#51 - Prevalence of anti-Neospora caninum (Apicomplexa: Sarcocystidae) antibodies in dairy cattle in rural properties of three microrregions of Maranhão, Brazil, 30(9):729-734

E-mail: whaubtyfranct@yahoo.com.br The objective in the present study was to research the prevalence of anti-Neospora caninum in 812 samples of blood serum of dairy cattle from farms of seven municipalities of microrregions of Itapecuru-Mirim, Middle Mearim and President Dutra, state of Maranhão, Brazil. For the calculation of sample size, it was considered a seroprevalence of 34.7% for N. caninum, with a maximum error of 9.5% and a confidence interval of 95%. To detect antibodies, it was used the technique of Indirect Immunofluorescence (IFI), with the cut-off of 1:200, using as antigen, tachyzoites strain NC-1, maintained in cell culture in the Laboratory of Diagnosis of Parasitism of the Animals, School of Veterinary Medicine of the Federal University of Bahia, Brazil. Of the total samples, it was obtained a prevalence of 50.74%. The titles ranged from 1:200 to 1:6400, distributed as follows: 108 (26.21%) serum samples showed title of 1:200; 132 (32.04%) 1:400; 94 (22.81%) 1:800; 46 (11.16%) of 1:1600; 23 (5.58%) of 1:3200 and nine (2.18%) with titers of 1:6400. Among the microrregiões the Itapecuru-Mirim showed the lowest percentage of animals seropositive (20.69%) and President Dutra the largest (47.66%). It was observed higher prevalence of seropositives in females (46.80%) than in males (52.46%). There was no significant difference (P> 0.05) for the microrregions variables, sex and age. Concluded that the dairy cattle of the regions studied are exposed to infection by N. caninum.

Para a detecção da presença de anticorpos da classe IgG, utilizou-se a técnica de Imunofluorescência Indireta (IFI), com ponto de corte 1:200, usando como antígeno, taquizoítos da cepa NC-1, mantida em cultura celular no Laboratório de Diagnóstico das Parasitoses dos Animais da Escola de Medicina Veterinária da UFBA. Do total de amostras analisadas, encontrou-se uma prevalência de 50,74%. Os títulos variaram de 1:200 a 1:6400, assim distribuídos: 108 (26,21%) amostras de soro apresentaram título de 1:200; 132 (32,04%) 1:400; 94 (22,81%) 1:800; 46 (11,16%) 1:1600; 23 (5,58%) 1:3200 e nove (2,18%) com títulos de 1:6400. Dentre as microrregiões, a Itapecuru-Mirim apresentou o menor percentual de animais soropositivos (20,69%) e Presidente Dutra o maior (47,66%). Com relação à variável sexo, observou-se maior prevalência de sororreagentes nas fêmeas (46,80%) do que nos machos (52,46%). Não se verificou diferença significativa (P>0,05) para as variáveis microrregiões, sexo e idade. Conclui-se que os bovinos leiteiros das regiões estudadas estão expostos à infecção por N. caninum.

E-mail: gugatoscan@hotmail.com Neospora caninum and N. hughesi are protozoa which can infect horses and can cause reproductive and neurological diseases, respectively. The pathogenesis of neosporosis in horses is poorly understood, as well as the sources of horizontal infection of N. hughesi. Furthermore, there are doubts about the role of the vertical transmission of Neospora spp. in maintenance of these parasites in equine populations. In this study, we evaluated: (1) the occurrence of infections by Neospora spp. in a population of mares (in reproductive age) on a farm of Crioula breed horses; and (2) the possible association between the serological status of mares and of their offspring, aiming to investigate, indirectly, the relevance of transplacental transmission for the occurrence of Neospora spp. in these horses. We found a highly significant association between the serological status of mares and their offspring. Although had been exposed to the same environmental risk factors, the descendants of seropositive mares had a higher percentage of seropositivity against Neospora spp. compared to the descendants of seronegative mares. The association between kinship and serological status indicates an influence of vertical (transplacental) infection raising the occurrence of Neospora spp. in the studied equine population.

