#1881 - Technological development of (40mg) furosemide tablets: a equivalence and bioavaibility study in dogs, p.462-466

E-mail: onaldo@cstr.ufcg.edu.br Furosemide (40mg) was administered to 20 street dogs, 10 males and 10 females, in two different pharmaceutical forms: (1) compressed furosemide 40mg formulated at the Federal University of Pernambuco (UFPE-tablet), and (2) a commercial formulation with equal bioequivalence produced by the Laboratory for Pharmaceutical Technology of Pernambuco State (LAFEPE), the LAFEPE-furosemide. The study aimed to evaluate the kinetics of dissolution of the UFPE-tablet in order to analyze the behavior of bioavailability of the best formulation for veterinary use. The plasmatic concentrations of furosemide for the determination of parameters of pharmacological kinetics were analyzed by high-performance liquid chromato-graphic method (HPLC). The in vitro study accomplished through physiochemical analyses demonstrated that the formulas of the furosemide tablets attained the pharmaceutical requirements in agreement with USP 23 and the Brazilian Pharmacopoeia. The evaluation accomplished in dogs with UFPE-tablets given in only dose demonstrated uniformity in blood levels indicating stability in maintenance of the pharmaceutical formulation and efficiency in absorption of the active compound. These values are not significantly different in relation to the 5% confidence limit. Regarding maximum concentration (Tmax) time and global bioavaibility assessed by AUC means, there were no considerable differences as well. UFPE-furosemide displayed 743.492µg/mL.h as AUC average value whereas LAFEPE-furosemide had an average of 537.284µg/mL.h.

It is necessary to identify very early any eye disease that could cause the failure of cataract surgery. The ocular ultrasonography or echography is a safe, non invasive, inexpensive and easy procedure that complements ocular examination. Two hundred and twenty five eyes of 123 dogs were evaluated using a 10 Mhz linear transducer ultrasound. The dogs were divided into three groups, as there is Group 1: 36 healthy control dogs; Group 2: 52 non diabetic dogs with cataracts; and Group 3: 35 diabetic dogs with cataracts; all of them were patients of the Veterinary Teaching Hospital, University of Sao Paulo. The ultrasonographic findings were in Group 1: vitreous degeneration in 43%, posterior vitreous detachment in 7.7%, asteroid hyalosis in 4.6% of the eyes; in Group 2: vitreous degeneration in 58.9%, posterior vitreous detachment in 8.4%, asteroid hyalosis in 2.1%, inflammatory or haemorrhagic process in 2.1%, retinal detachment in 4.3% of the eyes; and in Group 3: vitreous degeneration in 50.7%, posterior vitreous detachment in 13.8%, asteroid hyalosis in 12.3%, inflammatory or haemorrhagic process in 3.1%, retinal detachment in 3.1% of the eyes. It could be concluded that (1) diabetic dogs, mainly the ones with diabetes for more than 2 years, have greater chances to develop asteroid hyalosis; (2) there were no significant differences in retinal detachment frequencies for the diabetic and non diabetic dogs; and (3) the ocular ultrasonography is a safe, effective and important exam for the pre-operatory evaluation of dogs with cataracts.

In the Northern hemisphere, where the disease is classically recognized, bovine coronavirus (BCoV) has been assigned as a major etiologic agent of the disease. Nonetheless, in the Southern hemisphere, an in-deep etiological survey on WD cases had not been carried out. This study aimed to survey for BCoV by nested-RT-PCR, rotavirus by polyacrylamide gel electrophoresis (PAGE) and ELISA, bacteria by classical bacteriological methods and PCR for virulence factors and parasites by sugar flotation test on fecal samples of 21 cows from a farm during an outbreak of WD in São Paulo state, Southeastern Brazil. BCoV was detected in all 21 samples, while rotavirus was detected in two symptomatic cows. Escherichia coli, Yersinia intermedia, Providencia rustigianii Proteus penneri, Klebsiella terrigena and Enterobacter aglomerans were detected in samples from both asymptomatic and healthy cows in different associations. The study of E. coli virulence factors revealed that the strains isolated were all apathogenic. Cysts of Eimeria sp. and eggs of Strongyloidea were detected at low numbers in four of the symptomatic cows, with one co-infestation. These results suggest BCoV as the main etiologic agent of the cases of WD in Brazil, a conclusion that, with the clinical and epidemiological patterns of the disease studied herein, match those already described elsewhere. These findings give basis to the development of preventive measures and contribute to the understanding of the etiology of WD.

