This caused compressive atrophy with thinning and medial deviation of the corresponding submaxillary bone and exposition of the roots of molar teeth. The seeds appear to be of good palatability to the buffaloes.

#92 - Produção de progesterona in vitro pelas células do corpo lúteo bovino ao longo da gestação, p.370-376

[In vitro progesterone production from bovine corpus luteum throughout gestation.] Produção de progesterona in vitro pelas células do corpo lúteo bovino ao longo da gestação. E-mail: ppapa@usp.br The aim was to test the hypothesis that cultivated bovine luteal cells from three different thirds of pregnancy behave the same way as in vivo luteal cells relative to P4 production. Corpus luteum samples from days 90 (n=3), 150 (n=3) and 210 (n=3) of pregnancy were obtained at a local slaughterhouse. Under aseptic conditions cells were mechanically dispersed and cultivated in a 96 wells-plate. After 24 hours of culture, cells were washed and the precursor pregnenolone was added. Experiments were conducted eight times for each studied time period (24, 48 and 96 h) and three times for each gestational age. Culture medium and cells were collected after 24, 48 and 96 hours of precursor addition and kept frozen at -20oC until processing. Progesterone was measured by RIA and protein content by Lowry’s method. Results were statistically analyzed and considered different when p <0.05. A higher P4 production was observed on day 90 of gestation (35.277±0.075), then this production was decreased at day 150 (28.820±0.231) and increased again at day 210 (32.777±0.099). After 24 hours of culture, luteal cells P4 production reached maximum values in the group of 90 days (2.912±0.047) when compared to 150 (2.669±0.137) and 210 days (2.741±0.088). At 48 and 96 hours of culture, bovine luteal cells from day 90 of gestation produced more P4 than cells from day 210 (2.934±0.029 and 2.976±0.121 respectively x 2.760±0.059 and 2.695±0.149, respectively; p<0.05), which in turn, produced more P4 than cells from day 150 (2.334±0.084 for 48 h and 2.205±0.136 for 96 h). Luteal cells from day 150 of gestation presented a decreasing P4 production throughout the 96 hours of culture. These differences could be explained by differential gene expression of enzymes and/or factors belonging to the esteroidogenic cascade in accordance to the gestational period. The established luteal cell culture model could be used for further functional studies once P4 secretion pattern in vitro resembled what occurs in vivo.

#93 - Experimental poisoning by Phalaris angusta (Gramineae) in cattle., 19(3/4):116-122

ABSTRACT.- Sousa R.S. & lrigoyen L.F. 1999. [Experimental poisoning by Phalaris angusta (Gramineae) in cattle.] Intoxicação experimental por Phalaris angusta (Gramineae) em bovinos. Pesquisa Veterinária Brasileira 19(3/4):116-122. Depto Patologia, Universidade Federal de Santa Maria, 97105-900 Santa Maria, RS, Brazil. To investigate the effects of the ingestion of different amounts of Phalaris angusta, eight 6-8-month-old calves were assigned to four treatment groups. Group I calves were fed only P. angusta. Group II calves were feda mixture of 75 % of P. angusta and 25% of oat (Avena sativa) and ryegrass (Lolium multiflorum), whereas group III calves received a mixture of 50% of P.angusta and 50% of oat and ryegrass. Group IV calves received only oat and ryegrass and served as contras. P. angusta was toxic for calves. One group I calf died 34 days after the beginning of the ingestion of the plant, and the other calves were euthanatized in extremis 18 to 32 days after de beginning of the experiment. The main clinical signs were locomotor disorders, generalized tremors, frequent falis, and convulsions. Gross findings were confined to the brain and consisted of focal areas of green-bluish discoloration in the thalamus, mesencephalon, and medulla oblongata. Microscopic lesions were characterized by the presence of a intracytoplasmic yellow-brown indole-like pigment in neurons from grossly affected areas. Ultrastructural changes consisted of swollen lysosomes containing membranous material with variable orientation and density. The amount of ingested P. angusta was not a determinant factor for the severity of clinical signs and lesions. The extension of gross and microscopic lesions was not directly related with the intensity of the clinical signs. P. angusta was exclusively neurotoxic and should be considered as a differential diagnosis in cases of neurological disease of cattle with clinical signs consistent with tremorgenic syndrome.

