E-mail: marilene@ufrrj.br Metternichia princeps, a tree that belongs to the Solanaceae family, occurs in the Atlantic Forest of Brazil, from the state of Rio de Janeiro to Bahia. In the county of Itaguaí, state of Rio de Janeiro, M. princeps was suspected to cause a lethal kidney disease in goats. The poisoning was experimentally reproduced in goats confirming the suspicion. Experiments were then performed in rabbits. Leaves of M. princeps were collected in the county of Itaguaí, where the poisoning in goats occurred. Eleven rabbits received the dried plant material, ten received the fresh plant and one rabbit served as control. The dehydrated leaves were ground and mixed with water. Initially the suspension was administered by stomach tube to nine rabbits and caused death of seven of them at doses from 0.125g/kg on. Two rabbits that got sick, but survived at doses of 0.0625g/kg and 0.125g/kg, received doses of 0.5 and 0.25g/kg respectively, two months later and died, indicating that no tolerance to the plant poisoning effects developed. Additionally the dried leaves administered six months after collection to two rabbits at doses of 0.5 and 1g/kg respectively, caused death only of the rabbit that received the dose of 1g/kg, indicating that the stored leaves lost toxicity. Fresh sprouts caused death in three of six rabbits at doses of 1.55g/kg to 3g/kg. Fresh mature leaves caused death in one of four rabbits at the dose of 2g/kg, what suggests that there is no difference in toxicity between mature leaves and sprouts. The dried leaves caused death of rabbits at smaller doses than fresh leaves, what can be explained by the way of administration of the plant; dried leaves were given within a few minutes, whilst fresh leaves were ingested during one or two days, a fact that may influence absorption of the toxic principle. In all experiments with the dried and fresh leaves (sprouts and mature leaves) of M. princeps, liver and heart lesions predominated. The course of the poisoning was peracute to acute. The rabbits became apathetic with pale and cyanotic mucosae and cold ears; they fell down with struggling movements, vocalization and spaced breathing and died. At postmortem examination, the liver was pale with marked hepatic lobules; the other organs were congested and blood vessels were ingurgitated. The histopathological examination revealed as the more important changes in the liver, intense, especially centrilobular congestion, severe swelling of the hepatocytes, and foci of incipient necrosis of hepatocytes. In the heart, beside congestion, small groups of eosiophilic cardiac fibers, with nuclear picnosis and vacuolation were found. In the kidney, congestion and swelling of epithelial cells was seen. The performed experiments show that rabbits are susceptible to poisoning with dried and fresh leaves of M. princeps. It is suggested that the toxic principle of M. princeps, responsible for the clinical and pathological picture in rabbits, may not be the same compound that causes poisoning in goats or that the rabbit reacts differently as react goats; rabbits showed liver and heart lesions whilst goats had kidney lesions.

