The green turtle Chelonia mydas feeds and nests in the Brazilian coastal area and is considered an endangered species by the World Conservation Union (IUCN 2009) and threatened by the Red List of Brazilian Fauna (Ministério do Meio Ambiente 2009). Fibropapillomatosis is a disease characterized by benign skin tumors (fibropapillomas), and it is one of the main threats to the survival of this species. Studies suggest the involvement of viruses as infectious agents associated with environmental and genetic factors. Blood samples were collected from 45 turtles captured in the coastal area of the state of Sao Paulo, Brazil. From these, 27 were affected by fibropapillomas and 18 were tumor free. Biometrical data on the turtles, size, location and quantity of tumors were recorded. The area occupied by fibropapillomas per animal was calculated and four groups were determined according to severity of the disease or its absence. The objective of the study was to compare hemogram results of the sea turtles classified in these four groups. The lowest hematocrit value was observed in severely affected animals. In the hemoglobin assay, the highest value was observed in the group of tumor free turtles and the lowest, in animals severely affected. Lymphocyte counts and curved carapace length were on the verge of statistical significance.

Chelonia mydas, denominada tartaruga verde, é uma tartaruga marinha que frequenta o litoral brasileiro para alimentação e nidificação e é considerada em perigo de extinção pela IUCN (World Conservation Union, 2009) e ameaçada pela Lista Vermelha da Fauna Brasileira (Ministério do Meio Ambiente, 2009). A fibropapilomatose, doença caracterizada por tumores cutâneos benignos (fibropapilomas), é uma das mais importantes ameaças à sobrevivência dessa espécie. Pesquisas sugerem o envolvimento de agentes infecciosos virais em associação com fatores ambientais e genéticos. Foram colhidas amostras sanguíneas de 45 tartarugas provenientes do litoral do estado de São Paulo, Brasil, sendo 18 sem fibropapilomas e 27 acometidas. Dados de biometria das tartarugas, quantidade, localização e tamanho dos tumores foram anotados. Foi realizado o cálculo da área de fibropapilomas por animal e foram estipulados 4 grupos de acordo com o grau de acometimento e sua ausência. O objetivo foi realizar uma comparação entre os hemogramas das tartarugas marinhas classificadas nos 4 grupos. Animais de grau grave apresentaram o menor valor para hematócrito. Para dosagem de hemoglobina, observou-se que o maior valor foi para o grupo de tartarugas sem fibropapilomas e o menor para o de grau grave. Os valores de linfócitos e comprimento curvilíneo da carapaça beiraram a significância estatística.

Mazama nana is the least known of Brazilian deer species. The male reproductive parameters are still unknown, but apparently they did not show reproductive seasonality. In this work were used nine males of M. nana in reproductive age, kept in captivity. They were assessed for weight, height of wither, crown-rump length, the situation of horns, testicular volume and consistency. The sperm was collected by electro-ejaculation and subjected to analyses of motility, vigor and morphology. The correlation between age, weight, height of wither, crown-rump length, testicular volume and characteristics of the ejaculate (volume, motility, vigor and sperm concentration) were evaluated by the procedure Corr., of SAS®. The mean ± standard deviation observed for weight (kg), crown-rump length (cm) and height of wither (cm) were: 15.72±1.98, 74.9±3.05, and 48.5±2.06 respectively. For primary reproductive parameters were observed: volume of ejaculate (91.46±68.24ìl); motility (70±8.16%); vigor (3.0±0.67); sperm concentration (1536x106 ±351x106 spermatozoa per ml). For sperm morphology was found a high percentage of abnormal cells (40.90%), with predominant defects in the tail (25.95%).

