E-mail: miliane@ufrrj.br Mastitis is an inflammation of one or more mammary glands caused mainly by bacteria, among which the genus Staphylococcus plays an important role. Bacteria belonging to this genus are known to express virulence factors which allow their persistence and spread in the host. This study aimed to evaluate the phenotypic and genotypic aspects of virulence factors in Staphylococci spp. isolates from bovine mastitis clinical cases. A total of 272 milk samples from 8 farms in the South-Fluminense region of Rio de Janeiro were analyzed. The samples underwent conventional bacterial identification, yielding 250 Staphylococci spp. isolates. These were tested for the phenotypic detection of slime production by the microplate and Congo Red Agar methods. The hemolysins production, hemolytic synergism, caseinase and DNase production were also evaluated. The isolates were then assayed through the Polymerase Chain Reaction method to detect genes associated with virulence factors such as: capsule (cap5, cap8), fibronectin (fnbA, fnbB), slime (icaA, icaD) and hemolysins (hla e hlb). Regarding the number of isolates assessed, 58% (145/250) were identified as coagulase-negative Staphylococcus spp. and 42% (105/250) as coagulase-positive Staphylococcus spp. The latter comprised 36.2% (38/105) of isolates identified as S. aureus, 11.4% (12/105) as S. intermedius and 3.8% (4/105) belonging to the SIG group. The hemolisin production was not significant, whereas only 6,4% (16/250) produced alfa hemolysis, 4,8% (12/250) produced beta hemolysis and 1,6% (4/250) was able to produce both. Caseinase production was observed in 66.4% (166/250) and slime production assayed through the microplate method was positive in 76,8% (192/250). DNAse was detected in coagulase-negative Staphylococcus spp. (38/145) and in S. aureus (14/38). Low association between genetic detection of icaA (38/250) and icaD (54/250) and slime phenotypic expression (192/250) suggest that others genetic markers can be involved in this expression. Regarding gene amplification, the isolates did not show significant correlation between the genetic detection of icaA (38/250) and icaD (54/250) and slime production (192/250), indicating that other genetic markers may be involved in this trait expression. The frequency of the occurrence of the others studied genes was of 4% (10/250) for cap5 and cap8, 32,8% (82/250) for fnbA, 4,4% (11/250) for fnbB, 19,2% (48/250) for hla and 18% (45/250) for hlb. The major circulating strain profile on the farms encompassed slime and caseinase producer strains. The spaA gene was found in all of the S. aureus isolates, presenting varying amplicons sizes, with 300bp being the prevalent size. The amplification of the coa gene showed nine polymorphic variants, with 600bp being the prevalent amplicon. The agr gene was also detected in every S. aureus isolate, with an amplicon of 200bp. It was noticed that the presence or absence of the virulence genes assayed in this study were not correlated with the 6 distinct electrophoretic profiles obtained by PFGE.

