#1 - Pathological changes in palatine tonsils of slaughtered water buffaloes (Bubalus bubalis)

Abstract in English:

This study evaluated palatine tonsils from 327 water buffaloes collected in a slaughterhouse located in the city of Belém, Pará State, Brazil, over four months (nine sample collecting visits). The overwhelming majority (84.71%) of these buffaloes came from Marajó Island, Pará State, Brazil. The main pathological findings were crypt dilatations/keratin cysts (96.0%, 314 cases), mild to moderate follicular lymphoid hyperplasia (93.0%, 304 cases), crypt abscesses (79.8%, 261 cases), tonsillar corpuscles (31.2%, 102 cases) and tuberculoid granulomas (1.53%, 5 cases). Acid-fast bacilli (AFB) were observed in 20% (1 out of 5 cases) of these granulomas. The study indicates that pathological evaluation of tonsils in buffaloes can provide details of structural features of the organ in the species and information on subclinical or asymptomatic tonsillitis.

No presente estudo foram avaliadas tonsilas palatinas de 327 bubalinos, coletadas em nove visitas a um abatedouro frigorífico localizado na cidade de Belém, no estado do Pará, Brasil, em um período de quatro meses. Estes bubalinos, em sua ampla maioria (84,71%), eram procedentes da Ilha de Marajó, no estado do Pará, Brasil. Os principais achados patológicos foram dilatações de criptas/cistos de queratina (96,0%, 314 casos), hiperplasia linfoide folicular leve a moderada (93,0%, 304 casos), abscessos de criptas (79,8%, 261 casos), corpúsculos tonsilares (31,2%, 102 casos) e granulomas tuberculoides (1,53%, 5 casos). Bacilos álcool-ácido resistentes (BAAR) foram observados em 20% (1 de 5 casos) destes granulomas. O estudo indica que a avaliação patológica das tonsilas de bubalinos pode fornecer detalhes de características estruturais do órgão na espécie e informações de tonsilites subclínicas ou assintomáticas.


#2 - Safety and efficacy of allogeneic bone marrow mesenchymal stem cells for treatment of canine leukopenia induced by canine parvovirus infection

Abstract in English:

This study aims to establish a therapy strategy for canine leukopenia induced by canine parvovirus (CPV) infection through intravenous infusion of allogeneic bone marrow mesenchymal stem cells (BMMSCs) and to evaluate the therapeutic effect of BMMSCs on canine parvovirus. Forty healthy 2-month-old dogs were randomly divided into four groups including the BMMSC treatment group (A), conventional treatment group (B), CPV infection group (C), and a normal control group (D). Then the A, B, and C groups were orally infected with CPV (103.25 TCID50/mL) at 1mL/kg, and the D group received the same dose of saline. After the onset of infection, Group A received mesenchymal stem cells (MSCs) and rehydration as the treatment; Group B was treated with anti-inflammatory therapeutics and rehydration; and Group C and D were injected with the same dose of physiological saline. The level of leukocytes rebounded significantly after the treatment with BMMSCs and returned to reference numbers on Day 3 after treatment, which was significantly higher than that in the conventional treatment group. The concentrations of IL-2 and IFN-α were gradually increased during the treatment, and the BMMSC treatment group exhibited significantly higher IL-2 and IFN-α concentrations than the conventional treatment group on Days 3 and 4. The expression of the virus in the blood gradually decreased during the treatment, and the BMMSC treatment group displayed a faster decrease than the conventional treatment group. These results showed the advantages of BMMSC treatment over conventional treatment. This study provides a new BMMSC treatment strategy for canine leukopenia induced by CPV infection and reveals the mechanism by which BMMSC increases leukocytes after CPV infection.

