#1191 - Phenotypical characterization of Candida spp. isolated from crop of parrots (Amazona spp.), p.452-456

Abstract in English:

ABSTRACT.- Renata G. Vieira R.G. & Acqua Coutinho S.D. 2009. Phenotypical characterization of Candida spp. isolated from crop of parrots (Amazona spp.). Pesquisa Veterinária Brasileira 29(6):452-456. Curso de Pós-Graduação em Imunopatologia Veterinária da Universidade Paulista, Rua Agariba 48, São Paulo, SP 05053-010, Brazil. E-mail: selene@uol.com.br The purpose of this study was to characterize Candida isolates from crop of parrots. Forty baby parrots of genus Amazona, species aestiva and amazonica that were apprehended from wild animal traffic were used: 18 presented ingluvitis and 22 other alterations, but showing general debilitation. Samples were seeded on Sabouraud dextrose agar with chloramphenicol after be obtained by the introduction of urethral probe through the esophagus. Based on morphology and biochemical reactions (API 20C) Candida was confirmed; it was still searched the production of proteinase and phospholipase, virulence factors for Candida species. Candida spp. were isolated from 57.5% parrots, being 72.2% from birds with ingluvitis and 45.5% from without ones. Twenty-five strains of Candida were isolated, 60% and 40%, respectively from parrots with and without ingluvitis, and were speciated: 28% C. humicola, 24% C. parapsilosis, 20% C. guilliermondii, 20% C. famata, and 8% C. albicans. These results demonstrate that C. albicans is not the most frequent species isolated, and it is the first report that shows C. guilliermondii, C. famata, and C. humicola causing infection in parrots. Many isolates presented filamentation (76%), 100% produced proteinase and 68% phospholipase. The observation of Candida spp. producing virulence factors reinforce the pathogenic role of these yeasts in the cases studied.

#1192 - An outbreak of malignant catarrhal fever in Murrah buffaloes in Minas Gerais, Brazil, p.395-400

Abstract in English:

ABSTRACT.- Costa E.A., Bastianetto E., Vasconcelos A.C., Bomfim M.R.Q., Fonseca F.G., Gomes A.D., Leite R.C. & Resende M. 2009. An outbreak of malignant catarrhal fever in Murrah buffaloes in Minas Gerais, Brazil. Pesquisa Veterinária Brasileira 29(5):395-400. Departamento de Microbiologia, Instituto de Ciências Biológicas, Universidade Federal de Minas Gerais, Av. Antônio Carlos 6627, Belo Horizonte, MG 31270-901, Brazil. E-mail: lvc@icb.ufmg.br An outbreak of Malignant Catarrhal Fever (MCF) resulted in death of five female buffaloes and one domestic cow from the same farm. Four buffaloes died 10-15 days after the appearance of clinical signs, while the fifth was euthanized in extremis, after similar clinical signs. Histopathological lesions included multifocal histiolymphocytic epicarditis, myocarditis and lymphocytic interstitial pneumonia, which are commonly seen in cases of MCF in buffaloes. Furthermore, lymphocytic vasculitis centered in the adventitia, with occasional fibrinoid necrosis in the muscular layer, was found in the kidneys, liver, spleen, lymph nodes and brain. Nucleotide sequencing of DNA fragments from the central nervous system amplified by PCR revealed 98% similarity with known OHV-2 sequences from Genbank. Additionally, PCR analysis also revealed the presence of OHV-2 DNA in the peripheral mononuclear blood cells of two clinically healthy buffaloes. The diagnosis of MCF was based on epidemiological, clinical, gross and histopathological findings and on the results of a semi-nested PCR followed by nucleotide sequencing.

E-mail: davetpat@ufrgs.br Leguminous bloat may occur in cattle which graze pastures consisting of lush forages. In a dairy farm located on the municipality of São Francisco de Paula, Rio Grande do Sul, eight out of 66 cows died suddenly after being transferred to a paddock whose pastures were composed of Trifolium repens and Trifolium pratense. Animals were found dead in the morning of the next day after being transferred; no clinical signs were noticed. Main gross findings included enhanced abdominal volume, protrusion and congestion of the tongue and vagina, ruminal distension, pale liver, and enhanced spleen. Histologically, there were lung congestion and edema, and splenic lymphoid hyperplasia. The evidence of leguminous forages consumption associated with the pathological findings and the absence of growth on bacteriology confirmed the diagnosis.

E-mail: diomedes@ufpa.br A condition of buffaloes characterized by an increased volume of one of the cheeks was shown to be due to accumulation of the seeds of the palm-tree “mucaja” (Acrocomia aculeata) and of grass, in the oral vestibulum during rumination. This caused compressive atrophy with thinning and medial deviation of the corresponding submaxillary bone and exposition of the roots of molar teeth. The seeds appear to be of good palatability to the buffaloes.

