E-mail: thiagoalmeida2@hotmail.com Sheep farming has increased significantly in Brazil during the last decades. Concurrently, research groups and diagnostic laboratories compile data and perform retrospective studies to provide important insight for professionals. A prevalence study from January 1996 to December 2010 was carried out in the archives of Laboratório de Anatomia Patológica Animal (LAP), Universidade Federal de Mato Grosso do Sul (UFMS). Laboratório de Bacteriologia, UFMS, and Setor de Patologia Veterinária at Universidade Federal do Rio Grande do Sul helped on the diagnostic of pulmonary mannheimiosis and scrapie respectively. The reports for sheep were reviewed and grouped into conclusive and inconclusive ones. The conclusive cases were classified according to the etiology of the disease. In the period, 331 exams (3.97%) were done. Sixty-four experimental cases and materials from other states or countries (19.3%) were excluded. Remaining cases (267), eighty-seven (32.6%) were inconclusive and 180 (67.4%) were considered conclusive reports, were classified according to the etiology: 60 (33.3%) infectious and parasitary diseases; 45 (25%) were poisonings and toxi-infections; 41 (22.8%) were summarized as “injuries without apparent cause”; 22 (12.2%) cases of metabolic and nutritional diseases; 10 (5.6%) were classified as “other disorders” and 2 (1.1%) case of neoplasms. Haemonchosis, fibrinonecrotic or fibrinopurulent pleuropneumonia, bronchopneumonia and pneumonia, poisonings by Brachiaria spp. and copper poisoning were the most prevalent diseases in sheep. Two cases of scrapie have been diagnosed in this period.

Concomitantemente, grupos de pesquisas e laboratórios de diagnósticos realizam estudos retrospectivos com a finalidade de fornecer subsídios técnico-científicos para os médicos veterinários. Desta forma, realizou-se um estudo de prevalência nos arquivos do Laboratório de Anatomia Patológica Animal (LAP) da Universidade Federal de Mato Grosso do Sul (UFMS) no período de Janeiro de 1996 a Dezembro de 2010. O Laboratório de Bacteriologia da UFMS e o Setor de Patologia Veterinária da Universidade Federal do Rio Grande do Sul forneceram apoio diagnóstico nos casos de mannheimiose pulmonar e scrapie, respectivamente. Os laudos da espécie ovina foram revisados e agrupados em conclusivos e inconclusivos, dos quais foram excluídos os casos experimentais e de outros estados e países. Os casos conclusivos foram classificados de acordo com a etiologia da doença. Os exames da espécie ovina somaram 331 laudos (3,97 %) de um total de 8.333 casos diagnosticados no período. Destes, foram excluídos sessenta e quatro (19,3%) casos experimentais e materiais oriundos de outros estados ou países. Dos 267 casos remanescentes, 87 (32,6%) foram inconclusivos e 180 (67,4%) considerados conclusivos, sendo 60 (33,3%) doenças infecciosas e parasitárias; 45 (25%) intoxicações e toxi-infecções; 41 (22,8%) “lesões sem causa definida”; 22 (12,2%) doenças metabólicas e nutricionais; 10 (5,6%) foram classificadas como “outros distúrbios” e 2 (1,1%) neoplasmas. A hemoncose, intoxicação por Brachiaria spp., pleuropneumonias, broncopneumonias, pneumonias fibrinonecrosante ou fibrinossupurativa sem causa definida e a intoxicação por cobre foram as doenças mais prevalentes no período estudado. Dois casos de scrapie foram diagnosticados no período.

The diagnosis of Mycoplasma hyopneumoniae infection is often performed through histopathology, immunohistochemistry (IHC) and polymerase chain reaction (PCR) or a combination of these techniques. PCR can be performed on samples using several conservation methods, including swabs, frozen tissue or formalin-fixed and paraffin-embedded (FFPE) tissue. However, the formalin fixation process often inhibits DNA amplification. To evaluate whether M. hyopneumoniae DNA could be recovered from FFPE tissues, 15 lungs with cranioventral consolidation lesions were collected in a slaughterhouse from swine bred in herds with respiratory disease. Bronchial swabs and fresh lung tissue were collected, and a fragment of the corresponding lung section was placed in neutral buffered formalin for 48 hours. A PCR assay was performed to compare FFPE tissue samples with samples that were only refrigerated (bronchial swabs) or frozen (tissue pieces). M. hyopneumoniae was detected by PCR in all 15 samples of the swab and frozen tissue, while it was detected in only 11 of the 15 FFPE samples. Histological features of M. hyopneumoniae infection were presented in 11 cases and 7 of these samples stained positive in IHC. Concordance between the histological features and detection results was observed in 13 of the FFPE tissue samples. PCR was the most sensitive technique. Comparison of different sample conservation methods indicated that it is possible to detect M. hyopneumoniae from FFPE tissue. It is important to conduct further research using archived material because the efficiency of PCR could be compromised under these conditions.