Este estudo tem como objetivo estabelecer uma estratégia terapêutica de leucopenia canina induzida pela infecção por parvovírus canino (CPV) através de infusão intravenosa de células tronco mesenquimais da medula óssea alogênica (BMMSCs) e avaliar o efeito terapêutico de BMMSCs no parvovírus canino. Quarenta cães saudáveis de dois meses de idade foram divididos aleatoriamente em quatro grupos: o grupo de tratamento de BMMSCs (A), o grupo de terapia convencional (B), o grupo de infecção por CPV (C) e um grupo controle (D). Os grupos A, B e C foram infectados oralmente com CPV (103.25 TCID50/mL) a 1mL/kg e o D recebeu a mesma dose de soro fisiológico. Após o início da infecção, o grupo A recebeu uma dose de derivadas da medula óssea (CTMMO) e hidratação como tratamento, o grupo B foi tratado com terapia anti-inflamatória e hidratação, e grupos C e D foram injetados com a mesma dose de soro fisiológico. As concentrações de IL-2 e IFN-α aumentaram gradualmente durante o tratamento, e o grupo de tratamento BMMSC mostrou concentrações significativamente maiores de IL-2 e IFN-α do que o grupo de tratamento convencional nos Dias 3 e 4. A expressão de vírus no sangue diminuiu gradualmente durante o tratamento, e o grupo de tratamento BMMSC mostrou uma diminuição mais rápida do que o grupo de tratamento convencional. Esses resultados mostraram as vantagens do tratamento BMMSC em relação ao tratamento convencional. Este estudo fornece uma nova estratégia de tratamento da BMMSC para leucopenia canina induzida pela infecção por VCP e revela o mecanismo pelo qual a BMMSC aumenta os leucócitos após infecção por VCP.


#3 - Fungi isolated from wild birds in the Marambaia Island, Rio de Janeiro State, southeastern Brazil

In Brazil, the Atlantic Forest has been suffering from deforestation, which has had impacts on its flora, fauna, and microbiota. However, the fungal diversity present in these environments is little known and studied. In this study, a total of 90 samples of 45 wild birds (45 feathers and 45 feces) were collected in Ilha da Marambaia, southeastern Brazil. Filamentous fungi isolated from these samples were identified through macroscopic and microscopic characteristics. Some isolates were identified by molecular biology using the PCR technique. Acremonium, Alternaria, Aspergillus, Cunninghamella, Curvularia, Eurotium, Fusarium, Geotrichum, Neosartorya, Pestalotia, Paecilomyces, Penicillium, Rhizopus, Mucor and Syncephalastrum were identified. These results indicate the presence of saprophytic fungi species in the feathers and feces of wild birds of the capture site. Further studies should be conducted to elucidate if the mycobiota profile modifies with anthropization and if it interferes with bird health and environmental recovery.

No Brasil, a Mata Atlântica vem sofrendo com o desmatamento, que tem impactado sua flora, fauna e microbiota. No entanto, a diversidade fúngica presente nesses ambientes é pouco conhecida e estudada. Neste trabalho, um total de 90 amostras de 45 aves silvestres (45 penas e 45 fezes) foram coletadas na Ilha da Marambaia, Sudeste do Brasil. Fungos filamentosos isolados dessas amostras foram identificados por meio de características macroscópicas e microscópicas. Alguns isolados foram identificados por biologia molecular usando a técnica de PCR. Foram identificados Acremonium, Alternaria, Aspergillus, Cunninghamella, Curvularia, Eurotium, Fusarium, Geotrichum, Neosartorya, Pestalotia, Paecilomyces, Penicillium, Rhizopus, Mucor e Syncephalastrum. Esses resultados indicam a presença de espécies de fungos saprofíticos nas penas e fezes de aves silvestres do local de captura. Novos estudos devem ser realizados a fim de elucidar se o perfil da micobiota se modifica com a antropização e se interfere na saúde das aves e na recuperação ambiental.