E-mail: rinaldo.mota@hotmail.com The objective of this paper was to study the molecular performance and phenotypic characterization of Burkholderia mallei isolated from horses with clinical and serological diagnosis of glanders, originating from the Metropolitan District of Recife and Zona da Mata of Pernambuco and Alagoas. The isolation and biochemical identification of B. mallei was carried out by microbiological and molecular techniques of PCR-fingerprinting and RAPD-PCR. From the eight samples studied, four showed little phenotype variations. In the molecular tests, the samples formed 4 groups of different ribotype profiles and 4 genotype profiles. There was some association of PCR-fingerprinting with RAPD-PCR results. It was concluded that the slight biochemical variations were not associated with different molecular profiles. They also indicated that these differences show heterogeneity associated with the origin of the sample, indicating that the infection was caused by clones of different strains and that the polymorphism of DNA observed could make it

Pesquisa Veterinária Brasileira 29(4):303-311. Laboratório de Neurogastroenterologia Experimental, Universidade Paranaense, Praça Mascarenhas de Moraes 4286, Umuarama, PR 87502-210, Brazil. E-mail: eduardoaraujo@unipar.br The behavior of the gill epithelium of tilapias cultured in tanks at different altitudes and interconnected with PVC pipes was analyzed. Gill filaments of four specimens from four tanks (T1, T2, T3 e T4) sequentially interconnected were submitted to histological routine to obtain 5-mm-thick cuts that were stained with HE or submitted to histochemistry reactions PAS + diastase solution or Alcian Blue pH 2.5 or Alcian Blue pH 1.0. Considering the intermediary, apical and basal regions of the filaments, the lamellar area was measured and the amount of mucous cells was counted. It was verified that oxygen, pH, and temperature decreased progressively as the water flew from one tank to another. Thus, an increase was realized of the amount of mucous cells and the lamellar area in T2, as well as a progressive decrease of these measures on the tanks which received water from T2. Moreover, detachment of the gill epithelium, cellular hyperplasia in the interlamellar space, and telangectasias were observed in the fishes from T2, T3 e T4. It was concluded that the aquatic environment in tanks sequentially interconnected with PVC pipes suffers alterations from one tank to another, as that physico-chemical fluctuations reflect on the behavior of the gill epithelium through variations of the lamellar area and the amount of mucous cells.

#1199 - Evaluation of left ventricular diastolic echocardiographic parameters in healthy dogs by pulsed-wave Doppler, p.291-294

Abstract in English:

ABSTRACT.- Pereira G.G., Petrus L.C., Santos A.L.F., Yamaki F.L. & Larsson M.H.M.A. 2009. Evaluation of left ventricular diastolic echocardiographic parameters in healthy dogs by pulsed-wave Doppler. Pesquisa Veterinária Brasileira 29(4):291-294. Departamento de Clínica Médica, Faculdade de Medicina Veterinária e Zootecnia, Universidade de São Paulo, Av. Prof. Orlando Marques de Paiva 87, São Paulo, SP 05508-900, Brazil. E-mail: ggpereira2002@yahoo.com.br Left ventricular diastolic dysfunction plays an important role on heart failure progression. In order to obtain additional reference values of left ventricular diastolic parameters and investigate influence of common variables, peak E wave (peak E), peak A wave (peak A), E/A ratio (E/A), E wave deceleration time (EDT) and isovolumic relaxation time (IRVT) were studied in 40 clinically healthy dogs, by pulsed wave Doppler. The following values were obtained: peak E = 0.747 ± 0.117 m/s, peak A = 0.487 ± 0.062 m/s, E/A = 1.533 ± 0.198, EDT = 88.7 ± 9.2 ms and IRVT = 0.080 ± 0.009 s. Some parameters were influenced by heart rate (peak E, peak A and IRVT), by age (peak A and E/A) and by body weight (TRIV). Gender influence was absent. Values obtained can be used as reference for canine specimens but its interpretation should consider on the influence of related variables.

It was found that: (1) at 4 months of age the seminiferous tubules were poorly developed, appeared as seminiferous cords without lumen, lined by a low epithelium, and showed undifferentiated Sertoli cells and scarce interstitial tissue; (2) at 5 months the seminiferous tubules began to differentiate with increase in tubular diameter and lumen, the other tubular structures remaining similar to those previous referred; (3) at 6 and 7 months of age spermatocytogenesis began to appear, Leydig cells were large, polyhedral in shape, with vacuolated cytoplasm and clear nuclei, resting on a sparse interstitial tissue with few blood vessels; (4) 1-year-old cats showed testicular histological features of an adult animal, had seminiferous tubules of large diameter and high seminiferous epithelium with small lumen, and Leydig cells of different sizes, with polyhedral shape, vacuolated cytoplasm, clear nuclei and evident nucleoli resting in a sparse interstitial tissue with some blood vessels; (5) in Group 1 the average diameter of the seminiferous tubules was 160.58µm, and 185.94µm in Group 2; (6) the height of the seminiferous epithelium was 49.51µm for Group 1 and 63.29µm for Group 2; (7) the largest measures of the analyzed parameters were found in animals of Group 2, with functional reproductive organs.