A patogenia da neosporose é pouco conhecida nos equinos, bem como as fontes de infecção horizontal de N. hughesi. Além disso, há dúvidas quanto ao papel da transmissão vertical de Neospora spp. na sua manutenção em populações equinas. Neste estudo avaliaram-se: (1) a ocorrência da infecção por Neospora spp. na população de éguas em idade reprodutiva em um haras de cavalos da raça Crioula; e (2) a possível associação entre o status sorológico destas éguas com o de suas crias, como meio de investigar, indiretamente, a relevância da transmissão transplacentária na ocorrência da infecção por Neospora spp. nestes animais. A associação entre o status sorológico das éguas e o de suas crias foi altamente significativa. Os animais descendentes de éguas soropositivas tiveram maior ocorrência de anticorpos anti-Neospora spp. do que os descendentes de éguas soronegativas, embora expostos aos mesmos fatores de risco ambientais. A associação entre parentesco em primeiro grau e status sorológico indica a influência da infecção vertical (transplacentária) na ocorrência de Neospora spp. na população equina estudada.

E-mail: eduardofurtadoflores@gmail.com Bovine herpesviruses 1 (BoHV-1) and 5 (BoHV-5) are antigenic and genetically related viruses associated with different clinical syndromes in cattle, including respiratory, reproductive, neurological disease and abortion. Epidemiological studies indicate the widespread distribution of both viruses among Brazilian cattle. Serological diagnosis, that allows the identification of latently infected animals, represents an important tool for individual and herd monitoring. The present article describes the standardization of a monoclonal antibody (MAb)-based immunoenzymatic test (ELISA) for detection of antibodies to BoHV-1 and/or BoHV-5. The initial steps involved the determination of the most suitable MAb, the appropriate dilutions of viral antigen and serum samples, and the cut-off value of the assay. After standardization, the ELISA was validated by testing 506 cattle serum samples previously tested for neutralizing antibodies to BoHV-1 and BoHV-5 by virus neutralizing assay (VN). Comparing to the VN for BoHV-1 antibodies, the ELISA presented sensitivity and specificity of 96.6% and 98.3%, respectively. Positive and negative predictive values were 97.6%, the concordance between the tests was 97.6% and the coefficient of correlation k (kappa) was 0.95, demonstrating an excellent correlation. Comparing to the VN for BoHV-5 antibodies, the ELISA presented 94.3% of sensitivity, 97.9% of specificity, 97.1% of positive predictive value, 95.9% negative predictive value, concordance of 96.4% and kappa coefficient of 0.92. These results demonstrate that the ELISA presents suitable specificity and sensitivity to be used for individual and herd serological diagnosis of BoHV-1 and BoHV-5, thus, representing an alternative for VN assays and imported ELISA kits.

O diagnóstico sorológico, que permite identificar animais portadores da infecção latente, se constitui em importante ferramenta para monitoramento individual e de rebanho. O presente artigo relata a padronização de um teste imunoenzimático do tipo ELISA, com base em anticorpo monoclonal (AcM), para a detecção de anticorpos séricos que reagem contra BoHV-1 e/ou BoHV-5. Inicialmente, determinou-se o AcM mais adequado para a sensibilização das placas, as diluições apropriadas do antígeno e dos soros-teste e o ponto de corte do ensaio. Após a padronização, o ensaio foi validado testando-se 506 amostras de soro bovino, previamente testadas para anticorpos neutralizantes contra BoHV-1 e/ou BoHV-5 pela técnica de soroneutralização (SN). Comparando-se com os resultados da SN frente a BoHV-1, o teste de ELISA apresentou sensibilidade e especificidade de 96,6% e 98,3%, respectivamente. Os valores preditivos positivo e negativo foram de 97,6%, a concordância foi de 97,6% e o índice de correlação kappa entre os testes foi de 0,95, o que indica uma excelente concordância. Comparando-se com os resultados da SN frente o BoHV-5, o ELISA apresentou 94,3% de sensibilidade; 97,9% de especificidade; 97,1% de valor preditivo positivo e 95,9% de valor preditivo negativo. Para BoHV-5, a concordância entre os testes foi de 96,4% e o índice de correlação foi de 0,92, também excelente. Esses resultados demonstram que o teste padronizado apresenta sensibilidade e especificidade adequados para o diagnóstico sorológico das infecções por BoHV-1 e BoHV-5 em nível individual e de rebanho. Dessa forma, o ensaio pode se constituir em alternativa para o teste de SN e para os kits de ELISA importados.