#4 - Vertebral osteomyelitis caused by Enterococcus faecalis in broiler chickens from southern Brazil

Abstract in English:

Enterococcal spondylitis affects poultry and causes progressive lameness. This study reports what seems to be the first case of vertebral osteomyelitis caused by Enterococcus in broiler chickens in southern Brazil. We also conducted an experimental infection to evaluate microorganismal characteristics and pathogenicity in broiler chickens. We performed bacterial isolation, identification, and histopathology. The isolates were tested for their growth and survival capacity at different temperatures, pH values, and antimicrobial resistance profiles. The experiment infection was conducted with broiler breeders (n=9). Group 1 = negative control, Group 2 = challenged orally, Group 3 = challenged via air sac. The autopsy was performed on the 50th day of life (DOL). The report showed spondylitis and fusion of thoracic vertebra, accompanied by spinal cord compression, and femoral head necrosis. We used the isolates (n=17) to test their growth at 10°C and 45°C, survival capacity for up to 60° for 30 min, and growth under pH levels from four to 12. Higher resistance was observed against macrolides and quinolones. On experimental infections, all animals expressed signs of lameness and “sitting on the hocks”. Enterococcus faecalis is the causal agent of enterococcal spondylitis in broilers in southern Brazil, which is an underreported and emerging pathological condition that requires attention.

A espondilite enterocócica afeta aves e causa claudicação progressiva. Este estudo relata o primeiro caso de osteomielite vertebral causada por Enterococcus em frangos de corte no sul do Brasil. Também conduzimos uma infecção experimental para avaliar as características microbianas e a patogenicidade em frangos de corte. Realizamos isolamento bacteriano, identificação e histopatologia. Os isolados foram testados quanto ao seu crescimento e capacidade de sobrevivência em diferentes temperaturas, valores de pH e perfil de resistência antimicrobiana. O experimento de infecção foi conduzido com matrizes de corte (n=9). Grupo 1 = controle negativo, Grupo 2 = provocado oralmente, Grupo 3 = provocado via saco aéreo. A autópsia foi realizada no 50º dia de vida (DOL). O laudo mostrou espondilite e fusão de vértebra torácica, acompanhada de compressão da medula espinhal e necrose da cabeça do fêmur. Usamos os isolados (n=17) para testar seu crescimento a 10°C e 45°C, capacidade de sobrevivência até 60° por 30 min e crescimento em níveis de pH de quatro a 12. A maior resistência foi observada contra macrolídeos e quinolonas. Na infecção experimental, todos os animais manifestaram sinais de claudicação e postura sentada sobre os jarretes. Enterococcus faecalis é o agente causal da espondilite enterocócica em frangos de corte no sul do Brasil, que é uma condição patológica emergente e subnotificada que requer atenção.


#5 - Male reproductive tract disorders in equids: 87 cases (2014-2022)

Abstract in English:

Male reproductive tract disorders (MRTDs) are common in equids and may represent a significant proportion of the caseload of equine practitioners. Herein, we determined the frequency, clinico-pathological findings, and therapeutics of MRTDs in equids from a 9-year survey of the clinical records of a Veterinary Teaching Hospital in Midwestern Brazil. During this period, 87 affected equids presented 100 MRTDs distributed in 17 different diagnoses. Forty-three (49.4%) equids presented MRTDs affecting the glans penis or preputial skin fold. Testicles, scrotum, and spermatic cords disorders were diagnosed in 32 (36.8%) horses. Of the remaining 12 (13.8%) horses, six presented two different MRTDs on two distinct anatomical areas, and five animals exhibited the same disease, affecting two different anatomical locations. Only one horse presented three distinct MRTDs on two anatomical regions. Habronemiasis was the most frequent MRTD detected in 26% of the diagnoses, followed by miscellaneous wounds (14%), cryptorchidism (13%), inguinal hernia (13%), funiculitis (9%), paraphimosis (6%), squamous cell carcinoma (5%), urethrolithiasis (3%), preputial abscesses, and orchitis (2% each). Phimosis, ulcerative posthitis, epididymitis, hydrocele, papillomatosis, teratoma, and testicular torsion accounted for 1% each. Sixty-three (72.5%) equids received hospital discharge, 18 (20.6%) were humanely euthanized, and six (6.9%) died. This study detected the most relevant MRTDs that affected over 7.5% in a survey of 1,154 equids referred for hospital care. Our finding highlights that MRTDs may be debilitating and life-threatening conditions, and they can potentially impact the reproduction of equids in the region. This knowledge may support equine practitioners in elaborating proper sanitary and management protocols to prevent and reduce the incidence of most MRTDs recorded, improving equid production and welfare.

