#11 - Characterization of subclinical mastitis in dairy goats herds raised on an organic system in Rio Grande do Sul, 29(9):774-778

E.mail: veronica.schmidt@ufrgs.br Milk and derivates are recognized as vehicle of different pathogens for humans. These micoorganisms may manifest as post-milking contamination or as in infections in the animal itself, particularly in mastitis. The aim of present study was characterizes the occurrence of mastitis and management aspects in dairy-goats raised in three farms of organic milk. The correlation between mastitis and goat husbandry was also investigated. The clinical exam of the mammary glands of 64 dairy-goats did not reveal the presence of clinical mastitis. Nevertheless, the California Mastitis Test (CMT) identified 54 (22.7%) reactive mammary halves (+ or ++). From 238 milk samples collected, 37 (15.6%) were positive for bacterial isolates. Coincident positive results for bacterial isolate tests and CMT were observed in only eight samples, which indicate a sensitivity index of 21.6% for the caprine subclinical mastitis diagnosis test. Coagulase-negative staphylococci (CNS) were identified as the most frequent pathogen in milk samples (83.8%). The in vitro sensitivity test revealed CNS strains resistant to cotrimoxazole (50%), ampicillin (48.1%), nitrofurantoine (7.7%), cefaclor (7.14%), and oxacilline (3.85%). Cefalotine, gentamicin, neomycin, streptomycin, and tetracycline were the most effective drugs. No correlation was observed between the occurrence of subclinical mastitis and race, lactation period, drinking water quality in farms, or milking system as adopted in the present study.

Foi estudada a ocorrência de mastite e aspectos do manejo em cabras de três propriedades criadas em sistema orgânico. O exame clínico da glândula mamária de 64 cabras em diferentes períodos de lactação, não acusou a presença de mastite clínica. Entretanto, o Califórnia Mastitis Test (CMT) identificou 54 (22,7%) metades mamárias reagentes (+ ou ++). Foram colhidas 238 amostras de leite, das quais houve isolamento bacteriano em 37 (15,6%). Em apenas oito amostras houve coincidência entre o isolamento bacteriano e o resultado do CMT, indicando sensibilidade de 21,6% para este teste no diagnóstico de mastite subclínica em caprinos. Staphylococcus coagulase negativa (SCN) foi o microrganismo mais freqüente (83,8%). O teste de sensibilidade microbiana in vitro revelou resistência das linhagens de SCN ao cotrimoxazol (50%), ampicilina (48,1%), nitrofurantoína (7,7%), cefaclor (7,14%) e oxacilina (3,85%). Cefalotina, gentamicina, neomicina, estreptomicina e tetraciclina foram os antimicrobianos mais efetivos frente aos isolados. Não se evidenciou relação entre a ocorrência de mastite subclínica com a raça, a fase de lactação, sistema de ordenha ou qualidade da água utilizada nas propriedades.

From the 291 isolates, 170 (58.4%) were identified as negative coagulase Staphylococcus (SCN), 84 (28.9%) as Staphylococcus aureus, and 37 (12.7%) as positive coagulase Staphylococcus (SCP). To study sensitivity to antimicrobials, the diffusion in disks method was used with 16 antimicrobial drugs commonly employed in the treatment of mastitis. The most efficient antibiotic in vitro was the combination of neomicine + bacitracine + tetracycline with percentages of 98.4%, 99.3%, and 89.7% for the A, B, and C regions, respectively. The least efficient was ampicillin, which was resistant to 56.5% of the isolates taken from region A, 72.8% from region B, and 71.8% from region C. These results indicate the need for periodic testing of sensitivity in vitro, as these variations can compromise the treatment of animals as well as control programs for bovine mastitis caused by Staphylococcus spp.

Tests were made during 2 years in Holstein cows of a dairy farm producing type C milk, where milking was performed once a day. Sensitivities of electrical conductivity and chloride concentration tests from mammary quarters, where Corynebacterium sp was isolated (65.3% and 78.3%, respectively), were superior to the found in mammary quarters where S. aureus was identified (55.4% and 68.2%, respectively). The efficacies of the two diagnostic tests were similar. Statistical significance was demonstrated with regression analysis of both tests of healthy mammary quarters and subclinical mastitis quarters infected with Staphylococcus aureus.