As BoHV-1 has been historically associated with respiratory and genital disease, herpesviruses isolated from these clinical syndromes have been tentatively – and sometimes definitively - diagnosed as BoHV-1. Likewise, cases of herpetic neurological infection in cattle have been generally attributed to BoHV-5. This study reports the identification of 40 herpesvirus isolates from different clinical specimens and syndromes in central-southern Brazil, Argentina and Uruguay (1987-2006) by the use of a PCR able to differentiate between BoHV-1 and BoHV-5. BoHV-1 isolates (n=16) were identified in cases of respiratory disease (n=3), vulvovaginitis and/or balanoposthitis (n=3), in semen of healthy bulls (n=5) and in cases of neurological disease (n=5). Viruses identified as BoHV-5 (n=24) were isolated predominantly from cases of neurological disease (n=21), but also from semen of healthy bulls (n=2) and from a spleen of a calf with systemic disease (n=1). These results show that both BoHV-1 and BoHV-5 are not strictly associated with their respective diseases; yet are frequently involved in clinical conditions otherwise attributed to the other virus. These findings also reinforce the need of correctly identifying the herpesvirus isolates as to better understand their pathogenesis and epidemiology.

#1885 - Experimental swainsonine poisoning in goats ingesting Ipomoea sericophylla and Ipomoea riedelii (Convolvulaceae), p.409-414

E-mail: franklin.riet@pq.cnpq.br Ipomoea sericophylla and Ipomoea riedelii cause a glycoprotein storage disease in goats. This paper reports the experimental poisoning in goats by dried I. sericophylla and I. riedelii containing 0.05% and 0.01% swainsonine, respectively. Three groups with four animals each were used. Group 1 received daily doses of 2g/kg body weight (bw) of dried I. sericophylla (150mg of swainsonine/kg). Goats from this group had clinical signs 36-38 days after the start of ingestion. Group 2 received dried I. riedelii daily doses of 2g/kg of I. riedelii (30mg of swainsonine/kg) for 70 days. No clinical signs were observed, therefore the swainsonine dose was increased to 60mg/kg for another 70 days. Goats from Group 2 had clinical signs 26-65 days after increase in swainsonine dose to 60mg/kg. Group 3 was used as control. In these experiments the minimum toxic dose was 60mg/kg which represents 0.0004% of the dry matter in goats ingesting 1.5% bw of the dry matter. For goats ingesting 2%-2.5% bw of dry matter this dose would be 0.00024%-0.0003% of the dry matter. After the end of the experiment two goats were euthanized and another six were observed for recovery of clinical signs. Four goats that continued to consume swainsonine containing plant for 39-89 days after the first clinical signs had non reversible signs, while two goats that ingested the plant for only 15 and 20 days after the first clinical signs recovered completely. These and previous results indicate that irreversible lesions due to neuronal loss occur in goats that continue to ingest the plants for about 30 days after the first clinical signs. Clinical signs and histological lesions were similar to those reported previously for goats poisoned by swainsonine containing plants. No significant alterations were found in packed cell volume, red and white blood cell counts, hemoglobin and mean corpuscular hemoglobin concentrations, mean corpuscular volume, and serum levels of glucose, total protein, and albumin, and the serum activities of gamma glutamyl transferase and aspartate aminotransferase. Swainsonine concentration of 0.05% in I. sericophylla and 0.01% in I. riedelii are different from samples of these plants used in previous experiments, which contained 0.14% and 0.5% swainsonine, respectively, demonstrating a wide variation in the toxicity of different samples.