Dos cervídeos brasileiros, a espécie Mazama nana é a menos conhecida. Os parâmetros reprodutivos para os machos ainda são desconhecidos, mas parece que não apresentam sazonalidade reprodutiva. Neste trabalho foram utilizados nove machos de Mazama nana em idade reprodutiva, mantidos em cativeiro. Foram avaliados quanto ao peso corporal, altura de cernelha, comprimento crânio-caudal, situação dos chifres, volume e consistência testicular. O sêmen foi colhido por eletroejaculação e submetido a análises de motilidade, vigor e morfologia espermática. As correlações entre idade, peso, comprimento crânio-caudal, altura de cernelha, volume testicular e características do ejaculado (volume, motilidade, vigor e concentração do sêmen) foram avaliadas pelo procedimento Corr., do SAS®. As médias ± desvio padrão observados para peso (kg), comprimento crânio-caudal (cm) e altura de cernelha (cm) foram: 15,72±1,98, 74,9±3,05 e 48,5±2,06, respectivamente. Em relação aos parâmetros reprodutivos primários foram observados: volume do ejaculado (91,46±68,24µl); motilidade (70±8,16%); vigor (3,0±0,67); concentração espermática (1536x106 ±351x106 espermatozóides por ml). Em relação à morfologia espermática, foi observada uma alta porcentagem de células anormais (40,90%), sendo predominante os defeitos de cauda (25,95%).

Brucella spp. are intracellular facultative gram-negative bacteria which are pathogenic for many species of mammals, causing brucellosis, a worldwide spread zoonosis. Therefore the search for more efficient alternatives of control, as the development of new potential immunogens is necessary. In this study, we knockouted virB10 from Brucella abortus S2308 strain, generating a mutant strain probably incapable to produce the corresponding native protein. The gene virB10 is part of an operon that codifies for type IV secretion system, which is essential for the intracellular survival and multiplication of the bacteria in host cells. The knockout was carried through by the construction of the suicidal plasmid pBlue: virB10: kan and eletroporation in eletrocompetent cells of B. abortus S2308, leading to the exchange of the wild gene for the interrupted gene, containing the gene of resistance to kanamycin, for double homologous recombination. BALB/c mice were inoculated with S19, RB-51, DvirB10 strains of B. abortus and S2308 wild strain; the results demonstrated that the BALB/c mice inoculated with S19 and BALB/c mice inoculated with S2308 presented faster fall of trend line, when compared with the too much groups, for bacterial recovery (BR) and esplenic weight (EW) respectively. The groups that received DvirB10 S2308 B. abortus and RB-51 demonstrated similar behavior for both the characteristics. In the sixth week postinoculation, the results for BR (log UFC ± standart deviations) and EW (esplenic weight ± standart deviations), respectively, showed: groups inoculated with strains S2308 (4,44±1,97 and 0,44±0,11), S19 (1,83±2,54 and 0,31±0,04), RB-51 (0,00±0,00 and 0,20±0,01) and DvirB10 S2308 (1,43±1,25 and 0,19±0,03). Considered the bacterial clearance, all the groups differed statistical from the group that received S2308 (p<0,0001), the group inoculated with DvirB10 S2308 B. abortus was similar to the S19 group (p=0,4302) and different of group RB-51 (p=0,0063). The evaluation of the persistence of the strains showed that virB10 is essential for the maintenance of the virulence. These results support other studies concerning the immunogenic potential of this mutant strain.

Brucella spp. são bactérias gram-negativas, intracelulares facultativas que são patogênicas para muitas espécies de mamíferos causando a brucelose, uma zoonose difundida mundialmente. Por isso a busca de alternativas de controle mais eficientes se faz necessário como o desenvolvimento de novas cepas que possam ser testadas como potenciais imunógenos. Neste estudo realizou-se a deleção do gene virB10 da cepa S2308 de Brucella abortus gerando uma cepa knockout provavelmente incapaz de produzir a proteína nativa correspondente. O gene virB10 faz parte de um operon que codifica para um sistema de secreção do tipo IV, essencial para a sobrevivência intracelular e multiplicação da bactéria em células hospedeiras. A deleção foi realizada pela construção do plasmídeo suicida pBlue:virB10:kan e eletroporação deste em células eletrocompetentes de B. abortus S2308, ocorrendo a troca do gene selvagem pelo gene interrompido, com o gene de resistência a canamicina, por recombinação homóloga dupla. Camundongos BALB/c foram inoculados com as cepas S19, RB-51, DvirB10 de B. abortus e B. abortus S2308 selvagem; os resultados demonstraram que camundongos BALB/c inoculados com S19 e camundongos BALB/c inoculados com S2308 apresentaram queda mais rápida de linha de tendência, quando comparadas aos demais grupos, para recuperação bacteriana (RB) e peso esplênico (PE) respectivamente. Os grupos que receberam DvirB10 S2308 de B. abortus e RB-51 demonstraram comportamento semelhante para ambas as características. Na sexta semana após a inoculação, os resultados para RB (log de UFC ± desvio padrão) e PE (peso esplênico ± desvio padrão), respectivamente, mostraram: grupos inoculados com as cepas S2308 (4,44±1,97 e 0,44±0,11), S19 (1,83±2,54 e 0,31±0,04), RB-51 (0,00±0,00 e 0,20±0,01) e DvirB10 S2308 (1,43±1,25 e 0,19±0,03). Considerado o clearance bacteriano, todos os grupos diferiram estatisticamente do grupo que recebeu S2308 (p<0,0001), o grupo inoculado com DvirB10 S2308 de B. abortus foi semelhante ao grupo S19 (p=0,4302) e diferente do grupo RB-51 (p=0,0063). A avaliação da persistência revelou que o gene virB10 é essencial para a manutenção da virulência da bactéria. Os resultados obtidos possibilitarão que outras pesquisas sejam realizadas avaliando o potencial imunogênico desta cepa mutante.