Distúrbios do trato reprodutivo masculino (DTRMs) são comuns em equídeos e podem representar uma proporção significativa da casuística de hipiatras. Determinamos a frequência, os achados clínico-patológicos e a terapêutica de DTRMs em equídeos a partir de um levantamento de nove anos dos registros clínicos em um Hospital Veterinário no Centro-Oeste do Brasil. Nesse período, 87 equinos afetados apresentaram 100 DTRMs distribuídos em 17 diagnósticos diferentes. Quarenta e três (49,4%) equídeos apresentaram DTRM afetando a glande peniana ou pregas de pele prepucial. Distúrbios dos testículos, escroto e cordão espermático foram diagnosticados em 32 (36,8%) equinos. Dos 12 (13,8%) equinos restantes, seis apresentaram dois DTRMs diferentes em duas áreas anatômicas distintas, e cinco animais exibiram a mesma doença em duas localizações anatômicas diferentes. Apenas um cavalo apresentou três DTRMs distintas em duas regiões anatômicas. A habronemíase foi a DTRM mais frequente detectada em 26% dos diagnósticos, seguida por feridas diversas (14%), criptorquidismo (13%), hérnia inguinal (13%), funiculite (9%), parafimose (6%), carcinoma de células escamosas (5%), uretrolitíase (3%), abscessos prepuciais e orquite (2% cada). Fimose, postite ulcerativa, epididimite, hidrocele, papilomatose, teratoma e torção testicular representaram 1% cada. Sessenta e três (72,5%) equídeos receberam alta hospitalar, 18 (20,6%) foram sacrificados humanitariamente e seis (6,9%) morreram. Este estudo detectou as DTRMs mais relevantes que afetaram 7,5% dos pacientes em um levantamento de 1.154 equídeos encaminhados para atendimento hospitalar. Este achado destaca que as DTRMs podem ser condições debilitantes e com risco de morte, e podem impactar potencialmente a reprodução de equídeos na região. Este conhecimento pode ajudar os hipiatras a elaborar protocolos sanitários e de manejo adequados para prevenir e reduzir a incidência da maioria das DTRMs registradas, melhorando a produção e o bem-estar dos equídeos.


#6 - Body condition, external morphology, parasitology, and histological and biometrical study of the gastrointestinal tract of Sporophila nigricollis and Sporophila caerulescens seized from trafficking in Northeastern Brazil

Abstract in English:

The objective was to analyze Sporophila nigricollis and Sporophila caerulescens in terms of body conditions, parasitological, bacteriological, external biometric measurements, and histological analysis of the gastrointestinal tract. We used 115 individuals apprehended from 2020 to 2021 and sent to the Wild Animal Triage Center, Paraíba, Brazil. Concerning weight, S. nigricollis, females and males presented 9.36±1.36g and 9.70±1.39g, and S. caerulescens with 10.5±0.70g and 11.75±1.28g, being the second heaviest. The body condition in 69.62% of S. nigricollis and 62.50% of S. caerulescens was good. In the fecal microbiological examination, 40.25% of S. nigricollis and 40% of S. caerulescens were positive for Isospora sp. Dispharynx sp. was reported for the first time in S. nigricollis and S. caerulescens. In fecal bacteriology, 95.65% of S. nigricollis were identified with Gram-positive cocci and 73.91% with Gram-positive bacilli. Forty-one animals died and were placed under refrigeration for 48 hours; of these, 31.16% of S. nigricollis and 37.50% of S. caerulescens presented inadequate body conditions. 19.51% had gastrointestinal tract engorgement with hemorrhagic points, and 14.63% had hepatic alteration due to yellowish coloration. The study contributes with subsidies for the taxonomic elucidation of the genus Sporophila, in addition to the knowledge of the conditions in which birds are found in sorting centers, once the destination of most of the seized animals is the release, thus, a potential source of pathogens to the natural environment.

