E-mail: arianneoria@ufba.br The purpose of this study was to establish reference values for selected ophthalmic diagnostic tests in New Zealand rabbits (Oryctolagus cuniculus). A total of 22 adult male rabbits were used. The ophthalmic tests included evaluation of tear production with Schirmer tear test 1(STT1) and Endodontic absorbent paper point tear test (EAPPTT) using two different commercial brand materials. Applanation tonometry, Culture of the conjunctival bacterial flora, , conjunctival cytology and conjunctival histology were also performed. Mean (±SD) for STT1, EAPPTTa, EAPPTTb and IOP was 7.27±2.51mm/min, 12.43±1.69mm/min, 15.24±2.07mm/min, 12.89±2.80mm Hg, respectively. Staphylococcus epidermidis, Staphylococcus sp. and Bacillus sp. were predominant. The cytological evaluation revealed the presence columnar epithelial cells, superficial squamous keratinized cells, lymphocytes, heterophils, red blood cells, mucus and bacteria. The histological analysis revealed a stratified epithelium, characterized by the presence of columnar epithelial cells with a large number of goblet cells. The reported data can be used for therapeutic or experimental purposes.

E-mail: arianneoria@ufba.br O objetivo deste estudo foi estabelecer valores de referência para testes diagnósticos oftálmicos em coelhos da raça Nova Zelândia (Oryctolagus cuniculus). 22 coelhos, machos, adultos foram utilizados. Foi mensurada a produção lacrimal através do teste lacrimal de Shirmer 1 (TLS1) e da Tira endodôntica de papel absorvente (EAPPTT) de duas marcas comerciais distintas. Tonometria de aplanação, identificação da microbiota conjuntival, , citologia e histologia conjuntival também foram realizadas. A média e desvio padrão do TLS1, EAPPTT1, EAPPTT2 e pressão intraocular foi 7,27±2,51 mm/min, 12,43±1,69 mm/min, 15,24±2,07 mm/min e 12,89±2,80 mmHg, respectivamente. Staphylococcus epidermidis, Staphylococcus sp. e Bacillus sp. mostraram-se predominantes. A citologia conjuntival evidenciou a presença de células epiteliais colunares, células escamosas superficiais queratinizadas, linfócitos, heterofilos, células sanguíneas, muco e bactérias. A histologia revelou epitélio estratificado caracterizado pela presença de células epiteliais colunares com grande número de células caliciformes. Os achados deste estudo poderão ser utilizados com fins terapêuticos ou experimentais.

E-mail: cnunesc@hotmail.com The aim of the study was to evaluate the efficacy of oral ivermectin in the control of Psoroptes ovis and Leporacarus gibbus in naturally infested rabbits. Twenty adult rabbits were selected; they were distributed in two groups with equal proportions of both sexes, containing ten animals per group. In the control group the same volume of the treatment was administered of saline solution, meanwhile the treated group received a single dose of oral ivermectin (400µg/kg). The diagnosis of the mites was made with a stereoscopic microscope, after the proper collection of material. For P. ovis it was performed by collecting of ear wax with swabs and for L. gibbus it was performed by collecting hairs in the dorsal part of the neck, lumbar right, lumbar left, ventral side of the tail and ventral abdomen. The evaluation of the efficiency and the clinical evaluation of the lesions, measured in scores (grade 0 to 4) was made in days 0, +3, +7, +4, +21, +28, and +35, after treatment. An efficiency of 100% was observed for P. ovis following day +7, and for L. gibbus following day +14, remaining negative until the final day of the study. The score of lesions in the ears of the treated group regressed following day +14 and on day +21 all animals reached a score of 0. In the control group, two animals presented an increase in ear lesion score, one rabbit presented an increase from score 1 to 2, and the other rabbit, from score 3 to 4. No adverse reactions were observed in the treated animals. The single dose administration of oral ivermectin was successful in the control of P. ovis and L. gibbus in naturally infested rabbits.

