E-mail: claudialautert@yahoo.com.br Mycotoxins are a group of chemically diverse naturally occurring substances resulting from the secondary metabolism of pathogenic filamentous fungi. They are produced mainly by the genera Fusarium, Alternaria, Aspergillus and Penicillium which can contaminate grains and cereals such as wheat, corn and soy. According to the nature and the concentration levels, mycotoxins can induce toxic effects in food-production animals and humans. An in vitro study was conducted to evaluate the susceptibility of broiler chickens lymphocytes to different concentrations of ochratoxin A, deoxynivalenol and zearalenone. Each toxin was added to the cell medium at different concentrations (0.001, 0.01, 0.1 and 1&#956;g/mL). Cell viability and ecto-adenosine deaminase activity were assessed at 24, 48 and 72 hours by colorimetric assays. Thus, it were used 0.7x105 lymphocytes/mL in RPMI 1640 medium, supplemented with 10% fetal bovine serum and 2.5 IU of penicillin/streptomycin per mL, incubated at 37°C in a 5% CO2 atmosphere. All the experiments were carried out in triplicate and the results were expressed as mean ± standard error of the mean. The results showed that OTA and DON induced lymphocyte proliferation and reduced enzymatic activity in vitro (P<0,05), whereas ZEA also promoted proliferation (P<0,05), but neither alteration on enzymatic activity (P>0,05). It was possible to correlate the results about viability cell and ecto-adenosine deaminase activity, suggesting that, at minimal concentrations, the evaluated mycotoxins do not stimulated the enzymatic activity, which has proinflammatory action and contributes for the immunosuppression process, thus, avoiding a decrease on the viability cell. This is the first in vitro study conducted with OTA, DON and ZON in broiler chickens lymphocytes evaluating these parameters.

E-mail: claudialautert@yahoo.com.br Micotoxinas representam um vasto grupo de contaminantes químicos naturais originados a partir do metabolismo secundário de fungos filamentosos patogênicos. Elas são produzidas, principalmente, pelos gêneros Fusarium, Alternaria, Aspergillus e Penicillium, os quais podem contaminar grãos e cereais, como trigo, milho e soja. Conforme sua natureza e níveis de concentração, micotoxinas podem induzir efeitos tóxicos em animais de produção e humanos. Um estudo in vitro foi realizado para avaliar a susceptibilidade das células linfocitárias de frangos de corte a diferentes concentrações de ocratoxina A, deoxinivalenol e zearalenona. Cada micotoxina foi adicionada ao meio celular em diferentes concentrações (0,001; 0,01; 0,1 e 1&#956;g/mL). A viabilidade celular e atividade de ecto-adenosina desaminase foram analisadas em 24, 48 e 72 horas através de ensaios colorimétricos. Para isso, foram utilizados 0,7x105 linfócitos/mL em meio RPMI 1640, suplementado com 10% de soro fetal bovino e 2,5 UI de penicilina/estreptomicina por mL, incubados em atmosfera de 5% de CO2 a 37 °C. Todos os experimentos foram realizados em triplicata e os resultados foram expressos como média e erro padrão da média. Os resultados obtidos demonstraram que tanto ocratoxina A como deoxinivalenol induziram proliferação linfocitária e baixa atividade enzimática in vitro (P<0,05), enquanto zearalenona também induziu proliferação (P<0,05), mas nenhuma alteração na atividade enzimática (P>0,05). Foi possível correlacionar os dados referentes à viabilidade celular e atividade de ecto-adenosina desaminase, sugerindo que, em concentrações mínimas, as micotoxinas testadas não estimularam a atividade da enzima, que possui ação pró-inflamatória e contribui para o processo de imunossupressão e, portanto, evitando um decréscimo na viabilidade celular. Este é o primeiro estudo feito com OCRA, DON e ZEA sobre linfócitos de frangos de corte em cultivos in vitro na avaliação desses parâmetros.