E-mail: spatroca@yahoo.com.br To investigate the occurrence of antibodies IgG anti-Neospora caninum and Toxoplasma gondii, blood samples were collected from 374 adult female water buffaloes originated from 14 production units located in 13 counties of the state of Pará. The sera were subjected to indirect fluorescent antibody (IFA), using titers of 200 and 64 as IFA cut off points for N. caninum and T. gondii, respectively. Hundred and fifty three (40.9%) buffaloes were sero-positive for N. caninum, with 100% of the properties being sources of infection. Four buffaloes (1.1%) in four farms were seropositive for T. gondii, and one buffalo cow (0.27%) was seropositive for the two parasites. The presence of antibodies indicates that these protozoan parasites are circulating among buffaloes from the properties studied, representing a source of infection to other animals, as well as a possible cause of reproductive disorders in this species.

Obteve-se 153 (40,9%) de animais soropositivos para N. caninum com 100% das propriedades com focos da infecção, enquanto que quatro búfalas (1,1%) foram soropositivas em quatro fazendas para T. gondii e uma búfala (0,27%) foi soropositiva para ambos parasitos. A presença de anticorpos é um indicativo da circulação desses protozoários em búfalos das propriedades estudadas, representando uma fonte de infecção para outros animais, assim como o possível envolvimento em distúrbios reprodutivos nessa espécie.


#55 - Risk factors and presence of antibodies to Toxoplasma gondii in dogs from the coast of São Paulo State, Brazil, 30(2):161-166

E-mail: silva_rcd@yahoo.com.br Toxoplasmosis is caused by the obligate intracellular protozoan parasite Toxoplasma gondii and affects warm-blooded vertebrates, including pets and man. Dogs are epidemio-logically important since they act as sentinels for the infection in humans. The present study aimed to determine the presence of antibodies to T. gondii in 205 serum samples from dogs in Ubatuba, Sao Paulo state, Brazil, through indirect immunofluorescence reaction (IFAT), as well as the risk factors related to toxoplasmosis in the animals such as breed, age, sex, access to outdoors, homemade food ingestion, access to untreated water, and contact with rodents. Toxoplasmosis-positive samples accounted for 52/205 (25.4%), with titers ranging from 16 to 256. The serological results presented significant association (P<0.05) with homemade food ingestion (45/118; 38.1%; CI95% 29.9%-47.2%) (OR=7.0; CI95% 3.0-16.6), and with access to outdoors where those that do not have access to the street were prevalent (37/121; 30.6%; CI95% 23.1%-39.3%) (OR=0.5; CI95% 0.2-1.0). These results show that toxoplasmosis in this region is related to problems of sanitary education, mainly concerning the appropriate cooking of foods, since most positive animals did not show significant association with the presence of rodents or untreated water consumption but showed, instead association with ingestion of homemade food. Thus, toxoplasmosis is a public health problem in the studied region, and sanitary measures are needed to control the infection due to the strict relationship between man and dog and the presented risk factors.

O presente estudo objetivou determinar a ocorrência de anticorpos para T. gondii em 205 amostras de soro de cães do município de Ubatuba, SP, Brasil, pela reação de imunofluorescência indireta (RIFI), assim como os fatores de risco relacionados à infecção nos animais, como raça, idade, sexo, acesso a rua, ingestão de comida caseira, acesso a água não tratada e presença de roedores. 52/205 (25,4%) amostras foram positivas para toxoplasmose, com títulos variando de 16 a 256. Os resultados sorológicos apresentaram associação significativa (P<0,05) com consumo de comida caseira (45/118; 38,1%; IC95% 29,9%-47,2%) (OR=7,0; CI95% 3,0-16,6), e acesso a rua, em que aqueles que não tinham acesso a rua foram prevalentes (37/121; 30,6%; IC95% 23,1%-39,3%) (OR=0,5; CI95% 0,2-1,0). Estes resultados demonstram que a toxoplasmose na região está relacionada com problema de educação sanitária, principalmente quanto ao adequado cozimento dos alimentos, visto que a maioria dos animais positivos não apresentou associação significativa com presença de roedores ou consumo de água não tratada, porém os mesmos permaneciam em casa aos quais fora oferecida comida caseira. Portanto, a toxoplasmose consiste em um problema de saúde pública na região estudada, sendo necessárias medidas sanitárias para o controle da infecção, visto a estreita relação homem-cão e os fatores de risco presentes.

