E-mail: mateus.costa@univasf.edu.br The aim of the present study was to evaluate the antimicrobial susceptibility pattern and to perform the phenotypic and genotypic characterization of the Staphylococcus spp. isolates from mastitis cases in cattle (n=30) and buffaloes (n=30). The susceptibility to antimicrobial drugs was performed by disk diffusion test and the presence of efflux pump was evaluated in Mueller Hinton (MH) Agar supplemented with ethidium bromide as well as by detection of msrA gene. Similarly, the PCR technique was done to detect mecA, blaZ and ermA, B e C genes, that were related after with the presence of antimicrobial resistance in disk diffusion test. The formation of biofilm was characterized using Congo Red Agar (CRA), microplate adherence and detection of icaD gene. Staphylococcus spp. isolates shown high antimicrobial susceptibility in disk diffusion test. The multidrug resistance (MDR) rate ranged from 0 and 0,5. In the efflux pump test, 26.7% of Staphylococcus spp. strains were positive in phenotypic method and 6.7% in PCR for msrA gene. The erm, mecA and blaZ genes were detected in 1.7%, 6.7% and 11.7% of Staphylococcus spp. strains respectively. In biofilm production tests, 23.3% of the samples were positive in CRA, 50% in microplate adherence test and 8.3% in icaD gene PCR. The cattle isolates were less sensitive to antimicrobial drugs when compared to the buffaloes ones. The characterization of these isolates is very important to guide a successful antimicrobial therapy. The biofilm presence in the isolates may be associated with other factors besides antimicrobial resistance.

A suscetibilidade foi avaliada pela técnica de disco-difusão e a presença de bomba de efluxo foi avaliada utilizando-se Ágar Mueller Hinton (MH) adicionado de brometo de etídeo e pesquisa do gene msrA. Pela técnica da Reação em Cadeia da Polimerase (PCR) ainda foram identificados os genes mecA, blaZ e ermA, B e C, que posteriormente foram associados com os métodos fenotípicos para a identificação de resistência a antimicrobianos. A caracterização da formação de biofilme foi realizada utilizando-se os métodos Ágar Vermelho Congo (CRA), Aderência em Placa e a identificação do gene icaD. Pelo método de disco-difusão, os Staphylococcus spp. apresentaram alta sensibilidade aos antimicrobianos. O índice de resistência múltipla aos antimicrobianos (IRMA) apresentou variação de 0 a 0,5. Na pesquisa de bomba de efluxo, 26,7% das amostras foram positivas ao método fenotípico e 6,7% ao método genotípico (gene msrA). Os genes erm, mecA e blaZ foram detectados, respectivamente, em 1,7%, 6,7% e 11,7% das amostras de Staphylococcus spp. Na produção de biofilme, 23,3% dos isolados foram considerados positivos no CRA, 50,0% na Aderência em Placas e 8,3% na PCR pela detecção do gene icaD. Observou-se que os isolados obtidos de amostras bovinas apresentaram uma menor sensibilidade aos antimicrobianos no teste de disco-difusão quando comparados com as amostras bubalinas. A caracterização destes isolados é importante para orientar uma antibioticoterapia bem planejada. A presença de biofilme nos isolados pode estar associada a outros fatores que não a resistência às drogas antimicrobianas.


#32 - Susceptibility to disinfectants and antimicrobial resistance profile in Escherichia coli isolates

Abstract in English:

Colibacillosis caused by Escherichia coli is the most important enteric disease in pig production, which may lead to death of the affected animal. The bacterium has a great ability to develop resistance to multiple antibiotics and disinfectants. Thus, investigation addressing mechanisms of resistance and profile of field samples is necessary. E. coli is widely used as a model for studies that explore the intrinsic and extrinsic resistance to multidrugs. In this paper, we attempt to associate the susceptibility profile of 62 isolates of E. coli to three disinfectants and 13 antimicrobials. Also 31 isolates were tested for the presence of efflux mechanism. Of the three disinfectants tested, alkyldimethylbenzylammonium chloride+nonyl phenoxy polyethoxy ethanol was the most effective (100%), followed by glutaraldehyde+alkyldimethylbenzylammonium chloride (95.2%) and alkyldimethylbenzylammonium chloride (88.8%). Among the antimicrobials tested, there was greater resistance to tetracycline (62.2%) and higher sensitivity to florfenicol (88.6%). The high sensitivity of the isolates against disinfectants may be related to the absence of efflux mechanism. The average index of multiple resistance to antimicrobials was 0.52, what demonstrates a profile of multidrug resistant isolates, showing the need for rational use of these drugs in pig production.