No grupo controle foi administrado o mesmo volume do tratamento de solução salina, enquanto o grupo tratado recebeu dose única de ivermectina oral (400 µg/Kg). O diagnóstico dos ácaros foi realizado com auxílio de microscópio estereoscópico após a devida coleta de material. Para P. ovis foi realizada através de coleta do cerúmen com auxílio de zaragatoas efetuadas nas orelhas e para por L. gibbus foi realizada coleta de pelos nas regiões do pescoço dorsal, lombar direita, lombar esquerda, cauda ventral e abdômen ventral. A avaliação da eficácia e a avaliação clínica das lesões, mensuradas em escores (grau 0 a 4) foi realizada nos dias 0, +3, +7, +14, +21, +28 e + 35, após o tratamento. Foi observada a eficácia de 100% no controle de P. ovis a partir do dia +7 e para L. gibbus a partir do dia +14, mantendo-se negativos até o final do período experimental. O escore da lesão das orelhas do grupo tratado regrediu a partir do dia +14 e no dia +21 todos os animais atingiram grau 0. No grupo controle, dois animais apresentaram aumento no escore da lesão das orelhas, um coelho apresentou aumento do escore de grau 1 para 2 e outro coelho de grau 3 para 4. Não foram observadas quaisquer reações adversas nos animais tratados. A ivermectina administrada por via oral em dose única foi eficaz no controle de P. ovis e L. gibbus em coelhos naturalmente infestados.

E-mail: shfreitas@terra.com.br A segmental defect of 6mm diameter was performed in the medial metaphyseal region of the tibia of 12 rabbits. For the bone defect reconstruction there was implanted a combination of micro and macro fragments of heterologous fragmented cortical bone matrix preserved in glycerin (98%) and polymethylmethacrylate, both autoclaved. Radiological and macroscopic evaluation was performed at 30, 60, 90 and 120 days. Adhesion in relation to time of the micro and macro composites to the recipient bed was observed in 100% of the cases. This showed that this implant is biologically biocompatible, since it promoted bone defects repair, with no signs of infection, migration and/or rejection. In this way, this implant can be classified as one more option of substitute to fill large bone defects.

E-mail: fernandomilori@yahoo.com.br Actually, technological advances have contributed to the development and improvement of new materials for implants. Despite the fundamental benefits provided by orthopedic materials, complications resulting from corrosion, degradation, infection, and others may occur. The knowledge of characteristics of the biomaterials is important to predict their behavior in vivo, supporting that the most suitable compound is chosen to reconstruct the bone defect. Metallic plates are the most common material used for repair of long bone fractures, and they are mechanically stable and biocompatible. However, the need of removal and bone weakening are their disadvantages. In this paper, plates made from equine cortical bone were used experimentally in osteotomized rabbit femur (bone-GO Group), in a comparative study with metal plates (metal GM Group). Radiographic evaluation was performed every 30 days during 120 days, at which time was then performed femur histological analysis. During all evaluated moments of the study, there was no statistically significant difference between the two groups in callus morphometry, and complete consolidation was observed in all animals. However, it was observed that the callus was lower in GO compared to GM, all times during the study. Moreover, most of the GO animals showed complete consolidation fracture at 90 days only, whereas this occurred in GM at 60 days. There were no foreign-body cells in the histopathology exam of the GO animals, but larger amount of fibrous tissue was identified, involving this biomaterial. The plate made from horse bone represents a low cost alternative and it is very feasible, it allowed adequate stabilization of femur fracture in rabbits. In this study, the lower periosteal callus associated with a longer time for consolidation in GO suggest greater fracture stability when the bone plate was used.

