#181 - Gross anatomical features of the oropharyngeal cavity of the ostrich (Struthio camelus), 31(6):543-550

E-mail: catarina.tivane@uem.mz Most descriptions of the ostrich oropharynx and oesophagus are superficial and supply little meaningful morphological data. The aim of this investigation is describe the ostrich oropharingeal cavity, in order to supply the deficiency of macroscopic data about this important animal. Five heads of 12 to 14-month-old ostriches of either sex were anatomically dissected to expose the oropharynx. The ostrich oropharynx was “bell-shaped” composed by the maxillary and mandibular ramphoteca. The roof and floor presented two distinct regions different in colour of the mucosa. The rostral region was pale pink contrasting to creamy-pink coloured caudal region. The median longitudinal ridge extended rostrally from the apex of the choana to the tip of the beak in the roof and it is clearly more prominent and rigid than the homolog in the floor that appeared thin and stretched rostrally, continuing caudally surrounding the tongue and the laryngeal mound eventually merging with the oesophageal mucosa. The floor was formed by the interramal region, tongue and laryngeal mound containing shield-shaped glottis. It can be concluded that the present study, in addition to confirming the basic features of the oropharynx previously described for the ostrich, clarified the contradictory information presented in the literature and also provided new, unreported morphological data, some of which may be important when studying nutrition and health in these birds.

E-mail: catarina.tivane@uem.mz Os estudos já realizados sobre a cavidade orofaríngea da avestruz são escassos e não elucidam completamente sua morfologia. O objetivo desse estudo foi descrever macroscopicamente a cavidade orofaríngea de avestruzes com o intuito de suprir a deficiência de informação nesta espécie. Foram utilizadas 5 cabeças de avestruzes com doze a quatorze meses de idade dissecadas seguindo padrões anatômicos de modo a expor a orofaringe. A orofaringe de avestruzes tem formato de sino e é composta pelas ranfotecas maxilar e mandibular. O teto e o assoalho apresentavam duas regiões distintas diferindo quanto à coloração da mucosa. A porção mais rostral era de coloração rosa pálida contrastando com a coloração mais fortemente rosada da porção mais caudal. A ruga palatina mediana estendia-se rostralmente do ápice da coana até a ponta do bico. No teto esta era mais proeminente e rígida que a do assoalho que se apresentava delgada e se estendia ao longo da porção rostral da região interramal, continuando caudalmente ao redor da coana e laringe e estendendo-se até o esôfago. O assoalho estava formado pela região interramal, língua e uma laringe com uma glote em forma de escudo. Pode-se concluir o presente estudo, além de confirmar as características básicas da orofaringe da avestruz previamente descritas, clarificam a informação contraditória presente na literatura e também novas informações morfológicas não previamente descritas são destacadas de forma a amparar outros estudos sobre a nutrição e saúde destas aves.

For this purpose, a total of 198 chicks were placed 11 per pen with three pens per treatment. The different concentrations of HCl (1000ppm, 2000ppm and 3000ppm) and amproilum (at the dose rate of 125ppm) were given to the experimental groups in drinking water from 10 to 19th days of age. One group was kept as infected non medicated control and one as non infected non medicated control. At the 12th day of age, all the groups were inoculated orally with 75,000 sporulated oocysts except non infected non medicated control. Anticoccidial activity was evaluated on the basis of performance (weight gain, feed conversion ratio) and pathogenic (oocyst score, lesion score and mortality %age) parameters. Among HCl medicated groups, the maximum anticoccidial effect was seen in the group medicated with 1000ppm HCl followed by 2000ppm and 3000ppm HCl medicated groups. Amprolium and 1000ppm HCl were almost equivalent in suppressing the negative performance and pathogenic effects associated with coccidiosis (Eimeria tenella) challenge. In summary, the lower doses of HCl have the potential to be used as alternative to chemotherapeutic drugs for Eimeria tenella control. It is therefore suggested that further studies should be carried out to determine the possible minimum safe levels of HCl with least toxic effects to be used as anticoccidial.