A colibacilose, causada por Escherichia coli, é a enfermidade entérica de maior impacto na produção de suínos, podendo levar à morte do animal. Esta bactéria possui grande capacidade de desenvolver resistência a múltiplos antimicrobianos e a desinfetantes. Desta forma, estudos que abordem mecanismos de resistência e perfil de amostras de campo tornam-se necessários. E. coli é amplamente utilizada como modelo de estudos que exploram a resistência intrínseca e extrínseca a multidrogas. Neste trabalho, buscou-se verificar o perfil de sensibilidade de 62 isolados de E. coli de suínos frente a três desinfetantes e a 13 antimicrobianos. Ainda, em 31 destes isolados foi pesquisada a presença de mecanismo de efluxo. Dos três desinfetantes avaliados, o cloreto de alquil dimetil benzil amônio+poliexietilenonilfenileter foi o que se mostrou mais eficaz (100%), seguido do glutaraldeído+cloreto de alquil dimetil benzil amônio (95,2%) e do cloreto de alquil dimetil benzil amônio (88,8%). Dentre os antimicrobianos testados, observou-se maior resistência para a tetraciclina (62,2%) e maior sensibilidade para o florfenicol (88,6%). A alta sensibilidade dos isolados frente aos desinfetantes pode estar relacionada à ausência de mecanismo de efluxo. O índice de resistência múltipla médio aos antimicrobianos foi de 0,52, o que demonstra um perfil multirresistente dos isolados, conduzindo para a necessidade do uso racional destas drogas em suinocultura.

E-mail: miliane@ufrrj.br The use of antibiotic in the control of intramammary infections and in the elimination of its possible sources in dairy farms is an important control measure. However, the inappropriate use of antibiotics can result in the appearance of resistant strains and compromise the efficiency of the treatment. Besides Staphylococcus spp. are among the main pathogens of bovine mastitis, they are often resistant to antibiotics, especially beta-lactamics, mainly by two distinct mechanisms: the production of extracellular enzyme beta-lactamase, encoded by the blaZ gene, and production of PBP2a or PBP2’ a penicillin-binding protein with low affinity, encoded by the mecA gene. The expression of mecA gene is constitutive or induced by beta-lactamic antibiotics, such as oxacillin and cefoxitin. The mecA gene is inserted into the chromosome through a staphylococcal mobile genetic element, called staphylococcal cassette chromosome mec (SCCmec). The present study evaluated the phenogenotypical resistance profile to beta-lactam antibiotics of 250 Staphylococcus spp isolates, using oxacillin and cefoxitin as markers in order to produce data to the knowledge of resistance in dairy farms located in the South-Fluminense and the Metropolitan regions of the State of Rio de Janeiro to support the implementation of measures to control this disease. The assessment of resistance was made through 8 different phenotypic tests and yielded 54 profiles. Disk diffusion and agar screen with oxacillin were used as “gold standard” for the calculation of sensitivity, specificity and prediction once they are recommended by the CLSI veterinarian as standardized tests. Disk diffusion with cefoxitin achieved the best performance in the prediction of oxacillin resistance. Genotypic detection of mecA do not provided any positive isolate, otherwise mecI and mecRI genes were also detected in 11.6% (29/250) of the studied Staphylococcus spp. Four cassette mec types were detected (I, II, III and IV), being type I the most disseminated one. Gene blaZ was detected in 5.2% (13/250) isolates. From these 13 blaZ positive isolates, the whole system comprising blaR1-blaI-blaZ was detected in 23.1% (3/13) isolates.