É conhecida popularmente com os nomes de “café-do-mato”, “trombeteira” e “jasmin-do-morro”. Em um estabelecimento no município de Itaguaí, RJ, suspeitou-se que M. princeps era responsável por doença renal letal em caprinos. A intoxicação foi reproduzida experimentalmente nessa espécie, confirmando a suspeita. Foram então realizados experimentos em coelhos. Folhas de M. princeps foram coletadas no município de Itaguaí/RJ, local onde foi diagnosticada a intoxicação em caprinos. Onze coelhos receberam a planta dessecada, dez receberam a planta fresca e um coelho serviu como controle. As folhas dessecadas de M. princeps foram trituradas e misturadas com água. Inicialmente, a suspensão foi administrada por via intragástrica através de sonda a nove coelhos e causou a morte de sete coelhos nas doses a partir de 0,125g/kg. Dois coelhos que adoeceram mas sobreviveram às doses de 0,0625 e de 0,125g/kg, dois meses mais tarde receberam doses de 0,5 e 0,25g/kg, respectivamente, e morreram, o que indica que não houve desenvolvimento de tolerância à toxidez da planta. Adicionalmente, a planta dessecada, administrada a dois coelhos nas doses de 0,5 e 1g/kg seis meses após a coleta, causou a morte somente do coelho que recebeu a dose de 1g/kg, indicando que a planta armazenada perde em toxidez. Os brotos frescos causaram a morte de três dos seis coelhos, nas doses de 1,55g/kg a 3g/kg. As folhas maduras frescas causaram a morte de um dos quatro coelhos, na dose de 2g/kg, o que sugere que não há diferença na toxidez entre a brotação e as folhas maduras. As folhas dessecadas causaram a morte dos coelhos em doses menores que as folhas frescas, o que talvez possa ser explicado pelo modo de administração da planta; as folhas dessecadas eram administradas em poucos minutos, enquanto que as folhas frescas eram ingeridas durante um a dois dias, o que influenciaria a rapidez da absorção do princípio tóxico da planta. Tanto nos experimentos com as folhas dessecadas quanto com as folhas frescas (brotos e folhas maduras), predominaram no quadro clínico-patológico alterações relativas ao coração e ao fígado. A evolução da intoxicação foi superaguda a aguda. Os coelhos tornaram-se apáticos, com mucosas pálidas a cianóticas e orelhas frias, até que subitamente se debatiam na gaiola, faziam movimentos de pedalagem, vocalizavam, apresentavam acentuada dispneia e morriam. À necropsia, o fígado apresentava-se com evidenciação da lobulação; os demais órgãos estavam congestos e os vasos sanguíneos ingurgitados. À histopatologia, as lesões mais relevantes foram: no fígado, intensa congestão, especialmente centrolobular, tumefação acentuada dos hepatócitos e focos de necrose incipiente de hepatócitos; no coração, além de congestão, fibras cardíacas com aumento de eosinofilia, com picnose nuclear, fibras cardíacas com vacuolização, e no rim, congestão e tumefação de células epiteliais. Esses experimentos demonstram que o coelho é sensível à intoxicação pelas folhas dessecadas e frescas de M. princeps. Sugere-se que o princípio tóxico responsável pelo quadro clínico-patológico da intoxicação por M. princeps em coelhos não seja o mesmo princípio responsável pela intoxicação nos caprinos, ou que os coelhos reagem de maneira diferente dos caprinos; nos coelhos predominam alterações cardíacas e hepáticas enquanto que em caprinos as lesões são renais.

E-mail: julianapradomedvet@gmail.com From 2007 to 2009 a nephrotoxic disease of subacute course and high mortality in goats occurred on a farm in the county of Itaguaí, Rio de Janeiro. Metternichia princeps, a plant of the Solanaceae family, was suspected to be the cause. Through experiments in goats the clinical-pathological picture of poisoning by the plant and the lethal dose were established. For the experiments 12 young and adult goats of different races, both sexes and with weights above 15 kg were used. The goats that received 30g/kg in 5 days, 15g/kg in 3 days, single doses of 10g/kg and 5g/kg, all died. Of two goats that received single doses of 2.5g/kg, one died but the other did not show any clinical sign; a goat that received the single dose of 1.25g/kg also did not show any symptom of poisoning. First clinical signs were observed from 7 hours to 46h45min after the plant intake. The clinical course varied from 3h6min to 126h40min. First clinical signs were loss of appetite, adipsia, apathy and reluctance to move. After the animals entered in sternal decubitus and when they were placed in standing position, they kept their front limbs flexed, supported only by the posterior limbs on the floor until evolving into flexion of all four limbs followed by lateral decubitus. Postmortem examination revealed edema of the perirenal adipose tissue, pale kidneys which on the cut-surface showed whitish stripes from the cortex to the medullar area. At histopathology, severe coagulative necrosis of epithelial cells of the kidney tubules was seen. Compared with the natural cases, the goats experimentally poisoned by M. princeps, presented a similar clinic-pathological picture. By these experiments it could be proved that Metternichia princeps is the toxic plant that causes a nephrotoxic disease in goats in the Rio de Janeiro area; the minor lethal dose was 2,5g/kg in the experiments.

