E-mail: apkipnis@gmail.com The present study aimed to assess the CD4, CD8 and gd blood levels for Curraleiro Pé-duro, as well as the specific IFN-g response after BCG vaccination using flow cytometry. The specific immune response against BCG was also evaluated by tuberculin skin test, performed before and 45 days after the vaccination. For comparison purposes, the same parameters were investigated on Nellore calves, an exotic bovine with resistance previously demonstrated. Naturally, Curraleiro Pé-duro animals had greater levels of CD4, CD8 and &#947;&#948; lymphocytes (p<0.05). In response to vaccine, Curraleiro Pé-duro showed greater ability to respond specifically to BCG, generating resistance profile (Th1), evidenced by greater number of antigen specific CD4+ cells producing IFN-&#947; (p<0.05) and also higher tuberculin skin test reaction (p<0.05). Additionally, vaccinated Curraleiro Pé-duro calves had higher CD4 cells numbers than both Nellore control (p<0.05) and vaccinated groups (p<0.05). Curraleiro Pé-duro calves’ higher basal lymphocytes blood level and stronger response in both IFN-&#947; and tuberculin skin test parameters probably play a positive role on protection/resistance to Mycobacterium bovis.

E-mail: apkipnis@gmail.com O presente estudo teve como objetivo avaliar níveis sanguíneos de células CD4, CD8 e &#947;&#948; no sangue periférico de bezerros Curraleiro Pé-Duro, bem como a produção específica de IFN-&#947; por essas células em resposta à vacinação com BCG, através de citometria de fluxo. A resposta imune específica contra BCG também foi avaliada por teste tuberculínico, realizado antes e 45 dias após a vacinação. Para fins de comparação, os mesmos parâmetros foram investigados em bezerros da raça Nelore, uma raça bovina exótica com resistência demonstrado anteriormente. Naturalmente, animais da raça Curraleiro Pé-Duro apresentaram maiores níveis de CD4, CD8 e linfócitos &#947;&#948;. Em resposta a vacina, Curraleiro Pé-duro mostrou maior capacidade de responder especificamente ao BCG, gerando perfil de resistência (Th1), evidenciado pelo maior número de células CD4+ específicas produtoras de IFN-&#947; e maior reação cutânea a por tuberculina. Os maiores níveis basais de linfócitos, maior produção de IFN-&#947; e reação cutânea à prova tuberculínica provavelmente desempenham um papel positivo na proteção/resistência ao Mycobacterium tuberculosis.

E-mail: angelita_sampaio@yahoo.com.br The study examined (1) the immune response in broiler chickens after oral immunization with recombinant flagellin (rFliC) from Salmonella Typhimurium conjugated with sodium alginate microparticles, and the immune response enhancement in association with recombinant cholera toxin B subunit protein (rCTB) and pool of Lactobacillus spp. (PL). The immune responses were evaluated by dosage of IgY serum and IgA from intestinal fluid and immunostaining of CD8+ T lymphocytes in the cecum. The immunized animals were challenged with Salmonella Typhimurium (ST) 21 days after treatment. In all immunized groups, a significant increase (p<0.05) was observed in IgA levels (mg/mL), especially three weeks after immunization. The serum IgY levels (mg/mL) were little affected by the treatments and differed significantly among groups only in the second post-immunization week (p<0.05). After the challenge, the number of CD8+ T cells differed significantly between the treatments and negative control. Retrieval of Salmonella Typhimurium was not detected at 48 hours after the challenge in T2 (rFliC+rCTb), T3 (rFliC+PL) and T4 (rFliC+rCTB PL). The rFliC administered orally with or without rCTB and Lactobacillus spp. produces significant induction of humoral immune response, and the immunized chickens were more effective in eliminating Salmonella after challenge.