Bactérias pertencentes a este gênero são caracterizadas por expressar fatores de virulência que permitem sua persistência e disseminação no hospedeiro. O presente trabalho teve por objetivo avaliar fenogenotipicamente os fatores de virulência de isolados de Staphylococcus spp. a partir de casos de mastite bovina. Foram analisadas 272 amostras de leite provenientes de oito propriedades da região Sul-Fluminense do Estado do Rio de Janeiro. Após identificação, obteve-se um total de 250 isolados de Staphylococcus spp. Estes foram submetidos às provas fenotípicas de detecção da produção de “slime” em microplaca e em ágar vermelho congo; produção de hemolisinas e sinergismo hemolítico; produção de caseinase e DNase. Posteriormente foram submetidos à técnica de PCR para detecção dos genes de produção de cápsula (cap5 e cap8), fibronectina (fnbA,e fnbB), “slime” (icaA e icaD) e hemolisinas (hla e hlb). Do total avaliado, 58% (145/250) foi identificado como Staphylococcus spp. coagulase-negativos e 42% (105/250) como Staphylococcus spp. coagulase-positivos, destes 36,2% (38/105) foram identificados como S. aureus, 11,4% (12/105) como S. intermedius e 3,8% (4/105) como pertencentes ao grupo SIG. Apenas 6,4% (16/250) dos isolados foram produtores de a-hemólise, 4,8% (12/250) de b-hemólise e, 1,6% (4/250) de a e b-hemólise. A produção de caseinase foi observada em 66,4% (166/250), e a produção de “slime” avaliada pela técnica da microplaca em 76,8% (192/250) dos isolados, respectivamente. A DNase foi detectada em ECNs (38/145) e S. aureus (14/38). Os marcadores genéticos avaliados para a produção de slime, icaA e icaD apresentaram nenhuma ou leve concordância com a produção fenotípica, respectivamente, utilizando o coeficiente Kappa. Tal dado parece indicar que outros marcadores genéticos podem estar envolvidos com a expressão desta característica. Os demais genes detectados com frequência de 4% (10/250) para cap5 e para cap8, 32,8% (82/250) para fnbA, 4,4% (11/250) para fnbB, 19,2% (48/250) para hla e 18% (45/250) para hlb. O perfil circulante nas propriedades foi o 1: isolado produtor de “slime” e caseinase. O gene spaA foi positivo em todos os S. aureus, apresentando amplicons de tamanhos variados, sendo o tamanho prevalente o de 300pb. A amplificação do gene coa apresentou nove tipos polimórficos distintos, sendo prevalente o amplicon de 600pb. O gene agr foi detectado em todos os S. aureus, com amplicon de 200pb. Foi observado que os genes de virulência estudados estavam distribuídos de modo aleatório entreos 6 distintos perfis eletroforéticos obtidos através da Eletroforese em Gel de Campo Pulsado (PFGE).

E-mail: miliane@ufrrj.br The use of antibiotic in the control of intramammary infections and in the elimination of its possible sources in dairy farms is an important control measure. However, the inappropriate use of antibiotics can result in the appearance of resistant strains and compromise the efficiency of the treatment. Besides Staphylococcus spp. are among the main pathogens of bovine mastitis, they are often resistant to antibiotics, especially beta-lactamics, mainly by two distinct mechanisms: the production of extracellular enzyme beta-lactamase, encoded by the blaZ gene, and production of PBP2a or PBP2’ a penicillin-binding protein with low affinity, encoded by the mecA gene. The expression of mecA gene is constitutive or induced by beta-lactamic antibiotics, such as oxacillin and cefoxitin. The mecA gene is inserted into the chromosome through a staphylococcal mobile genetic element, called staphylococcal cassette chromosome mec (SCCmec). The present study evaluated the phenogenotypical resistance profile to beta-lactam antibiotics of 250 Staphylococcus spp isolates, using oxacillin and cefoxitin as markers in order to produce data to the knowledge of resistance in dairy farms located in the South-Fluminense and the Metropolitan regions of the State of Rio de Janeiro to support the implementation of measures to control this disease. The assessment of resistance was made through 8 different phenotypic tests and yielded 54 profiles. Disk diffusion and agar screen with oxacillin were used as “gold standard” for the calculation of sensitivity, specificity and prediction once they are recommended by the CLSI veterinarian as standardized tests. Disk diffusion with cefoxitin achieved the best performance in the prediction of oxacillin resistance. Genotypic detection of mecA do not provided any positive isolate, otherwise mecI and mecRI genes were also detected in 11.6% (29/250) of the studied Staphylococcus spp. Four cassette mec types were detected (I, II, III and IV), being type I the most disseminated one. Gene blaZ was detected in 5.2% (13/250) isolates. From these 13 blaZ positive isolates, the whole system comprising blaR1-blaI-blaZ was detected in 23.1% (3/13) isolates.