E-mail: julianapradomedvet@gmail.com Entre os anos de 2007 e 2009 ocorreu uma doença nefrotóxica de evolução subaguda com alta mortandade em caprinos em uma propriedade no município de Itaguaí, estado do Rio de Janeiro. Levantou-se a suspeita de que Metternichia princeps, planta pertencente à família Solanaceae, seria a causa. Através de experimentação em caprinos o quadro clínico-patológico de intoxicação por esta planta e a dose letal foram estabelecidos. Na experimentação foram utilizados 12 caprinos de diferentes raças, de ambos os sexos, jovens a adultos, com pesos acima de 15 kg. Os animais que receberam as doses de 30g/kg em 5 dias, 15g/kg em 3 dias, doses únicas de 10g/kg e de 5g/kg, morreram. Dos três animais que receberam as doses únicas de 2,5g/kg, dois morreram e um não apresentou sinais clínicos e o animal que recebeu a dose única de 1,25g/kg, também não apresentou sinais clínicos. O início dos sinais clínicos após a administração da planta variou entre 7h e 46h45min. A evolução variou entre 3h6min e 126h40min. Os primeiros sinais clínicos apresentados foram inapetência, adipsia, apatia e relutância ao movimento. Em seguida os animais entravam em decúbito esternal e ao serem colocados em estação, mantinham os membros anteriores flexionados, apoiavam apenas os posteriores no chão até evoluírem para flexão dos quatro membros e seguia-se o decúbito lateral. À necropsia destacaram-se o edema de tecido adiposo perirrenal, rins pálidos e, ao corte, com estriação esbranquiçada desde o córtex até a região medular. À histopatologia foi verificada acentuada necrose coagulativa das células epiteliais dos túbulos uriníferos. Comparativamente aos casos naturais, os caprinos intoxicados experimentalmente por M. princeps apresentaram quadro clínico-patológico semelhante. Desta maneira foi comprovado que Metternichia princeps é responsável pela doença nefrotóxica em caprinos no Rio de Janeiro; a menor dose que causou a morte dos caprinos nos experimentos foi 2,5g/kg.

E-mail: fabicientista@gmail.com Brucella spp. are intracellular facultative gram-negative bacteria which are pathogenic for many species of mammals, causing brucellosis, a worldwide spread zoonosis. Therefore the search for more efficient alternatives of control, as the development of new potential immunogens is necessary. In this study, we knockouted virB10 from Brucella abortus S2308 strain, generating a mutant strain probably incapable to produce the corresponding native protein. The gene virB10 is part of an operon that codifies for type IV secretion system, which is essential for the intracellular survival and multiplication of the bacteria in host cells. The knockout was carried through by the construction of the suicidal plasmid pBlue: virB10: kan and eletroporation in eletrocompetent cells of B. abortus S2308, leading to the exchange of the wild gene for the interrupted gene, containing the gene of resistance to kanamycin, for double homologous recombination. BALB/c mice were inoculated with S19, RB-51, DvirB10 strains of B. abortus and S2308 wild strain; the results demonstrated that the BALB/c mice inoculated with S19 and BALB/c mice inoculated with S2308 presented faster fall of trend line, when compared with the too much groups, for bacterial recovery (BR) and esplenic weight (EW) respectively. The groups that received DvirB10 S2308 B. abortus and RB-51 demonstrated similar behavior for both the characteristics. In the sixth week postinoculation, the results for BR (log UFC ± standart deviations) and EW (esplenic weight ± standart deviations), respectively, showed: groups inoculated with strains S2308 (4,44±1,97 and 0,44±0,11), S19 (1,83±2,54 and 0,31±0,04), RB-51 (0,00±0,00 and 0,20±0,01) and DvirB10 S2308 (1,43±1,25 and 0,19±0,03). Considered the bacterial clearance, all the groups differed statistical from the group that received S2308 (p<0,0001), the group inoculated with DvirB10 S2308 B. abortus was similar to the S19 group (p=0,4302) and different of group RB-51 (p=0,0063). The evaluation of the persistence of the strains showed that virB10 is essential for the maintenance of the virulence. These results support other studies concerning the immunogenic potential of this mutant strain.

