#21 - Anthelmintic intoxication in small ruminants diagnosed in Northeastern Brazil

Abstract in English:

The clinical, epidemiological and anatomopathological findings of anthelmintic poisoning in small ruminants diagnosed at the Veterinary Hospital of the Federal University of Campina Grande, Patos, Paraíba, from January 2000 to December 2022 are described. A total of 1,928 necropsies were carried out on small ruminants, of which 1,032 were goats and 896 sheep. Four outbreaks of anthelmintic poisoning were identified in these species: one outbreak of ivermectin in sheep, one outbreak of disophenol in goats and sheep and two outbreaks of nitroxinil in goats. These intoxications accounted for 0.78% and 0.23% of goat and sheep diseases diagnosed routinely. In the case of ivermectin poisoning, the animal had neurological clinical signs, and no macroscopic or histopathological alterations were observed. In the outbreaks of disophenol and nitroxinil, the animals showed anorexia, hyperthermia, tachycardia, tremors, dyspnea, incoordination and limb rigidity. Macroscopically, there were liver and kidney lesions associated with circulatory alterations, such as hemorrhages and edema. Microscopically, necrosis and degeneration of hepatocytes and tubular epithelial cells associated with hemorrhage and congestion were observed. The prevalence of these intoxications in small ruminants is considered low in the Semi-arid Northeast, but outbreaks can occur with high lethality in herds. The main risk factor for the occurrence of intoxications was overdose, combined with other factors such as the age of the animals, high environmental temperature, constant exercise and low body score, which increased the toxicity of anthelmintics.

Descrevem-se os achados clínicos, epidemiológicos e anatomopatológicos das intoxicações por anti-helmínticos em pequenos ruminantes diagnosticadas no Hospital Veterinário da Universidade Federal de Campina Grande, Patos, Paraíba, durante o período de janeiro de 2000 a dezembro de 2022. Foram realizadas 1.928 necropsias de pequenos ruminantes, sendo 1.032 caprinos e 896 ovinos. Destes, foram identificados quatro surtos de intoxicação por anti-helmínticos nessas espécies: um surto de intoxicação por ivermectina em ovino, um surto de disofenol em caprinos e ovinos e dois surtos por nitroxinil em caprinos. Essas intoxicações representaram 0,78% e 0,23% das doenças de caprinos e ovinos diagnosticadas na rotina, respectivamente. Na intoxicação por ivermectina o animal apresentava sinais clínicos neurológicos e não foram observadas alterações macroscópicas e histopatológicas. Nos surtos de disofenol e nitroxinil, os animais demonstravam anorexia, hipertermia, taquicardia, tremores, dispneia, incoordenação e rigidez de membros. Macroscopicamente observou-se lesões hepáticas e renais associadas a alterações circulatórias como hemorragias e edemas. Microscopicamente observou-se necrose e degeneração de hepatócitos e das células epiteliais tubulares associadas a hemorragia e congestão. A prevalência dessas intoxicações em pequenos ruminantes é considerada baixa no Semiárido nordestino, entretanto, os surtos podem ocorrer com alta letalidade nos rebanhos. O principal fator de risco para a ocorrência das intoxicações foi a sobredosagem aliada aos demais fatores como idade dos animais, temperatura ambiental alta, exercício constante e escore corporal baixo, que provocaram o aumento da toxicidade dos anti-helmínticos.