[Morphology of the male genital organs and cloaca of Rhea americana americana.] Morfologia dos órgãos genitais masculinos e da cloaca da ema (Rhea americana americana). Pesquisa Veterinária Brasileira 31(5):430-440. Departamento de Zootecnia, Universidade Estadual de Maringá, Av. Colombo 5790, Maringá, PR 87020-900, Brazil. E-mail: tcsantos@uem.br The rhea (Rhea americana americana) is a bird that belongs to the group of the Ratitas, order Rheiforme and family Rheidae. Macroscopic and microscopic morphology of the male genital organ (testes, epididymis, deferent ducts, and phallus) and the cloaca were analyzed in 23 emas, four chicken (2 weeks old), young (3 to 10 months old), and twelve adult ones (3 years old), from Cooperativa Emas do Brasil, RS and from CEMAS, Mossoró, RN. The testis of rhea had elongated shape and were located inside coelomatic cavity, in dorsal region of abdominal cavity, with medium length and width of 7.6±1.2cm and 2.6±0.7cm at adult animals; 4.5±1.5cm and 0.9±0.4cm at young animals; and 0.8±0.3cm, and 0.2±0.1cm at chicken. The testis were recovered by the tunica albuginea and its parenchyma had seminiferous tubules composed by spermatogenic epithelium and by sustentation cells, and also interstitial tissue, with interstitial endocrine cells, connective tissue and vessels. At the adult animals were observed all the cells from spermatogenic lineage, whilst at the youngs with 3 months the seminiferous tubules had a smale lumen with spermatogonia and undifferentiated sustentacular cells. The efferents ductus were composed by a cubic ciliated epithelium, while the epididimydis duct had a columnar epithelium. The epididymis was elongated and fusiform closely to medial testis board. The deferent duct had sinuous stretch at adult animals, rectilineae at young animal, convolute at its medium portion, decreasing its sigmoid shape at caudal portion, next to cloaca. The epithelium was pseudostratified ciliated, irregular lumen at adult animal, and circular at young animal, closely with urether. The cloaca was divided into three segments: coprodeum, urodeum and proctodeum. At urodeum the deferent ducts discharged into papillas at the ventral side wall, next to fibrous phallus’s insertion. The phallus was a lymphatic fibrous organ, located at ventral wall, at the cloaca floor, and was composed by two portions: one rigid forked and twisted, and another simple spiraled and flexible, which normally was inverted. In forced exposition, the phallus had 14 cm in length. In a general way the Rhea genital organs shared the morphology from others birds, mainly those described to the ostrich.

Os testículos da ema possuem formato alongado e localizam-se na cavidade celomática, na região intra-abdominal dorsal, com comprimento e larguras médias de 7,6±1,2cm e 2,6± 0,7cm nos adultos; 4,5±1,5cm e 0,9±0,4cm nos jovens; e 0,8±0,3cm, e 0,2±0,1cm nos filhotes. O testículo está envolto pela túnica albugínea e seu parênquima possui túbulos seminíferos irregulares, compostos por epitélio espermatogênico e por células de sustentação, e pelo tecido intersticial, com as células endócrinas intersticiais, tecido conjuntivo frouxo e vasos. Nos adultos observaram-se todas as células da linhagem espermatogênica, enquanto nos jovens com 3 meses, os testículos apresentaram túbulos seminíferos com luz reduzidas, espermatogônias e células de sustentação indiferenciadas. Os ductos eferentes possuem um epitélio cúbico ciliado, enquanto no ducto epididimário o epitélio é columnar. O epidídimo apresentou-se alongado e fusiforme junto a margem medial do testículo. O ducto deferentes apresentou trajeto sinuoso nos adultos, retilíneo nos jovens, convoluto na sua porção média, diminuindo seu formato sigmóide em sua porção caudal, próximo à cloaca. O epitélio é pseudoestratificado e reveste a luz irregular nos adultos e circular nos jovens, mantendo proximidade com o ureter. A cloaca dividiu-se em três segmentos: o coprodeu, o urodeo e o proctodeo. No urodeu os ductos deferentes desembocaram em papilas na parede ventro-lateral, próximo a inserção do falo fibroso. O falo é um órgão fibroso linfático, localizado na parede ventral, no assoalho da cloaca, e apresentou duas porções: uma rígida bifurcada e contorcida, e outra simples espiralada e flexível, a qual normalmente esteve invertida. Em exposição forçada, o falo teve 14 cm de comprimento. De forma geral os órgãos reprodutores das emas compartilharam da morfologia de outras aves, principalmente aquelas descritas para os avestruzes.

