#11 - Detection of Brucella abortus in bovine tissue using PCR and qPCR, 34(6):497-502

E-mail: gracia.rosinha@embrapa.br The aim of the study was to evaluate the technical polymerase chain reaction (PCR) and Real-Time PCR (qPCR) to detect Brucella abortus from bovine tissues with suggestive lesions of brucellosis. For this, 21 fragments of bovine tissues collected at abattoirs of Mato Grosso do Sul were processed and subjected to microbiological culture and extraction of genomic DNA to perform the PCR reactions and qPCR. Eight samples of microbiological culture showed bacterial growth and five samples were confirmed as B. abortus by PCR. DNA of Brucella (IS711 primers) was detected in 13 (61.9%) directly from tissue samples and 17 (81%) from tissue homogenate samples. With the species-specific set of primers BruAb2_0168F and BruAb2_0168R, 14 (66%) tissue samples and 18 (85.7%) tissue homogenate samples were positive. Six positive samples in the species-specific PCR were sequenced and the best hit in the BLASTn analysis was B. abortus. By qPCR, 21 (100%) tissue samples and 19 (90.5%) tissue homogenate samples were positive for B. abortus. Ten samples of DNA from bovine blood from an accredited-free herd were used as negative control in PCR and qPCR analysis using the primers BruAb2_0168F and BruAb2_0168R, and no one amplified by PCR, whereas two samples were amplified by qPCR (20%). In conclusion, both techniques detect the presence of B. abortus directly from tissues and homogenized, but the qPCR showed high sensitivity. The results indicate that qPCR can represent an alternative tool for faster and more accurate detection of B. abortus directly from tissues, and use in health surveillance programs by presenting satisfactory sensitivity and specificity.

E-mail: gracia.rosinha@embrapa.br Objetivou-se no presente estudo avaliar as técnicas reação em cadeia da polimerase (PCR) e PCR em Tempo Real (qPCR) para detectar Brucella abortus, a partir de tecidos bovinos com lesões sugestivas de brucelose. Para isto, 21 fragmentos de tecidos bovinos coletados em abatedouros de Mato Grosso do Sul foram processados e submetidos ao cultivo microbiológico e extração do DNA genômico para realização das reações de PCR e qPCR. No cultivo microbiológico, oito amostras apresentaram crescimento bacteriano e cinco foram confirmadas como B. abortus por PCR. Diretamente das amostras de tecido, DNA do gênero Brucella (oligonucleotídeos IS711) foi detectado em 13 (61,9%) amostras de tecido e 17 (81%) amostras de homogeneizado. Já com os oligonucleotídeos espécie-específicos BruAb2_0168F e BruAb2_0168R, 14 (66%) amostras de tecido e 18 (85,7%) amostras de homogeneizado foram amplificadas. Seis amostras positivas na PCR espécie-específica foram sequenciadas e o best hit na análise BLASTn foi B. abortus. Na qPCR, 21 (100%) amostras de tecidos e 19 (90,5%) amostras de homogeneizado foram positivas para B. abortus. Dez amostras de DNA de sangue bovino de rebanho certificado livre foram utilizadas como controle negativo nas análises de PCR e qPCR utilizando-se os oligonucleotídeos BruAb2_0168F e BruAb2_0168R. Na PCR nenhuma amostra amplificou, enquanto que na qPCR 2 (20%) amplificaram. Conclui-se que as duas técnicas detectam a presença de B. abortus diretamente de tecidos e homogeneizados, porém a qPCR apresentou maior sensibilidade. Os resultados obtidos indicam que a qPCR pode representar uma alternativa rápida e precisa para a detecção de B. abortus diretamente de tecidos, e ser utilizada em programas de vigilância sanitária, por apresentar sensibilidade e especificidade satisfatórias.

