#1 - Efficacy of disinfectants to inactivate H1N1 influenza A virus isolated from pigs

Abstract in English:

The aim of this study was to access the efficacy of four disinfectants to inactivate influenza A [H1N1] 0 hour and 72 hours after disinfectant dilution. A pandemic H1N1 influenza virus isolated from a pig with respiratory disease was used to obtain inoculums containing 6.4log10 EID50/mL; 5.4log10 EID50/mL; 4.4log10 EID50/mL and 3.4log10 EID50/mL. Suspension test was composed of 400µL of viral inoculum, 100µL of organic load and 500µL of each individually diluted disinfectant and incubated for ten minutes of contact time. After a neutralizing step, each mixture was filtered on a 0.22µm membrane and 0.2mL was inoculated in six 9-day-old embryo chicken egg through allantoic route. The allantoic fluid from eggs was harvest for RT-PCR and hemagglutination test. The experiment was repeated 72 hours after disinfectant dilution. On the first assessment with fresh disinfectant, influenza virus was inactivated by oxidizing compost disinfectant and phenolic disinfectant in all virus concentrations, the quaternary ammonium compound (QAC) and glutaraldehyde association inactivated the virus up to a concentration of 5.4log10 EID50/mL. QAC disinfectant did not eliminate virus viability. Seventy-two hours after disinfectants were diluted, oxidizing compost disinfectant and QAC and glutaraldehyde association disinfectant demonstrated the same result as the evaluation with fresh disinfectant solution. Phenolic disinfectant inactivated viral inoculum up to a concentration of 5.4log10 EID50/mL. QAC had no effect on inactivating 3.4log10 EID50/mL of influenza virus. In conclusion, three of the four disinfectants tested were effective to inactivate pandemic H1N1 influenza virus in the presence of organic load. Test result performed 72hours after disinfectant dilution suggest a decrease in the effectiveness of one disinfectant.

O objetivo deste trabalho foi avaliar a eficácia de quatro desinfetantes em inativar o vírus da influenza A [H1N1] 0-hora e 72-horas após a diluição dos produtos. Um vírus H1N1 pandêmico isolado previamente de um suíno com doença respiratória foi utilizado e foram obtidas quatro concentrações de inóculo contendo 6,4log10 EID50/mL; 5,4log10 EID50/mL; 4,4log10 EID50/mL and 3,4log10 EID50/mL. Para compor o teste em suspensão foram adicionados 400µL de inóculo viral, 100µL de matéria orgânica e 500µL de cada desinfetante diluído individualmente e a mesma foi incubada por 10 minutos. Após a etapa neutralizante, a suspensão foi filtrada em membrana 0,22µm e 0,2mL foi inoculado em seis ovos de galinha embrionados de nove dias de incubação, via rota alantóide. O fluido alantóide foi colhido após 72 horas para testes de hemaglutinação e RT-PCR. O mesmo protocolo experimental foi repetido usando as soluções desinfetantes 72 horas após a diluição. O vírus da influenza foi inativado pelo composto oxidante e também pelo desinfetante fenólico em todas as concentrações virais testadas 0-hora após diluição. O desinfetante com associação de amônia quaternária e glutaraldeído inativou o vírus na concentração de até 5,4log10 EID50/mL. O desinfetante à base de amônia quaternária não inativou o vírus. Os resultados 72-horas após a diluição não diferiram quando comparado com 0-hora, exceto o desinfetante fenólico, o qual inativou o vírus da influenza somente até a concentração 5,4log10 EID50/mL. Concluindo, três dos quatro desinfetantes testados foram efetivos ao inativar o vírus da influenza [H1N1] pandêmico na presença de matéria orgânica. Os resultados do teste com produtos diluídos após 72 horas sugerem redução da efetividade em, pelo menos, um desinfetante.

