Abstract in English:

Oliveira C.M.C., Barbosa J.D., Macedo R.S.C., Brito M.F., Peixoto P.V. & Tokarnia C.H. 2004. [A comparative study of the toxicity of Palicourea juruana (Rubiaceae) to buffalo and cattle.] E-mail: tokarnia@ufrrj.br An experimental study was performed to establish the toxicity of Palicourea juruana for buffaloes and to add new data on the toxicity of this plant for cattle. The clinical and pathological pictures were similar, but a comparison of the lethal doses for buffaloes (1-2g/kg) and cattle (0.25g/kg) shows that the buffalo is at least 4 times more resistant. In experiments performed 10 years earlier with the fresh leaves - collected on the same farm in July, at the beginning of the dry season, only 2 months later than in this second experimental series - the lethal dose for cattle was 2 g/kg. No explanation was found for the extremely high toxicity of the plant observed in the present study.

E-mail: davetpat@vortex.ufrgs.br The study assessed differences of pathogenicity among 19 strains of Brachyspira pilosicoli isolated from cases of diarrhea in swine in the State of Rio Grande do Sul, Brazil, using an one-day-old chick model. The chicken inoculated with live bacterial cultures were submitted to euthanasia 21 days later, and the cecum was examined histologically using hematoxylin-eosin, silver staining and immunohistochemistry with a polyclonal antibody anti-Brachyspira pilosicoli. With silver staining and immunohistochemistry, respectively, 21.59% and 70.96% of the chicken showed colonization of the cecal epithelium with B. pilosicoli. Differences in the type of colonization characterized by continuous adhesion, focal adhesion or free bacteria in the intestinal lumen were observed. Immunohistochemistry was more efficient to assess intestinal colonization, because 49.37% more colonized chicken could be detected with silver staining. With three strains, elongated figures inside the cytoplasm of epithelial cecal cells in the inoculated chicken were observed.

#603 - Comparative pathogenicity of bovine herpesviruses type 1 (BHV-1) subtypes 1 (BHV-1.1) and 2a (BHV-1.2a)

Abstract in English:

Spilki F.R, Esteves P.A., Lima M., Franco A.C., Chiminazzo C., Flores E.F., Weiblen R., Driemeier D. & Roehe P.M. 2004. Comparative pathogenicity of bovine herpesviruses type 1 (BHV-1) subtypes 1 (BHV-1.1) and 2a (BHV-1.2a). Pesquisa Veterinária Brasileira 24(1):43-49. Centro de Pesquisas Desidério Finamor, Fepagro Saúde Animal, Cx. Postal 47, Eldorado do Sul, RS 92990-000, Brazil. E-mail: proehe@ufrgs.br The study aimed to examine the capacity of two bovine herpesvirus type 1 (BHV-1) isolates of different subtypes (EVI 123/96, BHV-1.1; SV265/98, BHV-1.2a) to induce respiratory disease in calves. These two isolates are representative of the BHV-1 subtypes prevalent in Brazil. Viral subtypes were confirmed by monoclonal antibody analysis and by restriction enzyme digestion of viral genomes. The viruses were inoculated intranasally into seven 3 months old calves (four with BHV-1.1, three with BHV-1.2a). Three other calves of identical age and condition were kept as uninfected controls. In both groups of infected calves, the clinical signs observed were consistent with typical infectious bovine rhinothracheitis (IBR), including pyrexia, apathy, anorexia, nasal and ocular mucopurulent discharges, erosions on the nasal mucosa, conjunctivitis, lachrymation, redness of nasal mucosa, dyspnoea, coughing, tracheal stridor and enlargement of retropharingeal, submandibular and cervical lymphnodes. No significant differences were observed between the clinical scores attributed to both groups. Virus shedding in nasal and ocular secretions were also similar, apart from a significant difference in nasal virus shedding on day 1 to 3 post-inoculation, which was higher for BHV-1.1 than for BHV-1.2a. Following corticosteroid induced reactivation of the latent infection, recrudescence of clinical signs was also observed, with no significant differences on both groups. It was concluded that both subtypes BHV-1.1 and BHV-1.2a were able to induce clinically undistinguishable respiratory disease in calves, either subsequent to a primary infection or following reactivation.

E-mail: claudioslbarros@uol.com.br Cases of respiratory disease were diagnosed in five out of 23 cattle (21. 7%) after they wete fed moldy damaged sweet potatoes (lpomoea batatas) on a small farm in the county of São Vicente do Sul, state of Rio Grande do Sul, Brazil. Of those five cattle, three died spontaneously and another one was euthanatized for necropsy while showing advanced respiratory clinicai signs. The disease manifested itself approximately 24 hours after the ingestion of the sweet potatoes and lasted from 1 to 4 days. Clinicai signs included dyspnea (labored breathing and abdominal respiration), tachypnea, extended neck with Iow carriage of the head and rhythmical flaring of the nostrils. Two cows were necropsied. Necropsy findings included distended pale and rubbery lungs which failed to collapse when the thorax was open, and marked pulmonar interstitial emphysema and edema. Lymphoid hyperplasia was observed in the hilar nodes and spleen. Histologically, the lesions were those of interstitial pneumonia. Alveolar septa were thickened by fibroblasts and inflammatory cells, and there was hypertrophy and hyperplasia of type li pneumocytes; the interlobular septa were distended by edema and emphysema. The culture of the moldy sweet potatoes yielded Fusarium solani and Fusarium oxysporum.