E-mail: angelita_sampaio@yahoo.com.br The study examined (1) the immune response in broiler chickens after oral immunization with recombinant flagellin (rFliC) from Salmonella Typhimurium conjugated with sodium alginate microparticles, and the immune response enhancement in association with recombinant cholera toxin B subunit protein (rCTB) and pool of Lactobacillus spp. (PL). The immune responses were evaluated by dosage of IgY serum and IgA from intestinal fluid and immunostaining of CD8+ T lymphocytes in the cecum. The immunized animals were challenged with Salmonella Typhimurium (ST) 21 days after treatment. In all immunized groups, a significant increase (p<0.05) was observed in IgA levels (mg/mL), especially three weeks after immunization. The serum IgY levels (mg/mL) were little affected by the treatments and differed significantly among groups only in the second post-immunization week (p<0.05). After the challenge, the number of CD8+ T cells differed significantly between the treatments and negative control. Retrieval of Salmonella Typhimurium was not detected at 48 hours after the challenge in T2 (rFliC+rCTb), T3 (rFliC+PL) and T4 (rFliC+rCTB PL). The rFliC administered orally with or without rCTB and Lactobacillus spp. produces significant induction of humoral immune response, and the immunized chickens were more effective in eliminating Salmonella after challenge.

As respostas imunes foram avaliadas por dosagem de IgY sérica e IgA do fluído intestinal e imunomarcação de linfócitos T CD8+ presentes no ceco. Os animais imunizados foram desafiados aos 21 dias após tratamento com Salmonella Typhimurium (ST). Foi observado em todos os grupos imunizados um aumento significativo (p<0,05) nos níveis de IgA (mg/mL) principalmente três semanas após as imunizações. Os níveis de IgY sérica (mg/mL) foram pouco influenciados pelos tratamentos, apenas na segunda semana após imunização observou-se diferenças significativas (p<0,05) entre os grupos. Observou-se que o número de linfócitos T CD8+ apresentou diferença significativa entre os tratamentos e o controle negativo após o desafio. Quanto a recuperação de Salmonella Typhimurium, observou-se que 48 horas após o desafio já não havia detecção do agente nos grupos T2 (rFliC+rCTb), T3 (rFliC+PL) e T4 (rFliC+rCTB+PL). Concluí-se que rFliC administrada, via oral, associada ou não a Lactobacillus spp e rCTB, demonstrou induzir significativamente a resposta imune humoral e que as aves imunizadas foram mais eficientes na eliminação de Salmonella após desafio.

E-mail: rauber.vet@gmail.com The objective of this study was to determine the effects of three doses of fumonisin B1 (0, 100, and 200mg/kg of feed) on biological variables (relative weight of liver [RWL], total plasma protein [TPP], albumin [Alb], calcium [Ca], phosphorus [P], uric acid [UA], alanine aminotransferase [ALT], aspartate aminotransferase [AST], gamma glutamyltransferase [GGT], alkaline phosphatase [AP], total cholesterol [Chol], triglycerides [Tri], sphinganine-to-sphingosine ratio [SA:SO], and C-reactive protein [CRP]), morphological evaluation of the small intestine (villus height [VH], crypt depth [CD], and villus-to-crypt ratio [V:C]), histological evaluation, and on performance (body weight [BW], feed intake [FI], and feed conversion rate [FCR]) of broiler chickens. Significant effects of FB were observed on BW and FI (reduced), on RWL, TPP, Ca, ALT, AST, GGT, Chol, and Tri (increased) at both 14 and 28 days evaluations. In addition, significant increase was observed on FCR, Alb, P, SA:SO, and CRP and significant reduction in UA, VH, and V:C only at the 28 days evaluation. Significant histological lesions were observed on liver and kidney of FB inoculated broilers at 14 and 28 days. Those results show that FB has a significant effect on biological and histological variables and on performance of broiler chickens.

