#21 - Occurrence of Mollicutes and Ureaplasma spp. in outbreak of reproductive disease in cattle herds, State of Paraíba, Brazil, 33(3):315-318

Dom Manoel de Medeiros s/n, Recife, PE 52171-900, Brazil. E-mail: sanbsantos@gmail.com In March of 2012 was investigated a reproductive disease outbreak in cattle herds from Paraíba State, Brazil. Were examined 32 cows and two bulls Giroland breed. The cows showed signs and symptoms of reproductive failure such as repeat breeding, granular vulvovaginitis, infertility and abortions. Vaginal and preputial mucous samples were collected for analysis by PCR and isolation. The PCR reactions for Mollicutes and Ureaplasma spp. were realized with primers MGSO and GPO3, and UGP’F and UGP’R respectively. The nested PCR assay for Ureaplasma diversum was realized with primers UD1, UD2, UD3 and UD4. For bacteriologic isolation, obtained samples were diluted up to 10-1 at 10-5, inoculated into liquid and solid “UB” medium, and incubated for up to 21 days, at 37oC in microaerophilie jar. In the PCR reactions the frequency of Mollicutes detected in the analyzed vaginal mucous samples was 65.6, for Ureaplasma spp. was 50.0, while for U. diversum was 15.6. The frequency for isolation of Mollicutes was of 57.1 and for Ureaplasma spp. was of 28.6. In the UB agar was visualized growth of Mycoplasma spp. and Ureaplasma spp., associated in six of the samples. In the cows the presence of Mollicutes and Ureaplasma spp. was confirmed for the reproductive disease outbreak in the semiarid region of Paraiba.

E-mail: sanbsantos@gmail.com Em março de 2012 foi diagnosticado um surto de doença reprodutiva em rebanho bovino no Estado da Paraíba, Brasil. Foram examinadas 32 vacas e dois touros da raça Girolando. As vacas apresentaram sinais de doença reprodutiva como repetição de cio, vulvovaginite granular, infertilidade e abortos. As amostras de suabes vaginais e prepuciais foram colhidas e submetidas a isolamento bacteriano e PCR. As reações da PCR para Mollicutes e Ureaplasma spp. foram realizadas com os iniciadores MGSO-GPO3 e UGP’F-UGP’R, respectivamente. Na Nested PCR para Ureaplasma diversum, os iniciadores usados foram UD1, UD2, UD3 e UD4. Para isolamento bacteriano, as amostras foram diluídas de 10-1 até 10-5, semeadas em meio “UB”, líquido e placa, sendo incubadas por até 21 dias a 37oC em jarra de microaerofilia. A frequência de Mollicutes detectada na PCR foi de 65,6% e para Ureaplasma spp. foi de 50,0%, enquanto que para U. diversum foi de 15,6%. No isolamento a frequência de Mollicutes foi de 57,1% e para Ureaplasma spp. foi de 28,6%. No ágar “UB” foi visualizado o crescimento misto de Mycoplasma spp. e Ureaplasma spp. em seis amostras. Foi confirmado o envolvimento de micro-organismos da Classe Mollicutes em surto de doença reprodutiva em vacas no sertão paraibano.