Apesar dos fundamentais benefícios providos pelos dispositivos ortopédicos, complicações decorrentes de corrosão, degradação, infecção, além de outras podem ocorrer. O entendimento das características dos biomateriais é fundamental para a previsibilidade do seu comportamento in vivo, fornecendo subsídios para que o composto mais adequado seja escolhido na reconstrução do defeito ósseo. As placas de origem metálica são as mais utilizadas para o reparo de fraturas de ossos longos, sendo mecanicamente resistentes e biocompatíveis. No entanto, a necessidade de remoção e o enfraquecimento do osso são suas principais desvantagens. Neste trabalho, placas produzidas a partir de osso cortical equino foram empregadas experimentalmente em fêmur osteotomizado de coelhos (Grupo osso-GO), num estudo comparativo com placas de metal (Grupo metal-GM). A avaliação radiográfica foi realizada a cada 30 dias, durante 120 dias, momento em que foi então realizada análise histológica do material em estudo. Não houve diferença estatisticamente significante entre os dois grupos com relação à morfometria do calo ósseo e consolidação óssea em todos os momentos avaliados, sendo que ambas as placas permitiram a consolidação em todos os animais. Entretanto, observou-se que o calo ósseo foi menor no GO, em relação ao GM, em todos os momentos do estudo. Por outro lado, a maior parte dos animais do GO apresentou consolidação completa da fratura aos 90 dias, enquanto que no GM isto ocorreu aos 60 dias. Não foram evidenciadas células do tipo corpo estranho na histopatologia dos animais do GO, mas maior quantidade de tecido fibroso foi identificada, envolvendo este biomaterial. A placa confeccionada com osso equino representa uma alternativa de baixo custo e muito viável, uma vez que permitiu estabilização adequada para consolidação óssea de fratura de fêmur em coelhos. Neste estudo, a menor formação de calo periosteal, associada a um tempo superior para consolidação em GO sugerem maior estabilidade da fratura onde a placa de osso foi utilizada.

A 6mm of segmental defect was performed at the medial metaphyseal region of the left tibia of 12 rabbits and an association of fragmented heterologue mineralized bone matrix and methylmethacrylate conserved in glycerin (98%) was used as a graft to fill the bone defect. To evaluate the procedure morphological and radiological exams were performed after 30, 60, 90 and 120 days. There was gradual integration of the bone graft in the receptor bed in 100% of the cases showing that the material is biologically compatible as it promotes bone defect reparation without signs of infection, migration and/or rejection and can be considered one more option to be used as a substitute to fill bone defects.

E-mail: vetjulio@yahoo.com.br The aim of the present study was to evaluate the efficacy of the d-phenothrin/pyriproxyfen association against Psoroptes ovis, Cheyletiella parasitivorax, and Leporacarus gibbus infestations in naturally co-infested rabbits. Twenty crossbreed (New Zealand White x California) rabbits concurrently infested by the three mite species were randomly divided in two groups. All rabbits presented with hyperemia, erythema and formation of crusts in the ear canals caused by P. ovis. Infestations by both C. parasitivorax and L. gibbus were considered asymptomatic in all animals.Ten animals were treated with a 4.4% d-phenothrin and 0.148% pyriproxyfen spray formulation until have their body surface uniformly sprayed, including external ear canals. The other ten rabbits remained untreated, serving as control group. Observations were done on days +7, +14, +21, +28, and +35 post-treatment. The d-phenothrin/pyriproxyfen association showed 100% efficacy against the three mite species and was responsible for the remission of psoroptic mange lesions on treated animals. No signs of intoxication were observed. The results indicate that d-phenothrin/pyriproxyfen spray formulation in a single application is an effective and clinically safe option for the control of different mite infestations in rabbits.

Todos os coelhos infestados por apresentavam eritema, hiperemia e formação de crostas nas orelhas, causados por P. ovis. Infestações simultâneas por C. parasitivorax e L. gibbus foram considerados assintomáticas em todos os animais. Dez animais foram tratados com uma formulação spray contendo d-fenotrina a 4,4% e piriproxifen a 0,148%, pulverizando toda a superfície corporal de forma uniforme, incluindo a face interna das orelhas. Os outros 10 coelhos não foram t ratados, sendo mantidos como grupo controle. Os animais foram avaliados nos dias 7, 14, 21, 28 e 35 pós-tratamento. A associação de d-fenotrina e piriproxifen foi 100% eficaz no controle das três espécies de ácaros e foi responsável pela remissão das lesões de sarna psoróptica nos animais tratados. Não foram observados sinais de intoxicação. Os resultados indicam que a formulação spray de d-fenotrina e piriproxifen em uma única aplicação é uma opção clinicamente segura e eficaz no controle de infestações por ácaros em coelhos.