O objetivo foi analisar Sporophila nigricollis e Sporophila caerulescens quanto às condições corporais, parasitológicas, bacteriológicas, medidas biométricas externas e análise histológica do trato gastrointestinal. Foram utilizados 115 indivíduos apreendidos entre 2020 e 2021 e encaminhados ao Centro de Triagem de Animais Silvestres, Paraíba, Brasil. Quanto ao peso, S. nigricollis, fêmeas e machos apresentaram, 9,36±1,36g e 9,70±1,39g, e S. caerulescens com 10,5±0,70g e 11,75±1,28g, sendo o segundo mais pesado. A condição corporal em 69,62% de S. nigricollis e 62,50% de S. caerulescens foi boa. No exame microbiológico fecal, 40,25% de S. nigricollis e 40% de S. caerulescens foram positivos para Isospora sp. Dispharynx sp. foi relatada pela primeira vez em S. nigricollis e S. caerulescens. Na bacteriologia fecal, 95,65% dos S. nigricollis foram identificados com cocos Gram-positivos e 73,91% com bacilos Gram-positivos. Quarenta e um animais morreram e foram colocados sob refrigeração por 48 horas; destes, 31,16% de S. nigricollis e 37,50% de S. caerulescens apresentaram condições corporais inadequadas. 19,51% apresentaram ingurgitamento do trato gastrointestinal com pontos hemorrágicos e 14,63% alteração hepática devido à coloração amarelada. O estudo contribui com subsídios para a elucidação taxonômica do gênero Sporophila, além do conhecimento das condições em que as aves se encontram nos centros de triagem, uma vez que a maior parte dos animais apreendidos tem como destino a soltura, sendo assim, uma potencial fonte de patógenos para o ambiente natural.


#7 - Fusobacterium necrophorum predominates in the microbiota of mandibular dental abscess in Blastocerus dichotomus

Abstract in English:

Dental abscess in ruminants is an acute polymicrobial infection, usually resulting from periodontal disease or endodontic infection, with consequences for animal health and welfare. The present study aimed to describe the bacterial microbiota of dental abscesses in Blastocerus dichotomus. Biological material from mandibular dental abscesses, punctured with a sterile syringe and needle during routine veterinary curative procedures or necropsies, was collected from three ex-situ marsh deer. Bacteria were identified using high-throughput sequencing of the 16S ribosomal RNA gene. The three specimens had the presence of facial bulging, and two died because of severe emaciation with a history of progressive weight loss. Bacteroides (38.6%), Fusobacterium (36.65%), and Porphyromonas (7.49%) represented the most abundant genera and Fusobacterium necrophorum (35.69%), Porphyromonas levii (3.12%) and Porphyromonas gulae (1.78%) were among the ten most represented species in the microbiota of mandibular abscess in Blastocerus dichotomus. These molecular findings demonstrate a broader diversity of species in the polymicrobial nature of dental abscesses in B. dichotomus than was previously reported when culture-dependent methods were used in the diagnosis.