Bactérias do gênero Staphylococcus spp. estão entre os principais agentes etiológicos da mastite bovina e são freqüentemente resistentes aos antimicrobianos, em especial aos beta-lactâmicos, principalmente por dois mecanismos distintos: a produção da enzima extracelular beta-lactamase, codificada pelo gene blaZ, e a produção de PBP2a ou PBP2´, uma proteína ligante de penicilina de baixa afinidade, codificada pelo gene mecA. A expressão do gene mecA é constitutiva ou induzida por antibióticos betalactâmicos, como a oxacilina e cefoxitina. O gene mecA está inserido no cromossomo através de um elemento genético móvel, denominado cassete estafilocócico cromossômico mec (SCCmec). O presente estudo avaliou o perfil fenogenotípico de resistência aos beta-lactâmicos em 250 isolados de Staphylococcus spp., utilizando os marcadores oxacilina e cefoxitina, de modo a produzir dados que possam contribuir para o conhecimento da resistência antimicrobiana em algumas propriedades leiteiras das regiões Sul-Fluminense e Metropolitana do Estado do Rio de Janeiro com o objetivo de subsidiar a implementação de medidas de controle dessa enfermidade. A avaliação da resistência foi feita a partir de 8 diferentes testes fenotípicos, sendo obtidos 54 perfis. Os testes de difusão em disco simples e ágar screen com oxacilina foram utilizados como “padrão ouro” para os cálculos dos valores de sensibilidade, especificidade e predição por serem preconizados pelo CLSI veterinário. O teste de difusão em disco simples com cefoxitina foi o de melhor desempenho na predição da resistência a oxacilina. Na avaliação genotípica, não foi detectado qualquer isolado positivo para o gene mecA, já os genes mecI e mecRI foram detectados igualmente em 11,6% (29/250) dos Staphylococcus spp. avaliados. Foram detectados os quatro tipos de cassete mec analisados (I, II, III e IV), sendo o tipo I o que teve mais ampla distribuição entre as regiões estudadas. E-mail: miliane@ufrrj.br The present study evaluated the pheno- and genotypical antimicrobial resistance profile of coagulase-negative Staphylococcus (CNS) species isolated from dairy cows milk, specially concerning to oxacillin. Of 100 CNS isolates, the S. xylosus was the prevalent species, followed by S. cohnii, S. hominis, S. capitis and S. haemolyticus. Only 6% were phenotypically susceptible to the antimicrobial agents tested in disk diffusion assay. Penicillin and ampicillin resistance rates were significantly higher than others antimicrobials. Four isolates were positive to mecA gene (4%), all represented by the S. xylosus species. The blaZ gene was detected in 16% of the isolates (16/100). It was noticed that all mecA + were also positive to this gene and the presence of both genes was correlated to phenotypic beta-lactamic resistance. We conclude that CNS species from bovine milk presented significantly distinct antimicrobial resistance profiles, evaluated by phenotypic and genotypic tests, which has implications for treatment and management decisions.

Dos 100 isolados de ECN, S.xylosus foi a espécie predominante, seguida por S. cohnii, S. hominis, S. capitis and S. haemolyticus Apenas 6% dos isolados foram fenotipicamente suscetíveis aos agentes antimicrobianos testados no ensaio de difusão em disco. O percentual de resistência à penicilina e ampicilina foi significativamente maior que aos outros antimicrobianos. Quatro isolados foram positivos para o gene mecA (4%), sendo todos representados pela espécie S.xylosus. O gene blaZ foi detectado em 16% dos isolados (16/100), sendo todos mecA positivos e a presença de ambos os genes foi correlacionada com a resistência fenotípica aos beta-lactâmicos. Foi possível concluir que as espécies de ECN provenientes de leite bovino apresentaram distintos perfis de resistência antimicrobiana, avaliados por testes fenotípicos e genotípicos, podendo dificultar a adoção de medidas de tratamento e manejo dos animais.