E-mail: giseleveneroni@yahoo.com.br To verify the occurrence of caseous lymphadenitis in sheep and goats on farms of Pernambuco, Brazil, and in animals slaughtered in two Brazilian cities (Petrolina/PE and Juazeiro/BA), and to characterize the susceptibility profile of Corynebacterium pseudotuberculosis to disinfectants and antimicrobials, and its relationship with biofilm production were the objectives of this study. 398 samples were tested for sensitivity to antimicrobial drugs, disinfectants, and biofilm production. Among the 108 samples collected on the properties, 75% were positive for C. pseudotuberculosis. Slaughterhouse samples indicated an occurrence of caseous lymphadenitis in 15.66% and 6.31% for animals slaughtered in Petrolina and Juazeiro respectively. With respect to antimicrobials, the sensitivity obtained was 100% for florfenicol and tetracycline; 99.25% for enrofloxacin, ciprofloxacin and lincomycin; 98.99% for cephalothin; 98.74% for norfloxacin and sulfazotrim; 97.74% for gentamicin; 94.22% for ampicillin; 91.71% for amoxicillin; 91.21% for penicillin G; 89.19% for neomycin and 0% for novobiocin. In analyzes with disinfectants, the efficiency for chlorhexidine was 100%, 97.20% for quaternary ammonium, 87.40% for chlorine and 84.40% for iodine. 75% of the isolates were weak or non-biofilm producers. For the consolidated biofilm, found that iodine decreased biofilm formation in 13 isolates and quaternary ammonia in 11 isolates. The reduction of the biofilm formation was observed for iodine and quaternary ammonium in consolidated biofilm formation in 33% and 28% of the isolates, respectively. The results of this study highlight the importance of establishing measures to prevent and control the disease.

Entre as 108 amostras coletadas nas propriedades, 75% foram positivas para C. pseudotuberculosis. Amostras do matadouro indicaram a ocorrência de linfadenite caseosa em 15,66% e 6,31% para animais abatidos em Petrolina (PE) e Juazeiro (BA), respectivamente. Com relação aos antimicrobianos, a sensibilidade obtida foi de 100% para florfenicol e tetraciclina, 99,25% para a enrofloxacina, ciprofloxacina e lincomicina; 98,99% para cefalotina; 98,74% para norfloxacina e sulfazotrim; 97,74% para gentamicina; 94,22% para ampicilina, 91,71% para amoxicilina, 91,21% para a penicilina G, 89,19% para a neomicina e 0% para novobiocina. Nas análises com desinfetantes, a eficiência para a clorexidina foi de 100%, 97,20% para amônia quaternária, 87,40% para o cloro e 84,40% para o iodo. 75% dos isolados foram fracos produtores de biofilme. Para o biofilme consolidado, observou-se que o iodo diminuiu a formação de biofilme em 13 isolados e a amônia quaternária em 11 isolados. A redução da formação de biofilme foi observada no biofilme consolidado, para iodo e amônia quaternária, em 33% e 28% das amostras, respectivamente. Os resultados deste estudo destacam a importância de se estabelecer medidas para prevenir e controlar a doença.

E-mail: sampaio.elizabeth@gmail.com The objective of this study was to evaluate the in vitro efficacy of commercial disinfectants used in pre- and post-dipping, against Staphylococcus spp. isolated from milk originating from dairy cattle farms in the Wasteland and Forest Zone of Alagoas, Brazil. We used iodine (0.57%), chlorhexidine (2.0%), chlorine (2.5%) and quaternary ammonium compound (4.0%) at concentrations indicated, conventionally used as commercial disinfectants before and after dipping. We analyzed a total of 97 isolates of Staphylococcus spp. identified as S. aureus (16), coagulase positive Staphylococcus (7) and coagulase-negative Staphylococcus (74). The disinfectants were evaluated at three different times (15”, 30” and 60”). We found that 56.3% of Staphylococcus aureus was sensitive to iodine, 68.8% to chlorine, 87.5% to chlorhexidine, and 37.5% to the compound of ammonia, in time 60”. As for coagulase positive staphylococci (CPS), 100% of the isolates was resistant to chlorhexidine, 85.7% to the ammonia compound, 57.1% to chlorine, and 42.9% iodine, in time 60”. Regarding coagulase negative staphylococci (CNS), 91.9% was sensitive to chlorhexidine, 70.3% to chlorine, 66.2% to iodine, and 24.3% the ammonium compound, at time 60”. It is concluded from this study that the greatest disinfectant activity in vitro was with chlorhexidine and chlorine for S. aureus, with iodine and chlorine for SCP, and with chloride and chlorhexidine for SCN. Due to variations in the sensitivity and resistance profile found, it is necessary for regular assessments of the effectiveness of disinfectants used on the farms, to observe the effectiveness of the product and thus ensure the control of mastitis in the herd.

Analisou-se um total de 97 isolados de Staphylococcus spp. identificados como S. aureus (16), Staphylococcus coagulase positiva (7) e Staphylococcus coagulase negativa (74). Os desinfetantes foram avaliados em três tempos distintos (15”, 30” e 60”). Observou-se que 56,3% de Staphylococcus aureus foram sensíveis ao iodo, 68,8% sensíveis ao cloro, 87,5% à clorexidine e 37,5% ao composto de amônia no tempo de 60”. Quanto aos Staphylococcus coagulase positiva (SCP), 100% dos isolados foram resistentes ao clorexidine, 85,7% ao composto de amônio, 57,1% ao cloro, e 42,9 resistentes ao iodo no tempo de 60”. Conclui-se com esse estudo que a maior atividade desinfetante in vitro foi verificada para clorexidine e cloro frente aos S. aureus, iodo e cloro para os SCP e clorexidine e cloro para os SCN. Devido às variações no perfil de sensibilidade e resistência encontradas, é necessária a avaliação regular da eficiência dos desinfetantes usados nas propriedades, com o intuito de observar a eficácia do produto e assim garantir o controle da mastite no rebanho.