O abcesso dentário em ruminantes é uma infeção polimicrobiana aguda, geralmente resultante de doença periodontal ou infeção endodôntica, com consequências para a saúde e bem-estar animal. O presente estudo teve como objetivo descrever a microbiota bacteriana de abscessos dentários em Blastocerus dichotomus. Material biológico de abscessos dentários mandibulares, puncionados com seringa e agulha estéril durante procedimentos curativos veterinários de rotina ou necropsias, foi coletado de três cervos do pantanal criados ex situ. As bactérias foram identificadas usando sequenciamento de alto rendimento do gene 16S ribossomal RNA. Os três espécimes apresentavam abaulamento facial e dois faleceram por emagrecimento severo com histórico de emagrecimento progressivo. Bacteroides (38,6%), Fusobacterium (36,65%) e Porphyromonas (7,49%) representaram os gêneros mais abundantes e Fusobacterium necrophorum (35,69%), Porphyromonas levii (3,12%) e Porphyromonas gulae (1,78%) estavam entre as dez espécies mais representadas na microbiota do abscesso mandibular em Blastocerus dichotomus. Esses achados moleculares demonstram uma diversidade mais ampla de espécies na natureza polimicrobiana dos abscessos dentários em B. dichotomus, do que o relatado anteriormente quando métodos dependentes de cultura foram utilizados no diagnóstico.


#8 - Molecular epidemiology of Clostridioides difficile obtained from fecal samples of wild animals in Brazil

Abstract in English:

Clostridioides difficile is a strictly anaerobic, spore-forming Gram-positive bacterium associated with diarrhea, known as C. difficile infection (CDI). In domestic animals, C. difficile is considered an important pathogen mostly in pigs and horses, but there are also reports in other domestic species. In wild animals, the epidemiology of C. difficile is largely unknown, and the role of the bacterium as a cause of diarrhea is unclear. The aim of this study was to determine the prevalence of C. difficile in the feces of wild animals referred to the Center of Medicine and Research in Wild Animals (CEMPAS). Fecal samples obtained from 100 animals of 34 different species were subjected to qPCR for the detection of the C. difficile 16S rRNA gene and two major toxin genes (tcdA and tcdB) and to anaerobic bacterial isolation. A total of 63 animals (63%) were positive for C. difficile by qPCR, and 16 isolates were recovered. The opossum (Didelphis spp.) had the highest number of positive animals in both tests (from 21 samples, 19 were qPCR positive, and four isolates were recovered). Three toxigenic strains (RT 002, 004, and 014), all previously described as infecting humans and animals, were isolated in the following species: bearded dragon (Pogona vitticeps), pampas fox (Lycalopex vetulus), and marmoset (Callithrix sp.). The presence of C. difficile in the feces of wild animals highlights the importance of wildlife as potential carriers of infection for production animals or humans.

O Clostridioides difficile é uma bactéria Gram-positiva estritamente anaeróbica e formadora de esporos, associada à diarreia e conhecida como infecção por C. difficile (CID). Em animais domésticos, o C. difficile é considerado um patógeno importante principalmente em porcos e cavalos, mas também há relatos em outras espécies domésticas. Em animais selvagens, a epidemiologia do C. difficile é amplamente desconhecida, e o papel da bactéria como causa de diarreia não está claro. O objetivo deste estudo foi determinar a prevalência do C. difficile nas fezes de animais selvagens encaminhados ao Centro de Medicina e Pesquisa em Animais Selvagens (CEMPAS). Amostras de fezes obtidas de 100 animais de 34 espécies diferentes foram submetidas à qPCR para a detecção do gene 16S rRNA do C. difficile e dois principais genes de toxina (tcdA e tcdB), além de isolamento bacteriano anaeróbico. Um total de 63 animais (63%) foram positivos para C. difficile por qPCR, e 16 isolados foram recuperados. O gambá (Didelphis spp.) apresentou o maior número de animais positivos em ambos os testes (de 21 amostras, 19 foram positivas na qPCR, e quatro isolados foram recuperados). Três cepas toxigênicas (RT 002, 004 e 014), todas previamente descritas como infectando humanos e animais, foram isoladas nas seguintes espécies: dragão barbado (Pogona vitticeps), raposa-pampas (Lycalopex vetulus) e sagui (Callithrix sp.). A presença de C. difficile nas fezes de animais selvagens destaca a importância da vida selvagem como potencial portadora de infecção para animais de produção ou seres humanos.


