#21 - Prevalence and anatomopathological findings of primary cardiac neoplasms of the heart base tissues and metastases in dogs from southern Minas Gerais, Brazil (1994-2009)

Abstract in English:

The purpose of this study was to determine the prevalence and anatomopathological findings of primary and metastatic heart tumors and from heart base tumors diagnosed in dogs from southern Minas Gerais, Brazil. The diagnosis was based on histological characteristics and immunophenotyping of the tumors cells. From 1994 to 2009, 2397 necropsies of dogs were performed. Cardiac neoplasms were diagnosed in 31 dogs (1.29%), where seven (22.58%) were primary and 24 (77.42%) were metastatic. Neoplasms of the heart diagnosed as primary were hemangiosarcoma (1/7) and myxoma (1/7), and, primary of heart base, the chemodectoma (3/7) and ectopic thyroid carcinoma (2/7). Frequency was higher in male dogs (5/7), of Boxer breed (3/7) with mean age of 9.7±1.88 years. In metastatic neoplasms, the frequency was higher in female dogs (15/24), of Rottweiler breed (7/24) with the mean age of 7.5±3.23 years. The T cell lymphoma (7/27) was the most frequent neoplasm, diagnosed as metastatic.

E-mail: dantas.af@uol.com.br This paper describes the frequency of squamous cell carcinoma (SCC) diagnosed at the Laboratory (LPA) of Animal Pathology, Veterinary Hospital of the Federal University of Campina Grande (UFCG), in cattle, sheep, goats, and horses in the semiarid region of Paraíba, Brazil, from 1983 to 2010, analyzing epidemiological data and risk factors. We performed the analysis of risk factors, by the chi-square test, considering variables such as species, breed, sex, age and location of the tumor mass. During the period, 3153 diagnoses were recorded from biopsies and necropsies. From these, 81 cases (2.7%) were squamous cell carcinomas. The frequency by species was 4% (42/1052) in cattle, 2.5% (15/603) in horses, 1.7% (12/709) in sheep, and 1.5% (12/789) in goats, being significantly higher in cattle (p<0.001). All cases showed histological features of SCC, varying only the degree of cell differentiation. In cattle and goats, the frequency of SCC was significantly higher in adult animals (p<0.001 and <0.005, respectively). In cattle the preferred location was in the eyes and periocular region (p<0.001), in sheep in the skin (p=0.018), especially the head. In other species no significant differences were found in the tumor location. It is suggested that the higher frequency of SCC in cattle is due to the constitution of the herds, formed predominantly by females of the Holstein breed.

#23 - phological and immunohistochemical characterization of 172 cutaneous round cell tumours in dogs, 32(8):772-780

Abstract in English:

ABSTRACT.- Araújo M.R., Preis I.S., Lavalle G.E., Cassali G.D. & Ecco R. 2012. Histomorphological and immunohistochemical characterization of 172 cutaneous round cell tumours in dogs. Pesquisa Veterinária Brasileira 32(8):772-780. Setor de Patologia Veterinária, Escola de Veterinária, Universidade Federal de Minas Gerais, Av. Antônio Carlos 6627, Belo Horizonte, MG 31270-901, Brazil. E-mail: ecco@vet.ufmg.br This paper describes the use of a panel of antibodies (CD117, CD3, CD79a, CD45, cytokeratin, vimentin and E-cadherin) on formalin-fixed, paraffin-embedded sections of canine cutaneous round cell tumours. Neoplastic tumours were diagnosed by histology and histochemical stains and included 107 mast cell tumours, 31 cutaneous histiocytomas, two localized histiocytic sarcomas, 21 cutaneous lymphomas, three plasma cell tumours, one transmissible venereal tumour and seven unclassified round cell tumours. The histologic diagnosis was modified in 39.5% of the total 172 neoplasms. The staining for CD45 and E-cadherin were variable, and therefore, the final diagnoses of cutaneous histiocytoma and localized histiocytic sarcoma were made based on histology in association with negative results for CD3, CD79a, CD117 and cytokeratin. The cellular origin of unclassified round cell tumours was defined in all cases. Cutaneous B-cell lymphoma and plasma cell tumours were CD79a-positive and could be distinguished from each other by the morphological characteristics. Mast cell tumours and T cell lymphoma were CD117 and CD3 positive, respectively. The positive staining for vimentin and the negative staining for CD3, CD79a, CD117 and cytokeratin favoured the diagnosis of transmissible venereal tumours. Thus, the final diagnosis of cutaneous round cell tumours should be based on the interpretation of immunohistochemical results together with the cellular morphology observed by histology. Therefore, more studies to optimize the specific markers in formalin-fixed, paraffin-embedded tissues (especially for histiocytes) are required for definitive diagnosis of round cell tumours in dogs.