E-mail: mateus.costa@univasf.edu.br The study aimed to determine the antimicrobial resistance patterns and to identify molecular resistance markers in Staphylococcus spp. (n=210) isolated from small ruminant mastitis in Brazil. The antimicrobial resistance patterns were evaluated by the disk diffusion test and by detection of the presence of mecA, blaZ, ermA, ermB, ermC and msrA genes by PCR. The efflux pump test was performed using ethidium bromide and biofilm production was determined by Congo red agar test along with PCR for detection of the icaD gene. The isolates were most resistant to amoxicillin (50.0%), streptomycin (42.8%), tetracycline (40.4%), lincomycin (39.0%) and erythromycin (33.8%). Pan-susceptibility to all tested drugs was observed in 71 (33.8%) isolates and 41 Staphylococcus isolates were positive for the efflux pump. Although phenotypic resistance to oxacillin was observed in 12.8% of the isolates, none harbored the mecA gene. However, 45.7% of the isolates harbored blaZ indicating that beta-lactamase production was the main mechanism associated with staphylococci resistance to beta-lactams in the present study. The other determinants of resistance to antimicrobial agents ermA, ermB, ermC, and msrA were observed in 1.4%, 10.4%, 16.2%, and 0.9% of the isolates, respectively. In addition, the icaD gen was detected in 32.9% of the isolates. Seventy three isolates (54 from goats and 19 from sheep) were negative for all resistance genes tested and 69 isolates presented two or more resistance genes. Association among blaZ, ermA, ermB, ermC and efflux pump were observed in 17 isolates, 14 of which originated from goats and three from sheep. The data obtained in this study show the resistance of the isolates to beta-lactamics, which may be associated with the use of antimicrobial drugs without veterinary control.

Os padrões de resistência a agentes antimicrobianos foram avaliados pelo teste de difusão em disco e pela detecção da presença dos genes mecA, blaZ, ermA, ermB, ermC e msrA via PCR. O teste da bomba de efluxo foi realizado utilizando brometo de etídio e a produção de biofime foi determinada pelo teste do vermelho congo em paralelo com o PCR para detecção do gene icaD. Os isolados foram mais resistentes a amoxicilina (50,0%), estreptomicina (42,8%), tetraciclina (40,4%), lincomicina (39,0%) e eritromicina (33,8%). Setenta e um (33,8%) isolados foram sensíveis a todas as drogas testadas e 41 foram positivos para a bomba de efluxo. Embora a resistência fenotípica a oxacilina tenha sido observada observada em 12,8% dos isolados, nenhum possuiu o gene mecA. Entretanto, 45,7% dos isolados continham a gene blaZ, indicando que a produção de beta-lactamases foi o principal mecanismo associado com a resistência dos Staphylococcus aos beta-lactâmicos. Os outros determinantes de resistência a agentes antimicrobianos ermA, ermb, ermC e msrA foram observados em 1,4%, 10,4%, 16,2% e 0,9% dos isolados respectivamente. Além disso, o gene icaD foi detectado em 32,9% dos isolados. Setenta e três isolados (54 de cabras e 19 de ovelhas) foram negativos para todos os genes de resistência testados e 69 isolados apresentaram dois ou mais genes de resistência. A associação entre blaZ, ermA, ermB, ermC e bomba de efluxo foi observada em 17 isolados dos quais 14 eram oriundos de cabras e três de ovelhas. Os dados obtidos no presente estudo indicam a resistência dos isolados aos beta-lactâmicos, o que pode estar associado ao uso sem controle veterinário destas drogas nos animais.

E-mail: rinaldo.mota@hotmail.com Although exist poultry non-pathogenic Escherichia coli strains, many others have capacity to impose serious damages to this birds, being able to cause different infectious diseases. Pathogenic strains are termed Avian pathogenic Escherichia coli (APEC) strains. APEC strains harbor chromossomal and plasmid pathogenicity-related genes. The presence of resistance plasmids in avian E. coli strains could facilitate horizontal tranfer of virulence gene between pathogenic and non pathogenic strains. The aim of this paper was to determine the resistance level to 13 different antibacterial drugs of avian E. coli strains (35) isolated from commercial poultry of Pernambuco State, Brazil, and to correlate the detected resistance level to the presence of plasmids. The results show that 94.28% of strains were resistant to at least three different antibacterial drugs with the highest percentage to lincomycin. The Minimal Inibitory Concentration (MIC) showed that multi- resistance to various antibacterial drugs was present in these strains. Plasmids of several sizes, including plasmids of approximately 88Mda were detected in most of the studied strains. The results herein obtained suggest that the high resistance level observed could be due to the presence of plasmids, what could facilitate the transfer of pathogenicity related genes among pathogenic and non pathogenic strains; it is necessary to take a constant survey on the resistance level to antimicrobial drugs of avian E. coli strains to reach a better control of APEC strains and avoid transfer of pathogenicity related genes between strains.

