#11 - Genotyping and antimicrobial resistance in Escherichia coli from pig carcasses, 37(11):1253-1260

E-mail: mcardoso@ufrgs.br The increasing antimicrobial resistance observed worldwide in bacteria isolated from human and animals is a matter of extreme concern and has led to the monitoring of antimicrobial resistance in pathogenic and commensal bacteria. The aim of this study was to evaluate the antimicrobial resistance profile of Escherichia coli isolated from pig carcasses and to assess the occurrence of relevant resistance genes. A total of 319 E. coli isolates were tested for antimicrobial susceptibility against different antimicrobial agents. Moreover, the presence of extended-spectrum β-lactamase (ESBL) and inducible ampC-β-lactamase producers was investigated. Eighteen multi-resistant strains were chosen for resistance gene detection and PFGE characterization. The study showed that resistance to antimicrobials is widespread in E. coli isolated from pig carcasses, since 86.2% of the strains were resistant to at least one antimicrobial and 71.5% displayed multi-resistance profiles. No ampC-producing isolates were detected and only one ESBL-producing E. coli was identified. Genes strA (n=15), floR (n=14), aac(3)IVa (n=13), tetB (n=13), sul2 (n=12), tetA (n=11), aph(3)Ia (n=8) and sul3 (n=5) were detected by PCR. PFGE analysis of these multi-resistant E. coli strains showed less than 80% similarity among them. We conclude that antimicrobial multi-resistant E. coli strains are common on pig carcasses and present highly diverse genotypes and resistance phenotypes and genotypes.

O objetivo desse estudo foi avaliar o perfil de resistência em Escherichia coli isolada de carcaças suínas e descrever a ocorrência de alguns genes de resistência relevantes. Um total de 319 isolados de E. coli foi testado quanto à suscetibilidade frente a diversos antimicrobianos. A presença de isolados produtores de β-lactamases (ESBL) de espectro estendido e β-lactamase induzível do tipo ampC foi também investigada. Dezoito cepas multirresistentes foram escolhidas para investigação de genes de resistência e caracterização por macro-restrição (PFGE). Os resultados demonstraram que a resistência a antimicrobianos está disseminada, pois 86,2% dos isolados de E. coli foram resistentes ao menos a um antimicrobiano e 71,5% apresentaram perfil de multirresistência. Uma cepa de E. coli produtora de ESBL e nenhuma produtora de ampC induzível foram identificadas. Os genes strA (n=15); floR (n=14);aac(3)-IVa (n=13); tetB (n=13); sul2 (n=12); tetA (n=11); aph(3’)Ia (n=8); sul3 (n=5) foram detectados por PCR. A análise de PFGE demonstrou que cepas de E. coli multirresistentes apresentaram similaridade inferior a 80% entre si. Concluiu-se que cepas multirresistentes de E. coli são frequentes em carcaças de suínos e apresentam uma alta diversidade genotípica, bem como de fenótipos e genes de resistência.

The pulmonary system is most frequently affected, with lesions observed in the air sacs and lungs of a wide variety of bird species. The aim of this study was to confirm by molecular methods the identification and the genetic diversity of Aspergillus fumigatus isolates of lung’s samples from healthy broilers (Galus galus domesticus). Forty-four (9.5%) isolates of lung’s samples were confirmed as A. fumigatus by polymerase chain reaction (PCR) multiplex (amplification of β-tub and rodA gene fragments). Microsatellite typing for A. fumigatus was used to analyse all avian isolates. Among them, 40 genotypes (90.9%) were observed only one time. The results showed a high variability and multiple genotypes of de A. fumigatus collected from lung’s samples of broilers.

O sistema pulmonar de uma grande variedade de espécies de aves é o mais frequentemente afetado, com lesões nos sacos aéreos e pulmões. Objetivou-se confirmar por métodos moleculares a identificação e a diversidade genética de Aspergillus fumigatus isolados de amostras pulmonares de frangos de corte sadios (Galus galus domesticus). Quarenta e quatro (9,5%) isolados foram confirmados como A. fumigatus através de reação em cadeia da polimerase (PCR) multiplex (amplificação de fragmentos dos genes β-tub e rodA). Todos isolados foram tipificados, sendo quarenta (90,9%) observados apenas uma vez. Os resultados mostram uma alta variabilidade e múltiplos genótipos de A. fumigatus obtidos de amostras pulmonares de frangos, de corte.

