Abstract in English:

Canal C.W., Ferreira D.J., Macagnan M., Fallavena L.C.B., Moraes H.L.S. & Wald V.B. 2004. Prevalence of antibodies against chicken anaemia virus (CAV) in broiler breeders in Southern Brazil. Pesquisa Veterinária Brasileira 24(2):89-92. Centro de Diagnóstico e Pesquisa em Patologia Aviária (CDPA), Faculdade de Veterinária da Universidade Federal do Rio Grande do Sul, Porto Alegre, RS 91540-000, Brazil. E-mail: claudio.canal@ufrgs.br Chicks infected during the first two weeks of life with chicken anaemia virus (CAV) manifest clinical disease that can be avoided if the breeder hens transfer enough antibodies to their progeny. The objective of the present work was to establish the prevalence and titer of anti-CAV antibodies in some Brazilian broiler hen breeder flocks and verify in which phase of life the birds were infected. A total of 1,709 serum samples from 12 broiler hen flocks vaccinated against CAV and 64 unvaccinated flocks were analyzed for CAV antibodies with an enzyme-linked immunosorbent assay (ELISA). All non-vaccinated breeder flocks were found to be infected with CAV, with 89% of the hens tested presenting antibodies, 52% of these with titers considered high enough to protect their progeny against CAV infection. Likewise, all vaccinated hens had antibody titer to CAV capable of conferring protection to their progeny. Thus, vaccination of hens seems capable of conferring protection to chicks against clinically apparent CAV-associated disease.

E-mail: elporti@fcv.unlp.edu.ar Swine eperythrozoonosis is a haemotrophic disease caused by Eperythrozoon suis, actually called Mycoplasma suis, an extracellular bacterial organism that apparently adheres to pig erythrocyte membrane, inducing its deformation and damage. Since little is known about the ultrastructural and morphometrical aspects of this microorganism, the present work aimed to deal with these issues. The ultrastructural study revealed the presence of structures corresponding to tubules disseminated throughout the soma of M. suis. A variable separation between the microorganism membrane and that of the erythrocyte was also observed. The structural and positional attitude of M. suis could allow speculation about its mechanism of action.

Postal 15094, Porto Alegre, RS 91540-000, Brazil. E-mail: davetpat@vortex.ufrgs.br Green leaves of Trema micrantha were ground and mixed with water in a domestic blender and then administered by stomach tube to seven goats. One additional goat was fed ad libidum with the green leaves of T. micrantha. Clinical signs were observed in six goats that became ill 2 days after having been dosed with or fed the plant. There were five deaths, which occurred until 4 days after ingestion. Affected goats remained static during long periods and kept their heads low. Incoordination, rhythmical movements of the head, apathy, anorexia, and tenesmos were also noticed. Paddling movements and coma were seen in one goat. T. micrantha was toxic at dosages of 30 g/kg or higher. The most significant gross lesions were observed in the livers, which were yellowish, friable, and with pronounced lobular pattern. Their cut surfaces were reddened and depressed areas alternated with whitish ones. The tiver of one goat was slightly but homogeneously reddened but did not show accentuated lobulation. Petechial haemorrhages in the region between the chest and scapula, in the epicardium, mediastinum and serosal membranes of the abdominal organs were also observed. The most importante histologic finding was hepatic centrilobular coagulative necrosis, which was associated with congestion, haemorrhages and degenerative changes in the circumjacent hepatocytes. Additional microscopic lesions were found in the nervous system and included perineuronal and perivascular edema and swollen neurones, especially those of the frontal cortex.

Email: anemiaveterinaria@bol.com.br Onion poisoning is reported worlwide in several animal species. The toxic principie (npropyl dissulfide) present in onions causes the transformation of hemoglobin in methemoglobin. In order to study the laboratory, gross and histopathological tindings in onion poisoning in cats, tive 4-month-old cats were fed a single dose of 10g/kg of dried-onion each. Another cat of the sarne age did not receive the onion meal and served as control. AII tive cats developed clinical signs of the toxicosis; one of them died within 24 hours of the ingestion of the onion meal. Clinical signs included apathy, tachycardia, tachypnea, and cyanosis. Laboratory tindings included hemolytic anemia associated with Heinz bodies and methemoglobinemia. Main necropsy tindings were splenomegaly and brown discoloration of blood. Histopathological tindings included splenic and hepatic hemosiderosis and multifocal extramedullary hematopoiesis.

