E-mail: andre.meneses@ufra.edu.br To develop a model of hemodialysis (HD), 18 healthy dogs, without a defined breed, males, weighing 7-14 kg, were studied. A double lumen catheter was inserted into the jugular vein for vascular access. HD sessions, totalizing 5 for each dog, were performed with a proportional HD machine, controlled isovolemic ultrafiltration, standard dialysis solution and bicarbonate buffer. Sodium profile and sodium heparin were used. During HD sessions the dogs were kept anesthetized (levomepromazine and propofol). Hematological and biochemistry data, blood gas analysis, systemic arterial blood pressure, and activated clotting time were evaluated. Serum biochemistry e blood gas analysis showed, respectively, sodium and SO2 maintenance. Systemic blood pressure kept sustained during HD sessions. It could be concluded that it was possible to develop a HD dog model that is a safe and viable technique to be used in chronic renal failure patients.

E-mail: andre.meneses@ufra.edu.br Com o objetivo de desenvolver um modelo de hemodiálise (HD) em cães, foram estudados 18 animais, sem raça definida, machos, clinicamente sadios, com peso corporal variando entre sete e 14 kg. O acesso vascular foi obtido através de implantação do cateter de duplo lúmen em veia jugular externa. As sessões de HD, em número de cinco por animal, com até três horas de duração, foram realizadas em hemodialisadora de sistema proporcional com ultrafiltração (UF) controlada, com solução dialisante padrão e tampão bicarbonato. A UF foi ajustada para HD isovolêmica, utilizou-se perfil de sódio, e para anticoagulação heparina sódica. Os animais foram mantidos anestesiados com cloridrato de levomepromazina e propofol. Foram avaliados dados hematológicos, bioquímicos, hemogasometria, pressão arterial sistêmica e tempo de coagulação ativado. Foi observada diminuição do número global de hemácias, volume globular, hemoglobina e leucócitos. Em relação aos exames bioquímicos, houve manutenção nos níveis de sódio sérico, e quanto à hemogasometria, a manutenção da SO2. A pressão arterial sistêmica manteve-se constante. Os resultados obtidos no presente trabalho permitiram concluir que foi possível o desenvolvimento do modelo proposto e mostrou que a HD em cães é um método viável e seguro, que poderá contribuir para o tratamento clínico da insuficiência renal nesta espécie.


#72 - Effect of medroxy-progesterone acetate on follicular growth and endometrial cycloxygenase-2 (COX-2) expression during the bovine estrous cycle, 30(7):581-585

E-mail: valerio.portela@gmail.com The objective of this study was to evaluate the effect of medroxy-progesterone acetate (MAP) with or without estradiol benzoate (EB) on follicular growth during the estrous cycle in cattle. In the first experiment, Hereford cows were synchronized with a synthetic analogue of PGF2 alpha and were treated with two different doses of MAP (250 or 500 mg) with or without EB for 7 days starting on day 8 of the estrous cycle. Follicular growth was inhibited (P<0.05) in all cows except controls and those receiving 250mg MAP without EB. Seventy-five percent of the animals (15/20) showed estrus on days 21 and 22 of the cycle rather than at MAP withdrawal, demonstrating that these treatments did not induce estrus. To determine whether the EB treatment altered endometrial sensitivity to oxytocin and thus the luteolytic cascade, multiparous pre-synchronized cows received 5 mg of EB followed 6 hours later with 50 IU of oxytocin (OT; n=9). Eight hours after EB injection, endometrial fragments were collected from the cows on days 4, 13 and 17 of the estrous cycle and COX-2 gene expression was measured by PCR. EB increased COX-2 mRNA levels only on day 17 of the estrous cycle (P<0.05). In conclusion, MAP alone or associated with EB is able to suppress bovine follicular growth. However, EB in the presence of MAP is not efficient to induce luteolysis in cows when injected on day 8 of the estrous cycle.