No entanto, o uso inadequado de antibióticos no tratamento da doença pode selecionar cepas resistentes e comprometer a eficiência do tratamento. Bactérias do gênero Staphylococcus spp. estão entre os principais agentes etiológicos da mastite bovina e são freqüentemente resistentes aos antimicrobianos, em especial aos beta-lactâmicos, principalmente por dois mecanismos distintos: a produção da enzima extracelular beta-lactamase, codificada pelo gene blaZ, e a produção de PBP2a ou PBP2´, uma proteína ligante de penicilina de baixa afinidade, codificada pelo gene mecA. A expressão do gene mecA é constitutiva ou induzida por antibióticos betalactâmicos, como a oxacilina e cefoxitina. O gene mecA está inserido no cromossomo através de um elemento genético móvel, denominado cassete estafilocócico cromossômico mec (SCCmec). O presente estudo avaliou o perfil fenogenotípico de resistência aos beta-lactâmicos em 250 isolados de Staphylococcus spp., utilizando os marcadores oxacilina e cefoxitina, de modo a produzir dados que possam contribuir para o conhecimento da resistência antimicrobiana em algumas propriedades leiteiras das regiões Sul-Fluminense e Metropolitana do Estado do Rio de Janeiro com o objetivo de subsidiar a implementação de medidas de controle dessa enfermidade. A avaliação da resistência foi feita a partir de 8 diferentes testes fenotípicos, sendo obtidos 54 perfis. Os testes de difusão em disco simples e ágar screen com oxacilina foram utilizados como “padrão ouro” para os cálculos dos valores de sensibilidade, especificidade e predição por serem preconizados pelo CLSI veterinário. O teste de difusão em disco simples com cefoxitina foi o de melhor desempenho na predição da resistência a oxacilina. Na avaliação genotípica, não foi detectado qualquer isolado positivo para o gene mecA, já os genes mecI e mecRI foram detectados igualmente em 11,6% (29/250) dos Staphylococcus spp. avaliados. Foram detectados os quatro tipos de cassete mec analisados (I, II, III e IV), sendo o tipo I o que teve mais ampla distribuição entre as regiões estudadas. Gene blaZ foi detectado em 5,2% (13/250) dos isolados, sendo que nestes, todo o sistema blaZ-blaI- blaR1 foi detectado em 23,1% (3/13) dos isolados.

E-mail: miliane@ufrrj.br The present study evaluated the pheno- and genotypical antimicrobial resistance profile of coagulase-negative Staphylococcus (CNS) species isolated from dairy cows milk, specially concerning to oxacillin. Of 100 CNS isolates, the S. xylosus was the prevalent species, followed by S. cohnii, S. hominis, S. capitis and S. haemolyticus. Only 6% were phenotypically susceptible to the antimicrobial agents tested in disk diffusion assay. Penicillin and ampicillin resistance rates were significantly higher than others antimicrobials. Four isolates were positive to mecA gene (4%), all represented by the S. xylosus species. The blaZ gene was detected in 16% of the isolates (16/100). It was noticed that all mecA + were also positive to this gene and the presence of both genes was correlated to phenotypic beta-lactamic resistance. We conclude that CNS species from bovine milk presented significantly distinct antimicrobial resistance profiles, evaluated by phenotypic and genotypic tests, which has implications for treatment and management decisions.

Dos 100 isolados de ECN, S.xylosus foi a espécie predominante, seguida por S. cohnii, S. hominis, S. capitis and S. haemolyticus Apenas 6% dos isolados foram fenotipicamente suscetíveis aos agentes antimicrobianos testados no ensaio de difusão em disco. O percentual de resistência à penicilina e ampicilina foi significativamente maior que aos outros antimicrobianos. Quatro isolados foram positivos para o gene mecA (4%), sendo todos representados pela espécie S.xylosus. O gene blaZ foi detectado em 16% dos isolados (16/100), sendo todos mecA positivos e a presença de ambos os genes foi correlacionada com a resistência fenotípica aos beta-lactâmicos. Foi possível concluir que as espécies de ECN provenientes de leite bovino apresentaram distintos perfis de resistência antimicrobiana, avaliados por testes fenotípicos e genotípicos, podendo dificultar a adoção de medidas de tratamento e manejo dos animais.