Entretanto, o processo de fixação em formalina pode inibir a amplificação de DNA. Para avaliar se DNA de M. hyopneumoniae poderia ser recuperado de tecido FFEP, 15 pulmões com lesões de consolidação crânio-ventral de suínos oriundos de rebanhos com problemas respiratórios foram selecionados no abatedouro. Swabs bronquiais e pulmão fresco foram colhidos, e um fragmento da mesma porção de pulmão foi colocado por 48 horas em solução de formalina tamponada e posteriormente processado e embebido em parafina. PCR foi realizada comparando amostras de tecido fixado em formalina com amostras que passaram somente por refrigeração (swab bronquial) ou foram congeladas (fragmentos de tecido). A detecção de M. hyopneumoniae ocorreu em todas as 15 amostras de swabs e tecido congelado enquanto em amostras de tecido FFEP, o agente foi detectado somente em 11 das 15 amostras. Características histológicas de infecção por M. hyopneumoniae ocorreram em 11 casos e 7 destas amostras obtiveram marcação imuno-histoquímica positiva. Concordância entre histologia e detecção a partir de tecido FFEP foi observada em 13 casos. Dentre as técnicas analisadas, a PCR foi a mais sensível. A comparação de diferentes métodos de conservação de amostras indica que é possível detectar M. hyopneumoniae a partir de tecido FFEP, fato importante para pesquisa utilizando material arquivado, porém a eficácia do teste de PCR pode ficar comprometida sob essas condições.

A set of six primers were designed derived from the dsbE-like gene of A.pleuropneumoniae and validate the assay using 9 A. pleuropneumoniae reference/field strains, 132 clinical isolates and 9 other pathogens. The results indicated that positive reactions were confirmed for all A. pleuropneumoniae strains and specimens by LAMP at 63°C for 60 min and no cross-reactivity were observed from other non-A.pleuropneumoniae including Haemophilus parasuis, Escherichia coli, Pasteurella multocida, Bordetella bronchiseptica, Streptococcus suis, Salmonella enterica, Staphylococcus, porcine reproductive and respiratory syndrome virus (PRRSV), and Pseudorabies virus. The detection limit of the conventional PCR was 102 CFU per PCR test tube, while that of the LAMP was 5 copies per tube. Therefore, the sensitivity of LAMP was higher than that of PCR. Moreover, the LAMP assay provided a rapid yet simple test of A. pleuropneumoniae suitable for laboratory diagnosis and pen-side detection due to ease of operation and the requirement of only a regular water bath or heat block for the reaction.

E-mail: calderon@fmvz.unesp.br Exams of tracheobronchial secretions are widely used in research of lung diseases in several animal species, including men. The objective of this paper was to investigate feasibility of the technique for harvesting trachea-bronchial lavage in sheep and to verify its clinic-cytological relationship in healthy sheep and others suffering from pneumonia. In this study 33 sheep were used, 18 healthy and 15 sheep with clinical signs of respiratory disease, divided into groups GS and GD. Clinic examination was performed by obtaining tracheobronchial lavage through the nasal route. The crop was washed with the addition and aspiration of sterile saline. The samples were processed by cytospin and staining with Wright-Giemsa and Shorr. The total cell count and the number of red blood cells per milliliter was higher in the group of sheep with bronchopneumonia. In healthy animals predominance of macrophages was noted, followed by the number of epithelial cells, neutrophils and lymphocytes. In the group of sick animals fewer macrophages and predominance of the neutrophil population were noted. Because of its easy performance to obtain representative material, the technique studied was considered effective to obtain tracheobronchial fluid and therefore is a good method to harvest cells for research of the airways.