#22 - Detection of multiple circulating sandflies species and investigation of dogs and vectors naturally infected with Leishmania in the city of Ribeirão Vermelho, southeastern Brazil

Abstract in English:

Leishmaniasis is a zoonotic disease caused by parasites of the Leishmania genus, resulting in various clinical forms, including a highly lethal visceral form. This study aimed to identify Leishmania spp. in sandflies and dogs in a small Brazilian town. DPP® Visceral Canine Leishmaniasis (CVL) was used for screening, and ELISA and conventional PCR were used for confirmatory testing, while sandflies were captured using CDC light traps and conventional PCR targeting ITS1. The Hill series was used to identify the diversity profile of species in the sampled area using R software. The study identified a Leishmania spp. prevalence of 4.02% in dogs, exceeding the 2% limit required by the Brazilian Ministry of Health. A total of 443 sandflies belonging to 14 different species were identified, with Lutzomyia longipalpis being the most abundant (73.81%). Negligence regarding leishmaniasis in small towns can lead to late diagnosis, hence the need to implement effective strategies, including early diagnosis and treatment of human and canine cases, vector control programs, and awareness campaigns to educate the public about risks and preventive measures. These measures can help prevent the spread of leishmaniasis and improve health outcomes for affected individuals and animals.

A leishmaniose é uma doença zoonótica causada por parasitas do gênero Leishmania, resultando em várias formas clínicas, incluindo uma forma visceral altamente letal. Este estudo teve como objetivo identificar Leishmania spp. DNA em flebotomíneos e cães em uma pequena cidade brasileira. A investigação usou DPP® Visceral Canine Leishmaniasis (CVL) para triagem, e ELISA e PCR convencional para teste confirmatório, enquanto flebotomíneos foram capturados usando armadilhas de luz CDC e PCR convencional visando ITS1. A série Hill foi utilizada para identificar o perfil de diversidade das espécies na área amostrada, utilizando o software R. O estudo identificou uma prevalência de Leishmania spp. em 4,02% dos cães, ultrapassando o limite de 2% exigido pelo Ministério da Saúde do Brasil. Foi identificado um total de 443 flebotomíneos, pertencentes a 14 espécies diferentes, com Lutzomyia longipalpis sendo o mais abundante (73,81%). A negligência com a leishmaniose em cidades pequenas pode levar ao diagnóstico tardio, daí a necessidade de implementar estratégias eficazes, incluindo diagnóstico precoce e tratamento de casos humanos e caninos, programas de controle de vetores e campanhas de conscientização para educar o público sobre riscos e medidas preventivas. Essas medidas podem ajudar a prevenir a propagação da leishmaniose e melhorar os resultados de saúde para indivíduos e animais afetados.


#23 - Identification of enteropathogenic Escherichia coli as the cause of mastitis in cows from Brazil

Abstract in English:

Escherichia coli is recognized as one of the main microorganisms responsible for triggering clinical mastitis, a disease that causes considerable economic losses in the dairy industry. In this context, this study aimed to identify E. coli isolates present in individual milk samples collected from cows diagnosed with clinical mastitis from various regions of Brazil. Additionally, through polymerase chain reaction (PCR), the presence of virulence genes eae, bfpB, escN, aatA, aggR, ipaH, stx1, stx2, est, and eltA was investigated; all associated with the pathotypes of diarrheagenic Escherichia coli (DEC). As an integral part of the study, a comprehensive assessment of the sensitivity profile of the isolates to 11 different antimicrobials widely used in mastitis treatment was also conducted. A total of 198 milk samples were collected from cows diagnosed with clinical mastitis. Among these samples, 12 isolates (6.07%) demonstrated bacterial growth greater than three Colony-Forming Units (CFU) when grown on MacConkey agar medium and morphological characteristics of E. coli. The disc-diffusion test was used to evaluate the susceptibility of these isolates to antimicrobials, and the most predominant resistance was observed concerning streptomycin and tetracycline, affecting 16.67% of the strains analyzed. Notably, all isolates investigated did not demonstrate the presence of the genes eae, aatA, aggR, ipaH, stx1, stx2, est, and eltA. These results indicate that these isolates do not fit the pathotypes known as diarrheagenic Escherichia coli (DEC). However, one of the isolates tested was positive for the bfpB and escN genes. The detection of resistant E. coli associated with clinical mastitis points to possible gaps in the treatment of the disease. Additionally, the presence of resistance genes in E. coli strains indicates the potential to transmit these genes between animals and, perhaps, along the food chain.