E-mail: rinaldo.mota@hotmail.com Embora existam linhagens de Escherichia coli não patogênicas para aves, muitas outras possuem a capacidade de causar sérios danos à saúde das mesmas, sendo capazes de ocasionar diferentes tipos de processos infecciosos. As linhagens patogênicas são denominadas Avian Pathogenic Escherichia coli (APEC), possuindo genes relacionados ao processo de patogênese em epissomos (plasmídios) ou no cromossomo. A presença de plasmídios, contendo genes de resistência a antibióticos em linhagens aviárias, patogênicas ou não, indicam a possibilidade de transferência gênica lateral entre diferentes tipos de linhagens facilitando também a transferência de genes de patogenicidade ou virulência. Objetivou-se com este estudo avaliar o perfil de sensibilidade a antibióticos (13) de diferentes amostras (35) de E. coli isoladas de aves comerciais do Estado de Pernambuco apresentando, ou não, sinais clínicos de processos infecciosos e correlacionar esta resistência com a presença de plasmídios. Os testes utilizados demonstraram que 94,28% dos isolados foram resistentes a três ou mais antibióticos, com a lincomicina apresentando o maior percentual de resistência (100%). Na Concentração Inibitória Mínima (CIM) observou-se multirresistência a vários antimicrobianos. A presença de plasmídios foi detecada em 80,0% (28/35) dos isolados, com 16 isolados apresentando plasmídios com peso molecular aproximado de 88 MDa. Também foi verificada a presença de linhagens apresentando plasmídios de vários tamanhos. Concluiu-se que isolados de E. coli resistentes a antimicrobianos utilizados na avicultura estão presentes no Estado de Pernambuco, tanto em frangos de corte quanto em poedeiras comerciais. A presença de plasmídios detectados na maioria dos isolados pode estar associada à resistência aos antimicrobianos e sugere a presença de possíveis genes relacionados à patogenicidade. Monitorar a resistência a antibióticos em bactérias isoladas de animais torna-se um fator determinante para eleição e êxito do tratamento, bem como a possibilidade de eliminação daquelas que possuem plasmídios para se evitar a transferência de genes relacionados à patogenicidade.

E-mail: llovato@smail.ufsm.br Cracids are wildlife Galliformes which inhabits the America’s tropical forests. Fifty one cloacal swabs were collected from 10 different species of captive cracids from the Rio Grande do Sul State during 2007. The cloacal swab samples were submitted to bacterial isolation, identification and, subsequently; antimicrobial susceptibility testing. Ninety three bacterial isolates were obtained from the cracid population examined. The most prevalent among the isolates were Escherichia coli, and bacteria from the Staphylococcus and Streptococcus genera. All samples tested in this study were negative for Salmonella spp. The antimicrobial susceptibility tests showed that none of the 93 strains presented resistance to the antimicrobial imipinem. In addition, the lower percentages of resistance were observed against cloranfenicol and ciprofloxacine. The bacteria genus and species with the highest percentage of resistance to the different antimicrobials examined were E. coli, Serratia marcescens, Staphylococcus spp. and Streptococcus spp. In conclusion, the data presented in this article demonstrate that the cloacal microbiota of the reported cracid population is composed of several bacterial genera and species and multi-drug resistance may be a problem for the future, since some strains showed elevated percentage of resistance against several different antimicrobials.

E-mail: llovato@smail.ufsm.br Os cracídeos são aves silvestres que habitam as matas tropicais da América. Foram coletadas, no ano de 2007, amostras cloacais de 51 aves de dez espécies diferentes de cracídeos mantidos em cativeiros no Estado do Rio Grande do Sul. A partir dos swabs, colhidos assepticamente, foi realizado o isolamento e a caracterização bacteriana e o teste de susceptibilidade antimicrobiana dos isolados. Foram identificadas 93 cepas de bactérias. As bactérias mais frequentemente isoladas foram Escherichia coli, Staphylococcus spp. e Streptococcus spp. Todas as amostras foram negativas para o isolamento de Salmonella spp. O resultado do teste de sensibilidade mostrou que dentre as 93 cepas isoladas, todas foram sensíveis apenas ao imipinem. Adicionalmente, os menores percentuais de resistência foram observados frente ao cloranfenicol e ciprofloxacina. Os gêneros e espécies bacterianas com maior percentual de resistência a diferentes antibióticos testados foram Escherichia coli, Serratia marcescens, Staphylococcus aureus e Streptococcus spp. Com os resultados obtidos no presente trabalho, concluí-se, que a população de cracídeos estudada apresenta sua microbiota cloacal composta por vários gêneros e espécies bacterianas e que a multirresistencia pode ser um problema no futuro, uma vez que algumas cepas isoladas mostraram percentuais elevados de resistência a diferente antimicrobianos.