E-mail: jrwilton@uag.ufrpe.br The objective of this study was to analyze the risk factors for bovine mastitis in the micro-region of Garanhuns, Pernambuco, Brazil. For this, was submitted for microbiologic exam 1260 milk samples and on each farm was applied a questionnaire about characteristics of animal breeds, general data from the farm, handling of cattle and sanitary measures for milking. To analyze the risk factors, a logistic regression was made as a dependent variable of the microbiologic exam. The microbiologic exams showed 477 (37.9%) positive samples and 783 (62.1%) negatives ones. The risk factors for bovine mastitis were: the semi-intensive breeding system (OR=2.3, p<0.0001), feeding during milking (OR=1.3, p=0.0101), antimicrobial shift (OR=1.3, p=0.00770), and dry cow therapy (OR=1.5; p=0.0013). The results show that the main risk factors are associated with handling to which the cattle is submitted. The knowledge of these factors allow to adopt prevention and control measures to avoid mastitis to improve animal productivity and gain within the production system.

Para isso, foram submetidas ao exame microbiológico 1260 amostras de leite e em cada propriedade foi aplicado um questionário contendo informações relacionadas às características raciais dos animais, dados gerais sobre as propriedades, manejo do rebanho e higiênico-sanitário durante a ordenha. Para a análise dos fatores de risco foi realizada uma regressão logística considerando como variável dependente o exame microbiológico. Ao exame microbiológico observou-se 477 (37,9%) amostras positivas e 783 (62,1%) negativas. Os fatores de risco associados à mastite foram: sistema de criação semi-intensivo (OR=2,3; p<0,0001), alimentação dos animais durante a ordenha (OR=1,3; p=0,0101), rodízio de antimicrobianos (OR=1,3; p=0,0077) e o uso da terapia da vaca seca (OR=1,5; p=0,0013). Os resultados obtidos demonstram que os principais fatores de risco associados à mastite estão relacionados ao manejo higiênico-sanitário ao qual o rebanho é submetido. O conhecimento desses fatores fornece subsídios para a elaboração de medidas de prevenção e controle para a enfermidade em questão, melhorando a produtividade dos animais e, consequentemente, a lucratividade do sistema de produção.

[Prevalence and etiology of bovine mastitis in the dairy region of Rondon do Pará, state of Pará.] Prevalência e etiologia da mastite bovina em rebanhos leiteiros na região de Rondon do Pará, estado do Pará. Pesquisa Veterinária Brasileira 31(2):104-110. E-mail: cmagno@ufpa.br The prevalence and the etiology of bovine mastitis in the dairy region of the county of Rondon of Pará were investigated. The sensibility and the resistance of the isolated agents to the antimicrobiotics was evaluetad; 237 mixed-bred milk cows were used on nine properties, which were hand milked once a day and kept on Brachiaria brizantha pasture, with supply of mineral salt and water ad libitum. Clinical examination of the mammary gland, the test of the sieve and California Mastitis Test were performed. Of the 935 examined mammary quarters, 6.6% had subclínical mastitis, 1.3% clinical mastitis, and 92.1% were negative. The isolated bacteria in the clinical mastitis were coagulase negative Staphylococcus spp. (25%), Staphylococcus aureus (16.7%), Streptococcus spp. (8,3%), and Corynebacterium spp. (8.3%). In the subclínical mastitis coagulase negative Staphylococcus spp. (32.3%), Staphylococcus aureus (17.7%), Staphylococcus intermedius (1.6%), Streptococcus spp. (4.8%), Corynebacterium spp. (4.8%), and coagulase negative Staphylococcus spp./S.aureus (1.6%) were isolated. There was no microbial growth in 41.7% of the samples with clinical mastitis and in 37.1% with subclínical mastitis. In the antibiogram, 100% of the isolated negative coagulase Staphylococcus spp., S. aureus, S. intermedius, and Streptococcus spp. were sensitive to sulfazotrim. On the other hand, Corynebacterium spp. were 100% resistant to this same antimicrobiotic. Cefalotina, cefoxitina and gentamicina were efficient against the isolated Staphylococcus spp. which represent the greater part of the mastitis agents in this study. Mastitis was diagnosed in all flocks studied, however the number of affected animals was considered low; this probably is due to low milk production of the cows and to permanence of the calf with the mother after milking, what favors emptying the mammary gland. But hygienic sanitary measures and management practices have to be improved.