Escherichia coli é reconhecida como um dos principais microrganismos responsáveis pelo desencadeamento da mastite clínica, doença que causa perdas econômicas consideráveis na indústria de laticínios. Neste contexto, este estudo teve como objetivo principal a identificação de isolados de E. coli presentes em amostras individuais de leite coletadas de vacas com diagnóstico de mastite clínica, de diversas regiões do Brasil. Adicionalmente, por reação em cadeia da polimerase (PCR), foi investigada a presença dos genes de virulência eae, bfpB, escN, aatA, aggR, ipaH, stx1, stx2, est e eltA, todos associados aos patótipos de E. coli diarreiogênica (DEC). Como parte integrante do estudo, foi realizada uma avaliação abrangente do perfil de sensibilidade dos isolados a 11 antimicrobianos diferentes amplamente utilizados no tratamento da mastite. Foram coletadas 198 amostras de leite de vacas com diagnóstico de mastite clínica. Dentre essas amostras, 12 isolados (6,07%) demonstraram crescimento bacteriano superior a três Unidades Formadoras de Colônia (UFC) quando cultivadas em meio ágar MacConkey e características morfológicas de E. coli. Para avaliar a suscetibilidade desses isolados aos antimicrobianos foi utilizado o teste de disco-difusão, sendo observada a resistência mais predominante em relação à estreptomicina e à tetraciclina, afetando 16,67% das cepas analisadas. É relevante ressaltar que todos os isolados investigados não demonstraram a presença dos genes eae, aatA, aggR, ipaH, stx1, stx2, est e eltA. Estes resultados indicam que estes isolados não se enquadram nos patótipos conhecidos como Escherichia coli diarreiogénica (DEC). Porém, um dos isolados testados apresentou positividade para os genes bfpB e escN. A detecção de E. coli resistente associada à mastite clínica aponta para possíveis lacunas no tratamento da doença. Além disso, a presença de genes de resistência em estirpes de E. coli indica a capacidade potencial de transmitir estes genes entre animais e talvez ao longo da cadeia alimentar.


#24 - Crotalaria spectabilis Roth. (Fabaceae, Papilionoideae) seed poisoning in cattle and use of liver biopsy to diagnose subclinical disease

Abstract in English:

Crotalaria spectabilis seed poisoning has been reported in cattle on rural properties in Diamantino and Campo Verde municipalities in Mato Grosso, Brazil. The disease appeared 75 to 120 days after the animals were fed a diet contaminated with C. spectabilis seeds. Clinically, the animals showed a lack of appetite, progressive weight loss, apathy, photosensitivity, lateral recumbency, and death. Some animals developed neurological manifestations as a consequence of hepatic encephalopathy. In total, 37 cattle died, and six cattle were necropsied. The main macroscopic changes consisted of ascites and a firm and decreased liver with an irregular surface and multifocal pale yellowish discoloration. Microscopically, hepatocyte loss with fibrosis, megalocytosis and bile duct proliferation was observed. Animals with severe hepatic damage also had status spongiosus in the central nervous system. Furthermore, a hepatic biopsy of 15 cows who consumed contaminated grains but did not develop clinical disease revealed histologic liver changes similar to necropsied animals.

Relata-se dois surtos de intoxicação por sementes de Crotalaria spectabilis em bovinos em propriedades rurais nos municípios de Diamantino e Campo Verde, Mato Grosso, Brasil. A doença apareceu 75 a 120 dias depois que os animais foram alimentados com uma dieta contaminada com sementes de C. spectabilis. Clinicamente, foi observado falta de apetite, perda de peso progressiva, apatia, fotossensibilização, decúbito lateral e morte. Alguns animais desenvolveram manifestações neurológicas secundárias à encefalopatia hepática. No total, 37 bovinos morreram e seis deles foram submetidos à necropsia. As principais alterações macroscópicas consistiram em ascite e fígado diminuído em tamanho com superfície capsular irregular, difusamente firme e com áreas multifocais amarelo-pálidas. Microscopicamente, foi observado perda de hepatócitos acompanhada de fibrose hepática com megalocitose e proliferação de ductos biliares. No sistema nervoso central de bovinos com lesões hepáticas graves havia status spongiosus. A biópsia hepática de 15 bovinos que consumiram grãos contaminados, mas estavam clinicamente sadios, revelou alterações hepáticas similares às descritas nos animais submetidos à eutanásia.