#14 - Use of probiotics on the T cells activation and Salmonella Minnesota control in broiler chickens, 33(1):11-14

E-mail: santin@ufpr.br To evaluate the effect of probiotics on the immune response of broiler chickens challenged with Salmonella Minnesota (SM), 60 chickens were divided into three groups: CN - Birds that were not inoculated with SM (negative control), CP - birds inoculated with SM (positive control) and Probiotic- birds supplemented with probiotic consisting of Lactobacillus acidophilus, L. plantarium, L. rhamnosus, L. bulgaricus, Enterococcus faecium, Streptococcus thermophilus and Bifidobacterium bifidum in diet and inoculated with SM. At 14 days the birds were challenged with SM, and at 7 and 35 days were quantified goblet cells, CD4 + and CD8 + intestinal mucosa of the ileum and cecum. Birds supplemented with probiotics, at 7 days of age showed a significant increase (P&#8804;0.05) of goblet cells in the ileum and CD4 + and CD8 + cells in the cecum. At 35 days there were significant (P&#8804;0.05) of CD8 + cells in birds inoculated on CN and Probiotic. The use of probiotics provided a significant reduction (P&#8804;0.05) of Salmonella sp. counts.

A utilização de probióticos proporcionou redução significativa (P&#8804;0,05) da contagem de Salmonella sp.


#15 - Oregano, rosemery, cinnamon essential oil and pepper extract to control Salmonella, Eimeria and Clostridium in broiler chickens, 32(5):411-418

[Oregano, rosemery, cinnamon essential oil and pepper extract to control Salmonella, Eimeria and Clostridium in broiler chickens.] Óleo essencial de orégano, alecrim, canela e extrato de pimenta no controle de Salmonella, Eimeria e Clostridium em frangos de corte. Pesquisa Veterinária Brasileira 32(5):411-418. E-mail: larissapickler@yahoo.com.br The efficiency of a product in broiler feed containing essential oil of oregano, rosemary, cinnamon and extract of red pepper (plant compost) in the control of Salmonella, Eimeria and Clostridium was evaluated. Two experiments were carried out to evaluate the product. In the first experiment the efficiency of this product to control Clostridium perfringens after challenge with Eimeria acervulina, E. maxima and E. tenella was assessed. Day old chicks were allotted into three groups: T1 - control diet without growth promoter, T2 - diet with avilamycin (10ppm), and T3 - diet with addition of the plant compost (100ppm). The use of the plant compost in broiler diets reduced specific lesions of E. maxima and E. tenella at 14 days after inoculation and reduced the count of colony forming units (CFU) of Clostridium perfringens in the ceca comparing to the control group. In the second trial the efficiency of the same product in birds challenged with Salmonella Enteritidis was evaluated. Day old birds were submitted to three experimental diets: T1 - control diet without antibiotics growth promoter, T2 - diet with 10ppm Avilamycin, T3 - diet with 100ppm of the plant compost mentioned above. At 21 days of age all birds were inoculated with 105 CFU of Salmonella Enteritidis. The use of the plant compost and avilamycin decreased the excretion of Salmonella in poultry 72 hours after the inoculation. The use of the plant compost increased villous/CD3+ cells in the duodenum, compared to group avilamycin and control, but had no effect on the expression of these cells in the cecum.