Foram avaliadas 237 vacas mestiças de aptidão leiteira, pertencentes a nove propriedades, as quais utilizavam ordenha manual uma vez ao dia e sistema de criação extensivo em pastagens de Brachiaria brizantha, com fornecimento de sal mineral e água ad libitum. Realizou-se o exame clínico da glândula mamária, o teste da caneca telada e o California Mastitis Test. Dos 935 quartos mamários avaliados, 6,6% apresentaram mastite subclínica, 1,3% mastite clínica e 92,1% foram negativos. As bactérias isoladas na mastite clínica foram Staphylococcus spp. coagulase negativo (25%), Staphylococcus aureus (16,7%), Streptococcus spp. (8,3%) e Corynebacterium spp. (8,3%). Na mastite subclínica foram Staphylococcus spp. coagulase negativo (32,3%), Staphylococcus aureus (17,7%), Staphylococcus intermedius (1,6%), Streptococcus spp. (4,8%), Corynebacterium spp. (4,8%) e Staphylococcus spp. coagulase negativo/S. aureus (1,6%). Não houve crescimento microbiano em 41,7% das amostras com mastite clínica e 37,1% com mastite subclínica. No antibiograma, 100% dos isolados de Staphylococcus spp. coagulase negativo, S. aureus, S. intermedius, e Streptococcus spp. foram sensíveis ao sulfazotrim. Por outro lado Corynebacterium spp. foi 100% resistente ao mesmo antimicrobiano. A cefalotina, cefoxitina e gentamicina, apresentaram eficácia frente às bactérias isoladas do gênero Staphylococcus spp., as quais neste trabalho representam a grande maioria dos agentes causadores de mastite. A mastite foi diagnosticada em todos os rebanhos pesquisados, contudo o número de animais acometidos foi considerado baixo; isso provavelmente deve-se à baixa produção de leite dos animais e a permanência do bezerro ao pé após a ordenha, o que favorece o esvaziamento da glândula mamária. Diante disso, faz-se necessário que medidas higiênico-sanitárias e de manejo sejam adotadas.

PCR reactions were carried out using specific primers described previously for S. aureus (femA gene), S. intermedius (16S rDNA) and S. hyicus (16S-23S rDNA spacer region). In addition, products of amplification of variable regions of the 16S rDNA gene of the strains were sequenced. According to the results of the PCR, 83 strains were identified as S. aureus, 13 as S. intermedius, two as S. hyicus and two isolates were not identified. The sequencing of 16S rDNA was applied to 23 strains identified by PCR amplifications: six S. aureus and the strains identified as S. intermedius (n=13), S. hyicus (n=2) or not identified (n=2). The sequencing of 16S rDNA confirmed the six strains as S. aureus. The others 17 strains were identified as S. chromogenes (13 isolates) and S. hyicus (four isolates). Each sample was related to a specie according to the smallest E-value and highest similarity (³ 99%). The identification of S. hyicus and S. chromogenes was accomplished only by 16S rDNA sequencing.

Dos outros 17 isolados, 13 foram identificados como S. chromogenes e quatro como S. hyicus, com similaridade igual ou superior a 99%. Baseando-se nos resultados da reação de PCR do gene femA e do sequenciamento do rDNA 16S, foram identificados 83 S. aureus, 13 S. chromogenes e quatro S. hyicus. Neste estudo os oligonucleotídeos iniciadores empregados na reação de PCR para S. intermedius não foram específicos, pois amplificaram também S. chromogenes; e os empregados na reação de PCR para S. hyicus não foram sensíveis, pois falharam na identificação de dois isolados de S. hyicus. A identificação definitiva das duas últimas espécies somente foi possível pelo sequenciamento do rDNA 16S.