Sparrows (Passer domesticus L.) as intermediary hosts of Toxoplasma gondii in poultry farms from the “agreste” region of Pernambuco, Brazil. Pesquisa Veterinária Brasileira 31(2):169-172. Laboratório de Doenças Infecto-Contagiosas dos Animais Domésticos, Departamento de Medicina Veterinária, Universidade Federal Rural de Pernambuco, Rua Dom Manoel de Medeiros s/n, Dois Irmãos, Recife, PE 52171-900, Brazil. E-mail: rinaldo.mota@hotmail.com This paper aimed to identify Toxoplasma gondii infection in house sparrows (Passer domesticus, Linneaus 1758) coming from poultry farms in the “agreste” region of the Brazilian state of Pernambuco. 151 sparrows (Passer domesticus) captured in eight broiler, egg layer and commercial laying poultry farms, were used. Indirect hemagglutination test was used to research anti-T. gondii antibodies. Animals that presented titration of 1:16 were destined to DNA research through Polymerase Chain Reaction (PCR) technique, followed by Nested-PCR. It was observed that, from 151 analyzed samples. 91 (60.3%) were reagents and 60 (39.7%) were not reagents. It was verified, through analysis of the distribution of infected animals frequency per farm, that in only one farm (12.5%) no animal reagent to T. gondii was captured. It was also observed that three (30.00%) of the ten samples destined to DNA research for T. gondii were positive to PCR and four (40.00%) were positive to Nested-PCR. Anti-T gondii antibodies occurrence and the molecular identification of the agent confirmed natural T. gondii infection in sparrows from poultry farms in Brazil. Other studies must be carried out to highlight the real importance of these animals in the epidemiological chain and their efficiency in the transmission of the parasite to felines. Therefore, researches that use parasite isolation and molecular techniques to determine genomic profile of the agent present in these poultry farms are needed.

Para a pesquisa de anticorpos anti-T. gondi utilizou-se o teste de hemaglutinação indireta, aqueles animais que apresentaram titulação 1:16 foram encaminhados para pesquisa do DNA por meio da técnica de Reação em Cadeia da Polimerase (PCR) seguida do Nested-PCR. Das 151 amostras analisadas observou-se que 91 (60,3%) foram reagentes e 60 (39,7%) não reagentes. Na análise da distribuição de freqüência dos animais infectados por granja constatou-se que em apenas uma (12,5%) não foi capturado animal reagente para T. gondii. Das dez amostras que foram encaminhadas para pesquisa do DNA do T. gondii, observou-se que três (30,00%) foram positivas ao PCR e quatro (40,00%) ao Nested-PCR. A ocorrência de anticorpos anti-T. gondii e a identificação molecular do agente confirmam a infecção natural por T. gondii em pardais em granjas avícolas no Brasil. Outros estudos devem ser conduzidos para elucidar a real importância destes animais na cadeia epidemiológica e sua eficiência da transmissão do parasito para felinos. Para tal serão necessárias pesquisas que utilizem técnicas de isolamento do parasito e molecular para determinar o perfil genômico do agente presente nestas granjas.


#186 - Cloacal microbiota identification and evaluation of the antimicrobial resistance in captive cracids from Rio Grande do Sul, Brazil, 30(12):1077-1082

[Cloacal microbiota identification and evaluation of the antimicrobial resistance in captive cracids from Rio Grande do Sul, Brazil.] Microbiota cloacal de cracídeos cativos no Rio Grande do Sul e sua susceptibilidade a antimicrobianos. Pesquisa Veterinária Brasileira 30(12)1077-1082. Centro de Ciências Rurais, Universidade Federal de Santa Maria, Camobi, Santa Maria, RS 97105-900, Brazil. E-mail: llovato@smail.ufsm.br Cracids are wildlife Galliformes which inhabits the America’s tropical forests. Fifty one cloacal swabs were collected from 10 different species of captive cracids from the Rio Grande do Sul State during 2007. The cloacal swab samples were submitted to bacterial isolation, identification and, subsequently; antimicrobial susceptibility testing. Ninety three bacterial isolates were obtained from the cracid population examined. The most prevalent among the isolates were Escherichia coli, and bacteria from the Staphylococcus and Streptococcus genera. All samples tested in this study were negative for Salmonella spp. The antimicrobial susceptibility tests showed that none of the 93 strains presented resistance to the antimicrobial imipinem. In addition, the lower percentages of resistance were observed against cloranfenicol and ciprofloxacine. The bacteria genus and species with the highest percentage of resistance to the different antimicrobials examined were E. coli, Serratia marcescens, Staphylococcus spp. and Streptococcus spp. In conclusion, the data presented in this article demonstrate that the cloacal microbiota of the reported cracid population is composed of several bacterial genera and species and multi-drug resistance may be a problem for the future, since some strains showed elevated percentage of resistance against several different antimicrobials.