E-mail: alex@ufersa.edu.br The study aimed to determine the prevalence of brucellosis and its risk factors in the pig herd of Mossoró-RN. Comprised a cross-sectional quantitative approach and epidemiological study, where 412 blood samples of pigs from major producers in the municipality were collected and inspection of herds with interviews with producers were conducted. Antibodies to Brucella spp. were detected by the rose Bengal test (RBT) and confirmed by the complement fixation test (CFT). The prevalence of brucellosis in pigs was 27.0% in the RBT and 17.5% in the CFT. In 55% of the surveyed properties there was at least one positive animal, and these prevalence ranged from 6.7% to 80.0%. Risk factors that were influencing the occurrence of the disease were the presence of rats in the farms, contact with cattle and the young age of the animals. The study results showed that the causative agent of brucellosis was circulating in pigs in the municipality of Mossoró-RN, with high prevalence in the herd and on the properties, indicating the risk of transmission of this zoonosis to humans.

Compreendeu um estudo epidemiológico transversal e de abordagem quantitativa, no qual foram coletadas 412 amostras sanguíneas de suínos provenientes dos principais produtores do município e realizada inspeção das criações com entrevistas aos produtores. Anticorpos anti-Brucella spp. foram detectados pelo teste do antígeno acidificado tamponado (AAT) e confirmados pela reação de fixação de complemento (RFC). A prevalência da brucelose nos suínos foi de 27,0% no teste de AAT e 17,5% na RFC. Em 55% das propriedades pesquisadas havia pelo menos um animal positivo, e a prevalência nestas variou de 6,7% a 80,0%. Os fatores de risco que estavam influenciando a ocorrência da doença foram: a presença de ratos nas criações, o contato com bovinos e a faixa etária jovem dos animais. Os resultados do estudo permitiram concluir que o agente etiológico da brucelose estava circulando em suínos do município de Mossoró-RN, com elevada prevalência no rebanho e nas propriedades, evidenciando o risco de transmissão desta zoonose para o homem.

Reliable epidemiologic brucellosis data covering approximately 90% of the cattle population in Brazil have been recently published. Therefore, considering the scarcity of information regarding the economic impact of bovine brucellosis in Brazil, the goal of this study was to estimate economic impact of brucellosis on the Brazilian cattle industry. Several parameters including abortion and perinatal mortality rates, temporary infertility, replacement costs, mortality, veterinary costs, milk and meat losses were considered in the model. Bovine brucellosis in Brazil results in an estimated loss of R$ 420,12 or R$ 226,47 for each individual dairy or beef infected female above 24 months of age, respectively. The total estimated losses in Brazil attributed to bovine brucellosis were estimated to be approximately R$ 892 million (equivalent to about 448 million American dollars). Every 1% increase or decrease in prevalence is expected to increase or decrease the economic burden of brucellosis in approximately 155 million Reais.

E-mail: clebertja@uol.com.br The aim of this investigation was to determine the seroprevalence of Brucella ovis in sheep flocks and individual sheep in the Sertão mesorregion, Paraíba state, Northeastern Brazil, as well as to identify risk factors. Blood samples were collected from 1,134 sheep from 103 flocks in 17 counties. For the serological diagnosis of B. ovis infection the agar gel immunodiffusion test (AGID) was carried out. A flock was considered positive when there was at least one seropositive animal. Of the 103 flocks used, 21 (20.39%) presented at least one seropositive sheep, and of the 1,134 sheep examined 59 (5.20%) seropositive animals were diagnosed. Cleaning of facilities (odds ratio = 7.13; 95% CI=1.56-32.47; p=0.011) and purchase of animals (odds ratio = 6.06; 95% CI=1.39-26.48; p=0.017) were identified as risk factors. Based on the risk factor analysis, it is recommended the diagnosis of B. ovis infection prior to purchase of sheep and the periodic cleaning of the facilities on the farm.

Para o diagnóstico sorológico da infecção por B. ovis foi utilizado o teste de imunodifusão em gel de ágar (IDGA). Um rebanho foi considerado positivo quando apresentou pelo menos um animal soropositivo. Das 103 propriedades utilizadas 21 (20,39%) apresentaram pelo menos um animal soropositivo e dos 1.134 animais, 59 (5,20%) foram soropositivos. Realizar higiene nas instalações com periodicidade anual (odds ratio = 7,13; IC 95% = 1,56-32,47; p=0,011) e aquisição de animais (odds ratio = 6,06; IC 95% = 1,39-26,48; p=0,017) foram identificados como fatores de risco. Com base na análise de fatores de risco, recomenda-se a realização de diagnóstico da infecção por B. ovis previamente à aquisição de animais e realização periódica de higienização das instalações.