Alteration in the relative percentage of survival (RPS) rate of rainbow trout (Oncorhynchus mykiss) exposed to 5, 10 and 25µg ml-1 levamisole for 2 h against Yersinia ruckeri was investigated. The average weight of the 120 fish used in this study was 6.3g. Upon challenge with a virulent strain, the relative survival percentage of respectively 83.3%, 86.7% and 76.6% was recorded. The results suggest that the application of levamisole in fish farms could increase resistance to infection of fish and offer economic benefits.

This experiment was design to address the following points in relation to the acute poisoning by bracken fern (Pteridium aquilinum) in cattle: 1) the severity of the thrombocytopenia in different stages of the intoxication and its relationship to possible deficits in the secondary hemostasis, 2) the relationship between neutropenia and the morphological signs of septicemia occasionally found at necropsy, and 3) the mechanism of anemia and its relationship with blood loss, medium life of erythrocytes and the progress of the disease. The fresh green upper parts of P. aquilinum were fed to four mixed breed calves with average age of 18 months and weights ranging from 190 to 215 kg. A calf of similar age and weight was kept together with the other four under the same conditions, except for the ingestion of P. aquilinum. The four fern-fed calves died with typical features of acute bracken fern poisoning after being fed with the plant for 53-58 days daily doses of 8.0, 8.6, 10.2, and 10.6g/kg body weight totaling at the end of the experiment, respectively, 59.3%, 63.3%, 47.4%, and 47.5% of bracken fern in relation to their body weight. The disease was characterized by fever up to 42.5°C and varying degrees of hemorrhages observed clinically, at necropsy and on histological examination. Death occurred 6-7 days after the onset of fever. The hematological changes consisted mainly of marked thrombocytopenia and neutropenia. Two of the four affected calves had mild anemia. The deficits in secondary hemostasis were mild in each case. There were no significant changes thus permitting to rule out the role of secondary hemostasis in the pathogenesis of the hemorrhages in the bracken fern poisoning. The measurement of fibrin degradation products in the serum showed conflicting results and did no allow for a solid conclusion regarding the role of disseminated intravascular coagulation in the pathogenesis of the hemorrhages in fern poisoning. The cytology and the histopathology of bone marrow from the four poisoned calves revealed marked decrease in the numbers of hematopoietic cells from the three marrow lineages, which characterizes marrow failure due to aplasia; it was concluded that the events of the primary hemostasis due to thrombocytopenia are responsible for the hemorrhages. In blood culture from three affected calves there was growth of Klebsiella oxytoca, Staphylococcus hyicus and Staphylococcus aureus, indicating that septicemia, facilitated by neutropenia could have a role in the death of cattle acutely poisoned due to the ingestion of P. aquilinum. Additional points of the interest in the acute experimental poisoning by bracken fern in cattle reported here were the development of hematuria and the so called laryngeal form.