As respostas imunes foram avaliadas por dosagem de IgY sérica e IgA do fluído intestinal e imunomarcação de linfócitos T CD8+ presentes no ceco. Os animais imunizados foram desafiados aos 21 dias após tratamento com Salmonella Typhimurium (ST). Foi observado em todos os grupos imunizados um aumento significativo (p<0,05) nos níveis de IgA (mg/mL) principalmente três semanas após as imunizações. Os níveis de IgY sérica (mg/mL) foram pouco influenciados pelos tratamentos, apenas na segunda semana após imunização observou-se diferenças significativas (p<0,05) entre os grupos. Observou-se que o número de linfócitos T CD8+ apresentou diferença significativa entre os tratamentos e o controle negativo após o desafio. Quanto a recuperação de Salmonella Typhimurium, observou-se que 48 horas após o desafio já não havia detecção do agente nos grupos T2 (rFliC+rCTb), T3 (rFliC+PL) e T4 (rFliC+rCTB+PL). Concluí-se que rFliC administrada, via oral, associada ou não a Lactobacillus spp e rCTB, demonstrou induzir significativamente a resposta imune humoral e que as aves imunizadas foram mais eficientes na eliminação de Salmonella após desafio.

E-mail: edisio@pq.cnpq.br This study aimed to evaluate the efficacy of two inactivated vaccines against contagious agalactia containing adjuvant oily and watery. For this, 73 goats were divided in two experiments. In the experiment I was verified the vaccine safety and 15 goats were divided into three experimental groups of five animals each. A1 and B1 groups were immunized with vaccine containing either aluminum or oil, respectively, and group C was the control group without immunization. In the experiment II, the immune response against the vaccines was evaluated by immunization of 58 goats that were divided in to two groups: group A2, 28 animals were immunized with the aluminum based vaccine; and group B2, 30 animals were immunized with the oil based vaccine. In the experiment II, the animals received a third dose on 180 day after the second dose. Antibody levels were determined by indirect ELISA from ssamples collected on vaccination days and on 30 day after the second dose (Experiment I) and on 30 day after the third dose (Experiment II). The animals from B1 and B2 groups (oil based vaccine) demonstrated higher antibody levels (P<0.05) than A1 and A2 (aluminum based vaccine) in both experiments.

Para tanto, foram utilizados 73 caprinos, agrupados em dois experimentos. No experimento I, para avaliar a inocuidade das vacinas, foram utilizados 15 caprinos, subdivididos em três grupos de cinco animais cada, sendo que o grupo A1 foi imunizado com a vacina aquosa, o grupo B1 com a vacina oleosa e o grupo C não imunizado, foi o controle. No experimento II, para avaliar a resposta imune foram utilizados 58 caprinos, subdivididos em dois grupos, sendo o grupo A2, com 28 animais imunizados com a vacina aquosa e o grupo B2, com 30 animais imunizados com a vacina oleosa. Os animais do experimento II receberam uma terceira dose, 180 dias após a segunda dose vacinal. Os níveis de anticorpos foram determinados por ELISA indireto, realizado no dia de cada vacinação e 30 dias após a segunda dose (experimento I) e 30 dias após a terceira dose vacinal (experimento II). Os animais do grupo B1 e B2 (vacina oleosa) apresentaram níveis de anticorpos estatisticamente superiores (P<0,05) quando comparados aos dos grupos A1 e A2 (vacina aquosa) nos dois experimentos.