E-mail: sampaio.elizabeth@gmail.com The objective of this study was to evaluate the in vitro efficacy of commercial disinfectants used in pre- and post-dipping, against Staphylococcus spp. isolated from milk originating from dairy cattle farms in the Wasteland and Forest Zone of Alagoas, Brazil. We used iodine (0.57%), chlorhexidine (2.0%), chlorine (2.5%) and quaternary ammonium compound (4.0%) at concentrations indicated, conventionally used as commercial disinfectants before and after dipping. We analyzed a total of 97 isolates of Staphylococcus spp. identified as S. aureus (16), coagulase positive Staphylococcus (7) and coagulase-negative Staphylococcus (74). The disinfectants were evaluated at three different times (15”, 30” and 60”). We found that 56.3% of Staphylococcus aureus was sensitive to iodine, 68.8% to chlorine, 87.5% to chlorhexidine, and 37.5% to the compound of ammonia, in time 60”. As for coagulase positive staphylococci (CPS), 100% of the isolates was resistant to chlorhexidine, 85.7% to the ammonia compound, 57.1% to chlorine, and 42.9% iodine, in time 60”. Regarding coagulase negative staphylococci (CNS), 91.9% was sensitive to chlorhexidine, 70.3% to chlorine, 66.2% to iodine, and 24.3% the ammonium compound, at time 60”. It is concluded from this study that the greatest disinfectant activity in vitro was with chlorhexidine and chlorine for S. aureus, with iodine and chlorine for SCP, and with chloride and chlorhexidine for SCN. Due to variations in the sensitivity and resistance profile found, it is necessary for regular assessments of the effectiveness of disinfectants used on the farms, to observe the effectiveness of the product and thus ensure the control of mastitis in the herd.

Analisou-se um total de 97 isolados de Staphylococcus spp. identificados como S. aureus (16), Staphylococcus coagulase positiva (7) e Staphylococcus coagulase negativa (74). Os desinfetantes foram avaliados em três tempos distintos (15”, 30” e 60”). Observou-se que 56,3% de Staphylococcus aureus foram sensíveis ao iodo, 68,8% sensíveis ao cloro, 87,5% à clorexidine e 37,5% ao composto de amônia no tempo de 60”. Quanto aos Staphylococcus coagulase positiva (SCP), 100% dos isolados foram resistentes ao clorexidine, 85,7% ao composto de amônio, 57,1% ao cloro, e 42,9 resistentes ao iodo no tempo de 60”. Em relação aos Staphylococcus coagulase negativa (SCN) foi observado 91,9% de sensibilidade ao clorexidine, 70,3% sensíveis ao cloro, 66,2% ao iodo e 24,3% sensíveis ao composto de amônio no tempo de 60”. Conclui-se com esse estudo que a maior atividade desinfetante in vitro foi verificada para clorexidine e cloro frente aos S. aureus, iodo e cloro para os SCP e clorexidine e cloro para os SCN. Devido às variações no perfil de sensibilidade e resistência encontradas, é necessária a avaliação regular da eficiência dos desinfetantes usados nas propriedades, com o intuito de observar a eficácia do produto e assim garantir o controle da mastite no rebanho.

E-mail: jrwilton@uag.ufrpe.br The objective of the study was to characterize the epidemiological situation of Toxoplasma gondii infection in equids from Brejo Paraibano microregion, Northeastern Brazil. Antibodies against T. gondii were investigated in samples of 257 equids (204 horses, 46 mules and seven donkeys) in 26 properties. For serological diagnosis was used the Indirect Fluorescent Antibody Test (IFAT) and a cut-off of 1:64. The number of foci was found to be 46.1%. In the samples analyzed, the overall prevalence was 7.8% (C.I. 4.8-8.8). The prevalence was 8.3% (C.I. 4.9-13.0) for horses, 2.2% (C.I. 0.1-11.5) for mules and 28.6% (C.I. 3.7-71.0) among donkeys. Logistic regression of the variables showed that the water source was a risk factor, because in those properties that supplied running water to the animals the risk of infection was 4.4 times higher than in those properties which provided standing water (OR 4.4; C.I. 1.0-19.0). This is the first report of the presence of antibodies against T. gondii in equids in this micro-region of Paraiba State. To reduce the risk of infection in these species, good quality water should be given to the animals, as well as access of cats to water sources and facilities where animals are kept must be avoided.