Os objetivos desta pesquisa foram a viabilização da técnica de colheita de lavado traqueobrônquico na espécie ovina e o estudo da relação clínico-citológica do lavado de ovinos portadores de broncopneumonia e sadios. Foram utilizados 33 ovinos, 18 sadios e 15 portadores de enfermidade respiratória com sinais clínicos de envolvimento das vias aéreas, divididos nos respectivos grupos, GS e GD. Após o exame físico foi realizado o lavado traqueobrônquico por via nasotraqueal. A colheita do lavado foi feita com a inoculação e aspiração de solução fisiológica estéril. As amostras foram processadas citologicamente através de citocentrifugação e coradas pelos métodos Wright-Giemsa e Shorr. Tanto a contagem total de células epiteliais quanto o número de hemácias por mililitro foi maior no grupo de animais com broncopneumonia. Nos animais sadios notou-se predomínio de macrófagos, seguido por células epiteliais cilíndricas, neutrófilos e linfócitos. No grupo de animais doentes havia menor número de macrófagos, e predomínio da população de neutrófilos. Por ser de fácil realização, pouco dispendiosa e pela obtenção representativa de material, a técnica estudada mostrou-se eficaz na obtenção de fluidos traqueobrônquicos e, portanto um bom método de colheita de células para uso nas pesquisas de vias aéreas.

E-mail: andrezamedvet@yahoo.com.br The natural occurrence of bronchopneumonia in calves and the effect of DL-a-tocopherol in the prophylaxis and treatment of this disease is studied. We evaluated 60 male calves, aged up to 10 days, divided into two groups: GSV (group without vitamin) and GCV (group with vitamin). Supplementation with a single dose of 4,500 IU of DL-a-tocopherol administered intramuscularly (IM) was preceded by clinical examination, complete blood count (CBC), determination of serum proteins, globulins, gamaglutamyltransferase and tracheobronchial lavage cytology (D0) to check sanity and homogenization of the groups. The animals were kept two by two in individual 2.40 m2 stalls, where they remained until day 21 (D0 to D21). The animals were examined daily and, in the presence of clinical signs of bronchopneumonia (DX), taken from stalls, evaluated by CBC and tracheobronchial lavage cytology, and treated with enrofloxacin (5 mg/kg, IM). During the treatment, clinical examination was performed daily by the CBC and tracheobronchial lavage cytology repeated one week after the end of treatment (DY). In healthy animals, CBC and tracheobronchial lavage cytology were repeated on the last day of the experiment (D21). No significant difference for the blood variables gamaglutamiltrasfrase (p=0.09), serum protein (p=0.27) and globulin (p=0.10) and the age of animals (p=0.15) were observed in both of groups. Animals in group GSV and GCV took an average of 11 and 12 days for get sick, respectively. However, no statistical difference between was observed in both groups (p=0.68). In 34 (56.66%) animals were diagnosed bronchopneumonia, and 17 of these calves (50%) belonged to group GCV and 17 (50%) to group GSV. With respect to clinical signs, there was no significant difference between groups in any of the times studied. Out of 34 animals which were affected, 73.52% died of the disease, 64.7% were from GCV group and 82.35% were from GSV group. Cytology of tracheobronchial lavage showed no difference between groups in any of the times studied (D0, DX, DY and D21). We conclude that supplementation with vitamin E had no influence in the prophylaxis and Treatment of bronchopneumonia in calves.

Foram avaliados 60 bezerros machos, com idade máxima de 10 dias, divididos em dois grupos experimentais: GSV (grupo sem vitamina) e GCV (grupo com vitamina). A suplementação com dose única de 4.500 UI de acetato de DL-a-tocoferol por via intramuscular (IM) foi precedida por exame físico, perfil hematológico, determinação de proteínas séricas, globulinas, gamaglutamiltransferase e citologia do lavado traqueobrônquico (D0) para verificar a sanidade e homogeinização dos grupos. Os bezerros foram mantidos em bezerreiro e distribuídos aleatoriamente, 2 a 2 em baias individuais de 2,40m2, onde permaneceram até o 21º dia (D0 ao D21). Foram avaliados por exame físico diário e, na presença de sinais clínicos indicativos de broncopneumonia (DX), retirados do bezerreiro, avaliados por hemograma e citologia do lavado traqueobrônquico, e tratados com enrofloxacina (5mg/kg, IM). Durante o tratamento, o exame físico foi realizado diariamente, e o hemograma e a citologia do lavado traqueobrônquico repetidos uma semana após seu término (DY). Nos bezerros sadios foram repetidos hemograma e citologia do lavado traqueobrônquico, no último dia (D21) do experimento. Não houve diferença significativa quanto às variáveis gamaglutamiltrasfrase (p=0,09), proteínas séricas (p=0,27) e globulinas (p=0,10) e a idade dos bezerros (p=0,15) entre os grupos. Os bezerros do grupo GSV e GCV levaram em média 11 e 12 dias para adoecerem, respectivamente. Contudo, não houve diferença estatística significativa entre os grupos (p=0,68). Em 34 bezerros (56,66%) foi diagnosticado broncopneumonia, sendo que 17 destes bezerros (50%) pertenciam ao grupo GCV e 17 (50%) ao grupo GSV. Com relação aos sinais clínicos, não houve diferença significativa entre os grupos em nenhum dos momentos estudados. Dos 34 bezerros que adoeceram 73,52% morreram pela doença, sendo 64,7% do GCV e 82,35% do GSV. A citologia do lavado traqueobrônquico não apresentou diferença significativa entre os grupos em nenhum dos momentos estudados (D0, DX, DY e D21). A suplementação com vitamina E não teve influência na profilaxia e no tratamento de broncopneumonia de bezerros.