Este experimento foi delineado para investigar os seguintes pontos em relação à intoxicação aguda por samambaia (Pteridium aquilinum) em bovinos: 1) a intensidade da trombocitopenia em diferentes momentos da intoxicação e sua relação com possíveis déficits na hemostasia secundária, 2) a relação da neutropenia com as manifestações morfológicas de septicemia ocasionalmente observadas na necropsia, e 3) o mecanismo da anemia e sua relação com a perda de sangue, a vida média eritróide e a evolução da doença. As hastes superiores mais verdes de P. aquilinum foram administradas a quatro bovinos sem raça definida, com idade média de 1,5 ano e pesos entre 190-215 kg. Um bovino de idade e peso semelhantes foi usado como controle e, exceto por não ter recebido P. aquilinum, foi mantido nas mesmas condições que os outros quatro. Os quatro bovinos que receberam a planta morreram com quadro característico da intoxicação aguda por samambaia após receberem durante 53-58 dias, doses diárias de 8,0, 8,6, 10,2 e 10,6g/kg de peso corporal, que totalizaram, ao final do experimento, respectivamente, 112,7, 107,6, 85,7, 90,15 kg da planta, o que corresponde, respectivamente, a 59,3%, 63,3%, 47,4%, 47,5% da planta em relação ao peso dos bovinos. A doença caracterizou-se por febre de até 42,5°C e diversos graus de hemorragias observadas clinicamente, na necropsia e na histopatologia. A morte ocorria 6-7 dias após o início do quadro febril. As alterações hematológicas revelaram trombocitopenia e neutropenia acentuadas. Em dois dos quatro bovinos havia anemia leve. Não houve variações significativas nos tempos de coagulação dos bovinos intoxicados, quando avaliados os fatores de coagulação (secundária), excluindo-se assim a possibilidade da participação de distúrbios da hemostasia secundária na patogênese das hemorragias nessa intoxicação. A determinação dos produtos da degradação da fibrina no soro revelou dados conflitantes, não permitindo concluir se a coagulação intravascular disseminada tem participação na patogênese das hemorragias nessa intoxicação. A citopatologia e histopatologia da medula óssea dos quatro bovinos intoxicados revelaram acentuada diminuição no número de células hematopoéticas das três linhagens medulares, caracterizando insuficiência medular por aplasia; conclui-se que apenas eventos da hemostasia primária devidos a trombocitopenia são responsáveis pelas hemorragias. Na hemocultura de três dos bovinos intoxicados houve crescimento de Klebsiella oxytoca, Staphylococcus hyicus e Staphylococcus aureus, indicando que a septicemia, facilitada pela neutropenia, pode ter participação na causa da morte de bovinos na intoxicação aguda pela ingestão de P. aquilinum. Aspectos adicionais de interesse na reprodução da intoxicação aguda por samambaia em bovinos deste relato incluem o desenvolvimento de hematúria na doença aguda e a apresentação da chamada forma laríngea da doença.

Among the systemic signs of toad venom (bufotoxin) poisoning in dogs, the cardiotoxic effects are one of the most important. Thus, the objective of this experiment was to evaluate potential changes in the cardiac muscle in dogs poisoned experimentally by toad venom and to observe the eletrolyte alterations which may occur in this condition. Twenty dogs divided into control group (n=5) and poisoned group (n=15) were utilized. The toad venom was extracted by manual compression of the paratoidic glands. After general anesthesia, dogs in the control group received placebo and dogs in the poisoned group received the venom by orogastric catheter. Samples for dosage were collected 6 hours and 24 hours after poisoning and 0, 2, 4, 6 and 12 hours after poisoning for electrolytes dosage. The Man-Withney test was used for statistical analysis (P<0.05). The poisoned dogs showed (saline) elevated levels of cardiac markers CK-MB and TnIc, confirming the cardiotoxic effect of the bufotoxin. Hypokalemia and hypocalcemia were also observed.