E-mail: dayseabreu@predialnet.com.br The pathogenicity of Escherichia coli strains is partially related to the expression of virulence factors genes, present in genetic elements called plasmids. APEC strains responsible for diseases in birds may present the iss gene which increases the resistance of E. coli strains to the lityc effect of the host’s serum, besides resistance to several antimicrobials. This study was conduced in order to detect E. coli in tracheae of meat-type quails and to evaluate, by the presence of the iss gene and the profile of antimicrobial susceptibility, the pathogenic potential of the isolated samples for birds and humans. One hundred and eighty tracheae of quails were collected for detection of E. coli, antimicrobial sensitivity tests, and for polymerase chain reaction (PCR), for detection of iss gene. From the examined quails, 8.9 % (16/180) were positive for E. coli, from which 20 strains of this bacterium were obtained. Most of them were resistant to Tetracycline (16/20), followed by Ceftadizime (13/20) and Nalidixic-acid (12/20) and only one isolate was resistant to Amoxicillin. The detection of iss gene occurred in 55% (11/20) of the isolates, indicating that these strains had the potential to be pathogenic not only for quails, but also for other kinds of birds, other animals and even human beings that would be in contact with these E. coli isolates.

Foram coletadas 180 traquéias de codornas para detecção de E. coli, determinação do perfil de resistência a agentes antimicrobianos e posterior detecção, por reação em cadeia da polimerase (PCR), do gene iss. Das traquéias analisadas, 8,9 % (16/180) foram positivas para E. coli, sendo obtidos 20 isolados deste agente. A maioria dos isolados foi resistente à Tetraciclina (16/20), seguida pela Ceftazidima (13/20) e Ácido Nalidíxico (12/20), sendo apenas um resistente à Amoxicilina. A detecção do gene iss ocorreu em 55% (11/20) dos isolados. A presença do gene iss e a resistência a múltiplos antimicrobianos dos isolados obtidos neste estudo pode indicar um possível potencial patogênico das cepas de E. coli tanto para codornas quanto para outros tipos de aves e animais e mesmo para o ser humano que fique em contato com as mesmas.

All hemolytic isolates presented beta-hemolysin, and 38% of the non-hemolytic isolates were able to express hemolysins in the presence of a beta-hemolytic strain. The amplification of the coa-gene displayed four different size polymorphisms with about 400 bp, 600 bp, 700 bp and 900 bp. The spaA gene that encodes the IgG-binding region of protein A revealed sizes of 700 bp and 900 bp. The amplification of region X from spaA yielded a single amplicon for each isolate with the prevalent amplicon size being of 180 bp. Amplification of sae gene yielded an amplicon size of 920 bp in 71% of the isolates. Antibiotic resistance pattern revealed that 42% S. aureus were susceptible to all antimicrobials tested. Seven different antibiotic patterns were observed. Our results indicated that 47% and 25% of S. aureus strains exhibited resistance to penicillin and oxacillin respectively. All oxacillin-resistant isolates were mecA-positive.

All strains of Salmonella were subjected to bacterial conjugation, and the E. coli K12 Nalr Lac+ F standard strain was used as receptor, with the purpose to verify the ability to transfer the resistance marks. Gene transfer phenomenon was detected in seven strains, and except Salmonella Typhimurium which transconjugated to Sm, Tc and Su, the remaining ones were characterized by transferring mark Su only. By plasmidial analysis of strains used and their respective transconjugants, 63 Kb plasmid was found, which was probably related to S. Typhimurium resistance.