E-mail: ecco@vet.ufmg.br This paper describes an outbreak of pneumonia in a sheep herd in the central region of Minas Gerais, Brazil. Clinically, the animals presented apathy, exhibited respiratory difficulty during 2 to 3 days or sudden death. The animals were not medicated and found dead. Grossly, the pulmonary findings were similar in all sheep. The pulmonary cranial lobes and the ventral portion of caudal lobes were consolidated and purulent exsudate streamed out of the airways. In the parenchyma of the cranial lobes there were white slightly prominent multifocal to coalescent areas with 0.2 to 0.5cm in diameter intercalated with dark red areas. Consolidated lesions occupied 70 to 80% of the lungs. Fibrinous pleuritis was observed in sheep 1, 2 and 3. Microscopically, the findings were fibrinopurulent bronchopneumonia with intense hyperemia, areas with intra-alveolar hemorrhage and thickening of interlobular septa with numerous neutrophils, cellular rests and scattering fibrin. Multifocal areas with liquefaction necrosis containing numerous bacterial colonies were observed in sheep 1, 2 and 3. In the cranial lobes of these sheep, there were areas with degenerated neutrophils forming clusters of basophilic cells with alongated nuclei (“oat cells”) associated with bacterial colonies. The histological findings were characteristic of pneumonia caused by Mannheimia (M.) haemolytica. Samples of the cranial lobes were sent for bacterial culture, and M. haemolytica was isolated and identified in all animals. This is the first report correlating pathological findings and the isolation of M. haemolytica as cause of bronchopneumonia in sheep in the country.

Clinicamente os animais apresentavam apatia, mostravam dificuldade respiratória durante dois ou três dias ou morriam subitamente. À necropsia as alterações pulmonares eram similares em todos os ovinos. Havia consolidação dos lobos craniais e da parte ventral dos lobos caudais e ao corte fluía exsudato mucopurulento da traquéia e dos brônquios. No parênquima dos lobos craniais havia áreas brancas multifocais a coalescentes com 0,2-0,5cm de diâmetro, levemente proeminentes e intercaladas por áreas vermelho-escuras. Pleurite fibrinosa foi observada nos Ovinos 1, 2 e 3. As lesões de consolidação ocupavam cerca de 70-80% da extensão pulmonar. Microscopicamente, as alterações eram de broncopneumonia fibrinopurulenta com intensa hiperemia, áreas com hemorragia intra-alveolar e espessamento dos septos interlobulares por inúmeros neutrófilos, restos celulares e intensa exsudação de fibrina. Áreas multifocais com necrose de liquefação contendo numerosas colônias bacterianas foram observadas no Ovino 3. Nos lobos craniais dos Ovinos 1, 2 e 3, haviam áreas com neutrófilos degenerados formando aglomerados de células alongadas com formato de “grãos de aveia” associados a colônias bacterianas. As alterações histológicas foram características de pneumonia causada por Mannheimia (M.) haemolytica. Amostras dos lobos craniais de todos os ovinos foram encaminhadas para cultivo bacteriológico e M. haemolytica foi isolada e identificada em todos os animais. Este é o primeiro relato correlacionando os achados patológicos e o isolamento de M. haemolytica como causa de broncopneumonia em ovinos no Brasil.

Clinically, the pigs presented reduced growth rate, anorexia, lethargy, cough and dyspnea, especially after they were moved. High body temperature (40oC in average) was verified in some animals. Auscultation revealed moderate pulmonary crepitation and stertors. Pulmonary gross lesions were typical of lobular bronchopneumonia. Lung lesions were characterized by ventral-cranial consolidation in the majority of the cases. The color of affected pulmonary areas varied from diffuse dark red to mosaic pattern (dark red lobule intercalate by grayish lobule) or diffusely grayish. The majority of the lungs had mucopurulent exsudate in the bronchial lumen that also drained from the parenchyma cut surface. Upon microscopy, the changes were characterized by purulent and histiocytic bronchopneumonia with necrotic foci. In some animals, there was BALT hyperplasia associated with perivascular and peribronchial plasma cells and lymphocytes infiltration in most of these cases. Bordetella bronchiseptica and Streptococcus spp. were the most frequently isolated bacteria. Immunohistochemistry evaluation demonstrated Mycoplasma hyopneumoniae on the luminal surface of bronchial and bronchiolar epithelial cells, and the DNA of bacteria was detected by PCR. This is the first report of bronchopneumonia in wild boars associated with M. hyopneumoniae infection.