[Profile of antimicrobial susceptibility in strains of Gram positive cocci, negative catalase, isolated from buffalo subclinical mastitis.] Perfil de susceptibilidade a antimicrobianos em amostras de cocos Gram-positivos, catalase negativos, isoladas de mastite subclínica bubalina. Pesquisa Veterinária Brasileira 23(2):47-51. Depto Epidemiologia e Saúde Pública, Instituto de Veterinária, UFRRJ, Seropédica, RJ 23890-000, Brazil. The susceptibility of antimicrobials was studied in Gram positive and catalase negative cocci (21 samples of Lactococcus garvieae and 6 Enterococcus gallinarum), isolated from the milk of cows with subclinical mastitis, belonging to six buffalo herds in the State of Rio de Janeiro. The test used was diffusion of disks in agar Müller Hinton, according to recommendations of the National Committee for Clinicai Laboratory Standards - NCCLS. There were tested disks with ampicillin (10mg), cefalotin (30mg), cefotaxime (30mg), cefoxitin (30mg), doranfenicol (30mg), eritromycin (15mg), gentamycin (10mg), nitrofurantoin (300mg), norfloxacin (10mg), penicillin (1 O IU), tetracydin (30mg) and vancomycin (30mg). The results showed that with Lactococcus garvieae, the most efficient antimicrobial was nitrofurantoin, revealing 85.71% sensibility, followed by cefotaxime (61.90%), vancomycin (52.38%), norfloxacin (47.62%) and cefalotin (47.62%). The highest resistance was developed against penicillin and ampicillin, with 95.24% resistance for the two antimicrobials. The susceptibility profile developed by the strains of Enterococcus gallinarum showed low sensibility against the tested antimicrobials; the highest resistance observed was against eritromycin and gentamycin, with 33.34% sensibility for both. The antimicrobial evaluation showed 100% resistance against vancomycin and tetracyclin, followed by cloranfenicol, penicillin, ampicillin, cefoxitin, cefotaxim, norfloxacin and nitrofurantoin; all of them showed a resistance of 83.33% with the samples tested.

Estudou-se o perfil de susceptibilidade a antimicrobianos em cocos Gram-positivos catalase negativos (21 amostras de Lactococcus garvieae e 6 de Enterococcus gallinarum), isoladas do leite de fêmeas com mastite subdínica e pertencentes a uma população composta por seis rebanhos bubalinos localizados no Estado do Rio de Janeiro. O teste utilizado foi o da difusão de discos em agar Müller Hinton, segundo recomendações do National Committee for Clinicai Laboratory Standards - NCCLS, tendo sido testados discos com ampicilina (10mg), cefalotina (30mg), cefotaxima (30mg), cefoxitina (30mg), cloranfenicol (30mg), eritromicina (15mg), gentamicina (10mg), nitrofurantoína (300mg), norfloxacina (10mg), penicilina (10 UI), tetracidina (30mg) e vancomicina (30mg). Os resultados evidenciaram que em se tratando de Lactococcus garvieae, o antimicrobiano mais eficiente foi o nitrofurantoína com 85,71% de sensibilidade, seguido da cefotaxima (61,90%), vancomicina (52,38%), norfloxacina (47,62%) e cefalotina (47,62%). A maior resistência foi desenvolvida frente a penicilina e ampicilina, com 95,24% de resistência para os dois antimicrobianos testados. O perfil de susceptibilidade desenvolvido pelas amostras de Enterococcus gallinarum, mostrou baixa sensibilidade frente aos antimicrobianos testados, onde os maiores índices foram observados frente eritromicina e gentamicina, com 33,34% de sensibilidade para ambos; quanto à resistência desenvolvida, foi possível observar 100% de resistência com relação a vancomicina e tetraciclina, seguindo-se cloranfenicol, penicilina, ampicilina, cefoxitina, cefalotina, cefotaxima, norfloxacina e nitrofurantoína, todas evidenciando uma resistência de 83,33% das amostras testadas.