Por isso a busca de alternativas de controle mais eficientes se faz necessário como o desenvolvimento de novas cepas que possam ser testadas como potenciais imunógenos. Neste estudo realizou-se a deleção do gene virB10 da cepa S2308 de Brucella abortus gerando uma cepa knockout provavelmente incapaz de produzir a proteína nativa correspondente. O gene virB10 faz parte de um operon que codifica para um sistema de secreção do tipo IV, essencial para a sobrevivência intracelular e multiplicação da bactéria em células hospedeiras. A deleção foi realizada pela construção do plasmídeo suicida pBlue:virB10:kan e eletroporação deste em células eletrocompetentes de B. abortus S2308, ocorrendo a troca do gene selvagem pelo gene interrompido, com o gene de resistência a canamicina, por recombinação homóloga dupla. Camundongos BALB/c foram inoculados com as cepas S19, RB-51, DvirB10 de B. abortus e B. abortus S2308 selvagem; os resultados demonstraram que camundongos BALB/c inoculados com S19 e camundongos BALB/c inoculados com S2308 apresentaram queda mais rápida de linha de tendência, quando comparadas aos demais grupos, para recuperação bacteriana (RB) e peso esplênico (PE) respectivamente. Os grupos que receberam DvirB10 S2308 de B. abortus e RB-51 demonstraram comportamento semelhante para ambas as características. Na sexta semana após a inoculação, os resultados para RB (log de UFC ± desvio padrão) e PE (peso esplênico ± desvio padrão), respectivamente, mostraram: grupos inoculados com as cepas S2308 (4,44±1,97 e 0,44±0,11), S19 (1,83±2,54 e 0,31±0,04), RB-51 (0,00±0,00 e 0,20±0,01) e DvirB10 S2308 (1,43±1,25 e 0,19±0,03). Considerado o clearance bacteriano, todos os grupos diferiram estatisticamente do grupo que recebeu S2308 (p<0,0001), o grupo inoculado com DvirB10 S2308 de B. abortus foi semelhante ao grupo S19 (p=0,4302) e diferente do grupo RB-51 (p=0,0063). A avaliação da persistência revelou que o gene virB10 é essencial para a manutenção da virulência da bactéria. Os resultados obtidos possibilitarão que outras pesquisas sejam realizadas avaliando o potencial imunogênico desta cepa mutante.

The myenteric plexus arrangement and the number of neurons were comparatively evaluated among the different portions of the cecum. The neurons from the apical and basal regions were distributed in classes at intervals of 100µm2, the means of the corresponding intervals being compared. The ganglia, in both techniques, were often connected by fine bundles, which became thicker in the mesenteric region and in the region next to the cecal ampulla. The number of positive NADH-d neurons was higher than that of NADPH-d neurons in all portions, from both regions. The numbers of reactive NADH-d e NADPH-d neurons were significantly different among the different portions of the cecum, except for the antimesenteric basal and intermediate basal regions, considering the NADH-d neurons. The profile area for the reactive NADH-d e NADPH-d neurons was higher in the apical region than in the basal area. Differences in arrangement, distribution and size of positive NADH-d e NADPH-d neurons in the different cecum portions evidenced the importance of the subdivision of the analyzed organ.