RESUMO.- Sousa R.S. & lrigoyen L.F. 1999. [Experimental poisoning by Phalaris angusta (Gramineae) in cattle.] Intoxicação experimental por Phalaris angusta (Gramineae) em bovinos. Pesquisa Veterinária Brasileira 19(3/4):116-122. Depto Patologia, Universidade Federal de Santa Maria, 97105-900 Santa Maria, RS, Brazil. Para investigar os efeitos da ingestão de diferentes quantidades da planta Phalaris angusta em bovinos, oito bezerros, com idade variando entre 6-8 meses, foram divididos em 4 grupos com 2 animais cada. Os animais do grupo I receberam somente P. angusta na alimentação, enquanto que os animais do grupo II receberam P. angusta (75%), aveia (Avena sativa) e azevém (Lolium multiflorum) (25%). Os animais do grupo III receberam P. angusta (50%), aveia e azevém (50%) e os animais do grupo IV receberam somente aveia e azevém e serviram como controles. Todos os animais que ingeriram P. angusta adoeceram. Um animal do grupo I morreu 34 dias após o início da ingestão da planta e os outros animais foram sacrificados, in extremis, em um período que variou de 18 a 32 dias após o início do experimento. Os principais sinais clínicos observados foram alterações de locomoção, tremores generalizados, quedas e crises convulsivas. Alterações macroscópicas foram observadas apenas no encéfalo e eram caracterizadas por focos de coloração verde-azulada no tálamo, mesencéfalo e medula oblonga. Microscopicamente observou-se pigmento granular marrom-amarelado no citoplasma de neurônios das regiões macroscopicamente afetadas. Alterações ultra-estruturais consistiram de lisossomos contendo material com densidade e orientação variáveis. A quantidade de P. angusta ingerida não foi um fator determinante na gravidade do quadro clínico, nem na intensidade das lesões observadas. A intensidade dos sinais clínicos também não teve uma relação direta com a severidade das lesões macro e microscópicas. Phalaris angusta demonstrou ter ação exclusivamente neurotóxica e deve ser considerada no diagnóstico diferencial em casos de animais com sinais clínicos de origem nervosa, consistentes com síndrome tremorgênica.

Pesquisa Veterinária Brasileira 19(1):35-38. Depto Clínica e Patologia, Centro de Ciências Agroveterinárias, Universidade do Estado de Santa Catarina, Av. Camões 2090, Lages, SC 88520-000, Brazil. Two outbreaks of spontaneous poisoning in cattle by Phalaris angusta Neês ex Trin. (&quot;aveia louca&quot;, &quot;aveia de sangue&quot;) are reported in cattle. The outbreaks occurred in the State of Santa Catarina, Brazil, during the winters of 1993 and 1996. In both outbreaks the animals were held in paddocks where P. angusta was a prevalent forage. Clinical sigos in affected cattle included generalized tremors, alertness, hypermetria, ataxia and convulsions. Gross lesions were restricted to the brairi and consisted of gray to greenish discoloration in the thalamus and mesencephalon. The disease was experimentally reproduced in cattle by administration of P. angusta.

São relatados dois surtos de intoxicação natural por Phalaris angusta (&quot;aveia-louca&quot; ou &quot;aveia-de-sangue&quot;) em bovinos no Estado de Santa Catarina, nos invernos de 1993 e 1996. Nos dois surtos os animais estavam lotados em piquetes onde P. angusta era a planta dominante. Os sinais clínicos incluíam tremores generalizados, olhar atento, hipermetria, ataxia e convulsões. Alterações macroscópicas eram restritas ao encéfalo e se caracterizavam por coloração cinza-esverdeada no tálamo e mesencéfalo. A doença foi reproduzida experimentalmente em bovinos pela administração de P. angusta.

[Sudden death in cattle by Mascagnia sp (Malpighiaceae) in the State of Santa Catarina, Brazil.] Mortes súbitas em bovinos causadas pela ingestão de Mascagnia sp (Malpighiaceae), no Estado de Santa Catarina. Pesquisa Veterinária Brasileira 18(1):16-20. Laboratório de Patologia Animal, Centro de Ciências Agroveterinárias, UDESC, Av. Luiz de Camões 2090, Lages, SC 88520-000, Brazil. In the coastal areas of the State of Santa Catarina occurs a disease in cattle characterized by &quot;sudden death&quot;. The disease was reproduced in bovines by oral administration of Mascagnia sp. Single doses of 5 g/kg of the fresch leaves of Mascagnia sp caused non-lethal poisoning; single doses of 7,5 g/kg caused letharpoisoning in one of two bovines and doses of 10 g/kg death in two others. In the experiments clinical manifestations were observed when the animals were exercised. They consisted in weakness, swollen jugular vein, slight muscular tremors and sometimes sudden contractions; tachycardia, already observed before the exercise, became more intense. At the end the animals suddenly laid down or fell on their side. The course of poisoning in those two animals which were followed up till death, was 40 and 75 minutes. The highest doses given (15 and 20 g/kg) caused a protratecd course; the animals got slow and apathic, avoiding any movements. They were found dead, 7h45min and 21 hours after the first clinical signs. These two last experiments show the importance of exercise regarding the onset of symptoms and the occurrence of &quot;sudden death&quot;. The main post-mortem findings were intense red discoloration of the mucosa of the small intestine and oedema of the gallbladder wall. The main histological alteration was hydropic vacuolar degeneration of the tubular epithelium of the kidney in three of the five bovines which died.