#9 - Spontaneous and experimental poisoning by Myrocarpus frondosus in cattle

Abstract in English:

An outbreak of photosensitization in Holstein and Jersey cattle occurred after the passage of an extratropical cyclone in Southern Brazil. On that occasion, several trees fell, including some of the species Myrocarpus frondosus (Fabaceae family) popularly known as “gabreúva”, “cabriúna” or “gabriúna”. Five heifers consumed the leaves of this plant, and all of them became ill. The main clinical signs were decreased appetite, permanence in the water, decreased ruminal movements, dry stools, congested and icteric sclera and conjunctivae, dark urine and restlessness when exposed to the sun. One heifer showed circling, incoordination, restlessness and death six days after the onset of clinical signs. From the seventh day, three heifers showed improvement; one remained apathetic, with jaundiced mucous membranes, ulceration, and scaling on the tongue, increased respiratory rate, and fever, and died 18 days after ingestion of the plant. At necropsy, photosensitivity lesions were observed on the depigmented skin, snout, teats, and ear tip, characterized by discontinuity of the skin with the formation of crusts, inflammatory exudate associated with redness of the skin, in addition to ulceration in the mouth, snout and ventral portion of the tongue and jaundice and edema in the submandibular region. The liver was enlarged, with an orange color, full gallbladder, and wall edema. Through microscopy, liver lesions were characterized by moderate and diffuse necrosis of hepatocytes, vacuolar and centrilobular degeneration, bilirubin retention and proliferation of biliary epithelium. The literature shows no reports of M. frondosus toxicity in cattle. Experiments were carried out in cattle to clarify the possible clinical-pathological picture produced by this plant. A survey of the epidemiological data of the diagnosed spontaneous outbreak was carried out, and subsequently, three cattle received the plant’s green leaves, and a fourth received the dried leaves. After administering the plant, blood samples were collected daily for blood count, measurement of liver enzymes (gamma glutamyl transferase, alkaline phosphatase, alanine aminotransferase, aspartate transferase), bilirubin, measurement of vital parameters and liver biopsy from the onset of signs. In case of death, a necropsy was performed with collection of viscera samples for macro and microscopic evaluation. The experimental part was developed at the Laboratory of Animal Pathology, CAV-UDESC. The single dose of 33g/kg of green leaves caused severe clinical signs, while the administration of 22g/kg and 11g/kg caused mild to moderate signs of the disease. The supply of desiccated leaves in a single dose did not cause clinical manifestations. M. frondosus can be blamed as a cause of spontaneous hepatogenous photosensitization in cattle. Experimentally, the ingestion of green leaves of this plant, in doses greater than 11g/kg can lead to clinical changes, with subsequent recovery, or progress to death with severe liver damage.

Abstract in Portuguese:

Um surto de fotossensibilização em bovinos da raça Holandesa e Jersey foi observado após a passagem de um ciclone extratropical no Sul do Brasil, quando houve queda de diversas árvores, dentre elas Myrocarpus frondosus (família Fabaceae), popularmente conhecida como “gabreúva, cabriúna ou gabriúna”. Cinco novilhas consumiram folhas dessa planta, e todas adoeceram. Os principais sinais clínicos foram diminuição do apetite, permanência dentro da água, diminuição dos movimentos ruminais, fezes ressecadas, esclera e conjuntivas congestas e ictéricas, urina escura e inquietação quando expostas ao sol. Uma novilha apresentou andar em círculos, incoordenação e inquietação e morte seis dias após início dos sinais clínicos. A partir do sétimo dia, três novilhas apresentaram melhora e uma permaneceu apática, com as mucosas ictéricas, ulceração e descamação na língua, aumento da frequência respiratória, febre e morreu 18 dias após ingestão da planta. Na necropsia foram observadas lesões de fotossensibilização na pele despigmentada, focinho, tetos, ponta da orelha, caracterizadas por descontinuidade da pele com formação de crostas, exsudato inflamatório associado a vermelhidão da pele além de ulceração na boca, focinho e porção ventral da língua e icterícia acentuada e edema, na região submandibular. O fígado estava aumentado de volume, de cor laranja, vesícula biliar repleta e com edema de parede. Através da microscopia foram observadas, lesões hepáticas, caracterizadas por necrose de hepatócitos, moderada e difusa, degeneração vacuolar, centrolobular, retenção de bilirrubina e proliferação de epitélio biliar. Na literatura, não há relatos da toxicidade de M. frondosus para bovinos. Com o objetivo de esclarecer o possível quadro clínico-patológico produzido por essa planta foram conduzidos experimentos em bovinos. Foi realizado um levantamento dos dados epidemiológicos do surto espontâneo diagnosticado e, posteriormente, três bovinos receberam as folhas verdes da planta e um quarto recebeu as folhas dessecadas. Após a administração da planta, foram coletadas amostras de sangue diariamente para hemograma, dosagem de enzimas hepáticas (gama glutamil transferase, fosfatase alcalina, alanina aminotransferase, aspartato transferase), bilirrubina, aferição dos parâmetros vitais e biópsia hepática a partir do surgimento dos sinais clínicos. Em caso de morte, foi realizada necropsia com coleta de amostras de vísceras para avaliação macro e microscópica. A parte experimental foi desenvolvida nas dependências do Laboratório de Patologia Animal, CAV-UDESC. A dose única de 33g/kg de folhas verdes causou graves sinais clínicos, enquanto a administração de 22g/kg e 11g/kg causou sinais leves a moderados da doença. O fornecimento de folhas dessecadas em dose única não causou manifestações clínicas. M. frondosus pode ser responsabilizada como causa de fotossensibilização hepatógena espontânea em bovinos. A ingestão de folhas verdes dessa planta em doses superiores a 11g/kg pode levar a alterações clínicas, com posterior recuperação, ou evoluir para morte com lesões hepáticas graves.


#10 - Detection of virulence genes, antibiotic resistance genes and antibiotic resistance of Escherichia coli isolated from diarrheic lambs in Anhui Province, China

Lamb diarrhea seriously restricts the development of the sheep industry. Infectious pathogens often cause diarrhea, and E. coli isolates are highly distributed in infectious diarrhea. The study aimed to investigate the prevalence of pathogenic E. coli in diarrhea lambs and determine the distribution of virulence genes, antibiotic resistance genes, and antibiotic resistance. One hundred seventy-eight E. coli isolates were isolated from the feces of 204 diarrhea lambs. The virulence genes mdh, hlyF, iss, ompA, fimC, iucD, st, lt, stx1, stx2, and antibiotic resistance genes including tetA, tetB, aac (6′)-II, blaCMY-2, qnr, aadA1, sul1, blaTEM, blaCTX-M were detected by polymerase chain reaction (PCR). Antibiotic resistance was determined by Kirby-Bauer Disc Diffusion method. There were 109 (61.24%, 109/178) enterotoxigenic E. coli (ETEC), 119 (66.85%, 119/178) Shiga toxin-producing E. coli (STEC), and 95 (53.37%, 95/178) hybrid STEC/ETEC isolates. The highest prevalent virulence genes were ompA (80.90%, 144/178) and fimC (67.98%, 121/178), and the lowest was iucD (8.99%, 16/178). The most commonly detected antibiotic resistance genes were tetA/tetB (92.70%, 165/178), qnr (75.84%, 135/178), and sul1 (62.92%, 112/178), no blaTEM or blaCTX-M genes were detected. All isolates had high antibiotic resistance to lincomycin (96.63%, 172/178), tetracycline (88.76%, 158/178), and co-trimoxazole (80.34%, 143/178), and the multidrug resistant (MDR) rate reached 93.82% (167/178). The high prevalence of ETEC and STEC indicates that E. coli is one of the critical pathogenic agents leading to diarrhea in lambs in the region, and its high antibiotic resistance, especially MDR, should be brought to our attention.