[Arterial vascularization of the sinoatrial node in swine hearts: origin, distribution and quantification.] Vascularização arterial da região do nó sinoatrial em corações suínos: origem, distribuição e quantificação. Pesquisa Veterinária Brasileira 28(2):113-118. E-mail: avidotti@usp.br The sinoatrial node, for being topographically installed as the initial component of the conduction system, is responsible for the production of the nervous impulses, which determines the cardiac contraction. There have been made studies related to the node’s morphology in order to know the origin, distribution and quantification of the vases in this tissue, however, in spite of the results and quantitative data of the nodal irrigation - arterial vascular conduct and arterial vascular density, at the nodal level - the literature is scarce. With this objective 27 SRD swine hearts, injected with colored resin for macroscopic analysis of the origin and distribution of ANSA (sinoatrial node artery), 3 others injected with watery solution of colloidal coal (dyed nanquim) were used, to mark the route of the vases on the node level for stereological analysis. The atrial arteries originated as well from the right coronary artery as from the left one, with predominance of the first (66.66% and 33.33%, respectively). When originated from the right coronary artery, there existed the following branches: AADAM (right cranial medial atrial artery) in 14 cases, AADAI (right cranial intermedial atrial artery) in 2 cases, and AADAL (right cranial lateral atrial artery) in 2 cases. In 9 cases (33.33%) the following branches originated from the left coronary artery: 4 through branch AASPL (left caudal lateral atrial artery), 2 through branch AASAI (left cranial intermedial atrial artery) and 3 through branch AASAM (left cranial medial atrial artery). Interarterial anastomoses with vessels responsible for irrigation of the sinoatrial node area were observed in most of the cases (25 hearts). The Volume or Reference Volume (V(ref)) of the hearts were 35.32x104µm3. For the stereological variables analyzed, the estimate density of the vessel length (Lv) was 766; the vessel length (L) - mm - was 27.06x105µm; the area’s surface density (Sv) was 182 and the area’s surface (S) - mm2 - was 64.3x106µm2. The estimate of the vascular numeric density (Nv(vasc)) - total of vessels per unit of volume (cm3) was 2.19 10-5, and the total number of vessels in the organ (N(vasc)), estimated by the physical dissector method in combination with the estimate of the number of Euler (Xv), was 773.6832 x10-2. The raised vascular density and total number of vases in the swine’s sinoatrial node area suggests the existence of a vascular perinodal network complex and dense, ratifying the importance of this mark-pass for its blood supply.

E-mail: avidotti@usp.br The sinoatrial node, for being topographically installed as the initial component of the conduction system, is responsible for the production of the nervous impulses, which determines the cardiac contraction. There have been made studies related to the node’s morphology in order to know the origin, distribution and quantification of the vases in this tissue, however, in spite of the results and quantitative data of the nodal irrigation - arterial vascular conduct and arterial vascular density, at the nodal level - the literature is scarce. With this objective 27 SRD swine hearts, injected with colored resin for macroscopic analysis of the origin and distribution of ANSA (sinoatrial node artery), 3 others injected with watery solution of colloidal coal (dyed nanquim) were used, to mark the route of the vases on the node level for stereological analysis. The atrial arteries originated as well from the right coronary artery as from the left one, with predominance of the first (66.66% and 33.33%, respectively). When originated from the right coronary artery, there existed the following branches: AADAM (right cranial medial atrial artery) in 14 cases, AADAI (right cranial intermedial atrial artery) in 2 cases, and AADAL (right cranial lateral atrial artery) in 2 cases. In 9 cases (33.33%) the following branches originated from the left coronary artery: 4 through branch AASPL (left caudal lateral atrial artery), 2 through branch AASAI (left cranial intermedial atrial artery) and 3 through branch AASAM (left cranial medial atrial artery). Interarterial anastomoses with vessels responsible for irrigation of the sinoatrial node area were observed in most of the cases (25 hearts). The Volume or Reference Volume (V(ref)) of the hearts were 35.32x104µm3. For the stereological variables analyzed, the estimate density of the vessel length (Lv) was 766; the vessel length (L) - mm - was 27.06x105µm; the area’s surface density (Sv) was 182 and the area’s surface (S) - mm2 - was 64.3x106µm2. The estimate of the vascular numeric density (Nv(vasc)) - total of vessels per unit of volume (cm3) was 2.19 10-5, and the total number of vessels in the organ (N(vasc)), estimated by the physical dissector method in combination with the estimate of the number of Euler (Xv), was 773.6832 x10-2. The raised vascular density and total number of vases in the swine’s sinoatrial node area suggests the existence of a vascular perinodal network complex and dense, ratifying the importance of this mark-pass for its blood supply.