#25 - Obesity outcomes on electrocardiographic, echocardiographic, and blood pressure parameters in cats

Abstract in English:

Obesity is considered a chronic inflammatory process that is related to metabolic impairment, respiratory distress, and cardiovascular disease. In cats, few studies have evaluated the implications of obesity on the cardiovascular system, and the existing literature is controversial. Therefore, the aim of this study is to detect pressure, electrocardiographic, and morphofunctional changes in overweight and obese cats. After clinical and laboratory evaluation, 45 animals were selected for the study. Cats were separated according to body condition score (BCS) into three groups (control, overweight, and obese). All animals underwent blood pressure measurement, electrocardiogram, and transthoracic echocardiogram. Results are presented as mean ± standard deviation (SD). Data were considered statistically significant at p<0.05. GraphPad Prism® 7.04 software was used for statistical analyses. Mean arterial pressure in obese cats was higher than in animals with ideal weight (CT 123.60 ± 8.97mmHg vs OB 143.00 ± 22.12mmHg, p<0.0138), but hypertension was not detected. On the electrocardiogram, P wave duration (CT 33.67 ± 1.56ms vs OB 37.76 ± 2.76ms; p<0.0003) and QRS complex (CT 48.14 ± 2.56ms vs OB 54.48 ± 5.51ms; p<0.002) were significantly higher in the obese group. There were no significant echocardiographic changes. There was a direct correlation between blood pressure and BCS (r:0.36, p<0.01). The P wave was positively correlated with the BCS (r:0.56, p<0.0001). Likewise, the QRS complex correlated directly with the BCS (r:0.52, p<0.0003). The results indicate cat obesity can directly affect the cardiovascular system, promoting pressure and electrocardiographic changes. Therefore, monitoring the cardiovascular system of cats with obesity is essential.

A obesidade é considerada um processo inflamatório crônico que está relacionada ao comprometimento metabólico, dificuldade respiratória e doenças cardiovasculares. Em gatos, poucos estudos avaliaram as implicações da obesidade no sistema cardiovascular e a literatura é controversa. Portanto, o objetivo deste trabalho é detectar as alterações pressóricas, eletrocardiográficas e morfofuncionais em gatos com sobrepeso e obesidade. Quarenta e cinco animais, após avaliação clínica e laboratorial, foram selecionados para o estudo. Os gatos foram separados conforme o escore de condição corporal (ECC) em três grupos (controle, sobrepeso e obesidade). Todos os animais realizaram mensuração de pressão arterial, eletrocardiograma e ecocardiograma transtorácico. Os resultados foram apresentados como média ± desvio padrão (DP). Os dados foram considerados estatisticamente significativos se p<0,05. O software GraphPad Prism® 7.04 foi usado para análises estatísticas. A pressão arterial média dos gatos obesos era maior que nos animais com peso ideal (CT 123,60 ± 8,97mmHg vs OB 143,00 ± 22,12mmHg, p<0,0138), porém não se detectou hipertensão. No eletrocardiograma, a duração da onda P (CT 33,67 ± 1,56ms vs OB 37,76 ± 2,76ms; p<0,0003) e complexo QRS (CT 48,14 ± 2,56ms vs OB 54,48 ± 5,51ms; p<0,002) foi significativamente maior no grupo obeso. Não houve alterações ecocardiográficas significativas. Houve correlação direta da pressão arterial com o ECC (r:0,36, p<0,01). A onda P correlacionou-se positivamente com o ECC (r:0,56, p<0,0001). Do mesmo modo, o complexo QRS se correlacionou diretamente com o ECC (r:0,52, p<0,0003). Os resultados indicam que a obesidade em gatos pode afetar diretamente o sistema cardiovascular, promovendo alterações pressóricas e eletrocardiográficas. Sendo assim, é indispensável o acompanhamento do sistema cardiovascular dos gatos com obesidade.