Foram coletadas, no ano de 2007, amostras cloacais de 51 aves de dez espécies diferentes de cracídeos mantidos em cativeiros no Estado do Rio Grande do Sul. A partir dos swabs, colhidos assepticamente, foi realizado o isolamento e a caracterização bacteriana e o teste de susceptibilidade antimicrobiana dos isolados. Foram identificadas 93 cepas de bactérias. As bactérias mais frequentemente isoladas foram Escherichia coli, Staphylococcus spp. e Streptococcus spp. Todas as amostras foram negativas para o isolamento de Salmonella spp. O resultado do teste de sensibilidade mostrou que dentre as 93 cepas isoladas, todas foram sensíveis apenas ao imipinem. Adicionalmente, os menores percentuais de resistência foram observados frente ao cloranfenicol e ciprofloxacina. Os gêneros e espécies bacterianas com maior percentual de resistência a diferentes antibióticos testados foram Escherichia coli, Serratia marcescens, Staphylococcus aureus e Streptococcus spp. Com os resultados obtidos no presente trabalho, concluí-se, que a população de cracídeos estudada apresenta sua microbiota cloacal composta por vários gêneros e espécies bacterianas e que a multirresistencia pode ser um problema no futuro, uma vez que algumas cepas isoladas mostraram percentuais elevados de resistência a diferente antimicrobianos.

E-mail: debora@unipar.br This paper aims to analyze the effects of the Toxoplasma gondii infection in the intestinal wall and myenteric plexus of chicken (Gallus gallus). Ten 36-day-old chickens were separated into two groups: control and experimental, orally inoculated with oocysts of the T. gondii strain M7741 genotype III. After 60 days the birds were submitted to euthanasia and had their duodenum removed. Part of the intestinal segments was submitted to histological routine, HE staining, PAS histochemical technique, and Alcian Blue. Qualitative analysis of the intestinal wall and comparative measurements among the groups with respect to total wall thickness, muscle tunic, mucosa, and tunica mucosa were carried out. Caliciform cells were quantified. The other part of the intestinal segments was fixed in formol acetic acid and dissected having the tunica mucosa and the tela submucosa removed. Neurons were stained with Giemsa, counted, and measured. Chickens from the experimental group presented diarrhea and inflammatory infiltrates in the tunica mucosa, thickness reduction of all the parameters assessed in the intestinal wall, and an increase of the number of caliciform cells. There was a ~70% reduction regarding the intensity of myenteric neurons; and the remaining cells presented a reduction of ~2.4% of the perikarion and ~40.5% of the nucleus (p<0.05). Chronic infection induced by T. gondii oocysts resulted in intestinal wall atrophy, mucin secretion increase, death and atrophy of chicken myenteric plexus neurons. Death and atrophy of myenteric plexus neurons may be related with the causes of diarrhea observed in chickens with toxoplasmosis.

Parte dos segmentos intestinais foi submetida à rotina histológica, coloração por HE e técnica histoquímica de PAS e Alcian Blue. Realizou-se uma avaliação qualitativa da parede intestinal e medidas comparativas entre os grupos da espessura da parede total, túnica muscular, muscular da mucosa e túnica mucosa. As células caliciformes foram quantificadas. Outra parte dos segmentos intestinais foi fixada em formol acético e dissecada retirando-se a túnica mucosa e a tela submucosa. Os neurônios foram corados pela técnica de Giemsa, contados e mensurados. Os animais do grupo experimental apresentaram diarréia e infiltrados inflamatórios na túnica mucosa, redução da espessura de todos os parâmetros avaliados da parede intestinal e aumento do número das células caliciformes. Houve uma redução de ~70% da densidade dos neurônios mientéricos e as células remanescentes sofreram redução de ~2,4% do pericário e ~40,5% do núcleo (p<0,05). A infecção crônica induzida por oocistos de T. gondii levou a atrofia da parede intestinal, aumento da secreção de mucinas, morte e atrofia dos neurônios do plexo mientérico de galinhas. A morte e atrofia dos neurônios do plexo mientérico podem estar envolvidas na causa da diarréia observada em galinhas com toxoplasmose.