E-mail: ceambrosio@usp.br Fifteen papillae from female water buffaloes and one bovine female were obtained in a slaughter house for this study, which consisted in morphologic and biometrical analyses, histology and histometric and mesoscopical analyses. The means were: right cranial papilla (RCP) 20.95g; right caudal papilla (RCaP) 21.49g; left cranial papilla (LCP) 23.52g; and left caudal papilla (LCaP) 21.35g. Papillary duct average length, numbers of folds and papillary cisterns length were, respectively: 0.67/5,5 e 2.17cm for RCP; 0.62/6.0 e 2.26cm for RCaP; and for the left antimere: RCP: 0.71/6.0 e 2.26cm and for RCaP: 0.74/5.7 e 2.57cm. Pearson’s analysis revealed negative correlation between body weight and mammary glandular weight. Sampling of proximal, middle and distal area was taken and fixed into a 10% formaldehyde solution, followed by mesoscopical investigation through 10, 20 and 40µm thickness cuts stained by Azan, F3BA Picrosirus and Weigert-Van Gieson techniques. Histological and morphometric analyses were applied on 5µm thick samples and showed that the muscular layer was disposed into two arrangements, internal circular and external longitudinal with a bi-stratified cuboidal epithelium. A well developed muscular layer were related to the middle portion revealing thickness of 22.90mm with a great amount of connective tissue (7.10mm).

E-mail: gustalima@hotmail.com Many studies about Haematobia irritans have been made due to the damages and worries it causes to cattle breeding in many countries. The common name of the fly is related to its behavior. 1111 Europe and the United States of America it is named horn-fly because it stays right on the base of the horns in order to protect itself from low temperatures; in Latin America, however, it is known as "mosca-da-paleta" because of its preferred localization on the shoulder of cattle. Biological aspects of the bovines can lead to different levels of fly infestation in the herd. Mean numbers of flies on different anatomic regions and individual infestations were evaluated in 60 Nelore bulls with the sarne zootechnic features during 1998 in Araçatuba city, State of São Paulo, Brazil. Both sides of the animals were filmed, and the number of flies was recorded in cassette tapes. Fly counts and their delimitation on 15 anatomic regions were performed from film viewing. During rainy days a significant increase (p<0.05) in number of flies in the abdominal region was observed. Shoulders, forelegs, back and ribs showed the highest number of flies (p<0.05). The evaluation of individual infestation revealed less than 50 flies on 50% of the cattle, 50 to 100 flies on 38% and over 100 flies on 12% of them.

Seu nome está relacionado com o local de permanência nos bovinos. É conhecida como "horn fly" (mosca-dos-chifres) na Europa e nos Estados Unidos da América e mosca-da-paleta na América Latina. Os fatores biológicos podem produzir em bovinos de um único rebanho, diferentes níveis de infestação da mosca. Durante o ano de 1998 em Araçatuba, estado de São Paulo, foram avaliados o número médio de mosca por região anatômica; bem como os diferentes níveis de infestação em 60 bovinos da raça Nelore. Os bovinos foram filmados de ambos os lados do corpo para registrar o número de mosca em fitas cassetes. As fitas foram assistidas para a contagem e demarcação da mosca em 15 regiões anatômicas. O maior número de mosca (p<0,05) foi observado nas regiões escapular, interescapular e costal; nos períodos chuvosos observou-se um aumento significativo (p<0,05) na região ventral. As avaliações individuais, demonstraram infestação com menos de 50 moscas em 50% dos bovinos, 50 a 100 moscas em 38% e acima de 100 moscas em 12% dos bovinos.