#26 - Measuring of the salivary immunoglobulin in Callithrix jacchus primates in captivity running title: salivary immunoglobulin in marmoset

Abstract in English:

The primary antibody in the mucous membranes and exocrine glands is a polymetric secretory immunoglobulin A. This immunoglobulin has been used as an indicator of stress in various animals, including nonhuman primates, and can also be used to monitor immune activity. The secretory immunoglobulin A is predominantly found in seromucous secretions such as saliva, tears, colostrum, milk, and liquor, as well as tracheobronchial, intestinal, and genitourinary secretions. This study aimed to measure the salivary IgA levels in Callithrix jacchus (common marmoset) by the enzyme-linked immunosorbent assay test (ELISA). Twelve pairs of animals were used, previously conditioned by the operant conditioning technique with positive reinforcement to saliva collection. Samples were collected once a week for six months. In this experiment, the salivary secretory immunoglobulin A concentrations in Callithrix jacchus were very low. We suggest new studies using other techniques to quantify the IgA quantities in the saliva of these animals.

O principal anticorpo presente nas mucosas e glândulas exócrinas é uma IgA polimétrica denominada sIgA. Esta imunoglobulina vem sendo utilizada como indicadora de estresse em diversos animais incluindo primatas não humanos, podendo também ser utilizada para o monitoramento da atividade imunológica. A imunoglobulina A secretora é encontrada predominantemente em secreções seromucosas, como saliva, lágrimas, colostro, leite, liquor, bem como secreções traqueobrônquicas, intestinais e geniturinárias. O objetivo deste estudo foi mensurar os níveis da IgA salivar em Callithrix jacchus (sagui comum) através do teste de imunoabsorção enzimática (ELISA). Doze pares de animais foram previamente condicionados pela técnica de condicionamento operante com reforço positivo à coleta de saliva. As amostras foram coletadas uma vez por semana por seis meses. Neste experimento as concentrações de imunoglobulina salivar A secretora em Callithrix jacchus foram muito baixas. Nós sugerimos novos estudos utilizando outras técnicas para quantificar a IgA na saliva desses animais.


#27 - Pathological aspects of a septicemic salmonellosis outbreak caused by Salmonella serotype Typhimurium in captive blue-fronted Amazon parrots (Amazona aestiva)

Abstract in English:

There are reports of septicemic salmonellosis in exotic birds caused by Salmonella enterica serotype Typhimurium, although they are scarce and do not address the anatomopathological and immunohistochemical aspects of this condition in blue-fronted Amazon parrots (Amazona aestiva). This study aims to report an outbreak of salmonellosis by S. Typhimurium in 15 blue-fronted parrots introduced to a rehabilitation center in the state of Mato Grosso do Sul, Brazil. Hepatomegaly and splenomegaly, accompanied by flat white areas in the capsule and parenchyma, were the most frequent gross changes, histologically represented by necrotic and heterophilic hepatitis and splenitis and associated with bacteria in the cytoplasm of macrophages and free in the affected tissue. S. Typhimurium was identified from bacterial culture, PCR, and DNA sequencing. Along with immunohistochemistry samples of liver, spleen, kidney, small intestine, pancreas, thyroid, heart, and lung, immunostaining was used for Salmonella spp. without previous bacterial isolation. The lethality of this outbreak demonstrates the relevance of salmonellosis in blue-fronted parrots, which are known to be carriers of this bacteria.