E-mail: wilmavet@gmail.com The aim of this study was to evaluate the progression of lesions in different stages of osteoarthritis (OA) experimental by radiography (RX), computed tomography (CT), macroscopic and histopathology, linking these different diagnostic methods, helped to provide information that helps the best time for the therapeutic approach. Four experimental periods were delineated at 3, 6, 9 and 12 weeks after induction of OA, known as PI, PII, PIII and PIV, respectively, each with six animals. We evaluated the five compartments of the femorotibial joint: medial femoral condyle (MFC), lateral femoral condyle (LFC), medial tibial plateau (MTP), lateral tibial plateau (LTP) and femoral trochlea (FT). Therefore we established an index by compartment (IC) and by adding such an index was estimated joint femorotibial (IFT). It was observed that the CFM was the compartment with the highest IC also differed significantly (p<0.05) from other compartments. Compartments showed no significant difference (p>0.05) between the PI and PII, however contrary fact occurred between the PII and PIII (p<0.05), PIII and PIV (p<0.01) and between PI and PIV (p<0.001). Similarly the IFT, showed a significant difference in the animals of PIV compared to PI (p<0.001), PII (p<0.001) and PIII (p<0.01), and there was no statistical difference (p> 0.05) between the PI and PII. In the variation of the average interval between periods, there was a higher value between the PIII PIV and for the other intervals of time periods (PI, PII, and PIII-PII). However, these intervals showed no statistically significant difference (p>0.05). Through the RX, CT, macroscopic and histopathological findings, we found similar patterns among individuals within the same period demonstrating a gradual progression of the disease. These results show that between 3 and 6 weeks progression of the lesion is slower and probably also can be reversed in comparison to other ranges where proved further progression between 9 and 12 weeks after induction of trauma OA. These results may provide a better therapeutic approach aimed at reversing the lesions in early stages of OA. We conclude that the interconnection of the four diagnostic methods individually classified into scores, which were unified in both indices in the evaluation by the femorotibial joint compartment and may represent a diagnostic condition closer to the true condition of the injury and its progression.

E-mail: wilmavet@gmail.com O objetivo deste estudo foi avaliar a progressão das lesões em diferentes períodos da osteoartrite (OA) experimental em coelhos, através da radiografia (RX), tomografia computadorizada (TC), macroscopia e histopatologia, interligar estes diferentes métodos diagnósticos, bem como trazer informações que ajudem na decisão do melhor momento para a abordagem terapêutica. Foram delineados quatro períodos experimentais às 3, 6, 9 e 12 semanas após a indução da OA, denominados como PI, PII, PIII e PIV, respectivamente, cada qual com seis animais. Foram avaliados os cinco compartimentos da articulação femorotibial: côndilo femoral medial (CFM), côndilo femoral lateral (CFL), platô tibial medial (PTM), platô tibial lateral (PTL) e tróclea femoral (TF). Por conseguinte, estabeleceu-se um índice por compartimento (IC) e através da soma destes foi obtido um índice da articulação femorotibial (IFT). Observou-se que, o CFM foi o compartimento com maior valor de IC, além disso, diferiu significativamente (p<0,05) dos demais compartimentos. Os compartimentos não apresentaram diferença significativa (p>0,05) entre o PI e o PII, no entanto fato contrário ocorreu entre o PII e o PIII (p<0,05), PIII e PIV (p<0,01) e entre o PI e PIV (p<0,001). Similarmente o IFT, apresentou diferença significativa nos animais do PIV em relação ao PI (p<0,001), ao PII (p<0,001) e ao PIII (p<0,01), e não houve diferença estatística (p>0,05) entre o PI e PII. Na variação média dos intervalos entre os períodos, observou-se um maior valor entre o PIII e o PIV em relação aos outros intervalos de períodos (PI-PII e PII-PIII). No entanto, estes intervalos não apresentaram diferença estatisticamente significativa (p>0.05). Através do RX, TC, macroscopia e histopatológico, verificamos padrões similares entre os indivíduos dentro do mesmo período demonstrando assim uma gradual progressão da doença. Tais resultados evidenciam que entre as 3 e 6 semanas a progressão da lesão é mais lenta e muito provavelmente ainda pode ser reversível em comparação aos outros intervalos onde se comprovou uma maior progressão entre as 9 e 12 semanas após a indução traumática da OA. Estes resultados podem propiciar uma melhor abordagem terapêutica objetivando a reversão das lesões em fases iniciais da OA. Concluímos que, a interligação dos quatro métodos diagnósticos, individualmente classificados em escores e que foram unificados em índices tanto na avaliação por compartimento quanto pela articulação femorotibial pode representar uma condição diagnóstica mais próxima à verdadeira condição da lesão quanto de sua progressão.