No primeiro experimento, vacas da raça Hereford foram sincronizadas com um análogo sintético de PGF2á e tratadas com duas doses diferentes de MAP (250 ou 500mg), com ou sem EB, durante 7 dias, iniciando-se no oitavo dia do ciclo estral. Observou-se uma inibição do crescimento folicular (P<0,05) em todas as vacas, exceto no grupo controle e no grupo que recebeu 250mg de MAP sem BE. Os 75% dos animais não exibiu estro no momento da remoção do MAP, mas sim nos dias 21 e 22 do ciclo, demonstrando que os tratamentos não induziram cio. Para se determinar se o tratamento com BE alterou a sensibilidade endometrial à ocitocina e, assim, a cascata luteolítica, vacas multíparas pré-sincronizadas receberam 5mg de BE, seguidos, após 6 horas, de 50 UI de ocitocina (OT; n=9). Oito horas após a administração de BE, colheram-se fragmentos endometriais das vacas, nos dias 4, 13 e 17 do ciclo estral, mensurando-se a expressão gênica de COX-2 através de PCR. O BE aumentou os níveis de RNAm de COX-2 apenas no dia 17 do ciclo estral (P<0,05). Em conclusão, o MAP isolado ou associado a BE é capaz de suprimir o crescimento folicular bovino. Entretanto, o BE, na presença de MAP é ineficaz na indução da luteólise bovina, quando injetado no oitavo dia do ciclo estral.

E-mail: savasco@usp.br Risk factors associated with leptospirosis were identified in cows in the state of Bahia, northeastern Brazil. A total of 10,823 cows with > 24 months of age from 1,414 herds were randomly sampled. For the serological diagnosis of Leptospira spp. infection, the microscopic agglutination test (MAT) was carried out using 24 serovars as antigens. A herd was considered positive when presented at least one seropositive animal. Of the 1,414 investigated herds, 1,076 (77.9%; 95% CI = 75.7-80.0%) presented at least one reactant animal at MAT to any serovar. Serovar Hardjo (Hardjoprajitno) was the most prevalent, with 34.49% (95% CI = 31.97-37.14%) of the positive herds. Presence of more than 28 bovine females in reproductive age (OR=2.11; p<0.001), presence of cervids (OR=2.02; p=0.010), animal purchase (OR=1.57; p<0.001), to slaughter animals in the property (OR=1.58; p=0.030) and to share pasture (OR=1.63; p<0.001) were identified as risk factors for leptospirosis due to any serovar. Risk factors for leptospirosis due to serovar Hardjo (Hardjoprajitno) were presence of swine (OR=1.28; p=0.040) and animal purchase (OR=1.48; p<0.001).

Das 1.414 propriedades investigadas, 1.076 (77,9%; IC 95% = 75,7-80,0%) apresentaram pelo menos um animal reagente na SAM para qualquer sorovar. O sorovar Hardjo (Hardjoprajitno) foi o mais prevalente, com 34,49% (IC 95% = 31,97-37,14%) das propriedades positivas. Presença de mais de 28 fêmeas bovinas em idade reprodutiva no rebanho (OR=2,11; p<0,001), presença de cervídeos (OR=2,02; p=0,010), compra de animais (OR=1,57; p<0,001), abate de animais na própria fazenda (OR=1,58; p=0,030) e utilização de partos compartilhados (OR=1,63; p<0,001) foram identificados como fatores de risco para leptospirose por qualquer sorovar. Pesquisa Veterinária Brasileira 30(4):340-344. Departamento de Medicina Animal, Universidade Federal do Rio Grande do Sul, Avenida Bento Gonçalves 8824, Porto Alegre, RS 91540-000, Brazil. E-mail: lucasbmoraes@gmail.com Fifty Bursa of Fabricius (BF) were examined by conventional optical microscopy and digital images were acquired and processed using Matlab® 6.5 software. The Artificial Neuronal Network (ANN) was generated using Neuroshell® Classifier software and the optical and digital data were compared. The ANN was able to make a comparable classification of digital and optical scores. The use of ANN was able to classify correctly the majority of the follicles, reaching sensibility and specificity of 89% and 96%, respectively. When the follicles were scored and grouped in a binary fashion the sensibility increased to 90% and obtained the maximum value for the specificity of 92%. These results demonstrate that the use of digital image analysis and ANN is a useful tool for the pathological classification of the BF lymphoid depletion. In addition it provides objective results that allow measuring the dimension of the error in the diagnosis and classification therefore making comparison between databases feasible.