#4 - Susceptibility to disinfectants and antimicrobial resistance profile in Escherichia coli isolates

Abstract in English:

Colibacillosis caused by Escherichia coli is the most important enteric disease in pig production, which may lead to death of the affected animal. The bacterium has a great ability to develop resistance to multiple antibiotics and disinfectants. Thus, investigation addressing mechanisms of resistance and profile of field samples is necessary. E. coli is widely used as a model for studies that explore the intrinsic and extrinsic resistance to multidrugs. In this paper, we attempt to associate the susceptibility profile of 62 isolates of E. coli to three disinfectants and 13 antimicrobials. Also 31 isolates were tested for the presence of efflux mechanism. Of the three disinfectants tested, alkyldimethylbenzylammonium chloride+nonyl phenoxy polyethoxy ethanol was the most effective (100%), followed by glutaraldehyde+alkyldimethylbenzylammonium chloride (95.2%) and alkyldimethylbenzylammonium chloride (88.8%). Among the antimicrobials tested, there was greater resistance to tetracycline (62.2%) and higher sensitivity to florfenicol (88.6%). The high sensitivity of the isolates against disinfectants may be related to the absence of efflux mechanism. The average index of multiple resistance to antimicrobials was 0.52, what demonstrates a profile of multidrug resistant isolates, showing the need for rational use of these drugs in pig production.

A colibacilose, causada por Escherichia coli, é a enfermidade entérica de maior impacto na produção de suínos, podendo levar à morte do animal. Esta bactéria possui grande capacidade de desenvolver resistência a múltiplos antimicrobianos e a desinfetantes. Desta forma, estudos que abordem mecanismos de resistência e perfil de amostras de campo tornam-se necessários. E. coli é amplamente utilizada como modelo de estudos que exploram a resistência intrínseca e extrínseca a multidrogas. Neste trabalho, buscou-se verificar o perfil de sensibilidade de 62 isolados de E. coli de suínos frente a três desinfetantes e a 13 antimicrobianos. Ainda, em 31 destes isolados foi pesquisada a presença de mecanismo de efluxo. Dos três desinfetantes avaliados, o cloreto de alquil dimetil benzil amônio+poliexietilenonilfenileter foi o que se mostrou mais eficaz (100%), seguido do glutaraldeído+cloreto de alquil dimetil benzil amônio (95,2%) e do cloreto de alquil dimetil benzil amônio (88,8%). Dentre os antimicrobianos testados, observou-se maior resistência para a tetraciclina (62,2%) e maior sensibilidade para o florfenicol (88,6%). A alta sensibilidade dos isolados frente aos desinfetantes pode estar relacionada à ausência de mecanismo de efluxo. O índice de resistência múltipla médio aos antimicrobianos foi de 0,52, o que demonstra um perfil multirresistente dos isolados, conduzindo para a necessidade do uso racional destas drogas em suinocultura.

After identification, 12 yeast isolates were tested with the following disinfectants: iodine at 0,57%, chlorine at 2,5%, chlorhexidine, lactic acid and quaternary ammonium at 2,0%, at different exposure times. It was observed that 100% of the isolates were sensitive to chlorhexidine at all exposure times. Iodine has obtained the second best result with the following percentages: 83,33% were susceptible at 15’’ exposure, 91,7% in 30’’ and 100% were sensitive at 60’’ 300’’ and 600’’ times. For lactic acid, the results were 41,67% sensitive at 15’’ of exposure, 58,33% at 30’’, 66,7% at 60’’ and 300’’ and 75% at 600’’. In the analysis of the results for quaternary ammonium, it was observed that 66,67% were sensitive in 15’’and 30’’ times and 91,67% at 60’’, 300’’ and 600’’. Regarding the chlorine activity, only 16,67% were sensitive at the 15’’ and 30’’ times, and 25% at 60’’, 300’’ and 600’’. It was concluded that chlorhexidine and iodine have disinfectant activity significantly superior to chlorine, against to yeast involved in infectious processes of the mammary gland in cattle. It is necessary to perform a periodic assessment of the disinfectant activity from products used in the pre and post-dipping routine against dairy farms microorganisms most commonly involved in cases of mastitis (bacterial or fungal) which is an effective alternative to reduce the frequency of mastitis in cattle.