Para tal, foram realizados dois experimentos. No primeiro avaliou-se a eficiência deste produto no controle de Clostridium perfringens após desafio com Eimeria acervulina, E. maxima e E. tenella. Aves de um dia de idade foram divididas em três grupos: T1 - dieta controle sem aditivo promotor de crescimento; T2 - dieta com adição de avilamicina (10ppm); e T3 - dieta com adição do composto vegetal (100ppm). O uso do composto vegetal na alimentação de frangos reduziu lesões específicas de E. maxima e E. tenella aos 14 dias pós-inoculação (PI) como também reduziram a contagem de unidades formadoras de colônias (UFC) de Clostridium perfringens no conteúdo do ceco das aves em relação ao grupo controle. No segundo experimento avaliou-se a eficiência deste mesmo produto em aves desafiadas com Salmonella Enteritidis. Aves de um dia de idade foram distribuídas em três tratamentos, sendo T1 - dieta controle sem adição de antibiótico promotor de crescimento, T2 - dieta com 10ppm de Avilamicina, T3 - dieta com 100ppm de um produto a base do composto vegetal acima citado. Aos 21 dias de idade todas as aves foram inoculadas com 105 UFC de Salmonella Enteritidis. A utilização do composto vegetal e avilamicina diminuiu a excreção de Salmonella nas aves 72 horas PI de Salmonella. A utilização do composto vegetal aumentou a relação vilo/células CD3+ no duodeno, em relação ao grupo avilamicina e controle, porém não teve efeito sobre a expressão destas células no ceco.

Occurrence of infection with Toxoplasma gondii and factors associated with transmission in broiler chickens and laying hens in different raising systems. E-mail: amendoei@ioc.fiocruz.br Toxoplasmosis is a zoonotic disease caused by the protozoan Toxoplasma gondii. The aim of the present study was to determine the occurrence and identify the risk factors associated with transmission of T. gondii to chickens raised in different systems (free-ranged and confined) to produce eggs or meat. The 810 animals were allocated in two experimental groups according to the production system purpose: 460 broiler chickens (Group 1) and 350 layer chickens (Group 2). In order to analyze the possible factors involved in T. gondii infection in the chickens, an epidemiological questionnaire was developed for all properties.The serological detection of anti-Toxoplasma gondii antibodies was performed by Indirect Immunofluorescence (IFAT) and by Enzime Linked Imunossorbent Assay (ELISA). Since the agreement index (kappa) between these two serological techniques was considered high, 21.2% of the 810 animals were considered reactive. In Group 1, 12.2% (56/460) were positive, while in the Group 2 the positivity rate was 33.1% (116/350). The production system may be influencing the seropositivity of the animals in both groups. However, only in Group 2 it was possible to notice a statistically significant relationship between the breeding system and the frequency of positive sera. This result indicates that, at least for laying hens, the production system is directly involved in T. gondii infection. The contact with cats in Group 1 did not influence the distribution of seroreactive animals, but in Group 2 a significant relationship was observed. The occurrence of anti-T. gondii antibodies was high in both groups (broiler and posture chickens). Free-ranged chickens raised for egg production proved to be the most exposed group to the T. gondii infection. This can be related to the fact that these animals stay for longer periods in the farms, in direct contact with possibly contaminated soil by the presence of domestic cats.

Ocorrência da infecção por Toxoplasma gon- dii e fatores associados à sua transmissão em aves de corte e postura produzidas em diferentes tipos de criação. E-mail: amendoei@ioc.fiocruz.br A toxoplasmose é uma zoonose causada pelo protozo- ário Toxoplasma gondii. O objetivo deste estudo foi determinar a ocorrência e identificar os fatores de risco associados à transmissão de T. gondii para frangos criados em diferentes sistemas (caipira e confinado) para produzir ovos ou carne. Os 810 animais foram divididos em dois grupos experimentais de acordo com o propósito do sistema de produção: 460 frangos de corte (Grupo 1) e 350 galinhas poedeiras (Grupo 2). A fim de analisar os possíveis fatores envolvidos na infecção pelo T. gondii nas galinhas, um questionário epidemiológico foi respondido por todos os proprietários. A detecção sorológica de anticorpos anti-Toxoplasma gondii foi realizada pela técnica de imunofluorescência indireta (RIFI) e Enzime Linked Assay Imunossorbent (ELISA). Uma vez que o índice de concordância (kappa) entre estas duas técnicas sorológicas foi considerada alta, 21,2% dos 810 animais foram considerados reativos. No Grupo 1, 12,2% (56/460) foram positivos, enquanto no Grupo 2 a taxa de positividade foi de 33,1% (116/350). O sistema de produção pode estar influenciando a soropositividade dos animais em ambos os grupos. No entanto, apenas no Grupo 2, foi possível notar uma relação estatisticamente significativa entre o sistema de produção e da freqüência de soros positivos. Este resultado indica que, pelo menos para as galinhas poedeiras, o sistema de produção está diretamente envolvido na infecção pelo T. gondii. O contato com os gatos no Grupo 1 não influenciou a distribuição dos animais sororreagentes, mas no Grupo 2 uma relação estatisticamente significativa foi observada. A ocorrência de anticorpos anti-T. gondii foi alta nos dois grupos (frangos de corte e postura). Galinhas cairpiras criadas para produção de ovos provou ser o grupo mais exposto à infecção T. gondii. Isto pode estar relacionado ao fato de que estes animais ficam por períodos mais longos nas fazendas, em contato direto com o solo possivelmente contaminado pela presença de gatos domésticos.