E-mail: lucasbmoraes@gmail.com Cinquenta Bursa de Fabrícius (BF) foram examinadas através de microscopia óptica convencional e imagens digitais foram obtidas e processadas através do software Matlab® 6.5. Redes Neurais Artificiais (ANN) foram geradas com a utilização do software Neuroshell® Classifier, e os dados das análises óptica e digital foram comparados. A ANN classificou corretamente a maioria dos folículos, atingindo sensibilidade e especificidade de 89% e 96%, respectivamente. Quando os folículos foram agrupados de forma binária houve um aumento da sensibilidade para 90% e obteve-se um valor máximo para a especificidade de 92%. Estes resultados demonstram que o uso da análise digital de imagem associada à ANN é uma ferramenta bastante útil para a classificação patológica da depleção linfóide da BF. Além disso, fornece resultados objetivos que permitem medir a dimensão do erro classificatório, tornando possível a comparação entre distintos bancos de dados.

[Anthelmintic resistance of gastrointestinal nematodes in sheep, Mato Grosso do Sul, Brazil.] Resistência anti-helmíntica de nematóides gastrintestinais em ovinos, Mato Grosso do Sul. Pesquisa Veterinária Brasileira 30(3):229-236. Departamento de Patologia, Centro de Ciências Biológicas e da Saúde, Universidade Federal de Mato Grosso do Sul, Cidade Universitária, Campo Grande, MS 79070-900, Brazil. E-mail: eurico_sczesny@hotmail.com Entre os métodos de controle da verminose gastrintestinal em ovinos, a utilização de produtos químicos é o mais empregado. Porém, o uso indiscriminado e continuado desses produtos tem selecionado populações de helmintos resistentes aos anti-helmínticos, fenômeno relatado no mundo todo. Este trabalho teve como objetivo identificar as espécies de parasitos gastrintestinais e diagnosticar a situação da resistência anti-helmíntica em ovinos no Estado de Mato Grosso do Sul. Foram realizados testes de redução na contagem de ovos por grama de fezes (OPG) em rebanhos de dezesseis propriedades rurais; as sete formulações utilizadas continham as seguintes bases farmacológicas: Albendazol, Ivermectina, Levamisole, Triclorfon, Moxidectina, Closantel e uma contendo as três primeiras associadas. As espécies identificadas nas necropsias, em ovinos adultos, foram: Haemonchus contortus, Trichostrongylus colubriformis, Cooperia curticei, C. punctata, C. pectinata e Oesophagostomum columbianum; em ordem de prevalência. As formulações contendo Albendazol e Ivermectina não apresentaram eficácia na redução de OPG nos rebanhos testados, com médias de redução de 0,7 e -19,6%, respectivamente. Closantel apresentou eficácia média de 6,7%; Levamisole, Moxidectina e Triclorfon de 28,7, 26,8 e 65%, respectivamente; a associação das três bases (Albendazol, Ivermectina e Levamisole), uma média de eficácia de 55,8%. As percentagens médias de larvas infectantes recuperadas nas coproculturas, tanto no pré como no pós-tratamento, foram semelhantes; indicando que a resistência às bases testadas está presente em todas as espécies citadas, em maior ou menor intensidade. Os dois gêneros predominantemente resistentes são Haemonchus sp., com 86,9%; seguido por Trichostrongylus sp., com média de 47,5%; Strongyloides sp. 33,6%; Oesophagostomum, sp. 21,4% e Cooperia sp. 19,7%.

E-mail: eurico_sczesny@hotmail.com Among the methods of control of gastrointestinal worms in sheep, the use of chemicals is the most common. However, the continued, and indiscriminate, use of these products has selected populations of resistant helminths to anthelmintics, a phenomenon reported in the whole world. This study aimed to identify the species of gastrointestinal parasites and diagnose the status of anthelmintic resistance in sheep in the State of Mato Grosso do Sul Brazil. Feacal egg count reduction tests (FECRT) were performed in flocks of sixteen farms, and the seven formulations used contained the following pharmacological bases: Albendazole, Ivermectin, Levamizol, Trichlorfon, Moxidectin, Closantel and one containing the first three in association. The species identified at necropsy, in adult sheep, were: Haemonchus contortus, Trichostrongylus colubriformis, Cooperia curticei, C. punctata, C. pectinata and Oesophagostomum columbianum, in order of prevalence. The formulations containing Albendazole and Ivermectin did not show efficacy in reducing the EPG in the flocks tested, with average reductions of 0.7 and -19.6%, respectively. Closantel presented an average efficacy of 6.7%; Levamisolee, Moxidectin and Trichlorfon, 28.7, 26.8 and 65% respectively, the combination of three bases (Albendazole, Ivermectin and Levamizol), an average efficacy of 55.8 %. The average percentages of infective larvae recovered in the faecal cultures, pre and post treatment were similar, indicating that resistance to the bases tested is present in all species cited, to a greater or lesser degree. The two genera predominantly resistant are Haemonchus sp., with 86.9%, followed by Trichostrongylus sp., with an average of 47.5%, Strongyloides sp. 33.6%, Oesophagostomum sp. 21.4% and Cooperia sp. 19.7%.

