E-mail: camilelug@gmail.com Cryptococcus neoformans and C. gattii are associated with dry bird excreta but rarely recovered from birds’ digestive tract. The objective of the present study was (1) to verify the existence of C. neoformans and C. gattii in crop and cloaca of wildlife and captivity birds hypothesizing about a possible primary source of this yeast in the excreta, and (2) to determine the fungi’s invasive capability in avian species through latex agglutination. For that purpose, 172 cloacal and 77 crop samples of domestic pigeon, Passerine, and Psittacine birds were collected. None of these samples was positive, suggesting that the yeast is not saprobiotic in the digestive tract of these birds. Only one out of 82 serum samples collected from pigeons and Psittacine birds was positive (title 1:2) showing that Cryptococcus sp. probably has a low invasive capability in birds, and is thus considered only a dry excreta colonizer.

Mainly during the dry season livestock browse its leaves from the shrubs or ingest the dry leaves fallen to the ground. Farmers report the spontaneous poisoning by this plant when livestock has access to shrubs or branches that had been cut. Different parts of the ground fresh plant, diluted in water, are used by people in the semiarid to hunt birds. To determine the toxicity of C. phyllacanthus, leaves of the non-spiny plant were fed by hand to a goat by putting small amount into its mouth. After the consumption of 4.7g/kg body weight, the goat had tachycardia, tachypnoea, dyspnoea, nystagmus, opisthotonos and sternal recumbence. The death occurred 30 minutes after the onset of clinical signs. Fresh leaves from the same plants were given to 8 goats at doses of 0.5-2.5g/kg without causing clinical signs. Three goats showed clinical signs after the ingestion of 3g/kg. Clinical signs were similar to those observed in cyanide poisoning. Two goats given 0.5mg/kg of a 20% solution of sodium thiosulphate recovered immediately after treatment. Another goat recovered spontaneously. Leaves from the same plants were sun-dried during variable periods for 8-30 days, and then given to 8 goats. The goat that ingested the plant dried at the sun during 8 days died after the ingestion of 3 g/kg. The goat that ingested the plant, exposed to the sun for 9 days, showed clinical signs after the ingestion of 1.13g/kg, but recovered spontaneously. The goats that ingested the leaves exposed to the sun during 10-29 days showed clinical signs after the ingestion of 3g/kg, but recovered spontaneously or after treatment with sodium thiosulphate. No clinical signs were observed in the goat that ingested the plant that had been exposed to the sun during 30 days. In another experiment ground leaves of the plant were dried at the sun for 1, 2, 3, and 4 days and given to goats at the dose of 3g/kg. Goats that received the plant dried for 1, 2 or 3 days showed clinical signs, but recovered spontaneously. The goat that ingested the plant dried 4 days showed no clinical signs. Leaves of the spineless C. phyllacanthus used in the experiments, 20 samples of leaves from spiny plants, 3 samples of the ground plant stem, and 2 samples of fruits were positive with the picrosodic paper test for HCN. These results demonstrated that C. phyllacanthus is a cyanogenic plant. After being cut the entire leaves maintain their toxicity for up to 30 days, and the ground leaves for up to 3 days. After the end of the raining season dry leaves fallen to the ground and tested within a short period were negative for HCN. These results have to be taken into account for the use of C. phyllacanthus as forage.

#203 - Co-infection by porcine circovirus type 2 and porcine parvovirus in aborted fetuses and stillborn piglets in southern Brazil, p.425-429

E-mail: davetpat@ufrgs.br Porcine circovirus types 1 and 2 (PCV1, PCV2) and porcine parvovirus (PPV) are widespread in pig populations around the world. Nevertheless, only PCV2 has been associated with different clinical syndromes, thus representing a major problem to the pig industry. The association of cases of swine abortions and stillborns with PCV1 and PCV2 and PPV was studied retrospectively (2005-2007). Additional pathogens were also investigated in lesioned fetuses. The studied litters included stillborn piglets and several mummified fetuses of varied sizes. Ventricular dilatation, myocardial pale areas, and mesocolic edema were the gross lesions. Escherichia coli was detected as co-infecting with PCV2 the cases in which mesocolic edema was seen. Microscopic lesions included non-suppurative myocarditis, myocardial necrosis and fibrosis, mineralization foci and intranuclear inclusion bodies in cardiomyocytes, and interstitial mononuclear pneumonia. Samples from 7 (5.78 per cent) of 121 aborted fetuses and stillborn piglets had lesions consistent with a viral cause and showed both positive anti-PCV2 immunostaining as well as PCV2-PCR. In samples from 3 (2.47 per cent) of these 7 fetuses, co-infection with PPV was confirmed by Nested-PCR. Both viruses were detected in fetuses at different stages of gestation. Viral antigens of PCV2 were detected by immunohistochemistry mainly in macrophages and myocytes. PCV1 individually was not detected in any of these affected fetuses, but it was associated with PCV2 and/or PPV in some of them. These findings indicate that PCV2 alone or in association with PPV should be kept in mind when investigating causes of infectious abortion in pigs in Brazil.