E-mail: gemoya@ufrrj.br The eradication of the screwworm, Cochliomyia hominivorax, from USA, Central America and the North of Africa has been achieved by rearing and releasing sterile males. Why we do not decide to eradicate this insect from the southamerican countries too? ln this paper the scientific, ecological and economic reasons before taking such a courageous decision are discussed. The human botfly, Dermatobia hominis, has not been eradicated from any country, because there is so far no technique available for mass production in artificial diets. Studies on dispersion, sexual behavior and aggregation sites of the adults in the field are also not known. Why the neotropical myiases will continue to be treated basically with chemicals, recommendations for insecticide resistance management and studies on alternative methods are discussed trying to apply in the future an ecologically based pest management of the screwworm and the human botfly.

E-mail: rmtmed@cstr.ufpb.br Three outbreaks ofnitrate poisoning are reported from the semiarid region ofthe state of Paràíba, northeastem Brazil. One outbreak caused by Echinochloa polystachya (capim-mandante) and two others caused by Pennisetum purpureum (capim-elefante) occurred at the end ofthe dry season, after the first raios. ln one ofthe outbreaks caused by Pennisetum purpureum part of the pasture had been fertilized with cattle manure. Five out of 11 cattle died on one farm, 27 out of81 on another, and 3 out of9 on a third one. Clinicai sigos were anorexia, respiratory distress, teeth grinding, depression or hyperexitability, tremors, abdominal contractions, salívation, nasal discharge, uncoordinated gait, cyanosis, and finally recumbency. Nitrates and nitrites in the blood of affected cattle and pastures were determined by the diphenylamine test. It is suggested that the main reason for nitrate accumulation in the grasses was the prolonged draught followed by rain. ln one ofthe outbreaks caused by Pennisetum purpureum, another reason was probably the fertilization of the soil with cattle manure.

#608 - Oxidative metabolism of the neutrophils in sheep treated with sodium monensin and experimentally submitted to ruminal acidosis, 22(4):129-134

Abstract in English:

ABSTRACT.- Afonso J.A.B., Ciarlini P.C., Kuchembuck M.R.G., Kohayagawa A., Feltrin L.P.Z., Ciarlini L.D.R.P., Laposy C.B., Mendonça C.L. & Takahira R.K. 2002. [Oxidative metabolism of the neutrophils in sheep treated with sodium monensin and experimentally submitted to ruminal acidosis.] Postal 152, Garanhuns, PE 55292-901, Brazil. Ruminal acidosis is due to excessive ingestion of carbohydrates of rapid fermentation without previous adaptation of the microorganisms, causing severe metabolic disturbances to the animals. The objective of the present study was to assess the neutrophilic oxidative metabolism in sheep treated with sodium monensin in experimentally induced ruminal lactic acidosis. A total of 18 mate sheep, half-bred (Ideal x Merino), fistulated in the rumen, were used; nine of them received 33 mg/kg of the ionophore diet per day, for 30 days; the others were controls. The acidosis was induced by supplying 15g of sucrose/kg of body weight. The clinical evaluation and the rumen and blood samples were obtained before (Oh) and at 6, 12, 24 and 48 hours post-induction. In both groups, all the animals presented clinical manifestations of ruminal lactic acidosis 6 hours after the induction. From this period on, a significant pH decrease (P<0.05) was observed in the ruminal fluid, which reached levels below 5. There were relevant differences (P<0.05) between the groups 12 hours after the induction, when the sheep treated with monensin had higher values than those of the control group. During this period, the oxidative metabolism of the neutrophils remained inhibited, and the reestablishment of this function only occurred in the sheep which received monensin. Blood pH, plasmatic glucose and the ionizable calcium suffered alterations within its levels. The seric cortisol concentration rose significantly (P<0.05) in both groups, although differences (P<0.05) between them were found at the end of the observation period. The treatment with monensin did not influence the oxidative metabolism of the neutrophils inhibited by the lactic acidosis; however, a faster recovery of this metabolism was verified in the animals treated with the ionophore.

Pesquisa Veterinária Brasileira 22(4):129-134. Clinica de Bovinos, UFRPE, Cx. Postal 152, Garanhuns, PE 55292-901, Brazil.