#345 - Emergence of Haematobia irritans in cattle dung pats in Seropédica county, Rio de janeiro, 21(2):77-80

Abstract in English:

ABSTRACT.- Macedo D.M., Brito L.G. & Moya Borja G.E. 2001. [Emergence of Haematobia irritans in cattle dung pats in Seropédica county, Rio de janeiro] Emergência de Haematobia irritans em fezes bovinas no município de Seropédica, Rio de Janeiro. Pesquisa Veterinária Brasileira 21(2):77-80. Depto Parasitologia Animal, Univ. Fed. Rural do Rio de janeiro, Seropédica, RJ 23851-970, Brazil. This studywas carried out in order to investigate the emergence of adult flies of Haematobia irritans in cattle dung maintained in the field and in the laboratory, as well as other flies associated with dung pats. Two groups of cattle dung were used, one in the field and another in the laboratory, each group consisting of five fresh dung pats ofmedium size (about 30 cm in diameter), opaque appearance, presence of a thin externai crust· and firm consistency. The dung pats were covered with emergence cages in pyramidal format, presenting a hole in the superior part where a removable flask was coupled, and the substitution of the dung was done every 2 weeks. A total of 355 specimens of H. irritans was collected, 151 of which were obtained in the field and 204 specimens in the laboratory. These differences probably were dueto smaller temperature variations in the laboratory than in the field, besides the action of arthropodal predators, competitors and parasites of horn fly pupae that probably interfered in H. irritans emergence in cattle dung under field conditions. A higher number of females were observed in both conditions. Besides H. irritans, there were collected other flies associated with bovine manure belonging to the following families: Aulacigastridae, Muscidae, Psychodidae, Sarcophagidae, Sepsidae, Tachinidae and Ulidiidae. Sepsidae was the most abundant, with 5,224 specimens of the total of 8,928 flies collected, followed by Sarcophagidae with 2,235, Muscidae with 1,357, Aulacigastridae with 54, Psychodidae with 46, Ulidiidae with 6 and Tachinidae with 5 specimens.

RESUMO.- Macedo D.M., Brito L.G. & Moya Borja G.E. 2001. [Emergence of Haematobia irritans in cattle dung pats in Seropédica county, Rio de janeiro] Emergência de Haematobia irritans em fezes bovinas no município de Seropédica, Rio de Janeiro. Pesquisa Veterinária Brasileira 21(2):77-80. Depto Parasitologia Animal, Univ. Fed.

#346 - Experiments in rabbits with cyanogenic plants, 20(2):65-70

Abstract in English:

ABSTRACT.- Brito M.F., França T.N., Oliveira K.D. & Cerqueira V.D. 2000. [Experiments in rabbits with cyanogenic plants.] Estudos experimentais em coelhos com plantas cianogênicas. Pesquisa Veterinária Brasileira 20(2):65-70. Projeto Sanidade Animal Embrapa/UFRRJ, Km 47, Seropédica, RJ 23851-970, Brazil. The dried leaves of Piptadenia macrocarpa Benth. 1 =Anadenanthera macrocarpa (Benth.) Brenan], Piptadenia viridiflora (Kunth.) Benth. and Holocalyx glaziovii Taub. [=Holocalyx balansae Micheli], all trees of the Leguminosae Mimosoideae family, were given orally to rabbits in single doses. In the case of H. glaziovii, leaves were fed after being stored in two diferente ways. In the first series of experiments, Piptadenia macrocarpa failed to poison rabbits although the leaves were collected when sprouting, the most toxic stage forcattle. The experiments were performed 4 to 6 months after the leaves had been collected and dried. In a second series of experiments, similar leaves were fed at monthly intervals after harvest. They were toxic for the first 5 months, but were innocuous after 6 months storage. In both series of experiments the leaves were dried in the shade and then kept in cotton sacks at room temperature. They always were milled at the occasion of the experiments. These experiments with rabbits confirm the results with cattle, in which severe poisoning was caused by leaves stored for 3 months but caused no ili effect after 4 months, even when fed in large amounts. Young leaves of P. viridiflora were lethal at 12 g/kg, 12 months after collection and drying. In a second experiment the lethal dose was only 6 g/kg with leaves stored for only 2 months. These results show that the dried leaves of these species lose toxicity with time. Holocalyx glaziovii leaves were milled 6 weeks after collecting and dried and stored in sealed containers. They were fed in. single doses to different rabbits at intervals during a whole year. Lethal doses were initially 0.75 g/kg to 1.5 g/kg but rose to 1.5 g/kg to 3 g/kg by the end of the year. In a second experiment, leaves stored in cotton sacks and milled just before feeding were compared with milled leaves stored in sealed glass containers. The plant material kept in cotton sacks had halfthe toxicity of the leaves stored.for 2 months in sealed containers. After 10 to 12 months it was only a little less toxic than that stored in the sealed containers but there were no differences after 14 months. The symptoms observed in the rabbits that were poisoned by the dried leaves of P. macrocarpa, P. viridiflora and H. glaziovii were neuromuscular. Post-mortem findings and histopathological findings were almost absent. On opening the stomach, a strong smell of bitter almonds was sometimes noticed. The degree of toxicity during the storage of the dried leaves of he three tested plants was generallywell correlated with the picrate testfor cyanide.