Apesar de existirem relatos sobre salmonelose septicêmica em aves silvestres por Salmonella enterica sorovar Typhimurium, eles são escassos e não abordam os aspectos anatomopatológicos e imuno-histoquímicos da condição em papagaios-verdadeiros (Amazona aestiva). Este trabalho visou relatar um surto de salmonelose por S. Typhimurium em 15 papagaios-verdadeiros introduzidos em um centro de reabilitação localizado no estado de Mato Grosso do Sul, Brasil. Hepatomegalia e esplenomegalia acompanhadas por áreas planas brancacentas na cápsula e no parênquima foram as alterações macroscópicas mais frequentes, representadas histologicamente por hepatite e esplenite necróticas e heterofílicas associadas a bactérias no citoplasma de macrófagos e livres no tecido afetado. S. Typhimurium foi identificada a partir de cultura bacteriana, PCR e sequenciamento de DNA. Na imuno-histoquímica, as amostras de fígado, baço, rim, intestino delgado, pâncreas, tireoide, coração e pulmão evidenciaram imunomarcação para Salmonella spp., mesmo quando não houve isolamento bacteriano prévio. A alta letalidade deste surto demonstra relevância da salmonelose em papagaios-verdadeiros, que são majoritariamente atribuídos como hospedeiros carreadores desta bactéria.


#28 - Serum and immunohistochemical analyses of troponin I in sheep experimentally poisoned with Palicourea marcgravii

Abstract in English:

Palicourea marcgravii is a lethal toxic plant widely distributed in Brazil. Ingestion of this plant causes cardiotoxic effects in animals, leading to acute heart failure without evident macroscopic changes in the heart due to the rapid progression of the disease. Currently, the diagnosis is confirmed based on microscopic identification of characteristic renal lesions. Although troponin is used as a biomarker for myocardial lesions in human and veterinary medicine, its serum levels in sheep poisoned with P. marcgravii remain unknown. The objective of this study was to determine serum levels of troponin I and evaluate its expression in incipient heart lesions in sheep. Eight male sheep were experimentally intoxicated with 1g kg-1 of fresh P. marcgravii plants. The animals were physically examined every two hours, and blood samples were collected before the administration of the plant material and during the agonizing period immediately before death. Additionally, necropsy and immunohistochemistry were performed on cardiac tissue samples. All animals presented minimal serum levels of troponin I before plant administration, with a significant increase in these levels immediately before death, indicating leakage due to the cardiac injury. These results confirm the hypothesis that troponin is released into the bloodstream before morphological changes in the myocardium can be observed through conventional microscopy and immunohistochemical testing. These findings contribute to understanding the pathological mechanisms of this toxicity and may assist in the early diagnosis and treatment of affected animals.

Palicourea marcgravii é uma planta tóxica letal e amplamente disseminada no Brasil. Sua ingestão causa efeitos cardiotóxicos em animais, levando à insuficiência cardíaca aguda sem alterações macroscópicas evidentes no coração devido à rápida progressão da doença. A confirmação do diagnóstico atual se baseia na identificação microscópica das lesões renais características. Embora a troponina seja usada como biomarcador de lesões miocárdicas na medicina humana e veterinária, seus níveis séricos em ovinos intoxicados por P. marcgravii ainda são desconhecidos. O objetivo deste estudo foi determinar os níveis séricos de troponina I e avaliar sua expressão em lesões incipientes no coração de ovinos. Oito ovinos machos foram experimentalmente intoxicados com 1g/kg da planta fresca. Os animais foram avaliados fisicamente a cada duas horas, e tiveram seu sangue coletado antes da administração da planta e durante a fase agônica, antes do óbito. Além disso, foram realizadas necropsia e imunohistoquímica em fragmentos cardíacos. Todos os animais apresentaram níveis mínimos de troponina antes da administração da planta, e mostraram aumento expressivo imediatamente antes do óbito, indicando extravasamento devido à lesão cardíaca. Esses resultados confirmam a hipótese de que a troponina é liberada na corrente sanguínea antes das alterações morfológicas no miocárdio serem observadas pela microscopia convencional e imunohistoquímica. Esses achados contribuem para a compreensão dos mecanismos patológicos dessa intoxicação e podem auxiliar no diagnóstico precoce e tratamento em animais afetados.