E-mail: cmuramoto@gmail.com Evaluation of lymph nodes is part of the staging of the mammary tumors and helps to establish a prognosis and therapy. B-mode ultrasonography was used to evaluate size, edge, nodal borders, shape, architecture, echotexture and echogenicity of lymph nodes and Doppler ultrasonography to evaluate the quantity and distribution of the vessels. The aims of this work were to identify which ultrasound features can be used to classify a lymph node as metastatic or non-metastatic; to establish its accuracy as a useful tool to this differentiation, and to establish procedures that permit the reproducibility of the results. Sixty seven lymph nodes of 30 female dogs with mammary tumors were examined by ultrasound and classified as metastatic or non-metastatic. The ultrasonographic diagnostic impression was confronted with the histopathologic results. The ultrasonographic classification of the lymph nodes was correct in 92.5% of the cases, with sensibility of 94.1%, specificity of 92%, positive predictive value of 0.8, and negative predictive value of 0.9787.The characteristics of the nodes to classify them as metastatic or non metastatic were registered. Ultrasound imaging of regional lymph nodes is important to detect metastases and it is suggested that it becomes a regular procedure in the staging of canine mammary tumors.

E-mail: debora.zuccari@famerp.br E-cadherin is a cell-cell adhesion molecule and low e-cadherin expression is related to invasiveness and may indicate a bad prognosis in mammary neoplasms. The expression of cell proliferation markers PCNA and especially Ki-67, has also proved to have a strong prognostic value in this tumor class. The expression of these markers was related to the clinical-pathological characteristics of 73 surgically removed mammary tumors in female dogs by immunohistochemistry. There was no statistical correlation between these markers and death by neoplasm, survival time and disease-free interval. However, the loss of e-cadherin expression and marked Ki-67 expression (p=0.016) were considered statistically significant for the diagnosis (p=0.032). When evaluated as independent factors, there was evidence of the relationship between the loss of e-cadherin expression and high PCNA expression with changes in the body status (divided into obese, normal and cachectic) of female dogs (p=0.030); there was also evidence of the relationship between pseudopregnancy and e-cadherin alone (p=0.021) and for ulceration and PCNA alone (p=0.035). The significant correlation between the markers expression and these well known prognostic factors used individually or in combination suggests their prognostic value in canine mammary tumors.

Pesquisa Veterinária Brasileira 26(2):112-122. Departamento de Patologia, Universidade Federal de Santa Maria, 97105-900 Santa Maria, RS, Brazil. E-mail: glaukommers@yahoo.com Thirty bovine with neoplasms of the upper digestive tract (UDT) associated with spontaneous ingestion of bracken fern (Pteridium aquilinum) were studied. They were from 27 farms, located in the municipalities of Jaguari (23) and Nova Esperança do Sul (4), Rio Grande do Sul, Brazil. The total cattle population in those farms was 1,090 and large amounts of bracken fern were found in the pastures. Twenty-six of the affected cattle were cows and four were castrated males, 3-13 years of age; most of them were 7-8 years old (46,6%). Clinical signs observed in the affected animals were progressive weight loss, absence of ruminal movements, cough, dysphagia, regurgitation, halitosis, diarrhea, and bloat. Less frequent signs were selective appetite, dyspnea, and salivation. Two bovine died and 28 were submitted to euthanasia in advanced stage of disease and necropsied. The main gross and microscopic alterations were found in identical areas of the UDT. They consisted of papillomas, transforming papillomas, and squamous cell carcinomas (SCCs). Metastases of SCCs to regional lymph nodes and other organs, such as liver and lungs, were also observed (18/30). Twenty-nine bovine had papillomas of various sizes in several areas of the UDT. The digestive papillomatosis ranged from mild (45%), to moderate (38%), to severe (17%). Three developing phases were observed microscopically in the examined papillomas: an early growing phase, a developing phase, and a regressing phase. In 16 cases, there was malignant transformation of papillomas into SCCs. The SCCs were solitary (12/30) or multiple (18/30) and were histologically well, moderately, or poorly differentiated. Grouping the distribution of SCCs of larger extension in the UDT into cranial region (base of the tongue, pharynx/oropharynx, and epiglottis), medial region (esophagus), and caudal region (cardia and rumen), the distribution was cranial in 39%, middle in 16%, and caudal in 45% of the cases. By the same grouping criteria, but considering the total number of times SCCs of varied extensions were diagnosed in the cranial, middle, and caudal regions, the percentages changed to 34%, 26%, and 40%, respectively. The epidemiological and histomorphological evidences found in this study are in agreement with the observations that point out the co-carcinogenesis between bovine papillomavirus type 4 infection and chemicals of bracken fern in the pathogenesis of the SCCs in the UDT of cattle. However, the presence of pre-neoplastic changes and SCCs in situ or in early stages of development, independently of the presence of papillomas, clearly indicates the possibility of development of SCCs from normal epithelium, probably due to the direct action of the chemical carcinogens contained in bracken fern.