E-mail: rosinha@cnpgc.embrapa.br Vaccine strains B19 and RB51 are administered to cattle for prevention against infection by Brucella abortus. However, there are reports that these vaccines can cause miscarriages. Thus, every miscarriage among vaccinated animals should be thoroughly studied to determine the cause. In Brazil, there are no records on the origin of B19 and RB51 samples used in the preparation of commercial vaccines. Therefore, a study is needed to determine possible mutations in relation to the USDA reference samples of B. abortus due to the fact that vaccine samples could revert to the virulence of the disease. The aim of the present study was to perform a genotype analysis of vaccine strains B19 and RB51 used in Brazil. The methodology was based on the genotyping of marker genes of these vaccine strains by amplification using polymerase chain reaction. The results allowed the identification of the genotype of the B19 and RB51 commercial vaccine available for use on cattle in Brazil. The absence of mutations in the samples tested confirmed the genetic quality of the vaccines and stability of genes analyzed.

A metodologia utilizada foi a genotipagem de genes marcadores destas amostras vacinais, por meio da amplificação pela reação em cadeia da polimerase. Os resultados obtidos permitiram a identificação do genótipo das vacinas comerciais B19 e RB51 disponíveis e utilizadas em bovinos no Brasil. A ausência de mutações nas vacinas testadas corrobora com a qualidade genética das mesmas, quanto à estabilidade dos genes analisados.

E-mail: jane@fmvz.unesp.br The aim of this investigation was to evaluate the seropositivity for brucellosis in pig slaughterhouses. Sera of 910 pigs from 30 farms and slaughtered at slaughterhouses, located in the central region of the state of São Paulo, were tested with Buffered Plate Acidified Antigen (BPAA) and 2-mercaptoethanol (2-Me) as confirmatory test to determine the serological positivity in this species. The results demonstrated that 25 (2.7%) of the pigs from 10 farms reacted to BPAA, corresponding to 36% of positive properties. From BPAA positive animals 16% and 52% presented titers of 25 incomplete and 25 respectively in the Tube agglutination test (TAT), 8% presented 50 and 25 incomplete, respectively, in TAT and 2-ME, and 8% resulted 50 and 25 positive in TAT and 2-ME, respectively. These results demonstrate the high percentage of positive farms for swine brucellosis in this region and reinforce the need for official and effective program implementation for the swine brucellosis control.

Foram coletados 910 soros de suínos, procedentes de 30 propriedades, abatidos em frigoríficos da região central do estado de São Paulo, e submetidos às provas de soroaglutinaçao com antígeno tamponado acidificado (AAT) e 2-Mercaptoetanol (2-Me) objetivando determinar a ocorrência da enfermidade nesta espécie. Do total de soros avaliados foram observados 25 (2,7%) animais reagentes ao AAT pertencentes a 10 propriedades, caracterizando 36% de propriedades positivas. Dos animais positivos ao AAT, 16% apresentaram titulo de 25 (incompleto) e 52% titulo de 25 na soroaglutinação lenta (SAL), 8% apresentaram titulo de 50 incompleto na SAL e 25 incompleto no 2-ME e 8% apresentaram titulo de 50 na SAL e 25 no 2-ME. Estes resultados demonstram o elevado percentual de propriedades positivas para brucelose nesta região e ressaltam a necessidade de implementação de programas oficiais efetivos para o controle da brucelose suína.