Nas amostras analisadas, a prevalência geral foi de 7,8% (I.C. 4,8-8,8). A prevalência foi de 8,3% (I.C. 4,9-13,0) para os equinos, 2,2% (I.C. 0,1-11,5) para os muares e 28,6% (I.C. 3,7-71,0) entre os asininos. Na regressão logística das variáveis observou-se que a fonte de água foi um fator de risco, pois naquelas propriedades que forneciam água corrente para os animais o risco de infecção foi 4,4 vezes maior do que naquelas propriedades que forneciam água parada (OR 4,4; I.C. 1,0-19,0). Este é o primeiro relato da presença de anticorpos contra T. gondii em equídeos nessa microrregião do estado da Paraíba. Para diminuir os riscos de infecção nestas espécies, deve-se fornecer aos animais uma água de boa qualidade, bem como evitar acesso de gatos a fontes de água e instalações onde os animais são mantidos.

E-mail: jrwilton@uag.ufrpe.br The objective of this study was to analyze the risk factors for bovine mastitis in the micro-region of Garanhuns, Pernambuco, Brazil. For this, was submitted for microbiologic exam 1260 milk samples and on each farm was applied a questionnaire about characteristics of animal breeds, general data from the farm, handling of cattle and sanitary measures for milking. To analyze the risk factors, a logistic regression was made as a dependent variable of the microbiologic exam. The microbiologic exams showed 477 (37.9%) positive samples and 783 (62.1%) negatives ones. The risk factors for bovine mastitis were: the semi-intensive breeding system (OR=2.3, p<0.0001), feeding during milking (OR=1.3, p=0.0101), antimicrobial shift (OR=1.3, p=0.00770), and dry cow therapy (OR=1.5; p=0.0013). The results show that the main risk factors are associated with handling to which the cattle is submitted. The knowledge of these factors allow to adopt prevention and control measures to avoid mastitis to improve animal productivity and gain within the production system.

Ao exame microbiológico observou-se 477 (37,9%) amostras positivas e 783 (62,1%) negativas. Os fatores de risco associados à mastite foram: sistema de criação semi-intensivo (OR=2,3; p<0,0001), alimentação dos animais durante a ordenha (OR=1,3; p=0,0101), rodízio de antimicrobianos (OR=1,3; p=0,0077) e o uso da terapia da vaca seca (OR=1,5; p=0,0013). Os resultados obtidos demonstram que os principais fatores de risco associados à mastite estão relacionados ao manejo higiênico-sanitário ao qual o rebanho é submetido. O conhecimento desses fatores fornece subsídios para a elaboração de medidas de prevenção e controle para a enfermidade em questão, melhorando a produtividade dos animais e, consequentemente, a lucratividade do sistema de produção.

E-mail: bayard@biorep.ufsm.br Artificial insemination is routinely used in the swine industry to reduce the costs of production through to increase the efficiency of the refrigerated boar semen process. The objective of this study was to evaluate the effect of different levels of cysteine (CYS) added to the Beltsville Thawing Solution (BTS) extender semen during cooling for up to 72 hours. Ejaculated from three boars were collected with the gloved-hand technique and semen aliquots were diluted in BTS as follow: BTS only (BTS), BTS + 0.1mM cysteine (CYS0.1), BTS + 0.5mM cysteine (CYS0.5), BTS + 1.0mM cysteine (CYS1.0), BTS + 2.5mM cysteine (CYS2.5), BTS + 5.0mM cysteine (CYS5.0), BTS + 10.0mM cysteine (CYS10.0), and BTS + 20.0mM cysteine (CYS20.0). Evaluation of sperm integrity were analyzed using 0.5mg/ml propidium iodide (plasma membrane), 100µg/ml isothiocynate-conjugated Pisum sativun agglutinin (acrosomal membrane) and 153µM 5,5’,6,6’-tetrachloro-1,1’,3,3’-tetraethylbenzimidazolyl carbocyanine iodide (mitochondria potential) after semen dilution at specific times (0, 24, 48 and 72 hours). Additionally, we also evaluated the effects of 5.0 mM CYS addition in the BTS extender on the maintenance of sperm quality and their influence on fertility in the swine production. After artificial insemination, animals were evaluated based on the estrous return and the number of piglet’s born. Cysteine at concentrations of 10.0 and 20.0mM resulted in more pronounced reductions even at the time zero. Semen viability decreased to levels below 10% at these high levels of CYS in the first 24 hour of storage at 17°C. At the end of the storage time, less than 65% of sperm cells had intact plasma membrane in all groups. The sperm viability decreased significantly when the semen was added at high concentrations of CYS (time “0”; CYS10.0 and CYS20.0; p<0.05), when compared to the other CYS concentrations. The BTS (10.20±0.39) treated group showed a lower rate of estrus return when compared to other (BTSCYS; 86.05±039), and it showed also the highest total number of piglets borne per treatment (12.71±3.38 vs. 9.00±3.38, respectively). In conclusion, the addition of CYS in the BTS semen extender did not maintain spermatic viability of boar cooled spermatozoa and it results in a higher percentage of return to estrus and lower number of piglets borne.