#15 - Sensitivity and specificity of the Califomia Mastitis Test as a diagnostic tool for subclinical mastitis in quarter somatic cell count estimation

Abstract in English:

The California Mastitis Test (CMT) is a simple and widely used diagnostic tool for subclinical mastitis. lt is used even in areas where laboratory facilities are available for diagnosis and monitoring program purposes. CMT usually score 1-5, where 1 indicates a completely negative reaction and 2-5 increasing degrees of inflammatory reaction of the udder. The reactions 2-5, 3-5 or 4-5 may be considered as indicative of subclinical mastitis, because they are related to an increase of the somatic cell count (SCC) in the milk. This variation, however, may produce either false-positive or false-negative results. The aim of this study was to assess the sensitivity and specificity of the CMT in comparison with SCC. A total of 3,012 quarter milk samples from 760 lactating cows were examined. CMT was evaluated at the moment of sampling at cow side and SCC in the laboratory, with a fluoro-opto-electronic method (Fossomatic 90). The average CCS (x 1,000 cells/ml) for CMT scores were: 1 (79.9), 2 (333.5), 3 (670.3), 4 (1,354.0) and 5 (4,455.6). Three options for CMT interpretation were evaluated in relation to a range of CCS, starting from 100,000 cells/ml: (a) 1 versus 2, 3, 4, and 5; (b) 1 and 2 versus 3, 4 and 5; and (e) 1, 2 and 3 versus 4 and 5. The sensitivities of CMT scoring to detect quarters with SCC >200,000/ml were 79%, 61% e 34%for options a, b ande, respectively. The sensitivities of CMT scores for SCC >500,000/ml for options a, b ande, were, respectively, 93%, 82% e 54%. The sensitivity of CMT scores to identify subclinical mastitis was satisfactory (above 80%) when options b ande were used. The interpretation of CMT score 3 as negative for subclinical mastitis was considered as adequate (sensitivity around 80%) only when CCS ranged between 1,200,000 and 1,400,000 cells/ml. The specificities of CMT scoring for CCS of 200,000 and 500,000 were, respectively, 90% and 80% (option a), 97% and 90% (option b) and 99% and 97% (option e).

O "California Mastitis Test" (CMT) estima o conteúdo de células somáticas no leite e é interpretado subjetivamente, estabelecendo-se escores que, na maioria dos casos, variam de 1 a 5. O escore 1 indica uma reação completamente negativa e os de 2-5 indicam graus crescentes de resposta inflamatória do úbere, sendo normalmente considerados como indicativos de mastite subclínica. Dependendo da interpretação dos escores, o CMT pode produzir resultados falso-positivos ou falso-negativos. Esse trabalho teve o objetivo de avaliar a sensibilidade e a especificidade do CMT em relação à contagem de células somáticas (CCS). Foram utilizadas 3.012 amostras de leite provenientes de 760 vacas Holandesas ou mestiças Holandês-Zebu, pertencentes a 15 rebanhos. Todas as amostras foram submetidas ao CMT e processadas para CCS em equipamento Fossomatic 90. Os valores médios de CCS (x 1.000 células/mi) obtidos para os escores de CMT foram 1 (79,9), 2 (333,5), 3 (670,3), 4 (1.354,0) e 5 (4.455,6). Três opções de interpretação (doente/não-doente) para o CMT foram testadas, em relação aos valores de CCS, iniciando com 100.000 células/ml: (a) 1 versus 2, 3, 4, e 5; (b) 1 e 2 versus 3, 4 e 5; (c) 1, 2, 3 versus 4 e 5. As sensibilidades do CMT em identificar corretamente quartos mamários acima de 200.000 células/mi foram 79%, 61% e 34%, para as opções a, b e e, respectivamente. Para identificar corretamente contagens acima de 500.000 células/mi, as sensibilidades do CMT, para as opções a, b e e, foram, respectivamente: 93%, 82% e 54%. A sensibilidade do CMT em identificar quartos mamários com mastite subclínica foi adequada (acima de 80%) quando a interpretação do teste foi mais rigorosa (opções a e b). A interpretação da reação 3 como negativa (opção e) só alcançou sensibilidade de 80% para contagens entre 1.200.000 e 1.400.000 células/mi. As especificidades do CMT, para CCS de 200.000 e 500.000 foram, respectivamente, 90% e 80% (opção a), 97% e 90% (opção b) e 99% e 97% (opção e).