Dentre os sinais sistêmicos causados pelo envenenamento por veneno de sapo (bufotoxina) em cães, os efeitos cardiotóxicos são um dos mais importantes. O objetivo deste estudo foi avaliar as potenciais alterações no músculo cardíaco de cães envenenados experimentalmente por veneno de sapo e observar as alterações eletrolíticas que podem ocorrer nesse tipo de envenenamento. Utilizaram-se 20 cães divididos em grupo controle (n=5) e grupo envenenado (n=15). O veneno de sapo foi extraído por meio de compressão manual das glândulas paratóides. Após anestesia geral, os cães do grupo controle receberam placebo (solução fisiológica) e os do grupo envenenado uma alíquota do veneno por sonda orogástrica. As colheitas de sangue para dosagem dos marcadores cardíacos foram realizadas seis e 24 horas após o envenenamento. As colheitas de sangue para dosagem dos eletrólitos foram realizadas antes e duas, quatro, seis e 12 horas após o envenenamento. A análise estatística empregada foi o teste não-paramétrico de Mann-Withney (P<0,05). Os cães envenenados por veneno de sapo apresentaram elevação dos níveis dos marcadores cardíacos CK-MB e TnIc, confirmando a cardiotoxicidade do veneno. Hipocalemia e hipocalcemia foram também observadas nos cães envenenados.

In the last few years, the use of laparoscopy in veterinary medicine has expanded and consequently so was the need for studies that establish the advantages, disadvantages and possible complications of each procedure. The purpose of the current study was to describe a laparoscopic splenectomy technique and the alterations due to this access, and compare it to the open procedure in dogs. A total of 15 healthy female mongrel dogs were used, with mean weight of 17.4±2.5kg. The animals were distributed into three groups: Group IA of open splenectomy (laparotomy) using double ligation of the vessels of the splenic hilum with poliglicolic acid, Group IB of open splenectomy (laparotomy) with bipolar electrocoagulation of the splenic hilum, and Group II of laparoscopic access with bipolar electrocoagulation of the splenic hilum. Operative time, blood loss, size of incisions, complications during and after surgery were evaluated. Other parameters included pain scores, white blood cell (WBC) counts and postoperative serum concentrations of alanine aminotransferase (ALT), alkaline phosphatase (ALP), creatine kinase (CK), C-reactive protein (CRP), glucose and cortisol. No differences were found in the evaluation of parameters between both open splenectomy techniques employed. Laparoscopic access presented significant differences (p<0,05) when compared with open surgery: Longer operative time, smaller abdominal access, decrease in blood loss, lower concentrations of CRP, higher levels of CK and ALP, and lower scores in the pain scale. Laparoscopic surgery showed fewer complications of the surgical wound. No significant differences were observed between groups in the postoperative temperature, WBC, ALT, cortisol and glucose concentrations. In conclusion, the laparoscopic technique is useful for splenectomy in dogs, being advantageous in terms of blood loss, surgical stress and surgical wounds. However, it expends more operative time and causes transitory increase in hepatic and muscular enzymes.

Nos últimos anos, a utilização da laparoscopia em Medicina Veterinária vem expandindo e, conseqüentemente, a necessidade de pesquisas que determinem as vantagens, desvantagens e possíveis complicações de cada procedimento. Este estudo teve como objetivo descrever uma técnica de esplenectomia laparoscópica, assim como as alterações decorrentes deste acesso, e compará-la ao procedimento convencional em cães. Foram utilizadas 15 cadelas hígidas, sem raça definida, com peso médio de 17,4 ±2,5kg. Os animais foram distribuídos em três grupos: Grupo IA de acesso convencional (por laparotomia) utilizando ligadura com ácido poliglicólico no selamento vascular do hilo esplênico, Grupo IB de acesso convencional (por laparotomia) com eletrocoagulador bipolar do hilo esplênico, e Grupo II de acesso laparoscópico com eletrocoagulador bipolar para selamento vascular dos ramos esplênicos. Estes grupos foram avaliados em relação ao tempo cirúrgico, à perda de sangue, ao tamanho das incisões e às complicações durante e após a cirurgia. Também foram comparadas as avaliações da escala de dor e as alterações no leucograma e nas concentrações séricas da alanina aminotransferase (ALT), da fosfatase alcalina (FA), da creatina quinase (CK), da proteína C-reativa (CRP), da glicose e do cortisol no pós-operatório. Os acessos convencionais não diferiram entre si nos parâmetros avaliados. O acesso laparoscópico apresentou diferenças significativas (p<0,05) quando comparado ao convencional: maior tempo cirúrgico, menor acesso abdominal, diminuição na perda de sangue, menores concentrações de CRP, maiores níveis de CK e FA, além de pontuação menor na escala de dor. A cirurgia laparoscópica apresentou menor número de complicações das feridas cirúrgicas. A ALT, o cortisol, a glicemia, o leucograma e a temperatura retal pós-operatórias não diferiram significativamente entre os acessos convencional e laparoscópico. Conclui-se que a cirurgia laparoscópica é viável para esplenectomia em cães, apresentando vantagem em relação à perda de sangue, ao estresse cirúrgico e às feridas cirúrgicas, embora apresente maior tempo cirúrgico e cause aumento transitório de enzimas hepática e muscular.