Nonetheless, cells active in the dry period are of fundamental importance for the involution of the mammary gland and its recovery for the following lactation. Since there are no specific drugs for dry goat therapy, the dry cow therapy drugs tend to be misused in goats despite of the anatomical and physiological differences apart from the particularities of the two species. The objective of the present study was to evaluate the influence of five drugs specific for dry cow therapy on the function of goat milk phagocytes. Somatic cells were isolated from 20 milk samples of 10 lactating goats that had not been treated for mastitis during the previous 30 days. Milk samples were collected properly and microbiological culture yielded negative results. Cells adherent to glass coverslips were treated with commercially available dry cow therapy drugs containing active principles such as Gentamicin (M1), Cephalonium anhydrous (M2), Ampicillin (M3), Cloxacillin benzathine (M4) and Cephapirin benzathine (M5). Phagocytosis of Zymosan particles was evaluated. Mean phagocytosis indexes of cells treated with M2 (15.12% ± 16.22), M3 (6.02% ± 7.96), M4 (4.54% ± 5.45) and M5 (2.47% ± 4.64) were lower (p<0.001) than mean phagocytosis index of the control group (40.67% ± 19.68). Mean phagocytosis index of cells treated with M2 was greater (p<0.05) than those treated with M3, M4 and M5, whereas means of the latter three treatments were statistically similar. M1-treated cells did not adhere adequately to the cover slips, making it impossible to evaluate phagocytosis in this group. The results obtained enable the conclusion that drugs used affected milk phagocytes negatively. However, interference on somatic cell function is not the sole factor determining an unsuccessful dry period therapy, since the efficacy of the active principle on pathogens responsible for infectious processes also has to be considered.

Para tal, fez-se o isolamento de células somáticas de 20 amostras de leite provenientes de 10 cabras lactantes, sem antecedentes de tratamento de mamite nos últimos 30 dias, sob condições higiênico-sanitárias de colheita e com resultados negativos ao cultivo microbiológico do leite. As células aderidas a lamínulas de vidro foram confrontadas com formulações contendo princípios ativos disponíveis no mercado como Gentamicina (M1), Cefalônio Anidro (M2), Ampicilina (M3), Cloxacilina Benzatínica (M4) e Cefapirina Benzatínica (M5). Avaliou-se, por microscopia, a fagocitose de partículas de Zymosan. As médias dos índices de fagocitose das células submetidas ao tratamento com M2 (15,12% ± 16,22), M3 (6,02% ± 7,96), M4 (4,54% ± 5,45) e M5 (2,47% ± 4,64) foram menores (p<0,001) que a média dos índices de fagocitose do grupo controle (40,67% ± 19,68). A média dos índices de fagocitose das células submetidas ao tratamento com M2 foi maior (p<0,05) que as médias dos tratamentos com M3, M4 e M5 enquanto estas foram estatisticamente iguais entre si. As amostras celulares submetidas ao medicamento M1 exibiram adesão insuficiente ou ausente às lamínulas, inviabilizando a avaliação da fagocitose por meio da técnica utilizada. Os resultados obtidos permitem concluir que os medicamentos estudados exerceram influência negativa sobre os fagócitos do leite, porém, esta interferência sobre as funções das células somáticas não pode por si só determinar o insucesso da terapia proposta para o período seco, pois deve ser considerada, outrossim, a eficácia do princípio ativo sobre o patógeno causador do processo infeccioso.

A total of 60 isolates of Staphylococcus spp., identified as Staphylococcus aureus (50) and Positive coagulase Staphylococcus (10) were obtained from the mammary glands of dairy cows with subclinical mastitis in the regions of Metropolitan Recife, the Agreste and the Zona da Mata of the State of Pernambuco, Brazil. As active ingredients were used a chlorine base (25%), iodine (0.6%), chlorhexidine (2.0%), quaternary ammonium (4.0%), and lactic acid (2.0%) at four specific intervals (15", 30", 60", and 300"). One hundred percent of S. aureus was found to be sensitive to iodine, 93.3% to chlorhexidine, 80% to ammonia, 35.6% to lactic acid, and 97.8% were resistant to chlorine at a 60-minute interval. With respect to the Positive coagulase Staphylococcus (SCP), 100.0% of the isolates were sensitive to iodine, 81.8% to quaternary ammonium, 99.9% to lactic acid, 72.7% to chlorhexidine, and 100% was resistant to chlorine at an interval of 60 minutes. It can be concluded that the highest disinfectant activity in vitro was found to be with iodine and chlorhexidine for S. aureus, and with iodine and lactic acid for SCP. #19 - Mastitis caused by Mycoplasma bovis in dairy cattle, 21(4):143-145