E-mail: vetjulio@yahoo.com.br The objective of the present study was to evaluate the efficacy of an emulsion containing 10% of neem (Azadirachta indica) oil on the control of Psoroptes ovis in naturally infested rabbits. Twelve rabbits were randomly divided in two groups of six animals. The control group remained without treatment; while animals in the other group were treated with the 10% neem extract formulation by spraying both ears daily for seven consecutive days. The animals were evaluated daily for the presence of adverse effects. Material from ears all animals was collected on days +3, +7, +14, +21, +28 and +35, and evaluated for the presence of living mites. Animals from control group presented mites in both ears along all days of observation. The treated group presented an efficacy of 41.7% on day +3 and 100% from day +7 to +35. The product containing 10% neem oil has demonstrated to be effective for the treatment of psoroptic mange on rabbits. However, treated animals presented dermatological reaction such as alopecia and hyperemia at the site of application, varying from low to medium severity.

Os coelhos do grupo controle apresentavam ácaros em ambos os condutos auditivos em todos os dias de observação. O grupo tratado apresentou eficácia de 41,7% no dia +3 e 100% a partir do dia +7 até o dia +35. O produto demonstrou ser eficaz no tratamento da sarna psoróptica em coelhos. Entretanto, todos os animais tratados apresentaram reações dermatológicas, tais como alopecia e hiperemia no local de aplicação do produto, variando de baixa a média severidade.


#29 - Experimental poisoning of rabbits by Metternichia princeps (Solanaceae), 32(9):872-880

