Embrapa Pantanal, Rua 21 de Setembro 1880, Corumbá, MS 79320-900, Brazil. E-mail: aiesca@cpap.embrapa.br An indirect enzyme-linked immunosorbent assay was developed to detect antigen-specific secretory IgA antibodies to Campylobacter fetus subsp. venerealis in bovine vaginal mucus with a protein extract of the Campylobacter fetus subsp. venerealis by the acid glycine extraction method. Mean optical density measurement (l=450 nm) was 0.143±0.9. The most immunoreactive protein bands of the Campylobacter fetus subsp. venerealis or Campylobacter fetus subsp. fetus recognized by IgA in immunoblotting, using bovine vaginal mucus samples, migrate at 42.6 kDa. The protein that migrates at 93 kDa was recognized exclusively for C. fetus subsp. venerealis. A positive vaginal mucus sample of a cow from negative herd recognized antigens of C. jejuni subsp. jejuni e C. fetus subsp. fetus.

#12 - Electrophoretic profile and concentration of immunoglobulins G (IgG) in blood serum of Saanen goats with experimental mastitis induced by Staphylococcus aureus suplemented with vitamin E, 30(1):79-86

Abstract in English:

ABSTRACT.- Fonteque J.H., Kohayagawa A., Mattoso C.R.S., Lopes S.T.A., Paes P.R.O., Cassetari M.L. & Langoni H. 2010. [Electrophoretic profile and concentration of immunoglobulins G (IgG) in blood serum of Saanen goats with experimental mastitis induced by Staphylococcus aureus suplemented with vitamin E.] Perfil eletroforético e concentração de imunoglobulinas G (IgG) do soro sanguíneo de cabras Saanen com mastite experimental induzida por Staphylococcus aureus suplementadas com vitamina E. Pesquisa Veterinária Brasileira 30(1):79-86. Departamento de Medicina Veterinária, Hospital de Clínica Veterinária, Centro de Ciências Agroveterinárias, Universidade do Es-tado de Santa Catarina, Lages, SC 88520-000, Brazil. E-mail: fonteque@cav.udesc.br The objective was to evaluate the electrophoretic profile of proteins and serum concentration of immunoglobulin G (IgG) in Saanen goats with mastitis experimentally induced by Staphylococcus aureus (dl-a-tocopherol acetated). 14 adult goats, (supplemented with vitamin E DL-a-tocopherol) primiparous pregnant, seronegative for caprine arthritis encephalitis (CAEV), clinically healthy, were divided into two groups of seven animals: Not supplemented group (G1) and group supplemented with 2.000 UI of DL-a-tocopherol (G2 Vit E), by intramuscular injection on the day of the parturition and seven days later. At the 9th day after delivery 300 UFCs of the S. aureus ATCC 225923 strain were inoculated into the left half of the mammary gland of each animal. The mastitis was determined through collection of milk samples for evidence of infection by means of bacteriological examination, somatic cell count (SCC) and California Mastitis Test (CMT). Then samples were collected after 12, 24, 48 and 72 hours, antimicrobial intra-mammary gland treatment was initiated, with new evaluation 48 hours after treatment. The electrophoretic profile of serum protein of the goats, showed five fractions, as follows: albumin and globulin (a, b1, b2 e g-globulin). There was an increase in the production of g-globulin and lower production of b2-globulin fraction 12 hours after infection, and faster decrease in the supplemented group, showing the influence of vitamin E in the production of acute phase proteins. There was no influence of vitamin E in the serum concentration of immunoglobulin G (IgG) in supplemented animals. The supplementation with vitamin E increased the concentration of immunoglobulin and decreased the production of acute phase proteins, probably the antioxidant effect minimizing the tissue injury during the inflammatory process in the mammary gland.

[Passive transfer of immunity in Nelore and Limousin calves and serum proteinogram in the first four months of life.] Postal 6001, Londrina, PR 86051-990, Brazil. E-mail: janlisboa@uel.br To study the passive transfer of immunity, 90 healthy calves, 45 Nelore and 45 Limousin, were grouped (n=15) according to their dam’s parity: first, second or third or more calvings. Blood samples were draw from each calf with 24 to 36 hours of life and 15, 30, 60, 90, and 120 days. The total serum (TSP) and plasma (TPP) proteins, gamaglutamiltransferase activity (GGT), serum albumin, alpha, beta and gammaglobulin by electrophoresis in agarose gel and IgG estimated by the zinc sulphate turbidity test were determined. Two-way-ANOVA was used to compare the data in the first age. The age behaviour was analysed through repeated measures ANOVA. Correlations were established between the variables. The passive transfer of immunity was successful in both breeds and the parity of the dam had no effect on the calves’ serum gammaglobulin concentration. The levels of gammaglobulins were higher at the end of the first day of life, and decreased until 60 days. From there on, the increase due to the active production of antibodies was precocious in taurine calves and slower in Zebu calves. At the end of the first day of life, the gammaglobulin was correlated with IgG (r=0,859), TPP (r=0,807), TSP (r=0,811) and GGT (r=0,399). The variation of serum proteins followed the normal pattern throughout the first four months of life, with little differences between taurine and Zebu calves.

Abstract in English:

Simões S.V.D., Costa R.G., Souza P.M., Medeiros A.N. & Vilar A.L.T. 2005. [Passive immunity, neonatal morbidity and performance of kids in different colostrum management.] E-mail: saravilar@bol.com.br The experiment was performed to determine the kid management that ensures the best passive immunity, and to evaluate the relationship between passive immunity, neonatal morbidity and performance. Blood samples were obtained from 58 Saanen kids before colostrum ingestion and 30h after. The kids were submitted to five treatments: (T1) kids were allowed to nurse the dam for 24 hours ingesting colostrum ad libitum; (T2) kids were bottle-fed with 200mL colostrum in the first hour of life; (T3) kids were bottle-fed with 400 mL colostrum, 200mL in the first hour and 200mL 8 hours after birth; (T4) kids were bottle-fed with 400mL colostrum, 200mL in the first hour and 200mL 14 hours after birth; (T5) kids were bottle-fed with 600mL of colostrum, 200mL in the first hour, 200ml 12 hours after birth, and 200mL at 24 hours of life. Serum levels of gamaglobulins were determined by electrophoresis. The occurrence of diseases was monitored from birth to 28 days. Daily weight gains were used to evaluate the performance until weaning. The lowest concentration of g-globulin was observed in Group 2 (1,57g/dL) and the highest in Group 3 (2,60g/dL). In spite of higher levels of passive immunity Group 3 showed more animals with scours, however no statistical differences were observed. The immune status at 30 hours postpartum was not associated with neonatal weight gain.
