quantitative data of the nodal irrigation - arterial vascular conduct and arterial vascular density, at the nodal level - the literature is scarce. With this objective 27 SRD swine hearts, injected with colored resin for macroscopic analysis of the origin and distribution of ANSA (sinoatrial node artery), 3 others injected with watery solution of colloidal coal (dyed nanquim) were used, to mark the route of the vases on the node level for stereological analysis. The atrial arteries originated as well from the right coronary artery as from the left one, with predominance of the first (66.66% and 33.33%, respectively). When originated from the right coronary artery, there existed the following branches: AADAM (right cranial medial atrial artery) in 14 cases, AADAI (right cranial intermedial atrial artery) in 2 cases, and AADAL (right cranial lateral atrial artery) in 2 cases. In 9 cases (33.33%) the following branches originated from the left coronary artery: 4 through branch AASPL (left caudal lateral atrial artery), 2 through branch AASAI (left cranial intermedial atrial artery) and 3 through branch AASAM (left cranial medial atrial artery). Interarterial anastomoses with vessels responsible for irrigation of the sinoatrial node area were observed in most of the cases (25 hearts). The Volume or Reference Volume (V(ref)) of the hearts were 35.32x104µm3. For the stereological variables analyzed, the estimate density of the vessel length (Lv) was 766; the vessel length (L) - mm - was 27.06x105µm; the area’s surface density (Sv) was 182 and the area’s surface (S) - mm2 - was 64.3x106µm2. The estimate of the vascular numeric density (Nv(vasc)) - total of vessels per unit of volume (cm3) was 2.19 10-5, and the total number of vessels in the organ (N(vasc)), estimated by the physical dissector method in combination with the estimate of the number of Euler (Xv), was 773.6832 x10-2. The raised vascular density and total number of vases in the swine’s sinoatrial node area suggests the existence of a vascular perinodal network complex and dense, ratifying the importance of this mark-pass for its blood supply.

#139 - Toxoplasma gondii: estudo soro-epidemiológico de suínos da região Sudoeste do Estado do Paraná, p.15-18

Abstract in English:

ABSTRACT.- Millar P.R., Daguer H., Vicente R.T., Costa T., Sobreiro L.G. & Amendoeira M.R.R. 2008. [Toxoplasma gondii: epidemiological study of pigs from southwestern Paraná, Brazil.] E-mail: patriciariddell@vm.uff.br This study evaluated the seroprevalence of toxoplasmosis in pigs bred and slaughtered for human consumption. The frequency of the antibody anti-Toxoplasma gondii was investigated in 408 animals from distinct piggeries of 25 cities in the southwestern region of the state of Paraná, Brazil. Serum samples were examined for IgG antibodies specific for T. gondii through the indirect fluorescent antibody test (IFAT). A frequency of 25.5% of serum samples tested positive for dilutions equal to or greater than 1:64. Statistically significant were not observed in relation to sex and age of the animals. The highest antibody level observed was 4,096 (3.0%) and the most frequent was 64 (74%), suggesting that a chronic infection occurred in the population. The high prevalence of the infection found in the swine herds of this region suggests that these herds can be a source of infection for individuals who typically consume this type of raw or badly cooked meat.

Abstract in English:

ABSTRACT.- Silva F.O.C., Rafael E.L.S., Severino R.S., Drummond S.S. & Bombonato P.P. 2007. [Arteries of the cervical wolves of the thymus in stillborns hogs of the lineage C40.] E-mail: eltonvete@yahoo.com.br The thymus is a lymphoid structure of great importance for young animals, excellently acting on the after-birth development and maintenance of their immunologic ability. It has been shown well-developed from the final phase of the prenatal period through the beginning of puberty, and is the largest lymphoid structure with high lymphopoietic function during that period. The objective was to study the arterial vascularization of the cervical part of the thymus of C40 pigs, to supply morphologic information for this structure in animals that are of economical importance in Brazil, and to furnish subsidies for comparative anatomy. Thirty pig embryos, males and females, were used originating from cases of abortion and natural death. Both sides of the cervical lobes of the thymus were found divided during the fetal phase into cranial and caudal portions, joined by an isthmus and situated along the medial face of the mandibulary glands up to the cranial edge of the first pair of ribs. Characteristic for each side of the thymus vascularization is that the cervical lobes of the left side were found irrigated by Arteria subclavia, A. carotida communis, A. thyreoidea caudalis, A. thyreoidea cranialis, A. thyreoidea cervicalis superficialis, A. laryngea caudalis, and A. laryngea cranialis. The cervical thymus lobes of the right side were irrigated by A. carotida communis, A. thyreoidea caudalis, A. thyreoidea cranialis, A. thyreoidea cervicalis superficialis, A. laryngea caudalis, and A. laryngea cranialis.