Observou-se que 100% dos isolados mostraram-se sensíveis ao clorexidine, em todos os tempos de exposição. O iodo obteve o segundo melhor resultado com os seguintes percentuais: 83,33% foram sensíveis em 15” de exposição, 91,67% em 30” e 100% foram sensíveis nos tempos 60”, 300” e 600”. Para o ácido láctico, os resultados foram: 41,67% sensíveis em 15” de exposição, 58,33% em 30”, 66,67% em 60” e 300” e 75% em 600”. Na Análise dos resultados para amônia quaternária, observou-se que 66,67% foram sensíveis nos tempos 15” e 30”, e 91,67% em 60”, 300” e 600”. Em relação ao princípio ativo cloro apenas 16,67% foram sensíveis no tempo de 15” e 30”, e 25% em 60”, 300” e 600”. Conclui-se que o clorexidine e o iodo apresentam atividade desinfetante significativamente superior ao cloro, frente a leveduras envolvidas nos processos infecciosos da glândula mamária em bovinos. É necessário realizar uma avaliação periódica da atividade desinfetante dos produtos utilizados na rotina do pré e pós-dipping nas propriedades leiteiras frente aos microrganismos mais comumente envolvidos nos casos de mastite (micótica ou bacteriana), pois esta é uma medida eficaz para reduzir a frequência de casos de mastite nos rebanhos.

E-mail: rinaldo.mota@hotmail.com The objective of this investigation was to evaluate the in vitro sensibility of Staphylococcus spp. to several commercially available disinfectants used for pre and post dipping. A total of 60 isolates of Staphylococcus spp., identified as Staphylococcus aureus (50) and Positive coagulase Staphylococcus (10) were obtained from the mammary glands of dairy cows with subclinical mastitis in the regions of Metropolitan Recife, the Agreste and the Zona da Mata of the State of Pernambuco, Brazil. As active ingredients were used a chlorine base (25%), iodine (0.6%), chlorhexidine (2.0%), quaternary ammonium (4.0%), and lactic acid (2.0%) at four specific intervals (15", 30", 60", and 300"). One hundred percent of S. aureus was found to be sensitive to iodine, 93.3% to chlorhexidine, 80% to ammonia, 35.6% to lactic acid, and 97.8% were resistant to chlorine at a 60-minute interval. With respect to the Positive coagulase Staphylococcus (SCP), 100.0% of the isolates were sensitive to iodine, 81.8% to quaternary ammonium, 99.9% to lactic acid, 72.7% to chlorhexidine, and 100% was resistant to chlorine at an interval of 60 minutes. It can be concluded that the highest disinfectant activity in vitro was found to be with iodine and chlorhexidine for S. aureus, and with iodine and lactic acid for SCP. A further conclusion was that it is important to undertake a periodic evaluation of the disinfectants used on the dairy properties in the regions studied, given the variety of sensibilities and resistance to disinfectants used, which may prejudice the control of bovine mastitis caused by Staphylococcus spp.

E-mail: rinaldo.mota@hotmail.com The objective of this investigation was to evaluate the in vitro sensibility of Staphylococcus spp. to several commercially available disinfectants used for pre and post dipping. A total of 60 isolates of Staphylococcus spp., identified as Staphylococcus aureus (50) and Positive coagulase Staphylococcus (10) were obtained from the mammary glands of dairy cows with subclinical mastitis in the regions of Metropolitan Recife, the Agreste and the Zona da Mata of the State of Pernambuco, Brazil. As active ingredients were used a chlorine base (25%), iodine (0.6%), chlorhexidine (2.0%), quaternary ammonium (4.0%), and lactic acid (2.0%) at four specific intervals (15", 30", 60", and 300"). One hundred percent of S. aureus was found to be sensitive to iodine, 93.3% to chlorhexidine, 80% to ammonia, 35.6% to lactic acid, and 97.8% were resistant to chlorine at a 60-minute interval. With respect to the Positive coagulase Staphylococcus (SCP), 100.0% of the isolates were sensitive to iodine, 81.8% to quaternary ammonium, 99.9% to lactic acid, 72.7% to chlorhexidine, and 100% was resistant to chlorine at an interval of 60 minutes. It can be concluded that the highest disinfectant activity in vitro was found to be with iodine and chlorhexidine for S. aureus, and with iodine and lactic acid for SCP. A further conclusion was that it is important to undertake a periodic evaluation of the disinfectants used on the dairy properties in the regions studied, given the variety of sensibilities and resistance to disinfectants used, which may prejudice the control of bovine mastitis caused by Staphylococcus spp.