Clinical cases and diagnostic specimens from ruminants received by the Veterinary Hospital of the University of Campina Grande in Patos, Paraíba, Brazil, from January 2000 to August 2007, were reviewed to evaluate some epidemiological aspects of parasitic diseases of ruminants in the semiarid region of Paraíba and neighboring states. The region has an annual rainfall of about 800mm, with irregular rains concentrated in a 3 to 4-month-period, and an average annual temperature of 26°C. During those years, 163 (5.31%) out of 3,064 ruminants were affected by some parasitic disease. The most important parasitosis in goats and sheep was gastrointestinal helminthiasis, mainly hemonchosis; goats were more affected (6.24% of cases in this species) than sheep (4.7% of cases). The higher frequency of the disease in goats may be associated with higher susceptibility or due to treatment mistakes, i.e. use of the same dose for both species, which for most anti-antihelmintics is insufficient for goats. Cattle have a low rate of gastrointestinal helminthiasis (1 out of 1,113 cases). This low frequency is probably due to the farming system in the semiarid, with low stocking rate, up to one adult bovine for every 13-16 ha, and permanence of calves (susceptible) with their mothers (resistant) for nearly one year before weaning. The occurrence of eimeriosis in goats and sheep was observed in 0.76% of the cases, involving only young animals. The main cattle disease was tick fever with 14 outbreaks. Outbreaks of tick fever occur, mainly at the end of the raining season in areas of the semiarid which are marginal for tick, like plateaus and mountains of the Borborema region, irrigated areas, and areas of the basins of two rivers of the region. In the drier areas of the semiarid Rhipicephalus (Boophilus) microplus do not survive during the dry period, but tick fever can occur when cattle with ticks are introduced at the onset of the raining season.

Mycoplasmas are important avian pathogens, which cause respiratory and joint diseases that result in large economic losses in Brazilian and world-wide poultry industry. This investigation regarding the main species of mycoplasmas, Mycoplasma gallisepticum (MG) and M. synoviae (MS), responsible for the above mentioned conditions, was carried out through PCR Multiplex analysis. One thousand and forty-six (1,046) samples of tracheal swabs and piped embryos were collected from 33 farms with laying hens, breeders, broilers or hatchery, located in the Brazilian states of São Paulo, Paraná and Pernambuco, where respiratory problems or drops in egg production had occurred. The MG and MS prevalence on the farms was 72.7%. These results indicated (1) high dissemination of mycoplasmas in the evaluated farms, with predominance of MS, either as single infectious agent or associated with other mycoplasmas in 20 farms (60.6%), and (2) an increase of MS and decrease of MG infection in Brazilian commercial poultry.

Initially, fronto-occipetal (FOD) and bizigomatic (BZD) diameters were measured using a caliper. The ophthalmologic transpalpebral B-mode ultrasonography (US) was performed to measure the internal ocular structures, as follows: The cornea thickness (D1), distance between cornea and anterior lens capsule (D2), distance between cornea and posterior lens capsule (D3), lens thickness (D4), lens diameter (D5), lens area (D6), distance between posterior lens capsule and retina (D7), distance between anterior lens capsule and retina (D8), and the distance between cornea and retina (D9). Except for D4, there were effect of FOD and BZD on the measures of the internal structures of GO. The Lineal Regression Analysis between the measures of the internal oculars structures and DFO and DBZ were significant for D1, D2, D3, D4, D5, D6, D7, D8 and D9.