O objetivo deste trabalho foi avaliar o efeito da adição de diferentes concentrações de cisteína (CIS) ao diluidor de sêmen Beltsville Thawing Solution (BTS) resfriado sobre a qualidade espermática por até 72 horas. Foram coletados ejaculados de três cachaços e as amostras de sêmen foram diluídas em BTS, conforme os seguintes tratamentos: BTS (grupo controle); CIS0,1 (BTS + 0,1mM de cisteína); CIS0,5 (BTS + 0,5mM de cisteína); CIS1,0 (BTS + 1,0mM de cisteína); CIS2,5 (BTS + 2,5mM de cisteína); CIS5,0 (BTS + 5,0mM de cisteína); CIS10,0 (BTS + 10,0mM de cisteína) e CIS20,0 (BTS + 20,0mM de cisteína). A avaliação da integridade espermática foi determinada através de sondas fluorescentes em uma combinação de 100&#956;g/mL FICT-PSA (isotiocinato de lecitina), 0.5mg/ml PI (iodeto de propidio), e 153&#956;M JC-1 (5,5’,6,6’-tetracloro-1,1’,3,3’-tetraetillbenzimidazolil iodeto de carbocianina). As avaliações dos tratamentos foram realizadas 0, 24, 48 e 72 horas após a diluição do sêmen. Adicionalmente, foi avaliado o efeito da adição de 5,0 mM de cisteína ao diluidor BTS na manutenção da qualidade espermática e no efeito na fertilidade em suínos. Após a inseminação artificial, as fêmeas foram avaliadas quanto a taxa de retorno e o tamanho da leitegada. Durante todos os períodos analizados, os grupos CIS10,0 e CIS20,0 apresentaram menor número de espermatozóides viáveis em relação aos demais grupos. A viabilidade espermática diminuiu a níveis abaixo de 10% nos tratamentos CIS10,0 e CIS20,0 nas primeiras 24 horas de armazenamento a 17ºC. Ao final do período de armazenamento todos os grupos apresentavam média inferior a 65% de espermatozóides com a membrana plasmática intacta. A viabilidade espermática diminuiu significativamente quando altas concentrações de CIS (hora “0”; CIS10,0 e CIS20,0; p<0.05) foram adicionadas ao sêmen comparadas com as demais concentrações. O grupo BTS (10,20±0,39) apresentou menor taxa de retorno ao estro comparado com BTSCIS (86,05±0,39), além de apresentar maior número de leitões nascidos (12,71±3,38 vs. 9,00±3,38, respectivamente). Portanto, podemos concluir que a adição de CIS ao diluidor BTS não mantém a qualidade espermática e resulta em maior taxa de retorno ao estro e menor número de leitões nascidos.