E-mail: marilene@ufrrj.br Metternichia princeps, a tree that belongs to the Solanaceae family, occurs in the Atlantic Forest of Brazil, from the state of Rio de Janeiro to Bahia. In the county of Itaguaí, state of Rio de Janeiro, M. princeps was suspected to cause a lethal kidney disease in goats. The poisoning was experimentally reproduced in goats confirming the suspicion. Experiments were then performed in rabbits. Leaves of M. princeps were collected in the county of Itaguaí, where the poisoning in goats occurred. Eleven rabbits received the dried plant material, ten received the fresh plant and one rabbit served as control. The dehydrated leaves were ground and mixed with water. Initially the suspension was administered by stomach tube to nine rabbits and caused death of seven of them at doses from 0.125g/kg on. Two rabbits that got sick, but survived at doses of 0.0625g/kg and 0.125g/kg, received doses of 0.5 and 0.25g/kg respectively, two months later and died, indicating that no tolerance to the plant poisoning effects developed. Additionally the dried leaves administered six months after collection to two rabbits at doses of 0.5 and 1g/kg respectively, caused death only of the rabbit that received the dose of 1g/kg, indicating that the stored leaves lost toxicity. Fresh sprouts caused death in three of six rabbits at doses of 1.55g/kg to 3g/kg. Fresh mature leaves caused death in one of four rabbits at the dose of 2g/kg, what suggests that there is no difference in toxicity between mature leaves and sprouts. The dried leaves caused death of rabbits at smaller doses than fresh leaves, what can be explained by the way of administration of the plant; dried leaves were given within a few minutes, whilst fresh leaves were ingested during one or two days, a fact that may influence absorption of the toxic principle. In all experiments with the dried and fresh leaves (sprouts and mature leaves) of M. princeps, liver and heart lesions predominated. The course of the poisoning was peracute to acute. The rabbits became apathetic with pale and cyanotic mucosae and cold ears; they fell down with struggling movements, vocalization and spaced breathing and died. At postmortem examination, the liver was pale with marked hepatic lobules; the other organs were congested and blood vessels were ingurgitated. The histopathological examination revealed as the more important changes in the liver, intense, especially centrilobular congestion, severe swelling of the hepatocytes, and foci of incipient necrosis of hepatocytes. In the heart, beside congestion, small groups of eosiophilic cardiac fibers, with nuclear picnosis and vacuolation were found. In the kidney, congestion and swelling of epithelial cells was seen. The performed experiments show that rabbits are susceptible to poisoning with dried and fresh leaves of M. princeps. It is suggested that the toxic principle of M. princeps, responsible for the clinical and pathological picture in rabbits, may not be the same compound that causes poisoning in goats or that the rabbit reacts differently as react goats; rabbits showed liver and heart lesions whilst goats had kidney lesions.

É conhecida popularmente com os nomes de “café-do-mato”, “trombeteira” e “jasmin-do-morro”. Em um estabelecimento no município de Itaguaí, RJ, suspeitou-se que M. princeps era responsável por doença renal letal em caprinos. A intoxicação foi reproduzida experimentalmente nessa espécie, confirmando a suspeita. Foram então realizados experimentos em coelhos. Folhas de M. princeps foram coletadas no município de Itaguaí/RJ, local onde foi diagnosticada a intoxicação em caprinos. Onze coelhos receberam a planta dessecada, dez receberam a planta fresca e um coelho serviu como controle. As folhas dessecadas de M. princeps foram trituradas e misturadas com água. Inicialmente, a suspensão foi administrada por via intragástrica através de sonda a nove coelhos e causou a morte de sete coelhos nas doses a partir de 0,125g/kg. Dois coelhos que adoeceram mas sobreviveram às doses de 0,0625 e de 0,125g/kg, dois meses mais tarde receberam doses de 0,5 e 0,25g/kg, respectivamente, e morreram, o que indica que não houve desenvolvimento de tolerância à toxidez da planta. Adicionalmente, a planta dessecada, administrada a dois coelhos nas doses de 0,5 e 1g/kg seis meses após a coleta, causou a morte somente do coelho que recebeu a dose de 1g/kg, indicando que a planta armazenada perde em toxidez. Os brotos frescos causaram a morte de três dos seis coelhos, nas doses de 1,55g/kg a 3g/kg. As folhas maduras frescas causaram a morte de um dos quatro coelhos, na dose de 2g/kg, o que sugere que não há diferença na toxidez entre a brotação e as folhas maduras. As folhas dessecadas causaram a morte dos coelhos em doses menores que as folhas frescas, o que talvez possa ser explicado pelo modo de administração da planta; as folhas dessecadas eram administradas em poucos minutos, enquanto que as folhas frescas eram ingeridas durante um a dois dias, o que influenciaria a rapidez da absorção do princípio tóxico da planta. Tanto nos experimentos com as folhas dessecadas quanto com as folhas frescas (brotos e folhas maduras), predominaram no quadro clínico-patológico alterações relativas ao coração e ao fígado. A evolução da intoxicação foi superaguda a aguda. Os coelhos tornaram-se apáticos, com mucosas pálidas a cianóticas e orelhas frias, até que subitamente se debatiam na gaiola, faziam movimentos de pedalagem, vocalizavam, apresentavam acentuada dispneia e morriam. À necropsia, o fígado apresentava-se com evidenciação da lobulação; os demais órgãos estavam congestos e os vasos sanguíneos ingurgitados. À histopatologia, as lesões mais relevantes foram: no fígado, intensa congestão, especialmente centrolobular, tumefação acentuada dos hepatócitos e focos de necrose incipiente de hepatócitos; no coração, além de congestão, fibras cardíacas com aumento de eosinofilia, com picnose nuclear, fibras cardíacas com vacuolização, e no rim, congestão e tumefação de células epiteliais. Esses experimentos demonstram que o coelho é sensível à intoxicação pelas folhas dessecadas e frescas de M. princeps. Sugere-se que o princípio tóxico responsável pelo quadro clínico-patológico da intoxicação por M. princeps em coelhos não seja o mesmo princípio responsável pela intoxicação nos caprinos, ou que os coelhos reagem de maneira diferente dos caprinos; nos coelhos predominam alterações cardíacas e hepáticas enquanto que em caprinos as lesões são renais.