E-mail: nancylenichaves@hotmail.com This study was carried out to evaluate the intercurrence of brucellosis and enzootic leucosis in buffaloes in extensive production system by establishing the prevalence of reagent buffaloes for specific diagnostic tests. We analyzed 232 animals serologically by the technique of double immunodiffusion in agarose gel (AGID) for diagnostic of leucosis and the tampon acidified antigen (TAA) test, followed by confirmatory tests of 2-mercaptoethanol (2-ME) and slow serum agglutination (SSA) in tubes for diagnostic of brucellosis. The prevalence of buffaloes that were positive for leucosis and brucellosis tests were 4.21% (10/232) and 5.18%(12/232) respectively. The results of this investigation indicated that the diseases occur in the population studied, however, with low prevalence and without depending on the occurrence of both.

Os resultados obtidos nesta pesquisa permitiram concluir que as infecções estudadas encontram-se presentes na população avaliada, entretanto, com baixa prevalência e sem dependência de ocorrência entre ambas.

E-mail: rosinha@cnpgc.embrapa.br The presence of Brucella spp. in wild animals can influence their reproduction rate and may be a source of infection for domestic animals and humans. The objective of this study was to identify the presence of Brucella spp. in 44 blood samples from the deer Ozotoceros bezoarticus in the southern Pantanal of Sul-Mato-Grossense, using the PCR technique. It was seen that 20.4% (9/44) of the samples were positive. The consensus sequence was obtained by sequencing these samples, which then showed 514 pb and 95% of identity with gene virB5 of B. abortus (best hits accession nr AF226278, e-value 0.0). The phylogenetic analysis of the sample isolated from deer revealed the Brucella to be very close to B. suis. The high percentage of positive samples suggests that brucellosis may be a concern in deer within the studied area, and that these animals may poses a risk for other domestic and wild ones.

O objetivo deste estudo foi identificar a presença de Brucella spp. em 44 amostras de sangue de veado campeiro (Ozotoceros bezoarticus) do Pantanal do Sul-Mato-Grossense, utilizando a técnica de PCR. Observou-se que 20,4% (9/44) das amostras foram positivas. A sequência consenso de nucleotídeo obtida no sequenciamento do isolado de veado campeiro apresentou 514 pb e 95% de identidade com virB5 de B. abortus (best hits acesso nr AF226278, e-value 0.0), já na análise filogenética a amostra de Brucella isolada de veado campeiro apresentou-se muito próximo de B. suis. A alta porcentagem de amostras positivas sugere que a brucelose pode ser um problema entre os veados campeiros na área estudada e que estes animais podem representar riscos para outros animais domésticos e silvestres.

All sera were examined by AGID test (screening test) and CFT (confirmatory test). From the total of examined herds, 8.59% (95% CI = 5.83%-12.48%) were seropositive for Brucella ovis. The prevalence of seropositive rams was 5.57% (95% CI = 3.86%-7.97%). The seropositivity was lower in herds where cleanliness was frequently made (p< 0.05).

For this technique the antigen is the same as the one used in the tube agglutination test. However, literature is poor in information about the stability of the whole cell Brucella antigen for use in the complement fixation test to establish a time of validity of the antigen. Hence the aim of this investigation was to evaluate the stability of this antigen under refrigeration for use in the complement fixation test. Fourteen batches of antigen prepared with Brucella abortus strain 1119/3, produced from 9 months to 23 years and 11 months before, were analysed. One hundred and sixty-seven cattle sera with varying titres of antibodies to Brucella were tested through the warm complement fixation microtechnique with five 50% haemolytic units of complement. Sera with at least 25% of complement fixation in dilution 1:4 were considered positive. The results with 13 of the antigen batches were compared with the results obtained with the batch produced 9 months before by the McNemar c2 test and kappa statistic. The oldest antigen batch gave a higher proportion of sera titres which were exactly the same observed with the 9-month-batch (90.4%), and the antigen produced 4 years and 3 months before the test gave de lowest proportion of sera with the same titre of the 9-month-antigen (73.7%). The comparison of the results after being classified as positive and negative showed that the highest proportion of agreed results was observed with the antigen produced 21 years and 4 months before (98.8%, kappa 0.98). The antigen with the lowest proportion of agreed results was the one produced 3 years and 2 months before (91.6%, kappa 0.84). The results of the study show that most sera gave very similar results with all antigen batches evaluated, and that there was no relationship between the period of antigen production and the difference in test results.