#29 - Supplementation with increasing doses of selenium associated with vitamin E in the treatment of bovines with enzootic hematuria

Abstract in English:

Bovine enzootic hematuria (BEH) is a clinical form of poisoning in cattle caused by the consumption of Pteridium spp. (bracken fern), which has no treatment. However, selenium (Se) and vitamin E supplementation are feasible. The aim of this study was to evaluate the effects of supplementation with increasing doses of selenium associated with vitamin E in cattle with BEH, compare the levels of Se in whole blood and blood serum, and evaluate the activity of glutathione peroxidase, total antioxidants, and the relative activity of the enzyme monoamine oxidase (MAO). Four groups of cattle with BEH were supplemented parenterally for 13 weeks with increasing doses of Se: Control group, Treatment group 1 (0.05mg/kg), Treatment group 2 (0.1mg/kg), and Treatment group 3 (0.2mg/kg). All groups received 500mg of vitamin E in combination with the Se supplementation. The measured variables included weight, hematuria intensity, hematocrit, total plasma protein, plasma fibrinogen, blood glutathione peroxidase activity, and total antioxidant levels. The blood concentrations of Se and relative MAO activity were evaluated every two weeks. Kruskal-Wallis and Friedman tests (P<0.05) were used to assess treatment and time effects, respectively, followed by Dunn’s multiple comparison test. For weight, total antioxidant concentration, and relative MAO, there was no significant difference (P>0.05) between the treatments, but there was a significant difference over time (P<0.05). For hematuria and hematocrit, there were significant treatment differences (P<0.05) but no significant time differences (P>0.05). For glutathione peroxidase, there was no significant difference (P>0.05) among the treatments, but there was a significant difference (P<0.05) between treatments M8 and M14 in Treatment group 3. There were no differences in the total plasma protein and fibrinogen levels between treatments or over time (P>0.05). Selenium levels were higher in Treatment group 1, reaching the highest concentration (235.3µg/L) in the blood serum at eight weeks. Therefore, supplementation with Se at a dose of 0.05mg/kg associated with vitamin E improved the clinical condition of hematuria but did not interfere with glutathione peroxidase activity or with levels of total antioxidants. MAO activity was reduced by vitamin E supplementation. These results indicate that the serum is the best site for measuring Se levels.