However, literature is poor in information about the stability of the whole cell Brucella antigen for use in the complement fixation test to establish a time of validity of the antigen. Hence the aim of this investigation was to evaluate the stability of this antigen under refrigeration for use in the complement fixation test. Fourteen batches of antigen prepared with Brucella abortus strain 1119/3, produced from 9 months to 23 years and 11 months before, were analysed. One hundred and sixty-seven cattle sera with varying titres of antibodies to Brucella were tested through the warm complement fixation microtechnique with five 50% haemolytic units of complement. Sera with at least 25% of complement fixation in dilution 1:4 were considered positive. The results with 13 of the antigen batches were compared with the results obtained with the batch produced 9 months before by the McNemar c2 test and kappa statistic. The oldest antigen batch gave a higher proportion of sera titres which were exactly the same observed with the 9-month-batch (90.4%), and the antigen produced 4 years and 3 months before the test gave de lowest proportion of sera with the same titre of the 9-month-antigen (73.7%). The comparison of the results after being classified as positive and negative showed that the highest proportion of agreed results was observed with the antigen produced 21 years and 4 months before (98.8%, kappa 0.98). The antigen with the lowest proportion of agreed results was the one produced 3 years and 2 months before (91.6%, kappa 0.84). The results of the study show that most sera gave very similar results with all antigen batches evaluated, and that there was no relationship between the period of antigen production and the difference in test results.

Seub., collected in the field one or two times a week and kept in the refrigerator for 1-4 days, was administered for 30 or more days as the only food ad libitum to 2 donkeys and one white sheep, and as the only forage ad libitum, to one white horse. No clinical signs were observed in those animals, and serum activities of aspartate-aminotransferase (AST), alanine-aminotransferase (AST), and gamma-glutamyltransferase (GGT) were within normal ranges. In another experiment, one sheep was tied by a rope, during the day, in a pasture of exclusively F. humboldtiana for a 26 days period. Skin lesions characteristic of photosensitization appeared 4-5 days after the start of the experiment and became severe until day 26. After the withdrawal of the animal from the pasture on day 26 it fully recovered within 30 days. In another experiment, 4 sheep were tied by ropes in the same pasture, during the day, for a 29 days period. Another 4 control sheep were maintained in a neighboring pasture without F. humboldtiana. Skin lesions characteristic of photosensitization were observed after 11-25 days of F. humboldtiana grazing. Serum activities of AST and GGT, and serum levels of bilirubin were within normal ranges. At the end of the 29 days grazing F. humboldtiana, 2 sheep were euthanized, and 2 were withdrawn from the pastures. No gross or histologic liver lesions were observed on the 2 sheep euthanized; the other 2 sheep recovered within 17 and 20 days after the end of grazing. One mare and its foal grazed in the same pasture during 44 days. The mare that had a pigmented skin had no dermatitis, but the foal showed dermatitis on the white areas of the skin after 25 days of grazing. The two horses were withdrawn from the pastures after 44 days grazing, and the foal fully recovered within 15 days. These experiments indicate that F. humboldtiana causes primary photosensitization in domestic animals. The absence of lesions in animals ingesting the plant collected in the field and kept refrigerated suggests that F. humboldtiana lost toxicity after collection. The absence of lesions in the eyes of affected animal, which are characteristic of primary photosensitization caused by furocoumarins containing plants, suggests that F. humboldtiana contains a naphthodianthrone derivate, similar to those found in Fagopyrum esculentum and Hypericum perforatum which do not cause eye lesions.

This experimental simulation of sensibilization by Mycobacterium bovis and Mycobacterium avium in cattle aimed to verify the immune response by both the cervical comparative test and the evolution of the specific production of gamma-interferon, and also to identify interference of unspecified reactions by M. avium on the test results. The results support that the experimental animals started a response of delayed hypersensitivity to the inactivated bacilli, and that both diagnostic tests for bovine tuberculosis were efficient for the identification of animals sensitized with M. bovis and for discrimination of reactions generated by inoculation of cattle with M. avium.