E-mail: valerio.portela@gmail.com The objective of this study was to evaluate the effect of medroxy-progesterone acetate (MAP) with or without estradiol benzoate (EB) on follicular growth during the estrous cycle in cattle. In the first experiment, Hereford cows were synchronized with a synthetic analogue of PGF2 alpha and were treated with two different doses of MAP (250 or 500 mg) with or without EB for 7 days starting on day 8 of the estrous cycle. Follicular growth was inhibited (P<0.05) in all cows except controls and those receiving 250mg MAP without EB. Seventy-five percent of the animals (15/20) showed estrus on days 21 and 22 of the cycle rather than at MAP withdrawal, demonstrating that these treatments did not induce estrus. To determine whether the EB treatment altered endometrial sensitivity to oxytocin and thus the luteolytic cascade, multiparous pre-synchronized cows received 5 mg of EB followed 6 hours later with 50 IU of oxytocin (OT; n=9). Eight hours after EB injection, endometrial fragments were collected from the cows on days 4, 13 and 17 of the estrous cycle and COX-2 gene expression was measured by PCR. EB increased COX-2 mRNA levels only on day 17 of the estrous cycle (P<0.05). In conclusion, MAP alone or associated with EB is able to suppress bovine follicular growth. However, EB in the presence of MAP is not efficient to induce luteolysis in cows when injected on day 8 of the estrous cycle.

No primeiro experimento, vacas da raça Hereford foram sincronizadas com um análogo sintético de PGF2á e tratadas com duas doses diferentes de MAP (250 ou 500mg), com ou sem EB, durante 7 dias, iniciando-se no oitavo dia do ciclo estral. Observou-se uma inibição do crescimento folicular (P<0,05) em todas as vacas, exceto no grupo controle e no grupo que recebeu 250mg de MAP sem BE. Os 75% dos animais não exibiu estro no momento da remoção do MAP, mas sim nos dias 21 e 22 do ciclo, demonstrando que os tratamentos não induziram cio. Para se determinar se o tratamento com BE alterou a sensibilidade endometrial à ocitocina e, assim, a cascata luteolítica, vacas multíparas pré-sincronizadas receberam 5mg de BE, seguidos, após 6 horas, de 50 UI de ocitocina (OT; n=9). Oito horas após a administração de BE, colheram-se fragmentos endometriais das vacas, nos dias 4, 13 e 17 do ciclo estral, mensurando-se a expressão gênica de COX-2 através de PCR. O BE aumentou os níveis de RNAm de COX-2 apenas no dia 17 do ciclo estral (P<0,05). Em conclusão, o MAP isolado ou associado a BE é capaz de suprimir o crescimento folicular bovino. Entretanto, o BE, na presença de MAP é ineficaz na indução da luteólise bovina, quando injetado no oitavo dia do ciclo estral.

E-mail: carlosanramos@yahoo.com.br Bovine anaplasmosis is a major disease in tropical and subtropical regions of the world by determine economical loss due mortality and productive reduction. The disease is caused by Anaplasma marginale, an intraerythrocytic rickettsia whose control requires, besides an efficient vaccine, the accurate identification of chronically infected cattle. Although the existence of diverse methods of diagnosis of this rickettsia, the serological methods, in particular the enzyme immunosorbent assays (ELISAs), are the most used due to its versatility and practice. However, due to the high number of antigens currently available, an evaluation becomes necessary to define which antigens present the better performance in the diagnosis of anaplasmosis. Sera from cattle positive or negative to A. marginale by PCR, and sera from cattle proceeding from Brazil and Costa Rica, were tested by ELISAs based in recombinant MSP1a, MSP2, and MSP5, a pool of the three recombinant proteins, and initial body lisate antigen (CI). Using sera from A. marginale positive cattle by PCR, the highest sensitivity was shown by CI ELISA. Nevertheless, the highest specificity, with sera from negative cattle by PCR, was shown by recombinants ELISAs. The percentiles of positive cattle from Brazil and Costa Rica were higher with CI ELISA. Reasons for such differences were discussed.

Soros de bovinos positivos e negativos para A. marginale por PCR, e soros de animais provenientes do Brasil e Costa Rica, foram testados em ELISAs baseados em MSP1a, MSP2 e MSP5 recombinantes, um pool das três proteínas recombinantes, e antígeno de lisado de corpúsculos iniciais da riquétsia (CI). Utilizando soro de bovinos positivos para A. marginale por PCR, uma maior sensibilidade foi observada no ELISA CI. No entanto, uma maior especificidade, com soro de bovinos negativos a PCR, foi observada com os ELISAs recombinantes. O porcentual de bovinos positivos do Brasil e Costa Rica foi maior com ELISA CI. Razões para essas diferenças são discutidas.