#28 - A comparative study between cytology and histopathology for the diagnosis of canine neoplasms, 21(1):23-32

Abstract in English:

ABSTRACT.- Magalhães A.M., Ramadinha, R.R., Barros C.S.L. & Peixoto, P.V. 2001. [A comparative study between cytology and histopathology for the diagnosis of canine neoplasms] Estudo comparativo entre citopatologia e histopatologia no diagnóstico de neoplasias caninas. Pesquisa Veterinária Brasileira 21(1):23-32. Universidade Federal Rural do Rio de janeiro, Seropédica, RJ 23851-970, Brazil. A comparative study between cytology and histopathology of various neoplasms from 150 dogs was carried out. Severa) staining methods including Wright, May-Gri.inwald-Giemsa, NewMethylene Blue and Papanicolau, and histological staining methods such as Hematoxilin & Eosin, van Gieson, Sudan, Toluidine Blue and PeriodicAcid Schiffwere used. The data obtained indicated an 85.3% accuracy for the cytological diagnosis when the histopathological diagnosis was considered as the correct parameter. In 4.0% of the cases only the embrionary origin of the neoplasrns was determined, and in 1.3% only the prognosis was established. The cytological diagnosis differed from the histopathological diagnosis in 8.1% of the cases. In two cases (1.3%), although the cytological anel histopathological diagnoses differed, a reexamination determined that the former was correct which increased its accuracy up to 86.6%. Fine needle aspiration biopsy proved to be the best technique to obtain samples for the study. Cytology was found not suitable for the diagnosis of mammary neoplasms due to the wide variation of the cytological aspects from different areas. The imprint on slides is a technique not recommended to evaluate mesenchymal tumors and should be replaced by exfoliative cytology. The Wright method proved to be the more efficient staining method. Adapted staining methods from histopathology such as van Gieson for leiomyomas and leiomyosarcomas, Sudan Black B for lipomas anel liposarcomas, and Periodic Acid of Schiff and Toluidine Blue for mastocytomas, were especially useful in visualizing the fine morphological details of various epithelial anel mesenchymal neoplasms examined.

RESUMO.- Magalhães A.M., Ramadinha, R.R., Barros C.S.L. & Peixoto, P.V. 2001. [A comparative study between cytology and histopathology for the diagnosis of canine neoplasms] Estudo comparativo entre citopatologia e histopatologia no diagnóstico de neoplasias caninas.

#29 - Squamous cell carcinomas in_sheep on a farm in southem Brazil

Abstract in English:

Squamous cell carcinomas in Polwarth sheep were studied during two years on a farm in southern Brazil. The first case was observed in December 1977. From this date to March 1980, 14 additional sheep in a flock of 792 were affected. Eighteen neoplasms were observed in 15 sheep. Twelve of them were located on the muzzle, three on the ears, two on the eyelids and one on the lower lip. Nine neoplasms were surgically removed and only one recidivated during the observation period. Macroscopic and histologic appearance of the neoplasms are described. No metastases were detected in six necropsied sheep. The high incidence of neoplasms was assumed to be a consequence of the large percentage of old sheep in the flock.