RESUMO.- Brito M.F., França T.N., Oliveira K.D. & Cerqueira V.D. 2000. [Experiments in rabbits with cyanogenic plants.] Estudos experimentais em coelhos com plantas cianogênicas. Pesquisa Veterinária Brasileira 20(2):65-70. Projeto Sanidade Animal Embrapa/UFRRJ, Km 47, Seropédica, RJ 23851-970, Brazil.

#347 - Relationship between foamy macrophages in the liver of cattle and the ingestion of Brachiaria spp, 19(2):79-83

Abstract in English:

ABSTRACT.- Driemeier D., Döbereiner J., Peixoto P.V. & Brito M.F. 1999. [Relationship between foamy macrophages in the liver of cattle and the ingestion of Brachiaria spp.] Relação entre macrófagos espumosos ("foam cells") no fígado de bovinos e ingestão de Brachiaria spp no Brasil. Pesquisa Veterinária Brasileira 19(2):79-83. Depto Patologia Clínica Veterinária, Faculdade de Veterinária, UFRGS, Cx. Postal 15094, Porto Alegre, RS 91540-000, Brazil. To establish the etiological relationship and the appearance of foamy macrophages in the liver of cattle from tropical regions of Brazil, Iiver samples from the files of the Pathology Section of Embrapa-Projeto Sanidade Animal, Rio de Janeiro, were reviewed. A total of 55 liver samples of cattle which died from different causes between 1970 and 1991 were reexamined. Only samples of animals which grazed known pastures were reviewed. Foamy macrophages were not seen in the samples from 1970 to 1975, although 40 samples (72%) were from this period. Foamy macrophages were observed from 1976 on, coinciding with the introduction of Brachiaria decumbens from Australian seeds into Brazil. Some samples were from cattle with histories of photosensitization, which were at that time attributed to Pithomyces chartarum. The results of this study indicate that the liver changes are related to prolonged ingestion of Brachiaria spp.

RESUMO.- Driemeier D., Döbereiner J., Peixoto P.V. & Brito M.F. 1999. [Relationship between foamy macrophages in the liver of cattle and the ingestion of Brachiaria spp.] Relação entre macrófagos espumosos ("foam cells") no fígado de bovinos e ingestão de Brachiaria spp no Brasil. Pesquisa Veterinária Brasileira 19(2):79-83. Depto Patologia Clínica Veterinária, Faculdade de Veterinária, UFRGS, Cx. Postal 15094, Porto Alegre, RS 91540-000, Brazil.

#348 - Experimental studies in cattle with cyanogenic plants, 19(2):84-90

Abstract in English:

ABSTRACT.- Tokarnia C.H., Peixoto P.V, Brito M.F., Duarte M.D. & Brust L.A.C. 1999. [Experimental studies in cattle with cyanogenic plants.] Estudos experimentais em bovinos com plantas cianogênicas. Pesquisa Veterinária Brasileira 19(2):84-90. Projeto Sanidade Animal Embrapa/UFRRJ, Km 47, Seropédica, RJ 23851-970, Brazil. These studies were performed in arder to obtain additional data on Brazilian cyanogenic plants. Three animals, severely poisoned by Piptadenia macrocarpa ( =Anadenanthera macrocarpa), recovered readilywhen treated intravenously with 660 mg/kg of sodium hyposulphite (sodium thiosulphate) plus 30 g dissolved in water given by stomach tube. Fresh leaves (in the sprouting phase), fed in October, were more poisonous than mature leaves fed in March. Dried leaves slowly lost their toxicity within a few months. Both the fresh and dried leaves of Piptadenia viridiflora showed similar toxic effects to P. macrocarpa. As this was also readily reversed by the administration of sodium hyposulphite in two animals, P. viridiflora can be included in the group of toxic cyanogenic plants. Four animals severely poisoned by the leaves of Holocalyx glaziovii ( =Holocalyx balansae) also recovered readily after receiving sodium hyposulphite. The dried leaves of H. glaziovii also slowly lost their toxicity within a few months. Two animals severely poisoned by the leaves of Manihot glaziovii also recovered readily with sodium hyposulphite. However, dried leaves were no longer poisonous. The leaves of H. glaziovii and of M. glaziovii always gave a quick (in less than 5 minutes) positive reaction to the picrate test for cyanide. Those of P. macrocarpa and P. viridiflora reacted more slowly (after 15 and 10 to 30 minutes respectively) when in the sprouting stage, and even slower when the leaves were mature. The slower reactions seen with Piptadenia spp indicate a slower breakdown of the hidrocyanic glycosides and this also explains the longer course of poisoning seen in cattle fed these species. The dried leaves of Piptadenia spp and also of H. glaziovii gave slower reactions than the fresh leaves. These data permit the conclusion that the picrate test for cyanide is of relative value when evaluating the amount of cyanogenic glycosides in plants.