E-mail: lgmv@alternet.com.br Rabbits, guinea pigs, rats and mice were used to reproduce the cardiac lesions observed in ruminants in the intoxication by Ateleia glazioviana. Four animals of each species were fed during four months with a ration containing 10% of dry A glazioviana. The pellets were made by mixing 700 g of commercial ration, 200 g of cornstarch, 1000 mi of distilled water and 100g of the dry plant. Later this mixture was dried at 100ºC during 16-20 hours. Four control animals of each species were given similar food, but containing dry ryegrass (Lolium multiflorum) instead of A glaziovidna. Water was offered ad libitum and a daily amount of green oats (Avena sativa) was given to rabbits and guinea pigs. No clinical signs of disease were observed during the experiment. Ali animals were weighted weekly in the last 5 weeks of the experiment and no differences were observed in weight gains between treated and control groups. At the end of dosing ali animals were euthanatized and necropsied. No macroscopic or histologic lesions were observed in the heart or other organs. The abortive properties of A glazioviana were studied in rats. One group of 11 pregnant rats was fed, from day 7 to day 21 of gestation, ration containing 10% of A. glazioviana, prepared in the sarne way than in the previous experiment. A control group received the ration with 10% ryegrass. Ali rats delivered normal litters. To determine a possible loss of toxicity of the plant during the pellet preparation, dry A. glazioviana was heated for 16-20 hours at 100ºC and administered orally to one ewe. This sheep ingested daily 2.65 g/kg bw of dry A. glazioviana for 26 days. During the experimental period the animal had tachycardia and cardiac arrhythmia. At the end of the experiment bradycardia and severe depression were observed. It was euthanatized 7 days after the last plant administration. The sheep was 2.5-3 months pregnant and the fetus was apparently normal with no autolysis. Ascites and hydrothorax were observed at necropsy. Whitish areas were observed in the myocardium and the liver was pale. Degeneration and necrosis of cardiac fibers and proliferation of fibrous tissue were observed histologically in the heart. The liver had centrilobular degeneration of hepatocytes and congestion. These experiments indicate that A. glazioviana is not cardiotoxic for laboratory animals and suggest that the plant has a heat-stable active principie similar to the active principie of plants causing cardiac fibrosis (gousiekte) in Southern Africa.

RESUMO.- Leite L.G., Riet-Correa F., Medeiros R.M.T., Piacenti A., Aragão M. & Schons S.V. 2002. Susceptibility of laboratory animals to the intoxication by Ateleia glazioviana (Leg. Papilionoideae).]

ABSTRACT.- Schenk M.A.M., Mendonça C.L., Madruga C.R., Kohayagawa A. & Araújo F.R. 2001. [Clinical and laboratorial evaluation of Nellore cattle experimentally infected with Trypanosoma vivax] Avaliação clínico and laboratorial de bovinos nelore infectados experimentalmente com Trypanosoma vivax. Pesquisa Veterinária Brasileira 21(4):157-161. Embrapa Gado de Corte, Cx. Postal 154, Campo Grande, MS 79002-970, Brazil. In arder to evaluate the clinical-laboratorial alterations, six Neil ore calves were inoculated with 107 Trypanosoma vivax isolated from Poconé region, Mato Grosso, Brazil. The animals were evaluated daily for rectal temperature, packed cell volume (PCV), parasitemia, antibody production, calor of mucous membranes, behavior and appetite. Blood and serum samples for biochemical evaluation for aspartate aminotransferase (AST), alkaline phbsphatase (AF), gamma glutamyltransferase (GGT), cholesterol, urea, creatinine, creatine kinase (CK), calcium, phosphorus and proteinogram were collected on days 4, 8, 12, 16, 23 and 30 post inoculation (DPI). During the following 6 months rectal temperature, PCV and parasitemia were evaluated weekly. T. vivax was evidenced from 1 DPI in all calves and persisted until day 30 in five of six animals. A remarkable decrease (p<0.05) of PCV mean value (25%) was observed on 10 DPI. The animals presented no alterations in their clinical or serum biochemical state during the trial. Seroconversion took place 6 and 8 DPI, and all the animals remained seropositive during the 30 days of experiment. In all the experimental animals the occurrence of T. vivax infection was verified, characterized by the increase of corporal temperature, presente of the blood protozoa and reduction of the globular volume, without altéations in the other variables analyzed. Nellore calves, when experimentally inoculated with T. vivax, are able to establish a balance between host-parasite relationship.

RESUMO.- Schenk M.A.M., Mendonça C.L., Madruga C.R., Kohayagawa A. & Araújo F.R. 2001. [Clinical and laboratorial evaluation of Nellore cattle experimentally infected with Trypanosoma vivax] Avaliação clínico and laboratorial de bovinos nelore infectados experimentalmente com Trypanosoma vivax. Pesquisa Veterinária Brasileira 21(4):157-161. Embrapa Gado de Corte, Cx. Postal 154, Campo Grande, MS 79002-970, Brazil.