#16 - Influence of subclinical staphylococci mastitis on physical, chemical and cellular milk characteristics

Abstract in English:

One hundred twenty-six milk samples from 63 apparently healthy cows, but positive in the California Mastitis Test were submitted to determinations of pH, acidity, density, butter-fat, total solids, non-fat solids, cryoscopic point, caseine level, chloride level And polymorphonuclear leukocytes. Forty-one cows were infected by coagulase-positive Staphylococcus and 22 by coagulase-negative Staphylococcus. The results obtained in the milk sample analysis from the healthy quarters and infected by coagulase-positive Staphylococcus showed variations of all constituents investigated. However only the pH values (F=4.17*) and the polymorphonuclear leukocytes (F=ll.35**) showed significant differences. On the other hand, between the milk samples from healthy quarters and infected quarters by coagulase-negative Staphylococcus, only the polymorphonuclear leukocytes (F=16.29**) showed significant differences.

Foram submetidas às determinações do pH, acidez titulável, densidade, teor de gordura, extrato seco total, extrato seco desengordurado, ponto crioscópico, teor de cloreto, teor de caseína e às contagens de leucócitos polimorfonucleares, 126 amostras de leite procedentes de 63 vacas aparentemente sadias, porém, positivas ao Califórnia Mastitis Test, das quais 41 mostraram-se infectadas por cepas de Staphylococcus coagulase positiva e 22 por cepas de Staphylococcus coagulase negativa. Apesar de ter sido observada a variação de todos os constituintes investigados entre as amostras de leite procedentes de quartos sadios e infectados por Staphylococcus coagulase positiva, apenas os valores de pH (F=4,17*) e das contagens de leucócitos polimorfonucleares (F=ll,35**), mostraram diferenças estatisticamente significativas. Por outro lado, verificou-se, também, que entre as amostras de leite oriundas de quartos sadios e infectados por Staphylococcus coagulase negativa, apenas os valores das contagens de leucócitos polimorfonucleares (F=l6,29**) apresentaram diferenças estatisticamente significativas.


#17 - Effect of the etiological agent of subclinical mastitis on the production of milk

Abstract in English:

Fifty-five cows were selected from three dairy farms with a high incidence of subclinical mastitis. The Califomia Mastitis Test (CMT) performed on the milk of these animais had revealed 70 pairs of quarters, only one of which reacted positively. During a three-day period, milk from the individual quarters was weighed, recorded and the mean calculated. A comparison of the milk production from these individual quarters revealed that those with subclinical mastites produced 25.4% less milk than the opposite normal quarters. Laboratory tests confirmed the presence of bacterial infections in samples of milk from 41 of the affected quarters. In 15 quarters infected with Streptococcus sp., production of milk was reduced 42.9%. In 26 quarters infected with Staphylococcus sp., the reduction was 27.4%. In contrast, 30 normal pairs of quarters showed a productivity difference of only 1.1 %.

Em três rebanhos com alta incidência de mastites subclínicas, foram escolhidas, ao acaso, 55 vacas que, pelo Teste de Mastite da Califórnia (CMT), revelaram 70 pares de quartos, anteriores e posteriores, dos quais um quarto reagia positivamente em vários graus de intensidade enquanto no oposto a reação era negativa. O exame bacteriológico confirmou a existência da infecção por Streptococcus agalactiae, S. dysgalactiae, S. uberis, Staphylococcus aureus e S. epidermidis nas amostras de leite dos quartos afetados. Durante três dias consecutivos, as vacas foram ordenhadas, manualmente, em baldes com quatro subdivisões móveis, pesando-se o leite de cada quarto e calculando-se a média diária por quarto. O efeito da mastite foi avaliado comparando-se a produção de leite dos quartos normais com a dos quartos opostos portadores da mastite subclínica causada pelos agentes etiológicos acima citados. A comparação da produção de leite dos 70 pares de quartos opostos revelou que os quartos com mastite subclínica diagnosticada pelo CMT produziram 25,4% menos leite do que nos quartos normais. A comparação da produção de leite de 41 pares de quartos opostos, cuja reação positiva ao CMT foi complementada pelo exame bacteriológico, revelou que a redução da produção de leite em quartos com mastite subclínica variou de acordo com o agente etiológico. Em 15 quartos infectados por Streptococcus (S. agalactiae 8 pares, S. dysgalactiae 6 pares e S. uberis l par), a redução foi de 42,9%. Em 26 quartos infectados por Staphylococcus (S. aureus 11 pares e S. epidermidis 15 pares) a redução da produção de leite foi de 27,4% em relação aos quartos opostos normais. A comparação de 15 pares de quartos anteriores e 15 posteriores normais, revelou apenas diferenças médias de 0,5% e 1, 7%, respectivamente, entre os quartos de cada par.