#1886 - Toxoplasma gondii in semen of experimentally infected swine, 430-434

E-mail: a2abm@cav.udesc.br Eight reproductive boars were divided into three groups and inoculated with Toxoplasma gondii [GI (n=3) 1.5x104 oocysts strain P; GII (n=3) 1.0x106 tachyzoites strain RH; and GIII (n=2) non-inoculated control]. Clinical, hematological, parasitemia and serological tests and studies of the parasite in the semen through bioassay and PCR, and in reproductive organs (Bioassay and immunohistochemical analyses) were conducted to evaluate the toxoplasmic infection. Blood and semen were collected on day -2, -1, 1, 3, 5, 7, 9, 11, 14 and weekly up to 84 days post-inoculation (DPI). No clinical or hematimetric alteration was observed in the boars. Parasitemia was detected in one boar inoculated with oocysts at the 7th DPI and in another boar infected with tachyzoites (GII) at the 3rd and 49th DPI. Serological tests revealed antibodies against T. gondii in animals inoculated with oocysts or tachyzoites at the 7th DPI with dilutions of 1:256 and 1:64, which reached peaks of 1:4096 at day 11 and 9, respectively. The bioassays revealed the presence of the parasite in semen samples of a boar inoculated with oocysts (GI) at 3, 49 and 56 DPI and from two boars infected with tachyzoites (GII), one animal at 5 and two animals at 49 days DPI. Mice inoculated with semen from the control group (GIII) remained serologically negative. E-mail: a2abm@cav.udesc.br Eight reproductive boars were divided into three groups and inoculated with Toxoplasma gondii [GI (n=3) 1.5x104 oocysts strain P; GII (n=3) 1.0x106 tachyzoites strain RH; and GIII (n=2) non-inoculated control]. Clinical, hematological, parasitemia and serological tests and studies of the parasite in the semen through bioassay and PCR, and in reproductive organs (Bioassay and immunohistochemical analyses) were conducted to evaluate the toxoplasmic infection. Blood and semen were collected on day -2, -1, 1, 3, 5, 7, 9, 11, 14 and weekly up to 84 days post-inoculation (DPI). No clinical or hematimetric alteration was observed in the boars. Parasitemia was detected in one boar inoculated with oocysts at the 7th DPI and in another boar infected with tachyzoites (GII) at the 3rd and 49th DPI. Serological tests revealed antibodies against T. gondii in animals inoculated with oocysts or tachyzoites at the 7th DPI with dilutions of 1:256 and 1:64, which reached peaks of 1:4096 at day 11 and 9, respectively. The bioassays revealed the presence of the parasite in semen samples of a boar inoculated with oocysts (GI) at 3, 49 and 56 DPI and from two boars infected with tachyzoites (GII), one animal at 5 and two animals at 49 days DPI. Mice inoculated with semen from the control group (GIII) remained serologically negative. These monkeys are usually submitted to predatory chase, increasing the need for preservation of this wild animal species. An ultrasonographic examination of 10 Brown Capuchins was made in order to describe the normal ultrasonographic anatomy of their abdominal cavity. The urinary bladder revealed its wall thickness with an average of 0.2cm, the topographic situation of which allowed close relation with the wall of uterus and descendent colon. Using caudal abdominal scan, images of aorta, caudal vena cava and right iliac vein were obtained. Liver was accessible for examination by sagittal and cross-section ultrasound, allowing visualization of gallbladder and hepatic vessels. Renal scan allowed accuracy to evidence the echogenicity differences between pelvis, renal sinus, as well as the cortical-medullary relationship. The mean length of the kidneys was 6.24±0.31cm, and no significant differences were observed between left and right kidney length (Student’s t-test and ANOVA). The renal volume obtained was 2.37±0.18cm3. Correlation Coefficients of Pearson between right and left renal length and between right and left renal volume were r = 0.74 and 0.51. Mean thickness for cortical and medullar regions was 0.75±0.11 and 0.39±0.06cm, respectively. Correlation Coefficient of corticomedullar relation between right and left renal was r = 0.19. Examination by ultrasound was considered an efficient, non-invasive, fast and repeatable technique which provides useful data for clinicians and surgeons engaged in wild animal medicine.

The aim of this study was to evaluate the mucosa subjected to that model of small colon distention by histomorphometry and scan electronic microscopy (SEM). Sixteen horses were used. In the distended group (DG), nine of them were subjected to distention of the small colon by a surgically implanted intraluminal balloon that was inflated with a pressure of 40mm Hg during 4 hours. In the sham-operated group (SG), the balloon was implanted but not inflated. Full-thickness intestinal samples were collected before and after obstruction and after 1.5 and 12 hours of decom-pression. By SEM, it was observed that the mucosa turned flat and smooth after distention and returned to the wrinkled original appearance after decompression. Twelve hours after decompression the mucosa had a more irregular appearance with points of fragmentation. There was a reduction in mucosa thickness after distention, returning to basal values after decompression. Instead of the fact that there were changes in appearance and thickness, it was concluded that the mucosa could borne up the compression caused by distention returning to the original characteristics without major lesions.

An immunolocalization study and ultrastructural analysis by transmission electron microscopy of olfactory epithelium of mongrel dogs were made using 3 males (one year old) and 2 females (three years old). Labeled cells with positive staining by Proliferating cell nuclear antigen (PCNA) were observed in specific areas of the olfactory epithelium, especially above the basal membrane. The ultrastructure revealed cells adjacent to the basal membrane with morphology resembling sustentacular cells, supporting the idea of renewal of sustentacular and olfactory sensorial cells. Olfactory epithelium contains basal cells committed to self-renewal, characterized by high metabolic activity, identified by positive reaction to PCNA. These results suggested the renewal of sustentacular and sensorial olfactory cells through the same pathway.