E-mail: cassianabreu@gmail.com Mazama nana is the least known of Brazilian deer species. The male reproductive parameters are still unknown, but apparently they did not show reproductive seasonality. In this work were used nine males of M. nana in reproductive age, kept in captivity. They were assessed for weight, height of wither, crown-rump length, the situation of horns, testicular volume and consistency. The sperm was collected by electro-ejaculation and subjected to analyses of motility, vigor and morphology. The correlation between age, weight, height of wither, crown-rump length, testicular volume and characteristics of the ejaculate (volume, motility, vigor and sperm concentration) were evaluated by the procedure Corr., of SAS®. The mean ± standard deviation observed for weight (kg), crown-rump length (cm) and height of wither (cm) were: 15.72±1.98, 74.9±3.05, and 48.5±2.06 respectively. For primary reproductive parameters were observed: volume of ejaculate (91.46±68.24ìl); motility (70±8.16%); vigor (3.0±0.67); sperm concentration (1536x106 ±351x106 spermatozoa per ml). For sperm morphology was found a high percentage of abnormal cells (40.90%), with predominant defects in the tail (25.95%).

Os parâmetros reprodutivos para os machos ainda são desconhecidos, mas parece que não apresentam sazonalidade reprodutiva. Neste trabalho foram utilizados nove machos de Mazama nana em idade reprodutiva, mantidos em cativeiro. Foram avaliados quanto ao peso corporal, altura de cernelha, comprimento crânio-caudal, situação dos chifres, volume e consistência testicular. O sêmen foi colhido por eletroejaculação e submetido a análises de motilidade, vigor e morfologia espermática. As correlações entre idade, peso, comprimento crânio-caudal, altura de cernelha, volume testicular e características do ejaculado (volume, motilidade, vigor e concentração do sêmen) foram avaliadas pelo procedimento Corr., do SAS®. As médias ± desvio padrão observados para peso (kg), comprimento crânio-caudal (cm) e altura de cernelha (cm) foram: 15,72±1,98, 74,9±3,05 e 48,5±2,06, respectivamente. Em relação aos parâmetros reprodutivos primários foram observados: volume do ejaculado (91,46±68,24µl); motilidade (70±8,16%); vigor (3,0±0,67); concentração espermática (1536x106 ±351x106 espermatozóides por ml). Em relação à morfologia espermática, foi observada uma alta porcentagem de células anormais (40,90%), sendo predominante os defeitos de cauda (25,95%).

E-mail: cassivetr@hotmail.com As formas de imobilização em bovinos mais utilizadas levam a uma manipulação excessiva e consequentemente estressante para o animal. A dor é uma experiência emocional de sensação desagradável que vem associada a um prejuízo tecidual. Surge no mercado um método de contenção física para bovinos que propõe a utilização de um aparelho que funciona com ondas eletromagnéticas de baixa intensidade promovendo imobilização sem estresse e ou estímulo doloroso para o animal. O objetivo deste projeto foi avaliar os parâmetros cardiorrespiratórios e hemogasométricos bem como os níveis de cortisol de bovinos submetidos à imobilização eletromagnética, a fim de observar se tal método é eficaz na diminuição do estresse e/ou estímulo doloroso. Foram utilizados 6 bovinos, mestiços, peso médio de 300,3 kg (±85,76). Os animais foram alocados aleatoriamente em dois grupos (controle dele mesmo): grupo controle (GC), introdução da sonda via retal (desligada); grupo Imobilizador (GI) introdução da sonda e permanência desta ligada por 5 minutos. Análise estatística utilizou o Teste T pareado entre os grupos (P<0,05) e a avaliação entre tempos dentro de cada grupo análise de variância de uma única via com repetições múltiplas (ANOVA - RM). Momentos avaliados M0: basal; M1: imediatamente após a retirada da sonda; M2 e M3 (30 e 60 minutos após M1 respectivamente). Houve um aumento significativo nas variáveis: frequências cardíaca e respiratória em (M1), na Pressão arterial média (M1, M2, M3) e níveis de cortisol nos momentos M1 e M2 no grupo imobilizador quando comparado ao grupo controle. Com relação à resposta ao estímulo, o GI teve resposta negativa no momento da estimulação, comparado ao GC (resposta máxima). De acordo com os resultados obtidos, podemos concluir que o imobilizador eletromagnético é um método, exclusivamente, de contenção física, não oferecendo nenhum suporte anestésico, para o conforto ou bem estar ao animal.