[Kinetics of the humoral immune response of dogs young immunized with Leptospira interrogans.] Cinética da resposta imune humoral de cães jovens imunizados contra Leptospira interrogans. Pesquisa Veterinária Brasileira 31(11):1000-1005. Faculdade de Medicina Veterinária, Universidade Federal de Uberlândia, Av. E-mail: jack_ufu@yahoo.com.br The objective was to analyze the humoral immune response against Leptospira interrogans using the microscopic agglutination test (MAT) in 26 young dogs, and 17 mixed breed (Group A) and nine mixed breed (Group B) of both sexes, pertaining to kennels and home environments in Uberlandia, Minas Gerais. Dogs were vaccinated with commercial inactivated polyvalent bacterin with Canicola, Grippotyphosa, Icterohaemorrhagiae and Pomona serovars. The immunization schedule was based on three immunizations. The first vaccination was performed at forty-five days of age, considered day zero, and after two boosters at intervals of thirty days each. Seven blood samples from each dog were made from time zero up to 180 days post-vaccination, at intervals of thirty days each. Titles have not been detected pre-vaccination against serovars Canicola, and Icterohaemorrhagiae Grippotyphosa on day zero. Only one dog in group A was reactive with a titer of 1:100 against Pomona in the first harvest. There was no statistical difference between the agglutinating titers between Groups A and B (p>0.05) induced by commercial vaccine used, except at harvest II (p<0.05), in which the group B presented evidence to serovar Autumnalis in all dogs evaluated, whereas in group A, 64.7% of dogs were not reactive to any serovar tested. There were on the 30th title to serovar Autumnalis that persisted until 180 days post-vaccination in both groups, varying only the intensity of immune response without significant statistical difference. Sete colheitas sanguíneas de cada cão foram efetuadas do dia zero até aos 180 dias pós-vacinais, com intervalos de trinta dias cada. Não foram detectados títulos pré-vacinais para os sorovares Canicola, Icterohaemorrhagiae e Grippotyphosa no dia zero. Apenas um cão do grupo A foi reagente com título 1:100 contra o sorovar Pomona na primeira colheita. Não houve diferença estatística entre os títulos de anticorpos aglutinantes entre os Grupos A e B (p>0,05), induzidos pela bacterina comercial utilizada, exceto na colheita II (p<0,05), na qual o grupo B apresentou títulos para o sorovar Autumnalis em todos os cães avaliados, enquanto que, no grupo A, 64,7% dos cães não foram reagentes a nenhum sorovar testado. No dia 30, títulos para o sorovar Autumnalis que persistiram até os 180 dias pós-vacinais, em ambos os grupos, variando apenas a intensidade da resposta imunológica sem diferença estatística significativa. Pesquisa Veterinária Brasileira 29(9):725-730, Universidade Estadual de Maringá, Av. Colombo 5790, Maringá, PR 87020-900, Brazil. E-mail: aemurakami@uem.br An experiment was carried out to evaluate the effect increasing levels of yeast wall and broiler breeders’ age levels on lymphoid organs weight, cellular immune response and hematological parameters in broilers. A total of 3,360 Cobb broilers were allotted, in a completely randomized design and a 2x5 factorial arrangement, and two controls, compound of two broiler breeders age (34 and 57 weeks of age) and five yeast wall levels (zero, one, two, three and four kg of yeast wall/ton of diet). Broiler breeders’ age affected all studied variables. The inclusion of 3kg of yeast wall/ton of meal increased, at 57 weeks age broiler breeders, more intense inflammatory reaction when compared to control diet; however the circulated heterophils and lymphocytes numbers were not increased. In conclusion, the use of yeast wall, associated or not to sorghum on broilers diet still needs complementary studies, for example, purified components of yeast wall (MOS and ß-glucano).

E-mail: aemurakami@uem.br Um experimento foi conduzido para avaliar o efeito de níveis crescentes de parede de levedura e idade das matrizes reprodutoras sobre o peso dos órgãos linfóides, a resposta imune celular e o perfil hematológico de frangos de corte. Foram utilizados 3.360 pintos de corte da linhagem Cobb, distribuídos em delineamento inteiramente casualisado, em esquema fatorial 2x5, mais dois controles, sendo duas idades de matrizes (34 e 57 semanas de idade) e cinco níveis de suplementação de parede de levedura (zero, um, dois, três e quatro kg de parede de levedura/tonelada de ração). A idade das matrizes influenciou a resposta de todas as variáveis. A inclusão de 3kg de parede de levedura/tonelada de ração promoveu, na progênie de reprodutoras de 57 semanas, reação inflamatória mais intensa quando comparada a dieta controle, no entanto não houve aumento significativo no número de heterófilos e linfócitos circulantes. Conclui-se que a utilização da parede de levedura associada ao sorgo ou não em rações de frangos de corte ainda necessita de estudos complementares, que incluam, por exemplo, os componentes purificados da parede de levedura (MOS e ß-glucano).