E-mail: danicazola@gmail.com Spoligotyping was performed in the present study to genotype Mycobacterium bovis isolates obtained from tissues of cattle that were positive in the comparative intradermal tuberculin test (CITT) in the state of Mato Grosso do Sul (Brazil). Tissue samples from 13 positive cattle from different municipalities of the state were cultured using a Stonebrink medium. The resulting colonies were subjected to Ziehl-Neelsen staining and all isolates exhibited the staining characteristics of AFB. The 13 isolates of AFB were identified by means of a multiplex PCR (mPCR) assay. The hsp65 gene was targeted for the identification of Mycobacterium spp., whereas the IS6110 insertion sequence was targeted for the identification of the Mycobacterium tuberculosis complex (MTC) and the rvd1rv2031c region was explored for the detection of Mycobacterium bovis. The spoligotyping assay was performed to genotype mycobacterial isolates. Of the 13 cattle, seven had at least one lesion suggestive of tuberculosis in the retropharyngeal, parotid and lung lymph nodes or lung. The remaining six exhibited no lesions suggestive of the disease. In the mPCR, 11 of the 13 isolates (84.6%) were positive for Mycobacterium spp., 8/13 (61.5%) were positive for the MTC and 7/13 (53.8%) were positive for M. bovis. Based on the spoligotyping, eight isolates were grouped into three different groups of genotypes and one isolate exhibited an orphan type. Four isolates exhibited spoligotype pattern SB0121, while two isolates were associated with the pattern SB1145, another two were associated with pattern SB0881 and one was associated with pattern SB0140. Spoligotyping confirmed the genetic diversity present among isolates found in the state of Mato Grosso do Sul. In addition, SB0121 was confirmed as the predominant profile.

Tecidos de 13 bovinos positivos, oriundos de diferentes municípios do estado, foram cultivados em meio de Stonebrink. As colônias resultantes foram submetidas à coloração de Ziehl-Neelsen e todos os isolados apresentaram características tintoriais de BAAR. Os 13 isolados de BAAR foram identificados por PCR multiplex (mPCR). O gene hsp65 foi alvo para identificação de Mycobacterium spp, a sequência de inserção IS6110 foi alvo para identificação de complexo Mycobacterium tuberculosis (CMT) e a região rvd1rv2031c foi explorada para detecção de M. bovis. Os isolados micobacterianos foram genotipados pela técnica de spoligotyping. Dos 13 bovinos, sete tinham pelo menos uma lesão sugestiva de tuberculose em linfonodos retrofaríngeos, parotídeos e pulmonares ou no pulmão, e em seis não foram encontradas lesões visíveis sugestivas da doença. Na mPCR, 11/13 (84,6%) isolados foram positivos para Mycobacterium spp; 8/13 (61,5%) positivos para CMT e 7/13 (53,8%) positivos para M. bovis. Com base no spoligotyping, oito isolados de BAAR foram agrupados dentro de três diferentes agrupamentos de genótipos e uma amostra remanescente apresentou perfil único, sendo quatro isolados com padrão de espoligotipo SB0121, dois SB1145, dois SB0881 e um SB0140. A técnica de spoligotyping demonstrou que há diversidade genética entre os espoligotipos presentes no estado de Mato Grosso do Sul, embora predomine o perfil SB0121.

E-mail: caio-patologia@usp.br Enzootic paraplexia or scrapie is a fatal neurodegenerative disease affecting mainly sheep and rarely goats. The disease is influenced by polymorphisms at codons 136, 154 and 171 of prnp gene that encodes the prion protein. The animals may be susceptible or resistant to the development of the disease according to the allelic sequences observed in these codons. In Brazil there were only cases of scrapie in imported animals, therefore the country is considered free of the disease. This study performed the genotyping of different polymorphisms associated to the development of scrapie. Then, based on these findings the animals were categorized in resistant and susceptible. A total of 118 samples were sequenced from the Santa Ines sheep raised on properties located in the State of Sao Paulo. From these samples, 6 alleles and 11 genotypes were identified (ARQ / ARQ, ARR / ARQ, ARQ / AHQ, ARQ / VRQ, AHQ / AHQ, ARR / ARR, ARR / AHQ, VRQ / VRQ, ARQ / TRQ, TRR / TRR, TRQ / TRQ), the genotype ARQ / ARQ presented a frequency of 56.7%. It was also detected the presence of tyrosine at codon 136, which may be considered a rare observation, since there is no report regarding Santa Ines breeding presenting this polymorphism. These results showed the great genetic variability in Santa Ines in Sao Paulo and only 1,69% of the genotypes observed are extremely resistant to scrapie. These data demonstrate that the Santa Ines sheep can be considered potentially susceptible to scrapie.