The ground seeds were administered orally to 9 bovines. Doses of 1g/kg/day, 2g/kg/day, 3g/kg/day each in one bovine, and 5g/kg/day in two of three bovines, given for 61- 63 days, did not cause poisoning. Doses of 5g/kg, in one bovine, 7.5g/kg in two bovines and 10g/kg in one bovine, given for 47-61 days, caused symptoms between 47 and 80 days after the first administration and caused death between 3 hours and 5 days after the onset of symptoms. The main clinical signs were positive venous pulse of the jugular vein, abdominal breath, tachycardia, loss of appetite, dry feces, sub-mandibular edema and weakness. Bovines that did not die, were slaughtered 8 or 9 months after first administration. At necropsy pulmonary paleness, hydropericardium, hydrothorax, hydroperitoneum, mesenteric edema, augmented hepatic consistency, discoloration of the liver, right cardiac ventricle dilatation and ruminal wall edema were seen. The main histological lesions were thickening of the alveolar walls and of the arterioles with narrowing of their lumen, and periarteriolar fibrosis, besides hepatic and cardiac lesions of minor importance. It is concluded, that the lesions caused by ingestion of the seeds of C. mucronata over a long period are caused by the difficulties of blood passage through the pulmonar vessels due to fibrosis and thickening of the arteriolar walls through the pneumotoxic action of the plant.

#40 - Interstitial pneumonia in cattle fed moldy sweet potatoes (Ipomoea batatas), 23(4):161-166

[Interstitial pneumonia in cattle fed moldy sweet potatoes (Ipomoea batatas).] Pneumonia intersticial em bovinos associada à ingestão de batata-doce (lpomoea batatas) mofada. Pesquisa Veterinária Brasileira 23(4):161-166. Depto Patologia, Universidade Federal de Santa Maria, 97105- 900 Santa Maria, RS, Brasil. E-mail: claudioslbarros@uol.com.br Cases of respiratory disease were diagnosed in five out of 23 cattle (21. 7%) after they wete fed moldy damaged sweet potatoes (lpomoea batatas) on a small farm in the county of São Vicente do Sul, state of Rio Grande do Sul, Brazil. Of those five cattle, three died spontaneously and another one was euthanatized for necropsy while showing advanced respiratory clinicai signs. The disease manifested itself approximately 24 hours after the ingestion of the sweet potatoes and lasted from 1 to 4 days. Clinicai signs included dyspnea (labored breathing and abdominal respiration), tachypnea, extended neck with Iow carriage of the head and rhythmical flaring of the nostrils. Two cows were necropsied. Necropsy findings included distended pale and rubbery lungs which failed to collapse when the thorax was open, and marked pulmonar interstitial emphysema and edema. Lymphoid hyperplasia was observed in the hilar nodes and spleen. Histologically, the lesions were those of interstitial pneumonia. Alveolar septa were thickened by fibroblasts and inflammatory cells, and there was hypertrophy and hyperplasia of type li pneumocytes; the interlobular septa were distended by edema and emphysema. The culture of the moldy sweet potatoes yielded Fusarium solani and Fusarium oxysporum.

Três dos cinco bovinos afetados morreram espontaneamente, e o quarto foi sacrificado para necropsia quando mostrava sinais clínicos respiratórios avançados. A manifestação clínica iniciara cerca de 24 horas após a ingestão das batatasdoces e a evolução clínica foi de 1 a 4 dias. Os sinais clínicos incluíam dispnéia (respiração laboriosa e abdominal), taquipnéia, pescoço estendido com cabeça baixa e dilatação ritmada das narinas. Dois bovinos foram necropsiados. Os achados de necropsia incluíam pulmões distendidos, pálidos e de consistência borrachenta, que não colapsavam quando o tórax era aberto; enfisema e edema acentuados eram evidentes no pulmão. Os linfonodos e o baço apresentavam alterações características de hiperplasia linfóide. Histologicamente, as lesões eram típicas de pneumonia intersticial. Os septos alveolares estavam espessados por fibroblastos e células inflamatórias, havia hipertrofia e hiperplasia de pneumócitos tipo II; os septos interlobulares estavam distendidos por edema e enfisema. A cultura de amostras das batatas-doces mofadas produziu Fusarium solani e F. oxysporum.