[Mastitis caused by Mycoplasma bovis in dairy cattle] Mastite bovina por Mycoplasma bovis em rebanhos leiteiros. Pesquisa Veterinária Brasileira 21(4):143-145. Depto Medicina Veterinária Preventiva, Universidade Estadual de Londrina, Cx. Postal 6001, Londrina, PR 86051-970, Brazil. In this study 713 cows were examined. The animals were from three dairy farms in northern Paraná and the southwest of the State of São Paulo. From these cows, 137 had mastitis. On the three farms, 8 cows (1-12%) with Mycoplasma bovis mastitis were detected. Four were treated with tylosin and oxytetracyçlin and three with enrofloxacin There was no response to the treatments, and these animals were culled during the lactation period. One cow treated with enrofloxacin almost totally recovered milk production, but elimination of M. bovis continued during the lactation, and there was no bacteriological cure. This cow had a normal milk production in the next lactation period, without elimination of M. bovis. Culling of positive animals, the bacteriological study anel correct application of preventive practices for contagious mastitis controlled the dissemination of M. bovis to other animals.

Foram examinadas 713 vacas de três rebanhos leiteiros localizados na região norte do Estado do Paraná e sudoeste do Estado de São Paulo, das quais 137 apresentaram mastite. Nas três propriedades foram detectados oito animais (1, 12%) com mastite clínica por Mycoplasma bovis. Destes animais, quatro tratados com oxitetraciclina e tilosina e três com enrofloxacina, não responderam ao tratamento e foram descartados no decorrer da lactação. Uma vaca medicada com enrofloxacina recuperou quase que totalmente a secreção láctea mas a eliminação de M. bovis persistiu por toda lactação. Esta vaca apresentou cura bacteriológica na lactação seguinte. O descarte dos animais positivos, monitoramento bacteriológico e a aplicação correta das me9idas de prevenção para as mastites contagiosas controlaram a disseminação de M. bovis nos rebanhos.


#20 - Evaluation of the sensitivity of bulk tank milk cultures for the isolation of contagious bovine mastitis pathogens, 18(1):39-44

[Evaluation of the sensitivity of bulk tank milk cultures for the isolation of contagious bovine mastitis pathogens.] Avaliação da sensibilidade da cultura de leite do tanque para isolamento de agentes contagiosos da mastite bovina. Pesquisa Veterinária Brasileira 18(1):39-44. Embrapa-Centro Nacional de Pesquisa de Gado de Leite, Juiz de Fora, MG 36038-330, Brazil. Samples of bulk tank milk from 33 herds were collected at the dairy processing plant and cultured, as a means of detecting specific (contagious) bovine mastitis pathogens. Somatic cell counts (SCC) were made on a Fossomatic 90. Two and three weekly consecutive samples were obtained from 13 and 12 herds, respectively. Only one sample was examined from eight herds. Three daily consecutive samples of bulk milk and individual quarter samples from all lactating cows from four herds (A, B, C and D) were also examined. Milk from individual quarters were cultured on blood agar while tank milk samples were cultured on TKT, Mannitol Salt, MacConkey agars and Sabouraud containing chloramphenicol. Staphylococcus aureus was recovered from 26 of the 33 herds sampled in the dairy processing plant. Nine of these samples also contained Streptococcus agalactiae. Nine herds had SCC above 500,000 ml1 The remaining 23 herds had SCC levels below 400,000 ml1 S. aureus and S. agalactiae were isolated from five of the nine herds with high SCC, S. agalactiae from one and S. aureus from three. Six herds had SSC below 200,000 ml1 S. aureus and S. agalactiae were isolated from one, S. aureus from three, while the other two were negative for both pathogens. The results of herds A, B, C and D sampled at the farms showed that S. aureus was isolated from 1.8%, 19.2%, 17.0% and 8.4% of the animals and 0.9%, 5.9%, 5.4% and 2.2% of the mammary quarters, respectively. S. agalactiae was isolated from herds A, C and D. Within these herds the percentages of isolation were, respectively, 1.8%, 10.6% and 8.4% for the cows and 0.46%, 3.8% and 3.7% for the mammary quarters. S. aureus was recovered from all three bulk tank cultures from herds A, B and D. Only the third sample from herd C was positive for S. aureus. S. agalactiae was recovered from all samples collected from herd D, two samples from herd C and one sample from herd A. Coliforms were isolated from all tank samples from herds A, B, C and D and from all but one sample collected in the processing plant. Yeasts were recovered from 16 herds sampled at the processing plant and from all tank samples from herds A, B, C, and D. Neither coliforms or yeasts were isolated from the individual animals of herds A, B, C and D. These findings indicate that the milk was contaminated during or after milking, probably due to deficient hygiene and cleaning procedures. The analysis of the bulk tank milk cultures showed that the test was sensitive enough to detect contagious mastitis pathogens. The sensitivity of the test increased when more than two consecutive samples were examined.