A hematúria enzoótica bovina (HEB) é uma forma clínica da intoxicação em bovinos causada pelo consumo de Pteridium sp. (samambaia) que não possui tratamento. Contudo, a suplementação utilizando selênio (Se) e vitamina E tem se mostrado viável. Objetivou-se avaliar os efeitos da suplementação com doses crescentes de selênio associado à vitamina E em bovinos com HEB, bem como, comparar os níveis de selênio em sangue total e soro sanguíneo, avaliar a atividade de glutationa peroxidase, os níveis de antioxidantes totais e a atividade relativa da enzima monoamina oxidase (MAO). Suplementou-se durante 13 semanas, por via parenteral, quatro grupos de bovinos com hematúria enzoótica (HEB), com doses crescentes de Se: Grupo controle, Grupo tratamento 1 (0,05mg/kg de Se), Grupo tratamento 2 (0,1mg/kg de Se) e Grupo tratamento 3 (0,2mg/kg de Se). Todos os grupos receberam em associação 500mg/animal de vitamina E. Quinzenalmente foram avaliadas as variáveis peso, intensidade de hematúria, hematócrito, proteína plasmática total, fibrinogênio plasmático, atividade sanguínea de glutationa peroxidase, níveis de antioxidantes totais, concentração sanguíneas de selênio e atividade relativa de monoamina oxidase. Utilizou-se os testes Kruskal Wallis e Friedman (P<0,05), para avaliar os efeitos dos tratamentos e do tempo, respectivamente, ambos seguidos do teste de comparação múltipla de Dunn. Para as variáveis peso, antioxidantes totais e atividade relativa de monoamina oxidase não houve diferença entre os tratamentos (P>0,05) e houve diferença ao longo do tempo (P<0,05). Para hematúria e hematócrito, houve diferença entre os tratamentos (P<0,05) e não houve diferença ao longo do tempo (P>0,05). Para a variável glutationa peroxidase, não houve diferença significativa entre os tratamentos (P>0,05), porém houve diferença entre os momentos M8 e M14, no Grupo tratamento 3 (P<0,05). Para proteína plasmática total e fibrinogênio não houve diferenças entre os tratamentos e nem ao longo do tempo (P>0,05). Os níveis de Se foram maiores no Grupo tratamento 1 atingindo a maior concentração no momento M8 no soro sanguíneo (235,3µg/L). Portanto, a suplementação com selênio na dose de 0,05mg/kg associada à vitamina E melhorou o quadro clínico de hematúria, mas não interferiu na atividade da glutationa peroxidase e nos níveis de antioxidantes totais. A atividade da monoamina oxidase foi reduzida globalmente pela suplementação de vitamina E. Estes resultados indicam que o soro foi o melhor local para medir o selênio.


#30 - Feline application/inflammation-associated sarcoma: Gross aspects and histomorphological

Feline injection-site sarcomas in felines account for more than 40% of cutaneous and subcutaneous neoplasms in felines. The present study aimed to describe the macroscopic and histomorphological findings of feline application/injection sarcomas. Samples from 31 feline tumors with a history of feline application/inflammation sarcoma were re-evaluated regarding histological subtype, mitotic index and score, depth of tissue invasion, and presence of inflammation considering the location, intensity and predominant cell types. Of the 31 samples from felines diagnosed with sarcoma at the application/inflammation site, 87.15% were cats with no defined breed (NDB), with a mean age of 8.5 years. The predominant anatomical sites were the back and flank/abdomen, both with 29% (9/31), and the prevalent histological subtype was fibrosarcoma at 77.4% (24/31), followed by anaplastic giant cell sarcoma at 12.9% (4/31) and myxosarcoma 9.6% (3/31). The histological grade with the highest number of cases was III (51.6%), followed by Grade II (35.4%) and I (12.9%). The mean of the longest axis measurements varied between the different tumor grades without being significant, with the average being 2.5±2.79cm in Grade I tumors and 3.2±2.28cm in Grade II tumors. and 4.68±2.07cm in Grade III tumors. Necrosis was observed in 74.2% of tumors. The tissue inflammation score was mild to moderate in 58% of cases and severe in 32.2%, with lymphocytic and lymphoplasmacytic infiltrates being prevalent, with 25.8% each, followed by lymphoplasmohistiocytic with 22.6%. The infiltration depth was 38.7% in muscle tissue, followed by 32.2% in the subcutaneous tissue. Pleomorphism was accentuated in 51.6%. Desmoplasia was moderate in 45.1%. Satellite nodules were present in 29% of cases, and 19.4% had macrophages with intracytoplasmic content suggestive of adjuvants. Surgical margins were infiltrated (M1) in 48.4% and narrowed in 25.8% (M2). The anatomical locations observed were different from those recommended by the Vaccine-Associated Feline Sarcoma Task Force (VAFST); in most cases, the adjuvanted macrophage was not present. From this data, we can suggest that sarcomas in felines are not only correlated to the vaccine application, corroborating the hypothesis that any material, whether liquid or solid, and any chronic inflammatory process in the subcutaneous tissue of cats can induce the entity if they are predisposed to do so. The detailed histomorphological data evaluated in this study were key points and provided important information about tumor behavior, being a tool for clinical-oncological decision-making.