E-mail: olney@uenf.br Pythiosis is caused by Pythium insidiosum and the occurrence of disease in horses was described in the North and Northwest State of Rio de Janeiro, Brazil. The disease was described in cattle, sheep, humans, and horses in different states and regions across the country. This paper describes the development of IgY and IgG polyclonal antibodies, in chicken and rabbits, respectively against proteins extracted from kunkers and hyphae of P. insidiosum from affected horses. The proteins were recognized by chicken, rabbit and horse antibodies by immunodiffusion and Western blot against majority bands of 27 and 43 KDa, and titrated by ELISA. The antibodies IgY developed by the first time against Brazilian strains of P. insidiosum may represent a valuable tool in the detection of antigens of the pathogen and contribute to further studies aimed at immunotherapy and knowledge about this disease in endemic areas in Rio de Janeiro and in Brazil.

A doença foi descrita em bovinos, ovelhas, humanos e cavalos em diferentes estados e regiões do país. Este trabalho descreve o desenvolvimento de anticorpos policlonais, IgY e IgG, em galinha e coelho, respectivamente, contra proteínas extraídas de kunkers e hifas de P. insidiosum de cavalos doentes. As proteínas foram reconhecidas por anticorpos de galinha, coelho e cavalos contra as bandas majoritárias de 27 e 34 KDa em imunodifusão e Western blot tituladas por ELISA. Os anticorpos IgY desenvolvidos pela primeira vez contra cepas brasileiras de P. insidiosum podem representar um valioso instrumento na detecção de antígenos de patógenos e contribuem para novos estudos baseados na imunoterapia e no entendimento sobre esta doença em áreas endêmicas no Rio de Janeiro e em todo o país.

The selection of the animals for the experimental groups was done based on a serological screening with 6 antigens of Leptospira sp. constituted by non-reagent animals. The vaccine titers were monitored using the microscopic agglutination test (MAT) for Canicola, Grippotyphosa, Hardjo, Icterohaemorrhagiae, Pomona and Wolffi serovars. All vaccines used were capable to product agglutinins for the Hardjo and Wolffi serovars observed at 3 days after vaccination, remaining until the 150th day; those serovars induced the highest titres of agglutinins. Vaccine D, in spite of not containing the Wolffi serovar, induced the production of agglutinins to this serovar. Agglutinins to the Canicola serovar were only observed in the animals vaccinated with the D bacterine. Vaccine D induced the highest average titers of antibodies to all tested serovars.

Serum samples of 3,102 cows (age ³ 24 months) from 373 herds were analyzed by complement fixation test. Totally, 44 (1.42%) animal were positive with titers ³ 32. The seroprevalence of Chlamydophila spp. in the herds was 8.82% (6.15%-12.17%). Four variables were associated with seroprevalence for Chlamydophila spp. in the final model of logistic regression: confined or semi-confined breeding (OR=3.339, P=0.004), farms with less than 35 cows (OR=3.339, P=0.017), abortion in the pasture (OR=2.372, P=0.037) and pasture rent (OR=3.398, P=0.006) were risk factors for Chlamydophila spp. This bacterium infected a small number of cattle from herds with abortion in Paraná state. Chlamydophila spp impact as abortion cause is reduced in this state.

A total of 300 equine sera from four municipalities: Ananideua (58), Belém (61), Castanhal (131), and Santa Izabel do Pará (50), were evaluated for Borrelia burgdorferi by an Elisa test. There were no significant differences (P<0.05) among municipalities, breed, sex or husbandry. A total of 80 (26.7%) horses were B. burgdorferi positive with titles of 1:800, 72 (90%) horses, 1:1.600, 6 (7.5%) horses, and 1:3.200, 2 (2.5%) horses. The results were similar to those in the USA, where related frequencies ranged from 7 to 75% in asymptomatic seropositive horses. The presence of anti-B.burgdorferi homologous antibodies in horses from four municipalities in the metropolitan mesorregion of Belém suggests the possibility of occurrence of human cases in the region.

Epidemiological and reproductive data were obtained through a questionnaire which was administered in all goat pens. Significance level was set at 5% for all statistical tests. N. caninum positivity rate was 19.77%. Only one of the properties had no seropositive animal revealing N. caninum diffusion in the state. Seropositivity frequency did not correlate with sex, age, or reproductive problems. However, the presence of dogs was associated with a higher frequency of N. caninum seropositivity. A choroplethic map with hatch patterns showing the geographical distribution of goats seropositive for Neospora caninum may considerably contribute to geographic epidemiological studies and the development of disease control strategies.