Os animais podem ser susceptíveis ou resistentes, de acordo com as sequências alélicas observadas nos referidos códons. No Brasil ocorreram apenas casos de animais que foram importados, sendo o país considerado livre da doença. Neste trabalho foi realizada a genotipagem dos diferentes polimorfismos associados ao desenvolvimento do scrapie e a categorização em animais susceptíveis e resistentes. Foram sequenciadas 118 amostras provenientes de ovinos da raça Santa Inês criados em propriedades localizadas no Estado de São Paulo. Destas amostras foram identificados 6 alelos e 11 genótipos (ARQ/ARQ, ARR/ARQ, ARQ/AHQ, ARQ/VRQ, AHQ/AHQ, ARR/ARR, ARR/AHQ, VRQ/VRQ, ARQ/TRQ, TRR/TRR, TRQ/TRQ), dentre os quais o genótipo ARQ/ARQ teve ocorrência de 56,7%. Em nosso estudo foi detectada a presença da tirosina no códon 136, observação rara na medida em que não existem relatos nacionais e internacionais envolvendo a raça Santa Inês descrevendo este polimorfismo. Com os resultados obtidos, foi possível determinar a existência de grande variabilidade genética relacionada à raça Santa Inês no Estado de São Paulo. Apesar da variabilidade, apenas 1,69% dos genótipos observados mostraram-se extremamente resistentes ao scrapie. Estes dados demonstram que a raça nativa Santa Inês pode ser considerada potencialmente susceptível ao scrapie.

E-mail: leonardo@usp.br Twelve Brazilian isolates and one reference vaccine strain of avian infectious bronchitis virus (IBV) were propagated in embryonating chicken eggs. The entire S1 glycoprotein gene of these viruses was analysed by reverse-transcriptase-polymerase chain reaction and restriction fragment length polymorphism (RT-PCR-RFLP), using the restriction enzymes HaeIII, XcmI and BstyI. The RFLP patterns led to the classification of these isolates into five distinct genotypes: A, B, C, D and Massachusetts. Five of twelve isolates were grouped in Massachusetts genotype and the remaining seven viruses were classified into four distinct genotypes: A (2), B (2), C (2) or D (1). Such genotyping classification agreed with previous immunological analysis for most of these viruses, highlighting the occurrence of a relevant variability among the IBV strains that are circulating in Brazilian commercial poultry flocks.

E-mail: jpessoa@ibb.unesp.br Feline immunodeficiency virus (FIV) is a lentivirus associated with immunologic disorders in domestic cats. Due to the high genetic variability of FIV, five subtypes (A to E) have been identified and diversity within each subtype is also frequent. The study of the genetic diversity can aid the understanding the pathogenesis and epidemiology of the disease. Therefore, the present work aimed to analyze phylogenetically FIV isolates of domestic cats from the state of São Paulo, Brazil. The sequencing of 658 bp of the gag gene from 23 samples was performed and the results were analyzed using the Tamura-Nei nucleotidic substitution method. The phylogenetic analysis showed that all viruses belong to subtype B, and clearly three subgroups were present within this subtype. Additionally, these results suggest a common ancestor between the FIV strains derived from Japan and one Brazilian virus. In conclusion, this work presents the first information about the genetic diversity of FIV in the state of São Paulo. Additional studies are necessary to characterize the real scenario of the distribution of FIV subtypes in the population of Brazilian cats.

E-mail: jpessoa@ibb.unesp.br Feline immunodeficiency virus (FIV) is a lentivirus associated with immunologic disorders in domestic cats. Due to the high genetic variability of FIV, five subtypes (A to E) have been identified and diversity within each subtype is also frequent. The study of the genetic diversity can aid the understanding the pathogenesis and epidemiology of the disease. Therefore, the present work aimed to analyze phylogenetically FIV isolates of domestic cats from the state of São Paulo, Brazil. The sequencing of 658 bp of the gag gene from 23 samples was performed and the results were analyzed using the Tamura-Nei nucleotidic substitution method. The phylogenetic analysis showed that all viruses belong to subtype B, and clearly three subgroups were present within this subtype. Additionally, these results suggest a common ancestor between the FIV strains derived from Japan and one Brazilian virus. In conclusion, this work presents the first information about the genetic diversity of FIV in the state of São Paulo. Additional studies are necessary to characterize the real scenario of the distribution of FIV subtypes in the population of Brazilian cats.