The sample comprised 178 herds from 22 counties in the state of Mato Grosso do Sul, Brazil. The risk factors associated with the presence of infeccion were investigated. A total of 2,573 blood serum samples were tested against 10 leptospira serovars using the microagglutination test (MAT). Titers of 100 or higher for one or more serovars were detected in 1,801 females (98.8%) from 161 herds (96.5%). Serovar Hardjo (65.6%) was the most frequent, followed by serovar Wolffi (12.3%). These results suggest that bovine leptospirosis is widespread in all the counties under study, with a high prevalence both at the animal and the herd level. Beef farms and the Zebu breed were associated to the higher risk of herd infection by leptospiras.

This oil is used for the treatment of skin wound due to its recognized antiinflammatory and wound healing effects. Despite, its popular use, there are few published data about the therapeutic effect of this medicinal plant. The aim of the study was to evaluate the topic treatment effect of the Copaíba oil on the process of skin repair inflammation induced by a foreign body subcutanously implanted. Sixty BALB/c mice were submitted to a 1cm linear incision and a 12mm circle coverslip was subcutaneously implanted. Four treatments groups were established: control, sterile saline (C); vehicle control, sterile mineral oil, (VC); treatment 1 (T1), mineral oil plus copaiba oil (V/V), and treatment 2 (T2) copaiba oil. The evaluations were performed at pre-determined time points (1, 3, 5, 7 and 14 days). It was possible to find fibroblasts, epithelial cells proliferation, re-epithelization and newly formed blood vessels in all groups, however, all oil treated groups (T1 and T2) did not present re-epithelization at three days post surgical incision. On days 5 and 7, a higher intensity of edema and hyperemia on the groups T1 and T2 was observed, besides that, the T1 and T2 groups presented a serous cellular scab on the wounds that was absent on the C and VC groups. The inflammatory reactions among the groups C and VC showed more mononuclear cells than the T1 and T2 groups that presented a mixed cell patter composed from both mono and polymorphonuclear cells. Although the surgical wounds were re-epithelizaded, in the groups T1 and T2, they were covered by a serous cellular crust and the dermis tissue still presented an intense mononuclear cell inflammatory focus. Fourteen days after of the surgical incision, the gross aspects on groups C and VC were similar and on groups T1 and T2, despite wound to be completely closed and without crusts, the skin those animals was thickened. Furthermore, the dermis on group T2 presented moderate fibrosis, while the other groups presented slightly ones. The results demonstrated that topical treatment with C. langsdorffii oil debilitated the normal process of a wound repair in the presence of a foreign body.

Although it was not clearly elucidated whether APV may cause economical losses in broiler flocks, it is known that APV infection can induce specific antibody production on these birds, and these serological reactions may provide some information about the epidemiological status of the APV infections. This work was carried out in search for antibodies to APV in broiler flocks in counties of Mato Grosso do Sul. Five hundred and thirty six serum samples from 54 broiler flocks at 42 and 51 days of age were tested with a commercially available enzyme-linked immunosorbent assay (ELISA). The results showed that 330 samples (61.6%) were negative, 108 (20.1%) were suspect and 98 (18.3%) were considered positive for the presence to APV antibodies. Of all the flocks analyzed, 49 (90.7%) were considered either positive or suspect. The ELISA test demonstrated that there was a similar percentage of positive or suspect flocks among those flocks between 42 and 46 days of age, and among those between 47 and 51 days. No seasonal differences were observed, since the percentages of positive or suspect flocks either in summer or in winter months were similar. Most of the flocks were considered positive despite the type of broiler housing (conventional, environmental controlled or semi-controlled). It is concluded that there are strong evidences indicating circulation of APV in Mato Grosso do Sul. The percentages of positive flocks were similar regardless of the age groups of the birds examined, the type of broiler housing and the season when sampling was performed.