RESUMO.- Tokarnia C.H., Peixoto P.V, Brito M.F., Duarte M.D. & Brust L.A.C. 1999. [Experimental studies in cattle with cyanogenic plants.] Estudos experimentais em bovinos com plantas cianogênicas. Pesquisa Veterinária Brasileira 19(2):84-90. Projeto Sanidade Animal Embrapa/UFRRJ, Km 47, Seropédica, RJ 23851-970, Brazil.

Samples of liver, kidney, spleen, and hepatic, mesenteric, retromandibular, prescapular and mediastinic lymph nades, collected from 12 groups of cattle of different ages from the State of Mato Grosso, were studied. A total of 120 bovines was examined. According to their history, the animals were kept in pastures where Brachiaria decumbens and Brachiaria brizantha were the predominant plants. Grossly there was a yellowish liver which was more evidente after 24 hr. fixation with formalin. Hepatic and mesenteric lymph nades had normal size, howeve1; on the cut surface there were whitish grooves, extending from the cortex to the medulal; and multiple small white nodular areas in the medulal: Beside these areas there were red, sometimes hemorrhagic, foci. Microscopically all animals had cells with foamy citoplasm in the liver and in the hepatic and mesenteric lymph nades, of ten forming multinucleated cells. In the lymph nodes these infiltrates were adjacent to necrotic, hemorrhagic areas. In the liver the foamy cells were irregularly disseminated throughout the parenchyma or formed nodules around the central vein. These cells did not stain with PAS but weakly with oil red. Ultrastructurally foamy cells and hepatocytes showed negative images of crystals involved partially or totally by membranes. Mediastinic, retromandibular and prescapular lymph nades of the sarne animals showed no gross and microscopic alterations.

[Histologic, histochemical and ultrastructural study of livers and lymph nodes with foamy macrophages (foam cells).)

#350 - Comparison of conjugates in the competitive enzyme immunoassay for the serological diagnostic of bovine brucellosis

Abstract in English:

The purpose of this study was to evaluate the competitive enzyme immunoassay used for the serological diagnosis of bovine brucellosis, comparing the results obtained using a conjugate prepared with monoclonal antibody and a conjugate prepared with policlonal sera. The results were also compareci with those obtained by a microtechnique of complement fixation. Two hundred and seventy-three sera from herds with a history of brucellosis, 205 sera from herds free of brucellosis and without vaccination with Brucella abortus strain 19 and 122 sera from herds without history of infection or vaccination were tested. Among sera from infected herds, the complement fixation test revealed antibody titres in 28.51%, the enzyme immunoassay using monoclonal conjugate in 39.19% and the enzyme immunoassay using policlonal conjugate revealed antibody titres in 37.0% of the sera. The agreement between complement fixation and the enzyme immunoassay using policlonal conjugate was 94.38% and the agreement between both of the enzyme immunoassays was 97.89%. Among the sera from vaccinated herds, the agreement between the complement fixation test and the enzyme immunoassay using policlonal conjugate was 69.39%, and the agreement between the enzyme immunoassays was 90.73%. The complement fixation test did not reveal any antibody titre in 67.86% of the sera from vaccinated herds, the enzyme immunoassay using policlonal conjugate did not reveal titres in 51.22% and the enzyme immunoassay with monoclonal conjugate did not reveal antibody titres in 41.95% of the sera from vaccinated herds. All the 122 sera from herds without infection or vaccination were negative to the three serological tests.