The pain is an emotional experience and unpleasant sensation in combination with tissue damage. The new method of physical restraint for cattle uses low intensity electromagnetic waves, promising immobilization without stress or painful stimulation for the animals. The aim of this study is to evaluate of cardiorespiratory effects and plasmatic cortisol concentration of cattle submitted to electromagnetic immobilization. Six healthy cows with average weight of 300.3 ± 85.76 kg (mean ± SD) were assigned to two treatments in a randomized design with a 15-day interval between experiments GC (control group) and GI (immobilization group). Data are presented as mean + SD. The statistical analyses used the t-Test between groups, and ANOVA random block design between times into same group. All statistical analyses were performed using P<0.05 as a significant level. Evaluation moments: M1 (basal); M1: after withdrawal probe, M2 and M3 (30 and 60 minutes after M1 respectively). There was a significant increase of FC and f in M1, PAM in the moments (M1, M2, M3) and in the levels of plasmatic cortisol concentration in M1 e M2 in the immobilization group when to compared with the control group. It could be concluded that this electromagnetic immobilization is a method of physic restraint only, offers no anesthetic support and favors comfort and animal welfare.

Blood samples were collected and processed in accordance with immunocytochemistry standard technique using lymphoid markers species-specific, as anti-CD3 (MM1A - BoCD3) and anti-LB (LCTB16A-clone B-B14), for T and B lymphocyte counting, respectively. The data were submitted to descriptive statistics, and to Kruskall Wallis and Mann-Whitney tests. The results showed decreased levels of leukocytes, lymphocytes, T lymphocytes and B lymphocytes along the age advance. Absolute values of leukocytes, lymphocytes and T lymphocytes were higher in males than females. None of the evaluated parameters were affected by differences of the management carried out at the two farms.

Hepatic fibrosis is characterized by a disruption of normal tissue architeture by cellular lesions, and may alter the GJIC. This work aimed to study the expression and distribution of Cx32 in liver fibrosis induced by the oral administration of dimethylnitrosamine in female Wistar rats. The necropsy of the rats was carried out after five weeks of drug administration. They presented a hepatic fibrosis state. Sections from livers with fibrosis and from control livers were submitted to immunohistochemical, Real Time-PCR and Western-Blot analysis to Cx32. In fibrotic livers the Cxs were diffusely scattered in the cytoplasm, contrasting with the control livers, where the Cx32 formed junction plaques at the cell membrane. Also it was found a decrease in the gene expression of Cx32 without reduction in the protein quantity when compared with controls. These results suggest that there the mechanism of intercellular communication between hepatocytes was reduced by the fibrotic process, which may predispose to the occurrence of a neoplastic process, taken in account that connexins are considered tumor suppressing genes.

This article describes the construction of a recombinant plasmid expressing the PCV2 structural protein and the evaluation of cellular and humoral immune responses produced by this recombinant vaccine in BALB/c mice. The vaccine candidate was obtained and analyzed in vivo, in an effort to determine the ability to induce a specific immune response in mice. DNA was extracted from a Brazilian PCV2 isolate and the gene coding for Cap protein was amplified by PCR and inserted into an expression plasmid. Groups of BALB/c mice were inoculated intra-muscularly and intradermally in a 15-day interval, with 100 µg and 50 µg of the vaccine construct, respectively. Another group was inoculated intramuscularly with 100 µg of empty plasmid, corresponding to the control group. Seroconversion and cellular response in BALB/c mice were compared and used for vaccine evaluation. Seroconversion was analyzed by ELISA. After a series of 3 immunizations the spleen cells of the immunized animals were used to perform lymphocyte proliferation assays. Seroconversion to PCV2 was detected by ELISA in the animals inoculated with the vaccine construct when compared with control groups. Lymphocyte proliferation assays showed a stronger cell proliferation in the inoculated animals compared with the control group. Thus, the vaccine candidate construct demonstrated to be able to induce both humoral and cellular responses in inoculated mice.