E-mail: larissapickler@yahoo.com.br Two experiments were carried out to evaluate effectiveness of organic acids against Salmonella enterica enterica serovars Enteritidis (SE) and Minnesota (SM) in broilers. In the first experiment three treatments were evaluated: T1 - feeding with organic acids, T2 - feeding with organic acids and organic acids in drink water, and T3 - control group. All animals were oral challenged with SE. Organic acids in diet (T1) and organic acids in diet and drink water (T2) reduced the shadding of Salmonella in crop and cecum 7 days post challenged with SE and reduced the CD3+ cells in jejunal mucosa of broilers. In the second experiment four treatments were evaluated, T1 - control group, T2 - control group oral challenged with Salmonella Minnesota (SM); T3 - oral challenged animals with SM and treated with organic acids in diet; T4 - oral challenged animals with SM and treated with organic acids in diet and in drink water. Organic acids in diet (T3) and organic acids in diet and in drink water (T4) reduced the shadding of SM in crop of challenged broilers, 7 days post inoculation The use of organic acids in diet and in water was more effective to control SE than SM.

No primeiro experimento foram avaliados 3 tratamentos: T1 - ração adicionada de ácido orgânico, T2 - ração adicionada de ácido orgânico e ácido orgânico na água de bebida, T3 - grupo controle. Todos os animais foram inoculados com SE, via oral. A utilização de ácidos orgânicos na ração (T1) e na ração e na água (T2) diminuíram a excreção de Salmonella no papo e no ceco 7 dias pós inoculação com SE e houve redução de células CD3+ no jejuno dos frangos. No segundo experimento foram avaliados 4 tratamentos sendo T1 - controle, T2 - controle inoculado via oral com Salmonella Minnesota (SM), T3 - animais inoculados via oral com SM e ácidos orgânicos na ração e T4 - animais inoculados via oral com SM e ácidos orgânicos na ração e na água de bebida. Ácidos orgânicos a ração (T3) e na ração e na água (T4) reduziram a excreção de SM em papo de frangos de corte desafiados, 7 dias após inoculação. O uso de ácidos orgânicos na ração e na ração e na água foram mais eficientes em reduzir SE do que SM.

Anticoccidial activity was evaluated on the basis of performance (weight gain, feed conversion ratio) and pathogenic (oocyst score, lesion score and mortality %age) parameters. Among HCl medicated groups, the maximum anticoccidial effect was seen in the group medicated with 1000ppm HCl followed by 2000ppm and 3000ppm HCl medicated groups. Amprolium and 1000ppm HCl were almost equivalent in suppressing the negative performance and pathogenic effects associated with coccidiosis (Eimeria tenella) challenge. In summary, the lower doses of HCl have the potential to be used as alternative to chemotherapeutic drugs for Eimeria tenella control. It is therefore suggested that further studies should be carried out to determine the possible minimum safe levels of HCl with least toxic effects to be used as anticoccidial.