The results support that the experimental animals started a response of delayed hypersensitivity to the inactivated bacilli, and that both diagnostic tests for bovine tuberculosis were efficient for the identification of animals sensitized with M. bovis and for discrimination of reactions generated by inoculation of cattle with M. avium.

The aim of the present work was to evaluate the association among the kinetics of humoral immune response, the recovery ofviable leptospiras and the intensity ofthe tissue lesions in mice selected for high (H) and low (L) production of antibodies (selection N-A) and in mice from the outbreed Balb/c Iine, inoculated with pathogenic Leptospira interrogans serovar icterohaemorrhagiae. Toe H and L lines showed modifications in some steps ofthe immune response, mainly in relation to the macrophagic activity, representing extreme phenotypes found in heterogeneous natural populations. Mice were sacrificed in eight moments after the infection. Analysis of the results revealed that from the 7th day after the infection, on Iine H mice presented antibodies titles significantly higher as compared to the L line mice, maintaining the multispecific effect Balb/c tine mice showed intermediàte results between the two Iines. Antibodies production worked as a timiting factor to the infection, becausewhen a greater leptospiras recovery was obtained, at the initial phase of the infection, the titles of antibodies were elevated. Inflammatory and degenerative process led to similar lesions in the organs ofinfected mice. Only a variation in the degree oftissue compromising was observed according to the tine. H tine mice showed more extensive lesions than L and Balb/c lines mice. For the Balb/c tine mice, the lesions were moderate. As a mie, H and Balb/c mice lines showed a Th2 profile of immune response, with higher indexes of antibody production and gravity ofthe lesions, while L line mice presented a Th 1 profile ofimmune response.

[Humoral iminune response, bacterial recovery and time lesions in mice geneticaly selected for high and low antibody production and in outbreed Balb/c mice face to Leptospira interrogans sorovar icterohaemorrhagiae.] Resposta humoral, recuperação bacteriana e lesões histológicas em camundongos geneticamente selecionados para bons e maus produtores de anticorpos e Balb/c, frente à infecção por Leptospira interrogans sorovar icterohaemorrhagiae. O presente trabalho teve por finalidade associar a cinética da resposta humoral à recuperação de leptospiras viáveis e à intensidade das lesões teciduais em camundongos geneticamente selecionados para bons (High) e maus (Low) produtores de anticorpos (seleção IV-A), além de camundongos outbreed, Balb/c, inoculados com amostra patogênica de Leptospira interrogans sorovar icterohaemorrhagiae. As linhagens High e Low (seleção IV-A) apresentam modificações em alguns compartimentos da resposta imune, principalmente em relação à atividade macrofágica, representando fenótipos extre mos encontrados em populações naturais heterogêneas. Os camundongos foram sacrificados em oito momentos após a infecção. A análise dos resultados revelou que a partir do 7º dia após a infecção, os camundongos da linhagem High apresentaram elevação nos títulos de anticorpos estatisticamente significantes quando comparados aos camundongos da linhagem Low, mantendo assim o efeito multiespecífico. Os camundongos Balb/c apresentaram resultados intermediários entre as duas linhagens. A produção de anticorpos colaborou como fator limitante à infecção, pois quando obtevese maior recuperação de leptospiras, na fase inicial da infecção, os títulos de anticorpos encontravam-se em elevação. As lesões observadas nos órgãos de camundongos infectados consistiram basicamente nos mesmos processos inflamatórios e degenerativos, que não se alteraram, variando apenas o grau de comprometimento tecidual, de acordo com a linhagem. A linhagem high apresentou lesões mais extensas que as apresentadas pelas linhagens low e Balb/c, sendo que nesta última as lesões foram moderadas. De forma geral a linhagem High e Balb/c apresentaram um perfil de resposta Th2, com o maior índice de produção de anticorpos e gravidade das lesões, enquanto a linhagem Low apresentou um perfil de resposta Th 1.