E-mail: dayseabreu@predialnet.com.br The pathogenicity of Escherichia coli strains is partially related to the expression of virulence factors genes, present in genetic elements called plasmids. APEC strains responsible for diseases in birds may present the iss gene which increases the resistance of E. coli strains to the lityc effect of the host’s serum, besides resistance to several antimicrobials. This study was conduced in order to detect E. coli in tracheae of meat-type quails and to evaluate, by the presence of the iss gene and the profile of antimicrobial susceptibility, the pathogenic potential of the isolated samples for birds and humans. One hundred and eighty tracheae of quails were collected for detection of E. coli, antimicrobial sensitivity tests, and for polymerase chain reaction (PCR), for detection of iss gene. From the examined quails, 8.9 % (16/180) were positive for E. coli, from which 20 strains of this bacterium were obtained. Most of them were resistant to Tetracycline (16/20), followed by Ceftadizime (13/20) and Nalidixic-acid (12/20) and only one isolate was resistant to Amoxicillin. The detection of iss gene occurred in 55% (11/20) of the isolates, indicating that these strains had the potential to be pathogenic not only for quails, but also for other kinds of birds, other animals and even human beings that would be in contact with these E. coli isolates.

Este estudo foi conduzido para detectar E. coli em traquéias de codornas destinadas ao abate e avaliar, pela presença do gene iss e o perfil de susceptibilidade antimicrobiana, o potencial patogênico para aves e humanos dos isolados obtidos. Foram coletadas 180 traquéias de codornas para detecção de E. coli, determinação do perfil de resistência a agentes antimicrobianos e posterior detecção, por reação em cadeia da polimerase (PCR), do gene iss. Das traquéias analisadas, 8,9 % (16/180) foram positivas para E. coli, sendo obtidos 20 isolados deste agente. A maioria dos isolados foi resistente à Tetraciclina (16/20), seguida pela Ceftazidima (13/20) e Ácido Nalidíxico (12/20), sendo apenas um resistente à Amoxicilina. A detecção do gene iss ocorreu em 55% (11/20) dos isolados. A presença do gene iss e a resistência a múltiplos antimicrobianos dos isolados obtidos neste estudo pode indicar um possível potencial patogênico das cepas de E. coli tanto para codornas quanto para outros tipos de aves e animais e mesmo para o ser humano que fique em contato com as mesmas.

E-mail: lucasbmoraes@gmail.com Fifty Bursa of Fabricius (BF) were examined by conventional optical microscopy and digital images were acquired and processed using Matlab® 6.5 software. The Artificial Neuronal Network (ANN) was generated using Neuroshell® Classifier software and the optical and digital data were compared. The ANN was able to make a comparable classification of digital and optical scores. The use of ANN was able to classify correctly the majority of the follicles, reaching sensibility and specificity of 89% and 96%, respectively. When the follicles were scored and grouped in a binary fashion the sensibility increased to 90% and obtained the maximum value for the specificity of 92%. These results demonstrate that the use of digital image analysis and ANN is a useful tool for the pathological classification of the BF lymphoid depletion. In addition it provides objective results that allow measuring the dimension of the error in the diagnosis and classification therefore making comparison between databases feasible.

Redes Neurais Artificiais (ANN) foram geradas com a utilização do software Neuroshell® Classifier, e os dados das análises óptica e digital foram comparados. A ANN classificou corretamente a maioria dos folículos, atingindo sensibilidade e especificidade de 89% e 96%, respectivamente. Quando os folículos foram agrupados de forma binária houve um aumento da sensibilidade para 90% e obteve-se um valor máximo para a especificidade de 92%. Estes resultados demonstram que o uso da análise digital de imagem associada à ANN é uma ferramenta bastante útil para a classificação patológica da depleção linfóide da BF. Além disso, fornece resultados objetivos que permitem medir a dimensão do erro classificatório, tornando possível a comparação entre distintos bancos de dados.

E-mail: olney@uenf.br Pythiosis is caused by Pythium insidiosum and the occurrence of disease in horses was described in the North and Northwest State of Rio de Janeiro, Brazil. The disease was described in cattle, sheep, humans, and horses in different states and regions across the country. This paper describes the development of IgY and IgG polyclonal antibodies, in chicken and rabbits, respectively against proteins extracted from kunkers and hyphae of P. insidiosum from affected horses. The proteins were recognized by chicken, rabbit and horse antibodies by immunodiffusion and Western blot against majority bands of 27 and 43 KDa, and titrated by ELISA. The antibodies IgY developed by the first time against Brazilian strains of P. insidiosum may represent a valuable tool in the detection of antigens of the pathogen and contribute to further studies aimed at immunotherapy and knowledge about this disease in endemic areas in Rio de Janeiro and in Brazil.