Finalmente os animais se deitavam ou caíram subitamente. A evolução da intoxicação nos dois animais em que foi acompanhada até a morte, foi de 40 e 75 minutos. As mais altas doses administradas (15 e 20 g/kg) provocaram um quadro de intoxicação protraída; os animais se mostraram lerdos e apáticos, evitando quaisquer movimentos; foram encontrados mortos 7h45min e 21 horas após terem sido observados os primeiros sinais clínicos. Esses dois últimos experimentos mostram que a movimentação é um fator importante para a manifestação da &quot;morte súbita&quot;. Os principais achados de necropsia foram coloração vermelha intensa da mucosa do intestino delgado e edema da parede da vesícula biliar. As mais importantes alterações histológicas foram degeneração hidrópico-vacuolar do epitélio tubular renal em três dos cinco bovinos que morreram.


#96 - Reproductive disturbances in Bos indicus, B. taurus and crossbred bulis, in the Amazon region

Abstract in English:

A total of 164 Bos indicus and crossbred maintained in extensive conditions and B. taurus bulis, in semi-extensive conditions, were submitted to at least three clinico-andrological examinations and semen collections using the electroejaculation method for semen evaluation. Ninety five (57.9%) of the bulis were of Nelore breed: 80 (84.2%) of them presented a normal reproductive status, but 4 (4.2%) showed testicular degeneration, 1(1.1%) hypoplasia, 3(3.2%) delayed sexual maturity, 3 (3.2%) seminal vesiculitis, 1 (1.1%) acrobustitis, 2 (2.1%) Brucellosis and 1 (1.1%) fibrosis in the epididymis. Among the B. taurus bulis, 29 (17.7%) of the Holstein Friesian type, 21 (72.4%) showed testicular degeneration and 1 (3.2%) fibrosis in the epididymis: in 11 (6.7%) crossed Holstein Friesian x Zebú, only 3 (27.3%) were normal, 7 (63.6%) had testicular degeneration, 1 (9.1 %) total unilateral hypoplasia. Among 20 (12.1 %) crossbred bulis (Chianina x Nelore) 13 (65.0%) were normal, 7 (35.0%) showed testicular degeneration. Among 9 (18.2%) Pitangueiras bulis (3/8 Red Danish x 5/8 Guzerá), 5 (55.6%) were normal, 1 (11.1%) showed testicular degeneration and 3 (33.3%) had delayed sexual maturity. Among the observations made by the clinico-andrological examination and the sperm picture, the volume, concentration, mass activity and impairment of the different parts of the genital system as well as sperm abnormalities were observed. In some cases, the bulis were slaughtered or castrated and the testis or other segments of the genital system were submitted to histological examination to confirm the diagnosis.

Foram examinados um total de 164 reprodutores Bos indicus e mestiços, submetidos a manejo extensivo e, animais B. taurus e mestiços submetidos a manejo semi-intensivo, recebendo alimentação suplementar ao cocho. Os animais foram submetidos a mais de três exames clínico-andrológicos seguidos de colheita de sêmen pelo método de eletroejaculação, com a realização de espermiogramas. Dentre os animais examinados 95 (57,9%) eram da raça Nelore, sendo que 80 (84,2%) eram normais, 4 (4,2%) apresentavam um quadro de degeneração testicular, 1 (1,1 %) com hiplasia bi-lateral total, 3 (3,2%) maturidade sexual rétardada, 3 (3,2%) vesiculite, 1 (1,1 %) acrobustite, 2 (2,1%) brucelose, 1 (1,0%) fibrose do epidídimo. De 29 (17,7%) animais B. taurus do tipo Holandês Preto e Branco, 21 (72,4%) apresentavam um quadro de degeneração testicular e 1 (3,4%) com fibrose do epidídimo. Dos animais mestiços com Holandês Preto e Branco, em 11 (6,7%) animais examinados somente 3 (27,3%), eram normais; em 7 (63,6%) foi detectado degeneração testicular e somente 1 (9,1 %) hiplasia do tipo uni-lateral total. Dos 20 (12,2%) touros mestiços de Chianino x Nelore, 13 (65,0%) eram normais; 7 (35,0%) apresentavam degeneração testicular, e finalmente nos 9 (18,2%) touros Pitangueiras (3/8 Red Danish x 5/8 Guzerá), 5 (55,6%) apresentavam-se normais, 1 (11,1%) com um quadro de degeneração testicular e 3 (33,3%) com maturidade sexual retardada. Em alguns casos os animais foram submetidos ao abate ou castração, tendo os testículos e outras partes do sistema genital submetidos a processamento histológico e examinados ao microscópio para confirmação dos diagnósticos.