The region studied has about 5.7 million head of cattle, corresponding to 23% of the total of 24.9 million cattle in the State of Mato Grosso do Sul. On 210 farms, between December 2003 and March 2004, 2,376 blood samples were collected from cows, aged 24 months or older, for serial diagnostic tests. Screening through the buffered acidified antigen test was confirmed by the 2-mercaptoetanol test. On the occasion of sample collection a questionnaire with information related to identification, kind of cattle and management practices was filled out. In individual animals the real prevalence was estimated at 5.6%, and in the cattle herds at 37.3%. The variables, which presented association through odds ratio (OR), univariate analysis and 95% confidence interval (CI) with serum positivity for brucellosis, were: the exploration of beef cattle (OR = 2.82, 95% CI = 1.49-5.34), Zebu breed (OR = 2.62, 95% CI = 1.40-4.88) and abortion (OR = 1.83, 95% CI = 1.01-3.33). The results shown here demonstrate, despite the prevalence of brucellosis in the extract of Mato Grosso do Sul studied, that the control of the disease may depend on adoption of a program focusing upon the exploration of beef cattle, the Zebu breed and the occurrence of abortion.

#208 - Intoxicação de bovinos por Tetrapterys multiglandulosa (Malpighiaceae) em Mato Grosso do Sul

Pesquisa Veterinária Brasileira 26(3):139-146. E-mail: eqrural@nin.ufms.br Two outbreaks of poisoning by Tetrapterys multiglandulosa in cattle and the experimental reproduction of the toxicosis in sheep are described. Both outbreaks occurred on the same farm in the municipality of Bataiporã, state of Mato Grosso do Sul, Brazil. The first outbreak occurred in July-October 2004 and involved a cattle population at risk of 290 pregnant cows, which were introduced into a 60 hectare pasture with a legal reservation area heavily infested by T. multiglandulosa. Of these, 230 cows (79.3%) aborted, had stillbirths or delivered weak calves that died few days after birth. Seven cows died, and one cow and a 10-day-old calf were necropsied. The second outbreak occurred in September-October 2005, 40 days after 285 2-year-old heifers were introduced into the same pasture infested by T. multiglandulosa and where the first outbreak had occurred in the previous year. Nine heifers got sick and died, and three of then were necropsied. Clinical signs of affected cattle, including a 10-day-old calf, were marked lethargy, loss of weight with distension of the abdomen (ascites), subcutaneous dependant edema, distended and pulsating jugular veins, dyspnea and cardiac arrhythmia. Necropsy findings included a round and dilated heart with whitish and firm areas in the myocardium, and changes related to cardiac failure such as cavitary edema, nutmeg liver, pulmonary edema, a large blood clot in the left ventricle. Histopathological changes included necrosis and fibrosis in the myocardium, chronic passive hepatic centrolobular congestion, pulmonary edema, and spongy degeneration in the white matter of the brain. Experimental sheep died 29 (Sheep 1) and 35 (Sheep 2) days after being fed average daily doses of T. multiglandulosa corresponding to 14g/kg (Sheep 1) and 7,5 g/kg (Sheep 2) per day. Clinical signs were observed from the 7th day (Sheep1) and the 4th day (Sheep 2) of the experiment and included tachycardia, arrhythmia, lethargy and head pressing. Necropsy and histopathologic findings in both experimental sheep were very similar to those observed in affected cattle of the two spontaneous outbreaks.

Regional research becomes important, in order to know the interrelation between the agent and its host. In this way, the occurrence of immunoglobulins of class G, anti-Babesia bovis and anti-Babesia bigemina in the Nelore (Bos indicus) and Hostein breed (Bos taurus), was investigated in two regions of the State of São Paulo, 300 km distant from each other. For the indirect method of ELISA, 1,161 bovine serum samples were tested. The medium frequencies of antibodies showed that in the two regions exists an enzootic stability for B. bovis in both breeds studied; even so there was a tendency of marginal area for the Nelore breed in one of the regions. Regarding B. bigemina, in both regions exists enzootic stability for the Hostein and enzootic instability for the Nelore breed. Therefore, acute cases of the disease or specific outbreaks by B. bigemina infection in the Nelore breed may occur in these regions.