E-mail: michel_abdallavet@yahoo.com.br This study aimed to evaluate the protective effect of acetamid in experimental poisoning by Pseudocalymma elegans in sheep, goats and rabbits, in order to prove indirectly that monofluoroacetate (MF) is responsible for the clinical signs and death of animals that ingested the plant. Experiments were performed to determine for sheep and goats the lethal dose of P. elegans collected in Rio Bonito, RJ, in different seasons, and to adjust the dose of acetamid to be administered. - In the first experiment, two sheep and two goats received 1.0g/kg of fresh P. elegans, and two (one sheep and one goat) were pretreated with 2.0g/kg of acetamid. None of the animals showed clinical signs or died. Possibly, the plant could be less toxic, since it was collected at the end of the rainy season. - In the second experiment, two sheep and two goats received 0.67 and 1.0g/kg of the dried plant, after pretreatment with 2.0 and 3.0g/kg of acetamid, respectively. All animals died, as the administered doses of P. elegans were very high. - In the third experiment, two sheep and two goats received 0.333g/kg of dried P. elegans after previous administration of 2.0g/kg of acetamid; a week later, the protocol above was repeated, but without the antidote. In experiments with rabbits, doses of 0.5 and 1.0g/kg of dried P. elegans were given after administration of 3.0g/kg of acetamid; seven days later, the same protocol was repeated, except the administration of acetamide. This procedure, when acetamid was administered before, prevented the appearance of clinical signs and death of sheep, goats and rabbits. But the animals not treated with acetamid showed symptoms of poisoning and died. Clinically, the sheep and goats had tachycardia, engorged jugular vein, positive venous pulse, lateral recumbence, and muscle tremors. In the “dramatic phase”, the animals fell into lateral position, stretched the limbs, were paddling and died within minutes. The rabbits showed apathy, muscle tremors, vocalization and lateral decumbence minutes before death. At postmortem examination, the sheep and goats had engorged jugular veins and atria, dilated vena cava cranialis and caudalis, as well as pulmonary edema, hepatic congestion and edema of the gallbladder subserosa. In rabbits, the main macroscopic alterations were dilated atria, engorged vena cava cranialis and caudalis, and congested liver and diaphragm vessels. Histopathology revealed, in two sheep and one goat, vacuolar-hydropic degeneration of the distal convoluted kidney tubules, together with caryopicnosis. In the rabbits, the liver showed severe congestion with numerous shock corpuscles. The experimental results show indirectly that MF is to be held responsible for death of the animals that ingested P. elegans; since “acetate donor” compounds, such as acetamid, are capable to reduce the competitive inhibition of MF for the same active site (Coenzyme A) which prevents the formation of fluorocitrate, its active metabolite, formed in the body through the so-called “lethal synthesis”.