Amostras de leite total (leite do tanque) de 33 rebanhos foram coletadas na plataforma de recepção da indústria laticinista e cultivadas para detectar patógenos específicos (contagiosos) da mastite. Foi feita a contagem de células somáticas (CCS) das amostras utilizando o equipamento Fossomatic 90. Em 13 e 12 rebanhos avaliaram-se duas e três amostras semanais consecutivas, respectivamente, e em oito avaliou-se apenas uma. Foram também examinadas três amostras diárias consecutivas do leite do tanque e amostras dos quartos mamários individuais, coletadas na própria fazenda, de todas as vacas em lactação de quatro rebanhos (A, B, C e D). As amostras de leite dos quartos mamários individuais foram cultivadas em ágar sangue e as amostras do tanque, em placas de TKT, Sal Manitol, MacConkey e Sabouraud contendo cloranfenicol. Dos 33 rebanhos cujas amostras foram obtidas na plataforma de recepção da indústria, isolou-se Staphylococcus aureus de 26, nove desses em associação com Streptococcus agalactiae e em três rebanhos isolou-se somente S. agalactiae. Nove rebanhos tiveram CCS acima de 500.000 ml1 e 21, abaixo de 400.000 ml1 Em cinco dos nove rebanhos com CCS acima de 500.000 m1-1 foram isolados S. aureus e S. agalactiae, em três, apenas S. aureus e em um, apenas S. agalactiae. Seis rebanhos apresentaram CCS abaixo de 200.000 ml·1; de um deles foram isolados S. aureus e S. agalactiae, de três, S. aureus e os outros dois foram negativos para estes dois patógenos. Os resultados encontrados nos quatro rebanhos cujas amostras foram coletadas na própria fazenda mostraram que S. aureus foi isolado nas seguintes porcentagens dos animais: 1,8%, 19,2%, 17,0% e 8,4% e dos quartos mamários: 0,9%, 5,9%, 5,4% e 2,2%, respectivamente, para os rebanhos A, B, C e D. S. agalactiae foi isolado dos rebanhos A, C e D. Leveduras foram isoladas de 16 amostras coletadas na indústria e de todas amostras do tanque dos rebanhos A, B, C e D. Não foram isolados coliformes ou leveduras dos quartos mamários dos animais destes rebanhos, sugerindo que ocorreu contaminação do leite durante ou após a ordenha, provavelmente devido a deficiências nos processos de limpeza e higienização. A análise dos resultados das culturas do leite do tanque mostrou que o exame foi específico para detectar os patógenos contagiosos da mastite. A sensibilidade do teste aumentou quando se examinaram mais de duas amostras consecutivas.
